▼ This thesis was conducted to investigate risk factors for
ketosis development in lactating dairy cattle and evaluate treatments for affected animals. Four main studies were carried out. A systematic review of the
ketosis treatment literature was performed to analyze the current body of literature available and provide guidance for treatments to be used in future studies. Secondly, a randomized clinical trial was performed on seventeen commercial dairy farms to determine the effectiveness of a combination butaphosphan cyanocobalamin product, insulin, and propylene glycol for
ketosis treatment. A second randomized clinical trial was performed on nine commercial dairy herds to further evaluate the usefulness of a combination butaphosphan cyanocobalamin product and two durations of propylene glycol treatment on
ketosis resolution and early lactation milk production. Finally, records from five commercial dairy farms were analyzed to evaluate individual cow risk factors associated with
ketosis development. Evaluation of the
ketosis treatment literature revealed the lack of well-designed
ketosis treatment studies and the need for further investigation to determine an effective treatment regimen. Both treatment trials showed an effect of blood glucose concentrations at the time of enrollment on the efficacy of study treatments that had not been previously described in the literature. Animals that had blood glucose < 2.2 mmol/L at the time of
ketosis diagnosis were more likely to cure and produced more milk when treated with insulin, butaphosphan cyanocobalamin, or extended duration of propylene glycol than untreated controls with blood glucose < 2.2 mmol/L. Treatment benefits did not extend to animals with blood glucose > 2.2 mmol/L at the time of enrollment. Older age at first calving, extended days open in the previous lactation, longer dry period, and increased parity increased
ketosis risk. Also, animals that were ketotic during a lactation were more likely to become ketotic in the subsequent lactation. The information contained in this thesis helps increase understanding of
ketosis risk and proper treatment. The novel interaction of the level of blood glucose and
ketosis sheds light on previous inconsistent results on
ketosis impacts and will change approaches to understanding and treatment of the condition in the field.

▼ The most common metabolic disease affecting the adipose tissue in sheep and dairy
cattle is
ketosis. In these days it is thought that food (poor quality or an inadequate
quantity) is one of the most important causes of
ketosis. In this study we used ewes (2
ketotic and 2 control animals) and dairy cattle (6 Holstein Frisian dairy cattle)
adipocyte tissue and blood.
The aim of the study in ewes is to see if the rate of lipolysis changes when we
incubated the adipocyte tissue with isoproterenol, insulin and beta-hydroxybutyrate
(BHB) and the combination of isoproterenol insulin and the combination of BHB and
insulin is different between ewes with or without
ketosis.
The aim of the study in dairy cattle is to see if there is an alteration in the lipolysis rate
during the transition period (last week of gestation until the third week of lactation).
We also tested of the differences are seen in the different incubations we did.
In the ewes isoproterenol stimulates the lipolysis rate, BHB inhibits the lipolysis rate
and insulin had little or no effect. The combinations of insulin with isoproterenol also
elevated the lipolysis rate and the combination of insulin and BHB reduces the
lipolysis rate. Not any of these effects where significantly. The blood samples where
taken to confirm if the ewe was ketotic or not.
In dairy cattle the incubations we tested are isoproterenol, insulin 50 pM, 100 pM, 500
pM, BHB and the combination of isoproterenol with insulin 50 pM and isoproterenol
combined with insulin 500 pM. Isoproterenol stimulates the lipolysis, insulin had little
or no effect, BHB caused a reduction in lipolysis activity and the combination of
isoproterenol and insulin elevated the lipolytic activity. Only the reduction of lipolysis
caused by BHB is significant.

▼ The ability of dairy cattle to adapt to the natural change of energy balance in early lactation is an important aspect of the transition period, as the demands for milk production cannot be met by feed intake alone. Cattle unable to adequately transition from late gestation to early lactation are at a higher risk for subclinical
ketosis (SCK), an excess of circulating ketone bodies without clinical signs of hyperketonemia. Cows with SCK are at an increased risk of postpartum diseases such as displaced abomasum and metritis, and SCK has been found to decrease milk yield in early lactation and may adversely affect reproduction. The objectives herein were to: 1) describe the epidemiology of SCK in cows diagnosed with SCK in early lactation; 2) determine important dry and parturient period risk factors of hyperketonemia development; 3) determine the effect of oral administration of propylene glycol in cows diagnosed with SCK on disease development, removal from the herd, reproduction, and milk production; and 4) estimate the cost per case of SCK and evaluate different on-farm testing and treatment strategies based on herd SCK incidence. Peak incidence and prevalence of SCK was found to occur at 5 days in milk (DIM) with a median time to resolution of 5 days. Cows developing SCK from 3 to 7 DIM were more likely to suffer from negative disease and production outcomes than cows that developed SCK from 8 to 16 DIM. Treatment of SCK positive cows with propylene glycol decreased disease incidence, improved reproduction, and enhanced milk production over non-treated control cows. Risk factors associated with development of hyperketonemia included advanced parity, high prepartum non-esterified fatty acid concentrations, and calving difficulty. The cost per case of SCK in the first 30 DIM was estimated at $67. Testing of fresh cows 2 days per week from 3 through 9 DIM and treatment of SCK positive cows with propylene glycol was found to be the most cost-effective treatment in herds with SCK incidences between 15 and 50%; above 50% blanket treatment of all fresh cows with propylene glycol was more economically beneficial.

▼ This thesis describes an evaluation of the impact of administering recombinant bovine somatotropin (rbST) before calving on health, milk production, reproductive performance and dry matter intake (DMI) of lactating dairy cows. In addition, this thesis includes an evaluation of the cost of subclinical
ketosis (SCK), economic value of using Rumensin® Controlled Release Capsules (CRC) for disease prevention and the economic value of using propylene glycol (PG) for treatment of SCK after being diagnosed with different cow-side tests. One randomized clinical trial was conducted on 4 commercial dairy herds in Southern Ontario and at the University of Guelph where rbST was administered every other week starting at 28-22 days before calving until cows calved. The key finding of this study was that pre-partum rbST administration had no significant clinical impact on health and performance of dairy cows after calving. A spreadsheet was constructed to perform the economic analysis of
ketosis cost and treatment. It was estimated that one case of SCK can cost up to $350. The economic value of using CRC to reduce
ketosis depends on the prevalence of
ketosis-related diseases in a herd. Using a cow-side test with high sensitivity and specificity to diagnose SCK pays off through minimizing the losses by classifying fewer cows as false negatives and therefore treating more sick cows and mitigating the negative impact of SCK. Results presented in this thesis highlight the insufficiency of rbST administration in the pre-partum period only, to improve health in the transition period and to improve cow performance after calving. Results of an economic analysis highlight the high cost impact of SCK and the importance of reducing its incidence through implementing management during the transition period.

