▼ The ability of tumour cells to avoid immune destruction (immune escape) and their acquired resistance to anti-cancer drugs constitute important barriers to the successful management of cancer. The interaction between specific molecules on the surface of tumour cells with their corresponding receptors on immune effector cells can result in inhibition of these effector cells, consequently allowing tumour cells to evade the host’s anti-tumour immune response. The interaction of the Programmed Death Ligand 1 (PD-L1) on the surface of tumour cells with the Programmed Death-1 (PD-1) receptor on cytotoxic T lymphocytes leads to inactivation of these immune effectors, and is a specific example of an immune escape mechanism tumour cells use to avoid immune destruction. Clinically, antibodies capable of blocking the PD-1/PD-L1 interaction have demonstrated significant therapeutic benefit, and are currently being used to help bolster patients’ immune response against malignant cells in a variety of cancer types. Here we show that the PD-1/PD-L1 interaction also leads to tumour cell resistance to conventional chemotherapeutic agents. Incubation of PD-L1-expressing human and mouse tumour cells with PD-1-expressing Jurkat T cells or purified recombinant PD-1 resulted in tumour cell resistance to doxorubicin and docetaxel. Interference with the PD-1/PD-L1 interaction using blocking anti-PD-1 or anti-PD-L1 antibody or shRNA-mediated gene silencing resulted in attenuation of PD-1/PD-L1-mediated drug resistance. Moreover, inhibition of the PD-1/PD-L1 signalling axis using anti-PD-1 antibody enhanced the effect of doxorubicin chemotherapy to inhibit 4T1 tumour cell metastasis in an in vivo mouse model of mammary carcinoma. These findings indicate that blockade of the PD-1/PD-L1 axis may be a useful approach to immunosensitize and chemosensitize tumours in cancer patients and provide a rationale for the use of anti-PD-1/PD-L1 antibodies as adjuvants to chemotherapy.

▼ Multiple linear regression analysis may be used to describe the relation of a variable
(response variable) based on the score of several other variables (independent variables).
The least squares estimate of the regression coefficients are unsteady in that
replicate samples can give widely differing values of the regression coefficients if the
predictor variables are highly correlated. Ridge and Lasso regression analysis are regularization
techniques for eliminating the effect of high covariance from the regression
analysis. They produce estimates that are biased but have smaller mean square errors
between the coefficients and their estimates. The lasso and ridge trace plot of the coefficients
versus _ and cross validation are some ways that helps to determine the value
of regularization constant _ and regression coefficients based on the data. Ridge regression
and Lasso regression help the analysis to a more trustable interpretation of the
results of multiple regression with highly correlated covariates.

▼ The programmed death ligand-1 (PD-L1) is an immune checkpoint protein expressed on a variety of antigen-presenting cells to normalize immune system. Recently, overexpression of PD-L1 on the surface of cancer cells has been proven to contribute to cancer immune evasion and tumor progression. Specifically, PD-L1 is found highly upregulated in Triple negative breast cancer (TNBC), a subtype of breast cancer that currently has no effective clinical therapies, allowing blockade immune checkpoint therapy a potentially effective treatment. However, the overall response rate for PD-L1 targeted immunotherapy in PD-L1 positive TNBC is less than 20%. The mechanism for this is largely unknown. It has been suggested that the levels of many other genes regulating PD-L1 may contribute to the sensitivity of cancers to immunotherapy. Therefore, there is a critical need to understand how PD-L1 is upregulated and interacts with its upstream regulators in the modulation of anti-PD-L1 immunotherapy response in TNBC. Over the past years, the literature that implicated the regulation of PD-L1 was primarily on transcription levels. For this reason, my project aims to identify post-translational modification of PD-L1 in efforts to better target it in TNBC immunotherapy.
To identify novel kinase regulators of PD-L1, we first performed a proteomic analysis of PD-L1 and identified several tyrosine kinases as novel PD-L1 interacting proteins. We provide evidence that some of these kinases (EphA3, FAK, FES, and FER) could phosphorylate PD-L1 in vitro and co-expression of FAK, FES, and FER could stabilize PD-L1 in ectopic co-expression models. Moreover, FAK inhibition, knockdown, or ectopic over-expression resulted in decreased or increased PD-L1 levels, respectively; and that inhibition of EphA2 was associated with reduced endogenous PD-L1 levels. Most significantly, knockout of FAK in BT549 TNBC cells reduces immune evasion by inhibiting cancer cells-induced T-cell apoptosis in coculture experiments. In summary, we have identified several novel kinase regulators of PD-L1 stability and immune evasion. This is a critical first step in unveiling the molecular mechanism underlying PD-L1 upregulation in TNBC immune evasion and tumorigenesis. These findings present exciting opportunities for developing a new potential target and effective combinational therapies for immunotherapy-resistant TNBC.

▼ A HeLa sejtvonal a daganatos sejtvonalak között is kiemelkedő túlélő képességekkel és osztódási sebességgel rendelkezik, gyorsan és agresszíven képes befertőzni más sejtvonalakat. Különböző sejttenyészetek egy sejttenyésztő inkubátorban való tárolása, illetve a nem megfelelő sejttenyésztési technikák alkalmazása miatt növekszik a száma a HeLa általi fertőzéseknek. Becslések szerint a laborokban tárolt sejtvonalak 18%-a fertőzött más sejtvonalakkal, a HeLa egyedül 25%-áért felelős ezeknek az átkereszteződéseknek (Rahbari,R. és mtsai. 2009). Eddig csak rendkívül költséges technikákat ismertünk a sejttenyészetek keresztfertőzésinek tesztelésére, melyek csak a nagyobb sejttenyésztést végző kutató központok és vállalatok számára megfizethetőek. Rahbari és mtsai 2009-ben javaslatot tettek az
L1 retrotanszpozon kimutatásán alapuló,alacsonyabb költségekkel járó, HeLa specifikus, PCR alapú gyorstesztre, amely nagyon alacsony szintű HeLa fertőzést is kimutat (<1%), illetve még intakt sejtekből is képes jelezni a HeLa szennyezést. Az
L1 retrotranszpozont a Hela sejt amisyin génjének inszeciójaként adjuk meg a primerek megtervezése során. Az
L1 retrotranszpozon nem fordul elő más emlős sejtvonal ugyanezen génjében, ezért az inszerciót bizonyító 325bp méretű DNS fragment lesz a HeLa marker a gyorsteszt során. Emellett átírásra került a gén
L1 nélküli, feltételezhetően működő 500 bp méretű fragmentuma is, amely egységesen minden sejtvonalban (a HeLa-ban is) megtalálható. A HeLa sejtekben az inzert nélküli és az inzertet kódoló szekvencia átíródása egyszerre várható. Szakdolgozatom során az Orvosi Vegytani intézetben tenyésztett HEK293T, HepG2 és A549 sejtvonalak HeLa fertőzöttségét vizsgáltam a HeLa specifikus gyorsteszt használatával. A sejtfeltárás optimalizálása, illetve a PCR reakció sikeres optimalizációja után megtaláltuk azt a legkisebb DNS koncentrációt minden mintánál, amelynél a detektálhatóak a keresett DNS szekvenciák.

