▼ Chromosomes of six Vernonia species of the family Asteraceae collected from central part of Ethiopia were studied. The chromosomes were prepared from actively growing root tips by conventional air-dry technique and stained in Giemsa stain. Chromosome counts were made from well spread metaphase plates. All the counts are from somatic cells so that the chromosome numbers are 2n numbers. Chromosome counts of 2n = 20 for Vernonia amygdalina, Vernonia adoensis, Vernonia myriantha and Vernonia schimperi and 2n = 18 for Vernonia leopoldii and Vernonia galamensis were recorded. The karyotypes of the species are more of symmetrical with many median, some submedian chromosomes and only one pair of subterminal chromosomes in Vernonia myriantha. Prometaphase chromosome showed chromosomes with satellite (secondary constriction). The numbers of satellited chromosomes observed were one pair in Vernonia myriantha, Vernonia schimperi, Vernonia leopoldii and Vernonia galamensis and two in Vernonia amygdalina and Vernonia adoensis. The results of this study agree with most of the previous studies on the genus.

▼ In this study, live specimens of six rodent species were captured from localities around Addis Ababa by using Sherman live-trap. Somatic metaphase chromosomes were prepared by bone marrow method and the diploid number of chromosomes (2n) and the number of autosomal arms (FNa) were recorded and karyotypes constructed for these species. The karyological results were recorded as follows: Mastomys natalensis (2n = 32 and FNa = 54), Arvicanthis abyssinicus (2n = 62 and FNa = 64), Stenocephalemys albipes (2n = 46 and FNa = 50), Rattus rattus (2n = 38 and FNa = 58), Mus musculus (2n = 40 and FNa = 38) and one unidentified species (designated as ARSI-X) (2n = 32 and FNa =50). These karyological results were compared with previous reports on the same species from different localities of Ethiopia and abroad; and found that they were largely in agreement. The unidentified species, ARSI-X, morphologically looked liked Mastomys species but with distinctly different
karyotype. It was suggested that this specimen, could probably be a sibling species or a new cryptic species in the genus Mastomys, but it requires further detail morphological and cytological studies in order to determine its proper taxonomic status.

▼ Bu çalışmada, Türkiye’de doğal olarak yetişen Onobrychis cinsi Onobrychis
seksiyonuna ait 7 endemik korunga (O. beata, O. cilicica, O. fallax, O. pisidica, O.
podperae, O.sulphurea ve O. lasistanica) türü üzerinde ezme preparat yöntemi
kullanılarak karyolojik çalışmalar yapılmıştır. Kök uçlarının boyanmasında, feulgen ve
hematoxylin-iron boyaları kullanılmıştır. Gözlem yapılan türlerin bir tanesinde (O.
lasistanica) ploidi seviyesi tetraploid (2n=28), diğerlerinde ise diploid (2n=14) olarak
belirlenmiştir. Temel kromozom sayısı ise incelenen tüm türlerde x=7 olarak tespit
edilmiştir. Bununla birlikte, kromozomlar sentromer pozisyonlarına göre median’dan
submedian’a kadar değişmiştir. Gözlem yapılan kromozom özelliklerinden elde edilen
veriler hiyerarşik kümeleme analizi yapılarak türler arasındaki akrabalık ilişkileri
belirlenmiştir. Bu analize göre türler 3 grup altında toplanmıştır. Birinci grupta O.
fallax, O. sulphurea, O. beata ve O. lasistanica türleri, ikinci grupta sadece O. cilicica
türü ve üçüncü gruptada O. pisidica ve O. podperae türleri yer almıştır.

▼ There is a vast diversity of karyotypes in nature, yet mechanisms that have facilitated such diversity are unclear. Alterations to an organism’s
karyotype can have major negative fitness consequences in meiosis through non-disjunction and aneuploidy. Here, I investigated the role of biased segregation in female meiosis, i.e., meiotic drive, as a force that contributes to the evolution of
karyotype form. The closely related species pair, Drosophila americana and Drosophila novamexicana, is an exemplar for understanding mechanisms of
karyotype evolution. Since their recent divergence nearly half a million years ago, D. americana has evolved two different centromeric fusions: one fusion between the 2nd and 3rd chromosomes (Muller elements C and D), and the other fusion between the X and 4th chromosomes (Muller elements A and B). The 2-3 fusion is fixed in D. americana. However, the X-4 centromeric fusion remains polymorphic within the species. I uncovered biased transmissions for both fused chromosomes in D. americana such that the X-4 fused chromosome was inherited by 57% of the offspring from heterozygous females and the 2-3 chromosome was inherited by 62% of the offspring. Introgression experiments shoed the fused X-4 and the unfused X and 4th chromosomes are segregating at a 50/50 ratio in D. novamexicana. I have isolated the fused X-4 centromeric region as a possible player in the observed meiotic drive. However, the centromere is not sufficient to cause meiotic drive without a secondary factor. I also measured heterochromatin content between the fused and unfused X and 4th homologs. No obvious size differences were uncovered, but possible compositional differences were revealed. This suggests that if the centromere itself is involved in meiotic drive, either differences in the number of centromeres or compositional differences between the centromeres are influencing meiotic drive. Overall, I have identified and characterized meiotic drive as a force driving
karyotype evolution in D. americana but appears to be absent in D. novamexicana, and I have begun to dissect the mechanisms of meiotic drive.

▼ Os loricariíneos são Siluriformes altamente derivados e contabilizam cerca de 210 espécies, apresentando uma irradiação adaptativa bem sucedida, tendo representantes na Costa Rica, no Panamá e em toda a América do Sul, e ocorrendo também em ambos os lados dos Andes. Discussões taxonômicas conflituosas são encontradas sobre esta subfamília, envolvendo muitos de seus gêneros, principalmente os mais numerosos, como é o caso de Rineloricaria. O elevado número de espécies (64), a ampla distribuição geográfica e a presença de características diagnósticas válidas para todos os gêneros da subfamília, levam à discussão a proposta de uma subdivisão no gênero Rineloricaria, além de indicar as dificuldades de identificação apresentadas pelo gênero. Em relação às características citogenéticas, Loricariinae apresenta-se como uma subfamília heterogênea e diversificada, com o número diplóide variando de 2n=36 cromossomos a 2n=74 cromossomos. O gênero Rineloricaria contribui intensamente para essa variação, com números cromossômicos variando de 2n=36 a 2n=70 cromossomos. A participação de rearranjos Robertsonianos na evolução cariotípica de Rineloricaria é algo claro, mas evidências que confirmem a ordenação desses eventos não existem. No presente trabalho, técnicas citogenéticas e moleculares foram utilizadas com a finalidade de estabelecer relações evolutivas entre os integrantes da subfamília Loricariinae, em especial do gênero Rineloricaria, entender a evolução cariotípica dessa subfamília e identificar marcadores citogenéticos e moleculares úteis na identificação e descrição de espécies de Rineloricaria. Foram analisados os cromossomos de doze espécies de Loricariinae, pertencentes aos gêneros Harttia, Loricariia, Loricariichthys e Rineloricaria, coletadas na Bacia do Alto Paraná e nas drenagens do Leste brasileiro. Três regiões do genoma mitocondrial (ATPase 6 e 8, Citocromo b e Citocromo c Oxidase I) foram investigadas nessas espécies no presente trabalho. As análises filogenéticas recuperaram o gênero Rineloricaria como um grupo monofilético. Os dados citogenéticos permitiram o estabelecimento de tendências de evolução cromossômica em Loricariinae e no gênero Rineloricaria. O presente trabalho reforça a importância da utilização de diferentes abordagens na realização de estudos taxonômicos e evolutivos, sugerindo uma nova revisão do gênero Rineloricaria que leve em consideração os dados genéticos obtidos.