▼ Eighty PVC mini silos were used to evaluate corn silage fermentation characteristics with four homo- or heterofermentative inoculants. Freshly chopped corn silage was untreated Control (CON), or treated with Promote HN-3 (PHN), Promote LC-1000 (PLC), or Vigorsile EBL II (VEBL) before packing. Silos were opened at d 30, 60, 90 and 120 d. Averages over all time points, PLC and VEBL reduced pH compared to CON, and PLC had greatest lactic acid concentration and In vitro starch digestibility compared to CON, PHN, and VEBL. Ninety-six PVC mini silos were used to evaluate corn silage fermentation characteristics with four homo- or heterofermentative inoculants. Freshly chopped corn silage was untreated Control (CON), or treated with Promote AP (AP), Promote EBL (EBL), or Promote BP (BP) before packing. Silos were opened at d 30 and 90. After 90 d ensiled, corn silage with BP had higher pH, lower lactate, higher acetate concentration, and increased aerobic stability compared to CON, AP, and EBL. A 2 × 2 factorial randomized block design was used with 17 cows/treatment or 8 primiparous cows/treatment resulting in four treatments; 1) low starch + carrier (LSC), 2) low starch + Propionibacterium freudenreichii strain P169 (P169) (LSM), 3) high starch + carrier (HSC), 4) high starch + P169 (HSM). Starch amount was 15.5% and 26.7% prepartum and 20.1% and 29.7% postpartum. Multiparous cows fed HSM produced the greatest milk yield. Milk yield was similar for LSM and HSC. Feed efficiency was increased with P169. High starch tended to increase 3.5% fat corrected milk and energy corrected milk yield in primiparous cows. Also, high starch increased postpartum liver total lipid and triacylglycerol content. A retrospective analysis was conducted using 161 multiparous dairy cows to investigate easily measured, low cost variables correlated to subclinical ketosis in fresh cows. In addition, 328 cows from two commercial dairy farms were utilized to develop a ketosis prediction model (Pre-K) using day of calving data: colostrum yield (CY), body condition score, and parity. An increase in CY correlated with an increase in beta-hydroxybutrate concentration on d1 and 7. Pre-K model resulted in a sensitivity of 60% and specificity of 64% for predicting cows at risk for ketosis.

▼ Ketosis is a severe metabolic disease caused by negative energy balance (NEB) that leads to decreased milk production. Thus, the main objectives were to investigate literature information on the genetic background of NEB, subclinical and clinical
ketosis through a systematic review and meta-analysis and to identify genomic regions associated with subclinical and clinical
ketosis in first and later lactations, and also for subclinical
ketosis in two periods in early lactation (5 to 21 DIM and 22 to 45 DIM) by performing genome wide association studies (GWAS). The weighted single-step GBLUP (wssGBLUP) method was used for GWAS to identify genetic regions associated with the traits. The genes identified within the top-20 genomic windows were used for functional analyses. Biological understanding of this metabolic disease will help enhance accuracy of selection in the Canadian dairy cattle breeding programs and help with the development of biomarkers for early diagnosis and prevention of
ketosis.

▼ The primary purpose of this investigation was to
compare performance adaptations in tactical athletes utilizing a
ketogenic diet (KD) vs. control (CON) during a 12-week strength and
conditioning program in conjunction with their standard military
fitness training. Little research has been conducted on the effects
of a ketogenic diet on strength and power outcome goals, yet a
ketogenic diet has demonstrated to be physiologically beneficial in
other populations. Participants self-selected to a diet
intervention KD (n=15), or CON (n=14). The training intervention
included 6 weeks of resistance training, emphasizing back squat and
bench press, and 6 weeks of power training, emphasizing Olympic
lifts and plyometric training. PRE and POST-intervention,
countermovement jump (CMJ), 1 RM back squat and bench press,
repeated sprint intervals, an obstacle course, VO2 Max, and body
composition measures were assessed. At Week 1 (MP1) and Week 6
(MP2) an abbreviated performance battery was conducted including
CMJ, 1 RMs, and the sprint intervals. For KD, BF% was significantly
higher PRE (25.6 ,SE 5.0) than POST (20.26, SE 4.9) compared to CON
PRE (22.0, SD 8.6) and POST (21.3, SD 8.4). For KD, FM was
significantly higher PRE (46.5, SE 9.8) than POST (33.6, SE 7.6)
compared to CON PRE (38.2, SD 17.5) and POST (36.9 SD 16.7). For
KD, LBM was significantly higher PRE (135.9, SE 13.7) than POST
(132.9, SE 14.8) compared to CON PRE (131.4, SD 9.5) and POST
(133.1, SD 10.1). There were no significant effects between groups
for performance measures. Absolute back squat PRE (mean 110.33, SE
5.41) and MP1 (mean 111.72, SE 4.76) were both significantly less
than MP2 (mean 122.53, SE 4.47) and POST (mean 125.90, SE 4.50).
Relative back squat PRE (mean 1.34, SE .06) and at MP1 (mean 1.37,
SE .06) were both significantly less than MP2 (mean 1.53, SE .06)
and POST (mean 1.59, SE .06). Relative bench press PRE (mean 1.10,
SE 0.07) and at MP1 (mean 1.11, SE 0.07) were both significantly
less than MP2 (mean 1.16, SE 0.07) and POST (mean 1.18, SE 0.07).
There was no effects of time or training on power measures. In
summary, resistance training two days per week significantly
improved leg strength but had little effect on upper body strength
and power. A ketogenic diet had similar strength gains and
performance measures compared to CON. This indicates a ketogenic
diet can be an effective weight management tool in tactical
athletes without causing detriment to power and strength-related
tasks.

▼ Background: The optimal dietary approach to weight loss is a hotly debated topic among health professionals and the lay public alike. An emerging body of evidence suggests that a higher level of adherence to a diet, regardless of the type of diet, is an important factor in weight loss success over the short and long term. The overall aim of this thesis was to examine how dietary interventions can be designed to improve and measure adherence and thereby increase the success of dietary weight loss interventions. Methods: Four studies were conducted to achieve this aim. Study 1 (Chapter 2) is a systematic review and meta-analysis of appetite responses to two types of ketogenic diets (very low energy diets, VLEDs; and ketogenic low carbohydrate diets, KLCDs). Study 2 (Chapter 3) is a modelling study that presents the design process and underlying rationale of the dietary intervention of a study comparing fast versus slow weight loss. Study 3 (Chapter 4) compares the nutritional content, nutritional adequacy and cost of all VLED products in Australia. Study 4 (Chapter 5) is a validation study of a new method that was developed to improve the accuracy of portion size estimation and in so doing, measures of adherence. Results and discussion: Study 1 found that VLEDs and KLCDs are indeed associated with a suppression of appetite and that ketosis is a plausible mechanism underlying the effect. Study 2 demonstrated the feasibility of designing a food-based, moderately energy-restricted diet that can be individualized to a person’s dietary preferences, while still aligning with dietary guidelines and containing adequate protein for women of various sizes. Study 3 provides practical clinical guidance by demonstrating how commercially available VLEDs can be optimised in protein, which could improve adherence via increased appetite control. Study 4 found that the new method of estimating portion sizes could improve the accuracy of portion size estimation methods which may improve adherence measures. Conclusions: There are several key targets that have the potential to improve and measure adherence to dietary weight loss interventions and thereby weight loss success. By examining the design of strategies that are practical, affordable and feasible, the findings of this thesis have direct implications for clinicians tackling the obesity epidemic in research and real world settings.

ketosis. Two Dutch farms provided the data for nearly four consecutive years of testing all cows with the Precision Xtra test. There was no reduction of
ketosis seen during this time. Lowering the energy level of the rations for the dry cows was effective on the B farm. There was a lower incidence of
ketosis seen on the A farm during the grazing season. Testing of all fresh cows for
ketosis was not useful for evaluating transition management due to frequent changes in this study.

