A Amaz?nia interpretada por Arnaldo Rebello no conjunto de sua obra ? parte importante da produ??o musical da regi?o norte do Brasil. As ocorr?ncias cient?fico-interpretativas da m?sica na Amaz?nia s?o ainda uma car?ncia significativa do ponto de vista da hist?ria da m?sica. Com intuito de identificar as ocorr?ncias de Amaz?nia na obra de Arnaldo Rebello, procedemos a cataloga??o da produ??o composicional para piano, elencamos as can??es, e localizamos as atua??es deste compositor como pianista, professor, divulgados da m?sica brasileira. A compreens?o do estado do campo e do habitus no qual interagiu o compositor foi a base te?rico-metodol?gica para o desvelamento das redes de op??es composicionais e concep??es amaz?nicas constru?das a partir do social. Das m?ltiplas interpreta??es de Amaz?nia analisamos o repert?rio pian?stico, localizando sonoridades, pessoas e paisagens no qual o compositor foi influenciado. Com tamb?m as influ?ncias do cen?rio musical ? ?poca composto especialmente pelas bandas de metais de Manaus do in?cio do s?culo XX e da intensa atividade pian?stica coeva. Conclu?mos que essa Amaz?nia sinest?sica ? muito presente neste repert?rio, as sonoridades, cores, sabores e sons, bem como as rela??es de sociabilidades de Arnaldo Rebello, presentes nos t?tulos, nas dedicat?rias, e especialmente nas sonoridades que s?o apresentadas, bem como a sua coloca??o no campo musical e nas variadas inst?ncias de valida??o.

The Amazon played by Arnaldo Rebello in his body of work is an important part of the musical production of northern Brazil. The scientific and interpretive occurrences music on Amazon are still a significant lack from the point of view of music history. In order to identify instances of Amazon in the work of Arnaldo Rebello, we proceeded to catalog the compositional output for piano, the songs list and locate the performances of this composer as pianist, teacher, published in Brazilian music. Understanding of the state of the field and habitus in which interacted the composer was the theoretical and methodological basis for the unveiling the options of compositional networks and Amazonian conceptions built up from the social. Of multiple interpretations of Amazon we analyze the pianistic repertoire, localizing sounds, people and landscapes in which the composer was influenced. With also the influences of the music scene at the time composed especially by metal bands from Manaus to the early 20th century and the intense activity old-fashioned compared piano. We conclude that synaesthetic Amazon is very present in this repertoire the sonorities, colors, flavors and sounds, as well as relations of Arnaldo Rebello sociabilities, present in the titles, in dedications, and especially in sonorities that are presented, as well as its placement in the musical field and in several instances of validation.

A (+)-rotundifolona ? uma cetona monoterpen?idica derivada da piperitenona. ? encontrada geralmente em esp?cies do g?nero Mentha e possui diversas atividades biol?gicas comprovadas, entretanto com resultados divergentes quanto a sua citotoxicidade. Essa mol?cula foi isolada do ?leo essencial das folhas de Lippia pedunculosa com alto rendimento. A sua presen?a e atividades biol?gicas s?o relatadas em diversos estudos, mas pouco tem sido publicado sobre a mesma na ?rea de s?ntese org?nica. Este trabalho visa realizar semissintese de derivados da (+)-rotundifolona e de alguns an?logos ((R)-(-)-carvona, (-)-mentona e (R)- (+)-pulegona). Os constituintes do ?leo e os produtos obtidos foram separados por CCDP e analisados por espectrometrias de RMN e de massas. A (+)-rotundifolona foi identificada como constituinte majorit?rio do ?leo essencial e apresentou um [?]20 ? = + 171,4. As rea??es foram realizadas com a (+)-rotundifolona e seus an?logos comerciais, (R)-(+)-pulegona, (R)-(- )-carvona e (-)-mentona. A redu??o com NaBH4gerou os ?lcoois corespondentes, pulegol e isopulegol, carveol e mentol. A (+)-rotundifolona foi reduzida ao rotundifolol um novo composto que o anel ep?xi ? mantido. A broma??o al?lica com NBS mostrou prefer?ncia a posi??o met?lica em rela??o ? substitui??o no anel, gerando assim os derivados substitu?dos nas posi??es 9 e 10, para a bromocarvona, 8 e 9, para a bromopulegona, 8 e 9, para a bromorotundifolona. Foram sintetizadas tr?s benzaldoximas a partir dos benzalde?dos correspondentes e dois compostos izosazol?nicos in?ditos a partir da cicloadi??o dessas ?ltimas com a (+)-rotundifolona.

