O presente trabalho de investigação baseia-se na hipótese das cadeias A e B de insulina apresentarem comportamentos e estabilidade diferentes quando expostas às mesmas condições como consequência das diferenças verificadas na sua sequência.
Assim, os objetivos estabelecidos foram em primeiro lugar a obtenção das duas cadeias com recurso à enzima responsável pela sua obtenção a nível biológico, a proteína dissulfito isomerase (PDI), seguida da determinação de possíveis produtos de interesse. Por fim, o último objetivo traçado foi a mimetização da reação que ocorre fisiologicamente e por conseguinte a obtenção das cadeias purificadas para avaliação da sua estabilidade.
A metodologia utilizada divide-se em três partes fundamentais. A primeira consiste na avaliação da atividade da PDI na presença de insulina, utilizando-se para o efeito o ensaio turbidimétrico e um ensaio enzimático. A segunda parte incide na análise dos produtos de degradação com recurso à técnica de cromatografia líquida de alta eficiência, de modo a identificar péptidos de interesse. Por fim, a última parte consiste na avaliação da estabilidade das cadeias A e B de insulina obtidas através de um método químico pela técnica de dispersão dinâmica da luz.
Os resultados obtidos permitiram constatar a hipótese estabelecida inicialmente, dado que as cadeias A e B de insulina apresentam comportamentos e estabilidade diferentes quando expostas às mesmas condições. Foi igualmente verificado que a agregação é uma característica inata à insulina e suas cadeias, sendo contudo necessários mais estudos relativamente à cadeia B para a compreensão deste comportamento.

▼ Anterior Gradient-2 protein (AGR2) has recently been linked to the onset of several pathologies including asthma and inflammatory bowel disease. Most interestingly, it has been discovered to influence the transformation of cells and metastatic growth essential to cancer development, and has subsequently been linked to the development of resistance to anti-cancer therapeutics. AGR2 protein is overexpressed in a diverse range of human cancer types, and has been detected secreted into the extracellular milieu. Thus, AGR2 protein represents a compelling pro-oncogenic signalling intermediate in tumour emergence and endurance. This thesis presents an interdisciplinary approach including structural biology, cell biology and synthetic biology, and clinical studies to shed more light on the role of AGR2 in cancer development. Synthetic cell based reagents were developed to define the dominant pathways that are reprogrammed in a cell as a result of AGR2 synthesis. A cell panel was engineered incorporating the AGR2 (and mutants thereof) allele into the AGR2-null A375 cell line. These tools were then coupled to quantitative proteomics (SILAC) to unravel the mechanism whereby introduction of AGR2 alters cell phenotype, allowing identification of dominant pathways affected by AGR2 signalling. Using pathway analysis tools, the dominant pathway suppressed by wt-AGR2 expression highlighted the p53-signalling axis. DNA damage induced p53 stabilisation and p21 induction by cisplatin treatment confirmed the influence of AGR2 gene expression. Further data analysis identified the outlying protein expression changes identified by SILAC was the anti-viral cell cycle regulator TSG101 (tumour susceptibility gene 101), and confirmed by immunoblotting. Transfection and silencing studies of TSG101 confirmed that TSG101 attenuates p53 function. These data provide a mechanism to explain the most dominant pathways reprogrammed by AGR2 expression, incorporating ER stress response, proliferation markers and p53 pathway attenuation. Further advances were made in analysis of the function, regulation, and drugability of AGR2 protein. Assays were devised to define the subunit structure of AGR2 as a dimer unit; subsequent functional studies defined intrinsically disordered motifs that regulate stability of the dimer. A two-site sandwich microtiter assay (2SMTA) was designed to screen for self-peptides and mutations that regulate oligomer stability. These assays were used to identify the first biochemical property of AGR2 being that the dimer unit is required for maximal binding to the AAA+ protein, and well characterised AGR2 interactor, Reptin. In addition, based on this dimeric structure, a novel solution based dimerisation assay was developed to identify natural products that are able to disrupt the dimer suggesting that AGR2 itself can be targeted in principle with small molecules for therapeutic purposes.

▼ Brain tumor claimed the lives of 14,080 people in
United States in the past year. Gliomas are one of the most
malignant and deadly forms of brain tumor. The average life
expectancy of patients diagnosed with glioblastoma multiforme (GBM)
is 14 months despite revolutionary discoveries in the field of
radiotherapy and chemotherapy. The current focus lies in
discovering targeted therapies on the basis of our understanding of
the events occurring in the molecular level within the brain. ❧
Protein disulfide isomerase, a protein mostly found in the
endoplasmic reticulum, is abundant in glioblastomas. Inhibiting
PDI
has previously been shown to trigger apoptosis and result in the
death of cancerous cells. We report the discovery of chalcones as
effective
PDI inhibitors with an activity in the micromolar range.
We also discovered the potency of a class of compounds known as
propynoic acid carbamoyl methyl amides in brain
cancer.

▼ Perylene diimides (PDIs) have attracted the great
interests of both academic and industrial people over decades of
years because of their chemical stability, thermal stability,
fluorescence, and photoactive property. They have been widely used
as dyes, pigments, n-type organic semiconductors and organic field
effect transistors (OFETs) and so on. However, in addition to
molecular chemical nature, the self-assembly structure also deeply
affected material’s macroscopic property and practical application.
In order to get highly ordered assembly structure, we investigated
the use of
PDI-based derivatives as building blocks of
supramolecular self-assembly and liquid crystals via modifying
PDI’s imide position by alkyl chains and functional groups. Three
tetrahedral
PDI-based molecules tethering with different length of
alkyl tails (decyl, dodecyl, tetradodecyl) and four
cyano-substituted PDIs were successfully synthesized via a
systematic and convenient method, getting rid of the low solubility
of perylene. The chemical structures of products were fully
characterized by proton nuclear magnetic resonance (1H NMR)
spectroscopy and matrix-assisted laser desorption ionization
time-of-flight (MALDI-TOF) mass spectroscopy. Their thermal
stability could be maintained until 320 ¿ according to the results
of thermogravimetric analysis (TGA). Differential scanning
calorimetry (DSC) showed that the modified PDIs had the potential
to form ordered structure by thermal annealing.

