The aim of this thesis is the experimental study and thermodynamic modeling of mixtures containing natural antioxidants, ionic liquids and organic solvents.Natural antioxidants are cinnamic acid derivatives, which constitute raw materials of high additive value for food, drugs and cosmetics industries. Ionic liquids, which are molten salts, are alternative solvents having the perspective of replacing the volatile organic ones used throughout the chemical industry.The experimental part of the thesis focuses on the determination of solvating capacity of various ionic liquids for two antioxidants; namely p-coumaric acid and caffeic acid and the comparison with the corresponding one of two classic organic solvents, t-pentanol and ethyl acetate, at the temperature range of (303 to 318) K. Experimental results revealed that ionic liquids based on BF4- and TFO- anions are better solvents than the organic ones, whereas those based on PF6- and TF2N- are worse. Moreover, the solution process is not spontaneous, is always endothermic and the entropy of solubilization is favorable for all systems studied. Melting temperatures, heats of fusion and temperatures of decomposition of the two acids were also determined and it was revealed that both acids decompose forthwith melt-ing.For the thermodynamic modeling of solid-liquid equilibrium of mixtures composed of natural antioxidants, ionic liquids and organic solvents the performance of three classic activity coefficient models (NRTL, UNIQUAC, UNIFAC) as well as that of the relatively new quantum chemical COSMO-RS model, which combines quantum chemical molecular simulations and statistical thermodynamics calculations for the prediction of various thermodynamic properties of pure compounds and mixtures, is studied.Successful correlation was obtained by both NRTL and UNIQUAC models. UNIFAC model has been applied for solubility prediction of the two cinnamic acids in the two organic solvents, since for the mixtures with ionic liquids it is not applicable but prediction results are relatively poor.COSMO-RS model, which is a unique a priori predictive method for thermodynamic properties that does not require any experimental data for its application, has been evaluated not only to the experimentally studied mixtures but also to binary and ternary mixtures of antioxidants, ionic liquids and organic solvents available in literature. From an overall assessment of the results of the COSMO-RS model of present thesis, it is concluded that although in some cases quantitative deviations from the experimental data are significant, the qualitative description in most cases is correct, indicating that COSMO-RS is a suitable model for a priori solvent screening.

テロメラーゼによるテロメアDNA伸長反応（テロメラーゼ反応）は細胞のがん化を導く。そこで本研究では、テロメラーゼ反応の阻害によるがん治療法及び、テロメラーゼ活性測定を介したがん診断法の開発を行った。１．がん治療法の開発：テロメアDNAにより形成されるG-quadruplex構造に結合するリガンドは、テロメラーゼ反応を阻害することが知られている。しかし従来のG-quadruplexリガンドの多くは、細胞内では染色体二本鎖DNAとも非特異的に結合するため、競合的にG-quadruplexへの結合が阻害される。さらに最近、細胞内の分子クラウディング（Molecular Crowding：MC）環境による水の活量低下も、G-quadruplexリガンドの結合を阻害することが報告された。そこでこれら阻害要因に対するG-quadruplexリガンドの官能基、中心金属、π平面の大きさの影響について系統的に解析した。その結果、アニオン性官能基を有するフタロシアニンは、二本鎖DNAとは静電的に反発するため、G-quadruplexへの特異性が高いことが見出された。さらにアニオン性フタロシアニンはG-quadruplexに結合する際に水分子を取り込まないため、水の活量低下によるG-quadruplexへの結合阻害が生じないことが示された。そのためアニオン性フタロシアニンは、大量の二本鎖DNA存在下及びMC条件下においてもテロメラーゼ反応を効率よく阻害することが示された。以上の結果は、アニオン性フタロシアニンが細胞核内においても効率よくテロメアDNAのG-quadruplexに結合し、テロメラーゼ反応を阻害することを示唆している。２．がん診断法の開発：従来のテロメラーゼ活性測定技術は、テロメラーゼ反応とPolymerase Chain Reaction（PCR）を組み合わせた手法である。そのため生体試料中のPCR阻害分子によって擬陰性結果が生じることがあった。そこで本研究では、テロメラーゼ反応後の反応産物を磁性ビーズ上に固定化、洗浄することで、PCR阻害分子を取り除く手法を開発した。さらにその後に行うPCRの産物をサイクリングプローブ法によって高感度に検出する方法を開発した。その結果、これらの手法を組み合わせることによってPCR阻害分子である胆汁酸、ビリルビン、ヘモグロビン存在下においても、阻害の影響を受けることなくがん細胞中のテロメラーゼ活性を測定できることが示された。また、検出感度50細胞という非常に高い感度でがん細胞を検出できることが示された。

