▼ The existence of non-baryonic cold dark matter is well
established through a wealth of complementary observations such as
the Cosmic Microwave Background, Supernova Type Ia and Big Bang
Nucleosynthesis. Despite the overwhelming cosmological evidence,
the particle nature of dark matter remains unknown. The leading
candidate is a Weakly Interacting Massive Particle (WIMP), a
thermal relic from the Big Bang that only interacts through the
gravitational and weak forces. A terrestrial detector with high
target mass, low background levels and a low energy threshold (keV)
should be able to record the rare, faint interactions between
ordinary matter and galactic WIMP dark matter. The Large
Underground Xenon (
LUX) experiment is a 370 kg liquid xenon time
projection chamber that aims to detect WIMP recoils with xenon
nuclei. The experiment is located a mile underground at the Sanford
Underground Research Facility (SURF) in Lead, South Dakota.
LUX is
poised to become the world's most sensitive dark matter detector
after only 2 weeks of exposure and will surpass current detection
limits by an order of magnitude after 300 days. The
LUX
experimental program consists of three phases: (1) the LUX0.1
prototype detector during 2007– 2009, (2) the commissioning and
operation of the
LUX detector at the Sanford surface laboratory
during 2009–2012 and (3) the underground deployment and operation
of the detector beginning in 2012. This thesis describes the first
two phases of the
LUX program.
LUX utilizes 122 Hamamatsu R8778
photomultiplier tubes (PMTs) to detect photons produced by particle
interactions in liquid xenon. The
LUX PMT mounting, high voltage
distribution, analog signal shaping and gain calibration system are
described in detail. The PMTs are shown to have low radioactive
background levels, single-photon sensitivity and a 30% photon
detection efficiency at the 175 nm liquid xenon scintillation
wavelength. Handling protocols and vacuum integrity control based
on ion-initiated afterpulses are presented. Xenon purification and
energy calibration results are described for LUX0.1. During the
LUX
surface run, light collection and statistical position resolution
measurements were obtained. Additionally, the xenon circulation
path was imaged with alpha particles from a Rn222
injection.

▼ We propose a new technique for the calibration of
nuclear recoils in large noble element dual-phase time projection
chambers (TPCs) used to search for WIMP dark matter in the local
galactic halo. This technique provides a measurement of the
low-energy nuclear recoil response of the target media using the
measured scattering angle between multiple neutron interactions
within the detector volume. Several strategies for improving this
calibration technique are discussed, including the creation of a
new type of quasi-monoenergetic 272 keV neutron source. We report
results from a time-of-flight-based measurement of the neutron
energy spectrum produced by an Adelphi Technology, Inc. DD108
neutron generator, confirming its suitability for the proposed
calibration. The Large Underground Xenon (
LUX) experiment is a
dual-phase liquid xenon TPC operating at the Sanford Underground
Research Facility in Lead, South Dakota. Our proposed calibration
technique for nuclear recoils in liquid xenon was performed in situ
in the
LUX detector using a collimated beam of mono-energetic 2.45
MeV neutrons produced by the DD108 fusion source. The nuclear
recoil energy from the first neutron scatter in the TPC was
reconstructed using the measured scattering angle defined by
two-site neutron events within the active xenon volume. We measured
the absolute charge (Qy) and light (Ly) yields at an average
electric field of 180 V/cm for nuclear recoil energies spanning 0.7
to 74 keV and 1.1 to 74 keV, respectively. This calibration of the
nuclear recoil signal yields will permit the further refinement of
liquid xenon nuclear recoil signal models and clearly demonstrates
measurable ionization and scintillation signals in this medium at
recoil energies down to O(1 keV). The low-energy reach and reduced
systematics of this calibration have particular significance for
the low-mass WIMP sensitivity of several leading dark matter
experiments.

The 5 ' and 3 ' ends were mapped and two new lux genes, luxG and luxH, were discovered upstream of the termination site. A transcription terminator that was able to function bidirectionally was located downstream of only one new lux gene, luxG, in the lux operon of Vibrio fischeri. Differences in transcriptional start sites were detected when the V. harveyi lux mRNA was mapped in vivo and in vitro. Studies using recombinant Escherichia coli containing the V. harveyi lux operon demonstrated that the promoter in E. coli mapped to the same site as found in vitro. The in vivo start site mapped to 26 base pairs (bp) and the in vitro start site mapped to 123 bp upstream of the initiation codon of the luxC gene. The expression of luxR in recombinant E. coli shifted the start site from position -123 to position -26, demonstrating that LuxR is required for transcriptional activation of the -26 lux promoter. ▼ Despite years of intense study of bioluminescence in Vibrio fischeri, much remains unanswered regarding its benefit to the cell and the environmental factors affecting its regulation. In this dissertation, I show that bioluminescence leads
to a growth rate reduction under some culture conditions, yet it contributes to this bacterium’s ability to colonize one of its symbiotic hosts, the squid Euprymna scolopes. Based on the reaction catalyzed by the light-producing enzyme luciferase, two
conflicting hypotheses have been suggested to explain the benefit that bioluminescence provides symbiotic V. fischeri. One proposal is that luminescence provides an advantage by consuming oxygen and thus serving as an antioxidant to protect the cell from
toxic oxygen radicals. Conversely, a second proposal suggests that by consuming excess reductant luminescence acts as an electron sink. I examined the regulation of luminescence to help distinguish between these two hypotheses, assuming that luminescence
would be maximally expressed when it is most beneficial. I examined luminescence of mutants lacking the redox-responsive and oxygen-sensitive regulators ArcA and FNR, respectively. This was performed in the well-studied and naturally visibly luminescent
strain MJ1 as well as in the relatively dim strain ES114, which is a natural symbiont of E. scolopes. ArcA is most active under reducing conditions, and I found that it represses luminescence in both strains due to a conserved ArcA binding site in the
PluxI promoter, which controls the expression of the proteins necessary for light production. Similar to ArcA, FNR is active under anaerobic conditions and represses luminescence in MJ1, although no such effect is apparent in ES114. These results
demonstrate that luminescence is repressed under reduced conditions and therefore my data is consistent with the antioxidant model explaining the benefits of luminescence, but are inconsistent with the electron sink model. This work is the first to use
defined mutants in V. fischeri to elucidate a mechanism by which specific environmental factors alter the regulation of luminescence.

