▼ The effects of dietary folic acid supplementation on serum folate status, conceptus development and reproductive performance in gilts and sows was investigated in a series of experiments. The specific objectives of the study were to: 1) to study the serum folates profile following rapid consumption of a single meal containing different levels of supplemental folic acid; 2) to study the effects of maternal folic acid supplementation on conceptus survival, growth and development prior to mid-gestation in gilts and sows; and 3) to study the effects of multiple levels of dietary folic acid supplementation on sow reproductive performance over four successive parities. In the first experiment (Chapter III), gilts fed a single, rapidly consumed meal, had a rapid increase in serum folates concentration during the first hour postprandial. Within eight hours after feeding, serum folates in the gilts had returned to near prefeeding levels. The results also suggest that supplementing the diet with increasing levels of folic acid over a range of 0 to 4 ppm results in more rapid postprandial uptake and elimination of folic acid from general circulation. In the second experiment (Chapter IV), supplementing the diet of first parity and third parity breeding sows with 2 ppm folic acid had no effect on litter size at day 45 of gestation. However, several measurements associated with placental and fetal growth were increased with folic acid supplementation. Results of a fifth parity trial with four levels of supplemental folic acid were variable and inconclusive. The implications of increased placental and fetal growth up to day 45 of gestation in the first and third parity sows is discussed. In the third experiment (Chapter V), a decline in serum folate concentration in pregnant sows from mating to mid- to late gestation was clearly demonstrated. Supplementing the sow's diet with folic acid over a range of 0 to 4 ppm resulted in a linear increase in serum folate concentration at mating, during gestation and at weaning. However, under the conditions of this experiment, folic acid supplementation had no significant effect on sow reproductive performance. Advisors/Committee Members: Blodgett, Dennis J. ▼ The purpose of this study was to determine if there were differences in clothing values between Black and White middle-income women patronizing the primary and second-order markets, and to determine if there were differences in the clothing buying practices between these groups in the two markets. A self-administered questionnaire was distributed to a purposive sample of 250 Black and White women residing in three metropolitan areas. The clothing values measured were economic, political, religious, and conformity. Eighteen clothing buying practices were included in this study. N1ne practices were speclfical1y directed toward the second-order market shoppers. Variations were found in the rank order of clothing values within the two groups. Blacks ranked the religious value higher and conform1ty lower than the Whites. Blacks scored higher on the economic and re1igous clothing values while Whites scored higher on conformity. Significant differences between Black and White middle-income women in their clothing buying practices were found in: (1) the method used to acquire the majority of clothing; (2) the percentage of personal clothing items purchased in primary stores; and (3) buying pattern for a dress costing more than $50.00. Significant differences between Black and White middle-income women who used the second-order market were found in: (1) length of time respondents had purchased used-clothing; (2) satisfaction with price when making used-clothing purchases; (3) shopping the Salvation Army, Goodwill, and thrift stores; purchasing of (4) pants; and buying used-clothing to wear for (5) work and (6) school. Middle-income women who used the primary market exclusively scored higher on the conformity clothing value than did those who used the second-order market. ▼ Speech intelligibility tests using three headset systems, two passive and one with active noise reduction technology, were conducted. Nine listeners, six males and three females, responded to 180 50-word Modified Rhyme Word Lists presented under two noise conditions, two conditions of bilateral phase reversal of the speech signal, and the three headset systems. These three independent variables were studied in a full factorial within-subjects design. The noise conditions were pink noise and recordings from the troop carrying compartment of a u.S. Army M-2 Bradley Fighting Vehicle (tank noise having substantial low frequency energy). Phase reversal of the speech signal was also tested for its effect on speech intelligibility. The three headset systems were the David Clark Noise Attenuating Aviation Headset, and the Bose Aviation Headset used in both its passive and active mode. Active noise reduction technology employs the physical principle of constructive and destructive interference by creating signals which are identical to the noise under the earmuff but out of phase by 180 degrees. This technology is most effective at frequencies below 1000 Hz. Results showed that the Bose headset in its active mode required a significantly higher speech-to-noise ratio in