▼ Single crystals provide the purest form of samples
for basic research and also serve as the base components for large
number of devices. It has been realized that nonlinear optical
(NLO) effect in crystals could be exploited in the visible and
infrared region in many ways after the discovery of lasers. The
present thesis with 6 chapters discusses the details of
investigations carried out on the growth and characterization of
some amino acids (L-arginine, L-histidine and L-tyrosine) based
semiorganic nonlinear optical single crystals grown by low
temperature solution growth technique. Effect of neutral amino
acids on the growth and its properties of LAP single crystals have
been investigated. In order to study the improved characteristics
of L- arginine phosphate (LAP), amino acids mixed LAP crystals have
been grown by slow cooling method. Optical absorption studies have
been carried out using UV-Vis- NIR spectrophotometer. Thermal
behavior of the amino acids mixed LAP crystals has been studied by
TG and DTG analyses. Nonlinear optical characteristics have been
studied using Q switched Nd:YAG laser and#411;= 1064 nm). Optical
transmission studies confirm that LTHB is transparent in the entire
visible region and there is no absorption of light to any
appreciable extent in the visible range. Mechanical strength of the
LTHB crystal is found to be promising. Thermal analysis reveals
that the sample is thermally stable up to 284°C. The NLO property
studies confirmed that the second harmonic conversion efficiency of
the crystal is found to be better than KDP. Hence, it is concluded
that the optical, NLO and thermal properties confirm the
suitability of LTHB crystal for photonic device applications.
L-tyrosine hydrochloride single crystal has also been grown by slow
evaporation technique. The structural and thermal properties of the
grow crystal have been discussed. newline

▼ Nos últimos anos, a aplicação de DNA plasmídeo (pDNA) em terapia génica ou vacinas de DNA
tem-se revelado uma ferramenta com potencial aplicação no tratamento de inúmeras
doenças. Uma vez que esta abordagem recorre a vetores não-virais, é mais segura em termos
de ativação de oncogenes e apresenta uma diminuição de reações do sistema imunitário.
Deste modo, a indústria farmacêutica procura obter pDNA em maiores quantidades e isento
de contaminantes (RNA, DNA genómico, endotoxinas, proteínas). Uma técnica que permite
purificar o pDNA é a cromatografia de afinidade (AC). Esta metodologia baseia-se em
interações específicas entre ligandos e o pDNA, permitindo a eliminação de alguns passos de
purificação. A utilização de aminoácidos como ligandos de afinidade revelou ser uma
abordagem promissora para purificar ácidos nucleicos, pois baseia-se na bioespecificidade
aminoácido-nucleótido. A L-histidina e a L-arginina permitiram isolar a isoforma
superenrolada (sc) do pDNA, indicando a presença de interações específicas entre o pDNA e os
aminoácidos. Neste trabalho são apresentados os resultados de três ligandos, a L-tirosina e os
dipeptídeos L-tirosina-L-tirosina e L-tirosina-L-arginina. Os dipeptídeos foram sintetizados de
acordo com o método clássico de síntese química, que envolve a proteção do grupo amina e
do grupo carboxílico de cada aminoácido, e posterior formação da ligação peptídica e por fim
desproteção usando TFA (ácido trifluoracético). De seguida, os ligandos foram imobilizados na
Sepharose CL-6B através da ativação dos grupos epóxi com o reagente 1,4-butanediol
diglicidil éter. O suporte foi caracterizado por HR-MAS (Ressonância Magnética de Alta
Resolução de Ângulo Mágico) RMN. A técnica de STD-RMN (Diferença da Transferência de
saturação por ressonância magnética nuclear) permitiu identificar os grupos preferenciais dos
5´-mononucleótidos que interagem com os suportes Sepharose-L-tirosina, Sepharose-Ltirosina-
L-tirosina e Sepharose-L-tirosina-L-arginina. Utilizando a técnica de RPS (Ressonância
Plasmónica de Superfície), foi analisada a afinidade de cada um dos ligandos em estudo em
relação a cada uma das isoformas de pVAX1-LacZ a diferentes temperaturas (T=10°C e 25°C)
e usando diferentes tampões (Tris-HCl 10 mM e Hepes ácido 10 mM). Desta forma, a utilização
das técnicas STD-RMN e SPR permitiu fazer o screening das interações dos suportes-5’-
mononucleótidos e determinar a afinidade do complexo ligando: pDNA, para futura aplicação
em AC.
In the last years, the use of plasmid DNA (pDNA) in gene therapy or DNA vaccines has been
proved to be a tool with potential application in the treatment of diseases. Since this
approach uses non-viral vectors, it is safer in terms of oncogene activation and it presents a
decrease of the immune system response.
In this way, the pharmaceutical industry needs to obtain pDNA in larger quantities and free of

▼ Schizophrenia is a complex neurodevelopmental illness
that often requires a combination of treatments to adequately
control its symptoms. Despite the availability of several
neuroleptic medications on the market, many patients with
schizophrenia remain symptomatic. Research has shifted emphasis
towards impaired glutamate signaling as a major contributor to the
illness. Drug targets specific to the glutamate-nitric oxide (NO)
pathway may offer a promising new approach to treatment. The
therapeutic effect of the amino acid L-arginine (Arg), the
substrate for the NO-producing enzyme neuronal nitric oxide
synthase (nNOS) was examined in this thesis. As an augmenting
strategy, Arg was administered (at 6g per day) to patients with
schizophrenia in an attempt to improve the efficacy of
antipsychotic therapy. Responders to Arg therapy had a reduction in
anxiety symptoms that were previously resistant to prescribed
medication. The anxiolytic effect of Arg may be directly related to
an increase in endogenous central NO production and the downstream
induction of cyclic guanosine monophosphate (cGMP)-mediated
signaling cascades. L-Arginine also proved to be a safe strategy in
the treatment of schizophrenia and did not exacerbate any of the
known side-effects that were already present as a result of ongoing
antipsychotic therapy. This thesis also investigated genetic
influences on glutamate N-methyl-D-aspartate (NMDA) receptor
functioning and treatment response to Arg therapy. The Neuronal
Period-Arylhydrocarbon Nuclear Translocator (ARNT)-Single-minded
(PAS) domain containing 3 (NPAS3) gene codes for a protein that
functions as a transcription factor. NPAS3 and its related variants
may be important for the development of functional postsynaptic
density (PSD-95) scaffolding proteins that directly link the NMDA
receptor to the nNOS enzyme in the brain. Variants associated with
NPAS3 and schizophrenia were examined to elucidate the integrity of
the PSD-95 protein and used to predict the outcome of the effects
of adjuvant Arg therapy. Two carriers of a NPAS3 variant who did
not respond to Arg treatment may have had compromised NMDA receptor
function related to altered neurodevelopmental processes that
occurred during early brain development. Participants were also
genotyped for the catechol-O-methyltransferase (COMT ) enzyme
single nucleotide polymorphism (SNP) p.V158M to explore the
relationship between this genetic marker and clinical outcomes. No
significant gene-gene interaction between NPAS3 and COMT was
determined that would influence treatment response to Arg therapy.
Preclinical experiments were also included in this thesis to
determine the behavioural and neurochemical mechanisms by which Arg
was able to exert its therapeutic effect. Competing metabolic
enzymes that utilize Arg as a substrate were investigated by
measurement of related amino acids ornithine (Orn) and citrulline
(Cit). The immediate potentiation of whole brain Cit levels
following Arg administration suggested that NO is produced quickly
and NOS enzymes…

