▼ Abstract: Temporomandibular disorders (TMD) are associated with pain/inflammatory process, including biological and behavioral parameters. The aim of the present study was to evaluate the effect of two anti-inflammatories (parecoxib and dexamethasone) in the TMJ morphology of ovariectomized rats (OVX) with or without retrusive displacement of mandible (RDM). 72 rats were distributed into 4 groups: G1 - no RDM and no OVX; G2 - with RDM and no OVX; G3 - no RDM, but with OVX; G4 - with both RDM and OVX. The RDM induction was achieved by placing an oral device made with steel strips (orthodontic band) in the rat upperincisors. After 7 days of RDM, the animals were submitted to one of the following treatments: 0.9% NaCl (CONT); 0.30mg/kg/im/day parecoxib (PARE); or 0.1mg/kg/im/day dexamethasone (DEXA). After 7 days of treatment, the TMJs of both sides were removed. One of them was submitted to histological procedure (HE) after fixation and decalcification. The contralateral TMJ was macerated, centrifuged and the supernatant was submitted to the immunoassay (ELISA) in order to evaluate the pro-inflammatory cytokines (IL1-beta and IL-6). The readings were performed by spectrophotometry. All animals were weighed with the same age and before they were killed. It was possible to observe that the rats without RDM showed a mean (±SE) variation of body weight of -0.41±0.72%; the ones not submitted to RDM was -4.8±1.1%; with OVX of 26.9±1.2% and the ones submitted to both RDM and OVX was 8.3±2.5%. In general, these data showed a body weight gain in the animals with OVX, but not in the ones with RDM. Both PARE and DEXA decreased (Kruskal-Wallis, p<0.05) the articular cartilage area in G1 when compared to CONT, but PARE increased the area in relation to DEXA. None of the treatments influenced the IL1-beta levels in G1, but it was higher in G2 (Kruskal-Wallis, p<0.05) for DEXA. No differences were observed among treatments for groups 3 and 4 (p>0.05). IL1-beta levels were lower in G1 when compared to the other ones, disregarding of treatments. These levels were higher in G2 than G3 but not G4 (except for DEXA). None of the treatments affected the IL-6 levels considering each group. G2 showed the highest IL-6 levels than the other groups, irrespectively of the treatments. No statistically significant differences among the levels of IL-6 of G1, G3 and G4 were found. The morphological parameters were not affected by RDM or by OVX, but the treatments. We concluded that RDM increased the levels of both cytokines, being reduced by OVX. Both PARE and DEXA did not interfere with cytokines levels. The articular cartilage area was not influenced by RDM, but it increased this area when associated with PARE

▼ Menopause is an age-related loss of ovarian hormone production that has been linked with obesity-associated metabolic dysfunction, increased visceral adiposity, and inflammation, although the mechanisms remain unclear. Obesity has been strongly linked with profound shifts in the gastrointestinal (GI) microbiota, disrupted gut barrier function, and inflammation, but little is known regarding the involvement of the GI tract in obesity associated with loss of ovarian hormone production. Herein, we hypothesized that GI tract and the gut microbiota are involved in the weight gain and inflammation that occurs with the loss of ovarian hormone production and that dietary interventions may affect these responses.
The objective of aim 1 was to evaluate the impact of soy on metabolic health, adipose tissue inflammation, and the cecal microbiota in ovariectomized (OVX) rats bred for low-running capacity (LCR), a model that has been previously shown to mimic human menopause. Forty 27-wk old LCR rats were either OVX or sham-operated (SHM) and fed either soy-rich (soy) or soy-free (control) diets for 28 wk. Soy consumption reduced (p<0.05) body weight gain, adiposity, circulating cholesterol concentrations, and improved insulin sensitivity of LCR rats. Principal coordinates analysis (PCoA) of weighted and unweighted UniFrac distances of cecal microbiota revealed a sharp separation (p<0.05) between soy- and control-fed groups. The soy-fed group had a lower (p<0.001) Firmicutes:Bacteroidetes ratio compared to control.
The objective of aim 2 was to determine the energy metabolism, lipid accumulation, and inflammation in adipose and liver tissues of OVX female C57BL/6J mice in response to a high-fat diet (HFD). Forty 10-wk-old female C57BL/6J mice were fed either a high-fat diet (HFD; 60% kcal from fat) or a low-fat diet (LFD; 10% kcal from fat). After a 2-wk acclimation period, mice underwent surgical intervention (OVX or SHM). As expected, OVX mice fed HFD had substantially greater (p<0.05) body weight gain, adiposity, and hepatic triglyceride concentrations than OVX mice fed LFD. Compared to intact controls,
ovariectomy led to greater (p<0.05) adipose and hepatic tissue inflammation, macrophage infiltration, oxidative stress, hindered insulin signaling and glucose uptake, and altered lipid and energy metabolism. Moreover, HFD feeding of OVX mice led to greater (p<0.05) inflammation and macrophage infiltration in gonadal adipose tissue. The degree of adiposity and inflammation resulting from HFD in OVX mice vs. SHM mice was dramatically greater than that observed in OVX mice vs. SHM control mice fed LFD.

value-cut yields. Dressing percentage was higher (P < 0.01) in RAC-fed heifers than controls (CTL) and HCW, CCW, and REA tended (P < 0.10) to be increased by RAC feeding. Intact heifers had higher (P < 0.01) DP and larger (P = 0.05) REA and
tended to have higher (P = 0.09) bone maturity scores and lower (P = 0.09) YG than OVX heifers. The ribeye roll, shoulder clod, and gooseneck round were heavier (P < 0.05) and the tenderloin and knuckle tended (P < 0.10) to be heavier in RAC-fed
heifers compared to CTL. Conversely, OVX decreased (P = 0.04) the yield of the ribeye roll and tended (P = 0.06) to decrease the yields of the brisket and strip loin compared to INT heifers. The interaction of RAC supplementation and gender significantly
affected the value cuts shoulder top and top blade weights. These value cut weights were higher (P < 0.05) with RAC feeding in INT heifers, while weights were similar in OVX heifers.

▼ Laparoscopic surgeries are increasingly popular in exotic companion mammals, but information about their benefits or disadvantages is limited. The aims of this thesis are: 1) to assess the working space achieved with carbon dioxide pneumoperitoneum in rabbits; 2) to evaluate the cardiorespiratory effects of pneumoperitoneum; and 3) to compare intra-operative and post-operative outcomes between laparoscopic and open
ovariectomy in rabbits.
For the first two cross-over studies, six female New Zealand White rabbits were randomly assigned to sequences of intra-abdominal pressures (IAP). Computed tomography (CT) was used to measure working space after insufflation to 4, 8, and 12 mm Hg. From 4 to 8 mmHg, there was a 19% increase in working space, whereas from 8 to 12 mm Hg the increase was only 6.9%. The order of IAPs affected working space.
To assess the effect of pneumoperitoneum on cardiorespiratory parameters, the following outcomes were measured at 0, 4, and 8 mm Hg: blood pressures (ABP); cardiac output, cardiac and stroke volume indices (CI, SVI); heart rate; end-tidal CO2 (ETCO2); blood gases (PaCO2, PaO2); peak inspiratory pressure (PIP); and peripheral oxygen saturation (SpO2). Heart rate, SpO2, and ABP were unaffected by IAP. For PaO2, an interaction effect was seen between IAP
and pressure sequence. PaCO2 increased at 8 mm Hg, and ETCO2 and PIP were greater with each IAP. IAP decreased cardiac output and CI.
Intra-operative and post-operative outcomes between laparoscopic (LapOVE) and open
ovariectomy (OVE) were compared in female New Zealand White rabbits randomly allocated to surgical treatments (n=6 per group). Surgical and anaesthetic time were longer and incision length was shorter for LapOVE versus OVE. There were no significant differences in post-operative outcomes between the treatments, including ethograms evaluated by a blinded observer. Surgical complications associated with LapOVE included intestinal perforation, subcutaneous emphysema, and seroma formation. Incisional dehiscence occurred in both groups.
In summary, pneumoperitoneal pressures should be limited to 8 mm Hg in rabbits. When performing
ovariectomy in rabbits, laparoscopy may not provide as marked of an advantage over open approaches as in other mammals. However, further evaluation of laparoscopic techniques is warranted for this species.

