▼ The last few decades have seen growing concern about climate change caused by global warming, and it now seems that the very future of humanity depends on saving the environment. With recognition of CO2 emissions as the primary cause of global warming, their reduction has become critically important. An effective method of achieving this goal is to focus on the sectors that represent the greatest contribution to these emissions: electricity generation and transportation. For these reasons, the goal of the work presented in this thesis was to address the challenges associated with the accommodation of a high penetration of plug-in electric vehicles (PEVs) in combination with renewable energy sources. Every utility must consider how to manage the challenges created by PEVs. The current structure of distribution systems is capable of accommodating low PEV penetration; however, high penetration (20 % to 60 %) is expected over the next decades due to the accelerated growth in both the PEV market and emission reduction plans. The energy consumed by such a high penetration of PEVs is expected to add considerable loading on distribution networks, with consequences such as thermal overloading, higher losses, and equipment degradation. A further consideration is that renewable energy resources, which are neither exhaustible nor polluting, currently offer the only clean-energy option and should thus be utilized in place of conventional sources in order to supply the additional transportation-related demand. Otherwise, PEV technology would merely transfer emissions from the transportation sector to the electricity generation sector. As a means of facilitating the accommodation of high PEV penetration, this thesis proposes methodologies focused on two main themes: uncontrolled and coordinated charging. For uncontrolled charging, which represents current grid conditions, the proposal is to utilize dispatchable and renewable distributed generation (DG) units to address the high PEV penetration in a way that would not be counterproductive. This objective is achieved through three main steps. First, the benefits of allocating renewable DG in distribution systems are investigated, with different methodologies developed for their evaluation. The benefits are defined as the deferral of system upgrade investments, the reduction in the energy losses, and the reliability improvement. The research also includes a proposal for applying the developed methodologies for an assessment of the benefits of renewable DG in a planning approach for the optimal allocation of the DG units. The second step involves the development of a novel probabilistic energy consumption model for uncontrolled PEV charging, which includes consideration of the drivers’ behaviors and ambient temperature effect associated with vehicle usage. ▼ The Locus Control Region (LCR) of the hCD2 gene is able to confer T-cell specific, copy-number dependent and position-independent expression of linked reporter genes in transgenic mice. These properties of the LCR have been exploited for the generation of an expression vector potentially useful in the T-cell specific gene therapy protocols. The hCD2 LCR consists of a strong T-cell specific enhancer and an additional regulatory element without enhancer activity (designated HSS3) which is required for prevention of Position Effect Variegation (PEV) in transgenic mice. To identify trans-acting proteins binding to the HSS3 and, potentially, mediating the LCR function, in vitro footprint analysis of the HSS3 region was performed using thymus and liver nuclear protein extracts. Three of the identified footprinted regions (FT1-FT3) were used as baits in the one- hybrid yeast technique to isolate cDNAs from a T-cell specific library. Using the FT1 sequences as a bait, a 1.3 kb DNA insert coding for the HMG box of the human homologue of rat HBP1 protein was isolated. The rest of the open reading frame (ORF) was isolated from a Lamda-ZAP library. DNA- binding properties of the recombinant hHBP1 was characterised using DNAse I footprint, gel retardation and methylation interference experiments. It was found that HBP1 binds not to a canonical HMG consensus sequence, but to a novel TTCATTCATTCA motif which is higher in affinity than other recently reported HBP1 binding sites. To assess the contribution of FT1 to LCR function, the footprint sequences were deleted from the LCR and transgenic mice carrying a ΔFT1-LCR hCD2 minigene were generated. Expression patterns of the ΔFT1-LCR hCD2 transgenes were analysed by FACS and the integration sites were identified by FISH and Southern blot. The deletion of FT1 sequences resulted in variegation of expression of the ΔFT1-LCR hCD2 transgene when the transgene integrates into heterochromatic regions adjacent to the centromere or telomere. However, the degree of variegation was not as dramatic as one caused by the deletion of the whole of HSS3, indicating that the other HSS3-binding factors may also contribute to prevention of PEV. Sequences of cis regulatory elements other than the LCR were examined for affects on hCD2 transgene expression. This part of the work resulted in an improved version (with 10 x higher expression level) of the hCD2 expression cassette. Subsequently, related experiments have shown that introducing the fourth intron of the hCD2 gene into the improved vector coould result in further 4-5x increase in expression level of the cassette.