▼ Optimal immune function is crucial for the
best possible performance in elite athletes and the weekend warrior
alike. Research shows that sedentary people do not have as vigorous
an immune system as those who habitually exercise. However,
moderate exercise has been shown to drastically improve immune
response. The primary goal of this study was to
examine the effects of variable dosage of
l-
arginine on white blood
cell count (WBC) and platelet count before and after VO2max testing
of healthy males. The amount of
l-
arginine was also examined to
determine the effectiveness of supplementation per dose.
The current study comprised of twenty-five male
subjects of at least 18 years of age, but younger than 30 years of
age. After the initial visit the subjects were to return for three
subsequent visits, which consisted of a fasted pre-treatment blood
draw upon arrival. The treatment consisted of three ounces of lemon
juice, five ounces of water, one packet of lemonade flavoring, and
one of the three treatments, placebo, 3g or 6g. Treatment was then
given after the pre-treatment blood draw in a double blind randomly
assigned order. After a 25 minute absorption period had elapsed, a
second blood draw was taken. Testing then occurred, which included
a VO2max test utilizing a standard Bruce Protocol on a treadmill.
Upon completion of the VO2max test, a third blood draw was taken
one minute post-exercise. Blood samples were later sent for a
complete blood count (CBC) analysis to measure WBC and platelet
count. In this study, the level of significance
was set a priori at (P < 0.05). Post-hoc analyses was performed
where applicable to determine significant interactions. There was
no statistically significant relationship between dose and WBC.
With this information, neither the dose of 3g or 6g of
l-
arginine
nor the placebo showed a significant alteration to WBC, thus
indicating that no matter the dosage of
l-
arginine, there was no
significant effect on WBC among the subjects. There was a
statistically significant increase in WBC over time regardless of
dosage from pre-exercise to post-exercise testing. There was no
statistically significant relationship between treatment dose and
platelet count. Neither the dose of
l-
arginine nor the placebo
showed a significant alteration to platelet counts. There was a
statistically significant change of platelet count from
pre-exercise to post-exercise testing. Concluding that regardless
of dosage, platelet count increased post exercise.
The study concludes that
l-
arginine had no
significant effects on pre and post-exercise WBC and platelet
count, regardless of
l-
arginine dosage. The study did, however,
show a definitive relationship between time and both the WBC and
platelet count, thus indicating and proving that exercise itself
does evoke an immune response.

▼ Os aminoácidos L-arginina e L-glutamina foram analisados como protetores dos tecidos erétil do pênis contra os danos induzidos pela radiação. Grupos de ratos Wistar foram tratadas com: nenhuma intervenção, radiação pélvica e sacrifício 7 (RAD7) ou 15 (RAD15) dias; e radiação pélvica, a suplementação diária com L-arginina (A) ou L-glutamina (G), e sacrifício 7 (RAD7 + A, RAD7 + G) ou 15 (RAD15 + A, RAD15 + G) dias após irradiação. componentes estruturais do corpo cavernoso (CC), túnica albugínea do corpo esponjoso (TAC), e urotélio do pênis foram analisados através de métodos estereológicos e imuno-histoquímico. Os resultados mostraram que, no CC, o tecido conjuntivo foi maior nos RAD15 (p <0,04), mas essa mudança foi parcialmente revertido em RAD15 + G (p <0,05) e RAD15 + A (p <0,04). A matriz fibrosa das trabéculas CC estava manchada de colágeno tipo I. Nos RAD15, a intensidade da marcação foi aumentada, enquanto que em RAD15 + G + A e RAD15 a coloração era semelhante à dos controles. Nenhuma alteração de coloração foram observadas nos grupos que foram sacrificados sete dias após a radiação. Cavernosa teor de fibras elásticas na RAD15 foi aumentada (p <0,004), e este foi impedido de RAD15 + A (p <0,004), mas não em RAD15 + G. No TAC, os aminoácidos protegidos (p <0,02) contra o aumento da radiação induzida em fibras elásticas, mas apenas em RAD15. Densidade das células do urotélio e espessura urothellial, foram reduzidos em RAD15 (p <0,004), mas houve efeitos protetores dos dois aminoácidos. Em conclusão, a radiação induzida por alterações nas estruturas penianas tendem a ser mais pronunciado 15 dias após a sessão de radiação. Tanto A e G têm efeitos protetores contra estas alterações, sendo o primeiro um pouco mais eficaz.
We investigated whether arginine and glutamine protect penile tissues against radiation-induced damage. Groups of Wistar rats were treated with: no intervention; pelvic radiation, and sacrifice 7 (RAD7) or 15 (RAD15) days later; and pelvic radiation, daily supplementation with L-arginine (A) or L-glutamine (G), and sacrifice 7 (RAD7+A, RAD7+G) or 15 (RAD15+A, RAD15+G) days after radiation. Structural components in the corpus cavernosum (CC), tunica albuginea of the corpus spongiosum (TACS), and urothelium of the penis were analyzed using stereological and immunohistochemical methods. The results showed that in the CC, connective tissue was increased in RAD15 (p<0.04), but this change was partially prevented in RAD15+G (p<0.05) and RAD15+A (p<0.04). The fibrous matrix of the CC trabeculae was stained for collagen type I. In RAD15, the intensity of the labeling was increased, whereas in RAD15+G and RAD15+A the staining was similar to that of the controls. No staining changes were seen in the groups that were sacrificed seven days after radiation. Cavernosal elastic fiber content in RAD15 was increased (p<0.004), and this was prevented in RAD15+A (p<0.004), but not in RAD15+G. ▼ A radioterapia é amplamente empregada no tratamento de tumores abdominais. Entretanto, o seu emprego provoca danos indesejáveis ao tecido sadio, especialmente o intestinal, refletidos por meio de manifestações clínicas e histológicas. Buscando minimizar esses efeitos colaterais, inúmeras alternativas vêm sendo estudadas, dentre elas a suplementação dietética com aminoácidos. O objetivo deste estudo foi determinar o efeito da suplementação dietética com os aminoácidos L-glutamina, L-arginina e glicina nas camadas submucosa e muscular do íleo de ratos submetidos a irradiação abdominal. Cinqüenta ratos Wistar machos adultos foram distribuídos aleatoriamente em cinco grupos: I Controle não irradiado e sem suplementação de aminoácidos; II Controle irradiado e sem suplementação de aminoácidos; III irradiado e suplementado com L-glutamina; IV irradiado e suplementado com glicina; V irradiado e suplementado com L-arginina. O período de suplementação dietética foi de 14 dias, com a irradiação ocorrendo no 8. dia do experimento. A irradiação provocou diminuição da espessura da camada submucosa dos animais do grupo II, como também do conteúdo de colágeno, em comparação ao grupo controle não irradiado. A suplementação com L-arginina e glicina provocou aumento expressivo da espessura da camada submucosa, enquanto a suplementação com L-glutamina manteve a espessura dessa camada similar à observada nos animais não irradiados. Os animais com dieta suplementada com L-arginina e glicina também apresentaram aumento da espessura da camada muscular circular interna, em comparação aos ratos dos grupos I, não se observando diferenças no que se refere à camada muscular longitudinal externa. A suplementação com aminoácidos levou a aumento na intensidade de imunomarcação de colágeno tipo I nos animais do grupo IV, menos marcante no grupo III, e o grupo V foi o que mais se assemelhou ao observado nos animais não-irradiados. No tocante ao colágeno tipo III, a imunomarcação foi mais intensa no grupo IV, menos marcante no grupo V, e no grupo de ratos suplementado com L-glutamina, foi similar à encontrada nos animais não-irradiados. Os resultados sugerem que a suplementação dietética com glicina e L-arginina leva a aumento da quantidade de colágeno na parede ileal de ratos submetidos a radioterapia abdominal, eventualmente predispondo ao surgimento de fibrose intestinal, enquanto que a suplementação com L-glutamina propicia a manutenção da espessura da camada submucosa e pode contribuir para a atenuação dos efeitos colaterais decorrentes da irradiação.

