Neutrofilai ir makrofagai – vienos svarbiausių uždegiminiame procese dalyvaujančių ląstelių. Lėtinės obstrukcinės plaučių ligos (LOPL) metu aktyvintos šios ląstelės sintetina biologiškai aktyvias medžiagas, skatinančias intensyvesnį sisteminį ir kvėpavimo takuose vykstantį uždegimą. Iki šiol nėra aiškūs neutrofilų ir makrofagų sintetinamų žymenų (citokinų, proteinazių) ypatumai sergant LOPL ir atsižvelgiant į rūkymo įprotį. Nėra žinoma, kaip kraujo neutrofilų funkcines savybes tiesiogiai įtakoja pats sergančiųjų LOPL kvėpavimo takų sekretas. Šio tyrimo tikslas buvo įvertinti neutrofilų ir makrofagų funkcinių savybių ypatumus sergant lėtine obstrukcine plaučių liga ir galimus jų skirtumus priklausomai nuo rūkymo įpročio. Ištirti sergančių LOPL rūkančiųjų ir neberūkančiųjų matrikso metaloproteinazės-12 raiškos kvėpavimo takų sekreto makrofaguose ypatumai. Įvertintos sergančiųjų LOPL biologiniais ir cheminiais veiksniais aktyvintų kraujo neutrofilų funkcinės savybės (chemotaksis, fagocitozė, reaktyviųjų deguonies formų susidarymas), analizuotas potencialus moduliacinis kvėpavimo takų sekreto poveikis šioms funkcinėms savybėms. Nustatyta, jog sergančiųjų lėtine obstrukcine plaučių liga neutrofilai ir makrofagai buvo labiau aktyvūs nei sveikų asmenų ląstelės; rūkymo įprotis įtakojo kai kurias uždegimines neutrofilų ir makrofagų savybes kvėpavimo takų sekrete ir kraujyje.

Neutrophils and macrophages play an important role in inflammatory process. Activated during chronic obstructive pulmonary disease (COPD), these cells synthesize a variety of biologically active compounds, which may further amplify both systemic and local airway inflammation. The features of biologically active compounds (cytokines, proteinases) released by neutrophils and macrophages during COPD and according to smoking status are unknown. Still, a direct influence of airway mucous from COPD patients on functional neutrophil features is unknown. The aim of this study was to evaluate the functional features of neutrophils and macrophages during chronic obstructive pulmonary disease and possible differences according to smoking status. An expression of matrix metalloproteinase-12 in airway mucous of COPD smokers and ex-smokers was evaluated. The functional features (chemotaxis, phagocytosis, production of reactive oxygen species) of biologically and chemically activated blood neutrophils and a modulator effect of airway mucous were also analysed. The neutrophils and macrophages of patients with chronic obstructive pulmonary disease were more activated than the cells of healthy individuals; smoking status has influenced some inflammatory features of neutrophils and macrophages in airway mucous and blood.

Le RXP470.1 est l’un des premiers inhibiteurs puissants de la MMP-12, une métalloprotéase à zinc impliquée dans de nombreuses pathologies comme l’athéroclérose et la bronchopneumopathie obstructive chronique (BPCO). Pour comprendre les bases moléculaires contrôlant l’interaction de cet inhibiteur avec sa cible, des approches pluridisciplinaires associant des relations structure-activité, avec des études de cristallographie de complexes enzymes inhibiteurs et d’études de microcalorimétrie, décrivant les contributions enthalpiques et entropiques impliquées dans la formation des complexes, ont été réalisées dans ce travail de thèse. Les affinités de trois analogues du RXP470.1 ont été tout d’abord déterminées. Puis quatre structures cristallographiques de complexes enzyme/inhibiteur décrivant le mode d’interaction duRXP470.1 et de ces trois analogues ont été obtenues avec des résolutions de 1.15 Å, 1.50 Å, 1.50Å et 1.30 Å, respectivement. Parallèlement les études de microcalorimétrie ont été menées pour étudier les facteurs énergétiques contrôlant l’interaction du RXP470.1 avec la MMP-12. Les résultats indiquent que la présence d’une chaîne latérale très longue et hydrophobe en position P1’de l’inhibiteur s’insérant dans la cavité S1’ de la MMP-12 est essentielle à la très bonne affinité de cet inhibiteur pour la MMP-12. Cette interaction met essentiellement en jeu un effet entropique très important de - 4 kcal/mol. L’interaction du RXP470.1 est aussi essentiellement dirigée par une forte augmentation d’entropie (-TDS= -10 kal/mol) et une composante enthalpique beaucoup plus faible (DH= -2.5 kcal/mol), et ce malgré l’observation dans le cristal de nombreuses interactions entre l’inhibiteur et le site actif de la MMP-12. L’étude de microcalorimétrie met aussi en lumière la prise d’un proton au cours de la formation du complexe enzyme inhibiteur impliquant deux résidus chargés négativement en solution, le résidu catalytique Glu219 et le groupe phosphoryle chélatant du zinc dans l’inhibiteur. Cette étude révèle aussi que si le groupe phosphoryle est considéré comme un chélatant faible de l’atome de zinc, il impose néanmoins des contraintes directionnelles très importantes qui ont un impact sur le positionnement des autres parties de l’inhibiteur dans le site actif de l’enzyme. Ce dernier effet pourrait expliquer pourquoi un certain nombre d’interactions entre l’inhibiteur et l’enzyme ne sont pas optimisées et pourquoi la variation d’enthalpie pour former le complexe reste relativement faible. Cette étude ouvre maintenant la voie à d’autres études en plaçant au centre des futurs travaux le rôle du groupe chélatant dans la conception des inhibiteurs de MMP, ainsi de nouveaux inhibiteurs puissants et sélectifs d’autres MMP devraient voir le jour grâce à ce travail et aux résultats obtenus.

