▼ There is increasing evidence to suggest that fetal events can predetermine reproductive health and general wellbeing in adulthood, a process termed 'fetal programming'. This refers to the association between altered fetal growth/development and health disorders in adulthood e.g. the metabolic syndrome, which is linked to low male testosterone levels. Studies from both Europe and the USA have shown that adult male testosterone levels have been declining, independent of age. As low testosterone levels in aging men are associated with increased morbidity and mortality, this highlights the importance of investigating how testosterone levels are determined or potentially ‘programmed’ during fetal development. Evidence from human and rodent studies have shown that reduced fetal androgen exposure results in lower adult testosterone levels, although the mechanism(s) is unknown, to date. One way to explain how a fetal insult (e.g. androgen deficiency) could affect (testosterone producing) adult Leydig cells in adulthood, is if their progenitor cells were present during fetal life and were thus affected by such an insult. This hypothesis has been unexplored to date, due to the lack of a unifying/defining marker for adult Leydig progenitor cells. An earlier study promoted the hypothesis for the studies in this thesis, namely that chicken ovalbumin upstream promoter transcription factor-II (COUP-TFII) might constitute such a marker, as inducible knockout of COUP-TFII in pre-pubertal male mice results in failure of adult Leydig cells to develop. Therefore, the hypothesis which was explored in this thesis was that 'fetal programming' of COUP-TFII+ adult Leydig progenitor cells prior to their differentiation into adult Leydig cells, would explain how fetal events could predetermine adult testosterone levels. To investigate whether adult Leydig cells (ALC) develop from COUP-TFII+ interstitial cells, firstly an adult Leydig cell ablation/regeneration model was used, which involved a single injection of ethane dimethane sulphonate (EDS). This identified that in rats, ALC derive from COUP-TFII+ interstitial cells which do not express any other phenotypical adult Leydig or interstitial cell markers prior to differentiation. Secondly, COUP-TFII+ adult Leydig progenitor cells are abundant in the fetal testis and conserved across species, including man. Thirdly, fetal interstitial cells which differentiated into ALC, as evident from an ALC lineage tracer model, also expressed COUP-TFII. Overall, these findings suggest that the COUP-TFII+ interstitial cells which differentiate into ALC are 'adult Leydig progenitor cells'. The findings from this thesis also show that the identified adult Leydig progenitor cells express the androgen receptor (AR) in fetal life. Furthermore, experimental reduction of androgen action in fetal life in transgenic mice (AR knockout) or chemical manipulations to reduce fetal testosterone levels (di(n-butyl) phthalate; DBP exposure) resulted in a similar reduction (~40%) in progenitor cell numbers from birth through…

▼ Large amounts of agricultural chemicals are discharged into the environment in Brazil. These pesticides are often used to destroy or impair the activity of organisms that attack crops. There are some reports in the literature concerning the decline of many wildlife populations as a result of contamination of their habitats by pesticides. One of the putative affected species is the fruit bat Artibeus lituratus, which are efficient seed dispersers along tropical and subtropical regions, where they occur, helping in the balance of ecosystems in these regions. Furthermore, they can provide valuable insights on the patterns of contamination in mammals in general, because they are highly mobile and occupy a privileged position on the food chain. Endosulfan is a widely used insecticide and acaricide, and acts by contact and ingestion. Despite its broad spectrum of action, endosulfan is well tolerated by a number of beneficial insects and has been proven effective against a wide variety of pests. Nonetheless, studies showing its effect on the physiology of bats are needed, especially on the reproductive biology of these animals. The aim of this study was to evaluate the toxicity of the insecticide endosulfan on testicular morphology and morphometry in fruit bats A. lituratus. Twenty-six A. lituratus adult male bats were captured from Atlantic Forest remnants in the region of Viçosa, State of Minas Gerais, Brazil, using mist nets and kept in cages in an Animal House at the Museum of Zoology, Department of Animal Biology of UFV. They were randomly divided into 4 groups: 1) control (GC): receiving papaya without endosulfan; 2) spreader (GES): receiving papaya exposed to adhesive spreader Wil Fix, in a solution with concentration 0.015 g / L; 3) endosulfan ( GE I): receiving papaya exposed to an endosulfan solution with a concentration of 1.05 g / L; and 4) endosulfan (GE II): receiving papaya exposed to an endosulfan solution with a concentration of 2.1 g / L. Treatment lasted 35 days, after that, animals were euthanized and the testes were collected, weighed and then processed for routine histology in glycol methacrylate. Testis morphometry and stereology of both tubular and intertubular compartments were performed using images obtained from histological slides. The parameters evaluated were: seminiferous tubules diameter and seminiferous epithelium height, the volumetric proportion of the testicular parenchyma components and intertubular compartment, the total length of seminiferous tubules and the gonadosomatic (GSI), leydigosomatic (ILS) and tubulossomatic (ITS) indices, plus, the
Leydig cell volume. All the measurements were performed using the software Image-Pro Plus 4.5. Data was analyzed using ANOVA and Student-Newman-Keuls test to compare means, level of significance was 5%. Resumo: A castração é um método empregado para o controle populacional de animais, assim como para promover modificações comportamentais e corporais. Pode ser realizada de forma cirúrgica, segundo a qual ocorre a remoção das gônadas, ou por agentes quimioesterelizantes capazes de reduzir total ou parcialmente a produção de espermatozoides. Ambos os procedimentos, principalmente os métodos cirúrgicos, possuem limitações quanto à sua utilização em animais sinantrópicos - que convivem com o homem de forma indesejada e são considerados pragas. A adição de quimioesterelizantes na dieta desses animais poderia eliminar limitações que
métodos convencionais de castração proporcionam. Apesar de os quimioesterelizantes serem os mais indicados para tal finalidade, podem ocasionar graves reações farmacológicas adversas. O etano dimetanosulfonato (EDS) é um éster alquilante, considerado um quimioesterelizante que atua diretamente sobre as Células de Leydig promovendo infertilidade transitória, quando administrado por via injeção intraperitoneal. Assim, o objetivo do presente trabalho foi avaliar a viabilidade e eficácia na promoção da infertilidade através da administração oral do EDS. Para tanto, ratos machos adultos Wistar receberam EDS na menor dose (grupo EDS-F1), dose intermediária (grupo EDSF2) e maior dose (grupo EDS-F3) incorporado à ração. Parâmetros reprodutivos foram avaliados 7 e 70 dias após o final do tratamento com a finalidade de determinar a magnitude dos danos causados pelo EDS, além de estabelecer a menor dose efetiva capaz de induzir a infertilidade. Nossos resultados mostraram que a exposição ao EDS
provocou uma diminuição significativa na viabilidade espermática, no comportamento sexual, alteração nos níveis de hormônios sexuais, no potencial de fertilidade e na performance reprodutiva, sem alterar a saúde dos animais. Esses resultados demonstraram que a administração oral (pela dieta sólida) de EDS provocou efeitos esterilizantes semelhantes aos observados após injeções intraperitoneais, além de serem reversíveis em longo prazo. Portanto, a disponibilização de um regime alimentar contendo EDS parece ser uma forma segura e prática de quimioesterilização de roedores sinantrópicos, visando seu controle populacional