"ABSTRACT- Development of Combinatorial Strategies Employing Neural Stem Cells and Polymer Scaffold for Spinal Cord Repair Stem cell-based therapy holds promise to enhance functional recovery following spinal cord injury (SCI). Most evidence that transplantation of neural stem cells can produce beneficial outcomes after SCI has been derived from rodent models. Before being translated to human patients, it would be needed to examine the therapeutic effects in larger animal models where weight-bearing locomotion should be much more challenging. The present study examined therapeutic effects of a combinatorial strategy centering on human neural stem cells (hNSCs) in a canine hemisection SCI model. After traumatic injuries to the spinal cord, secondary injuries aggravate the extent of damage, expanding cystic cavities at the lesion site. Cavity wall is surrounded by glial scar and severely impedes axonal regrowth. Therefore, any regenerative strategy could be complemented with an attempt to bridge lesion cavities. In the current study, we implanted poly (lactic-co-glycolic acid) (PLGA) scaffolds to facilitate the delivery of neural stem cells. We sought to enhance therapeutic efficacy by ectopic expression of neurotrophin-3 (NT-3) in the hNSCs. NT-3 overexpressing hNSCs (NT3.F3) were produced by retroviral transduction of the immortalized hNSC line (F3) with human NT-3 cDNA. NT3.F3 cells were seeded into a predesigned PLGA (65:35) scaffolds, and the PLGA scaffolds with hNSCs (PLGA-NT3.F3) were implanted immediately after left hemisection at T11 in female dogs weighing 20-30 kg. The PLGA scaffold seemed to nicely fill the lesion cavity, showing a varying degree of biodegradation by 12 weeks. Survival of grafted cells was confirmed at 2 weeks, and some of the grafted cells migrated to the host tissue. There were very few regenerating axons into the scaffold in both groups. Moreover, the ventral horns caudal to the hemisected region were more profusely innervated by serotonergic axons in animals with PLGA-NT3.F3. These findings raise a possibility that implantation of PLGA-NT3.F3 can produce beneficial motor outcomes by promoting axonal remodeling below the lesion. This study suggests that the therapeutic strategy combining multidisciplinary approaches can be feasible and effective for spinal cord repair in larger species. MSCs have been also known to pose neuroprotective effects through secreting various cytotrophic factors. Based on our previous studies demonstrating neuroprotective effect of hMSCs in Parkinson’s disease, we extended our investigations into other parkinsonian disease in addition to properties of hMSCs on blood-brain barrier. . PART A. Human mesenchymal stem cells exerts neuroprotection in an animal model of double lesion-induced multiple system atrophy-parkinsonism (MSA-P). PART B. Inhibition of blood brain barrier (BBB) permeability by hMSCs. First, the double lesion-induced MSA-P was established with co-injections of MPTP (10mg/day, total dose 90 mg/kg for 9 days) and 3- NP (total dose 450 mg/kg for 9 days, 12hr interval). At one day after last injection, hMSCs were injected into the tail vain (1X106cells/ml). Three groups of mice were compared (control group, only MPTP+3-NP group, hMSC treatment in MPTP+3-NP group) through histopathological and behavioral analysis. Compared to only MPTP+3-NP-treated mice, hMSCs treatment in double lesion mice significantly increased survival of TH- and NeuN-immunoreactive cells in the substantia nigra and the striatum, respectively. Additionally, hMSC treatment significantly decreased Iba-1, GFAP and increased Calbindine immunoreactive cells in the substantia nigra and the striatum. Behavioral analysis showed that the decent times on the top of a vertical wooden pole was significantly decreased in hMSCs-treated double lesion mice, comparable to controls. This