▼ The superfamily Sauropodomorpha comprised some of the largest terrestrial herbivorous vertebrates that have ever walked on this planet. The study of these fascinating beings has been at the forefront of palaeontological research throughout the years. Especially in the last 20 years, due to the advent of modern techniques and technologies, significant steps have been made towards our understanding of their physiology. One of the sauropodomorphs’ central morphological characteristics is the expression of postcranial skeletal pneumaticity (PSP), a condition that is also expressed in the other superfamily of the order Saurischia, the Theropoda, as well as in their extant relatives, the Avians. This condition remodels the vertebrae and, occasionally the girdles and appendicular elements, producing perforations, depressions, excavations, cavities and internal chambers. This is a result of bone invasion and resorption from the development of the lung-air sac diverticula of the respiratory system. The various forms of this expression have been addressed across all subfamilies of sauropodomorphs mostly from an evolutionary aspect and, recently, from a developmental scope too. The resulting hypotheses state that PSP may have acted as a mechanism for weight reduction, allowing for sauropods to attain large sizes without having to suffer analogous gravitational constraints from equally attained masses. Any possible associations, though, between the expression of pneumaticity and body size have not been put under test. Through this study, a method of quantification and categorisation of PSP, and therefore, classification of the sauropodomorphs which express it, is created from the data retrieved from 61 taxa across all subfamilies, permitting us not only to trace any correlation between PSP and metric size data (body mass and body length) but also to visualise the evolution of PSP throughout Sauropodomorpha. This classification scheme from highest to lowest expression of PSP, ‘Alpha’, ‘Beta’, ‘Gamma’, ‘Delta’, and ‘Epsilon’ stems from the numerical estimate of pneumaticity in terms of a percentage, called the Pneumaticity Degree Index (PDI%). The revised scheme, Pneumaticity Degree Index (PDI%), takes account of the number of vertebral elements that are pneumatized in a single vertebra, the nature of pneumaticity traits, as well as the intensity of pneumatization in different body regions of interest (e.g. vertebral column), resulting in an integral and comprehensive measure of PSP. The proposed method ranks each pneumaticity trait with a value from 1 to 5 with 1 (100%) representing the most invasive unambiguous trait (e.g. camellae) and 5 (~ 10%) the least invasive and most ambiguous trait (i.e. fossa). By adding and dividing by their number all of the observed traits of every available vertebra of a region we retrieve a decimal numeric outcome and this outcome is translated to a percentage. ▼ A avaliação do sistema radicular é importante para melhor compreensão dos efeitos de manejo de nutrientes no solo e nutrição mineral de plantas. No entanto, essa avaliação tem sido uma atividade complexa, exaustiva e propícia a erros de amostragem e mensuração. Nesse contexto, o processamento digital de imagens e a análise de imagens podem auxiliar, pois têm sido utilizados em soluções de problemas similares. O objetivo deste trabalho foi desenvolver um método computacional para auxiliar na avaliação do crescimento radicular para amostras lavadas da cultura da soja, reduzindo o tempo gasto. Os resultados foram comparados com os métodos tradicionais (linha de interseção e massa fresca), e o software SAFIRA da Embrapa. O método proposto foi desenvolvido em plataforma Java, com aporte da biblioteca OpenCV por meio do plug-in JavaCV. A validação do método foi realizada comparando imagens (300 dpi) de amostras de fios de cobre de, 10, 20 e 50 mm de comprimento, com os valores obtidos manualmente com paquímetro, obtendo coeficientes de variação (CV) entre 0,01 e 2,99%. Das relações entre os comprimentos estimados com o método proposto e os obtidos pelo método tradicional obtiveram-se CVs variando entre 0,10 e 2,10%, que foram em média melhores que o SAFIRA (5,11% <CV <49,45%), em imagens de 300 dpi. O método também reduziu o tempo para obtenção dos atributos de raízes em mais de 53%. Dessa forma, o método proposto apresentou-se eficaz, principalmente para medidas de comprimento e área de amostras de raízes lavadas, sendo recomendado para estudos de raízes da cultura da soja.
The evaluation of the root system is important for better understanding of the effects of nutrient management on soil and plant mineral nutrition. However, this review has been a complex, comprehensive and conducive to sampling error and measurement activity. In this context, digital image processing and image analysis can help as they have been used in solutions of similar problems. The objective of this study was to develop a computational method to assist in the evaluation of root growth for washed samples of soybean, reducing the time spent. The results were compared with traditional methods (intersection line and fresh mass), and SAFIRA software Embrapa. The proposed method has been developed in Java platform with the OpenCV library supply through the plug-in JavaCV. The method validation was performed by comparing images (300 dpi) of samples of copper wires, 10, 20 and 50 mm in length, with values obtained manually using a caliper, obtaining coefficients of variation (CV) between 0.01 and 2.99%. Relations among the estimated lengths with the proposed method and those obtained by the traditional method yielded CVs ranging between 0.10 and 2.10 %, which were better than the average SAFIRA (5.11% <CV <49.45 %) at 300 dpi images. ▼ As mudanças ocorridas no mundo nos últimos anos vêm afetando de forma drástica pessoas, organizações e nações. Em uma velocidade crescente, são questionadas as bases nas quais se construíram as teorias econômicas, sociais e políticas vigentes. Na tentativa de substituí-las criam-se novas teorias, que também se tornam ultrapassadas rapidamente. Diversos autores afirmam que a civilização humana atravessa um período de mudança de paradigma. As ciências relacionadas à gestão das empresas estão entre as mais afetadas por essa mudança. A era da Informação proporcionou uma crescente disponibilização de informações, e conseqüentemente uma melhoria educacional. Isto provocou uma crescente conscientização da população na cobrança por bens e serviços de qualidade, em todos os segmentos do setor público, inclusive no meio educacional. O Governo Federal em reconhecimento dessa nova realidade, através do artigo 37 da Ementa Constitucional n 19/98, realizou a inclusão na Constituição de Federal de 1998 a eficiência como princípio da Administração Pública. A partir dessa institucionalização surgiram outras normatizações como: a Lei Complementar no. 101/00, evocada de Lei de Responsabilidade Fiscal que determina práticas que resultem em maior eficiência na alocação e execução dos gastos públicos. Dessa forma o governo editou a lei no. 10.861/2004 que estabelece o Sistema Nacional de Avaliação Superior SINAES, que possui dez diretrizes que devem ser observadas por todas as Instituições de Ensino Superior para que obtenham o credenciamento ou descredenciamento junto ao MEC, portanto essas dimensões por serem vitais são estratégicas para as instituições de Ensino Superior, logo devem estar associadas ao planejamento estratégico, gerenciamento por diretrizes e consequentemente ao PDI das Instituições de Ensino Superior. Nesta perspectiva foi realizada uma pesquisa qualitativa, bibliográfica, descritiva de caráter exploratório, junto a Universidade Federal de Santa Maria UFSM na qual associou-se ao modelo de planejamento estratégico já existente as dimensões do SINAES. Sendo que essas dimensões são diretrizes fundamentais a qualquer IES foi realizado um desdobramento destas na instituição em estudo. Constatou-se que com o desdobramento dessas diretrizes o Gestor da UFSM possuir uma ferramenta administrativa essencial para a sua gestão.