There is a high prevalence of hypertension in the African population and it is known that
vascular dysfunction (including nitric oxide (NO) bio-availability markers) play an important
role in the development of cardiovascular diseases. Since very little is known regarding the
role of markers of NO bio-availability in Africans, the aim of this study was to compare
markers of NO bio-availability (namely L-arginine, L-citrulline, asymmetric dimethylarginine
(ADMA) and symmetric dimethylarginine (SDMA)), ambulatory blood pressure (BP) and
markers of end organ damage between African and Caucasian school teachers. Additionally,
we also aimed to determine whether these markers of NO bio-availability are associated with
ambulatory BP and markers of end organ damage in both ethnic groups.
Methods
The SABPA (Sympathetic activity and Ambulatory Blood Pressure in Africans) study was a
cross-sectional study, including urbanised African (N=181) and Caucasian (N=209) men and
women, between the ages of 25 and 65 years. Cardiovascular measurements included
ambulatory blood pressure, pulse wave velocity (PWV), electrocardiographic Cornell product
and carotid intima media thickness (cIMT). Anthropometric measurements included height,
weight and waist circumference.
Various bio-markers were analysed, including glucose, L-arginine, ADMA, SDMA, Lcitrulline,
reactive oxygen species, albumin-to-creatinine ratio (ACR) and estimated
creatinine clearance (eCCR).
Characteristics of groups were compared with independent T-tests and Chi-square tests.
Single and partial analyses were used to investigate associations between NO bioavailability
markers with ambulatory BP measurements and markers of end organ damage.
Analyses of covariance (ANCOVA) were used for comparison of variables between groups
to determine significant differences, while adjusting for age, body mass index and antihypertensive
medication. Forward stepwise multiple regression analyses were performed to
determine if independent associations exist between ambulatory BP measurements or
markers of end organ damage with either- L-arginine, L-citrulline, ADMA or SDMA as the
main independent variable.
Results and conclusion
The Africans and Caucasians were of similar ages. However, the Africans had higher blood
pressure therefore their cardiovascular profile was unfavourable compared to that of the
Caucasians.
The inhibitors of NO biosynthesis, ADMA and SDMA, were significantly lower in the Africans
(p=0.046; p<0.001, respectively). However, the NO bio-availability markers, L-arginine and
L-citrulline, were higher in the African compared to the Caucasian participants (all p values
<0.05) regarded as significant.
When performing unadjusted analyses, we found significant negative associations between
eCCR and L-citrulline in all four subgroups: African men (r=-0.27; p=0.013), African women
(r=-0.24; p=0.021), Caucasian men (r=-0.21; p=0.044) and Caucasian women (r=-0.28;

▼ Mycobacterial disease plagues over one-third of the
global population. The most devastating of which – Mycobacterium
tuberculosis – kills more than 1.5 million annually and multi-drug
resistance increases its catastrophic reach. Host-directed therapy
is a promising counter-balance against drug resistant mycobacteria,
warranting further investigation into anti-mycobacterial host
defense. Key immune populations –macrophages and T cells – are
regulated by the availability of the amino acid
L-
arginine. Nitric
oxide (NO), a potent anti-mycobacterial agent deployed by
macrophages, and appropriate Th1 functions – e.g. proliferation,
IFN-γ production – demand
L-
arginine. However, mycobacterial
infection restricts
L-
arginine by inducing myeloid arginase,
resulting in diminished NO production by macrophages and
hyporesponsive T cells. Previously, our group found that synthesis
of
L-
arginine from the endogenous precursor,
L-citrulline, is a
necessary metabolic pathway for anti-mycobacterial immunity, but
the mechanism of action within the immune population was unknown.
Herein, we define the immunological consequences of
L-citrulline
metabolism during mycobacterial infection. We found that
L-citrulline metabolism endows macrophages and T cells with a
mechanism to circumvent arginase-mediated inhibition of
anti-mycobacterial function in vitro. Utilizing novel mouse models
of
L-citrulline metabolism deficiency, we uncovered an
unappreciated necessity of
L-citrulline metabolism for macrophage
and T cell immunity to mycobacterial infection in vivo.
Additionally, our data show that mice supplemented with
L-citrulline exhibit enhanced control of pulmonary M. bovis BCG
infection compared to vehicle-treated controls, revealing the
potential of this metabolic pathway as a novel therapeutic to
combat mycobacterial disease.

▼ A capacitação é um pré-requisito fisiológico importante para que a célula espermática fertilize o oócito. O óxido nítrico (NO) é sintetizado in vivo durante a conversão da
L-arginina em
L-citrulina por reações oxidativas catalisadas pela enzima óxido nítrico sintase (NOS) desempenhando um papel importante na regulação da motilidade e na capacitação dos espermatozoides. Estudos indicam que o NO é capaz de regular a concentração da AMP cíclico e, por conseguinte, através da atividade da adenil ciclase, estimular a capacitação espermática em várias espécies. O objetivo deste estudo foi avaliar a função do NO na capacitação de espermatozoides equinos criopreservados. Três ejaculados foram colhidos de três garanhões (n=9). O sêmen foi diluído em meio Botu-Crio® na concentração final de 200×106 células/mL, envasado em palhetas de 0,5 mL e criopreservado usando um sistema automatizado. Para cada análise, foram descongeladas quatro palhetas da mesma partida e do mesmo garanhão em banho-maria a 37 °C/30 s. e em seguida, o sêmen foi submetido à centrifugação em meio FIV. Posteriormente, o sêmen foi incubado neste mesmo meio na presença de
L-arginina, com ou sem inibidor da enzima óxido nítrico sintase o (
L-NAME), e com ou sem o removedor de NO (azul de metileno) nos tratamentos: 1) C= (FIV); 2) A=
L-arginina (10 mM); 3)
L =
L-NAME (1 mM); 4) M = azul de metileno (100 mM); 5) AL =
L-arginina (10 mM) +
L-NAME (1 mM); 6) AM =
L-arginina (10 mM) + azul de metileno (100 mM). As amostras foram incubadas a 38 °C e 5 % de CO2. Após a incubação realizou-se a análise computadorizada da motilidade do espermatozoide e as análises por citometria de fluxo. Para a análise computadorizada da motilidade espermática foram avaliados os tempos de incubação de 0, 60, 120 e 300 min. e para as análises por citometria de fluxo os tempos de 60, 120 e 300 min. Para avaliar a integridade das membranas plasmática e acrossomal usou-se a associação FITC-PSA e IP. Para a detecção da fosforilação do aminoácido tirosina, usou-se o anticorpo antifosfotirosina conjugado a uma fluoresceína (DAF-2). A fim de dosar a quantidade de NO produzido pelo espermatozoide equino criopreservado foi utilizada a sonda DAF e para avaliar a peroxidação lipídica da membrana espermática utilizou a sonda C11-BODIPY. A sonda H33342 foi usada com a finalidade de evitar que partículas do mesmo tamanho e granulosidade da célula espermática fossem incluídas na contagem das análises por citometria de fluxo. Os dados foram analisados por meio da ANOVA e a comparação das médias, dentro de cada tempo, pelo teste de Tukey, com o nível de significância de 5 %, usando o software SAS. A remoção do NO do meio de cultura inibiu a motilidade das células espermáticas em todos os tempos de incubação. A motilidade total e motilidade progressiva foram reduzidas nos grupos M e AM. ▼ O objetivo deste trabalho foi verificar o possível efeito protetor da L-glutamina e da L-arginina sobre a próstata ventral de ratos quando administradas por gavagem. Procurou-se simular as condições clinicas de pacientes submetidos à radioterapia pélvica tendo como órgão alvo outro órgão pélvico que não a próstata. Foram analisados os efeitos desta irradiação sobre a próstata considerando este órgão como normal. Foram utilizados ratos Wistar divididos em quatro grupos: Controle, animais não submetidos à irradiação (n= 10); Irradiado, submetidos à irradiação
abdominal e sem suplementação adicional de aminoácido por 21 dias (n= 10); Irradiado + Lglutamina, submetidos à irradiação abdominal e com suplementação adicional de L- glutamina por 21 dias (n= 10); e Irradiado + L-arginina, submetidos à irradiação abdominal e com suplementação adicional de L- arginina por 21 dias (n= 9). Os grupos foram mantidos em condições padrão de laboratório durante todas as etapas do experimento. Os animais submetidos à irradiação abdominal receberam uma dose única de 1000 cGy no dia 8 da experimentação. A Lglutamina e a L-arginina foram dissolvidas em água destilada e administrada por gavagem através da agulha IC-810. As próstatas foram removidas e processadas para inclusão em parafina. Foram estudados os seguintes parâmetros: estrutura acinar (área dos ácinos e altura do epitélio) e colágeno analisados por métodos morfométricos e peso corporal. O ganho de peso nos
grupos suplementados foi significativamente maior se comparado ao grupo irradiado. Houve redução da altura do epitélio no grupo irradiado quando comparado ao controle. A altura do epitélio no grupo suplementado com L-arginina foi significativamente maior do que nos grupos irradiado e suplementado com L-glutamina. Houve diminuição, de aproximadamente 18%, da área dos ácinos no grupo suplementado com L-glutamina. Já no grupo suplementado com Larginina o valor foi similar ao do controle. O efeito da L-glutamina sobre o parênquima prostático foi o de manter proporcionalmente o colágeno, preservando a integridade da matriz extracelular. No grupo suplementado com L-arginina, apesar da discreta redução na distribuição proporcional
de colágeno este também manteve índices semelhantes ao do controle. A radiação abdominal promoveu algumas modificações estruturais na próstata ventral de ratos. Essas modificações podem ser parcialmente prevenidas pela suplementação oral com L-glutamina e de L-arginina.