▼ Leydig cells produce testosterone in response to luteinizing hormone (LH) via the cyclic adenosine monophosphate (cAMP)/protein kinase A pathway. Additionally, these cells are responsible for producing insulin-like peptide 3 (INSL3), a peptide hormone that is essential for testicular descent. The
insl3 promoter in
Leydig cells can be activated by cAMP through the transcription factor Nur77, which has also been shown to regulate the promoters of the steroidogenic enzymes,
cyp17 and
3b-hsd. While the mechanism of LH action on testosterone production is well characterized, the effect of LH on
insl3 abundance has yet to be shown directly. The MA-10
Leydig cells treated with hCG exhibited a transient and robust increase in
nur77 mRNA, while
insl3 mRNA abundance remained unchanged. Further, cAMP failed to affect
insl3 mRNA, though
nur77 mRNA abundance was significantly increased. Inhibition of LH-receptor-linked signal transduction pathways in the presence of hCG implicated multiple signaling networks in the regulation of both
insl3 and
nur77. Treatment with hCG or cAMP did not affect the abundance of
3b-hsd mRNA. Interestingly, though the MA-10
cell line has been reported to lack CYP17 activity and mRNA and so produce progesterone instead of testosterone,
cyp17 mRNA was present and inducible by hCG and cAMP. The addition of hCG, testosterone, nor the combination of hCG and testosterone affected
insl3 mRNA abundance. Though hCG consistently increased
nur77 mRNA abundance, the addition of testosterone did not enhance the effects of hCG. Collectively, these results indicate that
insl3 is regulated by factors other than LH/CG and cAMP in the MA-10
cell line.

▼ Phthalate esters (phthalates) such as dibutylphthalate (DBP) are commonly used as plasticisers and pesticides in a variety of products such as children‟s plastic toys, food packaging, cosmetics, medical equipment (including surgical equipment), and acaricides. Because of their widespread use phthalates are ubiquitous environmental contaminants that humans are commonly exposed to. Phthalates are known endocrine-disrupting chemicals
(EDCs) that are well known to cause male reproductive defects such as cryptorchidism (failed descent of the testes) and hypospadias (malformations in the urethra) in a range of different species if they are exposed in utero. They do this by reducing testosterone production in Leydig cells, which are the primary site of testosterone biosynthesis in the male. Because phthalates are dose-additive they are considered to share the same mechanism
of toxicity. However, the details of phthalates mechanism of toxicity are not fully understood. The aim of this research was to investigate the effects of DBP on the steroidogenesis pathway using the cultured rat Leydig cell cancer line R2C as a Leydig cell model. R2C cells were exposed to a range of DBP concentrations (10 μg/mL, 5 μg/mL, 1 μg/mL, and 0.1 μg/mL)
and their steroid hormone production was analysed using reverse phase HPLC. R2C cells did not synthesise testosterone at detectable levels. However, DBP exposure stimulated cortisol biosynthesis at all concentrations but caused no change in progesterone biosynthesis. This cortisol stimulation in Leydig cells has not been observed before. Because cortisol and testosterone compete for precursors an increase in cortisol synthesis could starve testosterone synthesis of precursors. On top of this it has been shown that glucocorticoids including cortisol have an adverse effect on Leydig cell development reducing steroid production and even causing apoptosis. This could explain how DBP and other phthalates can cause male developmental defects such as cryptorchidism and hypospadias.

▼ A toxicidade de um agente químico depende de sua propensão inerente de produzir danos ao sistema biológico com o qual interage. O arsênio (As), um conhecido poluente ambiental, merece destaque como elemento potencialmente tóxico para a função reprodutiva masculina. A exposição ao As pode afetar o testículo, resultando em efeitos adversos na reprodução, e várias evidências indicam que sua ação tóxica está relacionada ao estresse oxidativo. Com o objetivo de avaliar os efeitos do As sobre os testículos de camundongos adultos, os indicadores: biometria corporal, compartimentos tubular e intertubular e atividade enzimática antioxidante da catalase (CAT) e da dismutase do superóxido (SOD) foram analisados após exposição dos indivíduos a diferentes doses de As. Trinta camundongos suíços adultos foram divididos em três grupos: um grupo controle que recebeu água potável, e dois outros que receberam solução de arsênio nas concentrações de 0,05 (As1) e 1,0 (As2) mg/L, oralmente. O tratamento em ambas as concentrações de arsênio não interferiu de forma significativa na maioria dos parâmetros referentes à biometria corporal e à morfometria tubular. Observou-se aumento na massa dos rins dos animais do grupo As2, sugerindo possível toxicidade neste órgão e maior atividade do mesmo no processo de desintoxicação; houve diminuição da proporção volumétrica da túnica própria e epitélio seminífero e aumento da proporção do lúmen dos animais do grupo As2, indicando possíveis efeitos adversos na espermatogênese, contudo, alterações significativas nas medidas do diâmetro tubular e do epitélio seminífero que confirmassem tais efeitos não foram observadas. No compartimento intertubular, ambas as concentrações de As promoveram redução no volume e diâmetro nucleares das células de Leydig e aumento nas proporções de vasos linfáticos. No entanto, apenas a maior concentração (As2) causou diminuição do volume de células de Leydig no intertúbulo e de sua relação nucleoplasmática. Com relação às defesas antioxidantes enzimáticas, aumento da SOD e diminuição da CAT, sugere-se que há produção de H2O2 aumentada concomitantemente a uma diminuição de defesa antioxidante de CAT, ocasionando estresse oxidativo. Os dados apresentados indicam que ingestão subcrônica de As provoca alterações nas defesas enzimáticas do testículo e afeta sua porção endócrina sem, noentanto, promover modificações relevantes na histologia do compartimento tubular que indiquem mudanças na atividade espermatogênica.