▼ The central melanocortin receptors have classically been implicated in the maintenance of energy homeostasis, though their expression in hypothalamus and brainstem. Two melanocortin receptors are predominantly expressed in the central nervous system, namely melanocortin -3 and -4 receptors (MC3R and MC4R). While the MC4R has been extensively studied by our lab and others, the function of the MC3R is less well understood. Through the utilization of a mouse model with the MC3R promoter driving green fluorescent protein expression (MC3R-GFP) and another with genetic knockout of the MC3R (MC3R KO), a more extensive study of the function of the MC3R was performed. High expression of the MC3R-GFP was noted in brain regions including the cortex, thalamus, hypothalamus, and ventral midbrain. Following up expression of the MC3R in the ventral tegmental area, MC3R deficiency was shown to alter dopamine homeostasis and sucrose preference in a female specific manner. The alteration in midbrain dopamine content in the MC3R KO female was normalized by
ovariectomy. In addition, the response to
ovariectomy in the MC3R KO uncovered a significant increase in body weight and body fat in response to surgery in MC3R KO animals. Further studies examining gender-specific control of energy homeostasis were conducted during pregnancy, when ovarian hormone signaling fluctuates. MC3R KO dams showed a deficiency in pregnancy induced food intake and a defect in the accumulation of fat stores in preparation for lactation. However, MC3R KO dams displayed normal body weight gain and food intake response to lactation. These findings demonstrate that the MC3R exerts multiple effects on the regulation of food intake and energy homeostasis that are specific to female animals.

▼ Diabetes e menopausa aumentam significativamente o risco de doença cardiovascular entre as mulheres. O objetivo do presente estudo foi investigar os efeitos do treinamento físico na disfunção autonômica cardiovascular e na incidência da mortalidade em ratas fêmeas diabéticas submetidas a privação dos hormônios ovariana. Ratos Wistar fêmeas foram divididas em grupos ooforectomizados: euglicêmicos sedentário e treinado (EOS e EOT), e diabéticos sedentário e treinado (DOS e DOT). O diabetes foi induzido por uma única injeção de estreptozotocina (50mg/kg i.v.) quinze dias antes da ooforectomia. A freqüência cardíaca (FC), a pressão arterial (PA), a variabilidade da freqüência cardíaca (VFC), a sensibilidade barorreflexa e o controle autonômico da FC foram avaliados após o período de treinamento físico em esteira ergométrica (8 semanas) iniciado uma semana após a ooforectomia. O grupo EOS apresentou o maior peso corporal ao final do protocolo (3467g), além dos maiores valores de PA média (1243 mmHg) e FC (36113 bpm) em relação aos demais grupos estudados. O animais EOT mostrou menor peso corporal (3257g), PA média (1132 mmHg) e de FC (3275 bpm) em comparação aos animais EOS. O grupo DOS apresentou reduzido peso corporal (1867g), bradicardia (2518 bpm) e hipotensão (1022 mmHg) em relação ao grupo EOS. Os animais DOT apresentaram atenuação dessas alterações. O diabetes induziu redução da sensibilidade barorreflexa (~55%). O treinamento físico induziu aumento das respostas de bradicárdica e taquicárdica no grupo euglicêmico (~40 e 74%) e diabético (~89 e 74%) em relação aos seus respectivos grupos sedentários. A VFC estava reduzida no grupo DOS (131,3 bpm) quando comparada ao grupo EOS (181,5 bpm). Os grupos treinados mostraram aumento da VFC (231 no EOT e 211,2 bpm no DOT) quando comparados aos grupos sedentários. O diabetes reduziu o tônus vagal (~68%). O grupo DOT apresentou o tônus vagal semelhante ao grupo EOS e reduzido tônus simpático quando comparado aos outros grupos. A FCI foi menor nas ratas DOS em relação as ratas EOS e o treinamento físico atenuou tal disfunção. Correlações foram obtidas entre a VFC e o tônus vagal com as respostas bradicárdica (r=-0,5 e r=-0,5) e taquicárdica (r=0,6 e r=0,6) induzidas por variações da PA. A mortalidade foi de 50% e de 25% entre os animais DOS e DOT, respectivamente. Em conjunto, esses achados fornecem evidências de que prejuízo na sensibilidade barorreflexa e na variabilidade da freqüência cardíaca são potenciais mecanismos responsáveis pelo aumento da mortalidade relacionada a doenças cardiovasculares observado em mulheres diabéticas, bem como que a melhora autonômica induzida pelo treinamento físico pode diminuir a incidência de mortalidade em mulheres diabéticas menopausadas.

▼ The oestrous cycle of the domestic bitch is unique with regard to its considerable length compared with that of most other domestic animals. A non-seasonal anoestrus, with a duration that may last from 2 to 10 months, follows each oestrous cycle. The anoestrus can be prolonged by oestrus-preventing drugs such as progestins. The general aim of the studies in this thesis was to get further insight into the endocrinology of the physiological and the progestin-induced canine anoestrus. Studies performed throughout recent years have made clear that during the course of canine anoestrus many endocrinological changes take place at the hypothalamic, pituitary, and ovarian level. Apart from these changes, there are indications of involvement of dopaminergic influences. The results of the study described in Chapter 3 provide forceful evidence that the bromocriptine-induced shortening of the interoestrous interval is not triggered by a decline in plasma prolactin concentration. The results of the study described in Chapter 4 provides additional evidence that the premature onset of oestrus brought about by a dopamine agonist must be due to some other dopamine-agonistic effect, probably increased FSH secretion. The study in Chapter 5 is a report on the effects of GnRH administration on the plasma concentrations of reproductive hormones in intact and OVX bitches. The data presented in this study demonstrate that provocative testing of the pituitary-ovarian axis using GnRH may be helpful in differentiating between bitches in anoestrus and neutered bitches. The study reported in Chapter 6 describes the basal and GnRH-induced plasma concentrations of FSH and LH in four anoestrous and four OVX bitches. In this report indications were found that measurement of the plasma FSH concentration in a single plasma sample may prove reliable for determination of neuter status. Taken together, the results of the studies described in Chapters 5 and 6 provide a basis for the diagnosis of remnant ovarian tissue and verification of neuter status in the bitch. Chapters 7 and 8 are reports on the effect of MPA on canine adenohypophyseal function. The results of both studies demonstrate that treatment with MPA affects the hypothalamic-pituitary-ovarian axis. Oestrus, ovulation, and a subsequent luteal phase did not occur in any of the bitches during treatment with MPA. The prevention of oestrus by MPA cannot be ascribed to a significant reduction in circulating levels of either FSH or LH. On the contrary, during the first months of MPA treatment there was an increase in basal plasma FSH and LH, which may be due to a direct oestrus-preventing effect of MPA at the ovarian level. With continuing MPA treatment, basal plasma gonadotrophin concentrations returned to pretreatment levels and the pituitary FSH response to GnRH stimulation decreased, while several LH pulses were not accompanied by a significant FSH pulse, suggesting that MPA treatment attenuated pituitary FSH sensitivity to endogenous GnRH. ▼ A ovariectomia bilateral em ratas é um dos modelos experimentais utilizados para se analisar a menopausa e as possíveis estratégias para amenizar os efeitos deletérios desta condição. A suplementação da dieta com antioxidantes vem sendo utilizada para diminuir o risco de estresse oxidativo, que está aumentado na menopausa. Nesse estudo, foram analisados os efeitos do ácido lipoico (AL) e dos ômegas-3 (ácido docosahexaenóico (DHA) e ácido eicosapentaenóico (EPA)) no fígado, através da suplementação da dieta de ratas ovariectomizadas. O ácido lipoico foi escolhido em virtude da sua capacidade antioxidante, pela conhecida característica de ser cofator de enzimas chave no metabolismo celular e por inibir alguns mecanismos pró- inflamatórios. Essas características são importantes em virtude da disfunção mitocondrial e do aumento dos níveis de citocinas inflamatórias, que estão presentes após a menopausa. Os demais ácidos graxos poli-insaturados foram utilizados por possuírem alta capacidade antioxidante e estarem bem estabelecidos como protetores endoteliais. Além disso, também exercem efeitos anti-inflamatórios e são capazes de normalizar os níveis de lipídios na circulação sanguínea. Os resultados demonstram que o ácido lipoico é capaz de atuar no fígado e recuperar a atividade da enzima fumarase e a atividade das enzimas antioxidantes superóxido dismutase mitocondrial (MnSOD) e glutationa peroxidase (GPx), além de diminuir o dano oxidativo em proteínas e manter os níveis de malondialdeído (MDA) semelhantes ao grupo controle. No entanto, não exerceu influência na atividade da superóxido dismutase citosólica (CuZnSOD). A suplementação com DHA e EPA restaurou os níveis das duas classes de superóxidos dismutase, mas, não conseguiu restaurar a atividade da GPx e da fumarase. Os níveis de proteínas danificadas foram menores em todos os animais suplementados com ômega-3 em relação ao grupo controle e ao grupo ovariectomizado sem suplementação. Além disso, os suplementos também atuaram na redução dos níveis de nitritos e nitratos pois possuem propriedades anti-inflamatórias, sobretudo o ácido lipoico, que inibe a oxido nítrico sintase induzível (ONi) de macrófagos. Os níveis de vitamina C não apresentaram diferença entre os grupos SHAM, OVX, AL e DHA. Mas, o grupo EPA apresentou níveis de vitamina C inferiores aos dos grupos SHAM e OVX. Além disso, os níveis de vitamina E se mostraram inferiores nos grupos suplementados com os ácidos graxos poli-insaturados de cadeia longa. Neste trabalho, os antioxidantes assumiram um papel importante na diminuição do dano oxidativo e na recuperação da atividade de enzimas importantes na homeostase celular, alterações essas, causadas pela diminuição drástica dos níveis de estrogênio no modelo de menopausa.