▼ ÇalıĢmamızın amacı nöropatik ağrı oluĢturulmuĢ sıçanlarda simvastatinin olası antinosiseptif etkisi ve bu etkiye nitrik oksidin katkısının değerlendirilmesidir.
Toplam sekiz grup olmak üzere 48 adet Sprague-Dawley, yetiĢkin (200-250g), erkek sıçan kullanıldı. Bu çalıĢma ESOGÜ Hayvan Deneyleri Yerel Etik Kurulu’nun onayıyla yapıldı. Siyatik sinirin ana dalının ligasyonuyla kronik konstriksiyon zedelenmesi oluĢturulmuĢ sıçanlara 10 gün süreyle simvastatin(10,20mg/kg,i.p.) uygulandı. Simvastatinin antinosiseptif etkisi 0,3,7,10. günlerde termal hiperaljezi, soğuk allodini ve mekanik allodini testleriyle değerlendirildi. Ayrıca aynı testler simvastatinin etkin bulunan dozu(20 mg/kg,i.p.) ile birlikte
L-NAME/
L-Arginin(10mg/kg) kullanılarak bu etkideki nitrik oksidin rolü de değerlendirildi. Pozitif kontrol olarak nöropatik ağrının tedavisinde klinikte rutin olarak kullanılan tramadol(10mg/kg) sıçanlara uygulandı. Tüm testlerde pençe çekme eĢikleri ve pençe çekme gecikmeleri ilacın uygulanmasından 30 dakika sonra kontrol ve sham gruplarına göre elde edildi. Ġstatistiksel analiz için grupların karĢılaĢtırılmasında normal dağılan verilerde Tek Faktör Tekrarlı Ġki Yönlü (ANOVA) Varyans Analizi kullanıldı. Çoklu karĢılaĢtırmalar Holm-Sidak metodu ile yapıldı.
Mekanik allodini testinde 3. günde Simvastatin 10-20mg/kg ve tramadol gruplarında; 10.günde tramadol ve simvastatin 20mg/kg gruplarında anlamlı olarak antinosiseptif etki saptanmıĢtır.
L-NAME(10mg/kg)+Simvastatin (20mg/kg’da) 10.günde pençe çekme gecikmesinde anlamlı bir azalma görülmüĢtür.
L-arginin+Simvastatin(20mg/kg) grubunda da 3.günde simvastatin(10-20mg/kg) ve tramadol grubuna göre pençe çekme eĢiği anlamalı olarak azalmıĢtır. Termal hiperaljezi testinde gruplar arasında günlere göre anlamlı bir farklılık görülmemiĢtir. Soğuk allodini testinde ise Simvastatin 20mg/kg grubunda 10.günde 3.güne ve kontrol grubuna göre pençe çekme gecikmesinde anlamlı bir artma görülmüĢtür(p≤ 0,01). ÇalıĢmamızda simvastatin 20mg/kg dozunda sıçanlarda nöropatik ağrıyı anlamlı bir Ģekilde azaltmıĢtır. Sonuçlarımıza göre simvastatinin bu antinosiseptif etkisinde nitrik oksid yolağının da katkısının olabileceği düĢünülmektedir.

arginine is one of the most versatile amino acids due to the fact that it serves as a precursor for many molecules which have important roles in bodily functions including mammalian reproduction. The current studies sought to further examine the role that
L-
arginine has in mammalian reproduction utilizing both <i>in vivo</i> and <i>in vitro</i> approaches. In the first study, a novel bioluminescent murine pregnancy model was developed to monitor VEGFR2 transcription activity non-invasively in the fetoplacental unit. Secondly, the effect that dietary
L-
arginine supplementation has during mouse gestation was examined.
L-
arginine supplementation increased weight gain during the latter third of gestation, total litter size, number of implantation sites, and litter birth weight. Additionally,
L-
arginine supplementation increased VEGFR2 transcription activity in the fetoplacental unit which may create a more favorable environment for fetal survival. Moreover, the increased number of implantation sites observed suggests an effect of
L-
arginine at the level of the endometrium. To this end, the effect that
L-
arginine has on apoptosis and cell proliferation in an established endometrial cell line was examined. The addition of
L-
arginine at physiological (200 micromolar) and supra-physiological (800
micromolar) concentrations increased cell proliferation , and this effect was achieved through biosynthesis of polyamines and nitric oxide.
L-
arginine also decreased the proportion of cells that were experiencing mitochondrial mediated apoptosis, and it was observed that this decrease in mitochondrial mediated apoptosis was concurrent with increased phosphorylation of BAD protein, which induces apoptosis when not phosphorylated. The final study examined the ability of porcine uterine epithelial (PUE) cells to synthesize
L-
arginine from
L-citrulline.
L-citrulline was able to support PUE cell proliferation in the absence of
L-
arginine. Additionally, ASS-1 and ASL,
L-
arginine synthesizing enzymes, were expressed in PUE cells and were regulated by the presence of
L-
arginine and
L-citrulline, respectively. This data would support the hypothesis that PUE cells may be able to convert
L-citrulline to
L-
arginine. Together, the current findings along with the plethora of relevant literature provide further evidence for the role of
L-
arginine in mammalian reproduction and allow for new questions to be investigated regarding this particular amino acids role in mammalian reproduction.

▼ Limited availability of donor lungs results in high mortality of patients waiting for lung transplantation. Modern techniques such as ex vivo lung perfusion (EVLP) or interleukin-10 (IL-10) gene therapy expand the donor pool and may improve the quality of donor organs and outcomes following lung transplantation remain inferior compared to other organ transplantations. Alterations in metabolic pathways, such as L-arginine/nitric oxide (NO) metabolism can contribute to post-transplantation organ dysfunction. We investigated the L-arginine/NO metabolism in pig models of lung transplantation. We found significant differences in the L-arginine/NO metabolism in lungs from brain death compared to non-brain death donors after prolonged hypothermic preservation. Moreover, we found that EVLP decreased NO metabolite concentrations in lung, a sustained effect after transplantation that was unaffected by IL-10 gene therapy during EVLP. In conclusion, donation circumstances and preservation methods may alter the L-arginine/NO metabolism in transplanted lungs, which may contribute to clinical outcomes after transplantation.

