ヒト脂肪由来幹細胞をストロンチウム(Sr)含有軟骨形成培地で培養すると、軟骨分化が促進されるか調べた。健康被験者の頬部脂肪体から脂肪組織を採取し、初代培養を行った。フローサイトメトリーにより脂肪由来幹細胞(ADSC)を単離し、基礎培地のみ、軟骨分化培地(CM)、Sr添加CM培地のそれぞれで培養した。軟骨形成はアルシアンブルー染色により調べた。培養細胞からtotal RNAを抽出し、逆転写反応によりcDNAを合成し、PCR法を用いてmRNAの発現を調べた。1.50mM Sr含有CMでは基礎培地のみとCMのみの場合と比べて、グリコサミノグリカン分泌と、SRY(sex determining region Y)-box 9およびcollagen type 2 alpha 1のmRNA発現が有意に高かった。脂質、骨芽細胞、肥大性軟骨への分化マーカーの発現増加は認めなかった。SrはADSCの軟骨分化を誘導する可能性が示唆された。

ラット両側上顎第1臼歯に人工的歯周欠損を作製し、アメロゲニンのエキソン5と同じ配列を有する新しい合成ペプチドの効果を調べた。ペプチド投与群では歯肉溝や接着上皮で上皮細胞増殖、上皮再生を示し、欠損部に抗III型コラーゲン抗体陽性の線維性構造が形成された。発現血管数の割合は3日目には有意差がなかったが、5,7,14日目には投与群で有意に高い割合を示した。今回作製した合成ペプチドは線維芽細胞の成長因子分泌を促進し、創傷表面の上皮細胞増殖から歯周欠損の治癒を早めることが示唆された。
