▼ Background: Over the last 20 years, cognitive and behavioural alterations in amyotrophic lateral sclerosis (ALS) have been recognized as an integral part of the disease. A proportion of patients present with a full-blown frontotemporal dementia syndrome, while selective cognitive changes are more commonly found, especially regarding frontal-executive abilities. Moreover, recent studies have highlighted a broader cognitive involvement in this population, concerning language and social cognition. Despite the increased awareness of ALS as a multisystem disorder and the importance of an accurate cognitive evaluation of these patients, the traditional neuropsychological ‘paper and pencil’ tools do not compensate for patients’ physical disability and can not be adequately used in the moderate-advanced stages of the disease. Objective: To investigate the use of P300-based Brain Computer Interface (BCI) and Eye Tracking (ET) technology for the administration of motor-verbal free cognitive measures in ALS. Materials and Methods: 34 patients diagnosed with ALS and 30 healthy subjects have been recruited. All participants underwent the BCI and ET-based neuropsychological assessment, together with three traditional cognitive screening tools (Frontal Assessment Battery - FAB; Montreal Cognitive Assessment – MoCA; Working Memory subtest of the Brief Assessment of Cognition in Schizophrenia), two psychological questionnaires (Beck Depression Inventory - BDI; State-Trate Anxiety Inventory - STAI-Y) and a usability questionnaire. For patients, also respiratory examination was performed, and the Frontal Behavioural Inventory - FBI was carried out with caregivers. Results: Significant correlations were observed between the traditional cognitive measures and the BCI- and ET-based neuropsychological assessment, mainly concerning accuracy and time-related variables in the ALS patients sample. Patients provided comparable rates than controls with regard to the BCI and ET usability. Conclusions: The developed motor-verbal free neuropsychological battery allows a longitudinal cognitive assessment during the course of the disease, also when traditional measures are not fully administrable, providing relevant information for clinical practice and ethical issues. ▼ According to official data released by Apple and Google, as well as market research agency Adjust and Localytics, there is an increasing number of dead apps just put away to the store. These apps are unattractive to users, so that users do not want to waste bandwidth and time to download, otherwise, delete or idle the app immediately. The reason is, apps yield explosion but users' attention is limited, so as hardware resources, such as screen, memory and storage. In this environment, software developers must continue to find any key breakthrough, or they will still create only zombie apps. I have been working in the industry due to software development for many years. In my experience, software companies or individual developers, always take the point of view of technical or functional modules to design software. This thinking model is exactly like manufacturing and processing. User experience is often overlooked in the process, not much effert for usersâ observation of their real needs and exploration. So that developers continue to create the software difficult to use, afterwards people spend more time to learn and adapt the software. In order to explore how to fully integrate the user experience design to software development process, this research will use IDEO's HCD Toolkit, implement Hear, Create and Deliver stages to understand user's needs and difficulties through rapid prototypes to carry out testing and evaluation. Technical development with program is finally the last stage. The whole process was focused on the exploration of user stories, insight into their needs, as well as fast and efficient prototype, frequent iterative testing, and appropriate references to Norman's design psychology theories, including Affordance and Mental Models. So the app will not be only in line with the needs of users, but also very intuitive to operate and understand. This research will develop an app for "Tainan 1616"âs shopping mechanism. The observation and interview ethnic is in the land of Tainan, like independent designers, farmers, fishermen, to help