▼ A ovariectomia, extração dos ovários, contribui para a carência de estrogênio e desta forma é um bom modelo para representar a diminuição deste hormônio que ocorre na menopausa, tornando as mulheres mais propícias aos sintomas e doenças desta fase. Após a menopausa pode-se observar maior estresse oxidativo em mulheres, resultado do desequilíbrio do sistema imunológico, esse estresse oxidativo pode ocorrer devido à influência que o estrogênio tem sobre o sistema imune e sua ação antioxidante. Neste cenário o zinco é considerado um elemento importante, uma vez que ele atua como cofator de diversas enzimas antioxidantes. O objetivo desta pesquisa foi avaliar os efeitos da suplementação de zinco associada ou não ao treinamento físico com carga no perfil lipídico e massa esplênica de ratas ovariectomizadas. Trata-se de um estudo experimental com 60 ratas wistar que foram separadas em cinco grupos com 12 animais cada, sendo estes grupos denominados SHAM (não ovariectomizado e sem tratamento), OX-C (ovariectomizado sem tratamento), OX-Z (ovariectomizado suplementado com zinco), OX-TF (ovariectomizado submetido à atividade física) e OX-ZTF (ovariectomizado suplementado com zinco e submetido à atividade física). O protocolo consistiu de 04 séries de 10 saltos dentro da água, com temperatura controlada de 32 2 C e a uma profundidade de 70 cm, e intervalo de 30 segundos entre as séries, foi utilizada uma sobrecarga acoplada em um colete específico. Nas primeira e segunda semanas de treinamento foi adicionada sobrecarga de 50% da massa corpórea das ratas, na terceira e quarta semanas, de 60%, quinta e sexta semana 70 % e nas duas últimas semanas 80% da massa corporal. O treinamento dos animais foi realizado três vezes por semana, durante o período de oito semanas, que coincidiu com o período de suplementação de zinco. Após a eutanásia, foram analisados perfil lipídico (colesterol total, LDL, HDL, VLDL, triglicerídeo), massa de órgãos (baço, coração e útero) e da gordura corporal. Os parâmetros foram analisados pelo teste-t não pareado e ANOVA univariada. Os resultados obtidos em relação à massa do útero e as dosagens de estradiol dos animais ovariectomizados foram significativamente menores em relação grupo controle. A supressão estrogênica provocou aumento no consumo de ração (p=0,0001) e massa corporal (p=0,0002), entretanto os grupos suplementados com zinco apresentou menores valores destes dados quando comparados com o grupo OX-C (p=0,0001/0,01 respectivamente). A massa do baço dos animais ovariectomizados sem nenhum tratamento foi significativamente maior que o SHAM (p=0,04), enquanto o grupo OX-ZTF, comparado com OX-C apresentou menor massa do baço (p=0,02). Observou-se que a supressão estrogênica elevou todas as concentrações séricas avaliadas, com diferença estatística apenas para as dosagens de colesterol total e LDL. Dietary xylitol supplementation increases bone calcium and
phosphorus concentrations in healthy rats, as well as protects
against the decrease of bone minerals and bone density during experimental
osteoporosis. This suggests that dietary xylitol might have a favorable
effect on the prevention of osteoporosis. However, before any conclusions
can be drawn about the usefulness of a compound, studies including
structural evaluation and biomechanical testing of bones must first
be performed.
Thus, the aim of the present study was to clarify whether
dietary xylitol affects bone resorption, bone structure, and bone
biomechanics in healthy rats, and whether dietary xylitol offers
some preventive effects against the increased bone resorption,
decreased bone trabeculation, and weakened bone biomechanical properties
during experimental osteoporosis.
Dietary xylitol reduced bone resorption in 3-mo old healthy
male rats, and protected significantly against the increase of
bone resorption in 3-mo old ovariectomized rats, as measured by
the urinary excretion of 3H following [3H]tetracycline-prelabeling.
In addition, increased trabecular bone volume of proximal tibia
in 4-mo old healthy male rats was detected after a 1-mo xylitol
feeding period, and significant protection against the decrease
of trabecular bone volume in 6-mo old ovariectomized rats was observed
after a 3-mo xylitol feeding period. Furthermore, dietary xylitol
increased the strength properties of long bones in 6-mo old healthy
male rats after a 3-mo feeding period, without affecting the bone
elastic properties as tested by three-point bending of tibia, torsion
of femur, and loading of femoral neck. Accordingly, dietary xylitol
protected significantly against the weakening of bone biomechanical
properties in 6-mo old ovariectomized rats after a 3-mo feeding
period.
In conclusion, the above results strongly support the hypothesis
that oral administration of xylitol protects effectively against
the progression of experimental osteoporosis. Dietary xylitol was
effective both in increasing bone mass in healthy rats, and in
preventing bone loss in ovariectomized rats, suggesting a favorable
effect of xylitol on both main targets in the prevention of osteoporosis.
As dietary xylitol was effective also in protecting against the
experimental osteoporosis-caused changes in bone structure and
weakening of bone biomechanical properties, oral xylitol administration
seems to provide interesting possibilities when searching for new
physiological choices for the prevention of osteoporosis.