▼ Bacteria use diffusible chemical signals to communicate within and between species and to coordinate certain group behaviors, including expression of host colonization factors. For Vibrio fischeri, a bioluminescent bacterium that colonizes
the Hawaiian bobtail squid Euprymna scolopes, bioluminescence is required to colonize the host fully, and it is controlled in part by the pheromone-dependent LuxR-LuxI regulatory system. While pheromone-dependent luminescence induction is influenced by
cell density, V. fischeri cells in culture are ~1000 times less luminescent and produce less pheromone than symbiotic cells, despite being at similar high cell densities. These observations and others suggest that density-independent factors also govern
LuxR-LuxI mediated pheromone signaling. In this dissertation I describe how certain environmental cues regulate the LuxR-LuxI regulatory system to control both pheromone production and bioluminescence in V. fischeri. I show evidence that the squid light
organ is a low iron environment and that low iron stimulates early induction and overall brighter luminescence in V. fischeri ES114 in culture. Moreover I demonstrate that inactivation of the redox-responsive ArcA/ArcB two-component regulatory system
derepresses pheromone production, initiating a positive feedback loop wherein pheromone can diffuse into surrounding cells to stimulate more pheromone synthesis and induce bright luminescence. I also describe how oxygen levels influence luminescence
through the oxygen-sensitive transcriptional regulator FNR, which regulates anaerobic respiration in V. fischeri in addition to bioluminescence. Finally, I show that citrate, a primary metabolite, regulates luminescence and this pathway requires the
Gac/Csr regulatory cascade, which is conserved throughout γ–proteobacteria and is a known regulator of both central carbon metabolism and host colonization factors. These findings show in greater detail how pheromone-mediated regulation in V. fischeri is
governed by more than cell density alone, and they suggest that pheromones can convey information about specific conditions, such as changes in redox or carbon flow, in addition to population size. This dissertation broadens our perspective of bacterial
pheromone-mediated communication and bacterial gene regulation in natural infections.

To próba ujęcia jego prozy w kontekstach antropologii kulturowo-wizualnej oraz propozycja wyznaczenia podstawowych struktur światów przedstawionych, geometrycznych wzorców modelowania przestrzeni. Istotną sprawą jest też określenie funkcji światła w powieściach, skategoryzowanego głównie na światło sztuczne/techniczne (lumen) oraz światło naturalne/światłość (
lux). Wstępnemu skalibrowaniu zagadnień związanych z przestrzenią, światłem i wizualnością służy rozdział rozdział Lumen,
lux, Lem. Od rozdziału Święte słupy analizie zostają poddane teksty Stanisława Lema z wszystkich etapów twórczości. Ich interpretacje i zestawienia umożliwiają dostrzeżenie powtarzających się wertykalno-koncentrycznych struktur, które przybierają rozmaite formy: statków kosmicznych, wieżowców, gmachów, stożków, bibliotek, kół, kuli, itp. LUX) Detector has recently finished a 332-day exposure and placed world-leading limits on the spin-independent WIMP-nucleon scattering cross section. In this work, we discuss the basic techniques to produce signal corrections, energy scale calibrations, and recoil band calibrations in a dark matter detector. We discuss a nonuniform electric field that was present during
LUX's 332-day exposure, and detail how such a field complicates these calibration techniques. Finally, we expand on the techniques presented earlier, such that they account for all of the complication introduced by the nonuniform electric field, and allow a limit to be produced from the data.

Integrations- und Segregationsprozesse in Grenzräumen. Absicht
dieser Arbeit ist es, der aktuellen Debatte über die europäische
Integration durch eine Analyse vergangener Integrations- und
Segregationsmechanismen die fehlende historische Grundlage zu
liefern. Diese Zielsetzung wird von einer essentiellen Überlegung
Georg Simmels geleitet: "Die Grenze ist nicht eine räumliche
Tatsache mit soziologischen Wirkungen, sondern eine soziologische
Tatsache, die sich räumlich formt." Eine Staatsgrenze wird somit
als ein erdachtes trennendes Konzept verstanden, als eine Idee, die
erst als konkrete Trennungslinie realisiert werden muss. Umgekehrt
kann eine "Grenze" also auch dort manifest werden, wo keine
Staatsgrenze existiert. Das Bestreben dieser Untersuchung ist es,
den Vorgang der Manifestation beziehungsweise Nichtmanifestation