these people lack the resources or information, to have the opportunity to embrace new technology applications in the app marketing. So they can sell their own special products in app, to solve the existing financial difficulties. This research found that, many of the recommendations by HCD Toolkit for design Thinking, will let programmers no longer just think about technical details or costs, but go deep into the real needs of users. Through users' talk, empathy and observation of their behavior and expression, developers can re-design a variety of possible solutions. Before actually entering the technichal programming, they can has sufficient observation with multiple users, quick and efficient iterative testing, for a variety of design concepts. Område: Utvecklingen idag går allt snabbare vilket gör att företag behöver bli mer konkurrenskraftiga, ett sätt att bli det är genom innovation. Det kräver ett aktivt ställningstagande och ett sätt att genomföra detta är med hjälp av innovationsprocesser. Ett annat konkurrensverktyg som blir alltmer viktigt är design. Dessa två begrepp går att bryta ner i en rad faktorer och kan ses ur flera persektiv. Syfte: Syftet med denna uppsats är att förstå skillnader och likheter mellan innovations- och designprocesser samt se hur de kan komplettera varandra. Metod: Vi har använt oss av en kvalitativ metod och har genomfört intervjuer med tre olika företag. Dessa tre företag har sedan analyserats och jämförts med teori och med varandra. Teorier inom respektive område har även jämförts med varandra. Slutsatser: Vi har kommit fram till att innovations- och designprocesser kan ses som olika nivåer. Innovationsprocesser är mer övergripande kring hela organisationen och designprocesser är snarare mer detaljerad kring produktutvecklingen. Därför kan designprocesserna även sägas vara en del av innovationsprocessen, ett verktyg. Vi såg även att det fanns nio faktorer i praktiken som var viktiga för innovationsprocessen, nämligen en innovationsfrämjande organisationskultur, stödjande ledarskap, personalens delaktighet och engagemang, projektbaserad innovation, multidisciplinära team, bra kommunikation, kundinvolvering, omvärldsbevakning och utvärderingar/reflektion. Utifrån dessa slutsatser skapade vi en egen modell med inspiration från befintliga modeller.

Field: Because of the faster development today, companies need to become more competitive and one way is through innovation. It requires an active standpoint and one way to achieve this is through innovation processes. Another competitive tool that is becoming increasingly important is design. These two concepts can be broken down into a number of factors and can be viewed from several perspectives. Purpose: The purpose of this thesis is to understand similarities and differences between innovation and design processes, and to see how they can complement each other. Method: We have used a qualitative method and have conducted interviews with three different companies. These three companies have then been analyzed and compared with theory and with each other. Theories in each field have also been compared with each other. Conclusions: We have come to the conclusion that innovation and design processes can be viewed as different levels. Innovation processes are more general and about the entire organization and design processes are rather more detailed on product development. The design process can therefore also be said to be part of the innovation process, a tool. ▼ The needs of an increasingly young and active orthotic patient population has led to advancements in ankle foot orthosis (AFO) design and materials to enable higher function. The Intrepid Dynamic Exoskeletal Orthosis (IDEO) is a custom energy-storing carbon fiber AFO that has demonstrated improved clinical function, allowing patients to return to high-intensity activities such as sports and military service. An improved understanding of AFO mechanical function will aid prescription and fitting, as well as assist in design modifications for different patient populations. This study investigated the mechanical properties of AFOs, specifically structural stiffness, rotational motion, and strut deflection, to discern design characteristics contributing to increased functional outcomes. Seven AFOs of different designs and materials were tested