▼ Die vorliegende Diplomarbeit untersucht, in welchem Ausmaß Schülerinnen und Schüler deutscher und serbischer Muttersprache englische phrasal verbs, welche vergleichbar sind mit deutschen Partikelverben, meiden. Englische Muttersprachler dienen hierfür als Bezugspunkt. Die Schülerinnen und Schüler sind zwischen 16 und 18 Jahre alt und weisen ein Englischniveau von ungefähr B1 auf. Die Studie erreichte eine Beteiligung von 143 Personen, die sich aus 23 englischsprachigen, 52 deutschsprachigen und 68 serbisch-sprachigen Teilnehmerinnen und Teilnehmern zusammensetzt. Ihnen wurde ein Multiple-Choice Test aus 25 Dialogen ausgehändigt, in welchen jeweils das betreffende Verb ausgelassen wurde. Sie sollten eine von vier vorgegebenen Antwortmöglichkeiten auswählen: dem richtigen Partikelverb oder dessen Synonym (ein einfaches Verb), einem falschen Partikelverb oder einem falschen Verb. Die Studie zeigt, dass deutschsprachige Schülerinnen und Schüler zu den Partikelverben tendieren, während serbische Muttersprachler eher dazu geneigt sind, sich für das einfache Verb zu entscheiden. Die englischen Muttersprachler zeigen die größte Abneigung gegenüber den phrasal verbs. Daraus folgt, dass die Muttersprache nicht die ausschlaggebende Rolle bei der Vermeidung der Partikelverben spielt. Es ist wahrscheinlicher, dass die feinen, aber dennoch vorhandenen, Unterschiede in der Bedeutung des Partikelverbes und dessen Ein-Wort-Synonyms dazu beitragen, dass eines der Beiden als etwas geeigneter im vorgegebenen Kontext empfunden wird. Wenn das Partikelverb und dessen Ein-Wort-Synonym als gleichermaßen geeignet empfunden wird, kann die Wahl auf persönliche Präferenzen und Sprachstil zurückgeführt werden. Es kann daher die Schlussfolgerung gemacht werden, dass der semantische Kontext, die persönliche Präferenz sowie der eigene Sprachstil den größten Einfluss darauf haben, ob die Wahl auf das Partikelverb oder dessen Ein-Wort-Synonym fällt.
This diploma thesis examines the degree of avoidance of English phrasal verbs by Serbian L1 English learners as well as avoidance and transfer of German L1 English learners. English native speakers serve as a point of reference for this purpose. The participants of the two English learner groups are high school students between the age of 16 and 18, whose level of English can be estimated to be intermediate, i.e. B1. The study reached a total number of 143 participants, i.e. 23 English native speakers, 52 German L1 and 68 Serbian L1 learners of English. A multiple-choice test of 25 dialogues was provided, whereby the verb in question was omitted so that participants had to decide on one out of four verbs: the appropriate phrasal verb (PV), its one-word equivalent (OWV), an incorrect phrasal verb or an incorrect one-word verb. The findings reveal that the German natives slightly tend to prefer PVs, while the Serbian natives tend to avoid the same. L1-adaptive
control methods for the closed-loop control of anesthesia delivery during surgery.
Our initial objective, described herein, is to design controllers that are robust to
inter-patient variability, such that patients follow a prespecified Bispectral Index
(BIS) profile. The controllers are then evaluated on their ability to reject the
disturbances effects on the patients??? BIS trajectory that arise during the course
of surgery. The controllers are designed using identification-based models constructed
using clinical trial data. Two different
L1-adaptive control schemes,
a state-feedback scheme and an output-feedback scheme, will be evaluated and
compared in terms of tracking performance, amount of anesthesia used, and robustness
to interpatient variability.

▼ Bromodomain and Extra-Terminal (BET) proteins are a conserved family of ‘epigenetic readers’ that bind to acetylated lysine residues on histone and non-histone proteins to modulate transcription. BET proteins are enriched at promoter and enhancer regions and recruit the positive transcription elongation factor b (P-TEFb) complex to activate RNA polymerase II. Anti-tumour responses elicited by BET inhibitors have been associated with the suppression of genes required for cellular proliferation and survival, including oncogenic transcription factors. Suppression of the proto-oncogene MYC was initially reported as a key mechanistic property of BET inhibitors, however more recent evidence suggests that additional target genes are mechanistically implicated.
In this thesis, the Eμ-Myc model of aggressive B-cell lymphoma was utilized to investigate the full repertoire of genes modulated by JQ1 and their functional significance in mediating therapeutic responses. JQ1 did not suppress the expression of transgenic Myc in this model, allowing the determinants of apoptosis induction to be assessed, independently of changes in Myc expression. This apoptotic response was p53-independent and associated with modulation in the ratio of pro- and antiapoptotic Bcl-2 family members to favor activation of the intrinsic mitochondrial apoptotic pathway. Therapeutic administration of JQ1 to mice bearing Eμ-Myc lymphomas led to robust clinical responses, however, universal treatment failure was observed despite ongoing therapy. Using RNA-Seq, disease progression and secondary JQ1 resistance was found to be associated with RAS pathway activation and Bcl-2 upregulation.
In addition, the efficacy of JQ1 was found to be dependent on an intact host immune system, where a 50% reduction in the survival advantage was observed upon transplantation into immune-deficient mice. Using RNA-Seq, the immune checkpoint ligand Cd274 (Pd-l1) was found to be potently suppressed by JQ1. Mechanistically, BET inhibition decreased Brd4 occupancy at the Cd274 promoter, leading to promoter-proximal pausing of RNA polymerase II, and loss of Cd274 mRNA production. Rapid epigenetic remodeling of the Cd274 locus in response to interferon gamma (IFN-γ) stimulation led to recruitment of Irf1, Brd4, RNA polymerase II, as well as increased local histone acetylation. Accordingly, BET inhibition suppressed constitutive and IFN-γ-induced PD-L1 expression in genetically diverse tumour models. Ectopic expression of PD-L1 in Eμ-Myc lymphomas was sufficient to reduce the efficacy of JQ1, demonstrating the significance of PD-L1 suppression to the observed therapeutic responses associated with BET inhibition. Finally, treatment of mice bearing Eμ-Myc lymphomas with JQ1 in combination with a checkpoint inhibitor (anti-PD-1) or immune stimulating antibody (anti-4-1BB/CD137) led to improved therapeutic responses.