▼ Very little information is known about the impact electrification has on driving behavior, or how drivers charge their electrified vehicles. The recent influx of electrified vehicles presents a new market of vehicles which allow drivers the option between electrical or conventional gasoline energy sources. The current battery capacity in full battery electric vehicles requires planning of routes not required of conventional vehicles, due to the limited range, extended charging times, and limited charging infrastructure. There is currently little information on how drivers react to these limitations. A number of current models of fully electric and plug-in hybrid electric vehicles, transmit data wirelessly on key-on, key-off, and charging events. The data includes battery state of charge, distance of miles driven on gasoline and electric, energy consumed, and many other parameters associated to driving and charging behavior. In this thesis, this data was then processed and analyzed to benchmark the performance and characteristics of driving and charging patterns. Vehicles were analyzed and contrasted based on model type, geographic location, length of ownership and other variables. This data was able to show benchmarks and parameters in aggregate for 56 weeks of electrified vehicle tracking. These parameters were compared to the EV Project, a large scale electrified vehicle study performed by Idaho National Labs, to confirm patterns of expected behavior. New parameters which were not present in the EV Project were analyzed and provided insight to charging and driving behavior not examined in any previous study on a large scale. This study provides benchmarks and conclusions on this new driving behavior, such as large scale analysis of brake regeneration performance and degradation of range anxiety. Analysis of the differences on charging and driving behavior between geographic regions and experience were examined, providing insight to how these variables affect performance and driving and charging patterns. Comparison of parameters established by the EV Project and new parameters analyzed in this report will help build a benchmark for future studies of electrified vehicles. ▼ Key battery, semiconductor, and software technologies have sufficiently progressed over the past few decades to enable viable plug-in electric vehicle (PEV) alternatives to conventional vehicles. Alternatives to petroleum-based fuels for transportation are sought to address concerns over energy security, foreign oil derived U.S. trade deficits, oil related geopolitical entanglements, and emissions. The various types of PEVs have substantially different characteristics. The types and key attributes of PEVs, charging standards, and charging locations are described. The likely scenario for PEV-Grid interactions over the next decade is synthesized from the analysis of the technologies available to and circumstances of vehicle manufacturers, utilities, and supplier firms. PEV adoption considerations are evolving. Many lessons have been learned from the first generation of PEVs that were introduced starting in late 2010. Technology, market, and policy drivers of emerging trends in the diffusion of PEVs are explored more in-depth. PEVs as electric loads are unique in that they are large, flexible, and intelligent. These attributes can not only provide utilities a new source of revenue, but also improve grid stability and economics. Actions, technologies, and policies that utilities can deploy to increase adoption are discussed. Actions are explored to make the overall PEV ownership experience superior to a conventional vehicle. This dissertation also describes research of the capability for PEVs in Vehicle to Home (V2H) scenarios, where the vehicle acts as a residential battery storage system and/or a backup generator in a residential microgrid configuration during a grid outage. Residential energy data collected from a smart grid testbed is used with a custom PEV model to assess the performance (in terms of duration and power output) of a BEV or PHEV used for backup power. Our earlier results quantify the extent to which photovoltaic (PV) generation and the characteristics of a PEV (battery size, gasoline availability) affect the backup duration during an electric grid outage. Strategies to further increase backup duration and non-continuous self-sustaining off-grid alternatives were found in our early V2H research. Varied amounts of load curtailment and PHEV engine-generator control improvements are modeled in subsequent research. When the low voltage electric grid is dimensioned electric loads are predicted by analyzing the area by certain factors such as geographical data, customer type, heating method etc. So far, the charging of Plugin Electric Vehicles (PEVs) is not