▼ To enhance the scientific utility and practical application of the honey bee
genome and assign the linkage groups to specific chromosomes, I identified
chromosomes and characterized the
karyotype of the sequenced strain DH4 of the honey
bee. The primary analysis of the
karyotype and ideogram construction was based on
banding and Fluorescence In Situ Hybridization (FISH) for rDNA detection. FISH
confirmed two locations for the NOR on telomeric regions of chromosomes 6 and 12
plus an additional less frequent signal on chromosome 1, all three of which were
confirmed with silver staining (AgNO3). 4?6-diamidino-2phenylindole (DAPI), and CBanding
methods were used to construct the primary ideograms that served as a basis to
further identify the chromosomes and locate important structures. The primary map was
compared with Giemsa banding, AgNO3-banding, Trypsin banding, and R-banding. The
karyotype of the honey bee was established as two metacentric chromosomes (1 and 10),
two submetacentric with ribosomal organizer (6 and 12), four submetacentric
heterochromatic chromosomes (16, 15, 4 and 13), four euchromatic subtelocentric
chromosomes (2, 8, 11 and 14) and four acrocentric chromosomes (3, 5, 7 and 9). In situ
nick-translation banding methods were used to verify the heterochromatin distribution.
The cytogenetic maps of the honey bee
karyotype represented in the ideograms were
subsequently used to place 35 mapped BACs (Solignac et. al. 2004) of Solignac?s BAC
library. As the BACs hybridized to multiple sites, the mapping was based on strength
and frequency of the signals. Location and position of the BACs was compared with those published in the different version of Map Viewer of the NCBI and BeeBase web
sites. 10 BACs were confirmed with the last version of Map Viewer V4, 12 BACs were
mapped based on high frequency and agreement with the earlier version of Map Viewer.
14 BACs were mapped as confirmed based on moderate frequency of the signal and
agreement with the last version of MVV, most of these BACs hits as a secondary signal.

▼ Karyotypes and nucleolar number of four species of the genus Commelina L. and two species of the genus Tradescantia L. (Commelinaceae) which were collected from different localities of central and south-western part of Ethiopia (Addis Ababa, Ginchi, Sebeta, Entoto and Jimma), were analysed. Somatic chromosomes were prepared from root tips that emerged from the nodes of stem cuttings that were made to stand submerged in water. The roots were pretreated in 8-hydroxyquinelin 3-5 hrs followed by fixation in 3:1 ethanol: acetic acid for 1-24 hrs at 4OC. Air-dry slides were prepared following cellulase and pectinase maceration at 37OC, the preparation was stained in Giemsa stain (PH 6.4), rinsed and mounted. Karyotype formula of the six species were found as C. africana (A.A) 2n=2x=30 (12m + 10sm + 8st), C. africana (Sebeta) 2n=2x=30 (14m + 10sm + 6st), C. benghalensis 2n=6x=66 (36m + 24sm + 6st), C. diffusa (Entoto) 2n=2x=30 (16m + 6sm + 8st), C. diffusa (Ginchi) 2n=66 (28m +26sm + 12st), C. diffusa (Jimma) 2n=2x=30 (10m + 8sm + 12st), C. subulata 2n=2x=30 (18m +10sm + 2st), T. zebrina 2n=2x=24 (4m + 6st + 14t), T. fluminensis (green) 2n=60 (8m + 6sm + 22st + 24t) and T. fluminensis (variegated) 2n=60. According to Stebbins
karyotype asymmetry, the karyotypes of C.africana (A.A), C. diffusa (Entoto), C. subulata were 2A type, while that of C. africana (Sebeta), C. benghalensis and C. diffusa (Ginchi) were 2B type. 3A asymmetry type was obtained for C. diffusa (Jimma) and 3B asymmetry type was also found for T. zebrina and T. fluminensis (green). Karotypes of Tradescantia were found to be bi-modal, while it is monomodal for the Commelina species of the studied plant materials. Satellites were observed for species C. africana, C. diffusa and T. zebrina with variation in number ranging from 2 to 6. The maxiumum number of nucleoli observed varies from two to four for Commelina, while, it is three for Tradescantia. This study reported
karyotype and nucleoli of the Ethiopian Commelinaceae for the first time. The current investigation can be considered as an additional
karyotype data to the earlier meiosis report for Ethiopian materials.

Mutacije u genima za izocitrat dehidrogenazu 1 i 2
(IDH1 i IDH2) su česte rekurentne molekularne promene kod akutne
mijeloidne leukemije sa normalnim kariotipom (AMLNK). Uticaj IDH
mutacija na kliničke karakteristike i ishod lečenja kod AML-NK su u
velikoj meri istražene, ali samo nekoliko studija je pratilo ove
mutacije u toku bolesti. Ciljevi: 1) Utvrđivanje učestalosti
prisustva IDH1 i IDH2 mutacija kod bolesnika sa AMLNK; 2)
Utvrđivanje uticaja prisustva IDH1 i IDH2 mutacija na učestalost i
dužinu remisije, ukupno preživljavanje i povezanosti IDH1 i IDH2
mutacija i drugih prognostičkih markera: parametri krvne slike,
biohemijski parametri, citološki tip bolesti, imunofenotipska i
molekularno-genetska obeležja (FLT3 i NPM1 mutacije); 3)
Utvrđivanje stabilnosti IDH1 i IDH2 mutacija u toku bolesti
poređenjem uzoraka na početku bolesti sa uzorcima u toku kompletne
remisije (KR), odnosno u recidivu bolesti. Materijal i metodologija
istraživanja: Uzorci od 110 starijih de novo AML-NK bolesnika bili
su ispitani na IDH1 i IDH2 mutaciju. Ispitana je njihova povezanost
sa drugim prognostičkim markerima i ishodom lečenja. Pored toga,
praćena je stabilnost ovih mutacija u KR i recidivu. Rezultati: IDH
mutacije su nađene kod 25 (23%) bolesnika: IDH1 je imalo 8 (7%)
bolesnika, svi od njih su imali IDHR132; IDH2 je imalo 17 (16%)
bolesnika (15 IDHR140; 2 IDHR172). IDH+ bolesnici su imali viši
broj trombocita (p=0.024), kao i veći procenat blasta u perifernoj
krvi (p=0.031) u odnosu na IDH- bolesnike. IDH+ bolesnici su imali
nižu stopu KR u poređenju sa IDH- bolesnicima (44% vs 60%,
p=0.152), ali to nije bilo statistički značajno. Osim toga, IDH
mutacije su imale samo neznatan uticaj na trajanje remisije bez
bolesti (IDH+-12 meseci vs IDH – 17 meseci; p=0.091), dok je ukupno
preživljavanje (OS) IDH+ bolesnika bilo kraće u poređenju sa IDH-
bolesnicima (2 vs 7 meseci; p=0.039). Nepovoljan uticaj IDH
mutacije na OS je naročito bio očigledan kod bolesnika koji nisu
imali NPM1 mutaciju (5 vs 12 meseci, p=0.050). IDH mutacije su
ispitivane sekvencionalno kod 19 IDH+ bolesnika koji su bili živi
posle indukcije...