▼ Biofilm formation by bacteria
provides them with a means to survive harsh environmental
conditions and promote colonization of a particular surface (biotic
or abiotic). Due to the resistance properties of biofilms,
biofilm-forming bacteria are difficult to treat in the context of
host infections. Thus, investigating the processes that control
biofilm formation will promote the development of novel
therapeutics. To study biofilm formation in the context of a host,
I utilized as a model system the marine bacterium Vibrio fischeri,
which must be competent to form a biofilm to colonize its squid
host Euprymna scolopes. Biofilm formation by V. fischeri requires
the syp polysaccharide locus, as well as the sensor kinase RscS and
the response regulators SypG, required for syp transcription, and
SypE, a negative regulator of biofilm formation. However, I
predicted that other factors were also involved in biofilm
formation. Therefore, my dissertation work focused on identifying
novel factors involved in this process. I first
performed random transposon mutagenesis on a biofilm-competent
strain of V. fischeri and screened for colonies that exhibited
defects in wrinkled colony formation, a biofilm phenotype. This
analysis permitted the identification of members of the
Lux
luminescence pathway (LuxQ and LuxU) as regulators of biofilm
formation. Overall, my data support a model in which the
Lux
pathway bifurcates at LuxU to regulate biofilm formation through
SypG. Next, I shifted my focus to better
understand SypG and its regulon. I asked whether this regulator was
the direct transcriptional activator of the syp locus. I found that
SypG recognizes and binds to a predicted enhancer sequence to
promote syp transcription. Additionally, I identified three new
putative members of the SypG regulon. The bam genes are involved in
biofilm maturation and, at least, BamA is a secreted product, while
the bal genes are involved in regulation of bioluminescence. Thus,
the work from my dissertation: 1) identified novel factors involved
in biofilm formation, 2) filled in critical gaps in our
understanding of how the syp locus is regulated, 3) identified the
first matrix protein in V. fischeri, and 4) provided new
information regarding the control of cellular
bioluminescence.

▼ Wind energy can be characterized as a cheap, clean, and renewable energy source that is absolutely
sustainable. With increasing demand for wind energy, it is productive to investigate the structural and operational
factors that undermine the proficiency and the characteristic performance of the wind turbine. Of
paramount importance to efficient wind energy generation is the aerodynamics of the wind turbine blades.
The aerodynamic factors, such as drag, airfoil pro files, and wake interactions that often reduce the performance of the wind turbines, can be investigated through computational mathematics using computational fluid dynamics (CFD). CFD offers basic techniques and tools for simulating physical processes and proffers
important insights into the
ow data, which are demanding and costly to measure experimentally.
In this thesis, we develop a simulation model in an open-source software package called OpenFOAM to investigate the performance characteristics of the
Lux Vertical Axis Wind Turbine (VAWT). The
Lux VAWT has a simpler design than its horizontal counterparts; however, its performance is affected by the unsteady
aerodynamic due to a complex flow field. The turbulent flow field is governed by the incompressible Navier-
Stokes equations. Simulations are carried out with an unsteady incompressible and dynamic
flow solver, PimpleDyMFoam, on an unstructured mesh surface of the
Lux VAWT geometry. The computational domain
includes both the stationary and rotating mesh domains to accommodate the rotating motion of the
turbine blades and the free-stream zone. The arbitrary mesh interface is applied as a boundary condition for the patches between the two domains to enable computation across disconnected but adjacent mesh domains.
Meshing was done using two separate meshing tools, snappyHexMesh and ANSYS Mesher. The
snappyHexMesh tool offered the most
flexible and effective control over the mesh generation and quality. In
order to derive the maximal power output from the
Lux VAWT simulations, the Unsteady Reynolds Averaged Navier – Stokes (URANS) equations are solved with different time-stepping methods; the objective is to reduce
the computational costs. While attempting to reduce the numerical diffusion from the non-transient terms
of URANS, a stabilized trapezoidal rule with a second-order backward differentiation formula (TR – BDF2) time-stepping method was implemented in OpenFOAM.
As a result, the transient aerodynamic forces of the blades, the torque, and power output are evaluated.
The findings demonstrate that most of the transient aerodynamic force is generated along the axis of rotation of the rotor during one complete revolution. Similarly, the computations indicate that the BDF2 method results in the least computational cost and predicts a turbine power that is somewhat comparable to the
experimental results. The difference between the simulation results and the experimental data is attributed partly to the pressure fluctuations on the turbine blades due to the mesh topology.

▼ The Gram-positive, soil dwelling bacteria of the genus Streptomyces produce greater than 50% of the clinically relevant antibiotics in use today. Thanks to the falling price of DNA sequencing, Streptomyces genomes are revealing that they encode more secondary metabolites (potential antibiotics) than they produce under standard laboratory conditions. By heterologously overexpressing the known pleiotropic regulators of antibiotic expression from Streptomyces coelicolor in several other Streptomyces species it has been shown that the secondary metabolite profile of these species can be influenced. While present-day methods of introducing genes (conjugation) and screening for antibiotics work well on a small scale, the low throughput nature of these protocols stand as a barrier to testing this hypothesis on a larger scale. The focus of the research presented here was to develop high throughput (HTP) methods of engineering and screening Streptomyces. With these two technologies in place, an attempt was to made to introduce three plasmids (pSET152-ermE*p-null, pSET152-ermE*p-atrA and pSET152-ermE*p-<em>lsr2NTD</em>) into 120 wild-isolate Streptomyces species from the Wright Actinomycete Collection. Exconjugants were successfully obtained for all three plasmids in 48 species of Streptomyces and were screened for increased antimicrobial activity using a HTP, lux-based bioassay. Numerous strains showed increased antimicrobial activity but WAC00206, WAC00230 and WAC00263 with pSET152-ermE*p-<em>lsr2NTD </em>showed the most promising improvement in antimicrobial activity. These hits have been designated as high priority for future investigation. These results suggest that HTP conjugation and the lux-based bioassay are powerful methods for introducing plasmids into and screening engineered streptomycetes.