considered as one of these factors. Approximately 30% of the distribution grid in Sweden is projected to need reinforcements due to the increased loads from PEVs during winters if the charging isn’t controlled. In addition to this Stockholm face the problem of capacity shortage from the transmission grid, limiting the flow of electricity into the city. This research is therefore conducted to analyze the impact that the increase of PEVs will have on the distribution grid in the future. This thesis simulates the electric grid for three substations located in Hammarby Sjöstad by using power flow analysis and electric grid data from 2016. To approach this problem a method to disaggregate the total power consumption per substation into power consumption responding to each building was developed. In addition to this the number of PEVs in the future was projected. Nine different scenarios were used to compare different outcomes for the future, namely the years of 2025 and 2040. In order to simulate the worst possible case for the electric grid all the PEVs were assumed to be charged at the same time, directly when arriving home on the Sunday when the power demand peaks in 2016. The results indicate that PEVs can have a considerable impact on the components of the low voltage distribution network and controlled charging should be implemented. By examining the impact on the simulated electric grid from the different scenarios the limit of PEV penetration is found. In the area of Hammarby this limit seems to be around 30 % of the total cars if there is no controlled charging. Without any controlled charging the peak power demand increases by 30% with a 30% share of PEVs, which is projected to happen in 2025. In 2040 when share of PEVs is projected to be about 95% the peak power is instead increased by more than 100% which shows the impact that PEVs can exert on the electric grid. Utilizing a simple method of controlled charging where the PEVs are instead charged during the night when the power demand is low, the peak power is not increased at all. This also results in the small cost benefit for PEV owners since the electricity is cheaper during the night and controlled charging can therefore save about 15% of the electricity charging cost. However, the main savings are for the grid owners since the need to reinforce the grid is heavily reduced. In addition to this the power losses are reduced heavily from about 14% down to 5% in the electric grid that is simulated.

▼ Plug-in electric vehicles (PEVs) represent a huge step forward in a green transportation system, contribute to the reduction of greenhouse gas emission, and reduce the dependence on fossil fuel. With the increasing popularity of PEVs, public electric-vehicle charging infrastructure (EVCI) becomes indispensable to meet the PEV user requirements. EVCI can consist of various types of charging technologies, offering multiple charging services for PEV users. Proper integration of the charging infrastructure into smart grid is key to promote widespread adoption of PEVs. Planning and operation of EVCI are technically challenging, since PEVs are characterized by their limited driving range, long charging duration, and high charging power, in addition to the randomness in driving patterns and charging decisions of PEV users. EVCI planning involves both the siting and capacity planning of charging facilities. Charging facility siting must ensure not only a satisfactory charging service for PEV users but also a high utilization and profitability for the chosen facility locations. Thus, the various types of charging facilities should be located based on an accurate location estimation of the potential PEV charging demand. Capacity planning of charging facilities must ensure a satisfactory charging service for PEV users in addition to a reliable operation of the power grid. During the operation of EVCI, price-based coordination mechanisms can be leveraged to dynamically preserve the quality-of-service (QoS) requirements of charging facilities and ensure the profitability of the charging service. This research is to investigate and develop solutions for integrating the EVCI into the smart grid. It consists of three research topics: First, we investigate PEV charging infrastructure siting. We propose a spatial-temporal flow capturing location model. This model determines the locations of various types of charging facilities based on the spatial-temporal distribution of traffic flows. In the proposed model, we consider transportation network dynamics and congestion, in addition to different characteristics and usage patterns of each charging facility type. Second, we propose a QoS aware capacity planning of EVCI. The proposed framework accounts for the link between the charging QoS and the power distribution network (PDN) capability. Towards this end, we firstly optimize charging facility sizes to achieve a targeted QoS level. Then, we minimize the integration cost for the PDN by attaining the most cost-effective allocation of the energy storage systems and/or upgrading the PDN substation and feeders. Additionally, we