Rotundifolone is a monoterpenoid ketone derived from piperitenone. It is generally found in species of the genus Mentha and has several proven biological activities, however with divergent results regarding its cytotoxicity. This molecule has been isolated from the essential oil of Lippia pedunculosa leaves with high yield. Its presence and activities are reported in several studies but little has been published about it in the organic synthesis area. This work aims to perform semisynthesis of rotundifolone derivatives and some analogues ((R)-(-)- carvone, (-)-mentone and (R)-(+)-pulegone). The oil constituents and products obtained were separated by CCDP and analyzed by NMR and mass spectrometry. Rotundifolone is identificate as a major compound of essential oil and its presents a [?]20 ? = + 171,4. The reactions were carried out with rotundifolone and its commercial analogues, (R)- (+)- pulegone, (R)-(-)-carvone and (-)-mentone. The reduction with NaBH4 generated the alcohols pulegol and isopulegol, carveol and menthol. Rotundifolone was reduced to rotundifolol a new compound that the epoxy ring is retained. Allyl bromination preferably showed the methyl position over ring substitution, thus generating the derivatives replaced in positions 9 and 10 for bromocarvone, 8 and 9, for bromopulegone, 8 and 9, for bromorotundifolone. A presente pesquisa ? um estudo sobre a atua??o do PIME (Pontif?cio Instituto das miss?es estrangeiras) em Parintins entre os anos de 1955-1980, o recorte temporal corresponde ? instala??o da Prelazia at? ? eleva??o ? categoria de Diocese, esse per?odo se caracteriza pelo grande fervor mission?rio e por iniciativas desses religiosos no campo social e religioso o que pode ser percebido nas in?meras obras por eles realizado. ? a partir do trabalho desses mission?rios que a Igreja cat?lica ganhar? envergadura n?o somente em Parintins como em outras localidades pertencentes ? Prelazia, esses desbravadores evangelizavam preocupados n?o s? com a quest?o religiosa, mas tamb?m com a social. Dessa forma, esse estudo tem como proposta analisar a atua??o do PIME em Parintins-AM. Para isso, procuramos entender como se deu o processo de evangeliza??o e quais os instrumentos utilizados pelos mission?rios na sua miss?o, tamb?m procuramos analisar o trabalho social do PIME como fator de fortalecimento do catolicismo em Parintins, sendo esse o objetivo geral da pesquisa. This research is a study about the performance of the PIME (Pontifical Institute of Foreign Missions) in Parintins between 1955-1980, the temporal cut corresponds to the installation of the Prelature until the elevation to the category of Diocese, this period is characterized by the great missionary fervor and the initiatives of these religious in the social and religious field, which can be seen in the many works they have done. It is from the work of these missionaries that the Catholic Church will gain a foothold in the city and the Middle Amazon, these Pathfinders evangelized concerned not only with the religious issue, but also with the social. Thus, this study aims to analyze the performance of PIME in the city of Parintins-AM. To this end, we seek to understand how the evangelization process took place and which instruments the missionaries used in their mission. We also sought to analyze the social work of PIME as a factor in strengthening Catholicism in Parintins, which is the general objective of this work. To meet this objective we will seek to: a) address the history of the Catholic Church in Parintins from the Jesuits to the PIME; b) to analyze the social and religious practices of PIME in the Parintins Prelature; c) identify through the narrative of the laity the meanings that the residents attribute to the work of PIME. ▼ The purpose of this dissertation is to analyse the institute of corporate criminal liability. The introduction of a rule by which not only natural but also legal persons (who have achieved or could have achieved unlawful material gain from an illegal act) could be punished by criminal sanctions is an important change in legal systems based on the continental European tradition. The legislation of most European countries (with