▼ Stroke is a major cause of death and the highest cause of morbidity worldwide. In NZ, the elderly and Maori and Pacific Island ethnicities are most affected. With failure of many neuroprotectant agents, current clinical therapy is limited to the use of thrombolytics to treat ischaemic stroke. Inflammation-dependent neurodegeneration occurs over a prolonged period after stroke. Melatonin has been shown to have anti-inflammatory, anti-oxidant, and neuroprotective effects after cerebral ischaemia reperfusion (IR) injury. This thesis focuses on the mechanisms of neuroprotection conferred by melatonin application after cerebral IR injury. Transient (2 hours) cerebral IR was achieved using the filament insertion model of middle cerebral artery occlusion (MCAO). The animals were first dosed (5 mg/kg i.p.) 1 hour after MCAO and two further doses administered over the next 48 hours. Melatonin administration was confirmed to reduce infarct size and be non-toxic. Immunohistochemistry was used to localise endogenous melatonin and its receptors (MT1 and MT2) in cerebral IR in MCAO and control animals. The majority of cells expressing melatonin and its receptors were found within the hypothalamus. The presence of melatonin and its receptors in the blood vessels, may suggest a role for melatonin in controlling immune cell infiltration after cerebral IR. Following MCAO, cellular immunoreactivity to melatonin receptor antibodies increased. Within the infarct, infiltrating inflammatory cells expressed melatonin and MT2 receptor. Following MCAO, major inducible enzymes such as nitric oxide synthase (iNOS), and cyclooxygenase (COX-II) were stimulated. Melatonin administration resulted in a significant decrease in iNOS activity as well as total NOS activity and a consequent decrease in nitrite levels. Melatonin administration also attenuated both the MCAO-induced increase in COX expression and activity. HT-1080 human fibrosarcoma fibroblasts was utilised to probe the effects of melatonin on arginase enzymes. Melatonin treatment decreased cell viability at high concentrations, and this effect was attributed to its pro-oxidant effect present at these concentrations in cancer cell lines. The pro-oxidant effect was associated with increased total NOS activity. On the other hand, both arginase II expression and activity were increased with higher concentrations of melatonin treatment. These results highlighted the possibility that melatonin may be able to stimulate eNOS and arginase enzymes, both of which are beneficial after cerebral IR. Inflammation occurring after cerebral IR has been linked closely to mitochondria-driven apoptosis. Increased oxidative stress was seen as indicated by inhibition of aconitase enzyme activity after MCAO. Consequently, most of the electron transport chain complexes measured were significantly impaired. Melatonin administration led to protection of electron transport chain complexes, thus providing evidence of mitochondrial protection after MCAO. ▼ Introduction: Impairment of cytosolic calcium ([Ca2+]i) signaling and in particular calcium overload has emerged as a possible unifying mechanism for precipitating acute pancreatitis (AP.) In the L-arginine (L-arg) experimental model of AP, nitric oxide (NO) has been implicated however the disease progression is largely unaffected by nitric oxide synthase (NOS) inhibitors (8). Additionally, L-ornithine (L-orn), a NOS-independent metabolite of L-arg, has been shown to be potent at inducing AP (28). Both L-arg and L-orn activate calcium-sensing like receptors (CaSR) (31) such as the GPRC6a which may be responsible for initiating the [Ca2+]i overload. The aim of this study is to investigate the effects of L-arg and L-orn on pancreatic acinar cells that maybe linked to the pathophysiology of AP. Furthermore to provide an alternative theory to the NO mediated ones, in particular that L-arg induces toxic changes in [Ca2+]i via a GPRC6a like receptor. Methods: Whole pancreata were harvested from male Sprague Dawley rats. Pancreatic acinar cells were isolated by collagenase digestion. [Ca2+]i was measured using fura-2 imaging, and cell viability assessed using physiological CCK. Oxidative stress was measured using dichlorofluorescein (DCF) and cell death was quantified using trypan blue exclusion. Results: L-arg and L-orn (100mM) induced spike-like, reversible increases in [Ca2+]i in 46% and 74% of cells and Ca2+ overload in 11% and 26% respectively. At 500 mM both induced Ca2+ overload in all cells however this was also seen with the osmotic control, mannitol. Isosmotic L-arg and L-orn (100mM) induced only reversible increases in [Ca2+]i. Neither L-arg nor L-orn had significant effects on CCK-evoked [Ca2+]i oscillations. Both L-arg and L-orn induced significant oxidative stress responses (22% and 37% of a maximum response seen with 3mM H202, respectively). Both L-arg and L-orn caused cell death in 76% +/- 4 and 89% +/- 7 at 3 hours respectively, compared to 35% +/- 4 and 40% +/- 3 with controls (Hepes, Glycine). Conclusion: The data suggests that the L-arg and L-orn causes significant increase in oxidative stress and cell death. The data suggests that although changes in [Ca2+]i were induced by both L-arg and L-orn the large concentrations used experimentally are likely to induce significant osmotic effects.

Tanto a obesidade quanto a SM aumentam o número de pacientes expostos ao risco de doença cardiovascular. Estudos recentes mostram que uma redução na biodisponipilidade de óxido nítrico (NO) é um dos principais fatores que contribui para a ação deletéria da insulina nos vasos de pacientes adultos com obesidade e SM. O NO, potente vasodilatador e anti-agregante plaquetário, tem como precursor o aminoácido catiônico
L-arginina que é transportado para o interior das plaquetas através do carreador y+
L. Uma família de enzimas denominadas NO sintases (NOS) catalisa a oxidação da
L-arginina em NO e
L-citrulina e é composta de três isoformas: neuronal (nNOS), induzível (iNOS) e endotelial (eNOS). Os objetivos principais do presente estudo são de investigar diferentes etapas da via
L-arginina-NO em plaquetas associando agregação plaquetária, concentração plasmática de
L-arginina, estresse oxidativo, marcadores metabólicos, hormonais,
clínicos e inflamatórios em pacientes adolescentes com obesidade e SM. Foram incluídos no estudo trinta adolescentes, sendo dez com obesidade, dez com SM, e dez controles saudáveis pareados por idade, sexo e classificação de Tanner (controles: n= 10, 15.6 0.7 anos; obesos: n= 10, 15 0.9 anos; SM: n= 10, 14.9 0.8 anos). O transporte de
L-arginina (pmol/109céls/min) através do sistema y+
L estava diminuído
nos pacientes com SM (18.4 3.8) e obesidade (20.8 4.7), comparados aos controles (52.3 14.8). Houve uma correlação positiva do influxo de
L-arginina via sistema y+
L com os níveis de HDL-Colesterol. Por outro lado, foi encontrada uma correlação
negativa do influxo de
L-arginina com os níveis de insulina, os índices Homa IR, relacionado a RI, Homa Beta, relacionado a função da célula beta e também com os índices de Leptina. Em relação a produção de NO, a obesidade e a SM não afetaram a
atividade e expressão das enzimas NOS. A atividade da superóxido dismutase (SOD), através da mensuração da inibição da auto-oxidação da adrenalina, mostrou diferença significativa nas plaquetas de pacientes com obesidade (4235 613,2 nMol/mg de proteína), quando comparada aos controles (1011 123,6 nmol/mg de proteína) e SM (1713 267,7 nmol/mg de proteína). A nível sistêmico, foi também evidenciada uma ativação desta enzima anti-oxidante no soro de pacientes obesos, em relação aos controles. A peroxidação lipídica avaliada pelas substâncias reativas ao ácido tiobarbitúrico (TBARS) estava inalterada no soro dos pacientes e controles. Estes resultados sugerem que o transporte de
L-arginina diminuído nas plaquetas de adolescentes obesos e com SM pode ser um marcador precoce de disfunção plaquetária. tratamento visível num futuro próximo, os números tendem a aumentar rapidamente. De
todas as doenças neurodegenerativas, a doença de Alzheimer (DA) é a mais predominante. Na Doença de Alzheimer há uma perda neuronal e sináptica generalizada que causa um declínio progressivo na memória e em outras funções cognitivas, conduzindo finalmente à demência.
Esta doença neurodegenerativa está associada a dois tipos de depósitos proteicos, placas
extracelulares β-amilóides (Aβ) e complexos neurofibrilares intraneuronais. Estudos recentes demonstraram que o cluster miR-29 pode diminuir a deposição das placas de Aβ atuando diretamente sobre a enzima que cliva os péptidos β-amilóides (BACE-1), a qual é responsável pela clivagem da proteína precursora e, consequentemente geração de espécies Aβ tóxicas.
Estes estudos apresentam a possibilidade da utilização do pré-miR-29 como um novo alvo
terapêutico no tratamento da DA, para qual é necessário garantir um elevado grau de pureza, estabilidade e integridade do miRNA. Assim, o principal objetivo deste estudo é o
desenvolvimento de uma nova estratégia de cromatografia de afinidade com uma matriz Ofosfo-
L-tirosina comparando duas metodologias diferentes – o desenho experimental e o método “tentativa/erro” - para obter o pré-miR29 com elevado grau de pureza e rendimento, visando a sua aplicação em terapia génica.