capture the correlation between the occupation levels of neighboring charging facilities and the blocked PEV user behaviors. ▼ Vehicle electrification is an effective way to reduce petroleum consumption, and the emissions caused by conventional vehicles. However, as market penetration of PEVs (Plug-in Electric Vehicle) increases, charging could present some serious challenges to the current electric power grid system. When considering the characteristics of electric power generation, transmission and distribution in the U.S., there is general agreement that local distribution is the most likely part to be adversely affected by the unregulated PEV charging. The increased load that can result from unregulated charging of PEVs could dramatically accelerate the aging of electrical transformers. The objective of this dissertation is to understand and model the aging behavior of residential transformers and to develop control strategies that can mitigate or eliminate the accelerated aging that could result from load peaks caused by PEV charging.The first step in this work is to quantitatively estimate the impact of PEV charging on residential circuits. To this end, a Monte Carlo simulation is developed that considers the stochastic inputs of usage patterns and vehicle characteristics and typical driving cycles to estimate the energy demand of a hypothetical PEV fleet. The result of the Monte Carlo simulation is then used to predict the impact of PEV charging on the life of residential distribution transformers. To estimate transformer life, an aging model obtained from established IEEE standards is used, together with a dynamic thermal model of the transformer used to estimate the transformer winding hot-spot temperature. Thermal modeling is a very important aspect, as aging is predominantly affected by transformer peak temperatures, which are very sensitive to transformer overloads during the warmer months of the year. Thus, the aging model links load patterns to transformer peak temperature through seasonal temperature fluctuations, and permits the assessment of transformer life under a broad range of practical conditions, that take into account regional variations in climate.The aging model makes it possible to develop charging control strategies that protect the transformer system while maximizing overall PEV charging quality. Charging quality is assessed based on the ability to fully satisfy the customer charging demand in a broad range of situations. In particular, the charging control policy makes use of load prediction algorithms using data-driven models that are based on actual electricity consumption data available from the utilityFinally, the dissertation extends the results obtained for the case of residential transformers to the case of public charging stations, wherein vehicle traffic and charging demand is not as easily predictable as in the case of residential dwellings. Queuing theory is employed to model vehicle random arrival and departure times to predict charging power demand and the expected waiting time for service in peak hours. ▼ This thesis has explored how effectively citizens and other stakeholders (community-based organisations (CBOs) and non-governmental organisations (NGOs)) in Peru's Loreto Region have been able to hold the oil company Petroperu accountable for environmental pollution. To do this, a theoretical framework called the political ecology of voice (PEV) has been developed which can be defined as an investigation into a specific temporal political, economic, social, and geographical environment in which citizens and other stakeholders voice over an environmental issue. Semi-structured interviews were concentrated predominantly on two oil-based communities; Barrio Florido adjacent to the Iquitos refinery, and Cuninico situated near the North Peruvian oil pipeline. The PEV theoretical framework has revealed many fascinating issues within the Loreto Region. Poor rural development, economic poverty, political disinterest and racialised class consciousness mean that both communities lack access to state institutions or their representatives to voice. Meanwhile, Petroperu strictly controls their access and relationship which resulted in no means of communication for Cuninico prior to a 2014 oil spill and limited dialogue for Barrio Florido. Citizen poverty and their economic dependency on Petroperu has also strengthened its power and influence over these communities allowing them to establish a climate of fear thus weakening citizen willingness to voice. For CBO/NGO state/resource extraction industry (REI) access and engagement, this can be positively co-operative (stemming from the former's mutual aims and/or conciliatory support) or negatively obstructive (resulting from the former's scrutiny-based voice) with those in the latter affected through state-orchestrated anti-NGO rhetoric and REI fragmentation of indigenous federations. Despite this, these actors perform many useful educational and accountability based roles which strengthen citizen voice. However, their involvement does not always guarantee success due to their own visions which can impact their representation of citizen voice. Overall, the PEV environment for Loreton stakeholders can be described as restrictive, intolerant and suppressive, particularly for citizens who must rely on the support of collective voice actors to hold REIs accountable for pollution.