▼ The freshwater snail Biomphalaria glabrata is an intermediate host for Schistosoma mansoni parasites, causing one of the most prevalent parasitic infections in mammals, known as schistosomiasis (Bilharzia). Due to its importance in the spread of the disease B. glabrata has been selected for whole genome sequencing and is now a molluscan model organism. In order to aid the sequencing project and to understand the structure and organisation of B. glabrata’s genome at the chromosomal level, a G-banded karyotype has been established. Unlike in any other previous reports, two heteromorphic chromosomes have been identified in the genome of B. glabrata and for the first time snail ideograms have been produced. In addition to characterising the snail chromosomes, a methodology for mapping single copy B. glabrata genes onto these chromosomes has also been established, and 4 genes have successfully been mapped using fluorescence in situ hybridisation. In the relationship between a parasite and a host organism, it is of fundamental importance to understand the basic biology and interfere with the life cycle to reveal how the parasite controls and elicits host gene expression for its own benefit. This study is also directly addressing this aspect of host – parasite interactions by investigating the effects of schistosome infection on the genome and cell nuclei of the host snail B. glabrata. Upon infection with S. mansoni miracidia, genes known to be involved in the host response to the parasite are dramatically relocated within the interphase snail nuclei. These events are in conjunction with the up-regulation of gene expression, indicating a parasite induced nuclear event. Moreover, a differential response between the schistosome-resistant and schistosome-susceptible snails is also reported. This is the first time this has been described in a host – pathogen relationship. The precise organisation of the genome is critical for its correct functioning. The genome is non-randomly organised and this level of organisation is very much influenced by the nuclear architecture. Being a molluscan model organism with the availability of a unique cell line, B. glabrata is a remarkable organism for the studies of nuclear and genome biology. For this reason, in this thesis the snail nuclear architecture was also investigated. For the first time PML bodies, transcription factories, and nuclear myosin 1 beta have been visualised in the snail nuclei. A heat shock system was also developed to study the role of these structures in the snail. Upon heat stimuli gene loci were found to reposition and co-localise with transcription factories, which was in parallel with the up-regulation of gene expression. The mechanism of this genome reorganisation was explored by investigating nuclear motor structures in the snail. seksiyonunda yer alan 2 tanesi endemik olmak üzere 6 farklı korunga türünün (O.
tournefortii, O. galegifolia, O. radiata, O. hypargyrea O. meschetica ve O. albiflora)
kromozom sayıları ve morfolojileri ezme preparasyon metodu ile incelenmiştir. 2 çeşit
boyama metodu kullanılmıştır (Feulgen ve Hematoxylin-iron). O. tournefortii türünde
Feulgen boyama metodu, diğer türlerde ise Hematoxylin-iron boyama metodu
kullanılmıştır. Araştırmada incelenen türlerin tamamında mitotik metafaz kromozom
sayıları, 2n=14 ve temel kromozom sayısı x=7 olarak tespit edilmiştir. Ancak,
kromozomlar sentromer pozisyonlarına göre median’dan submedian’a kadar
değişmiştir. Genel olarak türlerin hepsinde I veya IV numaralı kromozomlar üzerinde
satelit bulunurken, O. meschetica türünde satelite rastlanmamıştır.

sindromo fenotipo ryšį su nustatyta chromosomine konstitucija ir jo
įtaką šeiminei homeostazei. TS paplitimas, dažnis ir ligonių amžius
diagnozavimo metu iki šiol nebuvo aprašytas mūsų tirtoje
populiacijoje. KMU biologijos katedros citogenetikos laboratorijoje
buvo ištirti kariotipai 1271 asmeniui, kuriam galima buvo įtarti
TS: naujagimiai su įgimtomis sklaidos ydomis; mergaitės su fizinio
vystymosi atsilikimais; mergaitės su brendimo atsilikimu; moterys
su pirmine amenorėja; moterys su antrine amenorėja; moterys, tirtos
dėl persileidimų ir nevaisingumo. Kariotipo pakitimai, būdingi TS,
diagnozuoti 236 asmenims (18,6 proc.). Išskirti trys pagrindiniai
kariotipo pakitimų variantai ir įvertinta, ar vienoda jų įtaka
fenotipui. Tirta, ar yra skirtumas šių ligonių grupių fenotipe
pagal tirtuosius parametrus: nėštumo trukmę, naujagimių kūno ilgį
ir svorį, mergaičių ūgį ir svorį iki 18 metų amžiaus, tėvų amžių
gimstant vaikui, sibsų amžių, paciento, kuriam įtariamas TS, amžių
kariotipo tyrimo metu. Ištirta ir aprašyta keletas retų chromosomų
disbalanso variantų. Nustatyta, kad monosominiai ligoniai dažniau
vienintelę X chromosomą paveldi iš motinos (80,5 proc.), negu iš
tėvo. Pirmą kartą atliktas šeiminis daugiaveiksniškai paveldimų
požymių tyrimas (elektrokardiogramos ir gliukozės toleravimo
mėginys) ne tik probandams, sergantiems TS, bet ir jų pirmosios

The aim of this study is to evaluate the
link of chromosome constitution in Turner syndrome and in
disruption of familial homeostasis. The distribution and incidence
of TS in population nor the age of patients at the moment of
diagnosis were not described previously. Cytogenetical laboratory
at the Department of biology has investigated karyotypes for 1271.
The number of abnormal karyotypes found is 236 (18.6%). We divided
the patients into three groups according to logical, in our
opinion, changes in their karyotypes. The aim of our research was
to find out, if there were significant differences in phenotypes
within those groups. Such traits were chosen for the comparison:
the duration of pregnancy, length and weight of newborns, height
and weight of the girls under 18 years old, the age of parents at
birth of propositus, the age of the siblings, and the age at which
TS was diagnosed for the patient. We predicted, that in a case of
complete 45,X monosomy the clinical manifestation of the syndrome
should be more severe, and this group of patients would differ from
other karyological groups. We have found that in 80.5% of cases X
chromosome had the maternal origin and in the rest 19.5% –
paternal. To investigate the multifactorially inherited traits
(electrocardiograms and glucose tolerance test) in relatives of
Turner syndrome patients. We suggest that for the earlier diagnosis
of TS, it is reasonable to investigate all girls, whose height is
less than 3rd percentile.