▼ In this thesis some recent technological developments in human surgery are evaluated for their potential use in veterinary medicine by introducing them as surgical innovations for canine gonadectomy. Barbed sutures achieve wound apposition without surgical knot tying and thus avoid knot-associated negative consequences (lengthy placement, impaired wound healing around bulky knots, and the effect of unsightly knots on cosmetics). A study in 9 dogs found that celiotomy closure was easily achieved, no intraoperative or postoperative complications occurred, and ultrasonographic characteristics during wound healing were comparable to conventional sutures.
Ovariectomy is technically less complicated, less time consuming, and is associated with decreased morbidity (smaller incision, less intraoperative trauma, decreased discomfort) compared with ovariohysterectomy. For the incidence of long-term urogenital problems, including endometritis/pyometra and urinary incontinence, no differences between techniques were observed. This makes
ovariectomy the preferred method for elective gonadectomy. Neuromuscular blocking agents induce abdominal muscle wall relaxation and might lead to a larger intra-abdominal working space and improved surgical efficiency. However, the duration of ovarian pedicle transection in 40 dogs was not significantly changed (5.7 ± 2.3 versus 5.9 ± 1.9 minutes), arterial blood pressure decreased 5%, and the occurrence of intraoperative complications did not differ. There was no advantage to add NMB to the anesthetic protocol. Newly developed energy-based hemostatic surgical devices were evaluated during laparoscopic
ovariectomy. When a bipolar electrocoagulation grasping forceps was compared with monopolar electrocoagulation on 103 dogs, it decreased laparoscopic
ovariectomy time (41 versus 53 minutes), decreased intraoperative hemorrhage (8 versus 13% dogs), and facilitated exteriorization of the ovaries. Obesity (56 versus 42 minutes) and intraoperative mesovarial bleeding (56 versus 46 minutes) increased surgical time. Dog age, estrus, and pseudopregnancy did not influence surgical time. When a vessel sealing device was compared to the bipolar electrocoagulation grasping forceps in 40 dogs, the surgical time was reduced (3.5 ± 2.7 versus 8.1 ± 4.2 minutes), with less bleeding (0 versus 10%), and with less thermal coagulation damage based on histological scoring. Surgical times were influenced by surgeon experience, and ovarian pedicle fat scores. When the vessel sealing device was compared to a bipolar electrocoagulation cutting forceps in 21 dogs, excision of the ovaries was faster (2.5 ± 1.2 versus 4.8 ± 1.8 minutes), bleeding occurred less (0 versus 29%), and thermal coagulation damage was less (93.4 ± 85.8 versus 130.9 ± 101.2 µm). ▼ Ιntroduction: Osteoporosis is a major health threat and its prevalence is expected to rise dramatically in the coming decades. Hormone replacement therapy (HRT) plays a crucial role in the management and the prevention of osteoporosis, but prolonged use has been associated with adverse effects such as breast and uterine cancer, thromboembolic episodes and coronary heart disease. This has led to an increased demand for alternative therapies. In the present study, we assessed the phytoestrogenic potential of Amphimas pterocarpoides (AP) plant extract on the ovariectomy induced bone loss in rats in two different doses.Methods and materials: Fifty five female (10 month old) Wistar rats were used. Ovariectomy (OVX) was performed in 44 rats, while sham operation was performed in the remaining 11 rats (Control group). Ovariectomized animals were separated in the OVX group (n=13) and received drinking water ad libitum right after the surgery until the end of the experiment, while the treated groups received Amphimas pterocarpoides extract at 50 mg/kg (ALow group, n=15) and at 150 mg/kg (AHigh group, n=16). Measurements of Bone Mineral Density (BMD) were established using dual-energy X-ray absorptiometry (DEXA) and performed at baseline, 3 and 6 months after ovariectomy. After 6 months treatment, the rats were euthanized, and their femurs were extracted. The femoral biomechanical properties were evaluated using the 3-point bending test (3PB).Results: The percentage changes of the mean values of the proximal tibia BMD from baseline to 3 months for Control, OVX, ALow and AHigh, were 6.254%, -21.822%, -14.512% and -4.421% whereas, the percentage changes from baseline to 6 months were 8.108%, -32.134%, -23.985% and -8.144 accordingly. The 3PB test revealed that the maximal load before fracture (Fmax) demonstrated a significant higher value in the ALow group compare to the OVX group (112.69 Mpa vs. 96.9 Mpa, p<0.001). In addition the mean values for Stress in the OVX group was significant lower in contrast to ALow group (158.48 Mpa vs. 180.5 Mpa, p=0.011). AHigh group presented higher values in Fmax and Stress in contrast to the OVX group, but statistical significant values did not observed between groups.Conclusions: This study suggests that orally administered Amphimas pterocarpoides for 6 months, manage to prevent the rapid degradation ofbone and is effective in terms of improving both bone quantity (bone density) and quality (biomechanical parameters), surrogate markers of reduced risk fracture risk.

▼ Abstract: The aim of the present study was to evaluate the effect of sodium alendronate in the process of tissue repair of bone defects, artificially produced in estrogen-compromised rats irradiated with 15 Gy. We selected forty female adults rats (Rattus norvegicus, Wistar Albinus), aged 90 days. The animals were randomly divided into the following groups: ovariectomized; ovariectomized-alendronate; ovariectomized-irradiated; and ovariectomized-alendronate-irradiated. Sodium alendronate (5 mg/kg) was administered three times a week, for four weeks, in ovariectomized-alendronate and ovariectomized-alendronate-irradiated groups. Twelve days after the end of treatment with alendronate, a bone defect was created in animals' left tibia. Three days later, the posterior legs of ovariectomized-irradiated and ovariectomized-alendronate-irradiated group animals were irradiated with 15 Gy. The euthanasias occurred 10 and 33 days after bone defects creation. The presence of bone repair was analyzed by histomorphometry, and the bone microarchitecture was analyzed by micro computed tomography. In relationship to the process of tissue repair on osseous defects, the X radiation and the alendronate do not represent significant effects (p > 0,05). In relationship to the osseous microstructure, the X radiation increased the thickness of the cortical bone in the time of 10 days (p < 0,05); decreased the number of trabeculae per unit of length and increased the medium distance between the trabeculae in the time of 33 days (p < 0,05); the sodium alendronate inhibited the deleterious effect of the X radiation avoiding the increase in distance between the trabeculae and reduced the number of trabeculae per unit of length in the time of 33 days (p < 0,05). In these experimental conditions it is concluded that the radiation X had a deleterious effect on bone microarchitecture and alendronate sodium preserved bone tissue subjected to the radiation X

▼ Physical inactivity and increased consumption of energy dense, high fat (HF) foods leads to a state of positive energy balance. Regular exercise can facilitate the maintenance of a healthy body weight and mediate changes in dietary selection. Past studies using a two-diet choice (chow vs. HF) and voluntary wheel running paradigm found that upon simultaneous introduction of a novel HF diet and wheel running, male rats show complete and persistent HF diet avoidance whereas the majority of females reverse HF diet avoidance.
Ovariectomy (OVX) appears to decrease HF diet preference, bringing it closer to what is observed in male rats. Given that estrogen, but not progesterone, mediates changes in food intake and energy balance, we hypothesized that estrogen signaling is required for the reversal of HF diet avoidance in female rats. Accordingly, Experiment 1 compared the persistency of running-induced HF diet avoidance in males, sham-operated females, and OVX rats with replacement of oil vehicle, estrogen (E), progesterone (P), or both (E+P). Results showed that the number of rats and time point at which wheel running rats reversed HF diet avoidance varied. The reversal of HF diet avoidance in running females and OVX E+P rats occurred more rapidly and frequently than male running rats. E+P, but not E or P, replaced OVX wheel running rats significantly reversed HF diet avoidance. OVX oil rats avoided HF diet to the same extent as male rats for the first 11 days of diet choice and then rapidly increased HF diet intake and began preferring it. This incomplete elimination of sex differences suggests that developmental factors or androgens might play a role in sustaining running-induced HF diet avoidance. Subsequently, Experiment 2 aimed to determine the role of androgens in the persistency of running-associated HF diet avoidance with sham-operated and orchiectomized (GDX) male rats. The results showed that both intact and GDX male running rats persistently avoided the HF diet to the same extent. Taken together, these results suggest that both organizational and activational effects of ovarian hormones play a role in female specific running-induced changes in diet choice patterns. Importantly, the activational effects of androgens are not required for the expression of HF diet avoidance in males.