▼ Introdução: As abordagens dimensionais equacionam os delírios como situando-se num continuum com as crenças diárias, não estando circunscritos apenas à população clínica e podendo também ser encontrados na população geral. Devido à conceptualização multifacetada dos delírios, a análise das dimensões de desconforto, preocupação e convicção podem ser mais reveladoras do que o conteúdo da crença por si só, pelo que uma avaliação que incorpore estas dimensões é fundamental. Uma revisão de estudos mostrou uma lacuna de instrumentos para avaliar a ideação delirante na população geral, não existindo, no nosso conhecimento, nenhuma versão para a população portuguesa. Objetivos: Tradução, adaptação e estudo das propriedades psicométricas do Inventário de Ideação Delirante (PDI-21) para a população Portuguesa, com o intuito de avaliar a multidimensionalidade da ideação delirante na comunidade. Metodologia: A amostra é constituída por 249 adultos da população geral, com idades compreendidas entre 18 e 65 anos. Além do PDI-21, foi utilizado o Inventário de Sintomas Psicopatológicos (BSI), a subescala de Desejabilidade Social (DS) do Inventário de Personalidade de Eysenck – Forma Revista (EPQ-R) e o Instrumento de Avaliação da Qualidade de Vida da Organização Mundial de Saúde (WHOQOL-Bref). Resultados: A versão portuguesa do PDI-21 apresentou propriedades psicométricas adequadas no que respeita à sua consistência interna e estabilidade temporal. Demonstrou correlações positivas significativas com os sintomas psicopatológicos e associações negativas com a desejabilidade social e com a qualidade de vida, confirmando a sua validade convergente e divergente. A análise fatorial exploratória sugeriu a escala dicotómica do PDI ser constituída por um único fator. Por último, a análise da frequência de ideias delirantes (respostas positivas) para o total da amostra e em função do género revelou taxas de prevalência muito semelhantes às encontradas em estudos prévios. Discussão: A versão portuguesa do PDI-21 é caracterizada por propriedades psicométricas adequadas, podendo ser utilizada para avaliar a ideação delirante na população geral. Estudos futuros são necessários no sentido de replicar este estudo em populações mais jovens, bem como, comparar os resultados com uma amostra clínica. / Introduction: The dimensional approaches equate the delusions as placing themselves in a continuum with the daily beliefs, not being limited to the clinical population and it can also be found in the general population. Due to the delusions multifaceted conceptualization, the analysis of the dimensions of distress, preoccupation and conviction may be more revealing than the content of the belief by itself, whereby an evaluation that incorporates these dimensions is fundamental. ▼ O controle da reatividade de um reator nuclear à água pressurizada é realizado pelo uso
de barras de controle ou por diluição de boro na água do refrigerante do circuito primário. O
controle com diluição de boro apesar de inserir pequenas variações de reatividade no reator,
tem grande importância, uma vez que não afeta significativamente a distribuição do fluxo de
nêutrons.
Uma bancada experimental de baixa pressão, com seção de testes fabricada em acrílico,
foi montada em escala reduzida para simulação do pressurizador do reator nuclear à água
pressurizada IRIS (International Reactor Innovative and Secure). A bancada foi montada no
Centro Regional de Ciências Nucleares do Nordeste (CRCN-NE), Recife – PE, orgão
vinculado à Comissão Nacional de Energia Nuclear (CNEN). Para se estudar os processos de
diluição e homogeneização do boro, utilizou-se um corante com propriedades semelhantes ao
do ácido bórico, e assim foi possível realizar várias tarefas durante a execução dos
experimentos.
O presente trabalho tem como objetivos:
Elaborar procedimentos para estudo e análise da homogeneização do
boro no interior da seção de testes da bancada experimental;
Registrar a dispersão do boro (corante) através de fotografias digitais e vídeos;
Quantificar o valor da concentração de corante através de processamento
digital das imagens obtidas;
Os resultados obtidos experimentalmente estão de acordo com os das simulações do
modelo teórico.

▼ A inclusão dos jovens com Perturbação do Desenvolvimento Intelectual (PDI) no mercado de trabalho não se processa de uma forma natural, como seria de esperar numa sociedade inclusiva. Este estudo pretende recolher, analisar e determinar, sob a perspetiva dos empregadores, os fatores que facilitam ou dificultam a inserção profissional destes jovens. Pois é demarcando-os que se poderão equacionar e redefinir estratégias para que esta transição para a vida ativa das pessoas com PDI seja prática comum e cada vez mais uma realidade.
Realizaram-se entrevistas semidiretivas, a sete orientadores do trabalho destes jovens, de empresas do distrito do Porto, inseridas numa cultura de trabalho em rede/parceria com um Centro de Educação e Formação Profissional (CEFP). A partir da análise dos dados e de uma abordagem qualitativa, trabalhámos com valores, atitudes e opiniões para desenvolver conceções, ideias e perceções acerca desta temática.
Com base na análise dos dados, pudemos constatar que nas instituições visadas pelo nosso estudo, apesar de existirem ainda diversos fatores que não facilitam a transição para a vida ativa destes jovens, estes são superados pelos fatores que promovem este processo.
Este estudo evidenciou, assim, a importância, na ótica das empresas, de fatores subjacentes ao sucesso da inserção profissional de jovens com PDI, da intervenção dos supervisores que incidem a sua prática na inserção social e profissional destes jovens, da sua formação com vista à aquisição de competências específicas no local de trabalho e da adaptação da tarefa ao perfil do jovem com PDI. Por outro lado, mostrou a necessidade de se rever e redefinir algumas políticas e práticas que, nesta perspetiva, não respondem às necessidades das pessoas com PDI face ao mercado de trabalho.
The inclusion of teenagers with Disruptive Intellectual Development in the labour market doesn’t work in a natural way, as it would be expected in an inclusive society. This study aims at collecting, analysing and determining, according to the employers’ view, the factors which facilitate or not, the professional inclusion of theses teens. It is by focusing on them that we can question and redefine the strategies so that these teens’ transition to the labour market would be a real common procedure.
Semi-directing interviews were carried out with seven of these teens’ work supervisors, of companies from the district of Porto, inserted in a culture of networking/partnership with a Centre for Education and Vocational Training (CEFP). Based on data analysis and on a qualitative approach, we’ve worked with values, attitudes and opinions to develop concepts, ideas and perceptions about this issue.
Based on the data’s analysis, we could verify that in the institutions targeted by our study, although there are still several factors that do not facilitate these youngsters’ transition to their active life, those factors are overcome by those that promote this process.

This dissertation concerns the mechanism by which the mccPDI phenotype is regulated in the wild-type strains (Chapters 2), how the key effector protein (McpM) kills susceptible bacteria, and how the immunity protein (McpI) protects the mccPDI-producing bacteria (Chapter 3). Up-regulation of mccPDI is controlled by a two-component regulatory system (EnvZ/OmpR) that functions as an on/off switch. Low-salt conditions lead to upregulation of OmpR, which is a transcriptional regulator for both McpM and the cognate ligand on the susceptible cells, OmpF. Deletion of OmpR completely eliminates the mccPDI phenotype (Chapter 1). This on/off osmotic-sensing system is attenuated by the concentration of quorum sensing autoinducer-2 (AI-2) molecules in the culture media. Upregulation of mccPDI corresponds to increasing concentration of AI-2, while down regulation of the system corresponds to the decay of AI-2 as the bacterial population shifts from log-growth into stationary-growth phase. The kinetics of AI-2 expression correspond to the transcriptional pattern of mcpM, and deletion of the quorum sensing system eliminates upregulation of mcpM. It is very likely that sRNAs micC and micF also contribute to the mccPDI regulatory system (Chapter 2). A series of assays demonstrated that mccPDI inhibits the growth of susceptible bacteria when McpM permeabilizes the outer membrane, resulting in cell death. This phenotype can be blocked with the addition of low-molecular mass (200 – 600 Da) osmoprotectant to the culture media. The mechanism by which McpI protects mccPDI-producing cells is unknown, but experiments described in Chapter 3 show that McpI likely forms asymmetrical trimer structures that may serve to block pores that are formed by McpM.

