La détection d'ondes gravitationnelles en 2015 a posé la première pierre d'un tout nouveau pan de la physique de l'Univers. La multiplication et l'amélioration des détecteurs permettront, associées aux observations électromagnétiques, d'obtenir de précieuses informations sur les objets peu connus de l'Univers, comme les trous noirs. Dans ce contexte la mission LISA (Laser Interferometer Space Antenna) sera le premier détecteur interférométrique spatial à ondes gravitationnelles. Concrètement, l'effet d'une onde gravitationnelle est une déformation périodique de l'espace-temps, très faible, mais pouvant être détectée par mesure interférométrique. La mission LISA détectera ainsi des variations de longueur de l'ordre du picomètre pour une longueur de bras de l'ordre du million de kilomètres, soit une variation relative de 〖10〗^(-21). De tels niveaux de sensibilités nécessitent des techniques avancées de réductions de bruit qui doivent être testées numériquement et expérimentalement. Le bruit dominant sur la mesure sera le bruit de fréquence du laser, qui nécessitera, outre les techniques de stabilisation laser, l'utilisation d'une méthode de traitement numérique appelée interférométrie à temps retardés (TDI) qui permet de reformer virtuellement un interféromètre à bras égaux, ce qui a pour effet d'annuler le bruit de fréquence du laser. C'est particulièrement cette technique qui est testée par l'expérience faisant l'objet de cette thèse. Le LISA On Table (LOT) est un simulateur comportant une partie électronique et une partie optique permettant de simuler des signaux réalistes de LISA afin de tester TDI. Il permet de faire interférer des signaux retardés simulés représentatifs du temps de trajet de la lumière entre les satellites de la constellation LISA. L'expérience possédant ses propres bruits instrumentaux, il s'agit de travailler à leur compréhension et leur diminution. Le LOT permet en outre de tester d'autres points de la mission, comme le transfert de bruit d'horloge ou bien des prototypes d'instruments comme le phasemètre par exemple. Les travaux réalisés ont permis de faire fonctionner la partie optique du LOT sous vide, afin d'en limiter les bruits instrumentaux. L'expérience permet de tester et de valider TDI dans une configuration réaliste à bras inégaux de LISA. Des tests permettent aussi de valider la simulation de l'effet Doppler.

The first gravitational waves detection in 2015 opened a brand new window on astrophysics and cosmology. Developement and enhancement of new instruments, coupled with electromagnetic observations, will help with the understanding of quite unknow objects of the Universe, such as black holes. ▼ Although not immediately apparent, the structure of this thesis is based on some of the core notions of my art practice. My main goal is to redeem Lisa Nowak, the defamed astronaut, while inserting a somewhat personal discussion regarding domesticity with feminist underpinnings. The relationships between Lisa, the fluff and myself are not always linear and recognizable; the delineations of positions become entangled and non-hierarchical. This is much like the complicated relationship between the analyst and analysand, perhaps in a state of "hysteria"; with the fluff being object of fixation, or possibly, just the fix. It seems to be much like the problematic relationship between Lisa Nowak, William Oefelein and Colleen Shipman. I like to think of the fluff as an emulsion, and Lisa as the primer. The thesis is executed in a way that, much like looking at a painting or sculptural object, there is a focus on precise details and being completely engrossed, while simultaneously hovering as if the vantage point is from a far distance. Subsequently, the emerging image of the triangle is repeated throughout the thesis, as well as perceptual investigations. There are many embedded theories that remain of great interest for me as well, such as, abstraction, philosophy and psychology. This thesis is a good excuse to explore them more thoroughly in the name of fluff and Lisa Nowak. Both the fluff and Lisa become vehicles in order to explore the ideas of crossing boundaries and seeking new territories. With the excess of information that has subsumed our everyday lives in a Post-Modern era, the question as to where the location of a new topography exists. Contrary to a contemporary discussion of exhaustion within culture, this thesis also asserts that the sources that inform any artistic practice remain open, vast and endless, with possibilities of innovation, reformatting and shifting.; This thesis asks