the exception of some, such as Germany and Italy, which still oppose corporate criminal liability on principle) have been reformed (in the second half of the 20th century and especially in the last 30 years) in order to include norms of corporate criminal liability. The fact that a large number of states have decided to start punishing corporations with criminal sanctions is the result of an objective examination of social reality, which has unequivocally shown that serious crimes are continually committed on behalf of and for the gain of legal persons, and that those acts inflict serious damage to states, society and individuals. The problem of corporate crime and the legal response to it has become a global and regional issue, which has led to the adoption of many conventions and other legal documents that commit or recommend the signatory states to introduce efficient sanctions for corporate crimes. Serbian legislation introduced corporate criminal liability in late 2008, after it was adopted in most European legislations, including all former Yugoslav states. Nonetheless, four years after the adoption of the Corporate Criminal Liability Act, not a single legal proceeding against a legal person charged with committing a criminal act has been conducted. Bearing this in mind, a serious issue arises: is our legal system capable of providing an adequate response to corporate crime challenges? The problem described above could be divided into a series of questions and dilemmas (which range from questions of principle to almost purely practical matters) which demand academic examination and explication. The main objective of this dissertation is to provide a serious and academic analysis of corporate criminal liability in Serbian legislation, to highlight its advantages and disadvantages, and also to consider the extent to which it has been harmonized with international recommendations, comparative legislations, and leading theoretical approaches. Having this objective in mind, the introductory part of the dissertation analyses the criminological dimension of the phenomenon in order to show the extent of the social danger that it entails. The author also provides arguments in favour of and against punishing corporations with criminal sanctions, bearing the criminological facts in mind. The author then discusses different theoretical approaches and models of corporate criminal liability, considering the validity of their arguments and their practical applicability. Isso permite encontrarmos condi??es iniciais para recuperar uma superf?cie a partir de um sistema de equa??es diferenciais parciais.

In this work we studied geometric properties of surfaces with regular vertical projection and constant mean curvature in H^2xR. For the special case in which the constant mean curvature is 1/2, a certain map is constructed on the hyperbolic plane H², called Hyperbolic Gauss Map and the harmonicity of it is subsequently obtained. Another key point is that under certain conditions imposed on the surface, it is always possible from a given harmonic map G to recover a mean curvature surface H=1/2, such that G is its Hyperbolic Gauss Map and whose parameterization is given in terms of G. These results were obtained by Isabel Fern?ndez and Pablo Mira in "Harmonic Maps and Constant Mean Curvature Surfaces in H^2xR". The demonstrations of these results make use of conformal parameters and the use of techniques known in the theory of constant mean curvature surfaces. This allows you to find initial conditions for recovering a surface from a system of partial differential equations.