under cyclical loading to characterize their mechanical properties. These AFOs were fitted about a surrogate limb and underwent pseudo-static compressive testing using a materials testing system and motion analysis. Acquired data included: compressive force, vertical displacement, kinematic data, and ankle rotation. Testing was conducted at discrete orientations and loads corresponding to the latter sub-phases of stance: midstance, terminal stance, and pre-swing. The compressive stiffness, posterior strut deflection, and rotational motion of the various AFOs, as well as the ankle range of motion (ROM) of the surrogate limb, were characterized. The deformation of the various AFO designs during loading differed greatly, influencing the observed mechanical behavior. Traditional thermoplastic and carbon fiber designs deformed at the malleolar flares or rotationally at the ankle, demonstrating low proximal rotational motion of the AFO and large surrogate ankle ROM. The mechanical response of the IDEO was unique, with large deflection observed along the posterior strut, minimal footplate deformation, greater proximal rotational motion, and minimal ankle ROM. This design incorporates stiffer materials for fabrication, increasing the potential for energy storage, while restricting ankle motion. Enhanced knowledge of the mechanical behavior and energy storage/release mechanism may improve prescription, custom design and fitting of the IDEO. The highly variable demand of fast fashion causes retailers in the industry to face large uncertainties when allocating initial inventory batches across multiple locations. Further into the selling season, inventory imbalances might arise as an effect of deviations from expected demand. To mitigate these imbalances, retailers can make use of lateral transshipments of inventory from locations with excess stock to those facing the risk of a stockout. Such transshipments require models for determining what and how much to ship as well as when to ship it. This thesis investigates how such a model can be applied on replenishment warehouse level in a fast-fashion retail setting. The research was conducted through a quantitative case study at Hennes & Mauritz (H&M), one of the largest fast-fashion retailers in the world. An appropriate existing transshipment model based on the concept of service level was identified and adjusted to suit the characteristics of H&M. In contrast to the vast majority of models in literature, empirical probability distributions were used for dynamically modelling short-term demand. The proposed model was evaluated and found to suggest transshipments yielding significant revenue increase driven by lowered price reductions. This thesis differs from existing literature by providing a unique case-study of how a transshipment model can be applied in practice and how it performs on empirical data from one of the largest fast-fashion retailers in the world.

Den kraftigt varierande efterfrågan på snabbt mode gör att aktörer i branschen står inför stora osäkerheter vid initiala lagerallokeringar. Längre in i försäljningscykeln kan lagerobalanser uppstå till följd av avvikelser från den förväntade efterfrågan. För att hantera obalanserna kan företag flytta lager från de regioner med lageröverskott till de som i närtid riskerar att få slut i lager. Sådana flyttar kräver modeller för att avgöra vad och hur mycket som ska flyttas samt när det ska flyttas. Detta examensarbete undersöker hur en sådan modell kan appliceras inom branschen för snabbt mode. Studien genomfördes som en kvantitativ fallstudie i samarbete med Hennes & Mauritz (H&M), en av världens största aktörer inom snabbmodebranschen. En lämplig existerande modell som baseras på konceptet om servicenivå identifierades och anpassades efter H&M:s egenskaper. Till skillnad från majoriteten av modeller i literature användes empiriska sannolikhetsfördelningar för att dynamiskt modellera kortsiktig efterfrågan. Den föreslagna modellen utvärderades och visades föreslå flyttar som genererar signifikanta intäktsökningar drivet av lägre prisnedsättningar. Denna studie skiljer sig från existerande litteratur genom sitt bidrag med en unik fallstudie av hur en modell för lagerflyttar kan appliceras i praktiken samt hur den presterar på empirisk data från en av världens största snabbmodeaktörer.