▼ The utilization of plants with aphrodisiac action is a practice that has been increasing in the last years, a fact that awakes the interest of researchers from phytochemical, pharmacological and clinic investigations. Ouratea semiserrata is popularly used as astringent, anti- inflammatory and sexual stimulator. In this study, the goal was to evaluate the effects of aqueous extract from roots of O. semiserrata (AEROS) over the body and testicular biometry, seminiferous tubule and intertubule components of adult mice testes, also the extract being subjected to phytochemical prospection and the quantification of phenolics compounds. Twenty seven animals were used, divided into a control group (G1), which received distilled water, and two groups that received AEROS in the concentrations of 750 (G2) and 1500 (G3) mg/kg of body weight. The phytochemical prospection by thin layer chromatography revealed the presence of triterpenes/steroids, flavonoids, essential oils and tannins. The content of total polyphenols, proanthocyanidins, and flavonoids in the plant root were 3.57%, 2.07% e 0.11%, respectively. It is suggested that AEROS aphrodisiac potential is probably related to the presence of flavonoids and proanthocyanidins, compounds with vasodilatation activity. The treatment in both extract concentrations interfered in no significant way into body and testicular biometry and tubular morphometry, suggesting absence of alterations in the spermatogenic process and root toxicity. A reduction in volume proportion and connective tissue volume in both groups treated was present. The highest extract concentration promoted an increase in Leydig nuclear diameter and volume, and a decrease in macrophage proportion and blood vessels volume. Alterations in Leydig nuclear diameter and volume may be associated to increased levels of testosterone, probably, not enough to act over this testosterone-dependent reproductive organs weight.
A utilização de plantas com ação afrodisíaca é uma prática que tem aumentado nos últimos anos, o que desperta o interesse de pesquisadores em investigações fitoquímicas, farmacológicas e clínicas. Ouratea semiserrata é utilizada popularmente como adstringente, antiinflamatória e estimulante sexual. Neste trabalho, buscou-se avaliar os efeitos do extrato aquoso da raiz de O.semiserrata (EAROS) sobre a biometria corporal e testicular, túbulos seminíferos e elementos intertubulares dos testículos de camundongos adultos, sendo o extrato também submetido à prospecção fitoquímica e a quantificação de compostos fenólicos. Foram utilizados 27 animais, sendo um grupo controle (G1), que recebeu água destilada e dois que receberam o EAROS nas concentrações de 750 (G2) e 1500 (G3) mg/Kg de peso corporal. A prospecção fitoquímica por cromatografia em camada delgada revelou a presença de triterpenos/esteróides, flavonóides, óleos essenciais e taninos. ▼ Os roedores formam o grupo com maior diversidade de espécies na Classe Mammalia, abrigando indivíduos de poucos gramas como o camundongo, até espécies de 90 quilos como a capivara. A despeito de sua diversidade e do elevado número de representantes, poucos se sabe sobre a biologia reprodutiva da maioria dos animais deste grupo, o que dificulta a aplicação de medidas de conservação e manejo das mesmas. Por sua diversidade e pela facilidade de serem amostrados sistematicamente, os roedores silvestres tornaram-se modelos muito úteis para investigações. Assim, objetivou-se realizar estudos do processo espermatogênico de Oligoryzomys nigripes e Akodon cursor, ambos endêmicos da Mata Atlântica, visando obter dados básicos sobre a reprodução de roedores silvestres. Fragmentos testiculares foram processados para análise morfológica, imunohistoquímica e ultraestrutural. O índice gonadossomático encontrado para O. nigripes e A. cursor foi, respectivamente, de 0,68% e 0,74% enquanto o índice tubulossomático foi de 0,58% e 0,66%. Cada grama de testículo de O. nigripes e de A. cursor apresentou cerca de 23 e 27 metros, respectivamente. Em ambas as espécies, o ciclo do epitélio seminífero foi caracterizado em oito estádios de acordo com o método da morfologia tubular, e a duração de um ciclo foi de 3,93 dias em O. nigripes e 4,26 dias em A. cursor sendo necessários 17,68 e 19,15 dias para a formação de espermatozoides a partir de espermatogônias, respectivamente. A produção espermática diária por grama de testículo foi 148x106 células em O. nigripes e 142x106 células em A. cursor. Em O. nigripes e A. cursor, o padrão de organização dos elementos constituintes do intertúbulo apresentou células de Leydig isoladas ou agrupadas, geralmente associadas ao tecido conjuntivo e vasos sanguíneos e separadas dos túbulos seminíferos por um espaço linfático. Embora seja o elemento mais abundante do intertúbulo nestas espécies, o volume individual das células de Leydig foi considerado baixo, sendo de 804,60μm3 em O. nigripes e 858,7 μm3em A. cursor. O número destas células por grama de testículo foi, respectivamente, 25x106 e 38x106 células. O índice Leydigossomático (ILG), foi de 0,026% em O. nigripes e 0,044% em A. cursor. Pode-se concluir que as espécies aqui estudadas apresentaram baixo percentual de tecido intertubular e elevado percentual volumétrico de túbulo seminífero no parênquima testicular, o que juntamente com a curta duração do ciclo do epitélio seminífero, as coloca entre as espécies com produção espermática diária recorde entre os mamíferos. Tais caracteristicas podem ter estreita relação com o sistema de acasalamento destas espécies, indicando atividade poliândrica ou promiscua.