▼ Este estudo teve como objetivo investigar os efeitos de diferentes tipos de exercícios físicos (corrida e natação) sobre parâmetros ósseos em ratas ovariectomizadas (OVX) e inteiras (SHAM). Ratas Wistar com 20 semanas de idade, peso inicial de 271,42 17,6g (média desvio padrão) foram usadas. Dez animais foram sacrificados quinze dias após a cirurgia e denominados baseline (GB), os demais foram alocados aleatoriamente em um dos seis grupos (n = 12): corrida OVX (CO); corrida SHAM (CS); natação OVX (NO); natação SHAM (NS); sedentário OVX (CONO) e sedentário SHAM (CONS). Os animais foram alojados em gaiolas individuais, em sala com temperatura ambiente de 22 2 C e ciclo de 12 horas claro/escuro, onde receberam diariamente 20g de ração e água adlibitum. Os animais dos grupos CO e CS foram submetidos a um programa de treinamento de corrida em esteira durante 10 semanas (16 m/min, 60 min/dia, 5 dias/semana, 8 semanas com 0% de inclinação e 2 semanas com 10 de inclinação). Os animais dos grupos NO e NS foram submetidos a um programa de treinamento em natação durante 10 semanas em um tanque com água a 30 2 C, com sobrecarga de até 3% do peso corporal (60 min/dia, 5 dias/semana). Os animais dos grupos CONO e CONS permaneceram em gaiolas individuais, sem exercício programado, por 10 semanas. Após a eutanásia, o fêmur direito foi removido para avaliação da densidade mineral óssea (DMO), do conteúdo mineral ósseo (CMO) e da resistência óssea. O fêmur esquerdo foi retirado para determinação da morfometria óssea. Toda a gordura visceral foi removida e pesada, descartaram-se pele, vísceras, cabeça e pés, permanecendo apenas músculo e ossos (carcaça vazia) para a análise quantitativa de água, gordura, proteína e cinzas. Amostras de sangue foram retiradas para análise do biomarcador fosfatase alcalina óssea (ostase). Ao final do experimento os animais OVX apresentaram maior (p <0,05) peso corporal que os animais SHAM. Os programas de exercício aumentaram (p <0,05) a DMO e o CMO dos grupos CO e NO. O programa de exercício em esteira aumentou (p <0,05) atenacidade de flexão no fêmur dos animais do grupo CO e a força de flexão no fêmur do grupo CS. Não houve aumento (p >0,05) de osso trabecular nas regiões da cabeça e do trocânter do fêmur em CO e NO. O programa de natação aumentou (p <0,05) a espessura do osso cortical no grupo NO e NS. A gordura visceral foi reduzida (p <0,05) no grupo CO. Não houve diferença (p >0,05) entre os grupos experimentais na porcentagem de água, gordura, proteína e cinzas. Os programas de corrida em esteira e natação com sobrecarga não aumentaram (p >0,05) os valores de ostase em ratas OVX. Concluiu-se que: a) os programas de exercícios aumentaram a DMO e o CMO de ratas OVX; b) O programa de exercício em esteira aumentou a tenacidade do fêmur de ratas OVX; c) O programa de exercício em natação aumentou a espessura do osso cortical em ratas OVX e SHAM; d) O programa de exercício em esteira reduziu a gordura visceral em ratas OVX; e) Os programas de exercício não aumentaram os níveis de ostase em ratas…

▼ Limited research has been conducted to elucidate age-related changes in the female rat heart. Further, no studies to date have analyzed mitochondrial morphology in the aged estrogen (E2)-deficient female rat heart. To examine the mitochondrial morphological alterations associated with age and E2 loss, we studied F334 female rats (n=40) subjected to coronary artery ligation (CAL) to induce ischemia-reperfusion (I/R) injury. In order to offer a more physiological model of estrogen deficiency with aging, we also utilized bilateral
ovariectomy (OVX) in a subset of rats at 15 mo to be studied at 22-23 mo (MO OVX, n=16). Both adult and MO OVX rats underwent 31 min ischemia with reperfusion times 10 min, 6 hr, and 24 hr. 7 day reperfusion was studied in a subset of adult (n=2) and ovary-intact aged rats (n=2). Transmission electron microscopy was used to analyze mitochondrial area, density, and circularity. Western blot analysis was conducted on a subset of samples to assess changes in mitochondrial fission and fusion proteins dynamin-related protein 1 (Drp1), mitochondrial fission 1 (Fis1), mitofusin 1 (Mfn1), and optic atrophy 1 (OPA1). Aging with E2 deficiency resulted in smaller, more numerous mitochondria compared to adult. Increases in mitochondrial area and decreases in density were seen in adult and MO OVX within 6 hr of reperfusion, while reperfusion over 24 hr seemed to indicate recovery from ischemic injury, as mitochondrial areas returned to baseline in adult hearts. Mitochondrial Drp1 significantly increased with I/R, while no significant changes were seen in Fis1, Mfn1 or OPA1. For the first time, our mitochondrial morphological data suggests a temporal resolution for I/R injury in the female rat heart.

▼ In late gestation, the rate of fetal growth increases significantly which must be matched by proper nutrient delivery. Although there is little change in placental weight at this stage, there is a large increase in fetoplacental capillaries resulting in an increased surface area for maternal-fetal exchange. The objective of this thesis was to determine the role of maternal ovarian or fetal signals in mediating fetoplacental angiogenesis. Changes in fetoplacental vascularity were assessed upon the removal of one or both ovaries in late gestation, or upon removal of fetal factors by inducing fetal cardiac arrest. Results suggest that ovarian and fetal factors promote fetoplacental microvascular growth while diminishing non-blood space components in the exchange region of the placenta in late gestation. Further studies are required to elucidate the specific factors involved, which is imperative for understanding why and how blunted fetoplacental capillarization occurs in pregnancy complications such as intrauterine growth restriction.

▼ Physical activity may increase long bone calcium (Ca) content to preserve bone strength in postmenopausal women. This study determined the effect of compulsory swimming and voluntary running exercise on bone mineral density, bone Ca and phosphorus (P) content, and femoral neck and tibia strength in ovariectomized (OVX) retired breeder rats, as a model for postmenopausal women. Thirty-seven nine-month old Sprague Dawley rats were assigned randomly into one of four treatment groups for the nine-week study: OVX + running (OR; n=9); OVX + swimming (OS; n=10); OVX + no exercise (O; n=9); sham-surgery + no exercise (Sh; n=9). OR rats had free access to running wheels; OS rats were trained over one week to swim for one hour, five days a week. At sacrifice, femurs, tibias, humeri, and vertebrae were removed. Bone mineral density was analyzed using pDEXA, and bone Ca and P content were analyzed using atomic absorption spectrometry and colormetric assay, respectively. Femur and tibia strength was determined by Q-tester. Bone mineral density was significantly higher for all bones measured in the exercise groups compared to the sedentary groups. Mean grams (g) of Ca per dry femur weight for OS rats were higher than O rats (p=0.019). Tibias of the OR and OS rats were able to absorb significantly more energy to break load than the O rats (p=0.000; p=0.001, respectively), and energy absorption was significantly higher for the OR compared to Sh tibia (p=0.022). No other significant differences in parameters were observed among the four groups. Results of this study suggest that both types of exercise improve bone mineral density, that swim exercise may be beneficial in preserving femur Ca content, and that swim exercise and voluntary running may be beneficial in improving tibia strength in OVX rats.

▼ Estrogen (E2) loss results in increased visceral adiposity and impaired glucose tolerance. Estrogen replacement may restore insulin sensitivity if given shortly after menopause, but may be associated with side effects such as ovarian cancer. Other alternatives should be explored. Natural compounds such as resveratrol (RESV) and grape pomace (GP) have potential insulin sensitizing effects. To our knowledge, the efficacy of RESV and GP in restoring glucose tolerance in ovariectomized (OVX) rats has not been explored. Diet was administered ad libitum to sham control (SHAM) and OVX female rats until the onset of glucose intolerance in OVX rats as determined by an intraperitoneal glucose tolerance test (IPGTT). Subsequently, OVX animals were divided into treatment and control groups. The treatment groups received daily, one of i) a physiological oral dose of E2, ii) RESV, or iii) GP supplemented in the diet. Treatment continued for 6 wks followed by IPGTT and insulin tolerance tests. 2-3 d later, terminal surgeries were performed during which red and white gastrocnemius muscles, visceral adipose tissue (VAT) and liver were sampled to assess Akt response to insulin injection. VAT was stained to determine adipocyte size. None of the treatments restored normal glucose tolerance. Insulin tolerance was not worsened in OVX rats, and was unaffected with treatment. No difference in phosphorylation of the insulin signaling protein, Akt, was found after insulin injection in any tissue between SHAM and OVX rats. Adipocyte size was significantly increased in the OVX-E2 and OVX-RESV groups compared to SHAM. Delayed E2 replacement, RESV and GP were not effective in restoring normal glucose tolerance in OVX rats.