Les mécanismes de reconnaissance moléculaire sont à la base de nombreuses fonction biologiques essentielles (transduction du signal, régulation de l’expression génique, stimulation du système immunitaire,...). La compréhension des phénomènes physiques et chimiques à la base de ces phénomènes est fondamentale pour de nombreuses applications telles que la conception de médicaments, le développement d’outils diagnostiques ou tout autre procédé biotechnologique. Dans cette étude, nous avons étudié de manière extensive l’interaction entre la métalloélastase du macrophage (MMP-12) et le RXP470.1, un inhibiteur puissant et sélectif. En combinant des approches de cristallographie, de microcalorimétrie (ITC) et des tests enzymatiques, nous avons pu quantifier l’importance énergétique du transfert d’un seul proton suite à la liaison du RXP470.1, et mettre en lumière l’importance des contributions entropiques. Ainsi, la protonation du Glu219, un résidu catalytique, permet de compenser une enthalpie de liaison intrinsèque défavorable. Cette protonation est rendue possible par le large shift de pKa que subit le Glu219 en réponse à la liaison du RXP470.1 (pKalibre = 5.7 ± 0.1 / pKalié = 10 ± 0.04). Enfin, cette étude est la première, à notre connaissance, ayant combiné données d’affinité, thermodynamiques et structurales pour aborder le rôle du groupe chélatant. Nous avons ainsi étudié deux analogues du RXP470.1 variant seulement par la nature de leur pince à zinc. Ces modifications se traduisent par un effet marqué sur les profils d’affinité et de sélectivité ainsi que sur la signature énergétique des composés étudiés. L’étude des facteurs B, associés à l’analyse des structures cristallographiques, de ces inhibiteurs en complexe avec la MMP-12, suggère que des différences mineures de structures peuvent engendrer des variations de mobilité importantes au niveau des résidus impliqués dans l’interaction inhibiteur - enzyme. Ces différences trouvent leur origine dans un positionnement très légèrement différent du groupe chélatant par rapport au zinc.Pris dans leur ensemble, ces résultats pointent la nécessité de combiner un ensemble d’approches expérimentales pour décrire la complexité des interactions protéine/ligand. Ces associations doivent permettre d’évaluer le potentiel de méthodes théoriques capables de décrire des systèmes complexes.

