▼ Four experiments were conducted to investigate methods of producing transgenic bovine embryos entirely in vitro. Experiment 1 examined the effect of DNA microinjection at 11, 15 and 19 h after fertilization (haf) on survival rate and DNA detection frequency by polymerase chain reaction (PCR). There was no difference in transgene detection frequency between treatments (53% at 11; 50% at 15; 48% at 19 haf). Of all injected embryos developing to the morula or blastocyst stage after 7 d in culture, 89% tested positive for the presence of the transgene by PCR. Greater developmental efficiencies can be obtained when injection is performed early in pronuclear formation (7% (11/161) at 11; 4% (61159) at 15; 1 % (1/165) at 19 haf; p<0.05). Experiment 2 examined the effect of microinjection of DNA into the germinal vesicle (gv) of bovine oocytes on subsequent development and detection of the transgene. Injection of the transgene into the gv reduced developmental rates compared to controls (control=23% (89/384); non-injected=9% (23/250); GV injected=5% (12/259); p<O.05). Transgene detection frequency was 64% (37/58). Injection of bovine oocytes before fertilization results in viable embryos containing the transgene, although at low frequencies. Experiment 3 was designed to examine whether the frequency of microinjected DNA detection by peR In whole bovine embryos would decline over a 21 d culture period. At d 0, the transgene was detected in 100% (46/46) of embryos analyzed. At d 7, detection frequency was 84% (51/62) in viable embryos, at d 14 49% (18/37), and at d 21 38% (3/8). DNA detection frequency in microinjected bovine embryos by PCR analysis does not give a reliable indication of live transgenic birth rates until after 14 d in culture. Experiment 4 examined microinjected bovine embryos for their potential use as donor embryos in nuclear transfer, or cloning. There was no difference in development between embryos cloned from microinjected donor embryos and those from control donor embryos (injected=11 % (37/377); control=9% (7/81); p>0.05). Of the embryos developing from microinjected donors, 32% (12/37) were PCR positive. Microinjected embryos can be successfully used in a nuclear transfer program to produce more viable embryos, and the resulting embryos may be more reliably screened by PCR. The efficiency of producing viable bovine embryos positive for the injected gene may be increased by performing microinjection early in pronuclear formation, and entering the resulting embryos into a nuclear transfer program.

▼ In this research effort, we consider the linear complementarity problem (LCP) that arises in diverse areas including optimal control, economics, engineering, mechanics, and quadratic programming. This class of problems has posed a challenge to researchers for over three decades now. Most of the current algorithms designed to solve LCP are guaranteed to work only under some restrictive assumptions on the matrix <i>M</i> associated with LCP. In this research, we introduce two new algorithms based on an equivalent 0-1 mixed integer bilinear programming formulation of LCP.
In the first approach, we develop an enhanced intersection cutting plane algorithm for solving LCP. The matrix <i>M</i> is not assumed to possess any special structure, except that the corresponding feasible region is assumed to be bounded. A procedure is described to generate cuts that are deeper versions of Tuy's intersection cuts, based on a relaxation of the usual polar set. The proposed algorithm then attempts to find an LCP solution in the process of generating either a single or a pair of such strengthened intersection cuts. The process of generating these cuts involves a vertex ranking scheme that either finds an LCP solution, or else, these cuts eliminate the entire feasible region leading to the conclusion that no LCP solution exists. Based on the bilinear formulation, a heuristic is also proposed to front-end the algorithm, in order to possibly solve LCP.
In the second part of the dissertation, we present a global optimization algorithm based on a novel Reformulation-Linearization Technique (RLT). We do not place any restrictions on the matrix M associated with LCP in this case. This RLT scheme provides an equivalent linear, mixed integer programming formulation of LCP, that possesses a tight linear programming relaxation. The solution strategy developed is a composite impliCit enumeration-Lagrangian relaxation scheme. In addition to the bounds provided by the RLT-based relaxation, we further tighten these bounds at each node of the branch-and-bound tree through the use of strongest surrogate cuts, and strengthened intersection cuts. The heuristic developed in the previous algorithm is also invoked within this scheme.
Both algorithms have been implemented and tested on randomly generated test problems. These problems include both indefinite and negative definite defining matrices. The results of these tests indicate that both methods are effective in solving the LCP with the heuristic being quite effective in recovering LCP solutions when the matrix Mis negative definite. The Lagrangian dual approach for the implicit enumeration scheme proved to be computationally more efficient than a similar simplex based lower bounding scheme.