▼ Les populations d’enfants nés grands prématurés représentent 7% des naissances, soit 15 millions de bébés par an dont 40 % vont présenter des troubles neurodéveloppementaux. Les échecs scolaires sont surreprésentés par rapport à des enfants nés à terme et la lecture est alors constamment atteinte. Nous étudions, au sein d’un travail longitudinal en trois parties, leur langage oral puis leur lecture par les potentiels évoqués. La lecture s’acquiert sur des compétences langagières et visuo-attentionnelles. Nous avons émis l’hypothèse qu’ils présenteraient des troubles moteurs à minima, qui selon la théorie motrice de la perception de la parole, gêneraient le développement de la phonologie. Nous avons, dans une première partie, au sein d’un PHRC national, inclus des enfants nés grands prématurés de 3 ans pour caractériser et stimuler leur langage oral. Nous les avons randomisés en un groupe rééduqué (n=87) et un bras groupe non rééduqué (n= 78) (LAMOPRESCO 1). Les résultats ont montré l’efficacité de la stimulation courte et protocolisée sur des aspects sensorimoteurs du langage à 4 ans. Les enfants rééduqués ont une meilleure progression de leur score en phonologie (p=0.07), et des scores significativement améliorés en lexique de réception (p=0.02) et de production (p<0.01), et en compréhension (p<0.01). Les scores de phonologie étaient corrélés à des scores moteurs, motricité fine et de latéralité (p=0.019). Nous avons développé une méthodologie de PEV en lecture, avec des stimuli mots simples et contrastés dans l’hypothèse de distinguer la voie phonologique motrice de la voie visuelle. Les résultats en Décision Lexicale et en Oddball, de notre population normative adulte (n=19) et enfants normo lecteurs de 12 ans (n=19), questionnent l’onde N170. La population d’enfants appariés dyslexiques (n=16) enregistrée a permis de confirmer les fluctuations maturationnelles de l’onde N170 et de sa co-construction avec l’onde P100 attentionnelle. Le couple P1N1 semble être un marqueur neuro développemental précoce avant l’automatisation de la N170. Le décours temporel positif de P1N1 est corrélé (p=-0.51) pour l’ensemble des populations (n=48) de normo lecteurs et de dyslexiques à notre paramètre de leximétrie (VL). Au décours de cette deuxième partie, la N170, apparait bien être la spécialisation et l’automatisation de la lecture experte, mais le couple P1N1 nous renseigne sur sa construction neurodéveloppementale par le biais d’un probable effet top down maturationnel sur la P100. La position de la N170, est positive chez 56% des enfants nés grands prématurés appariés (n=19) alors qu’on la retrouve chez 39% des enfants dyslexiques. En Oddball, la latence de la P300 mots cibles est corrélée à la leximétrie VL (r=-0.39) dans toutes nos populations étudiées. Pour la troisième partie qui est l’enregistrement par les PEVc de nos enfants de la première partie, maintenant âgés de 8 ans, nous avons simplifié nos stimuli mots. Les recommandations de l’OMS sur la prise en charge des enfants nés de mères infectées par le VIH évoluent en fonction des nouvelles données scientifiques souvent issues d’essais thérapeutiques. Leur application en routine, surtout dans les pays à ressources limitées, devrait prendre en compte les contraintes opérationnelles locales pour guider les pratiques. Ce travail de thèse a été développé dans cette optique, avec pour objectif d’évaluer l’ensemble du processus de prise en charge de ces enfants avant l’âge de sept mois, depuis le dépistage précoce de l’infection VIH jusqu’à la mise en œuvre du traitement antirétroviral (ARV) systématiquement proposé dès le diagnostic de l’infection VIH, et leurs réponses aux vaccins administrés dans le Programme Elargi de Vaccination (PEV). Ce travail est issu de l’analyse des données collectées dans deux enquêtes : la cohorte ANRS-PEDIACAM initiée en 2007 et toujours en cours, menée dans trois hôpitaux urbains du Cameroun, et l’enquête transversale ACIP-EPIPEV, menée de novembre 2004 à Mai 2005 dans cinq hôpitaux du Cameroun et de la République Centrafricaine. Nos résultats ont montré que le diagnostic précoce du VIH et l’initiation des ARV chez les nourrissons étaient faisables et largement acceptés en routine dans les services de pédiatrie urbains. Parmi les nourrissons nés de mères séropositives inclus dans PEDIACAM, 89,7% ont été ramenés pour le test VIH à un âge médian de 1,5 mois (IQ : 1,4-1,6) et 83,9% ont complété le processus de diagnostic en revenant chercher les résultats du test avant l’âge de 7 mois. Les facteurs associés au processus incomplet reflétaient plus la qualité des soins prénataux et l’urgence du contexte obstétrical que les conditions environnementales. Parmi les nourrissons infectés par le VIH, 83,5% ont initié les ARV avant l’âge de 7 mois. Mais le processus d’initiation précoce des ARV a