Large amounts of agricultural chemicals are discharged into the environment in Brazil. These pesticides are often used to destroy or impair the activity of organisms that attack crops. There are some reports in the literature concerning the decline of many wildlife populations as a result of contamination of their habitats by pesticides. One of the putative affected species is the fruit bat Artibeus lituratus, which are efficient seed dispersers along tropical and subtropical regions, where they occur, helping in the balance of ecosystems in these regions. Furthermore, they can provide valuable insights on the patterns of contamination in mammals in general, because they are highly mobile and occupy a privileged position on the food chain. Endosulfan is a
widely used insecticide and acaricide, and acts by contact and ingestion. Despite its broad spectrum of action, endosulfan is well tolerated by a number of beneficial insects and has been proven effective against a wide variety of pests. Nonetheless, studies showing its effect on the physiology of bats are needed, especially on the reproductive biology of these animals. The aim of this study was to evaluate the toxicity of the insecticide endosulfan on testicular morphology and morphometry in fruit bats A. lituratus. Twenty-six A. lituratus adult male bats were captured from Atlantic Forest remnants in the region of Viçosa, State of Minas Gerais, Brazil, using mist nets and kept in cages in an Animal House at the Museum of Zoology, Department of Animal Biology of UFV. They were randomly divided into 4 groups: 1) control (GC): receiving papaya without endosulfan; 2) spreader (GES): receiving papaya exposed to adhesive spreader Wil Fix, in a solution with concentration 0.015 g / L; 3)
endosulfan ( GE I): receiving papaya exposed to an endosulfan solution with a concentration of 1.05 g / L; and 4) endosulfan (GE II): receiving papaya exposed to an endosulfan solution with a concentration of 2.1 g / L. Treatment lasted 35 days, after that, animals were euthanized and the testes were collected, weighed and then processed for routine histology in glycol methacrylate. Testis morphometry and stereology of both tubular and intertubular compartments were performed using images obtained from histological slides. The parameters evaluated were: seminiferous tubules diameter and seminiferous epithelium height, the volumetric proportion of the testicular parenchyma components and intertubular compartment, the total length of seminiferous tubules and the gonadosomatic (GSI), leydigosomatic (ILS) and tubulossomatic (ITS) indices, plus, the Leydig cell volume. ▼ O morcego Sturnira lilium possui hábito alimentar predominantemente frugívoro, sendo um importante polinizador, sobretudo para espécies de plantas que dependem dos morcegos para a polinização e dispersão de suas sementes. Pela sua importância, torna-se fundamental que se conheça como esta espécie de morcego se reproduz, bem como se há influência de fatores ambientais sobre seu ciclo reprodutivo, uma vez que esta influência tem sido reportada para diversas espécies de morcegos. Por existir pouca informação disponível sobre quantificação e caracterização da espermatogênese de morcegos objetivou-se, neste estudo, fornecer informações sobre a atividade testicular de S. lilium ao longo das estações climáticas anuais. Assim, foi realizada a caracterização morfológica e morfométrica, com cálculo dos índices indicativos de produção espermática, caracterização dos estádios que compõem o ciclo do epitélio seminífero e quantificação de sua capacidade androgênica, nos testículos de indivíduos adultos. Foram capturados 14 animais nas estações seca (n=7) e chuvosa (n=7), no município de Viçosa, MG. Após eutanásia, fragmentos testiculares foram processados histologicamente para avaliação morfológica, morfométrica e histoquímica, em microscópio de luz e para análise ultraestrutural, em microscópio eletrônico de transmissão. Os animais apresentaram IGS médio de 0,27 % e seus testículos seguiram a organização geral descrita para mamíferos. A análise ultraestrutural permitiu o acompanhamento da formação do capuz acrossômico nas espermátides arredondadas, além da caracterização de outras células que compõem os compartimentos tubular e intertubular. Considerando-se indivíduos de ambas as estações, o parênquima testicular foi composto por cerca de 85 % de túbulos seminíferos e 15 % de intertúbulo, sendo o comprimento tubular por grama de testículo de aproximadamente 80 m. A contagem de células em apoptose não revelou diferenças significativas entre as estações. A maior reserva espermática testicular foi observada na estação chuvosa, com 43,47x106 células, bem como a produção espermática diária, cuja média foi de 12,60x106 células na mesma estação. A presença de espermatócitos primários em zigóteno no estádio 1 do ciclo do epitélio seminífero, segundo o método da morfologia tubular, difere de muitos mamíferos descritos até o momento. Os animais deste estudo apresentaram índice mitótico médio de 15,4 células, índice meiótico de 2,9 células, rendimento geral da espermatogênese de 68,7 células e capacidade suporte das células de Sertoli de 7,0 células. The bat Sturnira lilium has a dietary habit predominantly frugivorous, being an important pollinator, especially for plant species that depend on bats for pollination and dispersal of their seeds. Due to its importance, it becomes essential to know how this bat species reproduces, as well as if there is influence of environmental factors on their reproductive cycle, since this influence has been reported for several bat species. Once little information is available about the quantification and characterization of spermatogenesis in bats, this study aimed to provide information about testicular activity of S. lilium throughout the annual climatic seasons. To this end, we performed a
morphological and morphometric characterization, with calculation of indices indicative of sperm production, characterization of the stages that compose the seminiferous epithelium cycle, and quantification of its androgenic capacity, in the testes of adult individuals. Thus, 14 animals were captured in the dry (n = 7) and rainy seasons (n = 7), in Viçosa city-MG. After euthanasia, testicular fragments were processed histologically for morphological, morphometric and histochemical analysis by light microscopy and ultrastructural analysis under transmission electron microscopy. The animals showed GSI average of 0.27 %, and their testes followed the general organization described for mammals. The ultrastructural analysis allowed observing the formation of the acrosomal cap in round spermatids, besides the characterization of other cells that compose the tubular compartment. Considering both seasons, the testicular parenchyma was composed of about 85 % of seminiferous tubules and 15 %
of intertubule, and the tubular length per gram of testis had approximately 80 m. There was no difference in the count of apoptotic cells between the seasons. The highest spermatic reserve of the testes was observed during the rainy season, with 43.47x106 cells, as well as the daily sperm production, whose average was 12.60x106 cells at the same season. The presence of primary spermatocytes at zygotene in the stage 1 of the seminiferous epithelium cycle, according to the tubular morphology method, differs from many mammalian described so far. The animals of this study showed an average mitotic index of 15.4 cells, meiotic index of 2.9, general yield of spermatogenesis of 68.7 cells, and support capacity of the Sertoli cells of 7.0 cells. The duration of one seminiferous epithelium cycle was 3.45 days, and approximately 15.52 days were required for development of sperm from spermatogonia. Rodents constitute the largest group of species diversity in the class Mammalia, individuals harboring a few grams such as the mouse, by species 90 kg as the capybara. Despite their diversity and high number of representatives, few is known about the reproductive biology of most animals of this group, which hinders the implementation of strategies for conservation and management of the same. For its diversity and the ease of being sampled systematically, wild rodents have become very useful models for research. Thus, the objective was to conduct studies of the spermatogenesis of Oligoryzomys nigripes and Akodon cursor, both endemic to the Atlantic Forest, in order to obtain basic data on the
reproduction of wild rodents. Testicular fragments were processed for histological, immunohistochemical and ultrastructural. Gonadosomatic index found to O. nigripes and A. cursor was respectively 0.68% and 0.74%, while the index tubulossomatic was 0.58% and 0.66%. Each gram of testis of O. nigripes and A. cursor forward about 23 and 27 meters, respectively. In both species, the cycle of seminiferous epithelium were characterized in eight stages, according to the method of the tubular morphology and the cycle length was 3.93 days O. nigripes days and 4.26 A. cursor being required 17.68 and 19.15 days for the formation of spermatozoids from spermatogonia, respectively. The daily sperm production per gram of testis was 148x106 cells in O. nigripes and 142x106células in A. cursor. In O. nigripes and A. cursor, the pattern of organization of elements of the intertúbulo presented Leydig cells isolated or groupedgenerally associated with connective tissue and blood vessels and the
seminiferous tubules separated by a lymphatic space. Although it is the most abundant element in the intertúbulo these species, the volume of individual Leydig cells was considered low, being 804.60 μm3 in O. nigripes and 858.7 μm3 in A. cursor. The number of these cells per gram of testis were, respectively, 25x106e 38x106 cells. The index leydigosomatic (ILG), was 0.026% in O. nigripes and 0.044% in A. cursor. It can be concluded that the species studied here showed a low percentage of intertubular tissue, and high volumetric percentage of seminiferous tubules in the testicular parenchyma, which together with the short duration of the seminiferous epithelium cycle, puts them among the species with daily sperm production record among mammals. Such characteristics may be closely related to the mating system of these species, indicating activity polyandrous or promiscuous.

Os tumores ovarianos secretores de andrógenos (TOSA) e a hipertecose do estroma ovariano (HPT) são as etiologias mais frequentes de hiperandrogenismo nesta faixa etária. A diferenciação entre estas duas condições é difícil, pois as manifestações clínicas são semelhantes e caracterizadas por hirsutismo, alopecia androgênica, clitoromegalia, hipertrofia muscular e agravamento da voz. O perfil hormonal das mulheres pós-menopausadas com TOSA e HPT pode não ser um parâmetro ideal para discriminar estas duas condições. Além disso, os estudos de imagem podem não caracterizar com precisão estas lesões ovarianas. Devido às dificuldades, em estabelecer o diagnóstico diferencial entre TOSA e HPT, a ooforectomia bilateral é a terapêutica indicada para as mulheres menopausadas com diagnóstico de hiperandrogenismo de origem
ovariana; embora na HPT o tratamento clínico com análogo do hormônio liberador de gonadotrofinas (aGnRH) possa ser uma opção terapêutica eficaz. Objetivos: O nosso objetivo foi avaliar a contribuição das características clínicas, do perfil hormonal e dos exames radiológicos para o diagnóstico diferencial entre TOSA e HPT em mulheres na pós-menopausa. Métodos: Trinta e quatro mulheres pós-menopausadas, na faixa etária de 52 a 80 anos de idade, que foram encaminhadas à Unidade de Endocrinologia do Hospital das Clinicas da Faculdade de Medicina da Universidade de São Paulo, entre 1999 e 2013 por hiperandrogenismo clínico e com diagnóstico histológico de TOSA (13 mulheres) e HPT (21 mulheres) foram avaliadas retrospectivamente. Os diagnósticos histológicos foram revisados e confirmados por um único patologista com experiência em patologia ginecológica. Os dados clínicos de hiperandrogenismo, o perfil hormonal (T, E2, LH, FSH) e as imagens radiológicas pélvicas (Ultrassom transvaginal e
Ressonância Magnética, RM) foram obtidos a partir da revisão de prontuários médicos. Resultados: Em relação aos dados da história clínica, não houve diferença significativa entre os dois grupos de pacientes para nenhuma das variáveis clínicas analisadas, exceto para o número de gestações, que foi significantemente maior no grupo com TOSA. Os sinais clínicos de hiperandrogenismo, especialmente agravamento da voz (p < 0,001) e hipertrofia muscular (p = 0,01), foram mais prevalentes no grupo de pacientes com TOSA do que o grupo de HPT. Embora na análise dos parâmetros hormonais, os pacientes do grupo com TOSA tenham apresentado níveis mais elevados de T e E2 e níveis mais baixos de gonadotrofinas (p < 0,01 e p <0,01, respectivamente) do que o grupo de pacientes com HPT, uma grande sobreposição nos níveis hormonais foi observada entre os pacientes dos dois grupos. ▼ The accumulation of androstenone in the fat of mature boars results in boar taint; the conjugation of androstenone would decrease this important meat quality problem by decreasing the accumulation and increasing the excretion of androstenone.
Leydig cells and testis microsomes from mature boars were incubated with radiolabeled pregnenolone, and the free and conjugated metabolites were examined by HPLC. Sulfated androstenone with a mass of 367 m/z was directly identified by MS, with a novel tentative structure of 3-keto-4- sulfoxy-androstenone. Addition of enolase to the microsomal incubations increased the formation of 3-keto-4-sulfoxy-androstenone. Overexpression of SULT2A1 in HEK cells resulted in the sulfoconjugation of dehydroepiandrosterone, but not androstenone, suggesting that SULT2A1 may not be involved in sulfoconjugation of androstenone. This thesis describes the novel direct characterization of androstenone sulfate and the importance of enolase in its formation. The relevance to boar taint metabolism is discussed.