▼ Svest je u poslednje tri decenije postala predmetom multidisciplinarnih studija svesti (consciousness studies), što je aktualizovalo značaj konceptualne analize pojma svesti i problema koji se javljaju u različitim istraţivačkim pristupima svesti. Da li različite discipline (pa čak i različiti pristupi u okviru iste discipline) govore uopšte o istom predmetu? Na šta se, na primer, odnose savremene neurološke teorije svesti, kakav teorijski status ima koncept svesti u okviru funkcionalnih dijagrama kognitivne psihologije, o čemu govore psihoterapeuti kada naglašavaju značaj svesnosti u psihoterapijskom procesu ili značaj osvešćivanja nesvesnog, šta imaju u vidu fenomenolozi ili praktičari kontemplativnih tradicija kada se bave neposrednim iskustvom i na kraju, šta pod svešću podrazumevaju različite ontološke pozicije o odnosu duha i tela? Konceptualna analiza koja je sprovedena u ovom radu ima za cilj poboljšanje komunikacije izmeĎu disciplina i lokalizaciju teškoća koje stoje na putu interdisciplinarnim istraţivačkim projektima. Analitička filozofija duha (philosophy of mind) je u poslednje četiri decenije izoštrila i locirala problematiku svesti, te razvila novi konceptualni aparat i misaone eksperimente koji omogućavaju preciznije razumevanje ograničenja u pokušaju prirodnonaučne rekonstukcije pojma duha (mind), kakvu su poslednjih decenija, inače sa velikim uspehom, preduzele tzv. mind sciences (poput kognitivne psihologije, veštačke inteligencije, lingvistike, neuronauke) . U radu autorka pokazuje kako različiti savremeni filozofi ukazuju na distinkcije dva lica svesti i duha – Dţekendofovo razlikovanje računarskog duha (computational mind) i fenomenološkog duha (phenomenological mind), Blokovo razlikovanje A-svesti (access consciousness) i P-svesti (phenomenal consciousness) i Čalmersovo razlikovanje „lakog“ i „teškog“ problema svesti. Dok se, na primer, A-svest, odnosi na svest u smislu pristupačnosti, odnosno dostupnosti odreĎenog sadrţaja (mentalne reprezentacije) u kontroli govora, rezonovanja i akcije, P-svest (fenomenalna svest) odnosi se na činjenicu da svesna mentalna stanja poseduju doţivljajne, kvalitativne odlike, koje su dostupne direktno jedino iz subjektivne perspektive prvog lica. Za ovaj aspekat svesti je filozofija duha uvela tehnički termin qualia (lat. sing. quale) i on čini svest drugačijom od ostalih predmeta istraţivanja u dominantnoj objektivističkoj naučnoj paradigmi. Listeria monocytogenes est une bactérie pathogène responsable de la listériose chez l’homme. Cette bactérie est capable d’envahir et de se répliquer dans des cellules phagocytaires et non-phagocytaires de l’hôte. De fait, Listeria interagit et interfère avec de nombreux composants cellulaires au cours de sa réplication. Mon projet de thèse s’est focalisé sur les altérations du protéome des cellules infectées par Listeria. Je me suis en particulier intéressé à l’activation de protéases de l’hôte et aux dégradations de certains facteurs cellulaires en réponse à l’infection.Une analyse protéomique de cellules humaines traitées avec la Listeriolysine O (LLO), une toxine secrétée par Listeria formant des pores dans les membranes cellulaires, nous a permis d’identifier plusieurs dizaines de facteurs cellulaires dégradés en réponse à l’exposition à cette toxine.Nous avons pu confirmer la dégradation de ces protéines dans le contexte d’une infection par Listeria in vitro (dans un modèle de cellules en culture) et in vivo (dans un modèle animal). Nous anticipons que la dégradation de ces protéines, impliquées dans différentes fonctions cellulaires, joue un rôle au cours de l’infection par Listeria en modifiant la physiologie des cellules hôte.Nous avons en parallèle mis en évidence que Listeria interfère avec le fonctionnement des lysosomes. Nous avons montré que la LLO déstabilise les lysosomes des cellules hôtes et induit leur perméabilisation. Ceci permet la libération de certaines protéases lysosomales, telles que les cathepsines, dans le cytosol des cellules hôte. Ces cathepsines, une fois libérées, pourraient alors cliver différentes protéines cytosoliques et ainsi modifier la physiologie des cellules infectées.L’ensemble de mes travaux a permis de montrer que Listeria interfère avec le fonctionnement de nombreuses protéases de l’hôte et induit la dégradation de plusieurs facteurs cellulaires. Ces cibles dégradées constituent une nouvelle classe de protéines dont nous pouvons maintenant tester le rôle potentiel dans l’infection. Mes travaux ont d’autre part permis de montrer que d’autres toxines bactériennes, similaires à la LLO, induisent également des altérations du protéome des cellules hôtes. Ceci suggère que les mécanismes identifiés avec Listeria sont conservés entre différentes classes de pathogènes bactériens.