▼ The lateral line, an external sensory system composed of a network of hair cells arranged along the skin of fishes and aquatic amphibians, plays an important role in orientation and navigation in the aquatic environment. The current work examines lateral line functioning in tadpoles of the frog, Rana catesbeiana, and highlights the functional diversity of this system in different animal species. Recently hatched, early larval tadpoles were divided into an untreated group and a treated group immersed in gentamicin, an ototoxin that is expected to inactivate the lateral line hair cells. Animals from the gentamicin treated group were further divided into one of three post-treatment recovery groups: No Recovery (1 hour), 3-Day Recovery, and 7-Day Recovery. All animals were assessed behaviorally while exposed to the presence or absence of flow. Following behavioral assessment, the skin of a subset of animals from each treatment and recovery group were processed with two vital dyes to visualize neuromast structure and integrity. In addition, brains of some animals were processed for immunohistochemical labeling of apoptotic cells within lateral line target nuclei of the medulla. The current work demonstrates that bullfrog tadpoles respond to water flow in a manner that is less stereotyped than the typical responses seen in many species of fish. I also show that damage to the lateral line neuromasts disrupts flow-sensing behaviors, but that these behaviors do not fully recover in parallel with structural recovery of neuromasts, indicating a lasting behavioral effect of systemic hair cell death. Damage to tadpole lateral line hair cells disrupts normal orientation behavior at all but very high flow speeds. This is consistent with the structural and organizational changes within tadpole neuromasts seen as a result of gentamicin treatment. Levels of apoptosis within medullary target nuclei of the lateral line did not vary between different treatment and recovery groups. Apoptosis was consistently high in auditory nuclei, suggesting that these nuclei are more plastic during early development than are lateral line nuclei. These data have implications for our understanding of how important environmental signals are processed during a period of rapid developmental change. ▼ Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease of the upper and lower motor neurons resulting in weakness, paralysis and death within two to five years. Frontal temporal dementia (FTD) results in neurodegeneration of the frontal, temporal and insular cortices causing behavioral and cognitive deficits. ALS and FTD exist along a spectrum (ALS-FTD). Characterizing early neurodegenerative disease phenotypes is critical in defining epochs of time during which the nervous system is most plastic and receptive to intervention. Such early events reveal pathways that are deranged prior to the onset of neurodegeneration as well as biomarkers indicative of disease. This information offers insight that allows for earlier diagnosis and the identification of new therapeutic targets, providing an opportunity for early intervention. Because neurodegeneration is a late-stage phenomenon, many contributing pathologies are already present. This is particularly a problem in fast-progressing diseases including ALS and FTD. To identify early phenotypes in ALS and FTD, we used a novel deep behavioral phenotyping technology, Automated Continuous Behavioral Monitoring (ACBM), to identify and quantify the behavior of the transgenic SOD1G93A mouse (B6.Cg-Tg(SOD1*G93A)1Gur/J), as well as the TDP-43Q331K knock-in mouse, a novel model of ALS-FTD. ACBM collects video data of individually housed mice over five days. Position, velocity, acceleration and motion features are extracted from the data and classified through a Support Vector Machine Hidden Markov Model (SVMHMM) as one of nine pre-defined behaviors. We first demonstrated that SOD1G93A mice exhibit neuromotor deficits as early as P30 (Chapter 2). To determine whether such deficits are present in the TDP-43Q331K mouse, we conducted five rounds of ACBM and found TDP-43Q331K deficits as early as P120 (earliest age measured) (Chapter 3). Lastly, to assess earlier phenotypes, we conducted ACBM in a second younger cohort of TDP-43Q331K mice at P30 and P120, and found gender-dependent phenotypes at both ages (Chapter 4). This work presents the earliest behavioral phenotypes identified in the SOD1G93A and TDP-43Q331K mice. ACBM can be used in the future to assess treatment efficacy of pharmacological, genetic or environmental approaches in mouse models of neurodegenerative disease. ▼ Amyotrophic lateral sclerosis (ALS) is a group of fatal, late onset, neurodegenerative disorders with progressive loss of motor neurons, with no cure or effective treatment. Patients die within 2-3 years. 5-10% of ALS cases are inherited, with a large proportion (20%) of these due to a mutation in the SOD1 gene. hSOD1-G93A mice are currently the only genetic mouse model showing all the ALS histopathological markers . In adult muscles, TGF-β2 is expressed at the neuromuscular junction (NMJ), with its receptors on the overlying nerve terminal. TGF-β2 injections substantially increase the time SOD1-G93A mice can run on a rotarod. Conversely, at wildtype NMJs, TGF-β2 increases vesicular filling and thereby, synaptic strength. I investigated if TGF-β2 also increased transmission at hSOD1-G93A NMJs. I undertook the first postnatal timecourse of NMJ disease development to determine when transmission abnormalities appear. mEPP (spontaneous single vesicle releases) and EPP (evoked multiple vesicle releases) amplitudes are reduced in hSOD1-G93A mice at early symptomatic stages (18-22 wk). Impaired synaptic transmission during stimulation trains occurred even earlier (12 wk). These synaptic impairments correlate with redistribution of acetylcholine receptors (AChRs) and overt motor symptoms in vivo. TGF-β2 superfusion reduced and often reversed the deficit in amplitudes in mEPPs, single EPPs and EPP trains. Although TGF-β2 can cross the blood-brain barrier, it had no acute effect on spinal cord motor neurones. This study shows for the first time that diseased motor neurones have impaired synaptic function even before motor symptoms in vivo. Our data indicate that in diseased motor neurones, TGF-β2 increases neuromuscular signalling, prolonging function. Strengthening neuromuscular transmission without acute central deleterious effects is encouraging and supports the use of TGF-β2 as a therapeutic. Boosting transmission at impaired NMJs with TGF-β2 could allow ALS patients motor neurones to function more effectively for longer.