▼ This study aims to investigate how second language (L2) learners’ first language (
L1) affects the acquisition of locative phrases in Chinese by examining the performance of L2 Chinese learners with different L1s (
L1 English and
L1 Korean learners). Locative phrases in Chinese introduced by preposition zài ‘at’ can occur in two positions in a sentence: between a
subject and a verb (preverbal position); after a verb (postverbal position). A preverbal locative phrase indicates the general location where an event happens. However, a postverbal locative phrase occurs with restrictions (Li and Thompson, 1981; Liu, 2009) and carries a distinctive semantic function indicating the location where an action ends up (Fan, 1986). These characteristics of locative phrases in Chinese cause difficulties for L2 learners. In the field of Chinese as Second Language or Foreign Language, the preposition zài ‘at’ is the most frequent preposition, but it also incurs the most errors among learners’ usage (Ding and Shen, 2001; Zhao, 2000). It has been assumed
L1 Influence is the main contributing factor (Cui, 2005; Ding and Shen, 2001), but to date relatively little empirical research has been done. The present study compares the performance between two
L1 groups. The participants were studying Chinese as a foreign language in the US and in South Korea respectively. Three experiments were conducted: a grammaticality judgment, a picture-meaning match, and an open-ended short essay. The grammaticality judgment experiment was designed to investigate how learners’
L1 affects their judgment of grammaticality; the picture-meaning match experiment explored learners’ knowledge on meaning differences between preverbal and postverbal locative phrases; the open-ended short essay experiment examined
L1 influence on the use of locative phrases in learners’ writings. Performance of the participants was compared in three ways, following the methodology suggested by Jarvis (2000): within each
L1 group (intra
L1 group), between
L1 English and
L1 Korean (inter
L1 group), and between each learner group and the NS group (inter
L1 group congruity). Results show that
L1 influence is significant only when there is a mismatch between
L1 and L2. The results confirm that preverbal locative phrases are acquired earlier than postverbal locative phrases, regardless of learners’
L1. Possible factors that may affect the acquisition sequence of Chinese locative phrases were also discussed. This study also finds evidence of avoidance (Laufer and Eliasson, 1993) in the usage of postverbal locative phrases among
L1 Korean learners.

▼ Çalışmanın amacı, 3 farklı kaynaktan (anne sütü, yeni doğan bebek feçesi, turşu suyu) izole edilen laktik asit bakterilerisinin (LAB) adiposit farklılaşması için karakterize edilen öncül adiposit 3T3-
L1 (ATCC CL-173) hücre hattı üzerindeki adipogenezi düzenleyici etkisinin belirlenmesidir. Çalışmada kullanılan suşların öncül adipositlerde adipogenezi düzenleyici etkisi; adiposit hücre farklılaşması boyunca, lipid depolanması ve bu yolda önemli bazı genlerin ifade seviyelerinin araştırılması neticesinde değerlendirilmiştir. Hücre canlılık sonuçları olgun adipositlerde; kontrol ile P. pentosaceus OZF, L. plantarum OZF, L.plantarum OZT uygulanan hücreler arasında önemli bir fark olmadığını göstermiştir. Adiposit farklılaşması boyunca, 13 gün içinde P. pentosaceus OZF, L. plantarum OZF, L.plantarum OZT varlığında hücre canlılık oranları sırasıyla % 91.15, % 88.30 ve % 85.05'in üzerinde olduğu belirlenmiştir. LAB'nin lipid akümülasyon üzerine inhibitör etkisi Oil Red O boyama ile ölçülmüştür. Sitoplazmik yağ damlacıkları üzerinde inhibitör etki P. pentosaceus OZF'de, L. plantarum OZF ve L. plantarum OZT suşlarından daha etkili olduğu belirlenmiştir. Adiposit farklılaşmasının, hücre içi doza bağlı inhibisyonu moleküler seviyede rt-PCR ile değerlendirilmiştir. Adipogenezde önemli rol oynayan bazı genlerin (transkripsiyon faktörleri PPARγ, C/EBP-α genleri, terminal adipojenik işaretleyici GPDH ve sitokin TNF-α geni) mRNA ekspresyon seviyeleri kontrolle karşılaştırıldığında, P. pentosaceus OZF uygulanan hücrelerde, L. plantarum OZF ve L. plantarum OZT'ye kıyasla anlamlı derecede azalma göstermiştir (p<0.05). Sitokin geni adiponektinin, P. Pentosaceus OZF ile muamele edilen hücrelerde diğer suşlara oranla daha fazla eksprese edildiği gözlenmiştir (p<0.05). Tüm sonuçlar birlikte ele alındığında, verilerimiz toksik olmayan konsantrasyonlarda P. pentosaceus OZF'nin öncül adipositlerin farklılaşmasını diğer suşlara oranla daha fazla inhibe ederek potansiyel anti-obezite etkileri sergileyebileceğini göstermektedir.
The aim of this study was to determine the adipogenesis regulating effect of lactic acid bacteria (LAB) isolated from three different sources (breast milk, newborn baby feces, pickle juice) on the preadipocyte 3T3-
L1 (ATCC CL-173) cell line specifically characterized for adipocyte differentiation. The adipogenesis regulating effect of the strains in the preadipocytes was evaluated as a result of the lipid storage during adipocyte cell differentiation and the investigation of the expression levels of some important genes in this pathway. Results of cell viability have shown that there is no significant difference in mature adipocytes between control and the cells treated with P. pentosaceus OZF, L. plantarum OZF, L.plantarum OZT. ▼ Os sistemas de aumento, conhecidos como SBAS (Satellite-Based Augmentation System) fornecem informações de integridade e acurácia em tempo real para seus usuários, utilizando-se das correções diferenciais que são transmitidas pelos satélites geoestacionários e estão disponíveis em algumas regiões do globo terrestre. Fora da sua região de abrangência o sistema ainda não disponibiliza os dados de correção em tempo real. Entretanto, o sinal é transmitido na mesma frequência L1 do GPS, o qual fica disponível aqui no Brasil, e alguns receptores GNSS possuem canais de frequência especifica que captam este sinal, com a perspectiva de que os dados destes satélites melhoram as condições do rastreio, além da melhoria na acurácia das coordenadas. Partindo deste princípio, foi utilizada a observável fase da onda portadora (L1) para inferir sobre a sua contribuição para o posicionamento. Os rastreios foram realizados em diferentes localidades devido à elevação dos satélites SBAS variar em função da latitude e longitude. Contudo, este estudo foi direcionado para os dados dos satélites PRN 138 (WAAS), PRN 120 e 124 (EGNOS), pois foram os únicos visíveis durante todo o rastreio, tendo simultaneidade dos dados entre os receptores base e móvel. Durante a etapa de processamento, o qual foi utilizado o software GNSS Solution, alguns experimentos foram realizados, tais como: 1) Processamento com todas as observáveis L1 dos satélites disponíveis (GPS e SBAS), análise dos desvios-padrão e comparação das coordenadas obtidas com as consideradas verdadeiras; 2) Processamento sem as observáveis L1 dos satélites SBAS, análises e comparações; 3) Retirada gradativa dos satélites GPS e reprocessamento em conjunto (GPS+SBAS) e separadamente (apenas GPS). A finalidade principal desses experimentos foi verificar a potencialidade da observável L1 dos satélites SBAS processadas em conjunto com a observável L1 do GPS, no posicionamento relativo. Análises estatísticas, como tendência e erro médio quadrático (RMS), foram aplicadas para verificar a existência de erros sistemáticos e a acurácia das coordenadas. Os resultados mostram que, especificamente nos locais de rastreio, as observáveis L1 dos satélites SBAS ao serem adicionados ao processamento, combinado com as observáveis L1 dos satélites GPS, não são determinantes para melhoria da acurácia das coordenadas.
The augmentation systems, known as SBAS (Satellite-Based Augmentation System) provide information for completeness and accuracy in real time to its users, using the differential corrections that are transmitted by geostationary satellites. They are available in specific regions of the globe such as USA, Canada and Europe. Outside their region of coverage the system still does not provide the correction data in real time. ▼ The principal intent of this research was to investigate classroom use of
L1, in this
case the Oromo language, in the English classroom at Adama Teachers College. To
this end, a descriptive research method was employed. Fifty first year regular
diploma students majoring in English and five instructors participated in the study.
Pertinent data were elicited through questionnaires and interview. Moreover,
classroom teachers' and pupils' talk were audio taped. While most of the data
gathered through the questionnaires and tape recording lessons were analyzed with
the aid of frequency and percentage, those collected through interview and the openended
items of the questionnaires were sorted out and summarized.
The study indicated that both teachers and students have positive attitude towards
the use of Oromo language at a college level. It also showed that certain amount of
Oromo language has been used in the EFL classroom: 1.28% of Oromo language,
which accounts for about 1.25% of the class time. However, the time devoted to the
Oromo language was found to be less than what was deemed acceptable by the
teachers and the students (i.e. 6-10 % of the class time) suggesting that there was a
gap between teachers' perception and their classroom practice.