O presente projeto de investigação pretende propor que a Substância Hepática Sensibilizadora de Insulina (HISS) consiste num dos produtos de degradação da insulina (cadeia A/B) nitrosilados, e que a enzima interveniente na formação de tais produtos é a Proteína Dissulfito Isomerase (PDI) presente nos hepatócitos. Adicionalmente sugere-se a ligação dos nitrosilados de insulina à albumina.
Com vista a tentar comprovar esta hipótese, a investigação efetuada teve o intuito de confirmar a existência de PDI ao nível dos hepatócitos, determinar a sua atividade e analisar os produtos de degradação originados. Outro dos objetivos foi efetuar a produção de nitrosotióis de cadeia A e B para posterior administração ex vivo e in vivo, e respetiva quantificação do potencial terapêutico.
Neste sentido, foram realizados diversos ensaios. Assim, numa fase inicial procedeu-se à monitorização da degradação enzimática da insulina por via in vitro com recurso à PDI purificada e a lisados de hepatócitos de rato recorrendo à técnica de turbidimetria. Seguidamente, foi realizada a análise dos produtos originados pela degradação da insulina efetuada pela PDI purificada através de eletroforese SDS-PAGE. Posteriormente, procedeu-se à separação química da cadeia A e B da insulina com posterior produção de nitrosotióis. Por último, estes produtos foram administrados a miotubos de rato e a ratos Wistar e o seu potencial terapêutico quantificado através da medição da captação de 2-deoxi-glucose marcada com trítio radioativo e através de sucessivos testes RIST, respetivamente.
Com efeito, os resultados desta investigação permitiram reforçar as ilações feitas pelos demais autores quanto à presença e atividade da PDI ao nível dos hepatócitos em relação à insulina, e para além disso confirmaram a suspeita inicial de que um dos derivados de insulina nitrosilados tem potencial efeito terapêutico ao nível da resistência à insulina apesar de não parecer estar relacionado com a estrutura da HISS.

▼ In eukaryotes, nascent proteins that are destined for the secretory pathway enter the endoplasmic reticulum (ER) in unfolded states and generally leave only after they have reached their native conformation. Protein folding in the ER is challenging and often error-prone. To prevent accumulation of misfolded proteins, the ER uses the ER-associated protein degradation (ERAD) machinery to detect and commit these defective proteins for retrotranslocation to cytosol for degradation via the ubiquitin-proteasome system. Previously, our group and others discovered an ERAD pathway that requires the generation of a specific N-glycan structure on misfolded luminal N-glycoproteins to permit their degradation. This N-glycan degradation signal can potentially serve as a mark to flag the defective states of the attached protein, but whether the generation of this N-glycan degradation signal is directly connected to the folding state of the attached protein has remained unknown.My thesis research work is focused on characterization of Htm1p, the mannosidase that is responsible for generating this N-glycan signal in Saccharomyces cerevisiae. Utilizing an in vitro reconstitution system of Htm1p in complex with its interacting partner, Pdi1p, and glycoprotein substrates whose structures can be closely manipulated, I find that unlike other members of the same mannosidase family, Htm1p-Pdi1p is a glycoprotein-specific mannosidase that has little activity against free glycan. I find that Htm1p-Pdi1p targets intrinsically and artificially misfolded proteins but not their native counterparts. Most importantly, I find that Htm1p-Pdi1p differentiates detailed folding states of misfolded proteins and preferentially acts on partially folded proteins to globally unfolded species. I further find that by forming a tight complex with Pdi1p, Htm1p blocks the intrinsic oxidoreductase function of Pdi1p but keeping the chaperone activity, suggesting a potential mechanism of how the Htm1p-Pdi1p complex recognizes misfolded proteins. In summary, this work demonstrates the tight correlation between the generation of the N-glycan degradation signal and the folding state of the attached protein, opening a future direction to investigate how the ER tells the bad from the good.

▼ Amine solvent losses are a significant issue for CO₂ capture by amine scrubbing. Solvent lost through aerosol emission represents an environmental hazard with adverse economic implications. This research focuses on developing analytical systems to quantify amine aerosol emissions. Fourier Transform Infrared Spectrometry quantified amine emissions and Phase Doppler Interferometry determined aerosol size and concentration. Baghouse pretreatment of the flue gas significantly reduced amine emissions through collection of aerosol nuclei. A baghouse at the National Carbon Capture Center (NCCC) reduced monoethanolamine (MEA) emission by over a factor of 10. An SO₃ generator was built to facilitate bench and pilot scale aerosol experiments by reacting SO₂ in air over vanadium pentoxide catalyst at 520 °C. Aerosol generation at UT-SRP produced up to 1.7 grams per minute of SO₃, with conversion exceeding 81 %. Bench scale experiments achieved conversion greater than 97 % and aerosol concentration up to 7E4 cm⁻³. SO₃ increased piperazine (PZ) emission by up to 7.6 mol PZ/ mol SO₃. SO₂ increased PZ emission by 1 mol/ mol SO₂, and increased MEA emissions by 3.9 mol/ mol SO₂. H₂SO₄ increased PZ emission by 3 mol/ mol H₂SO₄. PZ resisted aerosol emissions with lower SO₃ content; this is because a low inlet aerosol nuclei concentration results in rapid aerosol growth and subsequent collection by impaction. Higher process temperatures correlated with decreasing PZ emission, supporting the growth and capture theory. Increasing the solvent PZ content was shown to strongly correlate with increasing PZ emission. In bench scale experiments, PZ emission and aerosol size both increased as the PZ content in the solvent increased. Lowering the temperature bulge stage reduced PZ emission and the aerosol size. Increasing the inlet CO₂ correlated with larger aerosol. Increasing the solvent CO₂ loading and the inlet SO₃ resulted in greater aerosol concentration. Operations with a blower upstream of the absorber increased MEA aerosol emission. The upstream blower resulted in larger aerosol in greater quantities, containing a greater quantity of MEA Reduced MEA emission with an intermediate blower are probably due to collection of aerosol through impaction within the blower.