▼ Music and visuals have coexisted together in film for years, with millions of viewers and listeners attending film showings in a way of escaping their normal lives. This aim of escapism gives the viewer a chance to surround themselves in another world, apart from their own, and in doing so they can become immersed. To allow themselves the act of disbelief for a short period in order to find enjoyment and experience enhanced emotional involvement, where this could not be possible in day to day life. So what makes this combination of visuals and sound so appealing to the masses? What is it about the relationship between these two medias that culminates to create something truly captivating? When music and film connect in a perfect way it can make a masterpiece of audio-visual content, though there is a lot at play to make this happen. This dissertation delves into what it is that makes film so immersive to its audience, discovering multiple ways of composing music to film and how using similar formulas to those used in successful film, similar results can be yielded in terms of the ability to become immersed. However, an individual's personal opinion and experience can often be the deciding factor in that person becoming immersed. With this in mind this dissertation investigates the role of subjectivity in conjunction with immersion and film, exploring how subjectivity can affect how immersive a film can be to the viewer. The following films are analysed in detail: Birdman , dir. by Alejandro G. Iñárritu (Fox Searchlight Pictures, 2014), Fight Club , dir. by David Fincher (20th Century Fox, 1999), and Inception , dir. by Christopher Nolan (Warner Bros. Pictures, 2010). These three films have been chosen for analysis because they share similar narrative themes, that being the idea that the mind can influence the world, and in some cases create new worlds. These three films show their main characters interacting with parts of their mind, and in some cases being transported into their own minds.

▼ The submitted portfolio and accompanying thesis focus on the subject of doubles and duplication and their application as musical materials and compositional devices in my own work and, to a lesser extent, throughout the history of music and other artistic disciplines. In this study I look at the acoustic, structural, perceptual and aesthetic implications derived from the duplication of sonic materials, instruments, objects, setups, gestures and performers. From a general perspective, the use of duplication raises a number of issues of identical sounds by two or more sonic sources, the importance of synchronicity or asynchronicity in the suggestion of aural and visual analogies, the role of distance in the establishment of auditory correspondences, the degree of similarity required by specific materials to be identified as doubles, the role of the performer in creating a sense of gestural and performative duplication, the relationship between acoustic and acousmatic sources in the delineation of sonic parallels, etc. In this thesis, the subject of duplication is chiefly regarded from spatial, performative and sonic perspectives. The sections focusing on spatial issues examine the function of duplicated setups of objects and instruments in the space of performance. The analysis of performative elements addresses the role of gestural duplication and doppelgangers as musical and scenic resources. Finally, the sections on sound investigate the perceptual and aesthetic effects of duplicated sonic sources, echoic interactions and sonic displacements. This thesis is structured into two main parts. The first part (chapters 1 & 2) examines the different possible categories and models of duplication and their usage throughout music history. The second part (chapters 3 & 4) focuses on the application and different roles of duplication in my own music. The issue of duplication has been explored in the works composed throughout the research process. A total of six pieces (approximately 105 minutes of music) have been written over the course of this PhD. The issues present in these compositions have been thoroughly examined in my thesis, becoming the main object of study. Each of these works approaches duplication in a different manner, highlighting the versatility and potential compositional applications of this phenomenon.