▼ Cardiovascular disease is the largest single cause of mortality and its major underlying pathology is atherosclerosis. The proliferation of vascular smooth muscle cells (VSMC) is a key event in the pathogenesis of various vascular diseases, including atherosclerosis and hypertension. Thrombin is involved in the differentiation and abnormal proliferation of VSMC associated with atherosclerosis and hypertension. Thrombin stimulation results in extracellular signal-regulated kinase (ERK 1/2) activation through transactivation of the epidermal growth factor receptor (EGFR). Based on the studies in which PD98059 used to inhibit ERK1/2, we have shown previously that ERK1/2 was involved in the regulation by thrombin of VSMC’s proliferation. In addition, heparin-binding EGF-like growth factor (HB-EGF) and matrix metalloproteinases (MMPs) have also been detected in VSMC and shown to be regulated by thrombin. ADAMs (A Disintegrin And Metalloproteinase) are transmembrane metalloproteinases, belonging to adamalysin group, that are distinct from matrix metalloproteinases (MMPs) in that, they have an extracellular disintegrin domain and cytoplasmic domain that can associate with intracellular proteins. To the present knowledge there is no study that indicates the activation of an ADAM member in thrombin-induced VSMC proliferation. In this dissertation, the role of EGFR, ERK1/2, HB-EGF, general metalloproteinases, MMP-2 and ADAM 12, as well as PKCδ in mediating the mitogenic action of Thrombin in rat VSMC was investigated. Incubation of rat VSMC with Thrombin (1 U/ml) for 5 minutes resulted in significant increase of ERK1/2 phosphorylation by 8.7 ± 0.9 fold (p<0.001), EGFR phosphorylation by 8.5 ± 1.3 fold (p<0.001) and DNA synthesis by 3.6 ± 0.4 fold (p<0.001). Separate pretreatments for 30 minutes with EGFR tyrosine kinase irreversible inhibitor, 10 μM PD169540 (PD), and 20 μM anti-HB-EGF antibody, significantly reduced thrombinstimulated EGFR and ERK1/2 phosphorylation by 81 %, 72 % and by 48 % and 61%, respectively. Furthermore, same pretreatments with PD and anti-HB-EGF antibody reduced Thrombin-induced VSMC’s proliferation by 44% and 45%, respectively. In addition, pretreatments for 30 minutes with 10 μM KB-R7785 (KB), a specific ADAM 12 inhibitor or 10 μM specific MMP2 inhibitor significantly reduced thrombin-stimulated EGFR and ERK1/2 phosphorylation by 47 %, 47 % and by 25 % and 25 %, respectively. Furthermore, same pretreatments with either KB or MMP2 inhibitor reduced thrombin-induced VSMC’s proliferation by 27 % and 45 % respectively. Pretreatment of VSMC with 5 μM rottlerin, a specific inhibitor of PKCδ, for 30 min significantly reduced EGFR and ERK1/2 phosphorylation by 58 % and 55%, respectively and reduced significantly thrombin-induced VSMC’s proliferation by 32%. Kardiovaskularne bolesti predstavljaju najveći uzrok smrtnosti ljudske populacije, a njihova glavna i osnovna patološka komponenta je ateroskleroza. Proliferacija ili deoba glatkih mišićnih ćelija krvnog suda (VSMC) ključni je događaj u nastanku raznih vaskularnih oboljenja, uključujući aterosklerozu i hipertenziju. U procesu diferencijacije i abnormalne deobe VSMC povezanih sa hipertenzijom i aterosklerozom uključen je i trombin. Stimulisanje VSMC trombinom dovodi do aktivacije ekstracelularnim signalima regulisanih kinaza 1 i 2 (ERK1/2), preko transaktivacije receptora za epidermalni faktor rasta (EGFR). U ranijim studijama Isenović i saradnici potvrdili su na osnovu inhibicije ERK1/2 od strane PD9805 inhibitora, učešće ERK1/2 u regulaciji proliferacije VSMC izazvanoj trombinom. U nastavku, faktor rasta sličan epidermalnom faktoru rasta koji vezuje heparin (HB-EGF), protein kinaza C delta (PKCδ) i matriksne metaloproteinaze (MMP), nađene su u VSMC i pokazano je da je i njihova aktivnost regulisana trombinom. ADAM (engl. “A Disintegrin And Metalloproteinase”) transmembranske su metaloproteinaze koje pripadaju adamalizinskoj grupi i razlikuju se od matriksnih metaloproteinaza po tome što imaju vanćelijski dizintegrinski i citoplazmatski domen. C terminalni domen može da stupa u interakciju sa unutarćelijskim proteinima. Uvidom u literaturu, do sada ne postoji istraživanje koje ukazuje na prisustvo ADAM metaloproteinazne aktivnosti vezane za proliferaciju VSMC stimulisane trombinom. Predmet ove doktorske teze jeste proučavanje uloge EGFR, ERK1/2, HB-EGF, PKCδ, ukupnih metaloproteinaza, kao i specifičnih MMP-2 i ADAM 12, u posredovanju proliferativnog efekta trombina na VSMC pacova. Inkubacija VSMC pacova sa trombinom (1 U/ml) u periodu od 5 minuta rezultirala je u značajnom povećanju: fosforilacije ERK1/2 od 8.7 ± 0.9 puta (p<0.001), fosforilacije EGFR od 8.5 ± 1.3 puta (p<0.001) i sinteze DNK od 3.6 ± 0.4 puta (p<0.001). Prethodni pojedinačni tretmani ovih ćelija u trajanju od 30 minuta sa 10 μM PD169540 (PD), ireverzibilnim inhibitorom EGFR, i 20 μM antitelom protiv HB-EGF značajno su smanjili trombinom stimulisanu fosforilaciju EGFR za 81 % i 72 % i ERK1/2...