if indeed, this place is located within a proximity that is very close and readily accessible. This location would be the very impulse that seeks to find this new realm or enthusiasm, passion and imagination. Please keep in mind, this thesis is not blatant and does not have an aggressive agenda, it's fluffy. São investigados o comportamento estrutural e a resistência última à punção de ligações laje-pilar de regiões internas das lajes lisas, com um ou dois furos adjacentes ao pilar, e com ou sem transferência de momento fletor da laje ao pilar. As principais variáveis foram: o posicionamento e as dimensões dos furos, a taxa e a distribuição da armadura de flexão, a armadura de cisalhamento e a excentricidade de carga (relação M(momento transferido ao pilar)/V(força cortante)) na ligação. Foram ensaiadas 19 ligações internas laje-pilar, distribuídas em 2 grupos: o primeiro sem a transferência de momento (Série 1 10 lajes), com 1 (uma) ligação sem furo, 5 (cinco) com um furo, 2 (duas) com um furo e armadura de cisalhamento e 2 (duas) com dois furos, tendo como principais variáveis o tamanho e posicionamento dos furos, a taxa e a distribuição da armadura de flexão e a armadura de cisalhamento; e o segundo grupo com transferência de momento (Série 2 9 lajes), com 2 (duas) ligações sem furo, 4 (quatro) com um furo e 3 (três) com dois furos, tendo como principais variáveis o tamanho e posicionamento dos furos, a direção e sentido do momento transferido e a excentricidade de carga (relação M/V) igual a 0,50m ou 0,25m na ligação. Foram analisados os resultados de cargas últimas, fissuração, deslocamento vertical, deformações das armaduras de flexão, de cisalhamento e do concreto. O posicionamento dos furos em relação ao pilar, adjacente ao maior ou menor lado, as dimensões dos furos, a taxa e a distribuição da armadura de flexão, a armadura de cisalhamento, a variação da relação Mu/Vu em função do carregamento, e, por conseguinte, da excentricidade, influenciaram o comportamento e a carga de ruptura das lajes. Essas influências foram quantificadas. Os resultados experimentais foram comparados com os estimados pelas normas: CEB-FIP/MC1990, EC2/2004, ACI-318:2005 e NBR 6118:2003. O ACI e o EC2 apresentaram estimativas mais conservadoras, embora tenham apresentado algumas estimativas contra a segurança. A NBR 6118:2003, apesar de baseada em parte no EC2, apresentou estimativas menos conservadoras e com um número maior de estimativas contra a segurança. Recomenda-se que todas as normas passem a considerar o momento causado pela excentricidade do perímetro crítico em lajes com furos e, neste sentido, foram apresentadas propostas de modificações para todas as normas estudadas.

Os sistemas de canais de areia submarinos, ou canais turbidíticos, são, devido à sua natureza detrítica, potenciais reservatórios de hidrocarbonetos, normalmente com uma boa porosidade. No entanto, este tipo de reservatório caracteriza-se por uma acentuada heterogeneidade espacial, típica de uma situação não estacionária e que resulta do seu traçado meandriforme. Desta forma, compreender o padrão morfológico que define o corpo de um canal de areia é essencial para a caracterização destes reservatórios, o que por vezes se pode tornar uma tarefa difícil, devido à elevada variabilidade espacial e litológica. O objectivo deste trabalho é implementar um conjunto de novos atributos sísmicos que permitam identificar as características morfológicas e padrões espaciais em situações não estacionários típicos de um reservatório de hidrocarbonetos em canais de areia, tentando assim melhorar os processos de simulação estocásticos e de inversão sísmica deste tipo de reservatório. Para isso foi proposta a utilização dos estatísticos LISA (Local Indicators of Spatial Association) como novo atributo sísmico. Estes estatísticos são normalmente utilizados em aplicações com dados geográficos e permitem detectar padrões de associação espacial locais, que podem reflectir o corpo do canal de areia, uma vez que os LISA dão o grau de clustering para um conjunto de amostras. A aplicação dos LISA como atributo sísmico para a identificação de padrões morfológicos ou na identificação de lito-grupos, revelou-se ser uma boa ferramenta, em particular a utilização do índice Local Moran’s I, uma vez que permite a identificação dos padrões de associação espacial formados pelos corpos de areia e definir as direcções de maior anisotropia espacial. No entanto a sua integração nos processos de simulação estocásticos e no processo de inversão sísmica deverá ser complementada com outros condicionantes.