▼ Elevated ketone production and utilization results in a host of health benefits. The aim of this study was to assess the rate of ketone production during a prolonged fast and to evaluate how an initial bout of exercise influences this production. Mood and hunger, along with plasma insulin and glucagon, were also assessed. In this crossover study, 20 adult subjects (11 Male, 9 Female) completed two 36-hour fasts, with one protocol requiring the subject to complete an intense treadmill exercise session at the beginning of the fast. Ketone levels were assessed via blood ketone meter and recorded every two hours. Subjective mood and hunger ratings were also recorded every two hours. Venipuncture was performed every 12 hours to assess plasma insulin and glucagon. The mean area under the ketone production curve for the nonexercise intervention was 19.19 ± 2.59 mmol/L and 27.49 ± 2.59 mmol/L for the exercise intervention, resulting in a significant 8.30 mmol/L difference between conditions (95% probability interval was 1.94 to 14.82 mmol/L). The mean time to nutritional ketosis was 21.07 ± 2.95 hours with fasting alone, and 17.5 ± 1.69 hours when combined with exercise (posterior probability = 0.89). There was a significant decrease in insulin over time (F(3,133) = 61.75, p < 0.0001). There was also a significant increase in glucagon over time (F(3,133) = 21.10, p < 0.0001). Hunger and stomach discomfort did not differ between conditions. Anger (F(10,394) = 2.74, p = 0.0028), depression (F(10,394) = 2.91, p = 0.0016), tension (F(10,394) = 2.29, p = 0.0128), vigor (F(10,394) = 11.65, p < 0.0001), and fatigue (F(10,394) = 10.60, p = 0.0001) increased over the course of the fast, but did not differ between conditions. Completing aerobic exercise at the beginning of a 36-hour fast results in significantly more ketone production. The impact of exercise on ketone production comes at little or no impact on hunger, stomach discomfort and negative moods. A difference in time to achieving nutritional ketosis between conditions may exist, but this was not observed in this study.

ketosis (SCK) in transition dairy cows. Rumination time, lying behaviour, SCK, and other peripartum disorders were monitored from 2 wk before until 4 wk after calving for 339 cows. Lower rumination times and higher lying times in multiparous cows during the wk after calving (wk+1) were associated with increased odds of SCK with another health problem. Factors associated with lower odds of SCK in multiparous cows included: lower stall stocking density during wk+1, shorter dry period, lower milk yield during the previous lactation, and smaller BCS loss over transition. These results suggest monitoring behaviour may be useful in identifying multiparous cows with SCK and another health problem in wk+1; monitoring rumination behaviour, specifically, may aid in the early identification of multiparous cows at higher risk for developing SCK post-calving.

▼ Although dietary restriction usually results in weight loss, the majority of obese dieters eventually regain the weight that they have lost. Body weight is centrally regulated, with peripheral hormonal signals released from the gastrointestinal tract, pancreas and adipose tissue being integrated, primarily in the hypothalamus, to regulate food intake and energy expenditure. The peripheral modulators of appetite include leptin, ghrelin, cholecystokinin (CCK), peptide YY (PYY), insulin, pancreatic polypeptide (PP) and glucagon-like peptide 1 (GLP-1). Caloric restriction results in acute compensatory changes, including profound reductions in energy expenditure and circulating satiety hormones such as leptin, with increases in ghrelin and appetite, which encourage weight regain.
The regulation of body weight is not completely understood, and the present study was conducted to contribute to knowledge in the following areas:
1. Whether changes in appetite-regulating hormones after weight loss are sustained following prolonged weight maintenance.
2. Ketosis thought to suppresses hunger, but its effect on circulating hormonal mediators of appetite is largely unknown.
3. Few studies have examined psychological and behavioural factors as predictors of weight loss maintenance.
In the present study, 50 overweight or obese non-diabetic men and post-menopausal women undertook an 8-week ketogenic very low energy diet (VLED), in order to lose ≥10% of their initial weight. This was followed by a 2 week transition to regular foods (to maintain weight loss while reducing circulating ketones), and 52 weeks of follow-up, during which they were aiming to maintain their reduced weight. Prior to the commencement of the VLED, subjects completed five questionnaires assessing eating behaviours and psychosocial factors, which were used to predict eventual weight outcome.
Participants lost an average of 14% of their starting weight during the initial weight loss period. This was accompanied by significant reductions in the anorexigenic hormones leptin, insulin, CCK, PYY, GLP-1 and amylin, and increases in ghrelin, GIP and PP. Subjective ratings of appetite increased after weight loss. The majority of changes were in a direction which would encourage weight regain, and most persisted 12 months after initial weight loss.
When weight-reduced subjects were ketotic, the circulating concentrations of several hormones and nutrients which influence appetite were altered compared with after resolution of ketosis. Although there appeared to be a combination of orexigenic and anorexigenic effects of ketosis on circulating factors, subjective ratings of appetite were clearly lower in weight-reduced subjects during ketosis.

▼ Forty-one (10 primiparous and 31 multiparous) Holstein cows were used in an experiment to determine the effects of prepartum dietary energy (PDE) and supplemental Ca-propionate on the incidence of ketosis in transition dairy cows and to evaluate the usefulness of urine pH as a predictor for ketosis. Treatment factors were prepartum dietary energy concentration (normal vs. high) with or without 113.5g Ca-Propionate/day (Nutro Cal®, Kemin Industries, Inc., Des Moines, IA). All cows were fed the same basal diet postpartum. Ca-propionate supplementation continued postpartum. Cows were individually fed and dry matter intakes (DMI) were measured daily. Milk production was measured and sampled at each milking for 3 weeks postpartum and analyzed for % fat, % protein, and SCC (somatic cell count). Blood samples were collected prior to the afternoon milking 3x/ wk and analyzed for β-hydroxybutyrate (BHBA) and glucose concentrations. Urine was collected 3x/ wk and analyzed for pH, acetoacetate, and BHBA. Treatment did not affect mean DMI, milk yield, milk component production or percentage, 4% FCM, SCCS, BWT, urine pH, urine acetoacetate, urine BHBA, plasma BHBA, or plasma glucose. There was a statistical interaction of Ca-propionate supplementation and week for urine pH and BHBA. Correlation coefficients between urine BHBA, urine pH, and urine acetoacetate were not useful for prediction of ketosis across all cows. However, they were numerically higher when restricted to data from an individual cow. Correlation coefficients between BHBA and acetoacetate concentrations in urine were significant. These data suggest that PDE and Ca-propionate supplementation had no effect on the incidence of ketosis, however Ca-propionate supplementation allowed cows to recover faster from ketosis postpartum. The use of urine pH was not a useful tool for the prediction of ketosis across a herd, but may be a useful indicator of ketosis when compared within an individual animal. Due to factors unrelated to treatment diets, there was a high incidence of health disorders such as retained fetal membranes, metritis, displaced abomasums, and laminitis were observed during this trial that probably affected any observed treatment responses.