Even if the gap existed, even that small amount of Oromo language used could imply
that it has played a supportive and facilitating role in the English classroom though
the effect of
L1 on L2 learning needs further studies.
What is more, the amount of Oromo language that was evident in the classroom,
together with the positive attitude of teachers and students towards its use suggests
that certain amount of Oromo language ought to be used in the English classroom.

▼ Previous research has shown L1 attrition to be selective (Gürel 2004) and often restricted to structures at the interfaces between syntax and context/pragmatics, but not to occur with syntactic properties that do not involve such interfaces (Interface Hypothesis, Sorace & Filiaci 2006). This is supported by many studies exploring cross-linguistic influence effects in interface structures, such as the production and/or interpretation of null versus overt pronominal subjects, not only in L1 attriters (Tsimpli et al. 2004, Montrul 2004) but also in other bilingual groups with different language combinations, such as early bilinguals (Paradis & Navarro 2003, Sorace et al. 2009), and advanced late bilinguals (Belletti et al. 2007, Rothman 2009). The current hypothesis is that individual L1 attrition affects only the ability to process interface structures but not knowledge representations themselves (Sorace 2011). In this thesis, we first compared a well-studied syntax-pragmatics interface phenomenon (pronominal subjects in Spanish) with a non-interface structure (the Spanish personal preposition a, also known as Differential Object Marking, DOM). In Spanish, the distribution of null and overt subject pronouns is pragmatically constrained, whereas the presence of the preposition just depends on the animacy and specificity of the direct object. Participants included a group of attrited speakers of L1 Spanish who had been living in the UK for a minimum of 5 years, and a group of Spanish monolinguals. Using a naturalness judgment task and eye tracking while reading, participants were presented with anaphoric sentences in which number cues matched or mismatched predicted antecedent preferences (i.e. null pronoun: subject preference; overt pronoun: object preference). The DOM study also used a mismatch paradigm, crossing preposition presence (al vs. el) with animacy, where an animate object requires the prepositional form al and an inanimate object requires the article el. Offline ratings revealed equal mismatch sensitivity for both groups of participants with both structures. However, eye-tracking measures showed that monolinguals were reliably more sensitive than attriters to the pronoun mismatch, while both groups showed equal on-line sensitivity to the DOM mismatch, which reveals that attrition affects interface structures, but not non-interface structures. Second, we investigated the effects of recent (re)exposure to L1 input on attrition. A second group of attriters carried out the same experiment after having been exposed exclusively to Spanish in a monolingual Spanish-speaking environment for a minimum of a week. Their eye-tracking results patterned with the monolingual group. This novel manipulation shows that attrition effects decrease as a result of L1 exposure, which reveals that bilinguals are sensitive to input changes and that attrition affects online sensitivity rather than causing a permanent change in speakers’ L1 grammatical representations.