L'augmentation des températures ambiantes mondiales pose un défi important aux espèces de plantes sauvages et cultivées. La plupart des espèces de plantes ajustent leur cycle de reproduction et leur développement pour optimiser leur survie et leur forme par temps ambiant élevé (Barnabás et al., 2008; Fitter et Fitter, 2002; Willis et al., 2008). Ces adaptations conduisent généralement à des hypocotyles allongés, à une réduction du nombre de feuilles au moment de la floraison, à une transition accélérée des phases de croissance végétative à reproductive, à une réduction du nombre de graines, à des gousses plus petites et à une diminution de la surface foliaire. Face au changement climatique rapide, en particulier à l'augmentation de la température permissive à la croissance ambiante, il est urgent de régler la thermoréponse des usines pour adapter les installations aux changements climatiques et garantir la production alimentaire future.L'expression de PIF4 est contrôlée par le complexe du soir (EC), un complexe à 3 protéines comprenant ELF3, ELF4 et
LUX, en fonction de la température. La CE est capable de réprimer l'expression de PIF4 en se liant à des motifs spécifiques sur le promoteur de PIF4 à une température de croissance ambiante inférieure. Cependant, cette répression de l'expression de PIF4 est éliminée à une température ambiante de croissance plus élevée, ce qui entraîne un vieillissement prématuré des plantes.RésultatsLes trois protéines de la CE ont été produites en utilisant une stratégie d’expression différente, ELF4 et
LUX dans E. coli, et ELF3 dans des cellules d’insecte. Les méthodes de purification des protéines et les tampons ont été optimisés pour obtenir des ELF3 et

LUX étant la seule protéine de la CE à posséder un domaine de liaison à l'ADN connu, elle a été utilisée pour effectuer des tests de déplacement de la mobilité afin de comprendre l'affinité de liaison de
LUX pour ses motifs ADN cibles obtenus à partir de tests de puces de liaison de protéines. D'après les tests de décalage de mobilité, il a été observé que le domaine de liaison à l'ADN (DBD) seul pouvait se lier aux motifs d'ADN de son substrat à des concentrations molaires nano alors qu'une quantité de protéine excédentaire de 10 fois était nécessaire pour que la longueur totale de

▼ The Large Underground Xenon (
LUX) experiment has recently placed the most stringent limit for the spin-independent WIMP-nucleon scattering cross-section. The WIMP search limit was aided by an internal tritium source resulting in an unprecedented calibration and understanding of the electronic recoil background. Here we discuss corrections to the signals in
LUX, the energy scale calibration and present the methodology for extracting fundamental properties of electron recoils in liquid xenon. The tritium calibration is used to measure the ionization and scintillation yield of xenon down to 1 keV, the results is compared to other experiments. Recombination probability and its fluctuation is measured from 1 to 1000 keV, using betas from tritium and Compton scatters from an external 137-Cs source. Finally, the tritium source is described and the most recent results for ER discrimination in
LUX is presented.

▼ Invasive species are an environmental problem
throughout the world. In the midwestern United States, invasion by
Amur honeysuckle (Lonicera maackii) has been shown to reduce the
growth, reproduction, and diversity of native plants. An
interesting aspect of this species is its extended leaf phenology.
Amur honeysuckle leafs out earlier in the spring than most native
shrubs and understory trees and loses its leaves later in the fall.
Few studies, however, have determined whether extended leaf
phenology affects species richness by shading. To assess potential
impacts of extended leaf phenology, I first tested whether there
were differences in light intensity and temperature under Amur
honeysuckle compared to natural understory. I found that both light
levels and temperature were significantly lower under Amur
honeysuckle compared to natural understory, especially in early
spring. Based on these findings, I experimentally examined how
comparable levels of shading in the spring and over the entire
growing season affected the abundance and community patterns of
forest floor species. Shading reduced species diversity, but there
was no difference between the spring treatment and the full season
treatment. Overall the results indicate that extended leaf
phenology, particularly early leaf out in the spring, reduces light
and temperature levels which reduces the diversity of native
understory plants.

▼ El presente trabajo muestra el diseño de un plan de negocios para una nueva unidad estratégica
de negocios dentro de la empresa Lux Ltda., compañía existente desde 1943, y desde 1978
dirigida por los actuales dueños, actualmente tiene una facturación promedio anual de MM
US$21, y ve la necesidad de expandir sus líneas de productos de Iluminación para lograr
mantener esos niveles de venta.
Durante los últimos 40 años, Lux Ltda. se ha especializado como proveedor de empresas de retail
y distribuidores especialistas eléctricos, muchos de los cuales hoy en día están prefiriendo realizar
importaciones directas bajo sus propias marcas.
Por otro lado, se ha detectado un fuerte aumento de grandes proyectos industriales en Chile,
como el proyecto MAPA (Modernización y ampliación de la planta Arauco) de Celulosa Arauco,
mayor inversión forestal en Chile (MM US$2.350), el mega puerto que se construirá en San
Antonio, con una inversión total de MM US$3.380, entre algunos megaproyectos que se
desarrollaran en Chile durante los próximos años. Sumado a lo anterior, se aprecia una baja
cantidad de proveedores especialistas en iluminación heavy duty, hoy solo hay 5 proveedores
relevantes en el mercado.
Es por lo anterior que nace la oportunidad de negocio de crear una nueva unidad estratégica de
negocios, centrada en atender el mercado industrial heavy duty, donde hoy en día no está Lux
Ltda.
El mercado de iluminación heavy duty se estima en MM US$ 7,4 por año, por lo que se tiene
como objetivo para el primer año un 18% del actual mercado, lo que corresponde a MM US$1,3
e ir aumentando este valor cada año.
Financieramente el plan de negocio se evalúa con una inversión inicial el primer año de MM $361,
estimando flujos de cajas a 5 años, obteniendo un valor actual neto de MM $212, con una tasa
interna de retorno de 30,3% y un payback de 3,46 años.