▼ A oxidação do ácido úrico (7,9-diidro-1H-purina-2,6,8(3H)-triona) pela mieloperoxidase (MPO) gera o radical de urato que, em presença do radical ânion superóxido combinam para formar o hidroperóxido de urato (HOOU). Considerando os altos níveis de MPO, ácido úrico e superóxido na placa de ateroma espera-se que o ácido úrico seja oxidado a HOOU neste microambiente inflamatório. O HOOU é um oxidante mais forte que o peróxido de hidrogênio e pode oxidar grupos tiólicos de proteínas como a proteína dissulfeto isomerase (PDI). Como consequência à oxidação da PDI, ocorre uma modulação positiva sobre a NADPH oxidase (Nox) com aumento da produção de superóxido por neutrófilos. Sendo assim, a formação do HOOU no leito vascular poderia elucidar os mecanismos moleculares pelos quais o urato colabora para a progressão da aterosclerose em pacientes com hiperuricemia. Para investigar a interação do HOOU com a PDI, padronizou-se a síntese química do HOOU através de um sistema de fotooxidação do tipo I, utilizando urato, riboflavina (fotossensibilizador) e luz UVA. Inicialmente padronizou-se o tipo de irradiação e tempo de reação com o melhor rendimento para a síntese do HOOU. O HOOU formado e seu produto de redução o álcool 5-hidroxiisourato foram separados, identificados e caracterizados através de cromatografia liquida acoplada à espectrometria de massa (LC/MS). Após a purificação, determinou-se o coeficiente de absortividade molar em 308 nm (ε308 = 6537 ± 377 M-1.cm-1) e o tempo de meia-vida, aproximadamente 41 minutos à 22°C do HOOU. O HOOU foi capaz de reagir seletivamente com o aminoácido metionina e com o tripeptídeo glutationa. Além disso, o HOOU não forma adutos estáveis com a glutationa, sendo que toda glutationa consumida foi transformada em glutationa dissulfeto. Quando incubado com a PDI (10 µM), cerca de 70 e 100% do total de HOOU (3 µM) foi consumido após 30 e 120 segundos, respectivamente, enquanto que a PDI (23 µM) teve seus grupos tiólicos oxidados após a incubação com 140 µM HOOU por 30 min a 22°C. O HOOU oxidou os resíduos de cisteína dos dois sítios catalíticos da PDI com uma constante de velocidade da reação de 1,5 ± 0,04 x 103 M-1.s-1, demonstrando uma interação favorável com essa proteína no meio biológico e um possível papel modulatório do HOOU sobre a via PDI-Nox. Interessantemente, o ácido úrico aumentou a produção de superóxido e o consumo de oxigênio de células HL-60 diferenciadas em neutrófilos (dHL-60) e ativadas com acetato de miristato de forbol (PMA). ▼ E consenso que o crescente consumo de combustíveis fósseis na geração de potencia tem provocado uma maior degradação do meio ambiente. Para mitigar os efeitos adversos desse consumo, novas fontes e usos sustentáveis de energia são necessários. O uso de etanol na atual geração de motores de combustão interna demanda informações detalhadas sobre os processos de nebulização, evaporação, mistura e combustão das gotas desse combustível. Adicionalmente, novas estratégias de injeção de combustível em MCIs com injeção direta estão em desenvolvimento, como a injeção estraticada. Essa estratégia consiste na injeção de combustível em elevadas frequências durante as etapas de admissão e combustão do ciclo motor. Neste trabalho, técnicas avançadas de diagnostico a laser são utilizadas para analisar a inuência da frequência de injeção de combustível no processo de combustão de sprays pulsados de etanol, em que as chamas resultantes são estabilizadas por um swirler. A técnica de interferometria por efeito Doppler (
PDI) foi utilizada para medir o diâmetro e a componente axial da velocidade das gotas. O campo de velocidades do escoamento de ar foi avaliado pela técnica de velocimetria por imagens de partículas (PIV) em duas taxas de aquisição (7,4 Hz e 2.000 Hz) para avaliar tanto valores médios quanto rastrear estruturas coerentes no escoamento. O mesmo sistema PIV foi utilizado para identificar a posição de aglomerados de gotas pela técnica de espalhamento Mie ao longo de eventos individuais de injeção. Já a técnica de uorescência induzida por laser do radical hidroxila (LIF-OH) foi utilizada com taxa de aquisição de 4.700 Hz para rastrear a região com presença deste radical próximo ao queimador utilizado. Imagens da luminescência química espontânea foram obtidas para observar características globais das chamas. O queimador, posicionado em um ambiente aberto, e composto por um dispositivo swirler e um injetor automotivo. As frequências de injeção de 100 Hz, 250 Hz e 400 Hz foram escolhidas para as análises por produzirem chamas com características que variam desde uma chama estável e ancorada ao queimador até o comportamento similar a uma chama suspensa e instável. Etanol anidro líquido foi utilizado como combustível e fornecido ao injetor a pressão e vazão constantes para todos os casos. Os resultados indicaram que a variação na frequência de injeção produziu sprays com diferentes densidades. A menor frequência de injeção produziu um spray com maior densidade, em que as gotas formaram um aglomerado pouco sensível ao escoamento de ar na linha de centro do queimador. Esse aglomerado produz uma chama ancorada ao queimador com formato alongado e estreito. Já a maior frequência de injeção produziu um spray com menor densidade, permitindo que a dinâmica das gotas seja mais susceptível ao escoamento de ar. ▼ O ensino superior é considerado, atualmente, o precursor do desenvolvimento econômico e social dos países. Para desempenhar esse papel, as universidades têm buscado novas formas de gestão. Para auxiliá-las nesta missão, o planejamento estratégico tornou-se uma ferramenta importante neste sentido, utilizado tanto em instituições privadas quanto em públicas. As Instituições Federais de Ensino Superior (IFES) possuem o planejamento elaborado por meio de um modelo de Plano de Desenvolvimento Institucional (PDI) proposto pelo governo. No entanto, apesar dessa estrutura ser considerada como o planejamento estratégico, as universidades não se apropriam desse formato. Nesse cenário, as IFES carecem de um processo de planejamento que contemple as suas características. Dado isso, o objetivo desta dissertação é propor um modelo de processo de planejamento estratégico para IFES. Para atingir o objetivo, foram desenvolvidos três artigos. No primeiro artigo, foi elaborada uma revisão da literatura a fim de responder questões que embasem a proposição do modelo, enquanto no segundo artigo foi realizada uma análise documental acerca de PDIs e de Planos de Gestão (PGs) das IFES, a partir da qual foram identificados o relacionamento entre estes planos e as possíveis influências das variáveis no processo de planejamento sob as constatações detectadas na análise. Por fim, no terceiro artigo foi proposto o modelo de processo de planejamento completo, através de um estudo de caso com grupos focados aplicado em uma IFES. Como conclusão geral, identificaram-se elementos estratégicos que foram propostos com mais frequência em universidades. Confirmou-se que o modelo proposto pelo governo não atende a estas universidades, e verificou-se que os planos utilizados nas IFES não possuem elementos estratégicos. Além disso, identificaram-se as variáveis que podem influenciar o processo de planejamento estratégico e suas possíveis influências no caso em estudo. Por fim, propôs-se um modelo de processo de planejamento adaptado ao contexto das IFES, além das dinâmicas que podem ser utilizadas em cada fase desse processo.
Higher education is, nowadays, considered the precursor of economic and social development of the countries. To play this role, universities have been seeking new forms of management. To help them in this mission, the strategic planning has become a important tool in this sense, used in both private and public enterprises. The Brazilian public universities, particularly the federal ones, IFES, already have their planning drawn up by an Institutional Development Plan model (PDI) proposed by the government. However, despite this structure have been considered strategic planning, universities do not appropriated of this format. ▼ A exploração florestal tem afetado extensas áreas de florestas primárias na Amazônia brasileira. Esta atividade, quando praticada com base no manejo florestal, pode ser uma grande aliada no esforço para reduzir o desmatamento na região. O monitoramento florestal é fundamental para estabelecer planos de ação e estratégias que contribuam para o uso sustentável dos recursos naturais. Considerando a extensão da área sob exploração madeireira, a tecnologia de sensoriamento remoto orbital oferece meios para otimização do monitoramento. Neste contexto, este estudo realizado no norte do Mato Grosso analisou imagens de alta resolução espacial para caracterizar a geometria e o contraste radiométrico das feições mais conspícuas da atividade madeireira, que são os pátios de estocagem de madeira. Tamanho, forma dos pátios, distância entre eles e o contraste radiométrico entre pátios e a floresta em seu entorno foram caracterizados para fundamentar o desenvolvimento de um algoritmo de detecção de alvos pontuais a partir de técnicas de processamento de imagens aplicadas a dados de resolução espacial média. Foi realizada uma análise exploratória das imagens de alta resolução HRC/CBERS-2B para descrever características radiométricas e geométricas de pátios de estocagem e para avaliar sua utilidade como verdade terrestre para estudos relacionados ao monitoramento da exploração madeireira. Investigou-se a aplicação de modelo linear de mistura espectral, NDVI e transformações por IHS e principais componentes em imagens TM/LANDSAT e CCD/CBERS para detectar atividade de corte seletivo de madeira e foi desenvolvido um método para detectar pátios baseando-se no contexto da vizinhança dos pátios. Os resultados mostraram que os pátios têm brilho medido pela HRC 57% maior do que a floresta e apresentaram área média de 380 m2 distribuídos numa distância de em média 460 metros. Deste modo, em imagens de resolução de 20 a 30 metros (400 a 900 m2), os pátios se apresentam como objetos pontuais, o que justifica a proposição do uso de contexto de vizinhança para sua detecção. Foi definido limiar de decisão para definição do objeto "pátio"para cada imagem analisada e um algoritmo desenvolvido a partir da decisão de um "pátio"como um pixel de valor acima ou abaixo do limiar envolto de pixels com média abaixo ou acima deste limiar, dependendo da imagem analisada. A comparação das imagens antes e após a aplicação da metodologia proposta permitiu identificar áreas com ocorrência de exploração madeireira evidenciada a partir do realce de pátios de estocagem. Na análise de exatidão baseada na verdade terrestre derivada dos dados HRC 54% a 75% foram corretamente identificados pelas diferentes técnicas.