O presente estudo foi realizado numa exploração agropecuária situada na Freguesia de Benavente, Distrito de Santarém que se caracteriza por ser uma exploração de regime intensivo de produção de leite.
O estudo insere-se numa metodologia de estudo de caso e a amostra inclui 131 fêmeas selecionadas ao acaso entre novilhas e vacas adultas que concluíram a gestação durante o período do estudo. A recolha de dados iniciou-se com o parto através da medição da condição corporal (BCS). Posteriormente entre o décimo quinto (D15pp) e décimo oitavo dia pós-parto (D18pp) foi efetuada uma medição da concentração sanguínea de β-hidroxibutirato (BHB) e novamente avaliação BCS. As vacas leiteiras (VL) com concentrações sanguíneas de BHB acima de 1,4 mmol/l e sem sinais clínicos de cetose, eram sequencialmente incorporadas no grupo tratadas (GT), animais em que lhes era feito um tratamento, ou no grupo controlo (GC), animais que não era feito tratamento. O tratamento consistia na administração oral diária de 500 ml de propilenoglicol (PG), durante cinco dias e de um bolus endovenoso de 1L de soro glucosado 30% Braun® com 20 ml de Catosal® no primeiro e no terceiro dia de tratamento. Passados 5 dias após a primeira medição da concentração sanguínea de BHB esta voltava a ser mensurada nos animais pertencentes ao GT e GC.
Os resultados obtidos indicaram que as vacas multíparas sem cetose subclínica (CSC) dão mais leite (91,5 ± 38,28; p = 0,0185) do que as vacas com CSC. As VL com CSC têm maior probabilidade de desenvolver cetose clínica (p = 0,0001). Não existem diferenças significativas (p = 0,42) entre as vacas tratadas e as vacas não tratadas na produção de leite. Também se verificou que nas VL tratadas a concentração sanguínea de BHB diminui significativamente (p = 0,038) mas que o tratamento não tem qualquer influência na prevenção da cetose clínica (p = 0,6817), deslocamento do abomaso à esquerda (DAE) (p = 1) e mastite (p = 0,4828). Considerando apenas a produção de leite, o tratamento de vacas com CSC não aparenta ter retorno económico nos 15 dias após o tratamento (p = 0,16). No entanto outros estudos devem ser realizados de forma a contabilizar outros fatores que não apenas os associados à produção de leite momentânea e comparar esta medida com outras medidas preventivas e o seu impacto económico nas explorações tendo em conta os custos não contabilizados neste trabalho

▼ Le Ketosis-Prone Diabetes (KPD) est un phénotype de diabète intermédiaire entre le diabète de type 1 et le diabète de type 2, fréquemment rencontré chez le sujet d’origine noire africaine. Cette forme de diabète suscite un intérêt certain de par son évolution clinique marquée notamment par la restauration de l’insulinosécrétion initialement altérée. Sobngwi et coll. en 2008 ont établi une association entre le virus HHV8 et le KPD chez des sujets Africains vivant en France. Nulle part ailleurs l’étude n’a été reproduite. L’objectif de cette thèse était de rechercher la potentielle association entre l’infection à HHV8 et le KPD ; puis d’évaluer l’impact de l’infection à HHV8 sur le profil inflammatoire des phénotypes du diabète de type 2. L’étude s’appuie sur une population de patients Africains vivant en Afrique admis consécutivement pour une décompensation hyperglycémique (glycémie à jeun≥2,5g/l) au Centre National d’Obésité de l’Hôpital Central de Yaoundé. Plus spécifiquement, il était question de :• étudier la fréquence du diabète non auto-immun à tendance cétosique (KPD); • étudier l’association entre le virus HHV8 et le KPD;• rechercher si l’infection à HHV8 est associée à un profil inflammatoire pouvant participer aux phénotypes de diabète. Etait inclus dans l’étude tout patient diabétique âgé de plus de 18 ans présentant un diabète aigu avec syndrome cardinal et cétonurie (KPD1), ceux ayant présenté un diabète inaugural aigu avec syndrome cardinal et cétonurie et en rémission depuis plus de trois mois et sans cétonurie à l’inclusion (KPD2), et ceux présentant un diabète de type 2 connu sans cétonurie (DT2). Etait exclu de l’étude tout patient présentant des stigmates d’auto-immunité du diabète de type 1 A, un diabète « MODY », une endocrinopathie, une maladie du pancréas, ou un diabète de type auto-immun. Chez l’ensemble des participants admis, nous avons collecté les données cliniques (le poids, la taille, l’IMC, le rapport tour de taille sur tour de hanche, la pression artérielle, et le pourcentage de graisse) et des prélèvements à jeun ont été effectués (sérum et cellules mononuclées du sang périphérique) pour les analyses biologiques : la glycémie par glucose oxydase, l’HbA1c par HPLC, les paramètres du profil lipidique par des methodes enzymatiques, les concentrations d’insuline et de peptide-C par électrochimiluminescence, les anticorps anti-HHV8 par immunofluorescence, l’ADN viral HHV8 par PCR en temps réel, et les marqueurs de l’inflammation par le Luminex. The objectives were to 1.) determine if a higher betahydroxybutyrate(BHBA) level (≥1.2 mmol/L) measured at 5 days in milk (DIM) results in a lower first service conception success; 2.) study the relationship of BHBA ≥ 1.2 mmol/L measured at 5 DIM with the following variables: season, parity, body condition score at calving (BCSCavC) and 40 DIM (BCS40C), retained placenta (RP), metritis, mastitis, respiratory diseases (Resp), displacement abomasum (DA), lameness (Lame), clinical endometritis (CE), cyclic and AI at estrus. The null hypothesis is that there is no association between BHBA ≥ 1.2 mmol/L and first service conception success.
From November 2012 to august 2013, 1197 Holstein-Friesian cows were studied. Cows were enrolled at calving till the second pregnancy check on 60 ± 3 days after first insemination. Cows were excluded if sold or dead within 60 DIM or when data was incomplete. The definition of P32AI1 is the presence of an amniotic vesicle holding an embryo with heartbeat at 32±3 days after first artificial insemination (AI). Subclinical
ketosis (SCK) is defined as serum concentrations of BHBA ≥ 1.2 mmol/L. Cows were inseminated when standing heat was observed or after timed AI (TAI). All cows had serum blood collected on day 5 post-partum from the coccygeal artery or vein and were analyzed for concentrations of 3-OH-butyrate (BHBA; Autokit 3-HB, Wako). For the statistical analyses 1041 Holstein-Friesian cows were included. All analyses were performed with the SAS system version 9.3.
Elevated BHBA (≥1.2 mmol/L) concentrations measured with the Wako-kit on 5 DIM are not associated with pregnancy after first service. The logistic regression model with P32AI1 as response variable selected by backward elimination includes 2 variables that were significantly associated; RP and mastitis; 3 variables that have a tendency toward statistical differences; DA, Lameness, and CE. So, the probability to become pregnant after first service with RP, mastitis, DA, lameness or CE is smaller than to become pregnant without these diseases.
The logistic regression model with BHBA (≥1.2mmol/L) as response variable shows a statistically significantly relationship between season, parity, BCS at calving, mastitis, DA, and a tendency for statistical difference for BSC at 40 DIM and CE. The odds of having SCK in summer was 1.7 times greater than the odds of having SCK in winter. Primiparous had lesser odds of SCK than multiparous. Cows with a high (≥3.5) BCS at calving have 2.4 times greater odds of SCK than cows with a BCS lower than 3.0. Cows without DA were 0.5 times as likely to develop SCK as cows with DA. The odds of SCK in cows with BCS ≥ 3.5 at 40 DIM was 0.5 times less than the odds of SCK in cows with BCS < 3.0 at 40 DIM. The odds of SCK in cows without CE was 0.7 times smaller than the odds of SCK with CE. The odds of SCK without mastitis is 1.8 times more than the odds of SCK with mastitis. This is the first study reported such a relationship between SCK and mastitis.

