▼ Este trabalho apresenta as propriedades ópticas dos complexos Európio Tetraciclinas (EuTcs) na presença de LDL e de LDL oxidada com potenciais aplicações em análises clínicas. Foram escolhidos quatro elementos da família das Tetraciclinas: Tetraciclina (Tc), Clorotetraciclina (CTc), Metatetraciclina (MTc) e Oxitetraciclina (OTc) para fazerem parte dos complexos com o íon európio. As melhores condições para se formar os complexos eficientemente foram determinadas, através das medidas dos parâmetros ópticos como: absorção, emissão e de tempo de vida. As melhores concentrações de európio nos complexos EuTcs e possíveis influências de íons inorgânicos normalmente presentes no plasma sanguíneo também foram analisadas. As amostras foram preparadas em pH neutro e a luminescência visível do lantanídeo foi detectada após tempo de repouso das amostras de 15 minutos. Os resultados deste trabalho mostraram que as moléculas de LDL e de LDL oxidada apresentaram um importante papel no aumento da intensidade de emissão dos complexos das Tcs. As medidas realizadas com os complexos EuTcs não apresentaram deslocamentos nos comprimentos de onda dos espectros de absorção e de emissão na presença de LDL, o que demonstra a ausência de interação direta entre as moléculas de Tcs e as moléculas de LDL e LDL oxidada. No entanto, o íon európio pode interagir em diferentes sítios das moléculas de tetraciclinas o que diferenciou a intensidade de emissão de cada complexo. Comparando os resultados obtidos entre os complexos de EuTcs, o complexo EuTc foi o que apresentou perspectivas promissoras na quantificação de LDL e LDL oxidada.
This work presents the optical properties of europium complexes - Tetracyclines (EuTcs) in the presence of LDL and oxidized LDL with potential applications in clinical analysis. Four elements were chosen from the Tetracyclines family: Tetracycline (Tc), Chlortetracycline (CTc), Metatetraciclina (MTc) and Oxytetracycline (OTc) to be part of complexes with europium ion. The best conditions to form the complex efficiently were determined through measurements of optical parameters such as absorption, emission and lifetime. The best concentrations of europium complexes in EuTcs and possible influences of inorganic ions normally present in blood plasma were also analyzed. The samples were prepared at neutral pH and the visible luminescence of lanthanide was detected after resting time of the samples of 15 minutes. These results showed that the molecules of LDL and oxidized LDL have an important role in increase of the emission intensity for Tcs complexes . The measurements performed with the complex EuTcs showed no shifts in the wavelengths of the absorption and emission spectra in the presence of LDL, which demonstrates the absence of direct interaction between the molecules of Tcs and the molecules of LDL and oxidized LDL. ▼ A obesidade é considerada uma doença crônica e multifatorial, onde eventos como a inflamação de baixa intensidade e as modificações oxidativas estão presentes. A elevada prevalência de obesidade tem impacto direto no desenvolvimento precoce de diabetes mellitus, hipertensão e outros fatores de risco cardiovasculares. Esse perfil tem motivado a identificação de biomarcadores precoces, sendo o monitoramento da lipoproteína de baixa densidade oxidada (LDLox) e lipoproteína de baixa densidade eletronegativa [LDL(-)] potenciais candidatos. Diante do exposto, o objetivo deste estudo foi realizar a análise comparativa e de correlação entre o conteúdo de LDLox e [LDL(-)] em adolescentes. Foram selecionados 137 adolescentes de ambos sexos, com faixa etária de 10 a 19 anos e regularmente em matriculados em escolas públicas da cidade de São Paulo. O peso, altura e circunferência da cintura (CC) foram avaliados. Após jejum (12h-15h) foi coletada uma amostra de sangue e, a partir do plasma, foram realizadas as seguintes análises: glicose, insulina, perfil lipídico, apolipoproteína (AI e B), ácidos graxos não esterificados, tamanho de HDL, atividade da CETP e LDL(-) e LDLox. Os resultados encontrados foram analisados por meio do programa SPSS 15.0, considerando valor de significância de p< 0,05. Os 137 adolescentes foram distribuídos em dois grupos: 71 no Eutrófico (51,82%) e 66 no Obeso (48,18%), segundo a classificação do IMC. 48 (35,04%) dos adolescentes eram do sexo masculino e 89 (64,96%) do sexo feminino, com idade média de 14,2 (2,3) anos. Em relação à CC, observou-se que essa confirmou a classificação feita pelo IMC. Observou-se também uma maior prevalência de hipertensão (65% p = 0,011) e obesidade (64,7% p=0,041) nos antecedentes familiares do grupo Obeso quando comparado ao grupo Eutrófico. Os adolescentes obesos apresentaram maiores valores de triglicerídeos, HDL, APO B, CETP, insulina e LDL(-) e LDLox, quando comparados aos eutróficos. Perfil inverso foi observado para Apo AI. O conteúdo de LDLox e LD(-) variou significativamente em função do IMC. Entretanto, essas partículas de LDLs não se correlacionaram entre si, embora tenham apresentado associação com outros parâmetros cardiometabólicos. Os resultados obtidos confirmam o impacto negativo da obesidade sobre os parâmetros cardiometabólicos de adolescentes e, apesar do conteúdo de LDLox e LDL(-) ter aumentado em função do IMC, essas partículas parecem ser estruturalmente distintas. Essa possibilidade foi reforçada pelas diferentes associações dessas partículas com outros marcadores bioquímicos.

▼ 7-cetocoleterol (7KC) é um oxisterol conhecido por inibir a proliferação celular e por ser citotóxico. Desenvolvemos uma nanoemulsão contendo 7KC (LDE/7KC) que demonstrou efeito anti-proliferativo sobre as linhagens RPMI 8226 (mieloma) e melanoma (B16F10), in vitro, sendo preferencialmente captada via receptores de LDL. No presente trabalho avaliamos, in vivo, a cinética plasmática, biodistribuição, ação anti-tumoral e parâmetros tóxico-hematológicos em camundongos portadores de melanoma. A cinética plasmática apresentou um decaimento estatisticamente igual entre os animais portadores de melanoma e não portadores. Em relação à biodistribuição da nanoemulsão, houve um acúmulo de seus componentes radioativamente marcados, principalmente no fígado e no tumor, sugerindo sua captação via receptores de LDL. LDE/7KC promoveu uma redução superior a cinqüenta por cento do tamanho do tumor, que apresentou maior área de necrose e menor quantidade de vasos. Nos camundongos tratados com LDE/7KC houve um aumento da sobrevida. As análises toxico-hematológicas demonstraram que a nanoemulsão apresentou pouca ou nenhuma toxicidade. Os resultados demonstram a possibilidade da utilização da nanoemulsão LDE/7KC como um agente no tratamento do câncer, com poucos efeitos colaterais, devido a sua seletividade aos receptores da LDL.
7-ketocholesterol (7KC) is an oxysterol known to inhibit cell proliferation and to be cytotoxic. A nanoemulsion containing-7KC (LDE/7KC) was shown to have antiproliferative effects on RPMI 8226 myeloma cell line and melanoma (B16F10), in vitro. This particle is taken up mainly by LDL receptors. Here we have evaluated the plasma kinetic, biodistribution, anti-tumoral action and hematologic toxicity of LDE/7KC in melanoma bearing mice. The nanoemulsion accumulated in the liver and tumor, tissues with a high expression of LDL receptors. LDE/7KC promoted a tumor size reduction over fifty percent. An increased necrosis area and a decreased amount of blood vessels were found. An increased survival rate was observed. The hematolgic analyses demonstrated a lack of toxicicity. The results shows the possibility to use the LDE/7KC nanoemulsion as an agent for cancer treatment, with few collateral effects probably due to its internalization by LDL receptors.