▼ Research was conducted that culminated in the merging of the objectives of two tools: predetermined motion time systems and posture recording. This dissertation reports the development and testing of a computerized tool-the Work and Posture Analysis Sequence Technique (WAPAST)-used to collect methods, postural, and work measurement data. From the data collected, one can determine the method used to complete an activity by task identification, the time it takes to complete each task, as well as the time to complete an entire activity, and the postural considerations for each task and for the overall activity.
The tool is based on the Maynard Operations Sequence Technique (MOST) with some characteristics of the Ovako Working Posture Analysis
System (OWAS). Criteria established were speed of completion, accuracy, and reliability. A validation study was conducted to assess the data collected with WAPAST against data collected with known, validated tools. The study was an experiment that had several subjects use WAPAST to analyze videotaped work activities while several other subjects used MOST or OW AS to analyze the same videotaped activities. The data collected with WAPAST were compared with the data collected with MOST and OWAS. The work measurement data collected with WAPAST were found to be both accurate and reliable, but the postural data were not. Times to cornplete applications were extensive.
Recommendations are given for improving the tool, which include a different way of recording postural data and expanding the limitations of the tool.

▼ Numerous examples of non-linear survival curves, plotted as log surviving cells vs. time, for bacteria exposed to heat have been reported. Factors which may affect the shape of a survival curve and the heat resistance of bacteria include temperature, pH, and NaCl concentration. Many studies have examined the effect of these factors individually, but little information exists on the combined effects. The objective of this study was to mathematically model non-linear survival curves to account for these factors and their interactions simultaneously.
Heat resistance of Listeria monocytogenes(L. monocytogenes) was determined in O.lM KH₂P0₄ buffer and in infant formula at three temperatures (50, 55, and 60 C), three pH levels (5, 6, and 7), and three NaCl concentrations (0, 2, 4%). Survival curves were fit using linear regression, non-linear regression with a modified logistic equation, and non-linear regression with a modified Gompertz equation.

▼ Bisphenol A polycarbonate is an engineering thermosplastic which can be utilized for a variety of applications. Modifications to improve upon the properties of this useful material have been attempted since its invention. Three modifications of bisphenol A polycarbonate and its copolymers have been examined in this work.
The hydrolytic stability of copolymers of polydimethylsiloxane and polycarbonate was improved by the synthesis of pre-formed phenol functional polydimethylsiloxane oligomers in which the Si is attached directly to the aromatic ring of the hydroxyaryl functional component. <i>In situ</i> random block copolymers of polydimethylsiloxane and polycarbonate were synthesized by interfacial polymerization techniques. The Si-aryl bonds of the novel polydimethylsiloxane oligomers were found to impart good thermal and hydrolytiC stability to the random block copolymers.
Terminally reactive polycarbonate oligomers were also synthesized in order to form polycarbonate network materials. 4-Acetoxystyrene was utilized as a stable precursor to 4-vinylphenol which could then be used to quantitatvely control the molecular weight and functionality of the polycarbonate chains. The reactive oligomers were found to undergo a thermal cure above the respective glass transition temperatures to form insoluble networks. These network materials were found to have superior solvent resistance as compared to the linear material while retaining much of the useful mechanical properties (high fracture toughness, etc.) of bisphenol A polycarbonate. Furthermore, the glass transition temperatures of the networks were found to increase with a decrease in the molecular weight between crosslinks.
The fire resistance properties of polycarbonate were modified by the copolymerization of bisphenol A with phenol derivatives of triphenylphosphine oxide. The effect of changes in the composition were determined by differential scanning calorimetry and thermogravimetric analysis. The glass transition temperature, as well as the residual char yield at elevated temperatures was found to increase with an increase in the phosphorus monomer content as measured by the two respective methods. Cone calorimetry was also performed on the samples to determine the heat release rate, which reflected an increase in the fire resistance properties with an increase in the phosphorus content.