▼ O diabetes mellitus (DM) pode ser definido como um grupo heterogêneo de distúrbios metabólicos caracterizados pela hiperglicemia, resultando em disfunção, e última instância, insuficiência de vários órgãos, especialmente dos olhos, rins, nervos e sistema vascular. O presente estudo teve como objetivo avaliar os efeitos da Larginina (L-Arg), do exercício resistido (ER) e da associação entre L-Arg e o ER sobre as alterações dos fatores relaxantes derivados do endotélio (EDRFs) em anéis isolados da artéria mesentérica superior de ratos diabéticos induzidos por aloxano. Ratos Wistar (250-300g) foram divididos em 7 grupos: Controle (CON, n = 4), Estimulado Eletricamente (EE, n = 4), Controle Treinado (CT, n = 4), Diabético Sedentário (DS, n = 5), Diabético L-arginina (DL-Arg, n = 5), Diabético Treinado (DT, n = 5) e grupo Diabético Treinado + L-arginina (DT + L-Arg, n = 5). Os animais dos grupos diabéticos receberam aloxano 2 semanas antes do início dos protocolos de tratamento. O protocolo de ER consistiu em 3 séries de 10 agachamentos (no aparelho de Tamaki) com uma intensidade de 50% de uma repetição máxima (1RM). Os animais tratados com L-Arg receberam 1,25 mg/ml diariamente na água de beber. No final de 8 semanas, o vasorrelaxamento dependente de endotélio foi obtido em anéis pré-contraídos com L-fenilefrina (FEN, 10 μM) pela adição à cuba de concentrações crescentes e cumulativas de acetilcolina (ACh, 10-9 10-4 M). Após isto, curvas concentração-respostas foram construídas na condição controle ou na presença dos inibidores da síntese do óxido nítrico (LNAME), da ciclo-oxigenase (INDO) e dos canais para potássio (TEA). Os vasorrelaxamentos induzidos por ACh apresentaram uma redução significativa (p <0,05) na resposta máxima (Rmax) do DS comparado com o CON. Além disso, foi observado uma redução significativa (p <0,001) da sensibilidade (pD2) (de 6,8 0,12 para 6,2 0,11) do DL-Arg quando comparado com o DS. Na presença do LNAME o DL-Arg apresentou uma redução (p <0,001) dos vasorrelaxamentos 8 quando comparado com o DS. Após adição de L-NAME + INDO, o DT + L-Arg apresentou uma redução significativa (p <0,001) nos vasorrelaxamentos quando comparado com o DS. Já na presença de L-NAME + INDO + TEA, o DT e DT + LArg apresentaram uma redução significativa (p <0,001) dos vasorrelaxamentos quando comparados com o DS. Os resultados demonstram que apesar de não haver melhora nos vasorrelaxamentos induzidos por ACh, foi observado uma modificação na participação dos EDRFs em todos os grupos.

▼ Investigamos os efeitos dos exercícios físicos (EF) aeróbios e/ou suplementação de L-Arginina (L-ARG) sobre as possíveis alterações da composição corporal (CC) de ratas hipertensas (SHR) ovariectomizadas. As ratas (com aproximadamente 13 semanas de idade) foram submetidas a remoção cirúrgica dos ovários para podermos estudar o efeito da depleção de hormônios ovarianos, que ocorre na menopausa. Após 30 dias de ovariectomia, as ratas realizaram um protocolo de EF em esteira ergométrica de moderada intensidade ou mantidos sedentários por oito semanas e suplementadas com L-ARG. Enquanto o ganho de massa corporal (MC) relativa não foi afetada pelos EF ou L-ARG, o consumo alimentar (CA) aumentou significativamente nos grupos EXER (hipertenso treinado) e EXER-ARG (hipertenso treinado associado com suplementação de L-ARG), quando comparado ao grupo CONT (hipertenso sedentário - controle) Como esperado, os EF e a L-ARG, foram eficientes em elevar a capacidade física dos grupos EXER e EXER-ARG, com aumento extremamente significativo (p<0.001) da distância máxima alcançada na 8ª semana de EF, porém não causou alterações na massa úmida dos músculos estriados esqueléticos (MEE). Em associação, verificamos um aumento da massa úmida do coração nos grupos EXER e EXER-ARG, porém no grupo EXER-ARG observou-se uma discreta queda na PA (pressão arterial) final, ainda que não significante. Paralelamente, os dados mostraram que a L-ARG e/ou os EF não foram efetivos para a diminuição da reabsorção do osso ou a formação óssea crescente.
We investigated the effects of physical exercise (PE) aerobic and/or supplementation of L-arginine (L-ARG) on the possible changes in body composition (CC) of hypertensive rats (SHR) ovariectomized. The rats (approximately 13 weeks old) underwent surgical removal of the ovaries in order to study the effect of depletion of ovarian hormones at menopause occurring. After 30 days of ovariectomy, the rats underwent a protocol of PE treadmill of moderate intensity or kept sedentary for eight weeks and supplemented with L-ARG. While the gain in body mass (BM) was not affected by the relative PE or L-ARG, food consumption (CA) increased significantly in groups EXER (hypertensive trained) and EXER-ARG (hypertensive trained associated with supplementation of L-ARG) when compared to the CONT group (untrained - control) As expected, PE and L-ARG, were effective in increasing the physical capacity of groups EXER and EXER-ARG, with highly significant increase (p <0.001) the maximum distance reached the 8th week of PE, but caused no change in wet mass of striated skeletal muscle (ESM). In combination, we see an increase in wet weight of the heart in groups EXER and EXER-ARG, but in the group EXERARG showed a slight drop in BP (blood pressure) end, though not significant. Simultaneously, the data show that L-Arg and/or PE were not effective to decrease bone resorption or increasing bone formation.