study demonstrates that hMSCs treatment had a protective effect on loss of neurons in the substantia nigra and the striatum induced by MPTP+3-NP through a variety of mechanisms, such as anti-inflammatory actions, anti-apoptotic effect. Second, the neuroinflammation plays collectively suggest that excessive neutrophil infiltration and environmental factors, such as lower astrocyte density and higher BBB permeability, contribute to severe inflammation and neuronal death in the SNpc. At 4hour (4hr) after LPS injection, hMSCs were injected into the tail vain (1X106cells/ml) and three groups of rat were compared (control group, only LPS group, hMSC treatment in LPS group) through histopathological analysis after 12hr. Compared to only LPS-treated rats, hMSCs treatment in LPS-treated rats significantly showed increase of EBA-immunoreactive cells and reduction of Evans blue- immunoreactive cells in the substantia nigra. Interestingly, Compared to only LPS-treated rats, hMSCs treatment in LPS-treated rats significantly showed increase of the densities of astrocytes, assumed to significantly influence neurovascular structure and integrity and reduction of p-gp-immunoreactive cells, the one BBB transporter. "파울러자유아메바 (Naegleria fowleri)는 토양과 담수에서 자유생활을 하는 아메바로써 자연환경에서는 세균을 포식하며, 인체와 실험동물에서 원발성 아메바성 수막뇌염 (priamry amoebic meningoencephalitis)을 일으키는 것으로 알려져 있다. 숙주 세포로의 침입과 연관해서 파울러 자유아메바의 병원성 기전에서 표적 세포와의 접촉은 아주 중요하다. 본 연구실에서는 파울러 자유아메바에 대한 감염혈청 및 면역혈청을 생산하여 cDNA로부터 immunoscreenig을 통해 병원성 및 항원성과 관련이 있을 것으로 사료되며, 특히 위족(pseudopodia)에 특이성을 갖는 유전자 nfa1을 클로닝 해놓았다. 그리고 클로닝 된 nfa1유전자로부터 13.1 kDa의 재조합 단백질인 Nfa1을 얻었다. 본 연구의 목적은 위족 특이성을 갖는 nfa1 유전자가 파울러자유아메바의 병원성과 관련성이 있는지를 입증하기 위해서, 일차 배양한 쥐의 microglia와 함께 파울러자유아메바를 혼합 배양함으로써 아메바 영양형들의 병변효과 및 항Nfa1 항체의 투여효과를 광학, 주사, 및 투과 전자현미경을 이용해서 관찰하였다. 파울러자유아메바 영양형들은 활발한 위족과 food-cups 구조를 형성하여 microglia에 접촉하는 것을 확인하였다. 혼합 배양 3 시간 후, 소수의 microglia들은 자유아메바들에 의해서 파괴가 되었으며, 배양 시간이 길어짐에 따라 microglia들은 심하게 파괴되었다. 생화학적 방법인 ^(51)Cr 분비 실험결과, microglial 세포들에 대한 자유아메바의 세포독성은 배양 후 3, 6, 12 그리고 24 시간에 각각 17.8, 24.9, 54.6 그리고 98.0% 이었으며, 항 Nfa1 항체(1:100)를 투여하였을 경우, microglia들에 대한 세포독성이 각각 15.5, 20.3, 50.7 그리고 66.9%로 감소하였다. 한편, 파울러자유아메바와 혼합 배양한 microglia들은 방어적인 면역반응으로 TNF-α, IL-1β 그리고 IL-6등의 사이토카인(cytokine)들을 분비한다."

Naegleria fowleri, a free-living amoeba, causes fatal primary amoebic meningoencephalitis (PAME) in experimental animals and humans. On the pathogenic mechanisms of N. fowleri concerned with host tissue invasion, the adherence of amoeba to host cells is most important. The nfa1 gene (360 bp) was previously cloned from a cDNA library of pathogenic N. fowleri by immunoscreening, and produced a 13.1 kDa recombinant protein (rNfa1) that showed the pseudopodia specific localization by immunocytochemistry. On the basis of an idea that the pseudopodia-specific Nfa1 protein seems to be involved in the pathogenicity of N. fowleri, the cytopathic activity of N. fowleri trophozoites co-cultured with rat microglial cells was observed, and effects of an antiNfa1 antibody as treating on a co-culture system was elucidated. By a light, scanning and transmission electron microscope, N. fowleri trophozoites contacted with microglial cells produced vigorous pseudopodia and foodcups structure. Microglial cells were destroyed by N. fowleri trophozoites as showing the necrotic and apoptotic cell death in a timedependent manner. As the results of ^(51)Cr release assay, N. fowleri showed 17.8, 24.9, 54.6 and 98.0% cytotoxicity against microglial cells at 3, 6, 12 and 24 hr postincubation, respectively. On contrary, N. fowleri treated with an antiNfa1 antibody (1:100 dilution) showed 15.5, 20.3, 50.7 and 66.9% cytotoxicity, respectively. And then, microglial cells co-cultured with N. fowleri trophozoites secreted cytokines, TNF-α, IL-1β and IL-6, as the protective immune response.