both noise environments than the two passive headset systems to achieve the 70% level of speech intelligibility, the dependent variable in this study. The mean speech level under the Bose headset in its active mode was 2.8 dB higher in tank noise and 3.5 dB higher in pink noise than under the David Clark headset. The Bose headset in the active mode, however, provided a greater degree of broadband attenuation especially in the lower frequency range, e.g. < 630 Hz. Phase reversal proved to be of no benefit to performance in either noise environment. The pink noise proved to be the harsher environment for speech intelligibility than did the tank noise, primarily due to the increased levels in the speech bandwidth. Articulation Index scores for the three headset systems evidenced that the differences in performance among the three headset systems were in part the result of better earphone response characteristics in the principal speech bandwidth (600 - 4000 Hz) in conjunction with the strong attenuation performance of the David Clark headset in that same range. It is suggested that, in the absence of better earphone response characteristics and a broader bandwidth of active noise reduction performance, speech intelligibility is no better under active noise reduction headsets than under quality passive headsets. Further, the levels of speech intelligibility attained by active noise reduction headsets are at a cost of increased signal strength and higher purchase price. ▼ High teacher attrition rates coupled with critical personnel shortages in special education have augmented the need for research in teacher attrition/retention. Retaining quality personnel in special education teaching is vital to assuring that students with disabilities receive an appropriate education. The purpose of this study was to provide an in-depth exploration of factors influencing urban special education teachers' career plans. To gain a better understanding of urban special educators' commitment, job satisfaction, and career plans, qualitative research methods were employed throughout this study's data collection and analysis procedures. Based on the results of a screening instrument from another study, sixty special education teachers in the Memphis City Schools (MeS) were selected to participate in this study. These teachers were divided equally into three groups of special educators (i.e., stayers, leavers, undecideds) with specific career plans and attitudes (e.g., commitment, job satisfaction). Face-to-face interviews were conducted with each of the special educators using an interview guide. Cross-interview analyses were used to analyze the interviewees' responses to each of the questions on the interview guide. Patterns and themes that emerged from the data were identified and discussed. Specific teacher examples and verbatim quotes were also included to illustrate the study's findings. According to interviewees, various job-related factors (e.g., support, work assignment, student factors, work rewards) were most important to their commitment, job satisfaction, and plans to remain in and leave special education teaching in MCS. Support was more often mentioned as a reason for wanting to stay than any other factor. Reasons for wanting to leave special education teaching in MCS clustered around two major factors, work assignment and support. These findings suggest that special attention to job related factors may be particularly important to prevent attrition among these at-risk teachers. By listening to interviewees' recommendations for improving work conditions in MCS and including these teachers in the decision making process, school administrators may positively affect teachers' career plans and better retain their special education teaching force. ▼ The cell extract protein content of acetate- and methanol-grown Methanosarcina thermophila was examined by two-dimensional polyacrylamide gel electrophoresis to determine the extent of regulation by the growth substrate. More than 100 mutually-exclusive spots were present in acetate- and methanol-grown cells suggesting a high degree of regulation. Spots corresponding to acetate kinase, phosphotransacetylase, and the five subunits of the nickel-containing carbon monoxide dehydrogenase (CODH) complex were identified in acetate-grown cells. The nickel-containing methyl coenzyme M methylreductase from acetate-grown M. thermophila was purified 16-fold from a cell extract to apparent homogeneity. The enzyme had a native molecular weight of between 132,000 and 141,000 and contained three subunits with a configuration of a1B1y1-. The as-isolated enzyme was inactive, but could be reductively reactivated by either titanium (III) citrate or reduced ferredoxin. Reactivation with ferredoxin was a simplification over previously reported reactivation systems. ATP stimulated, but was not required for reactivation. The CO dehydrogenase enzyme complex from M. thermophila was purified and separated into its respective components: the CO-oxidizing nickel/iron-sulfur (Ni/Fe-S) component and the cobalt-containing corrinoid/iron sulfur (Co/Fe-S) component. EPR spectroscopy and spectroelectrochemical titration of the Fe-S centers of the Ni/Fe-S component indicated the presence of two low-potential [4Fe-4S]2+/1+ centers and third high-potential center whose Fe-S configuration is unknown. When reduced with CO, the NilFe-S component exhibited a previously unobserved Ni-Fe-C EPR signal. The Co/Fe-S component contained one [4Fe-4S]2+/1+ cluster, and the as-isolated corrinoid in the component was in the base-off conformation suggesting that modulation of the electron density of the cobalt ion may result in a modified reactivity of the active site of the corrin. The CODH enzyme complex and isolated Co/Fe-S component reductively dechlorinated trichloroethylene to cis-dichloroethylene, trans-dichloroethylene, 1,1-dichloroethylene, vinyl chloride, and ethylene. Factor III also catalyzed the dechlorination of trichloroethylene when in the presence of titanium (III) citrate. Reconstitution of the Co/Fe-S component with the CO-reduced NilFe-S component also allowed dechlorination demonstrating an electron transfer from the reduced Ni/Fe-S component to the Co/Fe-S component. ▼ Compressible turbulence in a high-speed, high Reynolds number, supersonic free shear layer was studied. A two-dimensional free mixing layer was chosen to study turbulence rather than a wall bounded flow due to the experimental fact that the effects of compressibility become significant at lower Mach numbers. The mixing layer was generated by supersonic injection of air <Ms = 1.8, Pts = 0.5 atm. Ttl = 295K. and Re/m = 7xlcf) through a rearward facing tangential slot, into a supersonic free stream (Moo = 4.0, Ptoo = 12.5 atm, Ttoo = 290K, and Re/m = 70xlcf). Flow visualization was accomplished by nanosecond Shadowgraph photography. The overall flow structure was documented with the Shadowgraph and conventional mean flow probes (Pitot pressure, cone-static pressure, and thermocouple probes). The turbulent structure of the flow field was also clearly depicted in the Shadowgraphs. Image processing techniques were developed in order to determine root-mean-square index of refraction (density) fluctuation levels from the Shadowgraph plates. Multiple overheat normal and cross-wire techniques were developed and/or improved for this study. The present research concentrated on the Reynolds averaged form of the Navier-Stokes equations. where the effects of compressibility are manifested through "apparent masstl terms (i.e. p'ufi). These terms appear in all of the Reynolds averaged Navier-Stokes equations (continuity, momentum, and energy). A new turbulence transformation, coupled with innovative experimental methods. allowed the full compressible Reynolds shear stress (the typical incompressible term, pJiU'j' as well as the apparent mass terms) to be directly measured. The full compressible heat flux and apparent mass terms were also estimated from the cross-wire results. Profiles were obtained at four downstream stations which were strategically located to map different levels of development of the shear flow. The frrst station was very close to the injector, about one free stream boundary layer thickness downstream (xJ8oo == I), hence, it is in the initial region. The second station was located at x/'Ooo == 28, which was near the beginning of the fully developed zone. The third station, x/'Ooo = 83, was just prior the shear layer and floor boundary layer merging. The last station was positioned just aft of the layer merging, x/'Ooo = 106. II In addition to physiological problems resulting from stress almost every psychosocial variable of importance is affected by stress in the workplace, including productivity, morale, and the .psychological well-being of workers" (Ivancevich and Matteson, 1980). Cooper and Marshall (1975) proposed two central features of stress at work, the interaction of which determines either coping or maladaptive behavior and stress related disease: (1) the characteristics of the persons (dispositional) and (2) the potential sources of stress in the work environment (situational). There is a growing need to use stress responses to cope with stressful situations encountered in occupations. In order to fulfill their responsibilities, employees should be aware of stressors that develop. The methods utilized in this study should provide an approach to identifying stressors and assessing counterproductive job situations. The purpose of this study was to measure and compare the degrees of stress experienced by administrators, teachers I and support personnel of the District of Columbia Public School System to selected occupational factors. This study utilized a descriptive research methodology and survey technique to gather data from a sample population of employees of the District of Columbia Public School System including: (a) administrators, (b) teachers, (c) support personnel f including engineers and clerical support personnel. It was hypothesized that: (1) there is no statistically significant difference in stress levels of administrators, teachers, and support personnel as measured by responses to measurements of perceived occupational stressors (change, clarity, tedium, control, intensity and conflict) according to reported situational and dispositional factors. Analysis of Variance procedures and Pearson Moment Correlation were used to test the hypothesis. Data for the study were obtained from five high schools in the