▼ Proprotein Convertase Subtilisin / Kexin Type-9 (PCSK9) has emerged as a major regulator of plasma cholesterol levels. PCSK9 is secreted mainly from the liver and circulates as a plasma protein. PCSK9 binds cell surface low-density lipoprotein (LDL) receptors and mediates their degradation upon endocytosis in the liver. This decreases the liver’s ability to clear LDL-cholesterol from the blood. PCSK9 is also capable of binding LDL particles themselves; this interaction inhibits the ability of PCSK9 to bind and mediate LDLR degradation in cultured hepatic cells, but its effect on PCSK9 function in vivo remains unknown. A disordered N-terminal region of the PCSK9 prodomain is necessary for binding to isolated LDL particles in vitro. This N-terminal region is also autoinhibitory to PCSK9-LDL receptor binding. We hypothesized that the N-terminal of the PCSK9 prodomain plays a role in an allosteric mechanism that regulates PCSK9 function. Through mutagenesis studies, we found that both a conserved stretch of acidic residues and an adjacent conserved stretch of hydrophobic residues are crucial for the PCSK9-LDL interaction; the hydrophobicity of the residue at position 38 (Tyr) within the conserved acidic stretch was also found to be important for this. Helical wheel modeling of the prodomain N-terminal sequence revealed the potential for a lipid-ordered amphipathic helix to form, which may aid PCSK9 docking onto LDL. Replacing residues A44 and L41 with helix-disrupting proline residues abolished LDL binding. Co-pelleting ultracentrifugation assays also show that wild-type PCSK9 is capable of associating with liposomes, while the A44P mutation disrupts this lipid association. The A44P-PCSK9 mutation, showing an 80-90% decrease in LDL association but with LDL receptor binding and degrading functions intact, may serve as an important tool in future studies investigating the PCSK9-LDL interaction in vivo. Elucidation of the mechanism by which LDL-binding naturally inhibits PCSK9 activity may also help to develop new anti-PCSK9 therapeutics in the future.

▼ The role of cell death in atherosclerosis remains ill-defined, however, a growing body of evidence suggests that cell death stimulates atherogenesis through the induction of inflammation and enlargement of the necrotic core. Although there is solid evidence to suggest that lipid oxidation and toxicity are linked, indications that protein oxidation may play an important role in cytotoxicity are numerous. The abundance of dead cells in atherosclerotic plaques and their co-localization with HOCl-modified proteins provides an opening for the suggestion that the products of protein oxidation may be at the heart of oxLDL-induced cell death. Examination of the modification of LDL and albumin by HOCl, and the cytotoxicity of these oxidised molecules were the focus of this study, along with the elucidation of their cell death mechanisms toward U937 cells.
Measurement of lipid peroxidation markers, TBARS and 7-ketocholesterol, showed no significant increase in HOCl-oxLDL compared to native levels although all α-tocopherol had been lost. In contrast there was a large loss of tyrosine, of which a small percentage went to dityrosine, indicating that the protein moiety of LDL was the main target of HOCl attack. Albumin became fragmented and smeared on SDS-PAGE gels with increasing HOCl/BSA molar ratios. In addition there was significant reduction in tyrosine levels and a small increase in dityrosine.
Both HOCl-oxLDL and oxidised albumin (oxALB) caused concentration-dependent cell viability loss in U937 cells following a significant drop in intracellular GSH concentration, coinciding with a peak in oxidative stress. Removal of chloramines with methionine significantly reduced the toxicity of oxALB, but at higher concentrations this effect was reduced. This was in contrast to HOCl-oxLDL where the removal of chloramines had no effect on its toxicity. Morphological observations of cell swelling, cell membrane integrity loss and rupture, along with flow cytometry results indicate that U937 cells underwent necrosis, but only after intracellular GSH was lost. Intracellular GSH and cell viability loss were prevented by 200 μM extracellular 7,8-dihydroneopterin (78NP), indicating that 78NP scavenging of ROS generated in response to the oxidised proteins was sufficient to prevent cell death.
This study demonstrates the cytotoxicity of HOCl-damaged LDL and albumin is likely due to a common oxidative product or structural motif which may be active within atherosclerotic plaques.

▼ Atherosclerotic cardiovascular disease is the leading cause of death of adults in North America. Diabetes is a classical risk factor for atherosclerotic cardiovascular disease. Plasminogen activator inhibitor-1 (PAI-1) is the major physiological inhibitor of fibrinolysis. Elevated levels of PAI-1, oxidized low-density lipoprotein (oxLDL) and glycated
LDL (glyLDL) were detected in patients with diabetes. Increased oxidative stress is associated with diabetic cardiovascular complications. Previous studies in our laboratory demonstrated that oxLDL or glyLDL increased the production of PAI-1 or reactive oxygen species (ROS) in vascular endothelial cells (EC). This study was undertaken to investigate transmembrane signaling mechanisms involved in oxLDL or glyLDL-induced upregulation of PAI-1 in cultured vascular EC. Further, we examined the mechanism for oxLDL or glyLDL-induced oxidative stress in EC.
The results of the present studies demonstrated novel transmembrane signaling pathway for oxLDL-induced PAI-1 production in vascular EC. We demonstrated that lectin-like oxLDL receptor-1, H-Ras, a small G-protein and Raf-1/ERK-1/2 mediate oxLDL-induced PAI-1 expression in cultured EC.
GlyLDL may activate EC via a distinct transmembrane signaling pathway. The results of the present study demonstrated that receptor for advanced glycation end products, NADPH oxidase and H-Ras/Raf-1 are implicated in the upregulation of heat shock factor-1 or PAI-1 in vascular EC under diabetes-associated metabolic stress.
We investigated the effects of oxLDL or glyLDL on mitochondrial function in EC. Treatment with oxLDL or glyLDL significantly impaired the activities of electron transport chain (ETC) enzymes and also increased mitochondria-associated ROS in EC. The findings suggest that oxLDL or glyLDL attenuated activity of ETC and increased ROS generation in EC, which potentially contributes to oxidative stress in vasculature.
In conclusion, diabetes-associated lipoproteins may upregulate stress response mediators and PAI-1 production via distinct transmembrane signaling pathways. OxLDL or glyLDL may increase ROS production via NOX activation and the impairment of mitochondrial ETC enzyme activity in EC. The understanding and identification of the regulatory mechanisms involved in diabetes-associated lipoprotein-induced signaling may help pharmacological design for the management of diabetic cardiovascular complications.

LDL oxidado. Objetivos: avaliar o papel da dieta rica em flavonoides no estresse oxidativo, na resposta inflamatória e no perfil lipídico de pacientes com doença cardiovascular submetidos a implante de stent coronariano. Métodos: trinta e três pacientes submetidos à angioplastia coronária com implante de stent foram randomizados no grupo dieta controle, com base nas recomendações do NCEP III e dieta antioxidante (AOX), rica em alimentos fontes de flavonoides, por 6 meses. As informações referentes à ingestão alimentar foram coletadas no início e término do seguimento, assim como, os níveis de estresse oxidativo (FRAP e sulfidril) e marcadores inflamatórios (Proteína C-reativa). Resultados: A orientação nutricional resultou numa redução da ingestão energética, de carboidratos e de lipídios no grupo dieta AOX, e um aumento na ingestão de flavonoides. Os marcadores de estresse oxidativo, FRAP e sulfidril e a proteína C-reativa não foram alterados no grupo dieta AOX, porém, os níveis de
LDL-colesterol apresentaram redução significativa quando comparados com o grupo controle. Conclusão: os resultados sugerem que uma dieta rica em alimentos antioxidantes à base de flavonoides, não está associada com a redução do estresse oxidativo e do perfil inflamatório em pacientes submetidos à angioplastia coronariana com implante de stent, a despeito de uma melhora no perfil lipídico destes pacientes.