Astrocytes can function as antigen-presenting cells (APC) by expressing class II major histocompatibility complex (MHC) antigens and co-stimulatory molecules upon activation. In vitro, astrocytes are activated by lipopolysaccharides (LPS), and interferon-gamma (IFN-gamma). We previously reported that gangliosides, membrane components of brain cells, especially neurons, can activate brain glial cells in vitro, thus could play roles in brain physiology and pathology. In this study, I tested whether gangliosides could induce the expression of immune molecules related to antigen presentation, using cultured rat brain glial cells, astrocytes and microglia. I treated cells with 50 ug/ml gangliosides mixture (Gmix) for 6 h to 3 d, and assayed the mRNA expression of class II transactivator (CIITA) and class II major histocompatibility (MHCII) molecules, using RT-PCR and real-time PCR. Gangliosides induce CIITA mRNA expression, followed by MHC II in astrocytes, but not in microglia. Critical for the development of mature APC capability is surface expression of class II MHC molecules and co-stimulatory molecules expression. Cell surface molecules of central importance to costimulation are CD40, ICAM-1, CD80 (B7-1) and CD86 (B7-2). In astrocytes, mRNA expressions of the above molecules were constitutive and that of CD86 was further induced by treatment of Gmix. Those expression patterns are the same in IFN-gamma-treated cells. Next, I tested the surface expression of MHC II by FACS and immunocytochemistry analysis. Gmix treatment failed to induce it in both FACS and immunocytochemistry analysis, in contrast to IFN-gamma treatment. This imply that Gmix could induce the mRNA expressions of CIITA and MHCII, but they could not induce surface expression of MHCII. To clarify the mechanisms, I assayed the molecules involved in it, including invariant chain (Ii) and the non-classical class II molecule RT1-DMs. Treatment of Gmix could not induce the mRNA expression of Ii, while IFN-gamma could. Still further studies are needed, I presumably suggest that Gmix are defective in fully presenting antigens due to a lack of expressing Ii molecules.

"신경교세포는 lipopolysaccharides(LPS)와 interferon-γ(IFN-γ) 자극에 의한 class II major histocompatibility complex (MHCII)와 동시 자극 물질(co-stimulatory molecules)의 활성화에 의해 항원 제시 세포(APC)로 작용할 수 있다. 최근 보고에 의하면 뇌세포의 막성분인 gangliosides(Gmix)에 신경교세포를 자극하여 활성화 시키나 이미 다른 자극제에 의해 활성화된 MHCII와 동시 자극 물질의 발현을 저해한다. 그러나 Gmix가 쥐 뇌의 신경교세포로부터 면역 관련 물질들을 발현시키는데 단독으로는 어떤 작용을 하는지에 대해서는 보고가 되어 있지 않다. 본 연구에서 Primary microglia와 astrocytes를 Gmix로 자극하면 CIITA와 MHCII는 astrocytes에서는 발현되나 microglia에서는 발현되지 않음을 RT-PCR로 확인하였으며, astrocytse에서 MHCII의 발현도는 Real-time PCR로 관찰하였다. 그러나 astrocytes에서 발현된 MHCII는 IFN-g와 LPS에 의해 발현된 MHCII와 달리 세포 표면으로 발현되지 않는 것을 FACS와 immunohistochemistry 실험에서 확인하였다. ▼ "Background: Buckwheat is one of the common food allergens in Korea. And it often causes severe and near fatal allergic reactions. At present allergen avoidance is the only therapeutic option for food allergies. Purpose: This study is designed to develop a murine model of IgE-mediated buckwheat hypersensitivity induced by intragastric sensitization with native buckwheat flour. During the procedure it was intended to determine not only the qualitative and quantitative differences of IgE production dependent upon amount of the sensitizing antigen, but also differences of cytokines produced by sensitizing antigen. At the same time, I evaluated a possibility of immunomodulatory effect of intranasal immunotherapy with buckwheat extract using established buckwheat allergy