Le développement d’une nouvelle sonde dite « activity-based probe » pour réaliser la détection de formes actives de protéases appartenant à la famille des protéases à zinc de la matrice (MMP) a été réalisé dans ce travail, en partant d’un inhibiteur phosphinique puissant des MMP dans lequel a été introduit un groupement photoactivable de type diazérine. Ce composé se révèle un inhibiteur puissant de plusieurs MMP avec des affinités nanomolaires. Ce composé incubé avec différentes MMP est par ailleurs capables de modifier de façon covalente un grand nombre de MMP au niveau de leur site actif, avec des rendements de modification variant de plus de 50% à 11%, selon la nature des MMP. En ayant choisi comme moyen de détection la radioactivité, nous démontrons qu’avec cette nouvelle sonde qu’il est possible de détecter des formes actives de MMP avec des sensibilités de l’ordre de la femtomole dans des systèmes modèles de protéomes complexes. Appliquée à l’analyse de lavages broncho alvéolaires de souris traitées par voie pulmonaire avec des nanoparticules pour induire une réponse inflammatoire, cette nouvelle sonde permet de mettre en évidence la présence de formes actives du domaine catalytique de la MMP-12, une métalloprotéase à zinc exprimée par les macrophages, mais pas dans les animaux contrôles. En revanche l’analyse de carotides humaines de patients souffrant d’athérosclérose ne nous pas conduit avec cette sonde à la détection de formes actives de MMP. Malgré ce résultat, il est à noter que la détection de forme active de MMP dans un fluide pathologique est une première dans ce domaine. Cette sonde étant validée pour sa capacité à détecter des formes actives de MMP, elle permettra dans l’avenir de tester d’autres fluides pathologiques d’origine humaine ou bien des extraits de tissu comme des tumeurs pour lesquels les MMP pourraient être des marqueurs de ces pathologies.

A new activity-based probe able to covalently modify the active site of proteases belonging to the matrix metalloprotease family (MMPs) has been developed in this thesis project. The probe was shown to behave as potent inhibitor of several MMPs, with nanomolar Ki values. This probe was also able to modify specifically only the free active site of MMPs, with particular high yields of cross-linking varying from 50 % to 11 %, depending of the MMPs tested. Using radioactivity as means of detection, this probe was able to detect active form of MMPs with a threshold of 1 femtomole. Applied to the study of bronchoalvelolar fluids (BAL) from mice exposed to nanoparticles by a lung aspiration protocol, this probe revealed the presence of the catalytic domain of MMP-12 under its active form, but not in control animals. When used to detect active form of MMPs from extracts obtained from human arteries of patient suffering from atherosclerosis, the probe was not able to detect such MMP active forms. ▼ Perinatal brain injury, as a result of hypoxia-ischemia (HI) or infection/HI, is a major cause of acute mortality and neurological morbidity in infants and children. The mechanisms of perinatal HI are not fully understood, which makes it difficult to find effective treatment. The aim of the thesis was to investigate the mechanisms of perinatal HI brain injury, and to evaluate different neuroprotective strategies; 1) effects of N-acetylcysteine and melatonin after LPS-sensitized HI, 2) effects of a GPE analogue (G-2mPE) after HI, 3) the involvement of matrix metalloproteinase (MMP) -9 and -12 after HI. An animal model of perinatal brain injury was used in the neonatal rat/mouse, i.e. permanent ligation of the left carotid artery, followed by exposure to a gas mixture with low oxygen content, either alone (HI) or in a combination with infection (LPS/HI). Neuroprotective effects of N-acetylcysteine and melatonin were investigated in neonatal pups after LPS/HI. The drugs were given in multiple doses and brain injury was evaluated 7 days after the HI insult. The neuroprotective effect of post HI administered G-2mPE was investigated in neonatal rats. MMP-9 gene deficient mice were used to evaluate the importance of MMP-9 after perinatal HI. MMP-12 expression after HI was investigated in wild type animals after perinatal brain injury. Marked neuroprotection was found with NAC treatment, which was associated with reduced isoprostane activation and nitrotyrosine formation, increased levels of the antioxidants glutathione and thioredoxin-2 and inhibition of caspase-3, calpain, and caspase-1 activation. A moderate reduction of brain damage was obtained after pre/post treatment with melatonin. Post-HI treatment with G-2mPE attenuated neuronal injury and promoted astrogliosis, as well as blood vessel growth. MMP-9 was shown to play an important role in the development of HI injury in the immature brain, particularly with regard to blood-brain barrier leakage and inflammation. MMP-12 may also be important for the development of brain injury, as the MMP-12 mRNA expression is up-regulated 24 hours after HI and an increased number of cells express MMP-12 in the damaged ipsilateral hemisphere.