Systems of submarine sand channels, or turbidites channels, are potential hydrocarbon reservoirs, due to its clastic nature, which normally provides a good porosity. However this type of reservoir is characterized by a high spatial heterogeneity typical of a non-stationary pattern in result of his sinuous shape. ▼ Death from breast cancer began a decline in the 1990’s. One widely accepted reason for this reduction was the discovery that breast cancer can be uniquely defined at the molecular level by oncogenes and tumor suppressor genes including, but not limited to, EGFR, AKT, ErbB2, PI3K, TP53, BRCA1/2, and PTEN. These genes and potential mutations within, can drive constitutive activation of aberrant signaling that can induce and sustain tumorigenesis. More importantly, these genes represent new avenues for possible markers and therapies. In the wake of this molecular biology discovery, there was an influx of new breast cancer treatments to the marketplace, most notably the launch of trastuzumab (Herceptin) in 1996. Subsequently, the incidence of death continued to decrease peaking from 2002 to 2003 when it dropped by 7% in that one year alone. However, the current challenge in oncology is how to translate the wealth of information contained in the molecular biology of cancer and translate it into patient care. While progress has been made in developing treatments, less progress has been made identifying new markers for breast cancer to get that treatment to the right patients or selecting them for treatment trials. Currently, the majority of providers and healthcare systems do not utilize molecular biology to determine risk of breast cancer recurrence and treatment decision making for breast cancer. Rather, they determine which patients require what adjuvant treatment based on some form of the breast cancer tumor–node–metastases (TNM) staging system and clinical-pathological criteria, such as lymphovascular invasion (LVI), nodal status, estrogen receptor (ER), progesterone receptor (PR), human epidermal growth factor receptor 2 (HER2/neu) status and more. As a result, potential differences exist worldwide in the selection of patients who require adjuvant chemotherapy based on their risk of breast cancer recurrence. All of these tests and criteria are based on the anatomical extent of the tumor, with little if any insight into the patient’s breast cancer biology. A significant percentage (30 to 50%) of all early stage breast cancer patients (50,000 to 75,000 patients annually in the US alone) have clinically ambiguous or confounding clinical-pathological characteristics, making it difficult for physicians to formulate clinical risk and supports the argument for development and utilization of molecular diagnostic testing. By looking at the molecular biology of cancer, companies, such as Agendia, seek to create technologies beyond clinical screening, imaging or cell surface staining. Technologies like MammaPrint and BluePrint, reveal who is truly at risk by differentiating responders from non-responders based on genetics. By interrogating single genes or overall patterns of gene expression, it is possible to better understand the various sequences of biological events that give rise to breast cancer and in turn, better develop and guide treatments. ▼ Subtle gravitational e ects can be measured by precisely monitoring the position of a test mass. Often this is done by measuring the displacement between two or more test objects. The Gravity Recovery and Climate Experiment (GRACE) satellites do just this, by continuously tracking changes in their separation with micron-level sensitivity. These displacement measurements are used to infer the gravitational potential of the Earth, which has enabled scientists to monitor key aspects of our climate since their launch in 2002. It is planned that the GRACE Follow-On satellites will include a laser ranging instrument as a technology demonstrator to improve the displacement measurement. Before science operation commences and measurements can begin, the laser on each satellite needs to be precisely