été considéré comme suboptimal chez environ un tiers d’entre eux. Les facteurs associés à ce retard étaient liés au système de soins, à la gestion des prélèvements et des résultats du test VIH. Enfin, dans l’étude EPIPEV, nous avons observé des proportions plus faibles de réponses vaccinales adéquates chez les enfants infectés que non infectés par le VIH, nés de mères séropositives. Cette différence était plus marquée pour le vaccin contre la rougeole (20% vs 60%). Le niveau de réponse observé chez les enfants non infectés nés de mères séropositives était néanmoins faible par rapport à celui attendu chez les enfants en population générale. Ce dernier résultat suggérerait que l’exposition au VIH pendant la grossesse pourrait entrainer un dysfonctionnement immunologique, même en l’absence de transmission du VIH à l’enfant. ▼ Plug-in Electric Vehicles (PEVs) have the potential of reducing gasoline consumption and greenhouse gas emissions in the transportation sector. The net impacts of PEVs – including upstream emissions from electricity generation and the impact these vehicles place on the electricity grid – depend on both the amount of travel conducted by PEV and locations that those PEVs are charged. This dissertation investigates the vehicle use choices and charging decisions of both battery electric vehicles (BEVs) and plug-in hybrid electric vehicles (PHEVs) for both home-based trip tours and long-distance trips using stated preference (SP) data. It presents a novel dynamic discrete choice modeling (DDCM) framework that explicitly accounts for the stochastic nature of the vehicle choice and charging decisions of PEV users: earlier choices on vehicle use and charging influence the utility of the future choices; the expectation of the future options influences those earlier decisions; and choices are made under uncertainty about actual energy consumption and availability of chargers. For home-based trip tours, my results show that BEV users are willing to pay 10-24 to avoid having to deviate from the originally planned route, which indicates that “range anxiety” of BEV owners – the fear of being stranded in the middle of a trip – is not a crucial issue for home-based trips. Using charging infrastructure development to encourage BEV adoption might be more beneficial than reducing “range anxiety” among the current users, which could entail building charging stations at locations that have more public exposure, such as public parking garages in a city center. When BEVs are on long-distance trips, the cost of deviation is significantly higher: 244, which indicates that BEV owners are likely to be more cautious and view finding a charger off the route much more costly when they are on long-distance trips. Comparing the cost of deviation for home-based tours and long-distance trips, to support the existing users, the most cost-effective places to invest in charging infrastructure are inter-city corridors instead of in-city locations. By comparing the relative size of the coefficient estimates, in this dissertation, I also analyze the monetary value of increasing charging power, moving the charging stations closer to highway exits, and having amenities such as restrooms, restaurants, and Wi-Fi near the charging stations. The comparison between the DDCMs and SDCMs based on simpler decision heuristics shows that for home-based tours, DDCMs only offer a little better prediction rate with a significant cost when it comes to computation time and complexity of model development. For the purpose of demand forecasting of a charging network or site selection for the charging facilities, the SDCMs based on simpler heuristics are recommended for home-based trip tours. However, the intermittency of most renewable sources and lack of sufficient storage in the current power grid means that reliable integration of significantly more renewables will be a challenging task. Moreover, increased integration of renewables not only increases uncertainty, but also reduces the fraction of traditional controllable generation capacity that is available to cope with supply-demand imbalances and uncertainties. Less traditional generation also means less rotating mass that provides very short term, yet very important, kinetic energy storage to the system and enables mitigation of the frequency drop subsequent to major contingencies but before controllable generation can increase production. Demand, on the other side, has been largely regarded as non-controllable and inelastic in the current setting. However, there is strong evidence that a considerable portion of the current and future demand, such as electric vehicle load, is flexible. That is, the instantaneous power delivered to it needs not to be bound to a specific trajectory. In this thesis, we focus on harnessing demand flexibility as a key to enabling more renewable integration and cost reduction. We start with a data driven analysis of the potential of flexible demands, particularly plug-in electric vehicle (PEV) load. We first show that, if left unmanaged, these loads can jeopardize grid reliability by exacerbating the peaks in