▼ Atherosclerotic-related cardiovascular disease (ASCVD) claims the lives of over 600,000 Americans yearly. Current methodologies of assessment do not distinguish lipoprotein density distributions and instead measure lipoprotein cholesterol, with lowdensity lipoprotein cholesterol (
LDL-C) and high-density lipoprotein cholesterol (HDLC) of primary clinical relevance.
Furthermore, the distinction between the effects of diet and exercise interventions on lipoproteins is frequently indiscernible due to the effects of energy deficit induced by interventions. High- performance lipoprotein density profiling (HPLDP) provides a cost-effective way to rapidly assess the efficacy of diet or exercise interventions. The purpose of this study is to characterize density distribution of HDL subclasses in response to diet or exercise using HPLDP.
Eighty-eight untrained subjects (74% male, average age of 54 years) were pooled from two lifestyle intervention studies that met the inclusion criteria. Following 12 weeks of diet or exercise interventions (750 kcal expenditure or 750 kcal deficit), subjects lost an average of 3.72 kg weight (-3.9%), lost 3.9 kg of body fat (-11.6%), increased lean mass 0.62 kg (+1.1%), and reduced body fat percentage by 3.41% (-9.1%). Average absolute VO2 max increased 0.16 liters O2/min (+7.1%) (p<0.05). Several lipoprotein density distributions were significantly different (p<0.05) between diet and exercise interventions (expressed as percent change from baseline): triglyceride-rich lipoproteins (TRL) (-17.14 vs 19.65), low-density lipoprotein subfraction 5 (LDL5) (-26.06 vs 8.14), high-density lipoprotein subfraction 3b (HDL3b) (-21.24 vs -0.71), and high-density lipoprotein subfraction 3c (HDL3c) (-17.88 vs 7.94). To elucidate the effects of cardiorespiratory fitness, subjects were further divided into categories of increased absolute VO2 max and decreased absolute VO2 max, with no changes between groups at baseline. Associations between absolute VO2 max percent change on TRL,
LDL-5, HDL-3b, and HDL-3c percent change remained significant after controlling for age, gender, and fat mass percent change. A regression equation was constructed from significant correlations and effectively predicted HDL-3c changes using absolute VO2 max measurements.
A significant linear relationship between improved absolute VO2 max and increased HDL-3c subfraction AUC exists; lipoprotein subfraction quantification may reveal positive effects of exercise overlooked using traditional clinical cholesterol assessment techniques.

Endothelfunktion. An Abgängen, Verzweigungen und Verengungen von Blutgefäßen
wirken geringere Scherkräfte und der Blutfluss verlangsamt sich, infolgedessen
treten an diesen Stellen vermehrt atherosklerotische Plauqes auf.
Verschiedene Krankheiten wie Diabetes, chronische Niereninsuffizienz und
Übergewicht gehen mit erhöhtem Blutcholesterolspiegel einher. Patienten haben
durch die Krankheit vermehrt modifizierte Lipoproteine im Blut.
Modifikationsprozesse von LDL stehen mit ersten Anzeichen der Atherosklerose in
Verbindung.
Humane koronare arterielle Endothelzellen (HCAEC) werden häufig im
Forschungsbereich der Atherosklerose, insbesondere der endothelialen Dysfunktion
eingesetzt, um die Wirkung von modifizierten LDLs und die dabei resultierende
Schädigung der Endothelzellen herauszufinden.
HDL und das weibliche Hormon Östrogen wirken Schädigungen entgegen und
werden als vasoprotektiv beschrieben.
Anhand von Toxizitätstests konnte die Lipotoxizität mit steigender Konzentration an
modifizierten LDL auf Endothelzellen gezeigt werden. Die Vermutung konnte
bekräftigt werden, dass HDL und 17-beta Estradiol (E2) den toxischen Effekt in
Zellkulturexperimenten vermindern konnten.
Frühere Studien befassten sich mit der Regulation von inflammatorischen Markern
und der Regulierung von Scavenger Rezeptoren, den sogenannten Fressrezeptoren.
Bisher ist wenig über die Regulation von LDL Rezeptor Familienmitgliedern im
vaskulären Endothelium bekannt.
Das Ziel meiner Diplomarbeit war, die Rolle von LDL Rezeptor Familienmitgliedern,
wie LDL Rezeptor, LRP1 und LRP2, in diesem Zusammenhang zu definieren.
Hierbei sollten HCAEC mit verschiedenen modifizierten LDLs (oxidiertem, glykiertem
und carbamyliertem LDL) behandelt werden. Weiters sollten geschlechtsspezifische
Unterschiede erfasst werden, indem Zellen zusätzlich zu modifizierten LDL mit E2
behandelt wurden.
Die Expressionsanalysen wurden unter statischen Bedingungen und unter
Einwirkung von Scherkräften in Zellkartuschen (Fiber Cell™ Model) durchgeführt.
Zusammenfassend konnte gezeigt werden, dass der Transkriptionslevel von LDL
Rezeptoren unter statischen Bedingungen und unter geringen Scherkräften
verringert wird. Erhöhter Blutfluss und Scherkräfte wirken aktiv auf Rezeptoren der
Endothelzellen. LRP2 mRNA konnte nur mit geringem Expressionstatus
nachgewiesen werden. LRP1 konnte auf Transkriptionsebene in
Zellkulturexperimenten nicht nachgewiesen werden, aber eine basale Aktivierung
unter Einfluss von Scherkräften.
In Zellkulturexperimenten zeigte die Inkubation von Endothelzellen mit E2 und
modifizierten LDL, einen geringen Antsieg von LRP1 und LRP2 auf mRNA Ebene.

▼ Les maladies cardiovasculaires représentent la première cause de mortalité en France avec une prévalence encore plus importante à La Réunion. La plupart des décès sont attribuables aux cardiopathies ischémiques due à la rupture d'une plaque d'athérosclérose et à la formation d'un caillot entrainant une ischémie cardiaque. L'oxydation des lipoprotéines de basse densité (LDL) et la dysfonction de l'endothélium représentent des étapes importantes dans la déstabilisation de la plaque, d'où l’intérêt de rechercher un traitement capable de diminuer l'oxydation des LDL ou de protéger les cellules endothéliales de leurs effets cytotoxiques. Les polyphénols sont des molécules antioxydantes très présentes dans le règne végétal. Des études préliminaires du laboratoire sur des adipocytes humain on montré que des extraits riches en polyphénols d'Antirhea borbonica et de Doratoxylon apetalum, deux espèces respectivement endémique et indigène de l'Île de La Réunion, possédaient des propriétés antioxydantes et anti-inflammatoires (Marimoutou, M et al. 2015). Le But de cette thèse était, dans un premier temps, de tester les capacités antioxydantes et cytoprotectrices d'extraits riches en polyphénols de ces deux espèces sur des cellules endothéliales. Puis dans un second temps nous avons testé l'extrait de Doratoxylon apetalum sur un modèle murin d'athérosclérose (souris ApoE KO) pour évaluer son effet anti-inflammatoire sur les plaques d'athérome.
Cardiovascular diseases are the leading cause of death in France with an even higher prevalence in Reunion. Most deaths are due to ischemic heart disease because of atherosclerotic plaque rupture and thrombus formation leading to cardiac ischemia. The oxidation of low density lipoproteins (LDL) and endothelial dysfunction represent important steps in the destabilization of plaque, hence the interest of a treatment capable of reducing the oxidation of LDL or of protecting endothelial cells from their cytotoxic effects. Polyphenols are antioxidant molecules very present in the plant kingdom. Preliminary studies on human adipocytes have shown that polyphenol-rich extracts of Antirhea borbonica and Doratoxylon apetalum, respectively endemic and indigenous species of Reunion Island, had antioxidant and anti-inflammatory properties (Marimoutou , M et al., 2015). The aim of this thesis was, first, to test the antioxidant and cytoprotective capacities of polyphenol rich extracts of these two species on endothelial cells. Then in a second time we tested the extract of Doratoxylon apetalum on a murine model of atherosclerosis (ApoE KO mice) to evaluate its anti-inflammatory effect on atheroma plaques.