pointed towards the opposite satellite, and thus the satellites must undergo an initial acquisition scan after launch to establish the laser link. This thesis is concerned with developing technology for the GRACE Follow-On laser ranging instrument and exploring interferometric techniques for future satellite missions. In the following chapters, we experimentally demonstrate an acquisition system with GRACE Follow-On-like parameters, requiring no additional hardware but relying on the photodetectors and signal processing equipment already required for science operation. This strategy was developed with multiple collaborators over several years led by C. Mahrdt at the Albert Einstein Institute. To establish the laser link, ve degrees of freedom must be optimized (pitch and yaw for each beam, and the frequency di erence between the two lasers). Laser steering and frequency scanning patterns are combined with a fast Fourier transform-based peak detection algorithm run on each satellite to nd the signal. We successfully demonstrate both stages (commissioning and reacquisition) of the proposed acquisition strategy. One of the core components needed for the GRACE Follow-On laser ranging instrument is the triple mirror assembly (TMA), a modi ed corner cube that symmetrically routes the laser beam around existing hardware about the satellite's center of mass. A prototype triple mirror assembly was designed and constructed by local and international collaborators, and we present optical tests demonstrating three of the performance requirements of the prototype. The path length stability of a beam traveling through the TMA was measured in a test bed resembling the measurement con guration of the GRACE Follow-On interferometer. The parallelism between the incoming and outgoing beams to/from the TMA is measured to the arc second level. Additional measurements quantify changes in the parallelism as the TMA prototype is heated and cooled. Finally, we give a brief overview of digitally-enhanced interferometry, a developing technique for optical metrology which has signi cant advantages over a conventional heterodyne system and could be employed for future space missions. With an in vehicle camera many different things can be done that are essential for ADAS or autonomous driving mode in a vehicle. First, it can be used for detection of general objects, for example cars, cyclists or pedestrians. Secondly, the camera can be used for traffic light recognition, which is localization of traffic light position and traffic light state recognition. No method exists at the moment able to perform general object detection and traffic light recognition at the same time, therefore this work proposes methods to combine general object detection and traffic light recognition. The novel method presented is including traffic light recognition in a general object detection framework. The single shot object detector YOLOv2 is used as base detector. As general object class dataset COCO is used and the traffic light dataset is LISA. Two different methods for combined detection are proposed: adaptive combined training and YOLOv2++. For combined training YOLOv2 is trained on both datasets with the YOLOv2 network unchanged and the loss function adapted to optimize training on both datasets. For YOLOv2++ the feature extractor of YOLOv2 pre-trained on COCO is used as feature extractor. On the features LISA traffic light states are trained with a small sub-network. It is concluded the best performing method is adaptive combined training which reaches for IOU 0.5 a AUC of 24.02% for binary and 21.23% for multi-class classification. For IOU of 0.1 this increases to 56.74% for binary and 41.87% for multi-class classification. The performance of the adaptive combined detector is 20% lower than the baseline performance of an detector only detecting LISA traffic light states and 5% lower than the baseline of a detector only detecting COCO classes, however detection of classes from both dataset is almost twice as fast as separate detection with different networks for both datasets.