▼ Ovarian secretions and growth hormone (GH) are major endocrine regulators of mammary growth and development in bovine mammary gland; but information on endocrine regulation during early prepubertal period is limited. Our overall objective was to study the regulation of mammary growth and development as well as transcript abundance in early prepubertal bovine mammary gland by ovarian secretions and exogenous bovine somatotropin (bST). In the first study, we determined the effect of staged
ovariectomy on mammary growth and development in two to four month old Holstein heifers. In the second study, effects of bST on mammary growth and development, and also on putative stem cell population were evaluated by beginning bST treatment in one month old Holstein heifers. Mammary growth and development was determined by mass of mammary tissue, biochemical analyses, histological examination, transcript abundance and protein expression in mammary parenchyma (PAR) and fat pad (MFP).
Ovariectomy reduced mass and lipid content of PAR without affecting the histological characteristics or rate of epithelial cell proliferation. There was a marked reduction in progesterone receptor expression both at the mRNA and protein level.
Ovariectomy also reduced transcript abundance in GH receptor (R), insulin-like growth factor (IGF)-1, IGF binding protein -6, estrogen responsive and proliferation marker genes, but increased the mRNA abundance of IGF-1R in PAR. On the other hand, administration of bST did not have an impact on PAR growth, epithelial proliferation, putative stem cell population or transcript abundance of IGF-axis genes. However, bST reduced the relative abundance of GHR, signal transducers and activators of transcription-5b and suppressors of cytokine signaling-2 in mammary PAR. Transcript abundance of IGF-axis molecules, estrogen responsive genes and proliferation markers in MFP was not affected by
ovariectomy or bST. Overall, our data suggest that ovary is a predominant regulator of mammary growth and development in prepubertal heifers and that exogenous bST is not effective as a mammary specific mitogen in very young prepubertal heifers.

▼ Osteoporosis is a skeletal disease that results in reduced bone mass, increased bone fragility and fracture risk. Osteoporotic patients who experience falls suffer serious fractures, hospitalization, and poor bone healing. Several different therapies have been developed for the treatment of osteoporosis, though many are unable to translate from the bench to the clinical population. A popular treatment being investigated is the application of mesenchymal stromal cells (MSC) for fracture repair and the reversal of osteoporotic bone losses. However, cells isolated from aged and osteoporotic patients have been shown to have deficient bone forming properties. Nevertheless, animal models of osteoporosis applying this treatment report amelioration of bone loss.
This work seeks to examine a more clinically relevant rat model of osteoporosis. Typical osteoporosis models use an
ovariectomy procedure to simulate post-menopausal bone loss on relatively young animals and conduct short-term studies. These studies may not accurately reflect the global regenerative changes in osteoporosis patients or the impaired MSC properties.
Adipose and bone marrow derived MSC from a long term
ovariectomy model were investigated for their regenerative potentials. MSC growth and bone forming potential was evaluated on keratin protein coated microcarriers in both static and perfusion cultures. Results from this study suggest that bone marrow derived MSC maintain their phenotype and functionality more effectively than adipose derived MSC in osteoporosis. Further preliminary results demonstrated the feasibility of MSC growth and differentiation on microcarrier based packed beds. These results highlight the complexity and challenges associated with the MSC regenerative strategy.

▼ Osteoporosis is an important systemic disorder, affecting mainly Caucasian women, with a diverse and multifactorial aetiology. A large variety of animal species, such as primates, sheep, swine, dogs, rabbits, rats and mice have been used as animal models in osteoporosis research. Their use enhanced our understanding on the etiology of osteoporosis. Additionally, many therapeutic regimes have been entered clinical practice after evaluation on animal models. Experimental protocols leading to bone loss, includes hormonal interventions (ovariectomy, orchidectomy, hypophysectomy, parathyroidectomy), immobilization, alcohol abuse osteoporosis and osteoporosis induced by food manipulation. Methods of bone mass evaluation, used on animal models are biochemical markers, densitometry, magnetic resonance imaging, histomorphometry and bone mechanical testing. The aim of the present study was to investigate the possible protective effect of the aqueous solution of the plant Sideritis euboea, with proven in vitro estrogenic action, on bone mass loss of the ovariectomized (OVX) rat experimental model of osteoporosis. Thirty-two intact female mature (10-month-old) Wistar rats were separated into 3 groups: sham-operated (Controls) (n=8), OVX (n=8) and OVX plus plant extract (Sid) (n=16). Sid administration in the drinking water commenced immediately after OVX. Bone mineral density (BMD) values, percentage change from the baseline measurement, pQCT parameters, and three point bending of the femurs, as well as body and uterine weight, were examined and compared between groups. Comparison of BMD absolute values of the whole tibia within OVX+Sid at 3 months revealed no statistical significant differences compared to baseline. While for the proximal tibial metaphysis mineral density absolute values did not significantly differ between Control and OVX+Sid groups demonstrating the plants beneficial effect for that period of time. When the % changes from baseline measurement of the whole tibia of OVX+Sid and Controls were compared, there was no significant difference at 3 months, demonstrating a highly protective effect. The respective comparisons of proximal tibia % changes in the OVX+Sid group although significant lower to the control group were significant less severe than the OVX group both at 3 and 6 months. pQCT evaluation showed a beneficial effect of the plant extract in the majority of the parameters evaluated. Body and uterine weight comparisons showed no significant difference between OVX and treated rats. The plant extract showed a protective effect on bones as evaluated by DEXA, pQCT, and three point bending. Furthermore the lack of significant effect on body and uterine weight is in favor of its selective estrogen receptor modulator-like activity, and merits further studies.