▼ Bevezetés: Az
LDL és HDL lipoproteinek fontos szerepet töltenek be az érelmeszesedés folyamatában. Ezen partikulumok nemcsak koleszterintartalmuk alapján jellemezhetők, hanem fizikai és kémiai tulajdonságukban, apolipoprotein és lipid tartalmukban, élettani szerepükben is különbözőek, heterogén szubfrakciókra oszthatók. Heterogenitásuk jellemzésére többféle analitikai módszer áll rendelkezésre.
Célkitűzés: Diplomamunkámban célul tűztem ki különböző szérum minták stabilitásának vizsgálatát alacsony hőmérsékleten történő, hosszú távú mintatárolás során, valamint különböző kardiovaszkuláris megbetegedésben szenvedő betegek lipoprotein szubfrakciós profiljának elemzését és egészséges személyek adataival való összehasonlítását.
Módszerek: A szérum minták lipoprotein szubfrakcióinak elkülönítése méretük alapján üvegcsöves akrilamid gélelektroforézissel, Lipoprint® rendszerrel történt. Az
LDL teszt esetében 7 különböző
LDL szubfrakciót választunk el. HDL teszt esetében 10 különböző HDL szubfrakció elválasztására van lehetőségünk.
Eredmények: 4 személytől vettünk mintát és a szérum mintából elválasztott lipoprotein szubfrakciók mintastabilitását vizsgáltuk a 0., 7. és 30. napon -70°C-on tárolt mintákból.
LDL szubfrakciók elválasztása során az egyes sávok között 0-3,2% közötti eltérést, a HDL szubfrakciók elválasztása esetében 0,08-2,33% közötti eltérést detektáltunk a 0. napon kapott eredményekhez képest. Vizsgálataink második felében egészséges személyek lipoprotein szubfrakciós profilját vetettük össze elhízott, ill. frissen diagnosztizált hiperlipidémiás személyek adataival, amely során a betegekben a kis, denz
LDL szubfrakció, ill. a kisméretű HDL szubfrakció nagyobb arányú jelenlétét figyeltük meg az egészséges személyekhez viszonyítva.
Következtetések: 30 napig történő, alacsony hőmérsékleten való mintatárolás során a szérumban nem léptek fel a lipoprotein szubfrakciós profil megváltozását előidéző érdemi változások. Az adatok pontosítása érdekében a méréseket érdemes kiterjeszteni hosszabb ideig tárolt mintákra is. A betegek szubfrakciós profiljában megfigyelhető eltérések fontos információkat szolgáltatnak a későbbi kardiovaszkuláris komplikációk és az érelmeszesedés folyamatának jellemzésére.

▼ Every blood vessel in the body is lined on its luminal aspect with a single layer of interconnected cells, known as endothelial cells. Together, these cells form a tissue known as the endothelium, a tightly regulated system that physically segregates the fluid in the blood from the peripheral tissues, and orchestrates the passage of nutrients, xenobiotics, immune cells, and fluids between distinct compartments in the body. In healthy, quiescent vasculature, passage of compounds across the vasculature occurs by two separate, yet equally important mechanisms. In the paracellular route, compounds that are small enough to pass between endothelial cell junctions freely diffuse down their concentration gradient, reaching equilibrium between the blood and sub-endothelial compartments. In contrast, transcytosis involves the regulated transit of compounds through endothelial cells by endocytosis, subcellular trafficking, and exocytosis, and in doing so, allows for the transport of larger compounds through the use of a number of specialized proteins. Under normal circumstances, these two mechanisms of endothelial permeability work in concert to maintain homeostasis. However it has recently become apparent that dysregulated endothelial permeability is the cause of a number of pathological conditions. Increased paracellular permeability of the lung endothelium leads to the acute respiratory distress syndrome (ARDS), a potentially fatal condition involving acute lung edema, respiratory failure, and death. Endothelial transcytosis of low-density lipoprotein (
LDL) leads to the accumulation of
LDL in the subendothelial intima, the initiating event in atherosclerotic lesion development. Although dysregulated endothelial permeability is central to the pathophysiology of these diseases, the paucity of literature on the
subject highlights the need for a detailed, mechanistic investigation.
In this thesis I present a body of work that uncovers several novel mechanisms that regulate endothelial permeability in the pathological contexts of pathogen-induced ARDS and
LDL transcytosis as a precursor to atherosclerotic lesion development. Results from these studies suggest that targeting endothelial permeability is novel and viable therapeutic strategy for management of these common diseases.

▼ Arsenic exposure in humans has been implicated in the development of a myriad of non-cancerous and cancerous diseases. A reductionist approach to understanding this unusual phenomenon would suggest that arsenic-induced perturbation of a small number of fundamental biological processes could manifest as this diverse array of disease endpoints. Endocytosis is a fundamental cellular process involved in the internalization and transport of various extracellular molecules and membranous components. BEAS-2B, a human bronchial epithelial cell line, was used to characterize the effects of chronic arsenite exposure on endocytosis. Fluorophore-labeled bovine albumin, human transferrin, and human low-density lipoprotein (
LDL) were the substrates utilized to measure endocytosis in BEAS-2B cells. The uptake of albumin in unexposed BEAS-2B cells is both dose-dependent and temperature sensitive. Chronic arsenite exposure in BEAS-2B cells increased the uptake of albumin by 3.4-fold after 8 hours of uptake relative to unexposed BEAS-2B cells. Pharmacological studies utilizing endocytosis inhibitors suggested that the uptake of albumin in both unexposed and arsenite-exposed BEAS-2B cells occurs through a combination of receptor-mediated endocytosis and macropinocytosis. Chronic arsenite exposure in BEAS-2B cells also increased the endocytic uptake of transferrin by 2.9-fold at 30 minutes and
LDL by 1.3-fold at 2 hours relative to unexposed BEAS-2B cells. Together, the data suggests that chronic arsenite exposure can increase the rate of endocytosis. This novel finding could add mechanistic insight to the conundrum of arsenic-associated human diseases.

▼ Atherosclerosis is characterised by inflammation, arterial stiffening and aberrant lipid transport by deposition of low-density lipoproteins (LDL) in the artery wall that is susceptible to oxidation. Oxidised LDL is taken up by endothelial cells, which can lead to impaired vasorelaxation. There is strong evidence for the role of myeloperoxidase (MPO), an inflammatory enzyme released by neutrophils, in oxidative damage to biomolecules such as LDL in atherosclerosis. MPO produces the oxidants HOCl and HOSCN. While it is well established that HOCl causes extensive LDL modification, less is known regarding HOSCN in atherosclerosis, which may be of relevance to smokers whom have significantly elevated plasma levels of the MPO substrate thiocyanate (SCN-), which can shift MPO to favour HOSCN production. Therefore, modifications of LDL by HOSCN may be a contributing factor to the deleterious effects of smoking in the development of atherosclerosis. This study examined modification of the LDL protein apoB-100 by HOSCN and found it selectively cleaved the N- and C-termini by free-radical mediated reaction. However, studies into the specific mechanism and determining the cleavage sites are still required. The role of HOSCN- or HOCl-modified LDL in inducing vascular cell dysfunction was also investigated. Exposing these modified LDL to endothelial cells did not induce inflammatory adhesion molecule or cytokine expression, unfolded protein response gene expression, or alter neutrophil adhesion. However, HOSCN- and HOCl-modified LDL did significantly impair nitric oxide (NO) production by uncoupling endothelial NO synthase (eNOS). This supported functional assessment of vasorelaxation by wire myography in rat aortic segments exposed to HOSCN- or HOCl-modified LDL ex vivo, which had impaired endothelium-dependent vasodilation. In cultured vascular smooth muscle cells (VSMC) exposed to these modified LDL, the gene expression of contractile markers α-smooth muscle actin and calponin were stunted, while the synthetic marker osteopontin was elevated. This is consistent with promotion of a synthetic VSMC phenotype. The data from this study support a role for HOSCN as well as HOCl in promoting vascular dysfunction by modification of LDL, which is an important mechanism in promoting atherosclerosis.