▼ Reconstructing sparse signals from undersampled measurements is a challenging problem that arises in many areas of data science, such as signal processing, circuit design, optical engineering and image processing. The most natural way to formulate such problems is by searching for sparse, or parsimonious, solutions in which the underlying phenomena can be represented using just a few parameters. Accordingly, a natural way to phrase such problems revolves around L0 minimization in which the sparsity of the desired solution is controlled by directly counting the number of non-zero parameters. However, due to the nonconvexity and discontinuity of the L0 norm such optimization problems can be quite difficult. One modern tactic to treat such problems is to leverage convex relaxations, such as exchanging the L0 norm for its convex analog, the
L1 norm. However, to guarantee accurate reconstructions for
L1 minimization, additional conditions must be imposed, such as the restricted isometry property. Accordingly, in this thesis, we propose a novel extension to current approaches revolving around truncated
L1 minimization and demonstrate that such approach can, in important cases, provide a better approximation of L0 minimization. Considering that the nonconvexity of the truncated
L1 norm makes truncated
l1 minimization unreliable in practice, we further generalize our method to partial
L1 minimization to combine the convexity of
L1 minimization and the robustness of L0 minimization. In addition, we provide a tractable iterative scheme via the augmented Lagrangian method to solve both optimization problems. Our empirical study on synthetic data and image data shows encouraging results of the proposed partial
L1 minimization in comparison to
L1 minimization.

▼ O Papiloma Vírus Humano - HPV é um dos mais importantes vírus relacionados com a carcinogênese, já sendo conhecido seu papel na gênese do carcinoma cervical uterino. A maioria dos casos de câncer gástrico está relacionada a um agente infeccioso: Helicobacter Pylori. Outros agentes infecciosos como os vírus Epsten-Barr e HPV são relatados associados ao câncer gástrico. Objetivo: Este estudo visa investigar a associação do HPV em pacientes com câncer gástrico e comparar a um grupo controle sem neoplasia. Método: Foram analisadas 40 amostras parafinadas provenientes de portadores de câncer gástrico e 40 amostras de pacientes do grupo controle submetidos à endoscopia e biópsia de mucosa gástrica normal e sangue periférico, no período de 2008 a 2009. O material coletado foi examinado por Reação de Cadeia de Polimerase - PCR para gene da região
L1-HPV. O DNA extraído foi amplificado em dois sistemas de reação utilizando-se dois pares de primers: MY09/MY11 (fragmento de 450pb) e GP+5/GP+6 (fragmento de 150pb). Foi utilizada água Mili-Q como controle negativo da reação, sendo o controle positivo uma mistura de 01μL de sangue humano acrescido de 0,25μL de plasmídeo p BR322(HPV16). Resultados: Foram identificados em quatro pacientes com CG a presença de HPV associado à mucosa tumoral, nos pacientes sem neoplasia gástrica foram identificados 10 casos de HPV associado à mucosa gástrica. Conclusão: No presente estudo não encontramos indícios de associação significativa do HPV ao câncer gástrico (p=0,077).

▼ Regenerating gene (REG)Iα is known to contribute to carcinogenesis and to be associated with a poor prognosis in various cancers. Programmed death-1 ligand(PD-L1) is a negative regulator of T cell activation thought to play an important role in tumor evasion from host immunity.In the present study, we tested the hypothesis that the pro-survival effects of REG Iα in cancer reflect enhanced expression of PD-L1.We found that PD-L1 mRNA expression tended to correspond to REG Iα mRNA levels in esophageal squamous cancer cells, and that REG Iα expression significantly increased expression of both PD-L1 mRNA and protein in TE-5 and TE-9 squamous esophageal cancer cells transfected with REG Iα.In addition, immunohistochemical analysis of squamous cell esophageal cancer specimens revealed that the spatial distribution of PD-L1 expression corresponded to that of REG Iα. These findings suggest that REG Iα may suppress antitumor immunity by inducing PD-L1 expression.

▼ Writing competently is a challenge for many college students. The need to improve writing at the postsecondary level has become a priority for business and education leaders alike in the United States. In the United States, for those who have not developed proficient English reading and writing skills, academic progress may be hampered on campuses where English is the primary language of instruction. Writing, especially for the diverse student population at the community college level, warrants attention in order to better guide instruction and assist students in achieving higher levels of proficient writing.
This study examined the writing of three groups of community college students: native English Language students (
L1, n = 146), English as a Second Language students (L2, n = 33), and English as a Second Language students who graduated from high school in the United States and have lived in the United States for four or more years (Generation 1.5, n = 72). Writing samples and biographical survey information were gathered from students and samples were analyzed using the online tool, Coh-Metrix. The research questions used to guide this investigation examined the differences between these groups in terms of lexical, syntactic, and cohesion characteristics of proficient writing, as well as declarative and procedural knowledge of writing, and the impact of age of acquisition (AOA), parental education levels, and number of years of education in the United States on L2 and Generation 1.5 students’ writing.
Results indicated significant differences in syntactic and lexical measures between all groups, with effect sizes ranging from small to large. The majority of differences related to proficient writing were found between
L1 and Generation 1.5. Responses regarding declarative knowledge, students focused mainly on clarity, audience, grammar, and spelling. Planning and revising were considered much less important. Responses regarding procedural knowledge focused on goal setting/planning, establishing purpose, writing, and revising, with less importance given to spelling and grammar. Finally, age of acquisition accounted for 10 % of the variation in students’ writing, specifically syntactic patterns, with parental education level and years of English studied in the United States having no significant impact.

▼ The intent of this survey study was to explore the use of
L1 (in this case Hadiyyisa) in the English classroom in Hadiya zone primary schools. To this end, a descriptive research method was employed in the study. Ten grade five English teachers and one hundred grade five students selected by availability and systematic sampling were participants of the study. Data were gathered through classroom observation, questionnaires and interview. The classroom teachers and students talk was recorded and transcribed. The data gathered through the questionnaires and the recorded observation were analyzed using frequency and percentage, while those collected through interview and open-ended items of the questionnaires were qualitatively analyzed. The study depicted that both teachers and students positively perceive the use of Hadiyyisa in the English classroom. The study also divulged that teachers and students believe in the various pedagogic uses of Hadiyyisa the major ones being: explaining new words, explaining difficult concepts, giving instructions, maintaining classroom discipline and checking students' comprehension. The study further revealed that most of the teachers (80%) and students (65%) appear to be in favor of using Hadiyyisa ‘sometimes’. However, actual classroom practices uncover excessive use of Hadiyyisa, about 20% out of the total English classroom talk. Finally, it was suggested that English language teachers should be given appropriate training in higher institutions .Further, appropriate workshops, seminars , etc. with regard to ‘how’, 'when', and 'how much'
L1 should be used in the L2 classroom.

▼ Leptin, a peptide hormone secreted by adipocytes, has been widely studied in the attempt to understand its contributions and relation to obesity. There is a sexual dimorphism in leptin levels between men and women, suggesting a possible influence of steroid hormones on leptin expression and signaling pathways. Likewise, evidence suggests that leptin gene expression is regulated by steroid hormones in humans and mice. Leptin signals through a membrane receptor, Ob-R, and expression of the Ob-R isoforms are also sexually dimorphic, and suggest that steroid hormones may also control receptor expression. The experiments described here attempt to better understand the potential relationship between steroid hormones and leptin receptor (Ob-R) gene expression. This hormonal regulation may result in leptin resistance in men and women, increasing risks for prolonged and irreversible obesity.