Listeria monocytogenes is a foodborne pathogen responsible for human listeriosis which invade and replicate in both phagocytic and non-phagocytic cells. Listeria intracellular life cycle involves interference with host cell components. My PhD project aims to characterize host cell proteome modifications in cells infected by Listeria. I focused my research on host proteases activation and the degradation of host cell proteins in response to infection. Les cellules humaines sont soumises à des stress induisant des cassures double-brin de l’ADN principalement réparées par la voie NHEJ, où la kinase DNA-PKcs joue un rôle central. L’activité de DNA-PKcs, régulée par de nombreuses phosphorylations, est cruciale pour le maintien de l’intégrité génomique. Plus récemment, il a été montré que cette protéine était également modifiée par l’O-GlcNAcylation dans la lignée COS7. Sachant l’équilibre existant entre phosphorylation et O-GlcNAcylation, nous avons étudié le rôle de cette nouvelle MPT dans la régulation de l’activité de DNA- PKcs. Nous avons montré que DNA-PKcs est O-GlcNAcylée dans les cellules HeLa. Puis nous avons montré que la modulation de l’O-GlcNAcylation de DNA-PKcs impacte son autophosphorylation en Ser2056, suggérant l’existence d’une balance O-GlcNAcylation /phosphorylation, ainsi que la capacité des cellules à réparer les DSBs par la voie NHEJ. De plus, nos résultats nous laissent envisager que cette modification puisse jouer un rôle dans la stabilité de la protéine. DNA-PKcs est une cible potentielle dans les stratégies de lutte contre le cancer. Nous avons étudié l’impact d’un composé sur DNA-PKcs. Cette molécule provoque une réduction de la quantité et de l’activité de DNA-PKcs, impliquant son ubiquitinylation et sa dégradation par le protéasome et menant à une sensibilisation des cellules à un traitement génotoxique. Dans ce contexte, nous avons développé une puce à anticorps pour évaluer le profil phosphoprotéique des voies de réparation de l’ADN et ainsi évaluer l’effet d’inhibiteurs de DNA-PKcs. L’ensemble de ces résultats contribuent à une meilleure compréhension de la régulation de DNA-PKcs.

Human cells are subjected to stresses inducing DNA double-strand breaks mainly repaired by the NHEJ pathway, where the kinase DNA-PKcs plays a central role. The activity of DNA-PKcs, regulated by numerous phosphorylations, is crucial for the maintenance of genomic integrity. More recently, it has been shown that this protein is also modified by O-GlcNAcylation in the COS7 cell line. Knowing the balance between phosphorylation and O-GlcNAcylation, we studied the role of this new PTM in the regulation of DNA-PKcs activity. We have shown that DNA-PKcs is O-GlcNAcylated in HeLa cells. We then showed that the modulation of DNA-PKcs O-GlcNAcylation affects its autophosphorylation on Ser2056, suggesting an O-GlcNAcylation/phosphorylation balance, as well as the ability of cells to repair DSBs by NHEJ pathway. Moreover, our results allow us to consider that this modification may play a role in protein stability. DNA-PKcs is a potential target in anticancer strategies. We studied the impact of a chemical compound on DNA-PKcs activity. This molecule causes a reduction in the amount and activity of DNA-PKcs, through its ubiquitinylation and its degradation by the proteasome and leading to sensitization of the cells to genotoxic treatment. L'insuffisance cardiaque (IC) touche 1-2% des adultes dans les pays développés. On distingue classiquement 2 formes d’IC : l’IC chronique, dont la mise en place est lente, et l’IC aigue dont l’apparition est rapide ou due à l'aggravation des symptômes et/ou des signes de l’IC. Quelles soient chroniques ou aigues, certaines formes d'IC n'ont pas de traitement efficace. L'insuffisance cardiaque à fraction d'éjection préservée (ICFEp) devient la forme prédominante d'IC chronique mais la compréhension de sa physiopathologie reste encore incomplète. Un modèle de rat transgénique sur-exprimant le récepteur β3- adrénergique humain au niveau endothélial a été développé dans l'équipe. Comme les patients, ce modèle développe une ICFEp avec l’âge qui se manifeste notamment par une dysfonction diastolique et une intolérance au stress. Dans le cadre de ma thèse et sur la base d'une étude transcriptomique, j'ai évalué les altérations du cycle du calcium, du métabolisme ainsi que l'apparition de fibrose et d'inflammation. Le choc septique est considéré comme une IC aigue particulièrement violente. Le deuxième axe de ma thèse a été de développer des modèles pertinents de choc septique et d’évaluer le potentiel thérapeutique d’une modification post-traductionnelle : la O-GlcNAcylation, déjà connue pour ses effets bénéfiques dans des formes d’IC aigues. Ainsi, une augmentation des niveaux de l'O-GlcNAc, grâce à un agent pharmacologique, le NButGT, améliore la fonction cardiovasculaire et la survie des animaux en choc septique.