▼ The spatiotemporal interactions and feedbacks of fire and vegetation in savanna systems are a key component of savanna structure, function, and can give insight into woody plant encroachment. The increased accessibility to remote sensing data and software via Google Earth Engine for fire analysis facilitates integrated savanna assessments for local-to-regional analyses and management. This study assesses spatial and temporal fire dynamics in the Botswana Kalahari at different extents and resolutions to test the hypothesis that fire and vegetation patterns interact via distinct processes at different extents and resolutions in savanna systems. MODIS burn products (MCD64A1 500meters) and in situ vegetation and fire measurements were used to test the affects of variables at different extents and resolutions on savanna fire and vegetation patterns. Spatiotemporal analysis at the extent of the Botswana Kalahari explored 1) the spatial autocorrelation of fire return time using a bivariate Moran's I analysis to observe how large patterns of fuel dynamics affect neighboring areas across time and space and 2) how variability of variables over small spaces influence larger patterns of fire occurrence using Geographically Weighted Regression or GWR. In situ grass and woody vegetation characteristics were measured and combined with MODIS fire history data to observe how vegetation at the square meter scale is influenced by regional affects of fire occurrence, grazing, and human influence using OLS regression. Fire intensity and post-fire vegetation mortality assessments were performed on sites that burned approximately two months after initial measurements were recorded to observe vegetation affects on fire intensity and the initial affects of fire on vegetation. Woody plant populations were projected across a number of fires under divergent fuel conditions to observe how fine resolution fire and vegetation patterns influence larger patterns. In the Botswana Kalahari study area, fire occurrence over time was heavily affected by fire presence in neighboring 500m pixels (first and second order), an indication that spatiotemporal patterns of fire are affected by patterns in neighboring areas. The temporal and spatial patterns observed suggest that temporally dynamic neighboring fuel conditions impact fire in a given location. Large scale patterns were observed between variables and fire occurrence using an OLS regression, and spatial variability of local coefficients was observed using a GWR model. Seasonality of precipitation, boreholes, and EVI had negative significant coefficients. Soil moisture, drought severity, and herbaceous cover had positive significant coefficients, but when examined locally using GWR there were high amounts of spatial variation – every variable ranged from positive to negative significant local coefficients except for seasonality of precipitation. ▼ Gravitational waves are a prediction of Einstein's General Relativity, the detection and analysis of which will open a new area in our understanding of the Universe. Indeed, these waves carry information from systems with strong gravity and are very difficult to observe with conventional instruments, which are based on the detection of light. Today, we have experimental evidence of the existence of gravitational waves, as the variation of the orbital period of the binary pulsar PSRB1913 + 16 (Nobel Prize 1993). However, there has been no detection. The scientific community is pursuing this detection with several observatories spread around the world: LIGO (USA), VIRGO (Italy, France), GEO600 (Germany, UK), LCGT (Japan ), etc. LISA (Laser Interferomer Space Antenna) is a proposal to put one of these observatories in heliocentric orbit using three satellites that contain masses in free fall. A laser interferometer is used to measure the distance between them. The satellites will be separated 1 Mkm, resulting in a triangular configuration that follows the Earth in its orbit. This proposal was accepted by the European Agency Espacia in 2013, as the subject of a mission to be launched in the early 2030's. This thesis is part of the LISA Pathfinder mission, which is the precursor mission of LISA and has a release date in 2015. This mission will test the technology required for the future LISA mission and therefore contains the main elements (laser, test mass, etc.) but in a smaller version and in a single satellite. The main scientific objective is to measure the relative acceleration between two masses in free fall to levels down to 10^(-14) m / s² / Hz ^(1/2) in