▼ Permeability glycoprotein (PGP) is a multidrug resistance 1 (MDR1) transporter and its over-expression protects cancer cells from chemotherapy and makes tumors drug-resistant. Therefore, it is important to understand the regulation of PGP expression in different tissues, including testes. Although PGP is expressed in different cells of the testis, the mechanism of regulation or if it is regulated in the different
cell types is not clear. Here we have treated adult male rats with ethane dimethane sulfonate (EDS), a
Leydig cell-specific toxicant, to deplete testosterone and investigate the role of testosterone in PGP expression in rat testes. For gene expression analysis, tissues were collected at 6 hr, 15 hr, 24 hr, 5 day and 7 day post-EDS treatment. Luteinizing hormone receptor (LHR) and insulin like peptide-3 (INSL3) are markers of
Leydig cells. Quantification of mRNAs for Lhr and Insl3 using real-time PCR was performed to analyze the rate of
Leydig cell loss post-EDS treatment. PGP expression was elevated significantly at 15 hr post-EDS and remained 10 fold above at 24 hr, and 7- to 8-fold higher at 5 and 7 day post-EDS. Although EDS treatment leads to testosterone depletion, testosterone replacement had a negligible effect on PGP expression at any of the time points evaluated. From this study, we conclude that PGP expression in rat testes is mostly likely not regulated by testosterone. Thus, PGP expression may be induced by selective chemical insults and these chemical insults may induce a constitutive expression of PGP on a prolonged basis after the primary chemical exposure.

clarify the rele of SCP2 in the regulation of steroid production
in rat Leydig cells.
The ra te of steroid production in the adrenal, the ovary,
the placenta and the testis is determined by the rate of
conversion of cholesterol to pregnenolone. The complex of
reactions and the localization of the enzymes involved in this so
called cholesterol side chain cleavage are well known. The P-
450scc enzyme complex catalyzing the production of pregnenolone
from cholesterol is located in the inner mitochondria! membrane
of steroidogenic cells (Fig.l.l). Several substances that regulate steroidogenesis, regulate the
rate of pregnenolone formation via an intracellular transducing
mechanism after binding to the plasma membrane of the cell.
Different "second messengers" are involved in the transduetion of
the signal from the plasma membrane to other processesjmessengers
in the cell which are involved in regulation of steroid production.
It is also known that protein synthesis and the transfer of
cholesterol to the inner mitochondria! membrane are important in
regulation of steroidogenesis. It is not known, however, if, and
in which way the induction of secend messenger systems in
steroidogenic cells is linked with protein synthesis, cholesterol
transfer and activatien of the cholesterol side chain cleavage
reaction inside the mitochondria. In this respect, sterol carrier
protein 2 (SCP2) is one of the possible proteins that could be
involved in transduetion of the signal from the secend messengers
to the cscc enzyme complex.

▼ Leydig-ove ćelije testisa su primarno mesto sinteze muških polnih hormona. Ovi hormoni su neophodani za reproduktivno, ali i za opšte zdravlje budući da su ozbiljni zdravstveni problemi često povezani sa njihovom smanjenom produkcijom. Insulin i insulinu sličan faktor rasta 1, IGF1
(engl. insulin like growth factor 1), i signalizacija koju pokreću preko svojih receptora (INSR i IGF1R), su jedan od ključnih faktora koji regulišu specifični razvoj tkiva, pa i samih gonada. Ipak, uloga i mehanizmi delovanja ovih receptora u steroidogenim tkivima nisu u potpunosti poznati. Stoga je istraživanje uokviru ove doktorske disertacije koncipirano sa ciljem da se, na modelu prepubertalnih (P21) i adultnih (P80) mužjaka miševa sa kondicionalnom delecijom
Insr i
Igf1r gena u steroidogenim ćelijama (Insr/Igf1r-DKO), definiše uloga INSR i IGF1R u regulisanju diferencijacije i steroidogene funkcije
Leydig-ovih ćelija. Pored toga, mužjaci i ženke P21 miševa sa istom delecijom su korišćeni za praćenje ekspresije glavnih markera mitohondrijalne biogeneze i fuzije/arhitekture u Leydigovim ćelijama, ovarijumima i nadbubrežnim žlezdama. Rezultati su potvrdili da delecija Insr i Igf1r u steroidogenim tkivima utiče na diferencijaciju i funkcionalne karakteristike
Leydig-ovih ćelija P21 i P80 miševa, upućujući na pojavu tzv. „feminizacije“. Broj
Leydig-ovih ćelija izolovanih iz P21 i P80 Insr/Igf1rDKO miševa bio je smanjen, a morfologija i ultrastruktura ovih ćelija izmenjene kod P21 Insr/Igf1rDKO miševa. ▼ The physiological role of multidrug resistance protein 4 (Mrp4) in the testes is unknown. It was discovered that Mrp4 is expressed primarily in mouse and human
Leydig cells; however, there is no current evidence that Mrp4 regulates testosterone biosynthesis. The role of Mrp4 was investigated in
Leydig cells where testosterone production is regulated by cAMP, an intracellular second messenger formed when the luteinizing hormone (LH) receptor (Lhr) is activated. As Mrp4 regulates cAMP, we compared testosterone levels in our Mrp4 WT and KO mice. Prepubertal KO mice had significantly reduced testicular testosterone, impaired gametogenesis, and disrupted cAMP homeostasis, resulting in decreased expression of genes involved in testosterone biosynthesis. Testosterone production was also impaired in adult KO mice but testicular morphology was normal. In vitro culture of primary KO
Leydig cells stimulated with LH revealed decreased intracellular cAMP concentrations and attenuated cAMP-response element binding protein (CREB) phosphorylation of downstream targets, notably several genes directly involved in testosterone biosynthesis. However, chemical inhibition of Mrp4 in WT
Leydig cells revealed a substantial elevation in intracellular cAMP concentration but paradoxically reduced testosterone production. The reduced testosterone production was related to decreased immunoreactive StAR expression, the rate limiting step in testosterone biosynthesis. Future analysis will focus on the mechanism controlling cAMP concentrations under these conditions, with a primary focus on cAMP regulation by phosphodiesterases (PDEs). Continued assessment of our KO animals revealed pre-pubertal animals had reduced systemic testosterone concentrations while adult mice had normal circulating levels. The difference is pre-pubertal KO mice have increased Cyp2b10, a known testosterone metabolizing enzyme, expression and catalytic activity due to disrupted testicular testosterone production. Therefore, defective testicular testosterone production deregulates hepatic Cyp-mediated testosterone metabolism to disrupt gametogenesis. The findings presented here have important implications for understanding the side effects of therapeutics that disrupt Mrp4 function and can alter androgen production.