▼ Primary reperfusion therapy in patients with acute myocardial infarction is the standard of care, limiting infarct size and reducing mortality. Paradoxically, this restoration of coronary flow can also contribute to cardiac damage and complications, reffered to as reperfusion injury.The myocardium possesses adaptive mechanisms that render it more resistant to potentially lethal ischemia. Postconditioning represents phenomenon in which a series of brief episodes of ischemia are performed just at the onset of reperfusion following a prolonged ischemic insult. Since the incidence of coronary heart disease in females changes with the menopausal status, ovarian hormones are believed to protect female hearts from coronary heart disease but clinical investigations failed to demonstrate the usefulness of estrogen replacement therapy in postmenopausal females. We studied the postconditioining effect in female rabbit. They were subjected to the following protocol: 30 min ischemia of the heart and three hours of reperfusion. For ischemically postconditioning hearts, six cycles of 10 sec ischemia foolowed by 10 sec reperfusion were performed. The rabbits were divided into four experimental groups : 1) control female hearts, without postconditioning (FemC) 2) Female postconditioned hearts (FemPost), 3) Female ovariectomized without postconditioning (OvFem) and 4) Female ovariectomized with postconditioning (OvPostc). Another serie of same groups underwent endomyocardial biopsy in the first reperfusion minute for ERKs, p38-MAPK and AKt estimation. All hearts were excised, the coronary snare was retightened and 2- to 9- μm fluorescent microspheres ( Duke Scientific, Palo Alto, California) were infused for 3-5 minutes to delineate the area at risk as the nonfluorescent myocardial region. Hearts were frozen for approximately a day at -20 °C, were sliced into 3mm transverse slices from the apex to the base and then were incubated for 20min in 1% triphenyltetrazolium chloride solution ( pH 7.4, 37 °C) . The slices were introduced in 10% formaldehyde solution for 24-48h to delineate the infracted areas. The borders between fluorescent and nonfluorescent regions were marked under ultraviolet light (wavelength 366 nm). Infarct and risk zone areas were planimetered (Scion Image Program) with the aid of a scanner (Abode Photoshop 6.0), and volumes calculated by multiplying by the slice thickness (3mm). Infarct size and risk area volumes were expressed as cm3 and the infarct size is presented as a percent of risk zone (I/R).The infarct size was reduced only in OvFemPost group (25.3±2.7%, vs 48.05± 2% in FemC and 55.1±2.7% in Postc groups respectively, p<0.05). The phosphorylation of ERKs, p38-MAPK, and AKT in the ischemic region of the heart, was significantly higher in the protected OvxPostc group. We conclude that the ineffectiveness of Postc to reduce the infarct size in anesthetized female rabbits is restored after ovariectomy probably via the RISK pathway. ▼ A osteoporose, uma doença óssea progressiva, é considerada um grave problema de saúde pública, sendo uma das condições mais importantes associadas ao envelhecimento e que afeta milhões de pessoas no mundo. Esta doença multifatorial é caracterizada pela densidade óssea reduzida e deterioração da microarquitetura óssea. O objetivo deste trabalho foi investigar as mudanças comportamentais em células mesenquimais da medula óssea e células osteoblásticas da calvária de ratas induzidas à osteoporose. Após aprovação da Comissão de Ética no Uso de Animais, 18 ratas Wistar foram utilizadas e divididas em grupos controle e ovariectomizadas. Após 150 dias, as ratas de ambos os grupos foram sacrificadas para coleta dos fêmures e fragmentos da calvária. As células recolhidas a partir da medula óssea e calvária foram cultivadas em placas de 24 poços (n = 5) para avaliação da proliferação e viabilidade celular, atividade de fosfatase alcalina (ALP), detecção e quantificação de nódulos mineralizados e análise da expressão gênica por meio de PCR em tempo real. Os dados foram submetidos ao testes de Kruskal-Wallis e Mann-Whitney, com nível de significância de 5%. As células da medula óssea do grupo controle (MC) mostraram uma diminuição significativa na proliferação quando comparado com as células do grupo controle da calvária (CC) em todos os períodos (p < 0,05). Por outro lado, as células da medula óssea de ratas com osteoporose (MO) revelaram um aumento significativo na taxa de proliferação após 7 e 10 dias (p < 0,01) em comparação às células da calvária de ratas ovariectomizadas (CO). A viabilidade celular foi maior em todos os períodos estudados dos grupos CC e CO em relação aos grupos MC e MO (p < 0,05). A atividade de fosfatase alcalina não foi significativamente diferente após 7 dias de cultura entre os grupos estudados; por outro lado, após 10 e 14 dias, observou-se uma diminuição da sua atividade no grupo MC quando comparado ao grupo CC (p < 0,01). Nas ratas com osteoporose, as células da medula óssea mostraram um aumento desta atividade quando comparada às células de calvária (p < 0,01). A análise dos nódulos mineralizados após 14 e 21 dias revelou que os grupos controle CC e MC não apresentaram diferenças significativas, ao passo que no grupo MO observou-se um aumento da mineralização quando comparado ao grupo CO nos mesmos períodos experimentais. Os resultados obtidos na análise de expressão gênica mostraram que para os genes Runx2, Oc, Alpl, Rank-l e Erα a expressão no grupo MO foi maior que no grupo MC (p < 0,05), enquanto que para as células da calvária, CC teve maior expressão gênica que CO (p < 0,05). Os genes Opg e Erβ apresentaram variações de acordo com o grupo avaliado. Frente aos resultados obtidos, sugere-se que existam alterações no metabolismo das células progenitoras e células diferenciadas após a indução da osteoporose e que as células da medula óssea apresentam um aumento da sua função como uma resposta compensatória neste modelo animal ovariectomizado.

Sabe-se também que a atividade física é capaz de trazer benefícios para este tipo de tecido, reduzindo os riscos de lesão articular. Contudo, existem poucos os trabalhos que relacionam estes dois fatores: deficiência estrogenica e atividade física regular. Foram utilizadas 12 ratas Wistar adultas, divididas em quatro grupos de três animais, de modo aleatório e sistematizado, sendo eles: ratas controle sedentárias (RCS), ratas controle treinadas (RCT), ovariectomizadas sedentárias (ROS) e ovariectomizadas treinadas (ROT). Cada grupo foi analisado pelas técnicas de microscopia de luz e microscopia eletrônica de transmissão. Os resultados foram obtidos a partir do método estereológico. Os valores foram estatisticamente significantes em relação a deficiencia estrogenica para os parametros: peso, condicionamento físico, densidade numérica de condrócitos, número total de condrócitos, densidade de volume dos condrócitos, densidade de volume da matriz extracelular, volume total de matriz extracelular e densidade de volume de colágeno. Em relação aos efeitos concomitantes da atividade física e da deficiência estrogênica, os resultados foram significativamente diferentes em relação a densidade numérica, volume total da cartilagem, volume médio dos condrócitos e densidade de volume de colágeno. Estes dados nos levam a concluir que a diminuição dos níveis de estrógeno afeta profundamente a cartilagem articular da cabeça do fêmur, enquanto a atividade fisica contínua é benéfica para esta cartilagem, incluseve quando há deficiência estrogênica.
Several studies show that estrogen has an important function in maintaining the structure of articular cartilage and that deficiency in women can cause an increase in the incidence of joint degeneration. It is also known that physical activity can bring benefits to this type of tissue, reducing the risk of joint damage. However, there are few works that relate these two factors: continuous physical activity and estrogen deficiency. The aim of this study is to verify the effects of estrogen deficiency on joint cartilage, its implications, and if frequent and moderate physical activity is able to reverse the natural process of joint degeneration. ▼ Osteoporosis a silent, progressive, systemic skeletal disease characterized by impaired bone architecture with decreased bone mass density affecting women in menopausal and after menopausal stage and men in the third age, mainly in the ratio 6:1. Periodontitis a chronic inflammatory, multifactorian disease has been associated with general osteoporosis .Statins, inhibitor of HMG-CoA-reductase, seems to interfere in bone formation exerting anabolic and antiresorptive effect. The study investigates the bone density in mandible and femur in an ovariectomized (OVX) rat model of osteoporosis in order to test the effect of simvastatin on bone size parameters.50 Wistar female rats aged 4-6 weeks, weighting 200-300g were randomized in five groups. Animals in Group 1,2 and 3 (n=10) were overiectomized and group 4 and 5 serve as contols. Group 2,3 and 5 were treated with simvastatin 0,5 mg/kg/daily p.o. for 3 months. Sizes parameters of the isolated femur, mandible and uterus were measured, BMD(bone mass density) via dual energy X-Ray absorptiometry (DEXA) and laboratory serum findings were estimated.Ovariectomy leads to experimental menopause with decreased uterus weight and reduction of all parameters in femur and mandible (weight, specific weight, bone weight /body weight ) that have been restored under the influence of statins. DEXA results showed significant decrease of BMD in ovariectomy group / controls while statins treatment decrease the severity of BMD lose. The serum IL-13 as osteogenesis index seems to be positive influenced by statins treatment.
H Οστεοπόρωση είναι σιωπηλή, εξελισσόμενη, συστηματική νόσος του σκελετού, που χαρακτηρίζεται από διαταραχή της αρχιτεκτονικής του οστού με ελάττωση της οστικής μάζας/πυκνότητας και προσβάλλει γυναίκες στην εμηνοπαυσιακή και μεταεμμηνοπαυσιακή φάση αλλά και άνδρες στην 3η ηλικία κυρίως σε αναλογία 6:1. H περιοδοντίτιδα χρόνια φλεγμονώδης, πολυπαραγοντικής προέλευσης, νόσος έχει συσχετιστεί με τη γενική οστεοπόρωση (απώλεια της οστικής πυκνότητας).Οι στατίνες, αναστολείς της HMG-CoA-αναγωγάσης, δρουν όχι μόνο ως παράγοντες μείωσης της χοληστερόλης, αλλά μπορεί να διεγείρουν σχηματισμό οστού με την αναβολική και αντιαπορροφητική τους δράση, όπως τεκμηριώνεται σε σειρά μελετών.Ο σκοπός της μελέτης ήταν να ερευνηθεί ο ρόλος των στατινών ως οστεοαναγεννετικών παραγόντων σε εγκατεστημένη οστεοπόρωση επιμύων (Wistar) λόγω έλλειψης οιστρογόνων (από ωοθηκεκτομή) και να εξεταστεί η προληπτική δράση αυτών σε ήδη οστεοπορωτικό υπόστρωμα. 50 θηλυκοί αρουραίοι τύπου Wistar ηλικίας 4-6 εβδομάδων, βάρους 200-300 gr χωρίστηκαν σε πέντε ομάδες. ▼ I conducted 4 field trials to evaluate population estimation methods and the application of immunocontraceptive vaccines or surgical sterilization to overabundant deer in urban/suburban communities where lethal removal is infeasible. I
demonstrated that sterilizing a high percentage of a deer population with no operational-related mortalities is possible, so surgical sterilization may be viable for some communities. I also evaluated non-baited camera surveys for monitoring deer
populations and concluded that they produced consistent trend data allowing for management decisions over time. To evaluate whether immunocontraceptive vaccines should be registered for dart-delivery, I presented data on both the injection dispersion and
efficacy, suggesting that dart-delivery should not be approved. Furthermore, I presented data on effort required to live-capture and hand-inject deer with immunocontraceptive vaccines, and the additional effort required to surgically sterilize deer. I
paired these effort data with deer approachability rankings and showed that effort increased significantly as deer approachability decreased.