▼ The dominant paradigm for cancer research focuses on the identification of specific genes for cancer causation and for the discovery of therapeutic targets. Alternatively, the current data emphasize the significance of
karyotype heterogeneity in cancer progression over specific gene-based causes of cancer. Variability of a magnitude significant to shift cell populations from homogeneous diploid cells to a mosaic of structural and numerical chromosome alterations reflects the characteristic low-fidelity genome transfer of cancer cell populations. This transition marks the departure from micro-evolutionary gene-level change to macro-evolutionary change that facilitates the generation of many unique karyotypes within a cell population. Considering cancer cell populations to be complex-adaptive systems, multi-level analyses were performed longitudinally including whole genome microarray, population
karyotype analysis, and determination of cell phenotype. As heterogeneity in ovarian cancer at each of these levels is linked to low survival, metastasis, and resistance to chemotherapy, a syngeinc model of spontaneous ovarian cancer development was employed. The significant findings of the current study are, 1) Genomic instability was apparent from the earliest stages of study, 2) Karyotypic heterogeneity was widespread, showed a pattern of expansion over time and preceded the acquisition of the transformed phenotype 3) a major karyotypic shift occurred between transformed cells in vivo and tumors formed in vitro, documenting the formation of a new system induced by environmental change 4) Chromosome copy number has greater impact on gene expression in early-stage cell populations, where karyotypes are beginning to depart from the diploid genome. A genome-centered paradigm for transformation is emphasized through the discovery of early large-scale increases in
karyotype heterogeneity. This occurred well before the appearance of the transformed phenotype, arose much faster in Brca1 conditionally inactivated cells, was linked to the largest shift in gene expression, and was linked to the transition from in vitro to in vivo survival facilitating tumorigenesis. These data demonstrate the significance, methodologies and rationale for quantifying
karyotype heterogeneity in transformation, tumorigenesis, and clinical cancers. Together, these findings support of a genome-centered evolutionary framework for cancer progression that emphasizes cell-to-cell genomic variability as the basis for macro-evolutionary selection and rapid phenotypic switching in response to new environments.

▼ The impact of cytogenetic findings in AML was
analyzed in the large population-based Swedish AML registry.
Karyotypic patterns differed by age: t(8;21), inv(16) and t(11q23)
were more common in younger patients, whereas loss of 5q, 7q and
17p, monosomal karyotype (MK) and complex karyotype (CK) were more
common in older patients. Patients with ≥5 chromosome abnormalities
had worse overall survival than those with fewer abnormalities or
normal karyotype in all age groups. Loss of 5q, 7q and/or 17p had,
in contrast to MK, a further negative impact on survival.
Multivariable analyses on risk factors in patients <80 years
with cytogenetic abnormalities and intensive treatment revealed
that age and performance status had the most significant impact on
survival (both P<0.001), followed by sex (P=0.0135) and a
karyotype including -7/del(7q) (P=0.048). We compared outcome of
AHD-AML and tAML, i.e., secondary (sAML) with de novo AML. The CR
rates were significantly lower but early death rates similar in
sAML vs de novo AML. In a multivariable analysis, AHD-AML (HR 1.51;
95% CI 1.26–1.79) and tAML (1.72; 1.38–2.15) were independent risk
factors for poor survival. The negative impact of AHD-AML and tAML
on survival was highly age dependent with a considerable impact in
younger patients, but without independent prognostic value in the
elderly. The frequencies of unsuccessful cytogenetics (UC) and
unperformed cytogenetics (UPC) were 2.1% and 2.0%, respectively.
The early death rates differed between the cytogenetic subgroups
(P=0.006) with the highest rates in patients with UC (14%) and UPC
(12%) followed by high-risk (HR) AML, intermediate risk (IR) and
standard risk (SR) cases successfully karyotyped (8.6%, 5.9%, and
5.8%, respectively). The CR rate was lower in UC and UPC and HR
compared with the other risk groups (P<0.001). The 5-year OS
rates were 25% for UC and 22% for UPC, whereas the corresponding
frequencies for SR, IR and HR AML patients without UC and UPC were
64%, 31% and 15%, respectively. Lack of cytogenetic data translates
into a poor prognosis. To ascertain the clinical implications of
high hyperdiploid (HH; 49–65 chromosomes) and triploid/tetraploid
(TT; >65 chromosomes) adult AML diagnosed 1997-2014, and 68
(1.9%) were HH (n=50)/TT (n=18). The OS was similar between
patients with HH/TT and CK AML (median 0.9 years vs. 0.6 years;
P=0.082), whereas OS was significantly longer (median 1.6 years;
P=0.028) for IR AML. The OS was shorter for cases with HH than with
TT (median 0.6 years vs. 1.4 years; P=0.032) and for HH/TT AMLs
with adverse abnormalities (median 0.8 years vs. 1.1 years;
P=0.044). HH/TT AML is associated with a poor outcome, but
chromosome numbers >65 and absence of adverse aberrations seem
to translate into a more favorable prognosis. Also, among 23
patients (0.4 %) with trisomy 13 with a median age of 72 years
(44-84), there was a striking male predominance (80%) with AML-M0
subtype in 37% of patients. Therapy-related AML and MDS/MPN/AML