▼ Diverging explanations of local multiparametric schemata are found in music of the common practice period (c. 1600–c. 1900). Associative statistical theories describe schemata as situated structures in particular times and places, whereas generative theories present these constructions as features formed through stability in universal and general rule systems. Associative-statistical theories of schemata elucidate the culturally conditioned relationships between features (distinctive attributes commonly used in grammars and schemata), but do not show the influence of universal psychological constraints; generative theories reveal the implicit structure of music, but do not formalise particular grammatical features and contexts. A synthesis of generative and associative-statistical approaches is necessary to model the interaction between universal and particular constraints of grammars and schemata. This dissertation focuses on a novel localised schema formed in the Classical instrumental grammar, termed the butterfly schema. It is posited that the butterfly schema is generated by a tendency for congruence that is manifest in and between the particular features of this grammar. Computational musicology and psychology provide interdisciplinary insight on the formal possibilities and limitations of grammatical structure. Computational models of schemata and grammars show how the congruent features of musical structure can be represented and formalised. However, they also highlight the difficulties found in the automatic analyses of multiparametric relationships, and may be limited on account of their inductive frameworks. Psychological approaches are important for establishing universal laws of cognition, but are limited in their potential to account for the diversity of musical structuring in grammars. The synthesis of associative-statistical and generative approaches in the present dissertation permits modelling the combination of the universal and particular attributes of butterfly schemata. Butterfly schemata are dependent on the particular grammars of periods of history, but are constrained by the tendency for congruence, which is proposed to be a cognitive universal. The features of the butterfly schema and the Classical instrumental grammar are examined and compared against the features of the Baroque and Romantic grammars, showing how they are formed from diverse types of congruent structuring. The butterfly schema is a congruent grammatical category of the Classical instrumental grammar that comprises: chords that are close to the tonic in pitch space (with a chiastic tension curve starting and ending on the tonic); a textural and metrical structure that is regular and forms a regular duple hierarchy at the level of regular functional harmonic change and at two immediately higher levels; and simple harmonic-rhythm ratios (1:1 and 3:1). ▼ This thesis examines the activity of the British musicians developing a practice of freely improvised music in the mid- to late-1960s, in conjunction with that of a group of British composers and performers contemporaneously exploring experimental possibilities within composed music; it investigates how these practices overlapped and interpenetrated for a period. The thesis identifies those characteristics of improvisation and experimentalism which favour a relationship between the two fields, but which ultimately underline the different expectations and objectives underlying each activity. The historical material is explored through a combination of archive research and interviews with musicians who were actively involved in the developments under examination. In addition the author draws upon his extensive personal experience as an improvising musician and composer, and as a performing associate of several of the key improvising musicians of the period. The first section of the thesis identifies the historical and social background, outlining the two key groups of participants working in the unmapped area between existing idiomatic improvisation and experimental composition practice, including brief studies of important figures who initiated or facilitated the exploration of shared activity during the period. A second section seeks to introduce further precision into discussion of improvised music by seeking to clarify the definition of taxonomic terms currently in use, and to extend these by identifying key characteristics of the wide range of approaches to playing improvised music. Section three explores the practical implications of the differing objectives of improvising musicians and composers. A series of archive case studies examining composing for improvising musicians during the 1960s and 1970s are discussed, along with an investigation of issues raised by the restoration of Derek Bailey’s Ping (prob. 1967/8) for contemporary performance by improvising musicians. The final section identifies fundamental differences of aspiration and approach within improvisation and composition, and examines the consequent implications for joint practice. It establishes why such differences are inevitable, and the insights they provide into the nature of artistic practice.