▼ O presente estudo teve como objetivo avaliar o efeito do laser de baixa potência com comprimento de onda de 660nm e 808nm sobre o processo de reparação de defeitos ósseos produzidos no fêmur de ratas submetidas à ovariectomia. Para tanto foi realizada ovariectomia bilateral em 18 ratas de linhagem Wistar distribuídas em grupo controle e irradiado após a comprovação da diminuição da massa óssea através de análise digital da densidade óssea, foi realizado uma perfuração padronizada no fêmur. Os grupos irradiados receberam 120J/cm2 dos lasers As-Ga-Al (660nm) e In-Ga-Al-P (808nm). Os animais foram submetidos à eutanásia em 14 e 21 dias após confecção do defeito ósseo. A avaliação histológica dos eventos ocorridos foi feita de forma descritiva, utilizando-se de um método semi-quantitativo através de microscopia óptica. Os resultados nos períodos de 14 e 21 dias mostraram que os grupos irradiados apresentaram superfície osteoblástica, fibroblástica e de osteócitos imaturos, significativamente maiores que o grupo controle (p<0,05). Análise de reação inflamatória mostrou que a TLB foi eficaz, pois, poucas células inflamatórias foram encontradas quando comparadas ao grupo controle com p<0,05. Conclui-se no modelo experimental estudado que ambos os comprimentos de ondas utilizados induziram a proliferação de células responsáveis pela consolidação óssea.

A biossÃntese desse hormÃnio à catalisada pela enzima aromatase e sua inibiÃÃo à importante para terapia do cÃncer de mama. Os pacientes que usam inibidores da aromatase como o Anastrozol (ANA) mostram maior nÃmero de fraturas e menor densidade Ãssea. Considerando, entÃo, o papel do estrÃgeno na resposta inflamatÃria e no metabolismo Ãsseo, alÃm dos efeitos adversos do ANA e da periodontite ser caracterizada por um processo inflamatÃrio que resulta em perda Ãssea alveolar, o objetivo desse estudo foi de investigar se o ANA afeta a periodontite em ratas ovariectomizadas. Para isto, o ANA (0,02, 0,1 e 0,5 mg/kg-v.o.) foi avaliado em um modelo de periodontite induzida por ligadura em ratos durante 11 dias. A periodontite foi analisada atravÃs de macroscopia, de histologia e da atividade de mieloperoxidase (MPO). As dosagens sÃricas de estrÃgeno, o leucograma, a variaÃÃo de massa corpÃrea e as funÃÃes hepÃtica, renal e esplÃnica tambÃm foram consideradas. A ovariectomia reduziu os nÃveis sÃricos de estrÃgeno apÃs 14 dias, mas nÃo exacerbou a periodontite ou aumentou a atividade de mieloperoxidase (MPO) quando comparada aos animais falso ovariectomizados (F-OVX). Embora o ANA nÃo tenha aumentado a perda Ãssea alveolar (POA), a administraÃÃo deste fÃrmaco por 11 dias aumentou a atividade de MPO. A ovariectomia combinada ou nÃo ao ANA nÃo promoveu nenhuma mudanÃa nas funÃÃes hepÃtica, renal ou esplÃnica, mas causou uma leucocitose promovida por cÃlulas mononucleares no 11 dia. Os animais ovariectomizados que receberam salina (SAL) tambÃm mostraram maior ganho de massa corpÃrea e a curva de peso do ANA foi similar à curva do SAL. Em conclusÃo, embora a reduÃÃo de estrÃgeno por 25 dias e a administraÃÃo de ANA por 11 dias nÃo tenha aumentado a POA, o ANA aumentou a atividade de MPO.
Estrogen deficiency was shown to increase bone remodeling, may affect some bone diseases as periodontitis. Biosynthesis of this hormone is catalyzed by aromatase enzyme and her inhibition is important for breast cancer therapy. The patients who use aromatase inhibitors as Anastrozole (ANA) show greater number of fractures and lower bone density. Since estrogen role in the inflammatory response and the bone metabolism, besides those adverse effects of ANA and periodontitis is characterized by an inflammatory process that results in alveolar bone loss, the aim of this study was to investigate whether ANA affects the periodontitis in ovariectomized rats. For this, ANA (0.02, 0.1 and 0.5 mg/kg-v.o.) was evaluated in ligature-induced periodontits for 11 days in ovariectomized rats. Periodontitis was analyzed through macroscopy, histology and by myeloperoxidase (MPO) activity. Serum dosage of estrogen, leukogram, corporal mass variation and liver, renal and spleen functions were also analyzed. ▼ Dentre as doenças neurodegenerativas, as que causam desmielinização do sistema nervoso central tem grande importância, como a esclerose múltipla (EM) que acomete adultos jovens e de meia-idade. Neste sentido, o 17-β estradiol e o resveratrol surgem como substâncias promissoras devido as suas ações antioxidante, antiinflamatória e neuroprotetora. Assim, o objetivo do presente estudo foi verificar o efeito do 17-β estradiol e do resveratrol nos parâmetros bioquímicos e hematológicos em ratas ovariectomizadas submetidas ao modelo de desmielinização pelo brometo de etídio (BE). No primeiro artigo, os animais foram separados em 3 grupos experimentais para cada idade. Os grupos consistiram de controles adultos (Sham-A) e de meia-idade, ratas ovariectomizadas adultas e de meia-idade e ratas ovariectomizadas com reposição por 30 dias com 17-β estradiol adultas e de meia-idade. Após este período, a atividade da AChE foi mensurada no encéfalo,sangue total e linfócitos, e a peroxidação lipídica foi mensurado apenas no encéfalo. Os resultados obtidos demonstraram que a atividade da acetilcolinesterase (AChE) aumentou no estriado (ST) de ratas adultas ovariectomizadas sem e com reposição estrogênica, e de meia-idade sem reposição. No hipocampo (HP) também foi observado um aumento na atividade desta enzima apenas nas ratas de meia idade sem reposição do hormônio. A inibição na atividade da AChE ocorreu no ST e no córtex cerebral (CC) de ratas adultas com reposição estrogênica e nas ratas de meia-idade com e sem reposição hormonal. A atividade da AChE aumentou no sangue total de ratas adultas com reposição estrogênica e diminuiu nas ratas de meia-idade sem 17-β estradiol. Nos linfócitos foi observado um aumento na atividade da AChE de ratas adultas com ou sem reposição de estrógeno, e inibida no grupo de meia-idade sem reposição estrogênica.Houve um aumento na peroxidação lipídica em todas as estrituras encefálicas dos animais adultos ovariectomizados e córtex cerebral e cerebelo dos animais de meia-idade ovariectomizados.. Nos manuscritos, os animais foram divididos em 5 grupos para avaliar a fase de desmielinização e outros 5 grupos para avaliar a fase de remielinização. Em cada fase, os grupos consistiram de ratas controle; ratas ovariectomizadas; ratas ovariectomizadas e desmielinizadas; ratas ovariectomizadas tratadas com 17-β estradiol (maunscrito 1) ou resveratrol (manuscrito 2); e ratas ovariectomizadas e desmielinizadas tratadas com 17-β estradiol (manuscrito 1) ou resveratrol (manuscrito 2). No manuscrito 1, investigou-se os efeitos do 17-β estradiol sobre os parâmetros relacionados a atividade da AChE encefálica, do sangue total e dos linfócitos, e atividade da butirilcolinesterase (BuChE) por um período de 7 ou 21 dias.