▼ Namen magistrske naloge je bil raziskati učinke dolgotrajnega in vivo tretiranja starih samcev laboratorijskih podgan s sildenafil citratom (zaviralec fosfodiesteraze tipa 5, PDE5) na produkcijo testosterona. Raziskave so potekale v laboratoriju na Naravoslovni-matematični fakulteti Univerze v Novem Sadu od januarja do julija 2015. Zanimalo nas je, ali se s starostjo zmanjšuje koncentracija serumskega testosterona in kako šestmesečno tretiranje s sildenafil citratom vpliva na produkcijo androgenov, nitritov in ključnega proteina, ki sodeluje pri sintezi androgenov, tj. steroidogenega akutno regulatornega proteina (StAR), ter encima solubilna gvanilil ciklaza (sGC-ß1), ki sintetizira cGMP. Analizirali smo ultrastrukturo Leydigovih celic netretiranih in tretiranih osebkov. Po šestmesečnem tretiranju s sildenafil citratatom (1,25 mg/kg telesne mase), smo podgane žrtvovali in merili koncentracije preiskovanih spremenljivk v serumu, testikularni
intersticijski tekočini (TIF) in Leydigovih celicah. S transmisijsko elektronsko mikroskopijo smo pregledali ultratanke tkivne rezine. Rezultati so bili skladni z navedbami v literaturi. Koncentracija androgenov se je s starostjo na splošno zniževala, medtem ko so se pri tretiranih osebkih te vrednosti bistveno povečale. Koncentracija nitritov se je s starostjo osebkov zviševala, po tretiranju pa zniževala. Koncentracija StAR se je po tretiranju povečala, medtem ko je koncentracija sGC-ß1 upadla. Vidne so bile tudi spremembe v ultrastrukturi Leydigovih celic
v tretiranih osebkih je s starostjo plazmalema postajala vse bolj gladka, z manj izrastki in v citoplazmi je bilo manj endoplazmatskega retikuluma kot v citoplazmi mlajših osebkov. V vseh tretiranih samcih, mlajših in starejših, smo opazili povečane mitohondrije zaradi večje steroidogene aktivnosti celic. Plazmalema Leydigovih celic mlajših tretiranih osebkov je bila mestoma diferencirana v izrastke, ki niso bili tako izraziti
kot pri netretiranih osebkih iste starosti. Plazmalema celic starejših tretiranih osebkov je bila nediferencirana, tako da je bil rob celice videti gladek. V citoplazmi Leydigovih celic ostarelih tretiranih in netretiranih osebkov so bili prisotni elektronsko gosti lipidni vključki.

Resumo: O gerbilo da Mongólia (Meriones unguiculatus) tem sido utilizado de maneira crescente em estudos sobre o sistema genital masculino. Alguns aspectos da espermatogênese e o ciclo do epitélio seminífero dessa espécie são conhecidos, mas investigações sobre o desenvolvimento pós-natal do testículo e diferenciação das suas principais populações celulares são incipientes. Assim, o objetivo desse estudo foi avaliar os eventos envolvidos com a diferenciação das células germinativas e de Leydig durante o desenvolvimento pós-natal, estabelecer o período de maturação testicular, e descrever a dinâmica das populações de células de Leydig do
nascimento à senilidade. Foram utilizados gerbilos machos com 1-6 dias, 1-8 semanas, 3 e 18 meses de idade. Os diferentes tipos celulares foram identificados com base em microscopia de luz de alta resolução, microscopia eletrônica de transmissão e imunocitoquímica para marcadores específicos da linhagem germinativa (VASA) e das células de Leydig adultas (enzimas 3ß-hidroxiesteróide desidrogenase - 3ß-HSD e 11ß-hidroxiesteróide esteróide desidrogenase 1- 11ß-HSD1). Reações imunocitoquímicas para o receptor de andrógeno (AR), para o marcador de células em proliferação (PCNA) e técnica para marcação de células apoptóticas (TUNEL), bem como análises estereológicas dos componentes testiculares e determinação dos níveis séricos de testosterona e estrógeno também foram efetuadas. O processo de migração dos gonócitos para a membrana basal no gerbilo se estende até a segunda semana pós-natal, sendo seguido da sua rápida diferenciação em proespermatogonias. Diferentemente de outros roedores,
os eventos relativos à maturação dos gonócitos e sua diferenciação em células da linhagem espermatogonial é mais longo, ocorrem assincronicamente entre os cordões seminíferos e estão associados à perda de sensibilidade ao andrógeno. O desenvolvimento da população de células de Leydig adultas (CLA) envolve quatro estágios progressivos de maturação: as células de Leydig adultas progenitoras, as recém-formadas, as imaturas e as maduras, as quais surgiram, respectivamente com duas, quatro, cinco e seis semanas de idade. As células de Leydig adultas maduras exibem núcleo excêntrico e irregular e um canalículo citoplasmático perinuclear. Também apresentam heterogeneidade funcional em relação à expressão do AR e da enzima 11ß-HSD1. As mudanças que ocorrem no insterstício testicular do gerbilo durante o desenvolvimento pós-natal são muito similares às encontradas em outros roedores, no entanto, o número de células de Leydig fetais permanece constante até a senilidade. Eurycoma longifolia (Tongkat Ali; TA) is a Malaysian shrub used to treat various
illnesses including male infertility. Considering that TA is also used to improve
male fertility and no report regarding its safety has been published, this study
investigated the effects of a patented, aqueous TA extract on various sperm and
testicular functions. Materials and Methods: This study encompasses two parts (part 1: on spermatozoa; part 2: on TM3-
Leydig and TM4-Sertoli cells). Part 1: Semen samples of 27 patients and 13 fertile donors were divided into two groups, washed and swim-up prepared spermatozoa, and incubated with different concentrations of TA (1, 10, 20, 100, 2000 μg/ml) for 1 hour at 37°C. A sample without addition of TA served as control. After incubation with TA, the following parameters were evaluated: viability (Eosin-Nigrosin test), total and progressive motility (CASA), acrosome reaction (triple stain technique), sperm production of reactive oxygen species (ROS; dihydroethidium test; DHE), sperm DNA fragmentation (TUNEL assay) and mitochondrial membrane potential (Δψm) (Depsipher kit). Part 2: TM3-
Leydig and TM4-Sertoli cells incubated with different
concentrations of TA (0.4, 0.8, 1.6, 3.125, 6.25, 12.5, 25, 50 μg/ml) and control (without extract) for 48 and 96 hours. After incubation with TA, the following parameters were evaluated: viability (XTT),
cell proliferation (protein assay),
testosterone (testosterone ELISA test) and pyruvate (pyruvate assay). Results
Part 1: For washed spermatozoa, significant dose-dependent trends were found
for viability, total motility, acrosome reaction and sperm ROS production.
However, these trends were only significant if the highest concentrations were
included in the calculation. In the swim-up spermatozoa, ROS production of spermatozoa showed a biphasic relationship with its lowest percentage at 10 μg/ml, yet, no significance could be observed (P=0.9505). No influence of TA could be observed for sperm DNA fragmentation nor Δψm. Part 2: The viability rates and protein production of TM3-
Leydig and TM4-Sertoli cells at 48-hour exposure to TA showed increases whereas at 96-hour incubation periods viability and protein production declined especially as from concentration 25 μg/ml TA. Similar results could be seen for TM4-Sertoli cells pyruvate production. The testosterone production at 48-hour exposure
marginally increased (P=0.0580) at the highest (50 μg/ml) concentration of TA.
However, at 96-hour exposure to TA the testosterone production significantly
(P=0.0065) increased. It is also apparent that after 96 hours the concentration of
testosterone has increased [12 x 10-4 ng/ml] when compared to 48-hour exposure
[6 x 10-7ng/ml] of Tongkat Ali. Conclusion: Part 1: Results indicate that the Tongkat Ali extract has no deleterious effects on sperm functions at therapeutically used concentrations (<2.5 μg/ml). Part 2: The cytotoxic effect of TA are only presented at higher concentration from 25 μg/ml. TM3-