▼ A toxemia da prenhez (TP) ocorre principalmente em ovelhas e cabras, suplementadas com concentrado, cuja manifestação se deve a um balanço energético negativo durante o período de transição, no qual as fêmeas apresentam quadro clínico de hipercetonemia, hipoglicemia, cetonúria, anorexia e sinais neurológicos que pode evoluir para a morte. Este trabalho se propõe a realizar um estudo do perfil bioquímico e hormonal em cabras e ovelhas, e anatomopatológico do parênquima hepático e renal em ovelhas com diagnóstico de TP. Foram utilizados dados (fichas clínicas) e material biológico (soro, plasma e tecido) de 45 ovelhas e 22 cabras que foram atendidas na Clínica de Bovinos, Campus Garanhuns/UFRPE, diagnosticadas com TP, no período de janeiro de 2007 a outubro de 2012. As análises realizadas no sangue foram: uréia, creatinina, albumina, proteína total, globulina, β-hidroxibutirato, ácidos graxos não esterificados (AGNE), glicose, frutosamina, amilase, aspartato aminotransferase (AST), gama glutamiltransferase (GGT), vitamina B12, ácido fólico, cortisol, insulina, Ca total e ionizável, P, relação Ca:P, Mg, Na, K. Cl. Além da caracterização dos achados macro e histopatológicos do fígado e rins das ovelhas que vieram a óbito. Na urina foi examinada a presença de corpos cetônicos empregando-se a fita reagente comercial. Das ovelhas submetidas à necropsia foram coletados fragmentos de fígado e rins para análise histopatológica. Marcadas alterações clínicas foram observadas nas cabras, com valores elevados para ureia, creatinina, AGNE, β-hidroxibutirato, amilase e cortisol, enquanto que a proteína total, albumina, cálcio total, cálcio ionizado, fósforo, a relação cálcio e fósforo, o sódio e a insulina encontraram-se abaixo dos valores de referência para a espécie. Quanto à glicemia, 77,28% das cabras com TP eram normoglicêmicas e/ou hiperglicêmicas, enquanto apenas 22,72% eram hipoglicêmicas. Os resultados obtidos das ovelhas revelaram valores elevados de ureia, creatinina, glicose, frutosamina, AGNEs, β-hidroxibutirato, cortisol, cloro, AST, GGT amilase e ácido fólico enquanto que a variável insulina, cálcio total, cálcio ionizado e potássio estavam com valores abaixo dos valores de referência para a espécie. Quanto à glicemia, 82,90% das ovelhas com TP eram normoglicêmicas e/ou hiperglicêmicas, enquanto apenas 17,10% eram hipoglicêmicas. Quanto aos achados macroscópicos no grupo de ovelhas com menor escore corporal, foi observado aumento do fígado, além de coloração amarelada na superfície e ao corte, enquanto que nas ovelhas com maior escore corporal estes achados foram mais intensos. Nos rins foi observada palidez na superfície em alguns dos animais magros e leve coloração amarelada em animais gordos. Quanto às alterações histológicas, verificou-se, nos animais com menor escore corporal, fígado com vacuolização que variou de rara a discreta. ▼ La fertilité des vaches laitières s’est beaucoup dégradée au cours des dernières décennies, tant à l’échelle nationale qu’internationale. L’objectif poursuivi est d’apporter un éclairage épidémiologique sur les relations dynamiques entre la conception (réussite à l’insémination artificielle ; IA), les mammites subcliniques et les déséquilibres métabolico-nutritionnels que sont la cétose subclinique et les déséquilibres azotés de la ration. Un premier chapitre bibliographique dresse l’état de l’art des liens entre performances de reproduction et (i) mammites, (ii) cétose subclinique et (iii) excès de protéines dégradables, en identifiant clairement les interactions et associations croisées entre ces différents composants, et en quantifiant autant que possible ces liens. Les trois parties suivantes mobilisent des données exhaustives du contrôle laitier français sur la période 2008-2012, qui ont été confrontées aux données d’IA. La variable d’intérêt retenue est la conception après IA, soit pour la première IA (IA1) ou pour toutes les IA. Les variations des concentrations de cellules somatiques (CCS) autour de l’IA ont été utilisées pour décrire la dynamique des mammites subcliniques autour de l’IA, selon 4 classes (Bas-Bas, Bas-Haut, Haut-Bas et Haut-Haut) et pour différents seuils de CCS. Le statut de cétose subclinique a été évalué grâce aux taux butyreux et protéique du lait. Les concentrations d’urée du lait sont utilisées pour caractériser les déséquilibres azotés. Le second chapitre propose une description succincte des résultats de production et de reproduction des troupeaux bovins laitiers français sur la période d’étude. Le troisième chapitre focalise sur l’interaction entre les CCS et la cétose subclinique dans un modèle de régression de Poisson expliquant la conception. Les chances de conception à l’IA1 sont réduites de 14% (Risque relatif = 0,86 [IC 95%=0,85–0,87]) pour les groupes Bas-Haut et Haut-Haut, comparé au groupe Bas-Bas, et de 3 à 17 %, selon les définitions retenues, lors de cétose subclinique comparé à son absence. Les résultats identifient et quantifient clairement l’interaction entre la cétose subclinique et les mammites subcliniques dans leur association avec la conception : la baisse de la conception est jusqu’à 2 fois supérieure lors de la présence simultanée d’une augmentation des CCS et d’une cétose subclinique par rapport à la situation où il y a seulement augmentation des CCS. Le quatrième chapitre, mobilisant des méthodes proches de celles du chapitre précédent, montre que la baisse de la concentration d’urée dans le lait autour de l’IA (en dessous de 150 mg/kg, 2,6 mmol/L) est associée à une baisse de la conception de 5 à 9% (Risque relatif = 0.91 (IC95%=0,87-0,96)]) par rapport à des concentrations d’urée du lait qui restent stables (250 - 450 mg/kg ou 4,3-7,7 mmol/L). O estágio realizou-se em três locais diferentes. A primeira parte foi realizada numa exploração Agro-pecuária de bovinos de leite em regime intensivo, permitindo aprofundar as práticas de maneio deste tipo de exploração. A segunda decorreu em clínica de sistema ambulatório em regime de minifúndio. A terceira parte consistiu no acompanhamento da clínica ambulatória em explorações de bovinos de leite em sistema intensivo. A revisão bibliográfica e caso clínico têm como tema “Cetose em bovinos de leite”. Esta doença afecta ruminantes em produção intensiva, quando a ingestão alimentar não é suficiente para repor os níveis energéticos exigidos. Apresenta uma alta prevalência nos efectivos, o que foi confirmado no estudo. O seu impacto económico é importante, propondo-se algumas medidas a instalar na prevenção desta afecção; ABSTRACT: This thesis here presented pretends to report the activities developed in the curricular stage made in Veterinary Medicine, under the theme Clinic of Farm Species. The stage was done in three different places. The first was developed in a dairy bovine farm, allowing me to understand the practical management of this kind of farm. The second part occurred in ambulatory clinic system, made in small-size farms. The third part had place in intensive dairy bovine farms. The bibliography revision and clinic case are under the theme “
Ketosis in dairy bovines”. This clinic condition affects high production dairy cows, when the energy intake is not enough to fulfill the energy levels required. It presents a high prevalence level, wich was conformed by the study. Its economic impact is important, reason why is proposed some prophylactic measures to take in the dairy farm.