▼ Cholesterol is an essential mammalian membrane component. In mammalian cells, cholesterol homeostasis needs to be tightly regulated since the aberrant intracellular cholesterol transport and storage are associated with many human diseases, including cardiovascular disease, Alzheimers disease, as well as a rare inherited neurodegenerative disorder, Niemann-Pick disease type C (NPC). One crucial cholesterol regulatory pathway involves endosomal sorting of low density lipoprotein (LDL)-derived cholesterol to the endoplasmic reticulum (ER), where cholesterol levels are sensed to exert a feedback regulation. However, the exact mechanisms underlying the transport of LDL-derived cholesterol from late endosome/lysosome (LE/Ly) to the ER remain unclear. This project identifies Hrs and Vps4 as novel regulators of endosomal cholesterol transport. Being part of, or associated with, the endosomal sorting complex required for transport (ESCRT) machinery, Hrs and Vps4 are two important proteins involved in the endosomal multivesicular body formation pathway. In this project, we show in HeLa cells that depletion of Hrs or Vps4 by RNAi results in cholesterol accumulation in LE/Ly, a cellular phenotype reminiscent of NPC disease caused by mutations in either NPC1 or NPC2 proteins. Endosomal transport of LDL-derived cholesterol to the ER is blocked in Vps4 knockdown or Hrs knockdown cells. Interestingly, the expression and localisation of NPC1 and NPC2 are not compromised upon Vps4 depletion. Other ESCRT subunits, including Tsg101 (ESCRT-I), EAP20 (ESCRT-II) and all ESCRT-III components investigated in this project do not have similar effects to those of Vps4 and Hrs, suggesting a novel role of these two ESCRT-associated molecules in endosomal cholesterol trafficking. This study provides mechanistic insights into how cholesterol is sorted out of LE/Ly, which may help understand the pathogenesis of cholesterol metabolism-related diseases.

▼ Introdução: O Diabetes mellitus (DM) está associado a complicações crônicas micro e macrovasculares. A medida da hemoglobina glicada (HbA1c) avalia o grau do controle glicêmico em pacientes diabéticos e seus níveis são capazes de prognosticar o risco de desenvolvimento dessas complicações. Entre as origens das complicações causadas pela hiperglicemia, há a hipótese dos produtos finais de glicação avançada (AGEs) e do estresse oxidativo. A reação de glicação não enzimática das proteínas também está relacionada com as complicações diabéticas e é responsável pela formação da HbA1c. Entretanto, tem sido demonstrado um aumento dessa glicação em pacientes não diabéticos. Um dos prováveis mecanismos para esse aumento é a peroxidação lipídica, com conseqüente aumento nos níveis de malondialdeído (MDA) que modifica a apolipoproteína B (APO B) do colesterol de baixa densidade (
LDL). A modificação oxidativa do
LDL confere propriedades específicas pró-aterogênicas. A presença do
LDL oxidado e o aumento da tendência do
LDL à peroxidação lipídica, podem contribuir para o aumento dos níveis de HbA1c. Objetivo: Verificar a associação entre os níveis de HbA1c com o Colesterol
LDL e
LDL oxidado em Indivíduos não-diabéticos. Métodos: Foi realizado um estudo transversal observacional, no qual um total de 196 indivíduos, classificados como não-diabéticos, foram analisados e divididos em três grupos, conforme os valores de HbA1c e glicemia de jejum (GJ): Grupo 1 (n =64) - HbA1c <5,7% e GJ <100 mg/dL; Grupo 2 (n =69) - HbA1c ≥5,7 e ≤6,4% e GJ <100 mg/dL; Grupo 3 (n =63) - HbA1c ≥5,7 e ≤6,4% e GJ ≥100 e <126mg/dL. Amostras de sangue total e soro foram coletadas. O

▼ Introdução: A doença cardiovascular é a principal causa de morbimortalidade precoce em todo o mundo, e responde por grande parte dos gastos dos recursos destinados aos programas de políticas públicas. Neste contexto, a dieta representa uma importante ferramenta na redução dos fatores de risco cardiovasculares. Tendo em vista que inúmeros estudos mostram que o consumo de ômega 9 ou alimento fonte modifica positivamente diversos fatores de risco cardiovascular clássicos, se torna importante avaliar seu efeito sobre propriedades físico-químicas da LDL e da HDL, marcadores cardiometabólicos e oxidativos em indivíduos brasileiros com diferentes níveis de risco cardiovascular. Objetivo: Avaliar o efeito do consumo de azeite de oliva sobre parâmetros cardiometabólicos clássicos e novos em indivíduos com diferentes níveis de risco cardiovascular. Métodos: O estudo foi do tipo clínico prospectivo, aleatorizado, placebo controlado, duplo cego baseado em intervenção nutricional. Indivíduos de ambos os sexos, distribuídos em grupos azeite de oliva (AO) e placebo (PL) receberam durante 8 semanas 3 g/d de azeite de oliva ou placebo. Todos os indivíduos foram classificados quanto ao risco cardiovascular, seguindo os critérios estabelecidos pelo Escore de Risco de Framingham (ERF). Nos momentos basal, T=4S e T=8S foram determinados o perfil clínico, antecedentes familiares de doenças, pressão arterial, consumo alimentar e nível de atividade física. A partir do plasma ou soro, obtidos após 12 h de jejum, foram determinados o perfil lipídico, as apolipoproteínas, o tamanho da HDL e da LDL, o conteúdo de LDL(-) e de NEFAS e atividade da paraoxonase. A aderência à intervenção foi monitorada por meios diretos (marcadores bioquímicos) e indiretos (registro de intercorrências). Resultados: O azeite de oliva foi efetivo em reduzir concentração de triacilglicerois dos indivíduos em alto risco cardiovascular (p=0,023 no T=4S e p=0,049 no T=8S) e a de LDL-C dos indivíduos com risco cardiovascular intermediário (p=0,045 no T=8S) no atual estudo. Observou-se também redução significativa na LDL(-), quando a amostra foi estratificada pelo ERF. Demais parâmetros permaneceram inalterados em função do tempo da intervenção e do ERF. Conclusão: Baixa suplementação (3 g/d) de azeite de oliva promoveu redução dos triacilglicerois, LDL-C e da LDL(-). Portanto, recomenda-se a incorporação de azeite de oliva na dieta brasileira ainda que em baixas doses. Sugere-se também que estudos adicionais usando doses maiores sejam realizados no sentido de identificar potenciais benefícios cardioprotetores adicionais associados ao consumo de azeite de oliva.

Avaliamos os perfis lipídico e de Ac anti-oxLDL e anti-pepD (pepD é um peptídeo derivado da apoB). Animais apoE-/- possuem maiores níveis de lípides sorológicos em comparação com animais C57Bl/6, exceção da HDL. A imunização oxLDL não alterou este perfil. A imunização aumentou os títulos dos Ac IgM, IgG e IgG1 anti-oxLDL. Animais C57Bl/6, mas não os apoE-/- tiveram aumento dos Ac IgM anti-pepD após a imunização. Acreditamos que o aumento de anticorpos IgG1 deve-se ao adjuvante usado. A aterosclerose pode ocorrer com maior freqüência, em indivíduos portadores de doenças renais crônicas. Para estudar essa interferência submetemos os animais a um modelo de isquemia e reperfusão renal (I/R). A I/R promoveu aumento da concentração de TG, podendo agravar a aterosclerose. O aumento de Ac IgG anti-oxLDL promovidos pela I/R sugere que o processo inflamatório desencadeado por este procedimento, aumenta a oxidação de LDL. Desta forma, concluímos que a I/R pode ser considerado um procedimento pró-aterosclerótico que não pode ser revertido pela imunização com oxLDL.
Oxidized LDL (oxLDL) antibodies (Ab) are involved in the development of the atherosclerotic lesion. We induced the formation of such Ab through immunization with oxLDL. We evaluate the lipids and antibody (anti-oxLDL and anti-pepD pep D is a peptide derived from apolipoprotein B) profile. The apoE-/- mice used have higher seric levels of most lipids than C57Bl/6 animals, the exception being HDL. The oxLDL immunization does not change this profile. The immunization increased anti-oxLDL Ab e IgM, IgG and IgG1 titers. Anti-pepD IgM Ab increased with immunization in C57Bl/6 animals only, and not in apoE-/-. We believe the increased IgG1 Ab due to the adjuvant used. Atherosclerosis has a higher incidence in individuals with chronic renal conditions. In order to study such interference, we submitted the animals to a renal ischemia and reperfusion model (I/R). The I/R procedure increased TG concentration, what can make atherosclerosis more severe. The increase in IgG anti-oxLDL antibody caused by I/R suggests that the inflammation process set by the procedure increases LDL oxidation. This way, we conclude that the I/R can be considered a pro-atherosclerotic procedure. The oxLDL immunization was not able to revert the atherogenic effect caused by I/R model.