Traditional reproductive toxicity testing is expensive and time-consuming with extensive animal use. High-content analysis (HCA) has emerged as a powerful tool for chemical toxicity profiling and screening. This dissertation project aimed to use HCA
assays to examine testicular toxicities of BPA and its analogs, bisphenol S (BPS), bisphenol AF (BPAF), and tetrabromobisphenol A (TBBPA) in single testicular cell type cultures and a testicular cell co-culture model. HCA assays were developed and
validated in mouse spermatogonial cell line C18-4. HCA revealed that BPAF and TBBPA exhibited higher toxicities than BPA or BPS, including reduction of cell number, alteration in nuclear morphology, perturbation of cell cycle progression and
cytoskeleton, and induction of DNA damage responses. A machine learning (ML)-powered HCA pipeline was developed to determine phenotypic changes in a testicular cell co-culture model utilizing mouse spermatogonial, Leydig, and Sertoli cell lines. Exposure
to BPA and its analogs resulted in the loss of spatial arrangement of three-dimensional (3D) structure and M phase arrest in the co-cultures. BPAF induced multinucleated cells, which were associated with increased DNA damage responses and impaired actin

lines showed cell-specific toxicities of tested chemicals. BPA and BPS exerted higher toxicities in Sertoli and Leydig cells, as compared to spermatogonial cells. BPAF exhibited distinct toxicity profiles among three types of cells at different doses.
TBBPA showed low toxicity in Leydig cells. In conclusion, BPA and its analogs affect multiple cellular and molecular events and show differential toxicities in three types of testicular cells and co-cultures, with testicular toxicities being cell-type
specific. The newly developed HCA assays, together with the ML algorithm, can be used in identification, screening, and comparison of potential reproductive toxins.

Leydig cell tumor formation in genetically susceptible strains of mice. The hypothesis that the mechanism of estrogen induced carcinogenesis includes a gene amplification event was tested using this
in vivo endocrine carcinogenesis model. High molecular weight DNA isolated from liver, hyperplastic chryptorchid testes, primary
Leydig cell tumors and transplanted tumor lines progressing to estrogen nondependence for growth, was studied using Southern blot analysis and the in-gel renaturation technique to identify amplification of characterized and anonymous sequences. No consistent gene amplification event could be demonstrated either in carcinogenesis or tumor progression to an estrogen nondependent state. These results suggest that estrogen mediated carcinogenesis does not depend on a gene amplification event.

Leydig cell tumors regress when the trophic stimulus is withdrawn. The second objective of this work was to characterize the early molecular events associated with endocrine induced regression. Using Northern blot analysis of regressing
Leydig cell tumor RNA, it was shown that c-fos, c-myc, TGFbeta and p53 are induced during regression. In contrast, the expression of the TRPM-2 gene (a marker for programmed
cell death) was constitutive in both growing and regressing
Leydig cell tumors; DNA isolated from growing and regressing tumors was of high molecular weight and did not exhibit the nucleosome ladder pattern typical of apoptosis (programmed
cell death). These results indicate that entry into the regressing state (diapause) is an active process involving the expression of several well characterized proliferation regulatory genes, and that entry into diapause does not involve an increase in the rate of programmed
cell death.
Three novel genes induced during regression (rig) were identified using a subtractive cloning and hybridization approach, rigl detects a 2.1 kb transcript that is induced 2 fold at 36 hr after estrogen removal. rig2 detects a 500 bp transcript expressed in growing tumors but induced at 36 hr and reaching a 10 fold induction at 96 hr after estrogen withdrawal. rig3 detects a 125 bp transcript that is not detectable in growing tumor cells but is detected at 12 hr after estrogen removal and is progressively induced to greater than 15 fold at 96 hr after estrogen withdrawal. These results suggest that entry into diapause is an active process involving the activation of a unique pattern of gene expression.

The utilization of plants with aphrodisiac action is a practice that has been increasing in the last years, a fact that awakes the interest of researchers from phytochemical, pharmacological and clinic investigations. Ouratea semiserrata is popularly used as astringent, anti- inflammatory and sexual stimulator. In this study, the goal was to evaluate the effects of aqueous extract from roots of O. semiserrata (AEROS) over the body and testicular biometry, seminiferous tubule and intertubule components of adult mice testes, also the extract being subjected to phytochemical prospection and the quantification of phenolics compounds. Twenty seven animals were used, divided into a control group (G1),
which received distilled water, and two groups that received AEROS in the concentrations of 750 (G2) and 1500 (G3) mg/kg of body weight. The phytochemical prospection by thin layer chromatography revealed the presence of triterpenes/steroids, flavonoids, essential oils and tannins. The content of total polyphenols, proanthocyanidins, and flavonoids in the plant root were 3.57%, 2.07% e 0.11%, respectively. It is suggested that AEROS aphrodisiac potential is probably related to the presence of flavonoids and proanthocyanidins, compounds with vasodilatation activity. The treatment in both extract concentrations interfered in no significant way into body and testicular biometry and tubular morphometry, suggesting absence of alterations in the spermatogenic process and root toxicity. A reduction in volume proportion and connective tissue volume in both groups treated was present. The highest extract concentration promoted an increase in Leydig nuclear diameter and volume, and a decrease
in macrophage proportion and blood vessels volume. Alterations in Leydig nuclear diameter and volume may be associated to increased levels of testosterone, probably, not enough to act over this testosterone-dependent reproductive organs weight.