This thesis was conducted to evaluate the effect of a commercially available proprietary feed additive that provides B vitamins and choline on the incidence of health disorders, milk yield and reproduction in early lactation in commercial dairy herds. A randomized controlled trial on three freestall dairy herds evaluated the use of 100 g/cow/d of dietary supplemental B vitamin, given 3 wk before to 3 wk after calving. Supplemental B vitamins did not have an impact on the incidence of
ketosis, clinical health disorders or anovulation, reproductive performance or udder edema. There was no improvement of milk production through the first three DHI test days with supplementation.

▼ The aim of this thesis was to evaluate the diagnostic accuracy for
ketosis (KET) detection methods, to estimate the prevalence of KET in Ontario, Canada and identify risk factors for increased within-herd prevalence and increased odds of KET at the cow-level. Finally, the benefit of including a glucocorticoid with propylene glycol in KET treatment was evaluated. A diagnostic test accuracy systematic review and meta- analysis was conducted for on-farm methods for the diagnosis of KET. The Precision Xtra handheld device that measures blood beta-hydroxybutyrate (BHBA) had the highest combined sensitivity (95%) and specificity (98%) of the methods evaluated. The ability of the Precision Xtra to accurately detect elevated prepartum blood BHBA at concentrations implicated in postpartum disease (0.6 to 0.8 mmol/L) was investigated. The device was moderately well correlated (0.77) with laboratory serum BHBA values, was in substantial agreement with laboratory NEFA identification of at-risk animals (κ = 0.64) and was associated with 2.2 fold greater odds of KET postpartum when prepartum BHBA ≥ 0.6 mmol/L. The within-herd and cow-level prevalence of KET was estimated to be 21% based on early lactation milk BHBA collected from herds participating in a dairy herd improvement program in Ontario, Canada. A longer calving interval, a longer dry period, being tested before 14 days in milk and having lower milk yield and fat percentages at the last test of the previous lactation was associated with increased odds of KET in multiparous animals. Increased odds of KET in primiparous animals were associated with being older at first calving. Being a Jersey increased odds of KET in all parities. Other factors associated with increased KET risk were calving in the spring and being from a herd with an automatic milking system. A randomized controlled trial evaluated the use of dexamethasone as an adjunctive therapy to propylene glycol treatment for KET. The addition of dexamethasone was associated with lower odds of KET in the week following treatment when blood BHBA was between 1.2 and 1.5 mmol/L at diagnosis, but the odds of KET after treatment were increased for the dexamethasone group when blood BHBA ≥ 3.2 mmol/L at diagnosis.

▼ There are several constraints to of the sustainability of the livestock industries in different areas of the world. The two most obvious hurdles to high dairy production are diseases during the transition period and heat stress (HS). Despite decades of research, the actual pathologies of ketosis and HS remain poorly understood. In the US, it was estimated that the economic losses from ketosis is 360/cow/cycle, and to HS was ~ 900 million per year. Thus, both severely jeopardize the competitiveness of animal agriculture. Regardless of the herd size, HS and ketosis affect every dairy region in the country. The biological investigations of ketosis and HS have been studied for more than 50 years, but the negative impacts of both are as severe today as they were 30 years ago.
In the current dissertation, ketosis in dairy cows and HS in pigs and steers were investigated to better understand the biology and etiology of both disorders. Study 1 (Chapter 2) was conducted to characterize biomarkers of inflammation during the transition period in healthy and clinically diagnosed ketotic cows. The results indicated that circulating NEFA and BHBA were increased and milk yield was decreased in ketotic relative to healthy cows. In addition, pre-calving circulating LPS was increased twofold in cows that were diagnosed with ketosis post-calving compared to healthy cows, but no LPS differences were detected post-partum. Post-calving LPB, SAA and Hp were increased when compared with their healthy counterparts. Our data suggest the development of ketosis may be intimately linked to inflammation and our selection criteria suggest that intestinal permeability may be the origin of maladaptation to lactation.
In Study 2 (Chapter 3) we investigated the effects of a mineral supplement (zinc amino acid complex) on temporal biomarkers of intestinal integrity and intestinal morphology in heat-stressed steers. As expected, HS increased thermal indices and decreased feed intake. However, steers supplemented with zinc amino acid complex had decreased rectal temperature, improved biomarkers of leaky gut (haptoglobin, and LBP), altered intestinal morphology (decreasing duodenum villi width, increasing jejunum villi height and jejunum and ileum villi height:crypt depth), and improvement in some of the blood gas variables relative to steers supplemented with zinc sulfate. Altogether, the findings of Study 2 demonstrated that a Zn-amino acid complex may mediate some of the negative effects of HS in a growing ruminant model.
Study 3 (Chapter 4) investigated the temporal pattern of metabolic variables and biomarkers associated with intestinal barrier dysfunction during recovery from HS in pigs. Similar to Study 2, HS increased thermal indices and decreased feed intake. Circulating glucose decreased during HS and remained low for 3 d following HS. ketosis; and to measure the effect of reducing milking frequency from 2 to 1 milking per day on
ketosis resolution and milk production in ketotic dairy cows. A study at the University of Guelph dairy research facility measured concentrations of ketones in the blood, milk, and urine daily in fresh dairy cows and evaluated a 2 wk period of once-daily milking (ODM), in conjunction with 5 d of 300 mL propylene glycol, as a treatment for
ketosis.
Ketosis was detected ~2 d later using milk and urine tests, and data suggested differences in the concentrations of ketones in milk and urine relative to changes in blood. ODM cows were more likely to decrease blood BHB concentrations below 1.2 mmol/L, and produced less milk than twice-daily milked cows.