▼ Jyrsijäharrastus on nykyään kasvussa nuorten keskuudessa Suomessa ja maailmalla. Lajeja, joita pidetään lemmikkeinä on useita. Gerbiilit ovat suosittuja helppohoitoisuutensa vuoksi. Maahamme on tuotu lemmikeiksi gerbiilejä, jotka ovat kotoisin kuivilta aroalueilta Aasiasta ja Afrikasta. Työssä käsitellään kahta gerbiililajia, mongoliangerbiiliä (Meriones unguiculatus) sekä perpallidusta (Gerbillus perpallidus).
Työn tarkoituksena oli vertailla mongoliangerbiiliä ja perpallidusta ulkonäön ja genomin avulla. Tavoitteena oli selvittää lajiutumista ja näiden kahden lajin välisiä sukulaisuussuhteita. Genomitutkimuksessa tarkasteltiin kromosomilukumäärää, kromosomien morfologiaa, heterokromatiinin määrää kromosomeissa sekä heterokromatiinin sijoittumista kromosomeihin. Tarkoitus oli myös varmistaa Suomeen tuotujen perpallidusten lajinmääritys (ovatko perpalliduksia).
Tutkimukseen saatiin kolme eläintä, yksi mongoliangerbiilinaaras sekä perpallidusuros ja -naaras. Eläimistä otettiin verta ja kudosnäytteitä soluviljelyjä varten. Soluviljelyistä saadusta materiaalista tehtiin preparaatteja metafaasien tarkastelua varten. Preparaatit värjättiin heterokromatiini- ja trypsiinivärjäyksillä.
Kaikkien gerbiililajien ulkonäkö on aikojen saatossa muokkautunut hyvin samankaltaiseksi. Syynä tähän on samanlainen kuiva elinympäristö. Myös lemmikkeinä pidettävät mongoliangerbiili ja perpallidus muistuttavat toisiaan paljon monilta osin. Eroavaisuuksia löytyy kuitenkin useita liittyen ruumiin painoon, hännän karvoitukseen, turkin laatuun, silmien kokoon jne. Käyttäytymiseltään mongoliangerbiili ja perpallidus eivät suuresti eroa toisistaan.
Genomitutkimuksessa tulokseksi saamamme diploidiset kromosomiluvut täsmäsivät kirjallisuuden kanssa molemmilla lajeilla (mongoliangerbiilin 2n=44, perpalliduksen 2n=40). Myös kromosomien rakenne vastasi aiemmissa tutkimuksissa saatuja tuloksia. Suuremman diploidisen kromosomiluvun ja suuremman akrosentristen kromosomien lukumäärän perusteella mongoliangerbiiliä voitaisiin mahdollisesti pitää alkukantaisempana kuin perpallidusta. Tämän johtopäätöksen varmistaminen vaatisi kuitenkin jatkotutkimuksia.
Sukupuolikromosomit olivat molemmilla lajeilla suuret, kuten kirjallisuuden perusteella saattoi odottaa. Aiemman tutkimuksen mukaan mongoliangerbiilin ja perpalliduksen X-kromosomin välillä olisi 3 poikkeamaa, joten lajit eivät todennäköisesti ole kovin läheistä sukua keskenään. Heterokromatiinin määrissä oli eroa: mongoliangerbiilillä oli vähän heterokromatiinia autosomeissaan, perpalliduksella oli runsaasti autosomaalista heterokromatiinia. Heterokromatiini oli molemmilla lajeilla pääasiassa sentromeerista. Sukupuolikromosomeissa heterokromatiinia oli molemmilla lajeilla vähän. Eroavaisuudet heterokromatiinissa tukevat oletusta, jonka mukaan lajit eivät olisi kovin lähisukuisia.
Tutkittujen perpallidusten ulkonäön, diploidisen kromosomiluvun ja kromosomien morfologian perusteella voidaan päätellä, että Suomeen tuodut yksilöt ovat Gerbillus perpallidus -lajia. Tiedot täsmäävät kirjallisuuden…