The toxicity of a chemical agent depends on its inherent propensity to produce damage to the biological system with which it interacts. Arsenic (As), a well known environmental pollutant, outstands as potentially toxic element to the male reproductive function. The exposure to As can affect the testicle, resulting in adverse effects on reproduction; several evidences indicate that its toxic action is related to oxidative stress. In order to evaluate the effects of As on the testes of adult mice, the indicators: body biometrics, tubular and intertubular compartments and antioxidant enzymatic activity of catalase (CAT) and superoxide dismutase (SOD) were analyzed after exposure of individuals to different doses of As. Thirty Swiss adult mice at reproductive
age were divided in three groups: a control group, which received drinking water, and two others, which received arsenic solution in the concentrations of 0.05 (As1) and 1.0 (As2) mg/L orally. The treatment with both arsenic concentrations did not significantly interfere in most parameters related to the body biometrics and tubular morphometry. There was increase in the mass of kidneys of animals who ingested the highest concentration of As, suggesting possible toxicity in this organ and his increased activity in the process of detoxification; decrease in the proportion of the tunica propria volume and seminiferous epithelium, and increase in the proportion of the lumen in the animals from group As2 were also observed, indicating possible adverse effects on the spermatogenesis, however, significant changes in the tubular diameter and seminiferous epithelium measurements that confirmed these effects were not observed. In the intertubular compartment, both concentrations of As promoted
a reduction in the volume and nuclear diameter of Leydig cells and increase in the proportions of lymphatic vessels. However, only the highest concentration (As2) caused decrease of volume of Leydig cell in the intertubular tissue and its relationship nucleoplasmatic. With respect to the enzymatic antioxidant defenses, increased SOD and decrease in the CAT were observed, suggesting a H2O2 production increase concomitantly with the reduction of the antioxidant defense of CAT, causing oxidative stress. The data indicate that subchronic ingestion of As can causes alterations inenzymatic defenses of the testis and affect its endocrine portion without, however, promoting relevant modifications in the tubular compartment histology which would indicate changes in the spermatogenic activity.

▼ Depuis plusieurs années, un nombre conséquent d’études décrivent une augmentation de l’incidence de pathologies liées à la fonction de reproduction masculine. Ces anomalies ont été regroupées sous le terme de « syndrome de dysgénésie testiculaire ». Ce syndrome aurait pour origine les effets délétères de polluants environnementaux sur le développement du testicule en période fœtale. Parmi ces polluants environnementaux, les phtalates et le bisphénol A (BPA) sont les plastifiants les plus produits et les plus répandus dans les objets de consommation courante. De nombreuses études leur sont consacrées et ont permis de les classer au rang de perturbateurs endocriniens en mettant notamment en cause leurs effets reprotoxiques. Mon travail de thèse est une étude des effets de ces deux perturbateurs endocriniens sur le développement du testicule fœtal.Nous avons réalisé une première étude concernant les effets du BPA sur le développement du testicule fœtal. Grâce au modèle de culture organotypique, nous avons développé notre étude dans trois espèces : le rat, la souris et l’Homme. Nous démontrons que le BPA diminue la sécrétion de testostérone dans le testicule fœtal humain à partir d’une concentration de 10-8M, alors que chez le rat et la souris, la sécrétion de testostérone n’est affectée qu’à partir de 10-5M de BPA. Nous avons également démontré une diminution de l’expression du gène de l’Insl-3, dans ces mêmes conditions. Ceci nous a permis de mettre en évidence une différence de sensibilité entre les espèces. Pour tenter de comprendre les mécanismes par lesquels le BPA exerce son effet toxique, nous avons comparé ses effets à ceux du DES, autre perturbateur endocrinien œstrogénomimétique. Contrairement au BPA, le DES diminue la sécrétion de testostérone fœtale chez les rongeurs, et non chez l’Homme. Ce résultat suggère l’implication de deux voies de signalisation différentes pour ces deux xéno-œstrogènes. Dans la continuité de plusieurs travaux réalisés dans notre laboratoire sur les effets du MEHP, nous avons tenté de comprendre les mécanismes par lesquels le MEHP induit un effet pro-apoptotique dans les cellules germinales mâles. ▼ Pituitary luteinizing hormone (LH) is required for the development of multiple cell types in both the male and female reproductive systems. In particular, LH promotes differentiation of testicular Leydig cells and stimulates steroid production by Leydig cells in the testis, theca, granulosa and luteal cells in the ovary. We have been interested in identifying nuclear transcription factors that are targets of LH signaling pathways in Leydig cells and concentrated on CCAAT/enhancer binding protein beta (C/EBPp). Our initial studies showed that C/EBPp is expressed in a differentiation specific pattern whose expression is stimulated by LH/human chorionic gonadotropin (hCG) and cyclic AMP (cAMP) in Leydig cells. We hypothesized that C/EBPp plays an important role in LH regulated Leydig cell development and steroidogenic function. To assess the specific roles of C/EBPp in Leydig cell function we have analyzed the steroidogenic capacity of Leydig cells from C/EBPp-deficient mice generated by gene targeting. This study revealed that testosterone production in male C/EBPp deficient mice is severally compromised suggesting that C/EBPp is essential for complete functional differentiation of Leydig cells. In order to understand how LH effects on Leydig cell differentiation and/or function may be mediated through C/EBPp, we attempted to identify genetic control elements that control C/EBPp transcription in steroidogenic and non-steroidogenic cells. We identified an evolutionarily conserved, steroidogenic cell-specific, distal enhancer element located in the C/EBPp 5'-flanking region. Our studies suggest that the activity of the enhancer may be, at least in part, controlled by as yet uncharacterized nuclear factors specifically detected in nuclear extracts of steroidogenic cells. These studies set the stage for elucidation of the molecular mechanisms controlling C/EBPp expression in steroidogenic cells in general, and may aid in uncovering alternative LH-dependent signaling pathways critical for functional maturation of Leydig cells

▼ Many environmental pollutants disrupt male reproductive function, inhibit steroid hormone biosynthesis, reduce serum testosterone levels, and decrease sperm counts. Although a disruption in steroidogenesis may underlie toxicant-induced reproductive toxicity, the mechanism by which these compounds inhibit steroid production remains unclear. The steroidogenic acute regulatory (StAR) protein mediates the rate-limiting and acutely-regulated step in steroidogenesis, the transfer of cholesterol from the outer to the inner mitochondrial membrane where die cytochrome P450 side chain cleavage (P450scc) enzyme initiates the synthesis of all steroid hormones. We hypothesized that environmental toxicants block steroidogenesis by disrupting StAR protein expression for several reasons: first, in contrast to the steroidogenic enzymes which have long half-lives and are chronically regulated, StAR protein is not an enzyme, is acutely regulated, its active form is highly labile and it must be continuously synthesized for steroid production to persist. Second, because StAR protein mediates the rate-limiting step in steroidogenesis, steroid production is very sensitive to disruptions in its expression. Finally, recent studies suggest that some environmental pollutants inhibit steroidogenesis by reducing cholesterol availability, implicating StAR protein as a potential target for these compounds.
Using mouse MA-10 Leydig tumor cells, we studied the effects of acute exposure (<4 h) to the organochlorine insecticide lindane (y-hexachlorocyclohexane; HCH), the organophosphate insecticide Dimethoate, the herbicide Roundup, and the imidazole antifungal drugs econazole and miconazole on: (1) steroid production, (2) total cellular protein synthesis, (3) StAR and steroidogenic enzyme activity and expression, and (4) protein kinase A (PKA) activity. Our studies demonstrate that these toxicants reversibly inhibited steroidogenesis without causing cell toxicity or affecting PKA activity. While lindane and Dimethoate reduced StAR protein expression by reducing StAR mRNA levels. Roundup, econazole and miconazole reduced StAR protein levels post-transcriptionally. Although Roundup and Dimethoate inhibited P450scc activity, a reduction in StAR protein expression alone could account for the effects of these compounds on steroidogenesis, suggesting that the inhibition of P450scc activity in these instances has little physiological significance, its activity being distal to StAR function. Our studies strongly suggest that several different classes of environmental pollutants inhibit steroidogenesis by reducing StAR protein expression, thus supporting our hypothesis.