▼ Site-directed mutagenesis and gene replacement procedures were used to isolate mutant strains of <i>Azotobacter vinelandii</i> that produce altered MoFe proteins where the α-subunit residue-195 position, normally occupied by a histidine residue, was individually substituted by a variety of other amino acids. Structural studies have revealed that this histidine residue is associated with the FeMo-cofactor binding domain and probably provides an NH→S hydrogen bond to a central bridging sulfide located within FeMo-cofactor. The present study investigates the role of the α-histidine-195 residue in nitrogenase catalysis by examining the altered MoFe proteins.
Comparisons of the catalytic and spectroscopic properties of altered MoFe proteins produced by the <i>Azotobacter vinelandii</i> mutant strains suggest that the α-histidine-195 residue has a structural role which serves to keep the FeMo-cofactor attached to the MoFe protein and to correctly position the FeMo-cofactor within the polypeptide matrix such that N₂ binding is accommodated. Substitution of the α-His-195 residue by a glutamine residue results in an altered MoFe protein that binds but does not reduce N₂, the physiological substrate. Stopped-flow spectroscopic analyses indicate that the α-195
gln MoFe protein is unable to reduce N₂ even though the altered MoFe protein can reach the redox state necessary for N₂ reduction. Although, N₂ is not a substrate for the altered MoFe protein, it is an inhibitor of both acetylene and proton reduction, both of which are otherwise effectively reduced by the altered MoFe protein. This result provides evidence that N₂ inhibits proton and acetylene reduction by simple occupancy of the active site. The α-195
gln MoFe protein catalyzes HD formation in the presence of N₂ and D₂. Moreover, N₂ binding at the active site of the altered MoFe protein is inhibited by the addition of D₂. These observations indicate that binding of nitrogen to the enzyme is necessary but its reduction is not required for the formation of HD. N₂ uncouples MgATP from proton reduction catalyzed by the α-195gln MoFe protein, but does so without lowering the overall rate of MgA TP hydrolysis. Thus, the quasi-unidirectional flow of electrons from the Fe protein to the MoFe protein that occurs during nitrogenase turnover is controlled, in part, by the substrate serving as an effective electron sink. N₂-induced uncoupling of ATP hydrolysis from substrate reduction by the α-195
gln MoFe protein is reversed by the addition of H₂ (D₂) in the assay atmosphere. This observation can successfully be explained if it-is assumed that the altered MoFe protein has a much greater binding affinity for H₂ (D₂) than for N₂. Substitution of the α-histidie-195 residue by glutamine also imparts hypersensitivity of acetylene reduction and N2 binding to inhibition by CO, indicating that the imidazole group of the α-histidine- 195 residue might protect an Fe contained within FeMo-cofactor from attack by CO.

However, the experimental data alone do not provide enough information about the behavior of the anchorage to be used exclusively in the design process. The results are used to modify the design of the anchorage system. It is shown that for a successful anchorage system the choice of material for the load transfer medium is crucial. A solution is presented to overcome the high stress concentration at the load entry area of the wire into the anchor.
A finite element analysis of the anchors for 1 and for 19 wires is used to assess the stress and strain fields inside the anchor, to validate the analytical model, and to determine locations of possible high stress concentrations. Three-dimensional and one dimensional models, that utilize axisymmetry, are evaluated. The results of the numerical analysis are used to demonstrate the improvement as a result of a change in material choice or design of the anchor. It is shown that the modification of the load transfer medium results in a decrease of 30 % of the average stress level.