▼ O presente estudo teve como objetivo relacionar os níveis de indicadores sanguíneos energéticos e de cálcio na ocorrência de afecções uterinas, mastite clínica e mastite subclínica em vacas de raça Holandesa. A presença de mastite foi monitorada através do California Mastitis Test (CMT) e da contagem de células somáticas, para a identificação de mastite subclínica, e do Teste da Caneca de Fundo Preta (TCFP), para identificação de mastite clínica. A presença das afecções uterinas foi identificada mediante exame clínico. Foram feitas coletas de sangue para a determinação dos níveis de cálcio (Ca), glicose e β-hidroxi-butirato (BHB). O CMT foi realizado nas duas primeiras semanas após o parto, o TCFP foi realizado conforme a rotina de ordenha, estabelecida pela propriedade, e as coletas de sangue foram realizadas na semana anterior ao parto e duas coletas até os 21 dias posteriores ao parto. A ocorrência de hipocalcemia subclínica foi determinada quando o Ca no soro atingiu valores ≤ 8,00 mg/dL, hipoglicemia com valores de glicose ≤ 50 mg/dL e hipercetonemia com valores de BHB ≥ 1,2 mmol/L. Nas vacas com mastite clínica foi realizado tratamento conforme estabelecido pelo protocolo de ordenha e tratamentos da fazenda. Os dados de mastite subclínica obtidos através do teste de CMT, por quarto mamário, foram validados através de cultura microbiológica realizada em laboratório de referência e em laboratório na fazenda para identificar crescimento bacteriano, bem como através da contagem de células somáticas (CCS) e da contagem bacteriana total (CBT). Não foi encontrada relação significativa entre a hipocalcemia subclínica, a hipoglicemia e a hipercetonemia com afecções uterinas e da glandula mamária nas vacas após o parto.
This study aimed to relate the levels of energy and calcium in the occurence of both uterine diseases, clinic and subclinic mastitis in Holstein cows. The presence of mastitis was monitored through California Mastitis Test (CMT) and the counting of somatic cells (CSC), for the identification of subclinic mastitis, and strip cup test, for the identification of clinic mastitis. The presence of uterine disorders was identified by clinical examination. Blood collections were made for determining calcium, glicose and β-hydroxy-butyrate (BHB). CMT was realized in the first two weeks after calving, BBMT was realized according to the routine of milking settled by the farm and the blood collections were made in the week before the calving and also two collections were made after it. The occurrence of subclinic hypocalcemia was determined when serum Ca reached ≤ 8,00 mg/dL, hypoglycemia with glucose values ≤ 50 mg/dL and hyperketonemia values with BHB BHB ≥ 1,2 mmol/L. ketosis and metabolic disorders caused by
negative energy balance in many modern dairy farms often are a reason for
insufficient milk yield, high rate of disease and impaired reproduction
performance. The aim of this study was to investigate the effects of feeding
propylene glycol, glycerin, and L-carnitin on metabolism and milk production.
97 Holstein Friesian cows were divided into four groups, at least five days
before calving. All four groups were fed with the same total-mixed-ration but
with different additions. Group 1 (control) received no addition, group 2
received 250 g propylene glycol/cow/day, group 3 received 312,5 g raw
glycerin/cow/day (corresponding to 250 g pure glycerin/cow/day), and group 4
received 312,5 g raw glycerin and 2 g L-carnitin/cow/day. The additions were
mixed into the TMR. Until day 100 post partum dry matter intake, milk yield
and body weight were recorded daily. Milk components were checked weekly.
Blood samples were collected on the day of division into the groups and on the
days 1, 3, 7, 14, 21, 28 and 56 p.p. and examined for certain metabolism
parameters. Determination of BCS and body-fat-thickness were taken on the day
of division and on the days 1, 14, 28, 56, 80 and 100 p.p.. Urine samples were
collected on the day of division and on the days 7, 21 and 56 p.p.. Liver
biopsies were taken on the day of division and on the days 7 and 21 p.p.. The
cows in group 2 (PG) had significant lower BHB- and NEFA-concentrations in
serum. Significantly lower were as well the milk fat/protein-quotient, liver
fat- and liver triyglyceride-content. In contrast to that group 3 (glycerin)
showed significant higher BHB- and NEFA-concentrations. Both milk fat/protein-
quotient and ECM-yield were significantly higher as in the groups 1 and 2. The
cows in group 3 needed more medical treatment. The cows in group 4 (glycerin
and L-carnitin) had the significant highest milk yield. Feed intake was
significantly greater in this group, too. In spite of the higher milk yield
all the other determined metabolism parameters did not differ. In this study
administrating of propylene glycol had the best suitability for the prevention
of
ketosis and for the improvement of the whole metabolic status. The addition
of glycerin and L-carnitin lead to an increased milk yield without making
energy balance worse.

▼ The transition period is characterized by an increase in the demand of nutrients in order to meet the requirements for the finals stages of gestation, and the initiation of lactation. The objectives of this study were to determine the association between dry matter intake (DMI) pre (-14 d) and postpartum (28 d) and postpartum diseases [dystocia, retained placenta (RP), metritis (MET), mastitis (MAST),
ketosis (KET), and displaced abomasum (DA)]. Data involving 294 cows from 7 experiments were collected. The data were analyzed with PROC MIXED and GLIMMIX of SAS. Random and repeated variables were cow, and day relative to calving, respectively. Models were adjusted for parity, BCS, evaporating cooling, experiment, and interactions between each disease or disorder and other covariates. P < or equal 0.05 were considered significant. Cows that had dystocia had a tendency to eat more during prepartum (P = 0.09) and ate less on days 11, 12, and 13 postpartum (dystocia x day P = 0.04). Cows that had RP ate less on d -3 and tendency on day -2 (RP x day P = 0.01), and ate less during the postpartum period (P < 0.01). Cows that had MET ate less on d -3, -2, and 1 (MET x day P = 0.03) before calving, and ate less during postpartum period (P < 0.01). Cows that had MAST ate less prepartum from days -5 to -1 and had tendencies on days -6 and -10 (MAST x day P < 0.01), and on d 1, 3, 4, 6, 7, 8, 9, 10, and 13 postpartum with tendencies on d 5, 12, 14 and 15 (MAST x day P<0.01). Cows that had KET ate less during prepartum except on d -8, -10, and -13 (KET x day < 0.01) and during postpartum (P < 0.01). Intake of cows that had DA did not differ prepartum (P = 0.70) but they ate less postpartum (P < 0.01). Cows with at least one disease ate less pre and postpartum (P < 0.01). Collectively, these data suggest that there is an association between DMI pre and postpartum and postpartum diseases. ( en )

▼ The overall objective of my thesis research was to develop a further understanding of how negative energy balance (NEB) relates to the behavior of dairy cows and how to minimize the risk of NEB in early lactation. In a first study, where cows were diet-induced into a state of NEB, it was demonstrated that greater NEB was associated with greater selection for the more energy-dense components of the diet and against the longer-less energy dense fractions of the diet, especially when fed a diet that is easily sorted. In a second study, it was determined that the supplementation of a molasses-based liquid feed through an automated milking system to fresh cows had a tendency to improve cow metabolic health. Cows receiving the additional energy supplement demonstrated improved energy balance and lost less body condition across the first 60 days in milk.