▼ Rearranjos cromossômicos envolvendo a presença de cromossomos marcadores extranumerário são achados citogenéticos freqüentes em pacientes que apresentam deficiência mental, alterações de crescimento, dismorfias e/ou malformações. A presença desse material é responsável por trissomia ou tetrassomia parcial de determinadas regiões cromossômicas, causando quadros clínicos distintos e inespecíficos. A variabilidade fenotípica está relacionada principalmente com os diferentes graus de mosaicismo, os genes presentes na região adicional, o cromossomo de origem, entre outros fatores. Sendo assim, a caracterização desse material cromossômico é de importância fundamental para a determinação do prognóstico e do aconselhamento genético dos pacientes e suas famílias. O presente estudo teve como objetivo a análise de cromossomos marcadores extranumerários por meio de técnicas de citogenética convencional e molecular. Foram selecionados onze pacientes que são acompanhados pelo o Serviço de Genética Médica do Hospital das Clínicas da Faculdade de Medicina de Ribeirão Preto USP, todos com diagnóstico citogenético convencional por bandeamentos GTG de cromossomo marcador extranumerário. Para determinação da origem e caracterização dos cromossomos marcadores foram aplicadas as técnicas de Cariótipo Espectral (SKY) e de Hibridação in situ Fluorescente (FISH). Em dez pacientes foi possível determinar a origem e composição dos marcadores. Dois pacientes apresentam cromossomos marcadores identificados como duplicações invertidas do cromossomo 15, com cariótipos definidos, respectivamente, como 47,XY,+idic(15)(pterq15::q15pter) e 47,XX,+idic(15)(pterq21::q21p11.2), um paciente possui cromossomo marcador derivativo do cromossomo 15, com cariótipo 47,XX,+der(15)(pterq21) e dois pacientes, sendo uma menina e seu pai, possuem cromossomos marcadores derivativos do cromossomo 15 com cariótipos, respectivamente, 48,XX,+2der(15)(pterq12) e 48,XY,+2der(15)(pterq12). Dois pacientes possuem cromossomos marcadores derivativos do cromossomo 9, com cariótipos definidos, respectivamente, como 47,XX,+der(9)(pterq21) e 47,XX,+der(9)(pterq32) e um paciente apresenta cromossomo derivativo do cromossomo 4 [47,XX,+der(4)(p16q21)[9]/48,XX,+der(4)(p16q21),+mar[91]]. Um paciente possui um cromossomo marcador translocado derivativo do cromossomo 22 [47,XY,+der(22)t(11;22)(q25:q11.2)] e outro paciente, um cromossomo translocado derivativo do cromossomo 15 [47,XY,+der(15)t(15;16)(q13;q13)], ambos herdados de mães portadoras de translocações aparente balanceadas. Em um caso, não foi possível a caracterização dos cromossomos marcadores por meio das técnicas aplicadas. Frequentemente, indivíduos com rearranjos equilibrados não apresentam manifestações clínicas, entretanto, na meiose, o pareamento entre cromossomos translocados forma uma figura quadrivalente em forma de cruz que torna a disjunção cromossômica incerta e dependendo do rearranjo, o individuo pode vir a ser infértil, apresentar um risco aumentado de abortamento espontâneo e/ou da prole apresentar alterações fenotípicas. Neste projeto, investigamos duas famílias de pacientes inférteis, portadores de translocações cromossômicas. O objetivo foi caracterizar as alterações citogenéticas e citogenômicas relacionadas à infertilidade masculina em pacientes portadores de rearranjos aparentemente equilibrados, associando técnicas de citogenética clássica (bandeamento GTG), citogenética molecular (FISH) e citogenômica (array-CGH). Foram estudados sete indivíduos da família 1, sendo diagnosticados três portadores da translocação (X;22), sendo um deles azoospérmico. Nesta família foram ainda detectados dois casos de mosaicismo para síndrome de Turner. A família 2 foi composta por dois irmãos oligozoospérmicos, portadores de translocação (8;13). Com a aplicação da técnica de FISH, definimos o cariótipo final dos portadores dos rearranjos como 46,XX ou 46,XY,t(X;22)(p22.3;q11.2) para a família 1 e 46,XY,t(8;13)(q13;q14)para a família 2. A técnica de array-CGH (plataforma 2x400K, Agilent) detectou alterações no número de cópias de alguns genes candidatos relacionados ao fenótipo de infertilidade, sendo a sequência 132 de piRNAs, os genes DDX11, Jagged 2 e ADAM18 na família 1 e os genes candidatos ADAM18 e POTE nos pacientes da família 2.
Reciprocal translocations are the most common balanced rearrangement in humans. Often individuals with balanced rearrangements show no clinical findings. However, in meiosis, the pairing between translocated chromosomes forms a quadrivalent cross-shaped figure which has the effect of making chromosome disjunction uncertain and, depending on the rearrangement, and on the segregation of the unbalanced chromosomes, the individual can be infertile, can present with an increased risk of spontaneous abortions or can have an offspring with abnormal phenotype. We have studied two families of infertile patients, who were carriers of chromosomal translocations. The objective was to characterize the cytogenetic and cytogenomic alterations related to male infertility in patients with apparently balanced rearrangements using classical cytogenetic techniques (GTG banding), molecular cytogenetics (FISH) and cytogenomics (array-CGH). Seven subjects of the family 1 were studied, including three carriers of translocation (X;22), one azoospermic. Two cases of mosaicism for Turner syndrome were detected in this family. ▼ The family Characidae is the largest and most complex among the Characiformes, including the majority of freshwater fish in Brazil, with species varying considerably in size, weight, shape, diet and behavior. Recently, this group was listed as Incertae Sedis due to lack of consistent evidence of monophyly going to have only 12 subfamilies beyond those cited groups above. With the objective of raising information that can be useful to assist in the understanding of this complex ictiofauna, cytogenetic analysis were performed on species of Astyanax aff. fasciatus from the region of Uberlândia-MG, Astyanax sp, Astyanax aff. bimaculatus and Bryconops sp the region of Monte do Carmo-TO. A. fasciatus showed diploide number of 46 chromosomes and a system of multiple Ag-NORs, which proved to positively stained Chromomycin A3 (CMA3). Astyanax sp and Astyanax bimaculatus showed 50 chromosomes, being that in the first one if it verified the presence from 0 to 6 chromosomes B and a system Ag-NORs simple. These small farms if had also shown heterochromatic. Moreover, conspicuous centromeric and pericentromeric heterochromatics markings had been also observed, Astyanax bimaculatus presented system Ag-NORs multiple with up to three pairs marked, some of which also CMA3 +. C-banding revealed centromeric marks in almost all chromosomes. Bryconops sp presented 2n = 48 chromosomes, Ag-NORs simple coinciding with the markings CMA3 and centromeric heterochromatic blocks. These results go of meeting to the idea that the group previously treated as Tetragonopterinae encompasses a diversity to groups sufficiently accented what it is a consequence of its polifiletic nature.
A família Characidae é a maior e mais complexa dentre os Characiformes, compreendendo a maioria dos peixes de água doce do Brasil, com espécies variando consideravelmente em tamanho, peso, forma, alimentação e comportamento. Recentemente, várias espécies deste grupo foram listadas como Incertae Sedis devido a falta de evidências consistentes de monofiletismo. Atualmente essa família é composta por 12 subfamílias, além daqueles grupos citados acima. Com o objetivo de levantar informações que possam ser úteis para auxiliar na compreensão dessa complexa ictiofauna, foram realizadas análises citogenéticas em espécies de Astyanax aff. fasciatus, provenientes da região de Uberlândia-MG, Astyanax sp, Astyanax aff. bimaculatus e Bryconops sp da região de Monte do Carmo-TO. A. fasciatus apresentou número diplóide igual a 46 cromossomos e um sistema de Ag- RONs múltiplas, as quais se mostraram positivamente coradas pela Cromomicina A3 (CMA3). Astyanax sp e Astyanax bimaculatus apresentaram 50 cromossomos, sendo que no primeiro se verificou a presença de 0 a 6 cromossomos B e um sistema de Ag-RONs simples. ▼ CHAPTER II: The Hypostominae subfamily comprises 45 genera, and in only five (Hypostomus, Liposarcus, Rhinielepis, Pogonopoma and Pterygoplichthys) some sort of cytogenetic work has been developed, being Hypostomus genus the largest in number of karyotyped species. Four species of the genus Hypostomus from Araguari and Tijuco rivers, Uberlândia-MG, were analyzed: H. regani, H. paulinus, H. margaritifer e H. strigaticeps, through cytogenetic techniques such as conventional staining, silver nitrate impregnation, C-banding and fluorochromes CMA3 and Hoechst 33258. In the conventional analysis H. regani presented 2n = 72, 10m+18sm+44st/a; H.strigaticeps 2n = 74, 6m+14sm+54st/a; H. margaritifer 2n = 74, 10m+24sm+40st/a, with an individual presenting an unusual heteromorphic metacentric pair on the size, and H. paulinus 2n=76, 6m+12sm+58st/a. The impregnation with silver nitrate showed multiple NORs, for the most species examined, except H. paulinus. The heterochromatin is distributed in the interstitial and telomeric regions. Staining with CMA3 showed positive markings for H. paulinus, H. margaritifer and H. strigaticeps, and negative bands in H. regani, H. margaritifer. Hoechst staining showed positive fluorescent bands in H. regani, H. strigaticeps. The results obtained in this study contribute to a better characterization of Hypostomus cytogenetics and understanding of evolution within the group and the Loricariidae family. H. regani and H. margaritifer. Hoechst staining showed fluorescent bands in H. regani and H. strigaticeps. karyotype of Squaliforma emarginata, which is cited by Artoni et al. (2001) as Hypostomus emarginatus and Pterygoplichthys joselimaianus. Specimens were collected in Porto Nacional -TO, the reservoir formed by damming the waters of the Tocantins River, for the implementation of the Power Plant Luis Eduardo Magalhaes. We performed cytogenetic studies by conventional staining, silver nitrate impregnation, C-banding, fluorochromes, CMA3 and Hoechst. The conventional analysis of S. emarginata showed 2n=52, 14m+2sm+36st and P. joselimaianus 2n=52, 20m+22sm+10st. Impregnation with silver nitrate in S. emarginata showed variation in the pattern of NORs between individuals, while P.joselimaianus showed single NORs. The heterochromatin is distributed in the interstitial and telomeric regions. ▼ Par une stratégie de dépistage combinant le caryotype et l’hybridation in situ en fluorescence (FISH), une insertion (X;6) présente chez des jumelles avec une leucémie myéloïde aiguë (LMA) et une translocation (12;13) dans deux cas de LMA et un cas de leucémie lymphoblastique aiguë (LLA) ont été mis en évidence. L’insertion (X;6) n’est pas rapportée et serait un variant de la translocation (X;6) rapportée dans 4 cas de LMA, dont un associe un gène de fusion MYB-GATA1. Nous avons mis en évidence la dérégulation de l’expression de ces gènes dans le cas d’insertion sans la présence de fusion MYB-GATA1. De plus, dans le premier cas de translocation (12;13) identifié, ETV6 serait fusionné à CDX2 ou FLT3. Le deuxième cas associe la délétion des gènes miR-15a et miR-16-1 à une fusion d’ETV6 et le troisième cas impliquerait une fusion ETV6- FOXO1.