adsorption filter. No significant change in total solids, total fat, and solids-not-fat percentages of skim and whole milk permeates was observed after filtration. Adsorption of Pseudomonas fluorescens at target concentrations of 103 , 102 , and 101 cells/ml was determined in 100ml of dairy standard methods buffer, nutrient broth, whole milk, and skim milk. The average percentage bacterial retentions were
95 ± 5.5%, 95 ± 2.6%, 28 ± 22.1%, and 62 ± 15.5%, respectively.
A treatment was developed for milk to increase the bacterial retention of ~ fluorescens after filtration. The preferred treatment for 100ml of skim milk involved the
following final concentrations (v/v): 0.80% disodium ethylenediamine-tetraacetic acid, 0.02% sodium dodecyl sulfate, pH to 7.5 with 1N sodium hydroxide. The average
bacterial retention of ~ fluorescens using the treatment was 91 ± 7.1%. Enumeration of bacteria adsorbed to the filter was then conducted using impedance microbiology. When milk was inoculated with ~ fluorescens at target concentrations of 103 , 102 , and 101 cells/ml, an average log bacterial increase of 1.4 ± 0.1 (25x) was obtained. This method will allow for rapid detection of microorganisms in milk by increasing
microbial load in the tested sample and eliminating the need for pre-enrichment.

▼ Effects of fat adaptation and NaHC0₃ supplementation on acid-base homeostasis were quantitated during repeated sprint exercise in horses. Contribution of strong ions ([SID]), Pco₂, and weak electrolytes ([A
tot]) to changes in plasma [H⁺] and the role of erythrocytes in acid-base balance were examined at rest and during exercise. Effects on plasma glucose and blood lactate [Lac⁻] concentrations due to sample handling were also assessed.
During exercise, blood [Lac⁻] was higher when horses received NaHC0₃ compared to water prior to exercise (P=0.0024), and in fat adapted horses compared to horses fed a control diet (P=0.0240). Blood [Lac-] was higher in fat adapted horses given NaHC0₃ compared to other diet/treatment combinations (P=0.0276). Plasma [SID] was higher during exercise when horses were given NaHC0₃ compared to plain water (P=0.0054), which contributed to decreasing [H⁺] and increasing [HC0₃⁻] during exercise (P=0.0001). Plasma Pco₂ contributed less to increasing plasma [H⁺] during exercise in fat-adapted horses compared to horses fed the control diet (P=0.0282). Intraerythrocyte [SID] decreased (P=0.0160) and [Atod increased (P=0.0002) which contributed to increasing [H⁺] within the cell (P=0.0228).

▼ Seed priming is a controlled hydration process, followed by dehydration, that allows pregerminative metabolic activity to proceed without germination. The objective of this research was to investigate the effects of priming on intrinsic characteristics of seed germination including temperature, water, and development, in order to understand how priming affects the germination of broccoli (<i>Brassica oleracea var. italica</i> Plenck) seeds. Priming of broccoli seeds consistently improved germination and emergence rate in the laboratory and field and was related to the accumulation of a specific level of hydropriming units expressed in MPa*h. Priming reduced the sensitivity of seed germination to temperature and increased the temperature range of germination but did not lower the minimum temperature for germination. Primed seeds leaked less electrolytes at supraoptimal temperatures (≥ 35°C) compared to nonprimed seeds. In the field, primed seeds produced a greater plant stand and yield under stressful emergence conditions. Under optimal conditions in the field for stand establishment, the advancement in emergence of primed seeds did not carry over to earlier, greater yields. Matric priming, using calcium silicate as the carrier in the ratio 1.0:0.8:1.8 (seed:carrier:water; by weight) for 7 d at 20°C, was superior to osmotic priming using polyethylene glycol (pEG 8000) at -1.2 :MPa in nearly all variables examined. This may be attributed to reduced respiration during priming of seeds in PEG or nutrient uptake by seeds in calcium silicate.
The testa was observed to be a barrier to broccoli seed germination. Priming-induced changes to the physical characteristics of broccoli seeds included increased volume (32%) and an irreversibly expanded, and weakened testa with some minute cracking near the area where the radicle emerges. Primed seeds germinated faster, in part by maintaining a lower hydrotime constant, and thus exhibited a greater progression towards germination per unit water potential at a constant temperature compared with nonprimed seeds. It was hypothesized that, since the testa threshold was reduced after priming by expansion and formation of free spaces, the reversibly expanded embryo of primed seeds does not become immediately appressed to the testa upon rehydration. Thus the yield threshold component controlling the rate of germination of primed seeds is lower by the amount of the testa threshold. The priming effect is more than just reducing the yield threshold as indicated by a significant invigoration of seeds with split testae. Despite the increased volume as a result of the formation of free spaces, primed broccoli seeds did not imbibe more water or have a greater turgor at full hydration. Priming did not lower the minimum water potential allowing germination, and primed seeds did not plateau in water uptake but, instead, moved immediately from imbibition to expansive growth. ▼ Geomagnetically induced currents (GIC) could saturate a transformer core. Two significant effects are the abnormal stray flux in transformers and extremely large harmonic contents in excitation currents, which can lead to serious equipment damage and power system misoperation. Such incidents have occurred during the March 1989 K-9 solar magnetic disturbance.
This dissertation starts with a systematically study of transformer magnetization under GIC. It reviews both dc and ac saturation patterns of five transformer core designs. Magnetic fields along various traverses for dc excitation are presented. Impedance matrix entries of a single phase transformer are compared for normal and dc operations. New observations have been formed based on the simulation results. The study helps to reveal the fundamental transformer magnetization mechanism under GIC in order to assess potential stray flux heating possibilities of geologically vulnerable transformer units.
Based on the finite element analysis, an improved method of modeling transformer excitation under dc bias using equivalent magnetic circuit is developed. There are two unique points in this approach: first, information of 3D finite element magnetic flux distribution analysis is used to construct and verify the circuit model; second, the effect of the transformer tank: is included The model is capable of simulating transformer excitation currents under different levels of dc bias with good accuracy. As a consequence, the complete variations of excitation current harmonics with respect to an extended range of dc bias are revealed. The sensitivity of transformer winding impedances and core loss on the excitation characteristics are examined. The saturated transformer under no-load and various loading conditions is simulated. A laboratory test is performed on a small scale transformer and compared with the model results. Excitation harmonics generated from dc biased three phase transformer banks with different types of equivalent loads are also simulated The effect of both unbalanced dc excitations and unbalanced loads are investigated The results of this study contribute in understanding transformers as harmonic sources and the impact on power systems during a geomagnetic disturbance.