▼ Objective To examine the relationship between maternal reflective functioning (RF) and child aggression and whether this relationship is mediated by child’s precursors of Theory of Mind (ToM). The influence of coaching on maternal reflective functioning and child aggression is also examined. Method The sample consisted of 85 mother-infant dyads. Mothers were allocated to a high or low risk group based on maternal characteristics. Half of the high risk mothers were randomly assigned to a coaching group. Maternal reflective functioning was assessed prenatally and when children were 20 months of age by Pregnancy Interview (PI) and Parental Development Interview (
PDI). Mothers were asked to aggressive symptoms in their child. Children’s precursors of ToM were measured at 20 months by an imitation task, a visual perspectives task and a discrepant desires task. Results Maternal prenatal reflective functioning was found to be negatively related to child aggressive behaviour. However maternal postnatal self-related RF was positively related to child aggression. In addition, maternal reflective functioning was found to be positively related to one precursor of ToM, but ToM did not mediate the relationship between maternal reflective functioning and child aggression. No effect of coaching on maternal reflective functioning and child aggression was found. Conclusion The present study provides evidence for the relationship between maternal reflective functioning and child aggression at a young age. This emphasizes the importance of well-developed maternal reflective functioning capacities for the behavioural development of children. This relationship could be mediated by child’s ToM, but this should be further researched. Coaching would be a suitable way to enhance maternal reflective functioning capacities, but the effects of coaching should be investigated after mothers received coaching for a longer period of time.

▼ A new glass-forming organic semiconductor material was synthesized using a known electron transport material, perylene diimide, and attaching it to a molecular glass in order to allow the material to be solution processed. Devices were made using a simple metal-semiconductor-metal structure and electrodes were selected to produce Schottky diodes. Experiments were carried out to characterize this new molecular glass perylene diimide. The new material shows evidence of traps, hysteresis, and other behaviours that are explored in this thesis. The material shows some potential as an electron transport layer with possibilities of memory storage behaviour.

▼ Currently the mammalian endoplasmic reticulum (ER) is known to contain at least 20 different protein disulphide isomerase (PDI) family members. The oxidoreductases in the PDI family are thought to catalyse the formation and rearrangement of disulphide bonds in newly synthesised proteins. The focus of this work was to characterise two of the PDI family members: PDI and ERp46. In vitro translation reactions of major histocompatibility complex (MHC), β1-integrin (β1-I), haemagglutinin (HA), procollagen α1(III) and preprolaction (pPL) were carried out in untreated or PDI-depleted cells. The depletion of PDI decreased the rate of folding of MHC and β1-I and also prevented the oligomerisation of HA, suggesting a role for PDI in folding these putative substrates. However, when PDI was depleted neither the folding of pPL or HA was affected, implying that they may not be substrates for PDI. To determine the role of ERp46 in the cell, a substrate-trapping approach was used. Here substrates interacting with ERp46 were trapped as mixed disulphides isolated by immunoprecipitation, separated by 2D SDS-PAGE and identified by mass spectrometry. It was demonstrated that ERp46 forms mixed disulphides with at least 23 proteins, including heavily secreted proteins such as laminins, integrins and collagens. In particular, interactions with Ero1, Prx IV, EDEM3 and ERAP2 were found and confirmed by immunoprecipitation of radiolabelled in vitro translated protein. Notably nine of these clients of ERp46 have previously been identified as substrates of ERp57 (Jessop, Watkins et al. 2009). This would support the hypothesis that several different oxidoreductases, working in concert, are required to fold certain substrate proteins. Also, it was confirmed that Prx IV and Ero1 each form a mixed disulphide with PDI. These results highlight the importance of PDI family members in recruiting co-factors to substrates. Additionally, the over-expression of ERp46 led to increased cell survival following DTT treatment, yet after depletion of ERp46, cells were less able to grow, perhaps suggesting a role for ERp46 in maintaining ER redox homeostasis and cell survival. This suggestion was supported by the finding that ERp46 is able to catalyse the reduction of Prx IV in the presence of glutathione. These results suggest that Prx IV provides a novel mechanism for the transfer of disulphide bonds to nascent proteins in the ER via PDI family members such as ERp46 and PDI.

▼ Coronary artery disease is the leading cause of death in the western world today. Although
induction of angiogenesis would appear to be an ideal therapeutic strategy, clinical trials of proangiogenic
factors have proved disappointing. Angiogenesis is a complex process involving many
signaling pathways and mediators, and further insights into the underlying cellular and molecular
mechanisms are urgently needed. Here, we used two mouse models, systemic hypoxia and
myocardial infarction (MI), to study the effects of hypoxia on angiogenesis in the myocardium,
and the cellular and molecular mechanisms involved.
Hypoxia-inducible factor-1α (HIF-1α) is an important transcriptional regulator of angiogenesis.
Small ubiquitin-related modifier-1 (SUMO-1) has been shown to stabilize transcription factors
and modulate their activity. In our mouse model of systemic hypoxia, we showed that SUMO-1
expression is enhanced by hypoxia in brain and heart. Furthermore, SUMO-1 co-localizes and
directly interacts with HIF-1α under hypoxic conditions, indicating that hypoxia-mediated
increases in SUMO-1 expression could modulate HIF-1α function.
We combined our mouse model of systemic hypoxia with a model of MI and showed that chronic
hypoxia protects the heart from infarct injury and promotes angiogenesis. A proteomics analysis
demonstrated that protein disulfide isomerase (PDI) is upregulated in the myocardial capillary
endothelial cells of mice exposed to chronic hypoxia. Furthermore, PDI knockdown in
endothelial cells in vitro increases apoptosis and inhibits migration and adhesion, indicating that
PDI may play an integral role in angiogenesis.
Endoglin is a co-receptor for transforming growth factor-β. In our mouse model of MI, we
showed increases in endoglin expression in endothelial cells in the heart one week after surgery.
Similarly, endoglin expression is increased in endothelial cells in vitro after exposure to hypoxia.
Furthermore, we showed that hypoxia promotes activation of the endoglin/ALK-1/SMAD1/5 but
not the endoglin/ALK-5/SMAD3 signaling pathway in endothelial cells. The induction of this
pathway represents another potential mechanism for regulation of angiogenic responses in
endothelial cells after MI.
The results presented advance our understanding of the complex pathways involved in hypoxiamediated
angiogenesis in the heart. Our findings could play a role in identifying new strategies
for the treatment of ischemic heart disease.