Heart failure (HF) affects 1-2% of adults in developed countries. There are two distinct forms of HF: chronic HF, with a slow development, and acute HF that appears rapidly or is due to an aggravation of the symptoms and/or signs of HF. For some forms of HF, acute or chronic, no effective treatment exists. Heart failure with preserved ejection fraction (HFpEF) is becoming the predominant form of chronic HF but the understanding of its pathophysiology remains incomplete. A transgenic rat model over-expressing the human endothelial β3-adrenergic receptor has been developed in the team. As in patients, this model develops HFpEF with age, and is characterized by diastolic dysfunction and stress intolerance. During my thesis and based on transcriptome data, I evaluated alterations of the calcium cycle, metabolism as well as the appearance of fibrosis and inflammation. Septic shock is considered to be a form of severe acute HF. The second axis of my thesis was to develop relevant models of septic shock and to evaluate the therapeutic potential of a post-translational modification: O-GlcNAcylation, already known for its beneficial effects in acute HF forms. Indeed, an increase in levels of O-GlcNAc, through a pharmacological agent, NButGT, improved cardiovascular function and survival of animals in septic shock.

Työhön kuuluu myös sähkötyöturvallisuuden huomioiminen tehdaskoestuksessa ja käyttöönotossa. Sähkötyöturvallisuuden varmistaminen huomioitiin testausohjeissa, ja sähkötyöturvallisuutta käsitellään raporttiosiossa. EU:n konedirektiivin ja siihen liittyvien standardien vaatimukset ovat myös osa tätä työtä. The topic of this thesis is Testing manuals for I/O interfaces and devices connected to fieldbus. Thesis was produced for Valmet Automation. Hardware of automation systems delivered by Valmet consists of I/O interfaces, I/O devices and devices connected to fieldbus. The purpose for testing manuals is to serve as universally comprehensive instructions for typical projects delivered by Valmet. Thorough testing of the hardware is essential part of the project delivery process of Valmet. Testing manuals aim at optimizing and securing the testing process of the hardware. Testing is done in two parts. At first, factory acceptance testing is done in facility of Valmet. Site acceptance testing is done after hardware has been installed in factory. This thesis includes testing manuals for the both phases of testing. Factory acceptance testing of several projects and site acceptance testing of one project were observed to produce testing manuals. Produced testing manuals are confidential and remain in the use of Valmet. Process of producing testing manuals and general content is included in published report of this thesis. Part of thesis was also electrical work safety in factory acceptance testing and deployment. Ensuring electrical work safety was acknowledged in testing manuals and electrical work safety is covered in report. ▼ The main topic of the thesis is the comprehensive and rich jurisprudence of the European Court regarding the right to life and the standards developed by the Court in this domain, to guarantee the protection of this right which passionates not only lawyers, barristers and judges, but also medical doctors, criminologists and philosophers. This right is not absolute and it is not unconditionally protected, as article 2 of the European Court of Human Rights does not rule out the death penalty when carried out in execution of the court judgment. The second paragraph of this article foresees some exceptional situations in which the deprivation of life is not considered illegal - when death results from the use of force which is no more than absolutely necessary: (a) in defense of any person from unlawful violence; (b) in order to effect a lawful arrest or to prevent the escape of a person lawfully detained; (c) in action lawfully taken for the purpose of quelling a riot or insurrection. The Court has considered several main groups of cases. The first ones are the cases of death as a consequence of the use of force. Therefore it is of utmost importance that such force be used in accordance with principles of proportionality, necessity and appropriateness. The second important group of cases is the one concerning forced disappearances, where states are obliged to take specific and effective measures in order to examine each case of disappearances