▼ This work is concerned with the processing of pellets of polypropylene (PP) containing pregenerated microfibrils of thermotropic liquid crystal polymers (TLCPs), referred to as microcomposites. The processing methods used are injection molding, sheet extrusion, and extrusion blow molding. The TLCPs used are HX6000 and Vectra A950. The microcomposites are produced by drawing strands of PP and TLCPs generated by means of a novel mixing technique and pelletizing the strands. The work was undertaken in an effort to improve on the properties observed for in situ composites in which the TLCP fibrils are generated in elongational flow fields that occur during processing. In situ composites usually exhibit highly anisotropic mechanical properties and the properties do not reflect the full reinforcing potential of the TLCP fibers. Factors considered include the effect of in situ composite strand properties on the properties of the injection molded composite, the melt temperature used in injection molding, TLCP concentration, and the melt temperature of the TLCP. It was found in this work that microcomposites can be processed by means of injection molding, sheet extrusion, and extrusion blow molding. It was necessary to process the materials at low temperatures to maintain the TLCP fibrils. However, HX6000, the higher melting TLCP allowed higher processing temperatures than Vectra A. When the TLCP fibrils were maintained, the properties of the TLCP reinforced composites did show reduced anisotropy as compared to an in situ composite. The tensile strength of the composites produced by all three methods was about equal. The modulus of the injection molded composites was slightly higher than that of the composite sheets, but the composite sheets showed a lower degree of anisotropy. In all three processing methods the modulus of the TLCP reinforced composite was a function of the modulus of the in situ composite strand used to produce the microcomposite. Therefore, it is recommended that to improve the properties of the microcomposites the properties of the in situ composite strands should be improved. Furthermore, the mechanical properties of the composites increased with increasing TLCP composition.
To provide a basis of comparison the properties of the extruded sheets and the injection molded composites were compared to both the predictions of composite theory and the properties of glass reinforced composites. It was found that the modulus of the 10 wt% composites approached the predictions of composite theory, but at higher TLCP loadings the modulus showed negative deviations from the predictions of composite theory. This is believed to be the result of a reduction of fiber aspect ratio due to poor fiber distribution and fiber breakup. The modulus of the TLCP reinforced composites was about the same as the modulus of the glass reinforced composites produced by both sheet extrusion and injection molding. ▼ Increasing individual's physical activity in our country is a primary health promotion and disease prevention concern. Yet, much of the population remains sedentary and thus, at increased risk for several diseases such as hypertension, coronary heart disease and hyperlipidemia. Unfortunately, programs promoting physical activity have had limited success in increasing activity and even less success in maintaining a program over time.
The "Exercise for Everyone" program was designed to assess the effectiveness of behavior change strategies developed for maintenance on physical activity using individuals recruited through a community setting. Any type of moderate physical activity (e.g., vacuuming, gardening, walking, swimming etc.) was considered appropriate and encouraged. The researcher used the following behavior change strategies: telephone prompting, and self-control strategies: self-monitoring, participant set goals, self-reinforcement, self feedback and problem-solving strategies. In this study, frequency of contact was varied from no contact, to once per every week, to once per every third week over 16 weeks and faded in the last four weeks. All participants were asked to self-monitor their physical activity, and all participants received either an information only class or a self-control strategies class. Survival analysis using five months of data points and using the criteria of first week of activity missed equaled "death" indicated an effect of greater adherence for individuals participating in a strategies class versus an information only class (46% vs. 14%), but not for the effect of frequency of contact (no contact, 33%; low contact, 31 %; high contact, 30% ) over all 18 weeks of the program. The results suggest the efficacy of a low cost class promoting self-control strategies for behavior change for adherence to an activity program and potentially for other health related behaviors.

