The present work used advanced methods of pulse-EPR to analyze the PSII of the cyanobacterium Thermosynechococcus elongatus. High-field EPR was used to record the signals of the water-oxidizing complex at W-Band frequencies, derive its g-tensor components, and compare it to model complexes and reference values gained from different frequency bands. Single-crystal EPR rotation patterns were taken to demonstrate the orientation dependency of the visible hyperfine structure to explain the drastic differences in appearance between the W-band spectra presented here and the well-known spectra at X-band frequencies. Pulsed ENDOR methods were employed under high-field conditions to characterize the couplings and orientations of and within a neutral radical found in PSII, the tyrosine-160 of the D2 subunit, Y160. Using single crystals, the molecular orientation with respect to the magnetic-field direction and within the crystal was resolved, thereby establishing the Y160 as a marker of the molecular orientation of PSII. Furthermore, high-field conditions allowed the reconstruction of the coupling strengths and orientation of the protons bound to the tyrosine. By selective deuteration, a proton known to be hydrogen-bound to Y160 was exchanged against a deuterium ion, thus separating its coupling signals due to the different gyromagnetic moment and free Larmor frequency. A reconstruction of this hydrogen bond, its strength and orientation offers direct spectroscopic proof of the bonding partner, a near-by histidine residue also bound to the D2-subunit.