Este estudo resgata as informações e a situação em que essa coletânea de textos foi criada, numa tentativa de remontar seu contexto histórico, momento literário e mercado editorial, buscando hipóteses que auxiliem no entendimento dos aparatos que a constituem, num esforço de aproximação do escasso material de divulgação localizado sobre obra, editora e organizador pesquisado nos periódicos da época e constante dos anexos , numa reflexão sobre os laços de interesse existentes entre o Brasil e a União Soviética.
The Anthology of Russian Short Stories, edited by Otto Maria Carpeaux and published in the early 1960s by Editôra Lux Ltda, marks the first time Russian short stories were published from direct translation of the original language to Portuguese. This study retrieved information about the Brazilian historical and literary context as well as its publishing market during the time the work was created in an effort to promote a better understanding of how it was influenced by them. The presented information about the work, publisher and editor )included in the attachments( was searched in the sparse number of publishing advertisements in the newspapers and magazines that circulated during that time. It allows a better understanding of the works content and promotes a deeper reflection on the common interests and relationship between Brazil and the USSR.

▼ Bonaventure et la lumière : la question semble au premier abord et pour la plupart des médiévistes résolue. Pourtant, la genèse de sa définition de la lumière (II Sent., d. XIII) – notamment la manière dont les idées de Robert Grosseteste lui parviennent – reste encore à déterminer. L’idée d’un usage métaphorique et analogique de la lumière et de sa dimension épistémique permet de mieux évaluer la théorie de la connaissance comme lumière, c’est-à-dire son amplitude réelle souvent réduite à l’illumination divine de l’homme tant intellectuelle que morale. Cette connaissance de la lumière permettant de considérer la connaissance comme lumière par le truchement de la métaphore et de l’analogie donc toute une épistémologie par la lumière se vérifie dans la cosmologie et dans la théorie de la beauté du frère mineur où la lumière joue bien un rôle principiel et paradigmatique. Enfin, que toutes ces implications philosophiques et théologiques de la lumière soient récapitulées dans le Christ compris selon les Écritures, comme splendor, sol iustitiae, lux mundi, compréhension nettement inspirée de la lecture du quatrième évangile et du Livre de la Sagesse par Bonaventure, bachelier biblique, demande encore à être élucidé. C’est en cherchant tant du côté des études à la Faculté des arts de Paris de 1235-1243 que du côté des écrits de Bonaventure, bachelier biblique puis sententiaire, que la question de la lumière dans son œuvre peut être interprétée. Nous proposons donc dans cet ouvrage d’ouvrir quelques pistes de compréhension de la via lucis bonaventurienne.
Bonaventure and light: the issue seems at first and for most medievalists resolved.Yet the establishment of the genesis of his definition of light (II Sent., d. XIII) — and in particular the way in which Robert Grosseteste's ideas reach him — is still to be determined. The idea of a metaphorical and analogical use of light and its epistemic dimension makes it possible to evaluate better the theory of knowledge as light, that is, its actual amplitude, often reduced to the divine enlightenment of Man, both intellectual and moral. This knowledge of light, which makes it possible to consider knowledge as light through metaphor and analogy, and thus a whole epistemology by light, is verified in the cosmology and in the theory of beauty of the Friar Minor in which light plays indeed a principle-like and paradigmatic role. Finally, that all these philosophical and theological implications of light are recapitulated in Christ understood according to the Scriptures as splendor, sol iustitiae, lux mundi, an understanding clearly inspired by the reading of the fourth Gospel and the Book of Wisdom by Bonaventure, bachelor of the Bible, still needs to be elucidated. It is by seeking as much on the side of studies at the Faculty of Arts of Paris from 1235 to 1243 that on the writings of Bonaventure, as baccalaureus biblicus and then baccalaureus sententiarus, that the question of light in his work can be interpreted. ▼ Das Ziel der vorliegenden Arbeit ist es, herauszufinden, ob die Steueraffären Lux Leaks, Panama Papers und Paradise Papers Anstöße für eine Europäisierung der Steuerpolitik im Bereich der Unternehmensbesteuerung gegeben haben. Dazu werden die Aktivitäten der EU-Institutionen für jeweils einen Zeitraum von vier Jahren vor und nach der Veröffentlichung von Lux Leaks Ende 2014 verglichen. Insbesondere kann eine deutlich höhere Anzahl an Gesetzesvorlagen der Europäischen Kommission festgestellt werden. Außerdem geht aus der Arbeit hervor, dass die EU-Kommission seit 2014 verstärkt ihre Kompetenz im Wettbewerbsrecht nutzt und in zahlreichen Beihilfeverfahren gegen Unternehmen wie auch EU-Mitgliedstaaten und ihre Steuervorbescheide vorgeht.
Beide Erkenntnisse lassen den Schluss zu, dass die Steueraffären Anstöße für eine Europäisierung gegeben haben. In beiden Fällen sind neofunktionalistische Dynamiken zu beobachten. Eine Verschiebung der institutionellen Balance kann nur insofern festgestellt werden, als dass die EU-Kommission nach Bekanntwerden der Steueraffären gewichtige Entscheidungen in Steuerprüfverfahren getroffen hat und dabei auch gegen die Interessen von Mitgliedstaaten entschied.
The aim of this paper is to investigate whether the tax affairs of Lux Leaks, Panama Papers and Paradise Papers provided an impetus for a Europeanisation of EU tax policy in the field of corporate taxation. Therefore, the activities of the EU institutions after the publication of Lux Leaks are being compared to those prior to the affair over a period of four years each. A much higher number of legislative proposals made by the European Commission was presented in the aftermath of Lux Leaks. In addition, the European Commission has been increasingly using its competence in competition law since 2014 and is acting in numerous state aid cases against companies as well as against EU member states and their approved tax rulings.
Both findings suggest that the tax affairs have provided an impetus for Europeanisation. In both cases neofunctionalist dynamics can be observed. A shift within the institutional balance can only be noted insofar as the Commission has made major decisions after formal investigations on state aid against the interests of individual member states.