▼ Stimulated Electromagnetic Emissions SEEs may provide important diagnostic information about space plasma composition, energetics, and dynamics during active experiments in which ground-based high powered radio waves are transmitted into the ionosphere. The nonlinear plasma processes producing this secondary radiation are not well understood particularly for some recent observations where the transmitter (pump) frequency is near the second harmonic of the electron gyro-frequency. New, more comprehensive, experimental observations of spectral features within 1kHz of the pump wave frequency are reported here in order to begin more careful comparisons of the experimental observations and a possible theoretical underpinning which is also provided. The experimental observations typically show two distinct types of secondary radiation spectra which are a) discrete narrowband harmonic spectral structures ordered by the ion gyro-frequency, so-called stimulated ion Bernstein Scatter (SIBS) and, b) broadband spectral structure with center frequency near 500 Hz and similar spectral bandwidth named ion acoustic parametric decay (IAPD). A theoretical model is provided that interprets these spectral features as resulting from parametric decay instabilities in which the pump field ultimately decays into high frequency upper hybrid/electron Bernstein and low frequency neutralized ion Bernstein and/or obliquely propagating ion acoustic waves at the upper hybrid interaction altitude. Detailed calculations of the threshold level, growth rate, unstable wavenumber and frequency bandwidth of the instabilities are provided for comparisons with experimental observations. An assessment of the effect of the critical instability parameters are provided including pump electric field strength, proximity of the pump frequency to the electron gyro-frequency and pump electric field geometry. The model shows quite reasonable agreement with the experimental observations.
Next, a two dimensional Particle-In-Cell Monte-Carlo Collision computational model (PIC-MCC) is employed in order to consider nonlinear aspects such as 1) electron acceleration through wave-particle heating, 2) more complex nonlinear wave-wave processes and 3) temporal evolution of electron irregularities through nonlinear saturation. The simulation results show that the IB associated parametric decay is primarily associated with electron acceleration perpendicular to the geomagnetic field. More gyro-harmonic lines are typically associated with more intense heating. Heating is reduced when the pump frequency is sufficiently close to 2fce. The IA associated parametric decay instability is primarily associated with electron tail heating along the magnetic field and heating is reduced when the pump frequency is sufficiently close to 2fce. Characteristics of caviton collapse behavior become prevalent in this case. Results are discussed within the context of some recent experimental observations. Efficient maturation of disulfide-containing proteins in the endoplasmic reticulum (ER) requires facilitated folding, aided by various folding catalysts and chaperones. As protein folding is a complex process, proteins may, even in the presence of endogenous folding factors, form misfolded states. Therefore, the cell has acquired several layers of quality control mechanisms to keep the folding load in the ER in balance with the folding capacity. The activity of folding catalysts is regulated to avoid stress connected to hyperoxidation. Other quality control mechanisms include the unfolded protein response (UPR) and ER-associated protein degradation (ERAD). The UPR consists of signaling cascades that sense unfolded protein stress and upregulate folding factors and other components to relieve the stress. ERAD directly removes non-native proteins.
The main folding catalysts that introduce disulfide bonds to folding proteins in the ER are members of the ER oxidoreductin 1 (Ero1) family and protein disulfide isomerases (PDI). Ero1 is a sulfhydryl oxidase that reduces molecular oxygen to hydrogen peroxide to form disulfides. The newly generated disulfides are then transferred during the catalytic cycle to PDI which catalyzes disulfide formation in folding proteins. The Ero1-PDI pathway has been studied extensively by cell-based techniques, and its activity is regulated in a redox dependent manner by rearranging regulatory disulfides. However, the kinetic aspects of these regulatory functions have remained poorly characterized.
In this study, we report comprehensive kinetic measurements for the Ero1-PDI pathway that reveal novel regulatory mechanisms for oxidative protein folding (OPF). These include a mechanism consisting of high affinity and apparent cooperativity for substrate oxygen to allow Ero1 to function efficiently at hypoxic conditions. We also show that different human Ero1 isoforms have differential dependence for substrate PDI with one of the isoforms, Ero1α, showing strong dependence for PDI for both redox activation as well as the catalytic cycle. This observation allowed us to describe a potential crosstalk between OPF and ERAD, in which OPF by the Ero1α-PDI pathway is downregulated, to potentially increase the efficiency of ERAD which requires reduced protein substrates. This work is concluded by a structural study of a novel chaperone, pERp1, that is crucial for our immunity, as it is involved in the folding of some immunoglobulins.

PDI; EC 5.3.4.1) and thioredoxin (Trx) in the marine macroalga Ulva fasciata Delile in response to hypersaline (90‰) for 1 h. 90‰ induced H2O2 accumulation, reflecting the occurrence of oxidative stress. The contents of free and protein-bound SH were increased by 90‰. Trx transcripts increased in response to 90‰.
PDI transcripts and enzyme activities increased in response to 90‰. H2O2 accumulation under 90‰ condition was increased by putrescine (Put) but decreased by spermidine (Spd) and spermine (Spm). By treatment of spermidine and spermine, the contents of free SH was increased and the contents of protein-bound SH decreased, showing that spermidine and spermine can increase free SH against oxidative stress. The gene expression and activity of
PDI were further increased by Spd and Spm. Overall, the gene expression of
PDI and Trx were responded to 90‰ for 1 hour and were adjusted proteinâSH in polyamines treatment.

▼ Converging evidence indicates that SOD1 aggregation is a common feature of mutant SOD1 (mSOD1)-linked FALS, and seems to be directly related to the gain-of-function toxic property. However, the mechanisms of protein aggregation are not fully understood. To study the contribution of modification on cysteine residues in SOD1 aggregation, we systematically examined the redox state of SOD1 cysteine residues in the G37R transgenic mouse at different stages of ALS and under oxidative stress induced by H2O2. Our data showed that under normal circumstances, cysteine 111 in SOD1 is free. Under oxidative stress, it is prone to oxidative modification by providing the thiolate anion (S-). With the progression of ALS, increased levels of oxidative insults facilitated the oxidation of thiol groups of cysteine residues. Human mutant SOD1 could generate an upper shifted band in SDS-PAGE, which turned out to be a Cys111-peroxidized SOD1 species. We also found that at different stages of ALS, accumulated oxidative stress facilitated the aggregates formation, which were not mediated by disulfide bond. The oxidative modification of cysteine 111 may promote the formation of disulfide bond-independent SOD1 aggregates.

PDI) in the mechanism of aggregates formation. Our data showed that up-regulated inducible nitric oxide synthase (iNOS) generated high levels of nitric oxide (NO), which induced S-nitrosylation of
PDI with the progression of ALS in the spinal cords of mSOD1 transgenic mice. This correlation was confirmed by treating SH-SY5Y cells with NO donor SNOC to trigger the formation of S-nitrosylated
PDI (SNO-
PDI). When mSOD1 was overexpressed in SH-SY5Y cells, iNOS expression was up-regulated, NO generation was increased consequently. Furthermore, both SNO-
PDI and mSOD1 aggregates were detected in these cells. Blocking NO generation with NOS inhibitor N-nitro-L-arginine (NNA) attenuated the S-nitrosylation of
PDI; the formation of mSOD1 aggregates was inhibited as well. We conclude that NO-mediated S-nitrosylation of
PDI is highly linked to the accumulation of mSOD1 aggregates in ALS.