District of Columbia. ▼ Angus cows (Bos taurus) with calves and 1/2 Dorset, 1/4 Finn and 1/4 Rambouillet ewes (Ovis aries) with lambs grazed Kentucky bluegrass (Poa pratensis) - white clover (Trifolium repens) in a randomized block design with three replications during 3 yr, to investigate effects of grazing cattle and sheep together and separately on soils, plants and animals. Additionally, changes in stocking rate, seasonal distribution of cool-season forages and varying nutritional requirements of animals were examined. There were 6 cow-calf pairs or 6 ewes with 11 lambs per replication of cattle alone and sheep alone treatments. Six cows plus six ewes, with their offspring, were allotted to each replication of mixed grazed pastures. Initial stocking rates were equilibrated among treatments to 1 cow and calf per .45 ha. Grazing sheep alone increased (P < .05) percentage bluegrass (58%) but decreased white clover (6%) and weeds (36%), compared to pastures grazed only by cattle (30, 16 and 53%, respectively, which was similar to initial composition). Soils in mixed grazed pastures remained higher (P < .05) in plant nutrients, pH, and percent organic matter and were less (P < .05) compacted, compared to soils where sheep or cattle grazed alone. Total forage mass and quality were higher (P < .05) in mixed grazed pastures, compared to pastures grazed by cattle and sheep alone. Daily gain (.23 kg/d), total gain (23 kg), and weaning weights (43 kg) of lambs were higher (P < .01) in pastures grazed by both animal species, compared to sheep alone (.18 kg/d, 19 and 38 kg, respectively). These animals also reached target weaning weights (43 kg) 14 d earlier (P < .05) than those grazed alone. Body condition of ewes was improved by the breeding season, compared to ewes grazed alone. Five measuring techniques were investigated to determine forage mass and botanical composition. Measurements based on small plots permanently located within pastures were as accurate as those based on the entire pasture. Visual estimations of forage mass were lower, compared to hand clipping, but reflected canopy height. Visual evaluations more accurately estimated presence of individual plant species than hand separated samples. Grazing ewes and cows with associated spring-born offspring on bluegrass-white clover improved the balance between plant growth and animal forage requirements and improved sustainability of the soil-plant-animal system. ▼ The first part of this research constituted the study of oligomeric incorporation into an inorganic network leading to the fonnation of a hybrid material by the sol-gel process. The oligomeric species chosen was silanol terminated poly(dimethylsiloxane) (PDMS) and the inorganic oxide precursor chosen was titanium tetra-isopropoxide (TIP). The rationale for choosing these materials stemmed from the desire to make materials for optical filters since a gradient of optical properties can be derived based on the composition and the initial reaction conditions. 29Si NMR studies were conducted to investigate the incorporation process which led to the conclusion that the PDMS chains were being degraded by the TIP. This problem led to the synthesis of TIP endcapped PDMS which was then used as the oligomeric component in the hybrid materials. The endcapping changed the functionality of the oligomer to six, and more importantly, its reactivity which was now closer to that of the TIP. Also, having the same functionality at the ends as the precursor affects the compatibility of the two components. Hybrid materials in the form of free standing films were fabricated from varying TIP/pDMS compositions and different initial reaction conditions. The hybrid materials synthesized were then subjected to a systematic structure-property relationship study. The degree of phase mixing or the encapsulation of the siloxane phase by the inorganic oxide increases with higher TIP content. This behaviour was seen by both the dielectric and dynamic mechanical studies. The modulus and the stress to break increased with higher TIP content while the strain to break decreased. At the 50/50 TIP/PDMS composition there was an abrupt change in the mechanical properties which signified a morphological change in these materials. The refractive indices of the materials also showed a trend similar to that of the mechanical properties. Three different cure procedures, viz., room temperature, thermal cure at 70 °C and microwave cure at 70 °C were utilized to fabricate the films. More phase separation occurred in the microwave cured over the thermally cured materials as evident from dielectric and dynamic mechanical studies. However, the effect diminished with higher TIP content in the materials. Changing the initial reaction conditions by varying the initial water and acid content changed the properties observed in the final products. Dielectric and dynamic mechanical studies showed that the phase separation was higher with increased water content. Also, the modulus of the materials made with increased water content was higher. Decreased acid content led to better dispersion of the two components as shown by the dielectric and dynamic mechanical experiments. The effect of using a less reactive inorganic component tetra-ethyl orthosilicate (TEOS), instead of TIP was also studied. The TEOS/pDMS materials showed more phase mixing than the TIPIPDMS materials, although, the dielectric tan Î´ peak maxima appeared at a lower temperature. Meprin A contains 90-kDa subunits (α subunits) and is expressed in random-bred and some inbred strains of mice. Meprin B contains subunits of 110 kDa (β subunits) in situ, and the enzyme from C3H/He mice, a strain that does not express α subunits, has been characterized. Evidence from this and previous studies indicate that β subunits are expressed in all mouse strains. Meprins were characterized with regard to their glycosylation by lectin blotting. Both meprin A and meprin B bound the lectins concanavalin A and the erythroagglutinin from Phaseolus vulgaris indicating that both enzymes contain high mannose and bisected biantennary complex type oligosaccharides. However, meprin A, but not meprin B, bound the agglutinins from Ricinus communis, Datura stramonium, and the leukoagglutinin from Phaseolus vulgaris, indicating that complex-type N-glycosylation differs in these proteinases. Lectin blots of membrane proteins from C57BL/6 mice indicated that there were differences between adult male and female mice in the glycosylation (specifically in the complex type oligosaccharides) of the α subunit of meprin A. A marked degree of carbohydrate heterogeneity was observed in meprin A from males as compared to the enzyme isolated from female mice. Additionally, the data indicated that at least three of the ten potential glycosylation sites in the meprin α subunit are glycosylated. Overall, these studies expand our understanding of how estrogens affect glycosylation of meprin A. The oligomeric organization of the meprins was examined in brush border membrane fractions from a random-bred strain (lCR) and two inbred strains of mice (C57BL/6 and C3H/He). The random-bred strain contained three oligomeric complexes of approximately 390, 440, and 490 kDa as determined after sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-P AGE) in the absence of reducing agents. The subuβnits in all three oligomers were linked by disulfide bridges. Western blotting using anti-α monoclonal antibodies revealed that α subunits (90 kDa) were present in the 390- and 440-kDa complexes. Western blotting with polyclonal antibodies specific for the β subunit (110 kDa) revealed the presence of these subunits in the 440- and 490-kDa complexes. Electroelution of the individual oligomers followed by SDS-PAGE under reducing conditions confirmed that the 390- and 490-kDa molecules are homotetramers of α and β subunits, respectively, and that the 440-kDa complex is a heterotetramer composed of disulfide-linked α and subunits. C57BL/6 mice expressed both α and β subunits and contained tetramers composed of α₄ and α₂β₂. C3H1He mice expressed only the 110-kDa β subunits and the β₄ oligomer. This type of multimeric organization of covalently-linked subunits is unique for the known endopeptidases. ▼ A series of RF plasmas was chosen to modify the surface chemical composition of Hercules IM7 carbon fibers. A two-liquid tensiometric method was used to determine the surface energy parameters y(ds) and I(psf) of the fibers. An XPS analysis of air and argon plasma treated fibers indicated a significant surface oxidation of the fibers which translated into low y(ds) values and high I(psf)values. An ammonia plasma was shown to remove an outer layer from the surface of the fibers. It also increased y(ds) compared to as-received fibers without affecting the non-dispersion (IPsf) XPS results indicated that methane and ethylene plasmas deposited a layer of low surface energy hydrocarbon on the fiber surface. A trifluoromethane plasma and a tetrafluoromethane plasma introduced a significant amount of fluorine containing groups in the fiber surface in the form of a fluorinated plasma polymer in the first case and through direct attack of the fiber surface by fluorine atoms in the second case. The surface chemical composition and the surface energy parameters of two series of commercially treated carbon fibers were also determined and compared to the results on IM7 carbon fibers. The adhesion of carbon fibers to polyethersulfone (PES) was measured by using the microbond pull-out test, and compared to the adhesion of the same fibers to an epoxy resin. The load required to debond the microdroplet was used as a measure of the bond strength. The data were also analyzed in terms of interfacial fracture energy accordIng to the model developed by Jiang and Penn (1992). The microbond pull-out test results showed no significant relation between the fiber surface chemical composition or the fiber surface energy, and the adhesion to PES. However, plasmas which have a strong ablative character such as the ammonia and the tetrafluoromethane plasmas did improve the fiber-PES adhesion, when compared to as-received fibers. The study of the fiber-epoxy systems revealed that a chemical effect contrlbuted to the adhesion improvement but to a lesser extent than the "cleaning" effect of the surface treatment. The results support the two part mechanism proposed by Drzal and extend its application to carbon fiber-thermoplastic systems, but in this case the chemical effect is minimal.