Most male individuals of all species of the genus Scarus which were reversed from females, are called âsecondary malesâ. However, a few individuals of âprimary malesâ would have maleâs reproductive ability, but never process sex reversal in their lifespan, and keeping femaleâs outlook (initial phase). Because parrotfishes have these two kinds of males existing in the same species, they belong to âdiandryâ protogynous hermaphroditisms. Previous studies suggested that the mechanisms that cause primary male might be due to social effect or genetic control. In our experiments, Blue barred parrotfish (Scarus ghobban) and rivulated parrotfish (Scarus rivulatus) were used to study the sex reversal model of parrotfish and the possible reasons that cause the juveniles develop to be primary males. This study including three major parts: (1) the relationship between body size and sexual types of two species. (2) the comparison of the tissue structures of the testes in both male types by histological observation. (3) the possible karyotypes difference, i.e. the genetic differences between primary and secondary male. This study shown that the body size of blue barred parrotfish (initial phase: 100-475 mm; terminal phase: 275-525 mm) were larger than rivulated parrotfish (initial phase: 126-270 mm; terminal phase: 246-350 mm), and there was a wider overlap zone between both color phases in Blue Barred parrotfish (S. ghobban: 47.2%, then S. rivulatus: 10.7%). Histological results showed that no matter primary or secondary males, they all had classical lobular testes. But, by contrast of the pure testis tissue of primary male, there were many mature or atric oocytes that remained in the testis of secondary male. Additionally, a few secondary males of the blue barred parrotfish were discovered whose process of sex reversal occurred earlier or faster than that for normal secondary males. Histological evidence further suggested that these individuals had never had a female reproductive function. Furthermore, these males which had no difference with other secondary males was found on the chromosome level. In our study, a heteromorphic chromosome was observed between primary males and secondary males of the rivulated parrotfish (Scarus rivulatus), which could prove that being a primary male is predetermined by a genetic factor. Based on the ecological interactive diversity of the two species, a preliminary hypothesis was put forth to explain those phenomena. Due to the spawning tactics used, the proportion of primary males in rivulated parrotfish populations is much higher than that in blue-barred parrotfish populations. On the other hand, the appearance of premature males was suggested to be a way of supplying sperm which was lacking in the population of the blue-barred parrotfish.

▼ Dentre os Chiropteros, a família Phyllostomidae constitui o clado mais diversificado do neotrópico e na Amazônia são encontradas cerca de 80 espécies. Do ponto de vista cromossômico, Phyllostomidae destaca-se por apresentar grande variação cariotípica, com números diplóides que vão de 2n=14 em Vampyressa melissa (Stenodermatinae) a 2n=46 em Macrotus waterhousii (Macrotinae). São vários os processos ou mecanismos genéticos que podem resultar em alterações cromossômicas numéricas/estruturais e muitas vezes sequências repetitivas de DNA estão envolvidas neste processo. Visando compreender a variedade e a evolução cariotípica desta família, foram realizadas análises citogenéticas clássicas e moleculares em cromossomos mitóticos de quatro espécies, pertencentes a quatro subfamílias de clados filogenéticos distintos: Artibeus obscurus, Carollia perspicillata, Desmodus rotundus e Phylostomus elongatus. Artibeus obscurus apresentou NF=56 e 2N=30/31, sendo 22m+6st+XX/XY1Y2; C. perspicillata NF=36 e 2N=20/21, sendo 14m-sm+4st+XX/XY1Y2; D. rotundus NF=52 e 2N=28, sendo 26m-sm+XX/XY; P. elongatus NF=60 e 2N=32, sendo 28m-sm+2a+XX/XY. O padrão de distribuição da heterocromatina constitutiva revelou marcação na região pericentromérica em todos os cromossomos das quatro espécies analisadas e variações intraespecíficas foram observadas quando comparados os resultados deste trabalho com o existente na literatura. Em relação à marcação de sítios de DNA ribossomal 18S e regiões organizadoras de nucléolo ativas, em A. obscurus foram evidenciados na região terminal dos pares 5, 6 e 7; em C. perspicillata na região pericentromérica do cromossomo X; em D. rotundus na região centromérica do par 8 e em P. elongatus na região centromérica/terminal do par 15. Marcações teloméricas conspícuas foram visualizadas em D. rotundus e P. elongatus, enquanto que em A. obscurus e C. perspicillata as marcações terminais são tênues. Sítios teloméricos intersticiais foram ausentes em P. elongatus e presente nas demais espécies, podendo ser indicativo de fusões. O retroelemento LINE-1 revelou marcação dispersas, porém em alguns cromossomos são coincidentes com os padrões de bandas escuras evidenciados na banda G, sendo também acumulados no cromossomo X. Se comparada a posição filogenética as espécies estudadas, nota-se que os táxons mais derivados, acumulam maior variação cariotípica, assim como os elementos repetitivos.

▼ The fishes of the order Perciformes are characterized as an important model for understanding the genetic structure of marine populations, because besides they present examples of conservation chromosomal, also they present the karyotype diversification for some groups. Gobiidae family is the most specious in the marine environment. Among its representatives, many species are part of a cryptic fauna little noticed and studied, a wide distribution with behavioral and reproductive characteristics, that make them conducive to the action of biogeographical barriers. Morphologically this family presents reduced body structures through simplification and regressions. Despite their importance in evolutionary inferences, cytogenetics data are incipient facing their species diversity, especially with western Atlantic species. In order to estimate the evolutionary diversity in Gobiidae, it were developed cytogenetic analysis and the standards body, through geometric morphometrics in five species on the Brazilian coast, Coryphopterus glaucofraenum, Bathygobius mystacium, B. soporator, Ctenogobius smaragdus e C. Boleosoma. The data show significant karyotype and morphological diversity among the species. The pericentric inversions and mergers play an important role in chromosomal evolution of this family, causing karyotypic structural and numerical differences in all species. Karyotypic and morphological comparisons among geographic samples of B. soporator from the coast of Maranhão, Rio Grande do Norte and Bahia showed cytogenetics patterns commons, but different morphological patterns. A sample from the Atol das Rocas revealed conspicuous morphological and karyotypic differentiation of another continental populations, confirming the presence of a new island species. The approaches done reveal diversification consistent with characteristics of a group of low vagile and largely able to environmental selection due from peculiar ecological requirements
Os peixes da ordem Perciformes caracterizam-se como um modelo importante para o entendimento da estrutura genética de populações marinhas, pois apresentam tanto exemplos de conservação cromossômica como de diversificação cariotípica para alguns grupos. Gobiidae é a família mais especiosa no ambiente marinho. Entre seus representantes, muitas espécies fazem parte de uma fauna críptica pouco percebida e estudada, apresentando ampla distribuição com características comportamentais e reprodutivas que as tornam propícias à ação de barreiras biogeográficas. Morfologicamente esta família apresenta estruturas corporais reduzidas através de simplificações e regressões. Dados citogenéticos, apesar da importância em inferências evolutivas, são incipientes frente sua diversidade de espécies, sobretudo para espécies do Atlântico Ocidental. ▼ O gênero Crotalaria possui aproximadamente 600 espécies descritas, localizadas principalmente nas regiões tropicais e subtropicais. Estas espécies estão classificadas em oito seções e nove subseções botânicas definidas principalmente com base em caracteres florais. Estas seções botânicas podem ser subdivididas em dois grupos principais: um de espécies com flores especializadas e outro de espécies sem especialização das flores. Estudos citogenéticos anteriores mostraram que as características cromossômicas são conservadas dentro de uma mesma seção ou subseção botânica e foi proposto que as espécies com especialização das flores teriam uma organização cromossômica diferente das espécies sem especialização das flores. Dentro deste contexto, foram descritos os cariótipos e a organização do genoma de três espécies e duas sinonímias, da seção Hedriocarpae, subseção Macrostachyae, com a utilização de coloração convencional dos cromossomos pelo método de Feulgen, de coloração com fluorocromos específicos às regiões cromossômicas ricas em GC ou AT e mapeamento físico por hibridação molecular in situ fluorescente (FISH) dos locos de DNA ribossômicos 45S e 5S. A obtenção de marcadores cromossômicos característicos para esta subseção e a construção de mapas citogenéticos possibilitaram a identificação de uma inversão pericêntrica, de eventos de transposição e da diferenciação na natureza das seqüências de DNA da heterocromatina pericentromérica, previamente descrita em espécies com flores especializadas como ricas em GC. Estes resultados revelaram alguns dos eventos citogenéticos ocorridos durante a diversificação do gênero, sendo que a subseção Macrostachyae é caracterizada por uma inversão no cromossomo 1 que pode ter uma origem antiga no gênero e que espécies sem especialização das flores, de fato possuem uma organização cromossômica distinta das espécies com flores especializadas, principalmente quanto à natureza das seqüências de DNA da heterocromatina pericentromérica.
Approximately 600 species of the Genus Crotalaria have been described, the majority is located in tropical and subtropical regions. These species are classified into eight botanical sections and nine subsections according to floral characteristics. The botanical sections can be divided into two major groups: one with species presenting flower specialization and the other without flower specialization. Previous cytogenetic studies showed that chromosomal features are conserved within the same botanical section or subsection, and it has been suggested that species with flower specialization have a different chromosomal organization from those species without flower specialization. In this context, the karyotypes and genome organization of five species (being three synonymy from the Hedriocarpae section, Macrostachyae subsection) were described by conventional chromosome staining using Feulgen, specific fluorochrome staining to GC or AT chromosomal rich regions and physical mapping by fluorescent in situ molecular hybridization of 45S and 5S ribosomal genes.…