model in this study. Materials & Methods: Female C3H/HeJ mice(4～5 weeks old, 6～8 per group) were sensitized and challenged via intragastric route with various doses of fresh buckwheat flour(Group 1: 1 ㎎/dose, Group 2: 5 ㎎/dose, Group 3: 25 ㎎/dose of proteins) mixed with cholera toxin as adjuvant. Sham-sensitized(Group 4) mice were sensitized with cholera toxin alone. Naive(Group 5) mice were not sensitized with buckwheat flour or cholera toxin. Three weeks later, mice of Group 1, Group 2, Group 3, and Group 4 were then challenged via intragastric route three weeks later with fresh buckwheat flour extract containing cholera toxin. Anaphylactic reactions were evaluated 30 minutes after challenge, and then B- and T-cell responses to a crude extract of buckwheat flour were characterized by evaluating buckwheat-specific IgE, IgG₁, and IgG_(2a) antibody concentrations, by performing splenocyte proliferation assays and by assaying of various cytokines with antigen stimulation. In the second stage of experiment, all the mice were sensitized by introducing buckwheat antigen(1 ㎎/dose of proteins) via intragastric route on Day 1, 2, 3, and 7. From the sensitization day 14 on, different mice were administered intranasally buckwheat antigens(Group 6: 2 ㎍/dose/mouse, Group 7: 20 ㎍/dose/mouse), or PBS(Group 8) by the same route, three times per week for 2 weeks. After that I evaluated the concentrations of antigen specific IgE, IgG, the production of various cytokines and the level of splenocyte proliferation by stimulation in vitro with buckwheat allergen. Results: Intragastric buckwheat challenges of sensitized mice provoked anaphylactic reactions such as severe scratch, perioral/periorbital swelling, or decreased activity. Reactions were associated with elevated levels of buckwheat-specific IgE antibodies. Splenocytes from buckwheat allergic mice exhibited significantly greater proliferative responses to buckwheat than non-allergic mice. Increased production of IL-4, IL-5, and IFN-γ by splenocyte induced by buckwheat were positively correlated with elevated levels of buckwheat-specific IgE and symptom scores in sensitized mice. 本論文は、浜松に本社を置くヤマハ株式会社(以下ヤマハ)を事例に取上げ、世界の演奏家認められる楽器を作るためにアーティストと協働する活動「アーティスト・リレション(AR)」に焦点を当て、音楽家が求めるの表現(暗黙知)を如何にして楽器として表出(形式知化)していくのか、そのナレッジ・マネメント(SECIモデル)と楽器作りの関係を中心にヤマハのAR活動の実際を明らかにすること目指した。更に、世界中の音楽家との関係性を築くことで「情報交流人口」を拡大させ、楽器産業中心とした音楽文化創造都市浜松をアピールする楽器企業の存在が、Global Talent Magnet として機能おり、浜松における音楽文化の特色のひとつであるという仮説検証を試みた。　研究は、まず音楽文化創造都市を目指す浜松の現状をアンケート調査によって明らかし、さらに浜松おける創造的専門職に該当すると考えられヤマハ関係者への多面的インタビューを実施した。インタビュー内容の構造分析結果から、AR活動を通じた楽器作りのプロセスはSECIモデルそのもであり、浜松楽器産業競争優位源泉となっているこが分かったが、AR活動が「情報交流人口」拡大に資する Global Talent Magnet としての機能を果たしているかの確認は不十分であったと考える。　加えて、質的調査(インタビュー)を進める中で浜松の音楽文化の特徴として浮かび上がった、アマチュア音楽家の存在がARと共に楽器作りに貢献し、更に浜松においては楽器企業の存在と合わせて創造的人材を吸引する Talent Magnet 機能を果たしていると考えられことが判明した。

少子高齢化の進むわが国では、高齢者が地域社会の担い手になることが期待される。高齢者の社会参加を促すものとして芸術文化活動の重要性は高まっているが、いまだ個人の問題として扱われがちである。本研究では近年注目されつつあるシニア演劇を題材に、高齢者の芸術文化活動を地域社会との関わりの中で捉え直し、その社会的な意義を明らかにすることを目的とする。調査対象は大阪府箕面市の劇団「すずしろ」とする。同劇団は、設立に行政が関与した点と、プロ俳優が指導する点に特徴があり、ニューヨーク公演や全国シニア演劇大会の実施などの特筆すべき実績を持つ。調査方法は劇団および行政へのインタビュー調査である。　事例分析では中川幾郎の「自治体文化政策基本モデル」を参考に、「蓄積」「表現」「交流」の3つの視点から、劇団および行政の成果と課題を整理した。その結果、「蓄積」「表現」の側面では一定の成果を上げていることが分かった。具体的には、団員の意識が地域社会に向けられるようになったこと、公演を通じて他の高齢者に影響を与えていること、市の知名度向上に寄与していることなどが挙げられる。一方、「交流」の側面では課題もあった。劇団が地域に開かれた存在になるために、他世代や他団体との交流を促進する必要性が浮かび上がった。事例調査・分析から、地域社会においてシニア演劇が担う役割として5つの可能性を指摘した。(1)社会参加のきっかけをつくること (2)他の高齢者のロールモデルになること (3)高齢社会の問題を地域で共有する場になること (4)シティ・プロモーションの役割を果たすこと (5)演劇文化の振興に寄与することである。最後にシニア演劇の発展条件として、高齢者、演劇人、行政の果たすべき役割をそれぞれ指摘した。