▼ The goal of this research project was to determine optimal breeding strategies for improving important traits of Marone species hybrids reared in recirculating aquaculture systems (RAS). Effects of hybridization and backcrossing among striped bass (Marone saxatilis), white bass (M chrysops) and yellow bass (M mississippiensis) on important traits were determined. This dissertation is described in four studies.
In the first study, perfonnance of striped bass female x white bass male (SBxWB) and striped bass female x yellow bass male (SBxYB) fingerlings reared in RAS was evaluated. SBxWB were superior to SBxYB for overall mean growth rate and feed conversion. Survival was higher for SBxYB than for SBxWB. Time x hybrid interactions for growth rate indicated that growth rate of SBxYB improved through time relative to that of SBxWB. Time x hybrid interactions for growth may have been due to differential responses ofSBxYB and SBxWB to deterioration of water quality. The slow growth of SBxYB limits its use for aquaculture.
In the second study, relationships among total weight, body shape, visceral components and fillet yield were examined in SBxWB and SBxYB in order to evaluate strategies for improving fillet yield. Fillets were heavier (5g per fish) for SBxYB than for SBxWB. Relative increase of fillet weight was slightly greater than total weight in both hybrids indicating that small increases in fillet yield can be achieved by rearing fish to a larger size. Low variation in fillet weight (SD ± 9.6 to 11.3g) and poor predictability of fillet weight from body shape (models with r2s of .27 to .43) in both hybrids indicated that potential for improving fillet yield within hybrids is limited. Identification of superior species or strain crosses appears to be the best strategy for improving fillet yield of Morone hybrids.

▼ A spatial linear stability analysis of circular and elliptical jets has been carried out. For the circular jet, the Michalke's shooting method has been extended to the case with a swirling basic flow. For the elliptical jet, an algebraic eigenvalue problem has been formulated by applying the Chebyshev and Fourier spectral collocation method to the disturbance equations of the flow expressed in a generalized cylindrical coordinate system. The resulting algebraic eigenvalue problem was solved using the inverse power method. The approach developed is capable of handling the influences of different distributions of the momentum thickness, swirl, and heat release. In addition to studying the stability properties of the elliptical jet in comparison with those of a circular jet, the linear stability analysis has been used to obtain the insight into the entrainment process. A possible way to enhance the entrainment of elliptical jets has been suggested.