▼ Com o intuito de contribuir para a compreensão da evolução cariotípica em mamíferos, realizamos estudos comparativos, utilizando a pintura cromossômica, em dois grupos basais de mamíferos, as preguiças e os marsupiais. Realizamos comparações entre os cromossomos humanos e os cromossomos das preguiças de três dedos Bradypus torquatus e Bradypus variegatus, estabelecendo as homologias. A análise conjunta de nossos dados e daqueles da literatura sobre pintura cromossômica em outras espécies de Xenarthra permitiu identificar ou confirmar sinapomorfias cromossômicas dos grupos assim como características ancestrais. Também realizamos comparações entre os cromossomos X das duas espécies de preguiça e entre os cromossomos X dos marsupiais americanos Marmosops incanus e Metachirus nudicaudatus. Os principais resultados e conclusões estão resumidos a seguir. 1. Os cariótipos de B. torquatus e B. variegatus são semelhantes quanto à correspondência com os cromossomos humanos. As duas espécies apresentaram em comum (a) a presença das associações dos cromossomos humanos (HSA) 4/8, 7/10, 7/16, 12/22, 14/15 e 17/19, (b) a conservação de HSA 5, 6, 9, 11, 13, 14, 15, 17, 18, 20 e X, (c) dois pares compartilhando homologia com HSA 2, 7, 10, 12, 19 e 22, (d) três pares, com segmentos homólogos a HSA 8 e (e) a ausência da associação ancestral de Eutheria HSA 16/19. 2. O cariótipo de B. variegatus (2n=54) é mais rearranjado em relação ao humano do que o de B. torquatus (2n=50), principalmente devido a fissões de cromossomos ancestrais, que levaram ao maior número diplóide dessa espécie. 3. Reunindo os dados para as preguiças B. variegatus e B. torquatus aos das demais espécies de Xenarthra que tiveram estabelecidas as correlações entre seus cromossomos e os cromossomos humanos, confirmamos como características presentes em todas as espécies dessa supraordem (a) a conservação de HSA 9, 13, 17, 18, 20 e X, (b) a presença de dois pares cromossômicos compartilhando homologia com HSA 19 e 22 e (c) a presença das associações HSA 4/8, 7/16, 12/22 e 14/15. 4. Confirmamos a associação HSA 7/10 e a divisão de HSA 8 em três blocos como assinaturas cromossômicas da supraordem Xenarthra, o que concorda com a monofilia do grupo. 5. Mostramos que a associação HSA 17/19, presente nos cariótipos de B. variegatus, B. torquatus e B. tridactylus, parece ser assinatura cromossômica do gênero Bradypus, apoiando a monofilia do grupo. 6. Mostramos que a associação HSA 12/22/16 parece ser uma sinapomorfia cromossômica, unindo as espécies B. variegatus e B. tridactylus. 7. Considerando a correspondência com os cromossomos humanos, verificamos que os cariótipos de B. variegatus e B. tridactylus são os mais semelhantes, no gênero Bradypus. 8. A análise das correspondências entre as sequências dos cromossomos humanos e as sequências dos cromossomos de grupos externos de mamíferos placentários (marsupial e galinha) disponíveis no banco de dados Ensembl, mostrou que a associação HSA 7/10 presente na supraordem Xenarthra também ocorre nesses grupos externos.…

▼ The fishes of the order Perciformes are characterized as an important model for
understanding the genetic structure of marine populations, because besides they
present examples of conservation chromosomal, also they present the
karyotype
diversification for some groups. Gobiidae family is the most specious in the marine
environment. Among its representatives, many species are part of a cryptic fauna
little noticed and studied, a wide distribution with behavioral and reproductive
characteristics, that make them conducive to the action of biogeographical barriers.
Morphologically this family presents reduced body structures through simplification
and regressions. Despite their importance in evolutionary inferences, cytogenetics
data are incipient facing their species diversity, especially with western Atlantic
species. In order to estimate the evolutionary diversity in Gobiidae, it were developed
cytogenetic analysis and the standards body, through geometric morphometrics in
five species on the Brazilian coast, Coryphopterus glaucofraenum, Bathygobius
mystacium, B. soporator, Ctenogobius smaragdus e C. Boleosoma. The data show
significant
karyotype and morphological diversity among the species. The pericentric
inversions and mergers play an important role in chromosomal evolution of this
family, causing karyotypic structural and numerical differences in all species.
Karyotypic and morphological comparisons among geographic samples of B.
soporator from the coast of Maranhão, Rio Grande do Norte and Bahia showed
cytogenetics patterns commons, but different morphological patterns. A sample from
the Atol das Rocas revealed conspicuous morphological and karyotypic
differentiation of another continental populations, confirming the presence of a new
island species. The approaches done reveal diversification consistent with
characteristics of a group of low vagile and largely able to environmental selection
due from peculiar ecological requirements