the low frequency band, i.e. 1mHz. This thesis develops the methods known as Markov Chain Monte Carlo (MCMC), which are used, among others, for the estimation of parameters. These techniques have been put to the test in recent years through various campaigns with simulated data and are currently part of the LTPDA, a MATLAB toolbox developed by the collaboration for the analysis of data from the mission. The work in this thesis describes in detail the application of these techniques to estimate the parameters that describe the dynamics of the test mass inside the satellite. Similarly, we have developed statistical techniques beyond the estimation of parameters in order to apply them to what is known as model selection, i.e. the evaluation of different statistical models to determine which one allows a better description of the data. In this respect, we have evaluated the different options available in the literature and, in particular, we have established the Reversible Jump Markov Chain Monte Carlo (RJMCMC) as a tool to carry out these studies during operations. This technique allows the comparison of models of different dimensionality, which represents a generalization of the MCMC methods. ▼ Piper aduncum (Piperaceae), nativa da região Amazônica, é conhecida como pimenta-de-macaco e aperta-ruão. É utilizada popularmente no tratamento de hemorragias, hemorroidas, diarreia, dor de dente e inflamação. Estudos fitoquímicos relatam o isolamento e identificação de mais de 47 compostos de P. aduncum, sendo o dilapiol (fenilpropanoide) o principal componente. No entanto, estudos farmacológicos com extratos padronizados ou substâncias isoladas são escassos e não há relatos na literatura científica da padronização química quantitativa dos extratos de Piper aduncum estudados. Considerando essas informações, este estudo propõe a investigação científica de extratos padronizados de P. aduncum avaliando a farmacodinâmica e os efeitos desses extratos no sistema cardiovascular e na musculatura lisa. Folhas de P. aduncum foram coletadas no minicampus da Universidade Federal do Amazonas (UFAM). A infusão do pó das folhas secas em água destilada originou o extrato aquoso (EA), que após partição em n-butanol originou a fração butanólica (FBut). EA e FBut foram padronizados em CLAE – UV 260 nm obtendo-se 9 picos principais com TR e amplitudes características no espectro de referência. A FBut foi purificada em CLAE preparativa, resultando 6 frações (F1 a F6). Nos testes biológicos, camundongos tratados com o EA (1 g/kg) por via oral não apresentaram efeitos que os diferenciassem dos animais controle tratados com água. Com a injeção intraperitoneal da FBut (0,2 e 0,5 g/kg), os animais apresentaram leve diminuição da deambulação e sedação nas primeiras 2 h após a injeção da FBut. Em ratos anestesiados, a injeção endovenosa do EA (1, 3 e 10 mg/mL) produziu hipertensão proporcional às doses; este efeito não foi observado com a injeção da FBut. A administração prévia do prazosin (1 mg/kg, e.v) inverteu o efeito do EA para hipotensão, sendo novamente revertido para hipertensão na presença do propranolol. A injeção endovenosa de adrenalina (ADR 0,1; 0,3 e 1,0 μg/mL) produziu efeito bifásico (hipertensão e hipotensão) na dependência da dose. Com a injeção prévia do prazosin, a ADR e o EA produziram hipotensão, que foi revertida para hipertensão após administração de propranolol. Em átrio direito isolado de rato, EA e FBut aumentaram a frequência de batimentos. Em átrio esquerdo isolado de rato, EA e FBut aumentaram a amplitude de contração; este efeito foi bloqueado pelo propranolol (10-6 M), indicando ação inotrópica positiva mediada por receptores β1 adrenérgicos. A atividade da Ca2+-ATPase extraída de músculo esquelético de coelho não foi inibida pela FBut. Em ducto deferente de rato (DDR), a FBut inibiu a contração máxima da noradrenalina de 35% sem alterar a CE50, indicando antagonismo do tipo não-competitivo e excluindo uma ação bloqueadora α-adrenérgica. Em DDR pré-contraído com ATP, a FBut inibiu o efeito do ATP nos receptores purinérgicos P1. ▼ ABSTRACT THE JOURNEY AN INTERNSHIP IN URBAN ACTIVISM, MUSIC VIDEOS: ZOMBIE AND BAD SYNE, AND A STUDY OF AFRO-PANAMANIAN IDENTITY & THE REGGAETÓN MUSIC MOVEMENT by Lisa Margaret Spencer The University of Wisconsin-Milwaukee, 2019 Under the Supervision of Co-Chairs: Dr. Theresa Kenney and Dr. Tami Williams PART I- Under the guidance of Dr. Jill Florence Lackey A major component of my doctorate included an internship in cultural anthropology at UrbAn in Milwaukee, Wisconsin with Dr. Jill Florence