Osteoporosis and atherosclerosis are two multifactorial and degenerative diseases, which constitute major problems to the public health. These age-related diseases, share common risk factors and because of that, several pathophysiological processes have been proposed. Diabetes mellitus, blood hypertension, cancer, smoking, alcohol consumption, low physical activity, are some of the common risk factors, which can cause atheromatic and osteoporotic disease. To investigate the genetic background of these diseases, researchers are studying genetic polymorphisms that either singularly (correlation study) or in combination (genetic sum) or in interaction with known risk factors may contribute to the emergence of these diseases.The first are based on connections of gene polymorphisms with the diseases in question, comparing the frequency of appearance of the polymorphisms that are studied, among patients and healthy people as far as it concerns the phenotype of the disease.The purpose of this study is to determine the possible association of gene polymorphisms MTHFR C677T, MTHFR A1298C, MTRR A66G, with atheromatic and osteoporotic disease in a Greek population living in Evros.An additional aim of this study is to determine the model of the genetic sum, in an effort to estimate the overall impact of the polymorphisms that are studied, on atheromatic disease and osteoporosis.Finally, we seek to analyze the interaction of the studied polymorphisms with coexisting pathological conditions of patients, such as, diabetes mellitus, hypertension, cancer, smoking habits, alcohol consumption and levels of physical exercise.Our sample comprises a total of 200 people. Regarding the study of atherosclerosis, 100 people represent a group of patients, of which 51 suffer from carotid stenosis, and 49 suffer from peripheral vascular disease. All of these patients suffered from heavily diagnosed atheromatic disease and entered the hospital in order to have surgical treatment to their problem. The remaining 100 participants constitute the control group, which contains healthy people as far as it concerns the atheromatic disease, with no clinical findings or medical history of other atheromatous diseases.Regarding the study of osteoporosis, a total of 61 women took part. Of these, 26 are the group of patients who suffer from diagnosed osteoporosis (tscore < -2,5) and 35 constitute the control group, which are healthy to osteoporosis individuals (tscore > -2,5).In all the participants, the C677T and A1298C polymorphisms of the MTHFR gene were analyzed as well as the A66G polymorphism of the MTRR gene.These genes play an important role to the metabolism of homocysteine and the polymorphisms that they are consisted of, are accused of the appearance of high concentration of homocysteine. Hyperhomocysteinemia is a possible risk factor for the appearance of atherosclerosis and osteoporosis.The analysis of the genetic polymorphisms was carried out with the Real Time PCR method and the statistical analysis was performed with the IBM SPSS 19.0 program.…

Anorexia nervosa (AN) is associated with distorted function of the hypothalamus- pituitary-gonadal axis, and thus adverse effects on skeletal integrity. We aimed to identify the combination of polymorphisms linked with deterioration of bone health in a sample of adolescent girls with AN. Patients and Methods: This cross-sectional study consisted of 40 young girls (12-21 years old), diagnosed with AN according to the American Psychiatry Association Criteria. Detailed medical history was recorded for all participants and blood samples were taken for hormonal evaluation and genotyping. Lumbar spine bone density (BMD) was evaluated using Dual energy X-ray absorptiometry. Results: Lower BMD values were observed in girls with: i) presence of the CTR-AluI polymorphism vs wild type (BMD, CC&CT vs TT genotypes: 0.84±0.21g/cm2vs 0.96±0.12g/cm2, p-value=0.028); ii) presence of the MTHFR677T polymorphism vs wild type (BMD, TT&CT vs CC genotypes: 0.86±0.23g/cm2vs 0.94±0.11g/cm2, p-value=0.047, adjusted for age, BMI, amenorrhoea). BMD measures exhibited a graded stepwise decrease with the addition of the CTR-AluI and/or MTHFR677 polymorphic variant (BMD, wild type vs one vs two polymorphic variants: 0.97±0.11g/cm2 vs 0.90±0.11g/cm2vs 0.75±0.33g/cm2, p-value for linear trend 0.011). Girls carrying the MTHFR677 polymorphism had 5.67- times higher odds of higher BMI (BMI >16.4kg/m2vs ≤16.4kg/m2, MTHFR677 polymorphism, CT&TT vs CC genotypes: OR=5.667, 95% CI: 1.254—25.606, p-value=0.024). Mean values of BMI Z-score differed between cases carrying the MTHFR A1298C polymorphism vs wild type (CA+CC vs AA genotypes: BMI Z-score: -2.51±1.39 vs 1.80±1.29, p-value=0.091). Multivariate regression analysis showed that AN-girls with higher BMI z-scores have significantly lower odds of carrying the MTHFR A1298C polymorphic variant compared to AN-girls with BMI z-scores within the lowest quartile (Q4 vs Q1, CA+CC genotype vs AA genotype: OR 0.047, p-value=0.032), adjusted for age, estrogen levels and duration of amenorrhoea. The prevalence of MTHFR C677T polymorphic variant or the combined prevalence of either MTHFR polymorphic variant (C677T or A1298C) did not differ between quartiles of BMI z-score adjusted for age. Conclusion: Combined presence of MTHFR677, CTR-AluI and MTHFR A1298C polymorphisms is associated with lower bone density in young girls with anorexia nervosa, implying a dose-response effect. The association between MTHFR677 and bone metabolism is likely mediatedby body weight.