▼ Sammanställning Denna studie bestod i att göra en ljusmätning på Kalmar sjöfartshögskolas fartyg, Calmare Nyckel, och jämföra dess resultat med riktvärden från Transportstyrelsen och Arbetsmiljöver-ket. Mätningarna har utförts i utrymmen som används i utbildningssyfte och som fartyg. Den typ av belysning som det har gjorts mätningar på är allmänbelysning och platsbelysning. Det har använts en luxmeter som har placerats på ett stativ för att utföra mätningarna. Ett rutnät av mätpunkter har räknats ut till varje utrymme. Antalet mätpunkter har varierat i de olika utrymmena på grund av storleken på utrymmet. Vissa av arbetsområden och utrymmen upp-fyller riktvärdena och kraven med hänsyn till platsbelysning och allmänbelysning. I de utrym-men som det har kompletterats med nya ljusarmaturer är värdena mycket bättre.
Abstract This study consists of a light measuring on Kalmar maritime university vessel,”Calmare Nyck-el”, and compares the results with benchmarks from Transportstyrelsen and Arbetsmiljöver-ket. The measurements can be analysed in two different aspects one as educational premises and the other one as a vessel. General illumination and spot illumination are the kind of light-ning that have been checked. The illumination has been gauged by a luxmeter placed on a tripod. Datum points have established a grid in all premises by directions from authorities, the amount of datum points is decided by the dimensions of the area. Some of the areas and spaces meet the guide standards and requirements with regards towards to general illumina-tion and spot illumination. The values were much better in the premises that have been sup-plemented with new light fixtures.

▼ Despite the importance of molecular recognition in nearly all aspects of protein function, the determinants of specificity for enzyme-substrate and protein-protein interactions are poorly understood. The majority of these complexes involving bacterial luciferase from V. harveyi have yet to be fully characterized. Luciferase catalyzes the reaction of molecular oxygen, FMNH2 and a long-chain aliphatic aldehyde yielding FMN, the corresponding carboxylic acid and blue-green light. In vivo, luciferase is thought to obtain FMNH2 following transfer from a transiently associated oxidoreductase. To identify the oxidoreductase responsible for providing FMNH2 in E. coli, bioluminescence was compared using single gene deletion strains deficient in either a homolog to the endogenous V. harveyi oxidoreductase (Frp) or an oxidoreductase distantly related to luxG from V. fischeri (Fre). Fre is responsible for reducing flavin in vivo but does not physically interact with luciferase. The association between luciferase and the flavin product is also described. Luciferase was crystallized and subjected to soaking with high concentrations of FMN. A model was obtained for luciferase bound to FMN. Using molecular dynamics, models for the enzyme:aldehyde, enzyme:FMNH2, and luciferase bound to several reaction intermediates are presented. Finally, a conserved loop region adjacent to the active center was investigated for the ability to facilitate protein:protein interaction between luciferase and the endogenous Frp oxidoreductase. Following alanine mutagenesis of the charged residues throughout this loop, it appears that the residues targeted by this study are not components of a docking platform but facilitate a lid-gating mechanism of paramount importance for catalytic function.

▼ Radiation portal monitors (RPMs) are employed worldwide at border crossings and other entry points as a method to detect the illegal transportation of radioactive material. Scintillating detectors composed of polyvinyl toluene (PVT) plastic are installed within the RPMs for vehicle and cargo screening, seeking gamma ray emissions that indicate the presence of radioactivity. A change in opacity referred to as fogging within PVT has been documented and is attributed to prolonged exposure in temperature and humidity fluctuations. The opacity changes lead to reduced light collection in the photomultiplier tube (PMT) and degradation in PVT over time due to irreparable microfractures. As part of an ongoing effort to combat PVT fogging, the purpose of this study was to develop a Opacity Monitoring System (OMS) capable of observing opacity changes in-situ. The OMS was tested, and the functionality demonstrated. The final design consisted of an array of multiple light emitting diodes (LEDs) as a light source, an optical sensor (OS), and a microcontroller board for data capture and transmission. The OMS was prototyped and tested at Texas A&M University for feasibility on small scale utilizing 0.038m x 0.152m x 0.762m (5 ⅞” x 3” x 1 ½”) PVT samples, and full scale testing on 0.88m x 0.15m x 0.104m (2.9 ft x 0.5 ft x 0.125 ft) PVT panels in an environmental chamber at Oak Ridge National Laboratories. Light intensity data obtained through the OMS during testing was analyzed and showed clear indication of opacity changes in the PVT detector throughout multiple hours of extreme temperature cycling, thereby demonstrated the capabilities of the design and endurance of the hardware throughout extreme environmental changes.