▼ Justificativa: Durante lesão de isquemia e reperfusão renal ocorre a inativação de óxido nítrico (NO), essa inativação diminui o relaxamento arterial endotélio-dependente e independente. O aumento nos níveis de NO poderia melhorar a disfunção endotelial e o relaxamento arterial diminuindo a lesão renal. O NO, sintetizado a partir da L-arginina pela enzima óxido nítrico sintase (NOS), parece exercer um efeito protetor sobre os rins durante a lesão de isquemia/reperfusão (I/R). O objetivo do estudo foi avaliar o efeito da L-arginina nos níveis dos biomarcadores de lesão (NGALp, NGALu, KIM-1 e IL-18) na função e na histopatologia renal em ratos submetidos a lesão de I/R.
Métodos: 32 ratos Wistar foram randomizados em 4 grupos: Sham (S): Laparotomia e nefrectomia direita. Controle (C): Laparotomia e nefrectomia direita, I/R em rim esquerdo. Controle L-arginina (CLA): Laparotomia e nefrectomia direita, L-arginina na dose de 800mg.kg-1.dose-1, 24 e 1 hora antes. L-arginina (LA): Laparotomia e nefrectomia direita, I/R em rim esquerdo, L-arginina na dose de 800mg.kg-1.dose-1, 24 e 1 hora antes. Foram analisados os níveis de NGALp, NGALu, KIM-1 e IL-18, creatinina e histopatologia renal. Nível de significância: p<0,05.
Resultados: Creatinina aumenta em todos os grupos com LA≈C>S≈CLA. A NGALp, aumenta em todos os grupos com evolução semelhante. A NGALu foi maior no grupo C e CLA em relação ao grupo LA e apresentou valores intermediários no grupo S. Os níveis de KIM-1 aumentam nos grupos S, CLA e LA, porém na análise de contrastes não há diferença na evolução dos grupos. A IL-18 apresentou valores maiores no grupo LA comparado aos demais grupos (C, S e CLA). Os grupos C e LA apresentaram grau de lesão no lado esquerdo maior que os grupos S e CLA. Não existe diferença histopatológica entre C e LA e nem entre S e CLA.
Conclusão: A L-arginina não exerce efeito protetor ou danoso aos rins submetidos a lesão de I/R na avaliação da função, da histopatologia renal e dos níveis de NGALp, KIM-1
Background: Nitric oxide (NO) inactivation occurs during renal ischaemia/reperfusion injury. This inactivation decreases endothelium-dependent and -independent arterial relaxation. Higher NO levels can improve endothelial dysfunction and arterial relaxation, thereby reducing the level of kidney injury. ▼ Introduction: Preeclampsia (PE) is a clinical complication of pregnancy characterised with the new onset of hypertension and proteinuria, and/or organ dysfunction. Globally, it is associated with maternal and perinatal morbidity and is a major cause for concern. If undiagnosed, preeclampsia can lead to eclampsia, which is characterised by an onset of seizures (convulsions). Thus, neurological consequences have been reported in both PE mothers and their offspring.
Materials and Methods: This study investigated the role of neuroinflammation and oxidative stress in the brain of an Nꭃ-nitro-
L-
arginine methyl (
L-NAME) induced Preeclamptic rat model through birth to late postnatal days in the mother and the offspring. Pregnant rats were divided into control, early onset and late onset PE groups. Blood pressure, urine volume and proteinuria were measured on gestational day (GD) 0, 12 and 17 to establish PE. At GD 19, postnatal day (PND) 1 and 60, rats and their pups were sacrificed and brain excised. Prior to sacrifice at PND 60, the offspring were subjected to neurobehavioural studies to test for memory performance and locomotor activity. Ionized calcium binding adaptor molecule 1 (IBA1) and Excitatory amino acid transporter 1 (EAAT1), and oligodendrocyte transcription factor 2 (OLIG2) expression in the cortex and cerebellum were analysed by immunohistochemistry. Additionally, cortical and cerebellar tissues were homogenised for further biochemical analysis of oxidative stress such as Nitric oxide (NO), lipid peroxidase superoxide dismutase (SOD), glutathione (GSH), lipid peroxidase (LPO) and purinergic enzyme activities at PND 60.
Results: We found an increase in blood pressure accompanied by proteinuria and a low foetal count in the
L-NAME treated groups. Neuroinflammation was evident in the treated group at birth and PND 60 as shown by an increase in the number of IBA1 expressing activated microglia with a simultaneous reduction in the immunoexpression of EAAT1. PE- induced axonal damage was noted as shown by a reduced number of oligodendrocytes. At PND 60, PE groups show alteration in oxidative stress markers, increased acetylcholinesterase activity, and decreased purinergic enzymes activities such as adenosine triphosphatase (ATPase) and ecto-nucleoside triphosphate diphosphohydrolase (ENTPDase). The offspring at PND 60 also display reduced memory performance and locomotor activity, which was accompanied by an increased number of activated microglia, down-regulated the immunoexpression of EAAT1 and reduced number of oligodendrocyte cells.
Discussion and conclusion: This study demonstrates neuroinflammation and axonal damage in PE at delivery which also persists into later life. This finding might be attributed to systemic inflammation and vulnerability of blood-brain barrier associated with PE which can cause crossing over of substances from the systemic environment thereby causing insult to the brain. ▼ A insuficiência cardíaca (IC) é uma síndrome clínica de elevada incidência e com um prognóstico ruim a longo prazo. Ela é a via final comum da maioria das doenças que acometem o coração, sendo um dos mais importantes desafios clínicos na área da saúde. O óxido nítrico (NO) representa, por intermédio de sua influência sobre o endotélio e as plaquetas, um importante papel na regulação da homeostase vascular. Este gás de meia-vida curta é sintetizado a partir do aminoácido
L-arginina, pela enzima NO sintase (NOS), levando à produção de guanosina monofosfato cíclica (GMPc). Estudos mostram que anormalidades na biodisponibilidade de NO em plaquetas podem contribuir para eventos trombóticos, não tendo sido ainda avaliada na IC. Na primeira parte do estudo, o objetivo foi investigar o efeito da IC na atividade e na expressão da NOS em plaquetas, no conteúdo intraplaquetário de GMPc, na agregação plaquetária, além dos parâmetros antropométricos e da composição corporal, das variáveis bioquímicas, dos aminoácidos plasmáticos, do estresse oxidativo (plasma e plaquetas) e da concentração sistêmica de marcadores inflamatórios em 15 pacientes com IC e 15 controles saudáveis. Na segunda parte do estudo, objetivou-se avaliar os efeitos do treinamento físico (TF) regular nessas mesmas variáveis (exceto a expressão da NOS) e nas variáveis hemodinâmicas e respiratórias. Para tal, foram avaliados 15 pacientes com IC que se mantiveram sedentários e 15 pacientes com IC que realizaram 30 minutos de atividade física aeróbia e treinamento contraresistência muscular localizada com pesos livres e máquinas, três vezes por semana, durante 24 semanas. Os resultados da primeira etapa do estudo demonstraram hiperagregabilidade plaquetária induzida tanto por colágeno como por ADP, com aumento do estresse oxidativo, da atividade basal da NOS e da concentração de GMPc, estando os níveis plasmáticos de
L-arginina em pacientes com IC diminuído. A expressão da iNOS, estava aumentada em plaquetas de pacientes com IC em relação aos controles saudáveis.Também foi observado aumento da resposta inflamatória, com maiores níveis sistêmicos de proteína C reativa, fibrinogênio, interleucina-6 e fator de necrose tumoral α. Após o período de TF, houve aumento do VO2 máximo e a agregação plaquetária induzida tanto por colágeno quanto por ADP estava diminuída. Ocorreu um aumento dos níveis plasmáticos de
L-arginina e, uma redução da atividade da NOS após o TF. Em relação ao estresse oxidativo, tanto a produção sistêmica, quanto a intraplaquetária de substâncias reativas ao ácido tiobarbitúrico (TBARs) e a carbonilação diminuíram na presença da atividade aumentada das enzimas superóxido dismutase (SOD) e da catalase após o TF. Houve uma diminuição da resposta inflamatória, uma diminuição do colesterol total, do LDL e dos triglicerídeos e um aumento do HDL, após oTF. ▼ A anorexia nervosa (AN) é um transtorno alimentar com alta morbi-mortalidade, que atinge principalmente adolescentes do sexo feminino. A AN é caracterizada por uma distorção na imagem corporal que leva a restrição alimentar, e, consequentemente, perda de peso e diversas complicações clínicas como um estado de hiperagregabilidade plaquetária. O óxido nítrico (NO) é produzido a partir do aminoácido catiônico L-arginina através de uma família de enzimas denominadas NO-sintases (NOS) e funciona como um protetor cardiovascular modulando várias funções como relaxamento endotelial e função plaquetária. O objetivo dessa dissertação é avaliar a via L-arginina-NO na AN, bem como investigar o ciclo da uréia, a função plaquetária, e o estresse oxidativo em pacientes com AN. O transporte de L-arginina, a produção de guanosina monofosfato cíclica (GMPc), a atividade e a expressão das isoformas da NOS, iNOS e eNOS, o estresse oxidativo (formação de substâncias reativas ao ácido tiobarbitúrico - TBARS e atividade da superóxido dismutase - SOD), bem como a atividade plaquetária foram avaliados nas plaquetas dos pacientes com AN. Nas hemácias, a atividade e a expressão da NOS, a atividade da arginase e o estresse oxidativo foram investigados. Os níveis plasmáticos de aminoácidos e marcadores de estresse oxidativo no soro foram também analisados. Nossos resultados demonstram que na AN o influxo de L-arginina via sistema y+L, a produção de GMPc e a atividade da NOS estão diminuídos nas plaquetas em relação aos controles. Ainda nas plaquetas dos pacientes com AN, a expressão das isoformas da NOS bem como a atividade plaquetária não se mostrou alterada em relação aos controles. Nas hemácias destes pacientes, observou-se reduzida atividade da NOS e uma elevada função da arginase. A expressão das isoformas da NOS em hemácias não foi afetada pela AN. A concentração plasmática de L-arginina estava reduzida em pacientes com AN. A formação de TBARS nas plaquetas, hemácias e no soro não estava alterada em pacientes com AN enquanto que a atividade da SOD mostrou-se alterada. Assim, apesar da baixa produção de NO, os pacientes anoréticos apresentam um mecanismo cardioprotetor compensatório independente do NO a ser investigado. As descobertas aqui apresentadas contribuem para uma melhor compreensão da fisiopatologia da AN.