▼ Em trabalhos anteriores, mostrou-se que uma nanoemulsão lipídica denomi-nada LDE após injeção endovenosa, em pacientes com carcinoma mamário e outros tumores sólidos concentra-se nos tecidos neoplásicos e pode direcionar especificamente agentes quimioterápicos ao tumor. Estudos clínicos mostraram que a LDE diminui acentuadamente os efeitos tóxicos desses agentes e em estudos com animais de experimentação não reduz seus efeitos antitumorais. No presente estudo, testamos a hipótese de a LDE injetada por via locorregional poderia concentrar-se no tumor de mama e nos linfonodos axilares da mama comprometida, visando futuras aplica-ções do sistema na quimioterapia neoadjuvante desse tumor. Três técnicas de injeção da LDE foram testadas em pacientes com carcinoma de mama avançado com tratamento cirúrgico pré-programado. A LDE marcada com colesterol radioativo foi injetada 12 horas antes da cirurgia, nas pacientes divididas em três grupos: Grupo 1 (G1, n=4): LDE injetada no parênquima mamário, a 5 cm da lesão ; Grupo 2 (G2, n=4): LDE injetada na região peri-tumoral; e Grupo 3 (G3 n=6): LDE injetada em região intratumoral. Este grupo foi subdividido em 2: em 2 pacientes realizaram cirurgia 2hs após a injeção da LDE e nas outras 4, 12hs após a injeção da LDE. Quantificou-se a capta-ção da LDE nos fragmentos de tecido tumoral e mamário normal e do linfonodo axilar retirados durante a cirurgia por contagem de radioatividade após extração lipídica dos tecidos. Os resultados evidenciaram que, em G1, houve maior captação da LDE em tecido normal, sugerindo se tratar de metodologia ina-dequada. Em G2, a captação da LDE foi quatro vezes maior no tecido tumo-ral do que no tecido normal, próximo, portanto, dos valores encontrados em estudos anteriores que utilizaram a injeção da LDE por via hematológica. Em G3, o valor médio de captação da LDE foi 53 vezes maior no tecido tumoral (>75%), com mínima captação pelo tecido mamário normal (<8%). A injeção intratumoral da LDE mostrou-se, portanto, metodologia ainda superior à inje-ção da LDE por via endovenosa para a utilização efetiva dessa nanoemulsão como terapia-alvo em tumores primários de mama.
In previous work had showed that a lipidic nanoemulsion called LDE after intravenous injection in patients with breast carcinoma and others solid tumors focuses in the neoplasic tissue and can target especially chemotherapist agents to the tumors. ▼ O efeito do fotossensibilizador na estrutura biológica não é apenas influenciado por suas propriedades fotofísicas, mas também por sua interação específica com biosistemas.Além disso, a localização do fotossensibilizador no tecido tumoral é um importante fator que resulta em diferentes mecanismos de destruição do tumor. Muitos fotossensibilizadores, após administração sistêmica, se ligam às proteínas plasmáticas e com isso são distribuídos em diferentes sítios no organismo. Os fotossensibilizadores hidrofílicos são largamente transportados por albuminas e globilinas e se acumulam preferencialmente no estroma vascular dos tumores. Entretanto, fotossensibilizadores mais hidrofóbicos se ligam às lipoproteínas, principalmente LDL, que promove a entrada do FS na célula através de endocitose mediado por receptor. Sendo assim, a localização do FS depende de sua ligação com as deferentes proteínas plasmáticas, sua farmacocinética e também é influenciada pela diferença entre o tecido normal e tumoral. O tecido tumoral tem pH mais baixo e maior expressão de receptores de LDL do que os tecidos normais, aumentando a seletividade dos FSs as células tumorais. A incorporação de FS hidrofóbicos em lipossomas para a administração sistêmica pode realçar ao transporte deste pelas lipoproteínas. No presente trabalho estudou-se a influência do pH na interação de fotossensibilizadores com lipossomas de DMPC e LDL. Os fotossensibilizadores utilizados nesse estudo foram Photofrin®, Photogem® e Photosan® que são derivados de hematoporfirinas. A metodologia empregada constitui de variação das concentrações de DMPC e LDL para os seguintes valores de pHs 5,0; 7,4 e 9,0, esse último pH utilizou-se somente para DMPC. O complexo FS - DMPC foi obtido por incubação dos FSs na concentração de 10 micro g.mL-1 com diferentes concentrações de DMPC (0 a 400 micro M) por trinta minutos no escuro. Isolou-se o LDL do plasma humano por ultracentrifugação por gradiente de densidade. Após a separação, o complexo FS - LDL foi obtido por incubação (12 horas no escuro) do FS na concentração 10 micro g.mL-1 com diferentes concentrações de LDL (0 a 0,04 micro M). O comportamento desses complexos foi analisado por espectroscopia de absorção ótica e por espectroscopia de fluorescência.
The effect of a photosensitizing compound on biological structures is governed not only by its photophysical properties but also by the specificity of its interaction with biosystems. ▼ Background: Oxidized low-density lipoprotein (oxLDL) has been shown to hold an important role in atherosclerosis initiation and progression. We investigated the association of circulating oxLDL to coronary and peripheral atherosclerosis severity among patients with recently diagnosed or suspected CAD.Methods: The study comprised 196 patients (91.8% men), all ≤ 65 year-old without comorbidities, who had their first coronary angiogram after an acute coronary syndrome (85.7%) or for recently diagnosed or suspected stable CAD (14.3%). The number of major coronary arteries with stenosis ≥50% (CS) and Hamsten coronary atherosclerosis score (HS) were used as indices of CAD severity and extent respectively. Arterial B-mode ultrasound was performed bilaterally to measure common carotid (CC) and common femoral (CF) maximal intima-media thickness (IMT) and maximal plaque thickness (PT). A full lipid panel was acquired and hs-CRP was measured. Circulating oxLDL was measured with a monoclonal antibody-based (4E6) enzyme-linked immunosorbent assay. Pearson’s and Spearman correlations, linear and logistic regression models and ROC curves were used for statistical analysis.Results: OxLDL had a significant positive correlation with total cholesterol (r=0.506, p <0.001), LDL (r=0.496, p <0.001), nonHDL (r=0.505, p <0.001) and apo B-100 (r=0.476, p <0.001) but not with other lipid parameters. Furthermore oxLDL correlated significantly and negatively with age (r=-0.213, p=0.004) and positively with body mass index (r=0.165, p=0.030). Considering the negative correlation of oxLDL with age and the cut-off point of 55 years that defines premature CAD in men further analysis focused on male patients. For the male population of the study (180 of 196 patients) a weak but significant relationship was found between oxLDL and CS (r=0.169, p=0.033) but not between oxLDL and HS (r=0.124, p=0.130). OxLDL did not correlate significantly with the carotid and femoral atherosclerosis indices used and no other lipid parameter or hs-CRP did correlate significantly with any of the coronary or peripheral atherosclerosis indices. OxLDL was significantly higher among men ≤ 55 year-old (n=102, mean age: 48±5 years) versus men > 55 year-old (n=78, mean age: 60±3 years): 99±46 U/L vs 84±32 U/L respectively, p=0.023. A) Among patients ≤55 year-old oxLDL significantly correlated with both CAD scores (CS: r=0.243, p=0.021 and HS: r=0.244, p=0.024). Those in the highest (≥107 U/L) compared to those in the lowest (<77 U/L) oxLDL concentration tertile, had higher CAD scores (CS: 1.9±0.9 vs 1.3±1, p=0.012 & HS: 2.6±1.7 vs 1.6±1.1, p=0.01) and more often multi-vessel CAD (in 70% vs 34.5% of cases respectively, p=0.009). They were also more likely to have multi-vessel CAD, after adjustment for risk factors, age, body mass index, statin treatment, CIMT and apoB-100 (adjusted OR: 3.146, 95% CI: 1.220-8.112, p=0.018). Furthermore, at ROC curve analysis oxLDL was found to be a significant albeit modest predictor of multi-vessel CAD (AUC=0.652, 95% CI: 0.539-0.765 p=0.013).…