Lackey. The non-profit organization was housed in the Lincoln Park Village neighborhood. I assisted in planning events in the agency’s the South Side Museum, implementing the South Side Farmer’s Market, executing, and naming, the first “Gathering of the Nations” cultural festival in Kosciusko Park, proofreading a potential multi-cultural studies curriculum for Milwaukee Public Schools, transcribing stories of the city’s homeless youth, and videotaping Milwaukee police officers for a Graduate course at Marquette University, and contributing to other agency projects. A key component of working as a cultural anthropologist is to connect groups and agencies through commonality that ultimately builds understanding and community. The methods and programs established by UrbAn were connected to many aspects of social justice, bringing voice and celebration to outsider, minority, and underprivileged peoples. Storytelling validates identity in personal, cultural, and unified modalities. I was able to use my film, social work, and Spanish language skills to maneuver through the community and serve it in ways best suggested by the agency’s founder and the inhabitants themselves. The project designed by UrbAn organized people to be in community together. Activities, events, and initiatives brought cultural groups together to celebrate their differences, rather than be divided by them. I took the footage and during my doctoral studies rented a film camera and expanded my footage, creating the narrative into a full music video. I entitled it, Zombie. The piece explores societal questions and inequities, as well as my relationship to these questions. I also began to explore the relationship between mind, body, and spirit on both a societal and personal level. The second video, Bad Syne, was created mid-way through my doctorate with the permission and supervision of Iverson White. My time working on this piece gave me deeper insight into the lens of a Puerto Rican graffiti and hip-hop artist in New York City. I found the connections to be personal. ▼ Dental Composites are essential for current dentistry. They serve not only as direct and substance-protecting filling material, but are also used as fixation material for indirect, labor-made construction. The organic phase of composites contains different monomers. The most important monomer of current composites is the triethylenglycolmethacrylat (TEGDMA), an ester composed of α,β-methacrylat acid and ethylenglycol. Many studies evidenced cytotoxic and genotoxic properties of the TEGDMA monomer connected with negative effect on cell vitality and on the differentiation of fibroblast pulp cells. At the moment the exact mechanisms of TEGDMA action are unknown. To study the influence of TEGDMA on differentiation of human cells LiSa-2 cells were treated with different concentrations of TEGDMA. LiSa-2 preadipocytes are a recognized and well-characterized model system for the investigation of adipocyte differentiation called adipogenesis. This process is controlled by a large number of regulatory proteins, the stability of which is determined by the ubiquitin proteasome system (UPS). One component of the UPS, the COP9 signalosome (CSN), is essential for adipogenesis. The CSN binds to cullin- RING-ubiquitin ligases (CRLs) and regulates their activity. CRLs ubiquitinate important regulators of adipogenesis, such as p27 and CHOP, and target them for proteolysis by the 26S proteasome. In the present study the following questions were answered: How does TEGDMA treatment of LiSa-2 cells influence the CSN, components of CRLs such as Cul1 and Cul3 and the substrates p27 and p53? How does TEGDMA influence the differentiation of LiSa-2 preadipocytes? Is there an effect of TEGDMA on the apoptosis in LiSa-2 cells? What is the mechanism of TEGDMA action in LiSa-2 cells? The data shows that TEGDMA treatment leads to a reduction of CSN subunits and of CSN-associated proteins. In dependence on the TEGDMA concentration a retardation of adipogenesis in LiSa-2 cells was observed. At high TEGDMA concentrations an increase apoptosis rate in LiSa-2 cells was measured and cell vitality drastically decreased. Interestingly, the decreased cell vitality induced by TEGDMA treatment was significantly rescued by using the antioxidant N-acetylcystein (NAC). Whether the reduction of CSN subunits in the presence of TEGDMA is due to the elevation of reactive oxygen species (ROS) has to be studied in the future. As a consequence for dentistry we conclude that the TEGDMA monomer has to be kept low by optimized polymerization rate.