▼ In this thesis, a flux switching permanent magnet generator (FSPMG) with three-stages skewing rotor applying in micro-scale hydropower generator was proposed. This innovation structure was arranged with a new type rotor called rotor skewing, which can reduce the cogging torque. Based on the inner holes design at rotor, there has six combinations of skewing angle in one rotor. The coil and magnets of FSPMG are located at the same side of outside stator. This arrangement was able to increase the speed adaptability of generator, which resolve the disadvantages of conventional generator only suitable for high speed. In research process, an Insert-mounted permanent magnet generator (IPMG) was used to compare the measurement and simulation results and established the appropriateness between the mathematical models and the actual structures. Then the flux density of magnet and the specification of FSPMG are derived by the method of equivalent magnetic circuit analysis. The dimension is limited at the diameter 12 cm of outer stator and the length 10 cm of stack length. The combination of stator slots and rotor poles are chosen for 12 and 10. The rated speed and the rated power of generator are 1200 rpm and 1 kW. A two-dimensional electromagnetic software ANSYS Maxwell was used to simulate the output performance of FSPMG structure such as flux density, back-EMF, harmonic distortion, core loss, strand loss, cogging torque and the output power in different rotor poles. The simulation results show that the structure of rotor tooth was optimized. At last, one set of FSPMG structure with 12 stator slots and 11 rotor poles are chosen for manufacturing the prototype. The experiment results show that the error of IPMG between the measurement and simulation can reach 2.6%, which confirms the accuracy of the numerical analysis. The simulation results show that the FSPMG can produce 1.1 kW with 6.9 Ω at 1200 rpm. Final, the prototype of FSPMG was measured with open-load test and loaded test with 6.9 Ω, the results are 75.6 V and 991 W. The error between simulation and measurement is 11.5% and 7.1%. The cogging torque measurement was measured in three skewing angle rotors: no skewing, equal difference skewing angle and equal ratio skewing angle. The results show that the cogging torque were 2.13 Nm, 0.85 Nm and 0.55 Nm, which decreased 60% and 74.2% respectively. It is confirmed that the skewing rotor can effectively reduce the cogging torque. This study provides a design and optimization process for FSPMG with a lower cost and high efficiency mathematical simulation analysis, and the skewing rotor effectively reduce the cogging torque and the torque ripple.

▼ The hunt for Dark Matter plays a truly critical role in contemporary physics. At both the largest and smallest scales, deep questions are being raised about the fundamental nature of the universe - questions that confirmation and then characterisation of particle dark matter will provide many answers to. This thesis presents some of the world's most sensitive searches to date for certain types of axion dark matter, axion-like particles, and two-neutrino double electron capture. These have been conducted using the Large Underground Xenon (LUX) experiment. Evidence for dark matter and physics beyond the Standard Model of particle physics is described in Chapter 1, while Chapter 2 gives an overview of proposed candidates for particle dark matter. The various experimental approaches being used to detect particle dark matter are presented in Chapter 3. Direct detection with time projection chambers plays a major role in this thesis, with particular interest in the LUX detector, that is described in its components and operations. Chapter 4 presents LUX direct searches for weakly interacting massive particles. Although I have contributed to these analyses, they are included for completeness only, as they are not part of my central work. The LUX collaboration's searches for axion dark matter and axion-like particle have delivered world-leading results on the axion-electron coupling constant. These results, that I personally led and which have been published in Physics Review Letters, are presented in Chapter 5, along with sensitivity studies, also led by me, made for the future LUX-ZEPLIN experiment. Finally, a search for two-neutrino double electron capture of 124Xe, that I performed using LUX data to extract a limit on the half life of the process, is presented in Chapter 6. Although being allowed by the Standard Model, two-neutrino double electron capture shares the matrix element calculation framework with the neutrinoless channel of the same process, becoming of great interest in the scope of neutrino physics. Conclusions follow and close the thesis.

▼ O presente relatório retrata a experiência vivida num estágio de marketing e promoção turística no grupo nacional hoteleiro
Lux Hotels.
Este relatório está estruturado em quatro capítulos de acordo com o estágio realizado. O estágio teve duração de nove meses, sendo que os primeiros sete meses decorreram no Lisboa Carmo Hotel, um dos hotéis do grupo, e os outros dois meses no departamento de marketing do grupo.
O primeiro capítulo aborda a fundamentação teórica, onde são expostos e desenvolvidos os conceitos de marketing turístico, a importância do marketing no setor hoteleiro, o marketing e as relações públicas e, por fim, a importância da internet e das redes sociais no turismo. Todos estes temas estão diretamente ligados com as tarefas realizadas ao longo do estágio.
O segundo capítulo descreve a empresa onde decorreu o estágio. Neste capítulo é apresentado, de forma pormenorizada, o grupo hoteleiro, a sua história, conceito e desenvolvimento, bem como uma análise pessoal ao marketing do grupo. Também neste capítulo é possível encontrar alguns aspetos personalizados acerca do Lisboa Carmo Hotel.
O terceiro capítulo descreve todas as tarefas realizadas ao longo do estágio, relacionadas com marketing e promoção no turismo.
Por último, o capítulo quatro são as conclusões, onde se pode ler uma análise critica acerca do estágio e tarefas desenvolvidas.
Com este trabalho pretendo mostrar as funções de um agente de marketing num grupo hoteleiro a nível nacional.