▼ No início da lactação, alterações metabólicas produto do severo balanço energético negativo (BEN) predispõem ao desenvolvimento de lipidose hepática e cetose. O objetivo deste trabalho foi avaliar, através de indicadores bioquímicos, a condição metabólica em vacas leiteiras, durante os primeiros 60 dias de lactação submetidas a três tratamentos preventivos que poderiam melhorar o status energético no início da lactação e por tanto, auxiliar na prevenção de lipidose hepática e cetose. Além disso, buscou-se definir quais dos parâmetros laboratoriais utilizados na avaliação da condição metabólica, seriam mais úteis no direcionamento do diagnóstico e prevenção de transtornos metabólicos por lipomobilização. Cinquenta e quatro vacas multíparas da raça Holandesa foram alocadas em quatro grupos: controle, precursor da glicose (1-2 propaneidol: propileno-glicol: 300 mL/48 horas), protetor hepático (Mercepton:20 mL/48 horas), e suplemento energético (ácidos graxos poli-insaturados protegidos: Megalac-E:250 g/vaca/dia), administrados durante os primeiros 30 dias após o parto. Amostras de sangue e urina foram coletadas nos dias 0, 7, 14, 21, 28, 35, 42 e 49 após o parto, no horário da tarde antes da segunda ordenha, e realizou-se avaliação do escore da condição corporal (ECC) em cada amostragem. Foram coletadas amostras de soro e plasma para análise de ácidos graxos não esterificados (NEFA), albumina, aspartato-aminotransferase (AST), ß-hidroxibutirato (BHB), colesterol, glicose, globulinas, proteína total, triglicerídeos e uréia, através de espectrofotometria utilizando kits diagnósticos. Na urina, determinou-se o pH e corpos cetônicos. No presente trabalho foram considerados pontos de corte para cetose subclínica concentrações séricas de BHB ≥1,4 mmol/L e de NEFA ≥ 0,7 mmol/L. Foi encontrada uma incidência de 24% para cetose subclínica. Os resultados do presente estudo não demonstram o efeito preventivo de nenhum dos três tratamentos, confirmando que até agora não existe tratamento para superar o BEN. Valores séricos de AST, colesterol total e BHB, assim como a determinação de corpos cetônicos na urina são bons parâmetros bioquímicos no diagnóstico e prognóstico de lipomobilização no pós-parto inicial.
In early lactation dairy cattle experience metabolic alterations, caused by severe negative energy balance (NEB). This disbalance predisposes the development of fat liver and ketosis. The aim of this study was to evaluate a metabolic condition by biochemical indicators in high yielding dairy cows (≥25 kg/day) during the first sixty days of lactation. Cows were subjected to three preventive treatments that we hypothesed could improve the energy status in early lactation and therefore aid in the prevention of fat liver and ketosis. ▼ Acetoacetato (AA) e 2-metilacetoacetato (MAA) são compostos β-cetoácidos acumulados em diversas desordens metabólicas como no diabetes e na isoleucinemia, respectivamente. Examinamos o mecanismo de oxidação aeróbica de AA e MAA iniciada por intermediários reativos de mioglobina de coração de cavalo (Mb) gerados pela adição de H2O2. Uma rota quimioluminescente que envolve um intermediário dioxetânico cuja termólise gera espécies α-dicarbonílicas (metilglioxal e biacetilo) foi proposta e estudada. Emissão de luz ultra fraca acompanha a reação, e sua intensidade aumenta linearmente pelo aumento da concentração tanto de Mb (10-500 µM) quando AA (10-100 mM). Estudos de consumo de oxigênio mostraram que MAA é, como esperado, quase uma ordem de grandeza mais reativo que AA. Estudos de EPR com captação de spin, utilizando MNP, possibilitaram detectar adutos de MAA atribuíveis a um radical centrado no Cα (aN = 1.55 mT) e ao radical acetila (aN = 0.83 mT). O sinal do radical acetila é totalmente suprimido por sorbato, um conhecido e eficiente supressor de espécies tripletes, o que é consistente com uma rota reacional envolvendo um intermediário dioxetânico. Clivagem-α da ligação carbonila-carbonila do produto biacetilo triplete produziria, de fato, radicais acetila. Além disso, utilizando AA como substrato para Mb/H2O2, um sinal de EPR atribuível ao aduto MNP-AA• (aN = 1.46 mT e aH = 0.34 mT) foi observado e confirmado por efeito isotópico. O consumo de oxigênio e o rendimento de compostos α-dicarbonílicos foram dose-dependentes à concentração de AA ou MAA (1-50 mM) bem como à concentração de H2O2 adicionado às misturas de reação contendo Mb (até 1:10 quando medido o consumo de oxigênio, e até 1:25 quando medido o rendimento de compostos α-dicarbonílicos) e tert-butilhidroperóxido (até 1:200). Os perfis de pH (5,8-7,8) para consumo de oxigênio e rendimento de compostos α-dicarbonílicos mostraram maiores rendimentos para baixos valores de pH, indicativo de ferrilMb formada no ciclo peroxidático da proteína. Avaliando os níveis de lesão de Mb, os β-cetoácidos diminuíram o nível de desorganização protéica na estrutura secundária e terciária elicitada por H2O2. Ainda, houve maior preservação da estrutura primária da proteína, sendo que MAA protegeu mais em comparação a AA, embora quando utilizado este último composto, foi mostrado que há acetilação dose-dependente de Mb. Acetoacetato aumentou a velocidade de descoramento da hemeproteína, provavelmente por ataque de espécies tripletes geradas no sistema. Músculos de rato, plantar e sóleo, expostos ex vivo a concentrações citotóxicas de glicose oxidase (GOX, gera H2O2 em fluxo), foram protegidas pelos ésteres etílicos AAE e MAAE. Foi detectado biacetilo no meio intracelular em músculos expostos a MAAE e GOX. ▼ O cruzamento com a raÃa Jersey vem sendo utilizado principalmente como alternativa para o aumento da concentraÃÃo de sÃlidos do leite em rebanhos da raÃa Holandesa, sendo a capacidade de produÃÃo destes animais fruto de diversos estudos. Entretanto, ainda existem limitadas informaÃÃes sobre diversos fatores relevantes para a tomada de decisÃo e para o manejo de rebanhos cruzados, tais como resistÃncia a doenÃas e facilidade de parto, sendo esta a motivaÃÃo do presente trabalho, o qual objetivou avaliar a sanidade, imunidade e dificuldade de parto de vacas mestiÃas HolandÃs x Jersey em relaÃÃo a vacas puras da raÃa Holandesa. Os dados foram obtidos de uma propriedade localizada no municÃpio de Carambeà âPR, a qual possui vacas puras da raÃa Holandesa e mestiÃas HolandÃs x Jersey em diferentes proporÃÃes de cada raÃa. Dados de dificuldade de parto, duraÃÃo da gestaÃÃo, retenÃÃo de placenta, indicadores sanguÃneos para doenÃas metabÃlicas pÃs-parto (cetose e paresia puerperal hipocalcÃmica) e de imunidade foram coletados no perÃodo de julho de 2010 a junho de 2011 e os dados de Contagem de CÃlulas SomÃticas (CCS) obtidos do controle leiteiro oficial, no perÃodo de 2005 a 2010. Para facilidade de parto adotou-se uma escala de variando de 1 (partos sem auxÃlio) a 5 (extrema dificuldade, cesariana ou morte) e a retenÃÃo de placenta quando transcorridos 24 horas do parto sem desprendimento dos envoltÃrios fetais. ConcentraÃÃo de β-hidroxibutirato (BHBA) no sangue, como indicador de cetose, cÃlcio total e iÃnico no soro sanguÃneo, como indicador de paresia puerperal hipocalcÃmica, e os indicadores de imunidade Imunoglobulina G (IgG) e proteÃna total no soro, foram determinados a partir de amostras de sangue coletadas no dia do parto, sendo repetido na nona semana apÃs o parto para BHBA. As variÃveis facilidade de parto e retenÃÃo de placenta foram avaliadas atravÃs de um modelo linear generalizado, com distribuiÃÃo binomial (regressÃo logÃstica) As variÃveis duraÃÃo da gestaÃÃo, BHBA, cÃlcio total e iÃnico, IgG e proteÃna total foram avaliadas atravÃs de delineamento experimental inteiramente casualizado, sendo os dados submetidos à anÃlise de variÃncia. Os dados de CCS foram transformados para Escore de CÃlulas SomÃticas (ECS) e submetidos à anÃlise de variÃncia atravÃs de medidas repetidas no tempo. O grupamento genÃtico nÃo afetou a facilidade de parto (P=0,4376), a retenÃÃo de placenta (P=0,7074) e a duraÃÃo da gestaÃÃo (P=0,2812). Vacas mestiÃas apresentaram maiores concentraÃÃes de IgG (1,776 contra1,456 mg/dL) e de proteÃna total (7,019 contra 6,525 mg/dL). Quanto à concentraÃÃo de BHBA, ocorreu diferenÃa somente no dia do parto, com valores mais altos para as vacas mestiÃas (0,580 contra 0,427 mmol/L). Observou-se diferenÃa entre grupamentos genÃticos para concentraÃÃo de cÃlcio iÃnico (P=0,082), com vacas mestiÃas apresentando concentraÃÃo mais baixa (4,3 contra 3,92 mg/dL).