▼ Xanthomonas citri subsp. citri é uma bactéria fitopatogénica que afeta os citrinos, causando o cancro cítrico. Esta doença tem distribuição global e representa perdas económicas significativas. Face à baixa eficácia e elevada toxicidade ambiental dos tratamentos com cobre atualmente praticados, existe a necessidade de desenvolver novas abordagens ao controle do patogénico que sejam simultaneamente eficazes, economicamente viáveis e ambientalmente sustentáveis. O presente trabalho teve como objetivo avaliar a aplicabilidade da inativação fotodinâmica (
PDI) de X. citri subsp. citri com um fotossensibilizador comercial, como estratégia de controle da bactéria e prevenção do cancro cítrico.
Foi avaliada a
PDI de X. citri subsp. citri em suspensões de células e em biofilmes construídos em suportes artificiais (ensaios in vitro) ou em folhas de laranjeira (ensaios ex-vivo). Foram testadas diferentes concentrações (1-100 μM) do fotossensibilizador azul de toluidina O (TBO) e do coadjuvante KI (1-100 mM) com irradiação durante 60 minutos com luz branca (400-700 nm, 150 mW.cm-2) artificial. Nos ensaios ex-vivo, foi também testada a irradiação com luz solar natural.
Os resultados mostraram uma redução de 5,9 log na concentração de células viáveis nas suspensões expostas 80 μM de PS e uma redução de apenas 1,7 log na concentração de células viáveis nos biofilmes, com 100 μM de TBO. No entanto, na presença de KI (10-100 mM), foi possível erradicar completamente os biofilmes (redução > 6 log) com 5 μM de TBO (in vitro) ou 10 μM de TBO (ex vivo). Com luz solar natural, conseguiu-se inativação completa de biofilmes em folhas de laranjeira tratadas com 50 μM de TBO e 100 mM de KI.
Os resultados mostram que embora os biofilmes de X. citri subsp. citri sejam menos suscetíveis à
PDI do que as células planctónicas, a presença de KI aumenta a eficiência de inativação sendo possível atingir inativação completa após 4 horas de irradiação das folhas com luz solar natural. A
PDI apresenta assim potencial de aplicação no controle do cancro cítrico em condições de campo, embora seja necessário aprofundar os estudos que permitam demonstrar que não há riscos para a fisiologia da planta ou para a qualidade dos frutos.

▼ The self-assembly of organic molecules has attracted
much interest because of the special properties and possible
technological applications of the mesoscopic materials which are
intermediate materials between molecules and solids.1 Structural
and spectroscopic studies on J- and H-aggregates present valuable
information for understanding interactions at the molecular level
in aggregation processes, and may also provide understanding that
leads to alternative molecular devices. The synthesis and
preliminary photophysical properties of two generations (G1-G2) of
glutamic acid-based peptide dendrimers containing free-base or zinc
porphyrin at the cores were performed. In this work, steady-state
absorption and fluorescence experiments for the free-base
dendrimers 59 and 63 were observed to have similar spectra relative
H2TPP in DMAc, while the absorption and fluorescence spectra of
zinc porphyrin dendrimers 60 and 64 were almost identical to that
of ZnTPP in DMAc. These dendrimers tend to not to aggregate in
polar solvents like DMAc, therefore, other nonpolar solvents will
be sought for self-assembly these dendrimers. Second, two electon
donor (D)- acceptor (A) dyads are composed of free-base porphyrin
(Fb) and perylene bisimide (
PDI) groups that are linked by leucine
(65) or polyleucine (n=2, the dyad 66) were prepared in this work.
Fb and
PDI were chosen because of the useful photophysical
properties each exhibits that will enable us to perform both energy
and electron transfer within each dyad. The absorption spectra of
the dyads indicate a close match with the sum of the component
spectra, indicating little or no electronic coupling occurs between
porphyrin and
PDI groups in the ground state. Due to the overlap of
the emission bands of porphyrin and perylene, fluorescence quantum
yield analysis could not be determined with any accuracy.
Fluorescence excitation spectra will therefore be performed in the
near future. Intramolecular FRET experiments will also be achieved
on these two dyads, whereas these measurements will allow us to
precisely determine the donor-acceptor distance at any given time.
After we measure the distance between the donor-acceptor dyad, we
will investigate how the rate of the electron transfer varies with
the distance, and what effect the dyad structure has in controlling
this rate constants.The photophysical and electrochemical
properties of covalently-linked arrays of N-confused porphyrins
(NCPs) are of interest to a variety of supramolecular and
biomimicking materials. While differing from the parent porphyrins
by inversion of only two atoms, NCPs exhibit physical and chemical
properties different than normal porphyrins. The synthesis and
preliminary photophysical properties of three dimeric and trimeric
N-confused porphyrin (NCP) arrays (75, 77 and 80) are presented.
The absorption and emission maxima for the NCP dimer (75) and
trimers (77 and 80) in DMAc were found to be broader and
red-shifted relative to that of NC-TPP. The fluorescence quantum

▼ The growing need for cheap, reliable,
economical, and environmentally safe sources of energy has prompted
extensive research into the field of organic photovoltaics.
Oligo(p-phenylenevinylenes) (OPVs) are model compounds for the
polymer poly (p-phenylenevinylenes). These compounds are a highly
conjugated, hole conductive materials used in both organic
photovoltaics and organic light emitting diodes. Unfortunately,
little research exists concerning the effects of nano- and
microstructure on organic photovoltaic performance. In other words,
how does the spatial relationship between adjacent OPV chains
influence photoconductivity? The first project of this dissertation
was comprised of two parts. The first part dealt with the need to
improve the asymmetric synthesis of OPV chains. By modifying the
current literature approach towards asymmetric OPVs, the total
overall yield of these chromophores has been optimized to values
that are 15 – 30% higher than current literature values. The
methodology and synthetic steps are explored. This project became
the basis for the second part, which was the synthesis of two OPV
units attached cofacially to bridging groups, in order to study
energy and electronic transfer between the two units as a function
of interchain distance, angle, and/or orbital
overlap. The second project of this dissertation
dealt with the creation of liquid crystalline perylene diimide
(
PDI) aggregates through non-covalent interactions. Perylene
diimides have been utilized as optimal n-type materials for
photoconductive organic devices, and have strong intermolecular π-π
interactions that induce specific packing orientations. The nature
of the packing can be influenced by the substituent at the cyclic
imide portion of the chromophore. This unit has no real effect on
the ground state electronic properties of the individual
chromophore because it is located at a node in the
PDI interacting
molecular orbitals. The
PDI therefore behaves as an isolated
chromophore with minimal to no ground state mixing with the imide
substituent. Nevertheless, the structure of that substituent can
greatly influence the bulk properties of the chromophore by
directly affecting the transverse and longitudinal packing in the
solid state. The utilization of a hydrazide linkage could direct
the self-assembly of appropriately substituted PDIs through both
hydrogen bonding and π-π stacking, into ordered arrays. The
synthesis and photophysical properties of these hydrazide-linked
PDIs are presented. The third and final project
in this dissertation involves the self-assembly of a
multichromophoric perylene diimide. Various self-assembly
techniques were used to construct this assembly, including hydrogen
bonding, π-π stacking, and ligation of metal-complexes, all of
which provide site-specific platforms for the organized assembly of
a multi-chromophoric system. The synthesis of this compound is
presented.