▼ Depressioon on psüühikahäire, mida iseloomustab ennekõike alanenud meeleolu, huvideringi kitsenemine, kehakaalu muutused, unehäired, keskendumise ja mõtlemise häired. Depressioon on sage ja palju-uuritud haigus, ent selle täpsed neurokeemilised mehhanismid on tänini ebaselged. Praegu kasutusel olevad antidepressandid ei ole piisava efektiivsusega- nii on probleemiks aeglane depressioonivastase toime algus ning suhteliselt suure protsendi haigete ravile mittereageerimine. Seega on vajalik depressioonimehhanismide parem mõistmine ning uute märklaudade leidmine selle raske haiguse raviks.
Lämmastikmonooksiid (NO) on tavatute omadustega virgatsaine närvisüsteemis. NO-d sünteesib L-arginiinist NO süntaas (NOS), lisaks lähtub L-arginiinist agmatiini süntees. Käesoleva uuringu eesmärkideks oli täpsustada NO osalemist depressiooni mehhanismides ning teise võimaliku mediaatormolekuli, agmatiini, rolli selgitamine.
Meie uuringutes omas agmatiin antidepressiivset toimet nii hiirte sundujumise testis kui ka efekti obsessiiv-kompulsiivse käitumise regulatsioonis nn kuulide matmise mudelis. Agmatiini toimed ei olnud vahendatud serotoniinergilise süsteemi ega NO süntaasi poolt.
Varem on leitud, et NO eellasmolekuli L-arginiini manustamine kõrvaldab mõningate antidepressantide efekti hiire sundujumise testis. On viidatud ka võimalusele, et mõned antidepressandid võivad toimida kui NOS-i inhibiitorid. Meie tulemused andsid kinnitust väitele, et osad antidepressandid on võimelised inhibeerima NO sünteesi ajus ning see mehhanism võib olla seotud NO eellasmolekuli L-arginiini võimega vähendada antidepressantide toimet. Kuna mõnedel antidepressantidel ei ole akuutselt manustatuna toimet sundujumise testis, ent need on efektiivsed kuulide matmise testis, siis uurisime lisaks, kas L-arginiini eelnev manustamine kõrvaldab ka selliste antidepressantide toime. L-arginiini eelnev manustamine vähendas antidperssantide toimet kuulide matmise testis, mille tulemusena võib öelda, et NO osaleb obsessiiv-kompulsiivse käitumise regulatsioonis.; Depression is a frequently seen psychiatric illness resulting in loss of psychosocial ability. The multiple symptoms of depression include anhedonia, depressed mood, inappropriate guilt, apathy, fatigue etc . Although a range of antidepressant medications are available, a substantial number of patients either do not respond adequately to these or are unable to tolerate their adverse effects. The limitations of current antidepressant drugs have warranted on-going research to identify pharmacological agents and strategies offering a greater therapeutic efficacy.
Nitric oxide (NO) is an unconventional neurotransmitter, involved in many physiological and pathological processes. NO is synthesized from L-arginine by NO synthases (NOS). ▼ This dissertation is written in chapter format following the guidelines of the Graduate School of The University of Georgia. Chapter 1 will serve as an introduction to provide readers with relevant literature review. Chapters 2, 3 and 4 are
copies of manuscripts prepared for submission to peer-reviewed journals. Chapter 5 provides a conclusion with emphasis on future research directions. Part of my research interest focuses on the design, synthesis and evaluation of mechanism-based
inhibitors for tryptophan indole-lyase (TIL) as an approach to a novel class of antibacterial treatment (Chapter 2). Our results indicated that homologation of the physiological substrate, L-tryptophan, at the Cα position effectively inhibited the
activity of TIL while displaying high selectivity in preference to tryptophan synthase. The accepted mechanism for TIL and tyrosine phenol-lyase (TPL) is remarkably similar. Therefore, we also extended our design and rationale in the search for potent
mechanism-based inhibitors of TPL. Synthesis, inhibition kinetics and pre-steady-state kinetic evaluation of inhibitors for TPL is discussed in Chapter 3. TIL can also catalyze the reverse of its physiological reaction, β-substitution of indole, to yield
L-tryptophan. As a proof-of-concept, we have demonstrated and validated the biosynthetic pathway for L-tryptophan from glycerol surplus by coupling natural glycerol metabolic enzymes of Escherichia coli with TIL expressed from tnaA plasmids (Chapter 4).
This work presented an attractive and convenient alternative to the multi-step chemical synthesis for L-tryptophan, using a “one-pot” approach with whole-cell catalysis. The final chapter will provide a discussion on future research direction for the
work reported in this dissertation.

platelet aggregation, lactic acid, and nitric oxide synthase in
normal health male subjects and the consequent results on maximal
oxygen consumption. The hypothesis of this study
is that with treatment of oral

will be a differential response of VO2max and lactic acid

hypercholesterolemia, hypertension, and heart disease. However,
l-
arginine supplementation has not been validated in terms of its
cardiovascular effect in healthy populations although it has been
studied for weight training. The benefits of using this supplement
in cardiovascular training may be related to NO formation which has
been shown to dilate blood vessels. Lactic acid accumulation may be
delayed or reduced because of better oxygen delivery. Should these
effects be substantiated by this study, use of
l-
arginine as a
cardiovascular endurance supplement could provide a legal and
healthy way for people to condition. Previous research has
demonstrated possible differential responses to
l-
arginine
supplementation and the physiological markers listed above. These
effects should cause increases in time to exhaustion and
VO
2max. Twenty-five
healthy males between the ages of eighteen and thirty were
recruited. Subjects who volunteered to participate in the research
completed an informed consent and health history questionnaire and
were measured for height, weight, body composition, and blood
pressure. Based on the answers to the questionnaire and the
measurements recorded, the subjects who qualified were allowed to
continue in the study. If the potential participant answered yes to
any question and/ or does not fall within acceptable quantified
limits, they were disqualified from the study. The three quantified
items on the Health history questionnaire are alcohol consumption
(< 3 drinks per day), excessive fatigue (excluded if still
feeling consistently fatigued after at least 7 hours of sleep in
previous 24 hour period) and if they have had mononucleosis within
six months of first trial date. Subjects were also excluded if they
are > 30% body fat, due to an increased chance of a cardiac
event, have a resting blood pressure > 140 mm/Hg systolic and
> 90 mm/Hg, again due to an increased chance of a cardiac event.
Females were also were excluded because of potential variations on
markers from the menstrual cycle. To provide