the load profile and increasing the negative correlation of demand with wind energy production. Then, we propose a simple local policy with very limited information and minimal coordination that besides avoiding undesired effects, has the positive side-effect of substantially increasing the correlation of flexible demand with wind energy production. Such local policies could be readily implemented as modifications to existing "grid friendly" charging modes of plug-in electric vehicles. We then propose improved localized charging policies that counter balance intermittency by autonomously responding to frequency deviations from the nominal frequency and show that PEV load can offer a substantial amount of such ancillary services. Next, we consider the case where real-time prices are employed to provide incentives for demand response. We consider a flexible load under such a pricing scheme and obtain the optimal policy for responding to stochastic price signals to minimize the expected cost of energy. We show that this optimal policy follows a multi-threshold form and propose a recursive method to obtain these thresholds. We then extend our results to obtain optimal policies for simultaneous energy consumption and ancillary service provision by flexible loads as well as optimal policies for operation of storage assets under similar real-time stochastic prices. We prove that the optimal policy in all these cases admits a computationally efficient form. Chamando atenÃÃo para o carÃter pedagÃgico das atividades realizadas nas AVD, esse atendimento passou a ser pensado nÃo somente como um treinamento, mas tambÃm como um importante instrumento favorecedor de aprendizagem, que podem levar a crianÃa com deficiÃncia visual a desenvolver-se de forma integral. Por esse carÃter pedagÃgico Ã que hoje o chamamos de Pevi. Para entender essa relaÃÃo, os referenciais teÃricos utilizados foram Arruda (2001) e Bruno (1997), Ã luz da teoria de desenvolvimento cognitivo de Piaget. Iniciamos essa investigaÃÃo com o estudo sobre as Pevi, a fim de descrever as suas principais atividades e os procedimentos necessÃrios a realizaÃÃo das mesmas. Em seguida, realizamos uma anÃlise prÃvia dos conteÃdos matemÃticos que podem ser abordados atravÃs da realizaÃÃo do passo a passo dessas atividades, paralelamente a um estudo piloto, o qual constou de observaÃÃes na sala da educaÃÃo infantil e na sala das AVD do Instituto dos Cegos de Fortaleza. ▼ Chromatin structure is a fundamental determinant of eukaryotic gene expression and it is composed of two chromatin environments, euchromatin and heterochromatin. Euchromatin provides an accessible platform for transcription factors; hence it is permissive for gene expression. Heterochromatin on the other hand is highly compacted and inaccessible, which in most cases leads to transcriptional repression. A locus that is composed of both of these environments is the ribosomal DNA (rDNA). In eukaryotes the rDNA is composed of hundreds to thousands of tandemly repeated genes where maintaining both silent and active copies is fundamental for the stability of the genome. The aim of this research was to investigate the role of the rDNA in gene expression in Drosophila melanogaster. In D. melanogaster the rDNA loci are present on the X and Y chromosomes. This research used the Y-linked rDNA array to investigate the role of this locus on gene expression. A genetic and molecular strategy was designed to create and quantify specific, graded and isogenic Y- linked rDNA deletions. Then the deletions were used to address the effect of rDNA deletions on gene expression using reporter genes sensitive to Position Effect Variegation (PEV). In addition, the effect of the deletions in nucleolus size and structure as well as the effect of spontaneous rDNA deletions on gene expression were tested in this study. This research found that changes in rDNA size change the chromatin balance, which resulted in increased expression of the reporter genes, decreased nucleolus volume, and altered nucleolus structure. These findings prompted a further research question on whether this effect on gene expression occured globally in the genome. This was addressed by performing microarray analysis where the results showed that rDNA deletions affect about half of the genes on the genome. Presented in this dissertation is evidence that suggest a novel role for the rDNA is a global modulator of gene expression and also is a contributor to the gene expression variance observed in natural populations. ▼ The rapid development of plug-in electric vehicles (PEV) brings both challenges and opportunities to the power system. This dissertation studies the modeling and optimal control of vehicle-grid integration (VGI) system with the consideration of both grid-side and PEV owners’ benefits. First of all, a phasor mode single-phase microgrid with PEVs and PV farm is modeled to study the cost effective optimal control of PEV charging / discharging, and cope with the power balancing problem. The linear programming (LP) and genetic algorithm (GA) are used to obtain optimized charging schedules. The time-of-use (TOU) price and discharge incentive are considered for cost minimization. Secondly, the impacts of the bi-directional power flow on battery degradation are investigated. An energy-throughput battery degradation model is applied to simulate the daily lifetime degradation. Case studies with different battery capacities, depth of discharge, temperature, and charging regime are investigated. Thirdly, a grid-tied charging system that enables both grid-to-vehicle (G2V) and vehicle-to-grid (V2G) is designed using SimPowerSystems in Matlab/Simulink. The bi-directional AC-DC and DC-DC converters are designed with PWM-driven bridges. A predictive current control (PCC) is designed to control the AC-DC converter. The phase-lock loop (PLL) technology and band-stop filter is applied to reduce the total harmonic distortion (THD) on grid current. In the previous studies, the control algorithms are designed base on phasor mode model and validated via offline simulation. To further investigate the optimal power management algorithm for real-time simulation, a detailed power electronics level VGI system is modeled with RT-Lab RTE-drive blocks. The real-time simulations with a rule-based algorithm are conducted in Hardware-In-The-Loop (HIL) enabled OPAL-RT simulator. To deal with a large-scale of PEVs charging, a piecewise consensus-based distributed control is presented to minimize the charging power loss and maximize the utilization of PEVs for V2G services. The graph theory is applied to represent the communication network among neighboring PEVs. A metropolis stochastic matrix is applied to specify the communication topology. Case studies with ten PEVs and one hundred PEVs are conducted and analyzed respectively. In the last, due to the rapid response of PEV batteries to the power change demand, PEVs are coordinated with a dual-level consensus-based frequency control method to support the primary frequency control (PFC). The upper-level control aims to minimize the frequency deviation of multi-area power system, and the lower-level control aims to minimize the frequency regulation cost and battery degradation cost for individual PEVs. The consensus-algorithm is applied to specify the information updating and exchange among neighboring individuals. The simulation with different load change are conducted and studied. Personliga eldrivna fordon (EV) är ett nytt teknikområde som är på väg att uppnå stort momentum på flera av världens marknader. Eftersom branschen fortfarande ligger i sin linda finns det i nuläget inga tydliga strukturer, som gäller för alla marknader världen över, gällande relationerna mellan aktörer, vilket leder till osäkerheter när det kommer till att ta strategiska beslut. Uppdragsgivaren för denna studie är Hong Kong EV Power Ltd. (EV Power), en Hongkong-baserad leverantör av laddningsstationer för elbilar och relaterade tjänster, som har ambitionen att inträda marknaden på det kinesiska fastlandet inom den närmaste framtiden. Emellertid har EV Power ännu inte bestämt sig vilken stad de vill rikta in sig på i det första skedet. Denna avhandling ämnar formulera en modell som kan användas för att utvärdera och jämföra geografiska marknader med avseende på lämpligheten för ett marknadsinträde av en leverantör av laddningsstationer för elbilar. Dessutom kommer modellen testas på tre städer på kinas fastland (Peking, Shanghai och Shenzhen), med syfte att komma fram till vilken stad som är mest attraktiv för EV Power, samt att utvärdera modellens funktionsduglighet. Sist kommer resultaten från utvärderingen av de tre städerna att tjäna som utgångspunkten för en analys som ämnar ta fram framgångsfaktorer för ett marknadsinträde på kinas fastland. För att uppnå detta har fyra olika datainsamlingsmetoder använts: Först studerades teori, med syfte att få bakgrundskunskap likväl som att få förståelse för specifika faktorer som påverkar ett marknadsinträde som detta. För det andra observerades EV Powers nuvarande verksamhet i Hong Kong, i avsikt att förstå vad som har lett till den framgång som företaget upplevt på sin hemmamarknad. För det tredje intervjuades branschexperter för att få ett perspektiv på branschen som helhet. Sist samlades sekundär data kring de tre städerna in, för att kunna utvärdera de olika faktorerna som ingår i den framtagna modellen. Den slutgiltiga modellen består av fem faktorer som påverkar en stads attraktivitet för ett marknadsinträde av en leverantör av laddningsstationer för elbilar. De identifierade faktorerna är: ’Marknadens tillgänglighet’, ’Kortsiktig efterfrågan’, ’Förväntad marknadsandel’, ’Vinstmarginal’ och ’Långsiktig produktpotential’. Dessa faktorer är i sin tur indelade i subfaktorer som har sina egna uppsättningar av drivare. Efter att ha använt modellen för att utvärdera de tre städerna konstaterades det att Shanghai är den lämpligaste staden för ett marknadsinträde av EV Power, främst på grund av stadens dominans på marknaden för privatanvända elbilar och ett gynnsamt regelverk. Slutligen hittades tre framgångsfaktorer för ett sådant inträde: ’Fokusera på tjänster’, ’Bibehåll partner-relationer’ och ’Inträd tidigt’.