▼ Because the act of supervisory feedback can critically affect a subordinate's
performance, it is imperative to explicate the various conditions in which the character of
feedback is determined. The purpose of the following research was to model the
conditions under which supervisors adopt a negative feedback bias (NFB). This bias was
first described by Kahneman and Tversky (1973), when they noted that Israeli flight
instructors found that praise of exceptionally good piloting was often followed by poorer
performance, while criticism of exceptional poor flying was usually followed by improved
performance. Thus, the flight instructors came to believe that negative feedback
motivated people effectively, while positive feedback appeared ineffective. Of course,
supervisors had erred by failing to recognize the natural variation of their students'
performance. In general, this error applies primarily to the conditions under which
supervisors acquire and interpret information. Two factors were hypothesized as
responsible for the emergence of a NFB: (a) limitations caused by opportunity-based
supervision, where only a certain amount of subordinate behavior can be sampled at any
given moment, and (b) supervisors find it difficult to recognize the natural variation,
random fluctuations, and regression to mean processes characteristic of performance
governed by common causes (cf. Deming, 1982; Hogarth, 1980 and Kahneman &
Tversky, 1973). Results indicated that NFB was an emergent process occurring over time
and under conditions where (a) supervisors managed highly inconsistent subordinate
performance and (b) supervisors had limited information regarding a subordinate's
performance per evaluation episode. Since this experimental approach and set-up is
relatively novel, the results are discussed from several conceptual perspectives. Finally, a
discussion regarding the ecological approach to feedback research, and the importance of
model building and testing is offered.

▼ A novel concept, the fracture efficiency, is proposed in this dissertation as a performance index of fracture specimen designs for adhesive bonds. Evaluated based on a simple quantity, the fracture efficiency parameter, the fracture efficiency represents how much crack driving force a certain specimen design can produce along the bondline for a given maximum non-singular stress in the adherends. In this dissertation, a study of membrane peeling specimens shows that various existing test geometries have little difference in the fracture efficiency. Furthermore, the study shows that it is unlikely to develop new n1embrane peeling tests with high fracture efficiency. Therefore, gross inelastic energy dissipation in the film specimen may occur when using the membrane peeling tests, and may result in large errors in the film adhesion measurement when only elastic material response is considered in the analysis. As a result, the use of a backing material for film adhesion measurement is suggested because of the significant increase in the fracture efficiency when a proper backing material is used. Besides the use as one of the guidelines for fracture specimen designs, it is also demonstrated in this dissertation that the fracture efficiency parameter can also be used to help one determine the validity of a measured bond fracture strength.

▼ This research examined the outsourcing decision for the logistics function of motor carrier transportation. A full factorial design was executed on a simulated transportation network to investigate how the efficiency of motor carrier transportation was affected by how it was structured (private/leased fleet versus contract carrier transportation) and the characteristics of the transportation activities. Transaction Cost Analysis (TCA) offered a useful theoretical framework for consideration of this make or buy decision by suggesting the independent variables of asset specificity, uncertainty, and frequency/volume.
Seven two-part research hypotheses examined the relationships among the independent variables to gain a greater understanding of the factors which drive the make versus buy decision for motor carrier transportation.

▼ It is observed for the first time that telechelic ionomers based on sulfonated polyisobutylene exhibit primary and secondary SAXS peaks when the molecular weight distribution is narrow (Mw/Mn ca. 1.15). These SAXS peaks are interpreted as due to non-global lamellar or cylindrical ionic domain order. The non-global ordering strongly affects the tensile and dynamic mechanical properties of the ionomer. This effect is attributed to local ionic phase continuity and the concept of restriction of chain motion by the ionic domain.
Triblock copolymer ionomers from t-butyl methacrylate (tBMA) end blocks and polybutadiene or tapered poly(styrenelbutadienel styrene) (tSBS) mid blocks exhibit complex SAXS patterns indicating dispersed spheroidal ionic end block domains in the matrix. TEM shows that ionization of the 9 mol % PtBMA-PBD triblock produces a morphological transformation from unoriented rodlike domains to ionic spheroids.

▼ Policy case studies are presented on two congressional proposals that, if implemented, may result in implicit or explicit federal government endorsement of particular standards for the licensure or certification of teachers. They are, (a) authorization for the United States Department of Education to award design and implementation grants for programs of alternative teacher credentialing, and (b) appropriation of federal funds to support the National Board for Professional Teaching Standards.
A discussion of the genesis of each proposal, a review of the legislative history of these two measures, and analysis of supporting rationale for each is included.
Using an analytic framework based on the works of Deborah Stone and Thomas Green, it was determined that unresolved policy conflicts over teacher education governance and content led to federal attention to matters of teacher certification and licensure.