▼ Abstract: Background/Objectives: Plasma level of low-density lipoprotein cholesterol (
LDL-C) has become a major marker and the central preventive target for cardiovascular disease in a large spectrum of clinical settings. However, recent reports have raised questions about the consistency of this concept in individuals aged of 80 years or older. Hence, the aim of this study was to evaluate in a carefully selected population if the association between lipid parameters and coronary atherosclerotic burden remains significant. Design, Setting, Participants, Measurements: Individuals aged of 80 years or older (n=208) who spontaneously sought primary prevention care and have never manifested cardiovascular disease, malnutrition, neoplastic or consumptive disease were enrolled for a cross-sectional analysis. Medical evaluation, anthropometric measurements, blood tests and cardiac computed tomography were obtained. Results: 1- In analyses adjusted for age, gender, diabetes, systolic and diastolic blood pressure, smoking and statin therapy no association was found between coronary calcium score (CCS) and
LDL-C (1.79 (0.75-4.29)). There was no association between TG and CCS. The association between HDL-C and CCS was significant and robust in unadjusted (0.32 (0.15-0.67)) as well as in the fully adjusted analysis (0.34 (0.15-0.75)). Results: 2-All the coronary segments (>3.000) were assessed to the presence of coronary atherosclerotic plaques. Coronary atherosclerotic plaques by CCTA were classified according to previous studies: positive remodeling (PR), not calcified plaques (NCP) and spotty calcification (SC). Individuals were categorized in the presence or absence of PR, NCP and SC and highest and lowest to the last quartile of the lipid profile (HDL-C (? 63 vs < 63mg/Dl),
LDL-C ? 138 vs < 138 mg/dL, TG (?156 vs <156 mg/dL)). Results-In adjusted analysis by age, gender, body fat, hypertension, creatinine, CRP, diabetes mellitus, smoking, physical activity and use of statin all type of plaques with instability phenotype were associated with age. No association was found between LDL_C or TG and atherosclerotic plaques. Association remains between remodeling plaques and HDL-C< 63 mg/dL in fully adjusted model (2.5(1.1,-6.3);0.040) Conclusion: The present study confirms in a healthy cohort of individuals aged of 80 years or more that while the association between

▼ Muitos estudos têm revelado que a oxidação da lipoproteína de baixa densidade (LDL) é um fator pró-aterogênico e que o estresse oxidativo tem um papel central na formação de LDL oxidada. As Tiossemicarbazonas são compostos usados em terapias anti-câncer, antiviral e antifúngica em humanos. No entanto, estudos sobre a atividade redox desta classe de moléculas ainda são bastante controversos. O objetivo deste trabalho foi avaliar a atividade antioxidante de um composto derivado de uma tiossemicarbazona: a isatina-3-N4-benziltiossemicarbazona (IBTC) e seu possível efeito protetor contra a oxidação da LDL in vitro. Tais atividades foram avaliadas através da formação de dienos conjugados em LDL e em soro humano induzidos por 2,2-azobis(2-amidinopropano diidroclorido) AAPH e por CuSO4; da perda da fluorescência do triptofano (Trp) em LDL induzida pelo CuSO4; da formação de substâncias reativas ao ácido tiobarbitúrico (TBARS) em LDL, plasma humano e em diferentes tecidos de ratos e do potencial antioxidante total (TRAP) do IBTC. A toxicidade do IBTC foi investigada em fatias da artéria aorta. Os resultados mostraram que a IBTC foi eficaz em reduzir a formação de dienos conjugados induzidos tanto pelo AAPH quanto pelo CuSO4, aumentando de modo concentração-dependente a fase lag. A IBTC também preveniu a perda acentuada da fluorescência do Trp, reduziu a formação de TBARS na LDL no plasma e nos tecidos isolados de ratos e não causou toxicidade em fatias da artéria aorta. Esses dados indicam que a IBTC pode ser considerado um promissor agente antioxidante e exibir propriedades anti-aterogênicas in vitro.
Several lines of evidence support the hypotheses that the oxidation of low density lipoprotein (LDL) is a proatherogenic factor. Oxidative stress is one of the causes of the overproduction of reactive species that increase the formation of oxidized LDL. Thiosemicarbazones are compounds used in anticancer, antiviral and antifungal therapy; however, their redox activities have been controversial. Thus, we tested, in vitro, the possible antioxidant activity of a thiosemicarbazone derivate, the isatin-3-N4-benzilthiosemicarbazone (IBTC). We measured the conjugated diene formation in serum and LDL induced by two oxidant agents, 2,2-azobis(2-amidinopropane dihydrochloride) (AAPH) and Cu2+, as well as the loss of tryptophan fluorescence in LDL induced by Cu2+. The total reactive antioxidant potential (TRAP), Thiobarbituric acid reactive substances (TBARS) formation in human LDL and plasma and in different rat tissues was also assessed. The toxicity of IBTC was measured using aortic slices viability assay. The results show that IBTC reduced the formation of conjugated dienes AAPH and Cu2+- induced increased in a dose-dependent manner the lag phase. IBTC also prevented the tryptophan fluorescence loss, and reduced the TBARS formation in LDL, plasma and rat tissues, showing no toxicity to aortic slices. These data indicate that IBTC is a good antioxidant and a promising antiatherogenic agent for further studies in vivo.