▼ L’analyse des processus de création de l’écrivain américain William Burroughs et de l’artiste brésilien Cildo Meireles construit deux figures exemplaires de l’artiste en tant que sujet politique. Les choix faits par ces deux artistes pendant le processus de réalisation de leurs œuvres déclarent en effet leur position face à la littérature, à l’art et à la réalité. Ils les construisent en tant que sujets-artistes et produisent des rencontres qui (re) configurent l’espace artistique. Ils renouvellent les pratiques de création. Nous étudions ces processus avec pour outils différentes notions théoriques. Outre celle de la politique, qui, selon Jacques Rancière, est le processus de l’égalité ; celle du sujet, définie par Alain Badiou, comme une construction qui se produit quand un événement arrive ; et celle de choix qui, selon Slavoj Zizek, ne laisse pas intacte la subjectivité du sujet. Nous essayons ainsi de définir le caractère politique des choix faits par ces deux artistes et de mettre en évidence l’idée de l’art et de la politique que leurs œuvres mobilisent.Nous étudions le processus de création de Burroughs, de son premier roman jusqu’à la découverte du cut-up et l’écriture de son texte Last Words. Nous analysons le processus de création des œuvres de Meireles : Insertions dans les circuits idéologiques, œuvre qui se constitue dans un processus en marche ; et Le sermon sur la montagne : Fiat lux. Construction d’une situation : transformation d’un espace en une bombe potentielle.Malgré leurs différences, ces œuvres ont une procédure de réalisation similaire, une sorte de travail tabulaire, de collage d’éléments hétérogènes.
The analysis of the creation process of both Americain writer Willam Burroughs and Brazilian artist Cildo Meireles construct two exemplary figures of the artist as political subject. The choices made by these artistes during the process of realization of their works declare indeed their position on literature, art and reality. Their choices construct them as artists-subjects and produce encounters that (re) configure the art space. They renew the creative practices. We study these processes with some theoretical tools: such as the notion of politics which according to Jacques Rancière, is the process of equality; the notion of subject, defined by Alain Badiou as a construction that is produced when an event happens; and the notions of choice which, according to Slavoj Zizek, does not leave intact the subjectivity of the subject. Thus we try to define the political nature of the choices made by these artists and to highlight the idea of art and politics that their works mobilize. We study the process of creation of Burroughs, from his first novel until his discovery of cut-up technique and the writing of his text Last Words. ▼ This research project investigates the creation of
immersive contemplative viewing experiences within my visual arts
practice by identifying and adapting the fundamental elements of
Japanese contemplative space into my aesthetic and conceptual
lexicon. Contemplative experience is an intrinsic aspect of the
human experience which has become increasingly scarce in this age
of perpetual overstimulation and increasing secularisation. The
primary goal of the research project is to provide a significant
opportunity for new audiences to engage in meaningful
contemplative experience informed by the centuries-old principles
of yūgen 幽玄, ma 間, and hikari to kage 光と影 within
Japanese architecture. The studio-based research first explores
the creation of contemplative objects constructed with glass,
followed by a series of maquettes of potential immersive
contemplative environments. The research culminates with the
immersive installation, Lux Mandala, which utilises the material
characteristics of glass microspheres to synthesise the ephemeral
optical phenomenon of the “glory” into a meditative encounter
with light, shadow, space, transience and profundity. By
aesthetically locating the viewer at the centre of this
phenomenon, the viewer’s presence and perceptual mechanisms
complete a participatory loop, allowing the phenomenon to enter
into existence while enabling the viewer to reflect upon its
nature, notions of perception and transience, and the
significance of their own presence within this evanescent
tableau. The outcomes of this research project represent a
significant new nexus between visual arts practice, immersive
experience, and the essence of Japanese contemplative space.

▼ Since its characterization, the definitive shortcoming of the bacterial luciferase (lux) bioluminescent reporter system has been its inability to express at a functional level in the eukaryotic cellular background. While recent developments have allowed for lux function in the lower eukaryote Saccharomyces cerevisiae, they have not provided for autonomous function in higher eukaryotes capable of serving as human biomedical proxies. Here it is reported for the first time that, through a process of poly-bicistronic expression of human codon-optimized lux genes, it is possible to autonomously produce a bioluminescent signal directly from mammalian cells. The low background of the bioluminescent signal, along with its characteristic lack of substrate amendment required for bioluminescent production, makes a mammalian-based lux reporter system ideal for real-time monitoring of cell culture or murine model systems. The delectability of a lux-based system provides for a functionally equivalent process to monitoring firefly luciferase-expressing cells under cell culture or subcutaneous imaging conditions without the well-documented uncertainties stemming from additional substrate introduction. However, the relatively blue-shifted emission wavelength of the lux reporter system, along with its low quantum yield, has been shown to reduce its effectiveness for use during deep tissue imaging of animal subjects. Despite these disadvantages, it has been demonstrated that a human cell line expressing the human codon-optimized lux genes can function as a biosensor for determination of human bioavailability of toxic compounds and that, by regulating the production of the luxC and luxE genes, the lux system can be employed as the first mammalian, real-time, fully autonomous bioreporter. These cell lines provide unique and efficient models for the detection and monitoring of human-relevant compounds of interest. The limiting reagent for bioluminescent production in the mammalian cellular background has been determined to be the cytosolic availability of the FMNH2 co-substrate and, in light of this evidence, directions for future optimization have been characterized and evaluated in respect to their ability to increase bioluminescent yield under these conditions.

gravitational galactic rotations based upon solar luminous intensity and the observed rotations of
galaxies. The cause of the observed increased rotational velocity in galaxies was ascribed to a
large invisible mass termed dark matter. Various indirect observations affirmed the notion of
dark matter and also suggested that the dark matter consists of non-baryonic material. Dark matter currently stands as one of the great mysteries in modern physics.
A leading candidate for dark matter is known as the Weakly Interacting Massive Particle
(WIMP) which nicely fits dark matter projections. The Large Underground Xenon (
LUX) experiment, located in the Homestake Mine of the Sanford Underground Research Facility (SURF), was a xenon-based direct detection experiment designed and built for the detection of WIMPs.
LUX had a dual-phase Time Projection Chamber (TPC) which allowed for precise energy and position reconstruction of events. Reanalyzing the
LUX data, it becomes possible to search for more exotic theoretical dark matter forms and new physics.
The Lightly Ionizing Particle (LIP), a theoretical particle with a charge of ∫ ∙ ⅇ with ∫ < 1 and
ⅇ the electron charge, arises in a variety of dark matter conceptions associated with additional dark sectors as well as other physical constructs such as monopoles and quantized charges. This work seeks to use data from the first underground run of
LUX Run03, and reanalyze it to search for tracks in the detector as a result of LIP interactions. Throughout the reanalysis new techniques in simulation, data analysis, and data processing are developed, including pulse chopping to separate merged signals. The LIP search, making use of traditional cut methods as well as machine learning for event classification, places new limits on LIP flux in the charge range 0.01ⅇ to 0.3ⅇ as low as 10⁻¹⁰ cm⁻² s⁻¹ sr⁻¹.