▼ A large-eddy simulation methodology (LES) has been developed for predicting the turbulent physics of an incompressible flow in simple and complex geometries. The Cartesian form of the governing equations was first verified, and then later used to investigate a three-dimensional shear-driven cavity flow. The investigation involved Reynolds numbers of 2000, 3200, 5000 and 10000 and focused on the unsteadiness and turbulent characteristics of the flow. At the low Reynolds numbers (Re ~ 5000) where the cavity flow is fully laminar, direct numerical simulations (DNS) were conducted whereas the LES methodology was adopted to predict the cavity flow at the higher Reynolds number (Re = 10000). Determining the parameters in the damped subgrid scale (SGS) turbulence model for this complex flow was guided by the DNS results at Re = 5000. The SGS model was also verified against DNS results at Re = 7500 where the cavity flow was known through laboratory experimentation to be locally transitional. The LES results using the damped SGS model verified the published experimental evidence as well as uncovered new flow features within the cavity.
LES computations were also carried-out of the three-dimensional shear driven cavity flow at a high Reynolds number where the SGS turbulent field was represented by a dynamic model. Lilly's least-squares expression was tested for determining Smagorinsky's coefficient in the model without ad hoc measures such as ensemble-averaging or filtering. However, zero cutoff of negative total viscosity (kinematic plus turbulent eddy viscosity) was necessary to maintain stable solutions. A discretized filter function was derived for the test filter. Both qualitative and quantitative comparisons to experimental data show that the dynamic model performed quite well. The dynamic model gave better comparisons to the experimental evidence than the damped model did. Vortex formation in the wake of a circular cylinder and their subsequent downstream transport was also numerically investigated by LES. Here however, the curvilinear form of the governing equations was necessary to perform the computations. A new generalized dynamic model was derived to represent the SGS stress field in the curvilinear space. This new model introduced the contravariant velocity components as part of the field variables.
New downstream boundary conditions were also formulated to permit the shed vortices to exit with minimum disturbance. The focus of the investigation was at Re = 5600 with some verification of the computations at Re = 200 and Re = 3000. At all of these Reynolds numbers. the upstream boundary layer was laminar but the wake was fully turbulent at Re = 3000 and Re = 5600. The LES results of the many interesting characteristics of the wake showed good comparisons to the experimental data.

▼ Most educational leaders of higher education recognize the importance of creating and sustaining a sense of community on college campuses to enhance the likelihood that students achieve their educational goals. Historical definitions of campus community often are vague in their characterizations and difficult to use to structure learning experiences. This study was concerned with creating definitions of campus community with greater specificity and utility than has been traditionally revealed through literature. Historical conceptualizations of community were reviewed and a consensually oriented process for operationally defining campus community in a particular setting was employed to arrive at a preferred definition of community.
The Hutchinson methodology for defining "fuzzy concepts" was used to arrive at a preferred definition of community. Student participants from a land-grant research university living in three distinct life style arrangements were consulted in workshops designed to achieve consensus to arrive at their operational definitions. The findings from this study should be useful especially to student affairs professionals who are concerned with structuring
out-of-class college environments to strengthen the learning opportunities for students.

▼ Virginia's mandated student outcomes assessment program, as created by Senate Joint Resolution No. 83 in 1986, allowed community colleges in the Commonwealth latitude in developing their own assessment strategies. These strategies have developed slowly and with some inconsistency.
The purpose of this study is to determine the congruency of beliefs about the implementation of Virginia's student outcomes assessment program between purposefully selected members of the Virginia General Assembly and community college assessment practitioners.
Data for this study were derived from the review of public documents, and the execution of a survey instrument, the results of which were used to develop interviews with designated state legislators and community college assessment practitioners. The interviews became the primary data source for the study.
Findings indicate that the legislative participants believe that used by all community colleges across Virginia would be more appropriate than approaches specifically geared to individual institutional missions. Practitioner respondents differ on this issue. The legislators generally believe that the use of a standardized testing approach administered across all community colleges could be beneficial. The practitioners beliefs differ, though not markedly, from the legislative group.