▼ Cette thèse étudie différents problèmes de minimisations avec des fonctions de régularisations qui promeuvent la parcimonie. Plus précisément, on souhaite reconstruire une image, que l'on suppose parcimonieuse et qui a subit une transformation après un opérateur linéaire, à l'aide de problèmes de minimisations. Dans ce manuscrit, on s'intéressera plus particulièrement à la minimisation l1 synthèse, analyse et bloc qui sont très utilisées pour reconstruction une image que l'on sait parcimonieuse. Ces minimisations produisent en pratique des résultats convaincants qui n'ont été compris théoriquement que récemment. Les différents travaux sur le sujet mettent en évidence le rôle d'un vecteur particulier appelé certificat dual. L'existence d'un certificat dual permet à la fois d'assurer la reconstruction exacte d'une image dans le cas où il n'y a pas de perturbations et d'estimer l'erreur de la reconstruction en présence de perturbations. Dans nos travaux, nous allons introduire l'existence d'un certifical dual optimal pour la minimisation l1 synthèse qui minimisent l'erreur de reconstruction. Ces résultats ayant une forte interprétation géométrique, nous avons développé un critère identifiabilité, c'est à dire que ce critère assure que l'image recherchée est l'unique solution du problème de minimisation. Ce critère permet d'étendre nos travaux à la minimisation l1 analyse, l1 bloc et à d'autres cas.
This thesis studies different minimization problems with sparses based regularization. More precisely, we want to reconstruct a sparses image, which undergone a linear transformation, with minimization problems. In this manuscript, we will be focused on l1 synthesis, analysis and block minimization which are widely used in sparse approximations. These problems offer competitive results which are theorietical understood only recntly. Different studies on the subject emphasized the contribution of a particular vector called dual certificate. The existence of this dual certificate allows simultaneously to guarantee the exact recovey of an image in noiseless case and to estimate the noise robustness in noisy case. In this work, we introduce eth existence of an optimal dual certificate for the l1 synthesis minimization which minimizes the reconstruction error. As those results have a strong geometrical interpretation, we develop an identifiability criterion which ensures the uniqueness of a solution. This criterion generalizes the work on l1 synthesis minimization tothe analysis case, block case and others.

▼ Osteosarcoma (OS) is a mesenchymal tumor that is the most common malignant bone tumor in children and young adults. Our objective was to identify novel pathways in OS that may be related to the expression of an immunosuppressive protein ‘programmed cell death ligand 1 (PD-L1)’. Using RT-qPCR, varying levels of relative PD-L1 gene expression was shown in 69 primary OS tumors. Ingenuity pathway analysis was utilized to analyze 400 significantly differentially expressed genes between the high and low PD-L1 expressing OS tumors and the elongation initiation factor 2 (EIF2) pathway was illustrated to be significantly down-regulated (p=3E-07; z-score=-2.5) and the ‘cell death of OS cells’ was predicted to be significantly activated in OS tumors with high PD-L1 expression (p=1E-04; z-score=2.53). Both IFN- and thapsigargin were utilized to observe the effects of attenuating the EIF2 pathway on PD-L1 expression. Overall, the down-regulation of the EIF2 pathway did not influence PD-L1 expression.

▼ This thesis aims at investigating occurrences of error types produced by
L1 Hungarian
learners of L2 English from the perspective of whether
L1 transfer errors represent an
outstanding figure among other categories and therefore serve as the most influential
obstacles in the SLA process overtaking intralingual and developmental error types.
Composing another interest area of the study, the hypothetical dominance of interlanguage,
that is transfer errors raise the issue whether grammatically incorrect and semantically weird
TL constructions can be, and if it is so, on what bases, attributed to two methods of L2
application: translating to the target language vs. thinking in the L2. The types of
inappropriate grammatical structures in L2 were scrutinized among elementary school
students progressing in the 8th grade and fitting to the requirements of the lower-intermediate
level. Two bilingual and two conventional language learner groups were tested by 3 different
tasks focusing on error inducement and error recognition which also intended to study the
methods of SL usage formerly described here. The participants were also asked about facts
regarding learning English even as which task(s) had proved to present difficulty. The test
results as well as statistics calculated let it draw the conclusion that although such tests were
prepared for participants that tried to eliminate the potential emergence of ungrammatical
constructions resulting from transfer by their nature, the majority of errors generated by the
learners compose
L1 transfer errors irrespective of bilingual or conventional classes.
Moreover, these results may be contributed to the assumption that the participants applied
mere translation instead of formulating their thoughts in the TL.

▼ Nonlinearity is one of major challenges in modern engineering system that we need to deal with in the control strategy design. In contrast to a linear system, the superposition principle normally cannot be simply applied to a nonlinear system. The feature of nonlinear system varies significantly from one to another, which brings more difficulty to conclude a uniform controller design methodology than the linear system. In addition, most of systems contain the feature of constraints for both input and/or output implicitly or explicitly, which makes it more complicated for the controller to maintain within required margins. The nonlinearity and constraints may not necessarily appear simultaneously in a system, but the resulted challenges in control system design cannot be ignored or avoided.
The basic design of
L1 adaptive control aims to solve the nonlinear system with guaranteed response. In the absence of constrained output, however, the traditional
L1 adaptive control design structure needs to be revised and adjusted by incorporating other control design strategy such as model predictive control so that the basic scope of
L1 adaptive control can be extended to provide effective solutions to such problems.
The objective of this thesis is to solve the controller design problem for the system in combination of nonlinearity, output constraints based on the fundamental
L1 adaptive control structure with adequate modifications. The system dynamic feature of the process that has been investigated in this thesis differs from case to case though, a uniform design method for the
L1 adaptive control has been generated to solve those problems as a whole.