▼ Com a intensificação do controle glicêmico e maior sobrevida dos pacientes com diabetes mellitus tipo 1 (DM1), houve mudança progressiva das causas de mortalidade, com maior destaque para a doença cardiovascular (DCV). Em função da LDL oxidada ser considerada ponto fundamental na patogênese da doença aterosclerótica, o presente estudo objetivou avaliar as absorbâncias obtidas nos diferentes comprimentos de onda dos fosfolipídios oxidados e o coeficiente de oxidação da partícula de LDL in vitro em pacientes com diabetes mellitus tipo 1. Foram avaliados 54 pacientes com DM1 com idade de 29,1 12,11 e de duração de doença de 14,6 8,43 e 28 voluntários não diabéticos. Os pacientes e voluntários foram submetidos à avaliação clínica e laboratorial onde foram mensurados perfil lipídico, glicêmico, ácido úrico, níveis pressóricos, excreção urinária de albumina e análise espectofotométrica da oxidação do LDL colesterol antes e 1, 3, 6 e 24 horas após adição de substância oxidante- sulfato de cobre. Os coeficientes de oxidação (Cox) do LDL dos dois grupos apresentaram leitura semelhante em todos os momentos com diferença significativa entre o tempo basal e após 1h (p<0, 001) e entre o momento após 3 e 6 horas após adição de sulfato de cobre ( p< 0,001) para os grupos avaliados. No modelo de regressão múltipla, o resultado obtido foi o modelo Cox = 8,246 13,93a -2,71b + 10,21c (R2=0,835), onde a, b e c representam as absorbâncias (205 nm, 232 nm e 280 nm, respectivamente). Concluímos que em nossa amostra os comprimentos de onda 232 nm e 280 nm referentes à leitura dos dienos conjugados e a fração protéica da LDL, respectivamente apresentaram maior importância na determinação do coeficiente de oxidação da partícula.
With the intensive glycemic control and longer survival of type 1 diabetes mellitus (T1DM) patients, cardiovascular disease has been the main cause of mortality. Oxidized LDL is considered a critical factor in the atherosclerotic process and the aim of our study was evaluate the absorbance of different wavelengths of oxidized phospholipids and oxidation rate of LDL particles in vitro in T1DM. We studied 54 patients aged 29.1 12.11 years and with a disease duration of 14.6 8.43 years and 28 non-diabetic volunteers. The two groups had carried where they were measured lipid profile, glucose, uric acid, blood pressure, urinary albumin excretion and spectrophotometry analysis of the oxidation of LDL cholesterol before and 1, 3, 6 and 24 hours after addition of substance oxidant copper sulfate. The coefficients of oxidation (Cox) of LDL from both groups had similar reading at all times with a significant difference between the baseline and after 1 hour (p <0 .001) and between the time after 3 and 6 hours after addition of sulfate copper (p <0 .001) for both groups. ▼ Dietary guidance recommends a healthy dietary pattern that is low in saturated fat (SFA) to lower low-density lipoprotein cholesterol (
LDL-C), a primary target for cardiovascular disease (CVD) risk reduction. Using either carbohydrates (CHO) or unsaturated fat to replace saturated fat in an average American diet may achieve a similar reduction in
LDL-C, but a high intake of dietary CHO may increase triglycerides (TG), lower high density lipoprotein cholesterol (HDL-C), and increase small, dense
LDL, along with promoting insulin resistance thereby increasing the risk of metabolic syndrome and type 2 diabetes. These cardio-metabolic risk factors also are targets for dietary intervention to achieve a greater CVD risk reduction. When SFA is replaced with unsaturated fat, the predominant fatty acid class is monounsaturated fatty acids (MUFA) because the recommended intake of polyunsaturated fatty acids (PUFA) is less than 10% of energy. Overall, the substitution of MUFA instead of CHO (especially refine grains and added sugar) for SFA calories may favorably affect other lipid risk factors of CVD.
The oxidative modification of
LDL particles is a key factor in the initial process of atherosclerosis. Dietary antioxidant vitamins, polyphenols, and other bioactive compounds from foods (e.g., fruits and vegetables) have been a focus of nutrition because of their role in improving antioxidant status and lowering
LDL oxidation. Identifying foods that can beneficially improve multiple cardio-metabolic risk factors is needed for the primary prevention of CVD.
Avocados are a nutrient-dense source of MUFA, rich in vitamins, minerals, fiber, phytosterols and polyphenols. It is unclear whether avocados affect cardio-metabolic risk factors beyond their fatty acid profile. This dissertation investigated the effects of a MUFA enriched moderate fat diets including one avocado per day on established and novel cardio-metabolic risk factors.
A randomized, cross-over, controlled feeding trial was conducted with 45 healthy overweight/obese participants with baseline
LDL-C levels in the 25-90th percentile. After a 2 week run-in average American diet (34% fat, 13% SFA, 11% MUFA, 51% carbohydrate, 16% protein) at baseline, three cholesterol-lowering diets (6-7% SFA) were fed (5 weeks each) with a random sequence: a lower-fat diet (LF: 24% fat, 7% SFA, 11% MUFA, 9% PUFA, 59% carbohydrate, 16-17% protein); and two moderate fat diets matched for macronutrients and fatty acids (34% fat, 6% SFA, 17% MUFA, 9% PUFA, 49% carbohydrate, 16-17% protein): the avocado diet included one fresh Hass avocado (136 g) per day, and the moderate fat diet provided mostly the same foods (as the avocado diet) but used high oleic acid oils and low fat dairy products to match the fatty acid profile of one avocado. O colesterol encontra-se em todas as células do organismo humano. Se, por um lado, é vital, por exemplo, para a construção de novas células, por outro, pode acumular-se no nosso organismo, levando à deposição de gordura na parede das artérias. Encontra-se bem fundamentada a relação entre o aumento drástico de mortes por doença cardiovascular, nos países desenvolvidos, e o elevado nível de colesterol que se verifica em parte substancial da sua população.
Sendo uma substância lipídica, necessita de transportadores específicos para circular no plasma, as lipoproteínas. Estas partículas designam-se de acordo com a sua densidade. As duas mais importantes são as lipoproteínas de alta densidade (HDL) e as lipoproteínas de baixa densidade (LDL). As primeiras têm como principal função remover o excesso de colesterol para o fígado. Pelo contrário, as segundas transportam o colesterol do fígado para as células.
Este trabalho visa estudar a problemática da hipercolesterolemia, na população infantil e adolescente. Neste segmento populacional, esta condição assume especial importância, sobretudo quando se verifica a conjugação de altos níveis de LDL colesterol com baixos níveis de HDL colesterol, pelas consequências na saúde a longo prazo. O aumento muito significativo da hipercolesterolemia relaciona-se intimamente com o estilo de vida actual, que contempla diversos factores de risco, a serem analisados com detalhe ao longo do trabalho.
Pela análise de uma grande variedade de dados estatísticos, procurou caracterizar-se cada um destes factores, no sentido de avaliar a sua efectiva contribuição para a prevalência da hipercolesterolemia, em crianças e jovens, e verificar a existência de possíveis correlações entre eles, que pudessem contribuir de forma decisiva para o aumento do risco. Cholesterol is found in all cells of the human organism. If, on the one hand, it is vital, for example, to build new cells, on the other hand, it can also accumulate in the body, leading to fat deposition in artery walls. There is a well-substantiated relationship between the dramatic increase in deaths caused by cardiovascular disease in developed countries and the high level of cholesterol occurring in a substantial share of its population.
As a lipid substance, it requires specific transporters making it possible to circulate in plasma, designated lipoproteins. These particles are classified according to their density. The two most important are the high-density lipoproteins (HDL) and the low-density lipoproteins (LDL). The former are mainly responsible for removing the excess of cholesterol to the liver. On the contrary, the latter carry cholesterol from the liver to the cells.

▼ Drei Vertreter der low-density Lipoprotein Rezeptor Familie, LDLR selbst, very-LDLR, und LDLR related protein (LRP) werden von minor group Rhinoviren des Menschen (HRVs) als Rezeptoren verwendet. Während ICAM-1, der Rezeptor der major group der HRVs, aktiv am viralen uncoating beteiligt ist, wurden minor group Rezeptoren als passive Vehikel betrachtet, die das Virus in Endosomen transportieren, wo sie ein Milieu mit niedrigem pH vorfinden. Für die YWTD β-Propeller Domaine des LDL-Rezeptors wurde jedoch gezeigt, dass sie an der Abdissoziation gebundenen LDLs bei niedrigem pH beteiligt ist. Wir wollten daher
herausfinden, ob sie auch aktiv an der Infektion mit HRVs teilnimmt. Es existiert keine humane Zell Linie, die LDLR in Abwesenheit von VLDLR oder LRP exprimiert. Daher wurden in unseren Untersuchungen CHO-ldla7 Zellen verwendet, die keinen endogenen LDL-Rezeptor exprimieren. Diese waren stabil transfiziert um entweder Wildtyp oder β-Propeller negative humane LDL-Rezeptoren zu exprimieren. Mit Hilfe dieser Zellen
untersuchten wir den möglichen Einfluss des β-Propellers auf die Konversion von HRV2 in subvirale Partikel als eine Funktion des niedrigen pH und die Freisetzung des Virus vom Rezeptor. Wenn HRV2 an propeller-negativen LDLR gebunden war ein niedrigerer pH wurde für die Konversion benötigt. Dies deutet darauf hin, dass die hoch-avide Rezeptorbindung das Virus in seiner nativen Konformation stabilisiert. Infektionsversuche zeigten eine zeitliche Verzögerung in der intrazellulären viralen Konversion, im Abbau und in der viralen de novo Synthese in der Abwesenheit der β-Propeller-Domaine. Wir schließen daraus, dass die Funktion des β-Propellers bei niedrigem pH auch dann signifikant ist, wenn der Ligand ein Rhinovirus ist. Er erleichtert die virale Konversion im Inneren von Endosomen, indem er den
Virus-stabilisierenden Effekt des mit hoher Avidität bindenden LDLR reduziert. Dadurch begünstigt er die Infektion.
Three members of the low-density lipoprotein receptor (LDLR) family, LDLR proper, very-LDLR (VLDLR) and LDLR-related protein (LRP) act as minor group human rhinovirus (HRV) receptors. Whereas ICAM-1, the receptor of the major group of HRVs, actively contributes to viral uncoating, minor group receptors were rather considered passive vehicles just delivering virus to the low pH environment of endosomes. However, the YWTDβ-propeller domain of LDLR has been shown to be involved in the dissociation of bound LDL at acidic pH. We thus aimed to clarify whether it also actively contributes to HRV infection. No human cell line expressing LDLR in the absence of VLDLR or LRP is available. Therefore, CHO-ldla7 cells which are deficient in endogenous LDLR and are stably transfected to express either wild-type or β-propeller negative human LDLR were used to investigate low pH-triggered conversion of HRV2 and its release from the receptor.