▼ The l∞ performance of Linear Time-Invariant (LTI) systems has been one of the corner stones of the robust control theory for over the past 30 years. The l∞ performance has been studied mostly for LTI systems and the scarcity of the results for other types of systems is prominent in this area. This dissertation aims to depart from LTI systems and investigate the l∞ performance for other classes of systems. In particular, the l∞ performance of Linear Switched Systems (LSS) and of linear systems with cone constraints is studied in the first and second part of this dissertation, respectively.
Part I: In Part I, we first consider the worst-case l∞ induced norm computation of LSS. That is, sup_σ‖G_σ‖, where G_σ is a LSS, σ is the switching sequence, and the norm, ‖.‖, is the l∞ induced norm. This problem can be linked to robustness of systems when the switching is arbitrary. We provide lower and upper bounds of this quantity. These bounds are hard to compute and in general conservative. Hence, we narrow our attention to special classes of LSS by defining the classes of input, output, and input-output LSS and show that for these classes, exact expressions for the worst-case l∞ induced norm can be found. Moreover, we introduce the class of generalized input-output LSS and show how their l∞ gains can be computed exactly via Linear Programming (LP). The class of generalized input-output LSS proves to be a sufficiently rich class as it is dense in the set of all stable LSS. We further derive new stability and stabilizability conditions and control synthesis in terms of LP utilizing generalized input-output LSS.
The other extreme from the worst-case norm is the minimal norm, i.e., inf_σ ‖G_σ‖. The interest in this type of problem is motivated by situations where there may be limited sensor and/or actuator resources for filtering and control. We show that for Finite Impulse Response (FIR)switching systems the minimizing switching sequence can be chosen to be periodic. For input-only or output-only switching systems an exact characterization of the minimal l∞ gain is provided, and it is shown that the minimizing switching sequence is constant, which, as also shown, is not true for input-output switching.
Moreover, we study Markov Linear Switched Systems (MLSS). These are LSS whose switching sequence is a Markov process. We introduce the notion of the stochastic l∞ gain and provide exact expression to compute it. However, this computation is challenging, as we show, and hence we resort to a more relaxed but tractable notion of l∞ mean performance. We provide tractable computation and control synthesis method with respect to the l∞ mean performance.
Part II: Part II of this dissertation deals with the l∞ gain of linear systems with positivity type of constraints. are related mainly to their polyphenol content. Catechins are the major component
of the green tea polyphenols (Balentine, Wiseman, & Bouwens, 1997) Catechins
are the major source of flavonols and they are comprised of various types of
catechins such as epigallo catechin 3 gallate (EGCG), epigallocatechin (EGC),
epicatechin gallate (ECG) (Sano, et al., 2001). Many studies have reported and
demonstrated that EGCG can exert anti-obesity effects.
Obesity is considered the major risk factor for type 2 diabetes mellitus
(T2DM), cardio-vascular diseases like hypertension and atherosclerosis (Serrano
Rios, M., 1998 (Serrano Rios, 1998). Being slightly overweight can also lead to
the onset of life-style related health diseases (Wolfram, Wang, & Thielecke,
2006). Considering all the health related diseases linked to obesity, it can lead to
metabolic syndrome hence, prevention of obesity is gaining the major attention
over the treatment.
Number of studies demonstrated that EGCG can inhibit lipid accumulation
in adipocytes (Lee, Kim, & Kim, 2009). But, it has not been proved to reduce the
fat mass in humans and research animals. The major reason behind this problem is
the EGCG bio-availability which is very low in animals as well as in humans.
Along with the problem of bio-availability, another factor is instability of EGCG
and higher metabolic degradation. Therefore, to gain maximum benefit of this
biologically functional molecule, it is important to use biocompatible and
biodegradable nanoparticles in order to increase its stability and absorption
thereby, enhancing its bio-availability.
Nanotechnology is promising area which offers higher EGCG bio- availability and stability, at which EGCG can be used to exert its beneficial health
effects in reducing lipid accumulation.

L1-cellular adhesion molecule (L1CAM, or
L1) and ankyrins B and G interaction. Neurite arborization is compromised due to a loss of L1CAM and ezrin-radixin-moesin (ERM) protein interaction. L1CAM and neuropilin-1(NP1) interaction leads to a loss of neurite directionality.
How these neurite growth changes affect neurogenesis throughout the brain, and lead to microcephaly (MiC), has not yet been modeled. Future drug discovery projects aiming to halt the progression of postnatal MiC in MASA syndrome could narrow the scope of focus if modeling software parameters were adjusted to reflect phenotypic changes to neurites lacking
L1 demonstrated in the literature.

▼ The somatic mutations found in melanomas reflect the biological processes that govern tumour development. They also help shape how tumours evolve and escape immune regulation. Studying these changes can therefore help to refine our understanding of melanoma progression with the added potential to offer new perspectives for disease management.
Most prior melanoma sequencing studies have focused on advanced disease. Thus, somatic alterations that influence the behaviour of early-stage tumours have not been fully explored. Consequently, in this thesis I study a collection of 524 primary melanomas on which extensive clinical data have been collected for almost two decades. I describe the mutational landscape of these tumours including driver genes, new recurrent variants, mutually exclusive genetic interactions and copy number alterations. I discuss and associate these features with aspects of tumour pathology, sun exposure, immunogenicity and patient outcomes. To identify genes required for melanoma survival, I intersect my genomic analysis with a dataset of CRISPR-Cas9 dropout screens and discover a melanoma-associated genetic vulnerability mediated by Interferon Regulatory Factor 4 (IRF4). I then begin to experimentally validate and explore the biological pathway by which IRF4 may function in the context of melanoma.
Checkpoint inhibitors have revolutionised melanoma care, yet only a minority of patients respond to these treatments and our comprehension of the mechanisms governing PD-L1 expression on melanoma cells is still limited. In the second part of this thesis, I examine the regulation of the key checkpoint receptor PD-L1, which is often upregulated in melanoma to facilitate tumour escape. To improve our understanding of the processes controlling PD-L1 expression, and how the PD-1/PD-L1 axis can be targeted to overcome immune evasion, I employ a genome-wide CRISPR-Cas9 screening approach. I identify genes which elicit downregulation of PD-L1 when disrupted in melanoma cells, capturing several central processes including basal transcription, N-linked glycosylation and intracellular transport. A second extensive screen in eight cancer cell lines of melanoma, bladder and lung cancer origin validate these findings and link novel candidate genes, including Sphingolipid Transporter 1 (SPNS1), to the control of PD-L1 cell surface expression. Additional work is required to further validate and understand the regulation of PD-L1 through SPNS1, as well as its contribution to immune surveillance.
In summary, I present the first comprehensive evaluation into the somatic alteration landscape of primary melanomas, gaining insight into the molecular architecture of these tumours. Additionally, I introduce novel genes and processes regulating PD-L1 gene transcription, processing and presentation on the cell surface. Collectively, these results improve our understanding of the genetic processes that govern primary melanomas, as well as providing valuable insights into the mechanism of PD-L1 regulation.

L1-regularization, Lasso, is a popular algorithm for recovering the sparsity pattern (also known as model selection) in linear models from observations contaminated by noise. We examine a scenario where a fraction of the zero co-variates are highly correlated with non-zero co-variates making sparsity recovery difficult. We propose two methods that adaptively increment the regularization parameter to prune the Lasso solution set. We prove that the algorithms achieve consistent model selection with high probability while using fewer samples than traditional Lasso. The algorithm can be extended to a broad set of
L1-regularized M-estimators for linear statistical models.

▼ Cardiovascular disease is the leading cause of mortality in all developed nations. Several independent studies have shown that cardiotrophin-1 (CT-1) serum levels are modulated in patients with various types of cardiovascular disease including ischemic heart disease, valvular heart disease, and accelerated arthrosclerosis. CT-1 is a member of the Interleukin-6 family, or gp130 family of cytokines. It is also known to induce cardiomyocyte hypertrophy in vitro and in vivo, and is a critical component for cardiomyocyte survival. CT-1 is a naturally occurring protein with a molecular mass of approximately 21.5 kD and a 200 amino acid long sequence, it was discovered in a cDNA screen of murine stem cells and was originally identified in cardiomyocytes. Since then, CT-1 expression has been reported in several other tissues including skeletal muscle, liver, ovary, kidney and lung. Interestingly, our studies reveal that CT-1 protein expression is significantly modulated in adipose tissue following high fat feeding in C57BL/6 mice. Of note, we did not observe regulation of CT-1 expression in white adipose tissue in human obesity and Type II Diabetes Mellitus (T2DM). We can readily detect CT-1 in the media of cultured adipocytes. This study suggests that CT-1 secretion from murine adipose tissue may be an important contributor to the levels of circulating CT-1. In summary, our studies demonstrate that CT-1 is an adipokine that is modulated in murine obesity and T2DM.

Losso Pages in thesis, 43. Words in abstract, 281. ABSTRACT Obesity has become a major health problem worldwide. Obesity increases the risk of hypertension, diabetes, and certain types of cancer. Quercetin is a bioactive compound widely found in a variety of foods that are consumed daily. Isorhamnetin is a bioactive compound found in some foods and also is a quercetin metabolite. Many studies have reported the anti-obesity and anti-inflammation properties of quercetin and isorhamnetin. The objective of this study was to test the effect of quercetin or isorhamnetin at physiological and supraphysiological concentrations on the inhibition of the differentiation process of 3T3-L1 pre-adipocyte to adipocyte. Cell viability results demonstrated no significant difference (P > 0.05) between non differentiated cells, control and quercetin or isorhamnetin treated cells. During adipocyte differentiation for 8 days in the presence of quercetin or isorhamnetin, cell viability was above 94.84% and 97.63%, respectively. Red oil O staining assay was performed in order to evaluate the inhibitory effect of quercetin or isorhamnetin on cytoplasmic lipid droplet accumulation. Significant differences (P < 0.05) were reported. Isorhamnetin was more effective than quercetin in inhibiting cytoplasmic lipid droplet accumulation. Neither quercetin nor isorhamnetin had an effect on the expression of macrophage chemoattractant protein -1 (MCP-1). CCAAT/enhancer binding protein α (C/EBP-α) was down-regulated by quercetin or isorhamnetin. Compared to control quercetin decreased PPAR-γ 1 and 2 expressions by 45.03 ± 3.17% and 27.58 ± 12.39%, while isorhamnetin decreased PPAR-γ 1 and 2 expressions by 41.48 ± 9.51% and 2.01 ± 32.46%, respectively. β-catenin was not dose dependent either for quercetin or isorhamnetin and did not follow a specific trend. Taken together, our data indicate that isorhamnetin more than quercetin can exert potential anti-obesity effects by inhibiting differentiation of pre-adipocytes at physiological concentrations.

▼ Adipocytes function as the major storage site for triacylglyerol (TAG) and have important roles as active endocrine cells. It is widely assumed that the endocrine function of fat cells is determined by their size. As adipocytes increase in size, they tend to release more pro-inflammatory secretions leading to the recruitment of macrophages, setting up a vicious cycle of inflammation. The aim of this study was to genetically manipulate the size of fat cells in culture to determine the effect on inflammatory behaviour. The size of adipocytes may be manipulated in vitro by increasing TAG storage in 3T3-L1 cells. Acyl-coenzyme A:diacylglycerol acyltransferase 1 (DGAT1) catalyses the final and only committed step of TAG synthesis. Therefore key strategies in this study were to achieve overexpression of DGAT1 in 3T3-L1 preadipocytes, assess the resulting phenotype following differentiation, and to examine the adipocyte response to inflammatory stimuli as well as interaction with RAW 264.7 macrophages. Interactions between the cells were simulated by incubating the 3T3- L1 adipocytes with secretions collected from RAW 264.7 macrophages and vice versa. DGAT1 was overexpressed in 3T3-L1 preadipocytes via recombinant lentivirus and resulted in a more than 16-fold increase in mRNA compared to control cells transduced with an “empty” reporter lentiviral vector. Prior to differentiation, the DGAT1 overexpressing preadipocytes showed elevated intracellular TAG accumulation and the formation of distinct lipid droplets, whereas the control cells did not demonstrate this behaviour. After differentiation, the increase in lipid was unexpectedly not maintained, characterised by a lower rate of lipogenesis and smaller adipocyte size. Assessment of adipocyte interactions with RAW 264.7 macrophages revealed both the recombinant DGAT1 and control vectors were associated with elevated inflammatory gene expression in response to macrophage secretions. Similarly, exposure to adipocyte secretions obtained from both populations of the transduced 3T3-L1 cells led to increased RAW 264.7 macrophage activation compared to secretory factors collected from non-transduced adipocytes. Genetic manipulation of 3T3-L1 cells by lentiviral transduction led to elevated inflammation which appeared to be due to activation of an immune response to the vector. Overexpression via the recombinant DGAT1 vector resulted in an unexpected reduction in adipocyte size which did not occur in the transduced control cells despite both populations having an impaired inflammatory profile. This loss of adipogenic behaviour was likely attributable to interference with the complex differentiation process due to an excessively high gene expression load, rather than the DGAT1 transgene itself. These results emphasise the need to exercise caution when using genetic manipulation techniques with the 3T3-L1 cell line, particularly because changes in inflammatory profile can occur without alterations in observable traits and may therefore be difficult to detect.