in detail. The thesis is also examining numerous legal and ethical dilemmas related to the third group of cases, concerning abortion and the rights of the unborn child. The European Convention does not define „everyone“ whose life needs to be protected. Some people believe that a life begins with conception, others that it begins only at birth. In this case the Court was very cautious and refused to decide whether article 2 of the European Convention applies to a foetus. The next group of the cases relates to euthanasia, which has been recently permitted in several European countries. The Court has repeated its well-known stance that there is an obligation for states to protect life and concluded that the right to life cannot be interpreted in its negation, i.e. as the right to die. Therefore, the main arguments for and against euthanasia had to be invoked. The last group of the cases before the Court is the one in which the deprivation of life had occurred as the consequence of factors related to environment and health care (including medical errors), where states had to improve actively the living conditions and promote the protection of life. The thesis elaborates the right to life as the principal human right, indispensable for all other rights, and it insists on positive obligations of state when it comes to its violation. Les Oligosaccharides soluble (OS) sont essentiellement générés durant le processus de dégradation des N-glycoprotéines nouvellement synthétisées et mal conformées (ERAD) et durant la voie « turn-over » des glycoprotéines matures. Dans le but de déterminer si la modification post-traductionnelle O-GlcNAc est effectivement impliquée dans le processus de dégradation des N-glycoprotéines, nous avons analysé par spectrométrie de masse les OS provenant des cellules CHO après traitement par l’inhibiteur PUGNAc. Le PUGNAc ou « O-(2-acetamido-2-deoxy-D-glucopyranosylidene) amino-N-phenylcarbanate » est un inhibiteur puissant de l’O-GlcNAcase (OGA) qui catalyse l’hydrolyse du résidu O-GlcNAc des résidus sérine et thréonine des protéines O-GlcNAcylées.L’analyse par spectrométrie de masse révèle l’apparition d’une population d’OS de structures anormaux dans les cellules CHO suite au traitement par le PUGNAc. Cette population a été identifiée comme ayant des structures possédant des résidus GlcNAc au niveau de leur extrémité non-réductrice issues d’une dégradation lysosomale incomplète des glycoprotéines. Contrairement au PUGNAc, le NButGT, un autre inhibiteur de l’OGA, n’aboutit pas à l’apparition de cette population. Ainsi, Nous avons démontré que ces structures s’accumulent exclusivement dans la fraction membranaire conséquence de l’inhibition des β-hexosaminidases lysosomaux par le PUGNAc. Notre étude avait permis, d’un part, de mettre en évidence la capacité du PUGNAc de mimer une maladie de surcharge lysosomale et, d’autre part, de montrer un autre aspect des effets indésirables induits par le PUGNAc et qui nécessite d’être pris en considération lors de l’utilisation de cet inhibiteur.

Free oligosaccharides (fOS) are generated as a result of glycoproteins catabolism that occurs in two principal distinct pathways: the endoplasmic reticulum-associated degradation (ERAD) of misfolded newly synthesized N-glycoproteins and the mature N-glycoproteins turnover pathway. We analyzed fOS by Mass spectrometry in PUGNAc CHO treated cells in order to investigate whether O-GlcNAc modified proteins were involved in N-glycoprotein degradation process. The O-(2-acetamido-2-deoxy-D-glucopyranosylidene) amino-N-phenylcarbanate (PUGNAc) is a potent inhibitor of the O-GlcNAcase (OGA) catalyzing the cleavage of β-O-linked 2-acetamido-2-deoxy-beta-D-glucopyranoside (O-GlcNAc) from serine and threonine residues of post-translationally modified proteins. Mass spectrometry (MS) analysis revealed the appearance of an unusual population of fOS after PUGNAc treatment. The structures representing this population have been identified as containing non-reducing end GlcNAc residues resulting from incomplete lysosomal fOS degradation. Only observed after PUGNAc treatment, the NButGt, another OGA inhibitor, did not lead to the appearance of the population. Le glioblastome multiforme (GBM) est la tumeur cérébrale maligne la plus fréquente et la plus agressive chez l’adulte. Malgré l’utilisation concomitante de la chirurgie avec la radiothérapie et la chimiothérapie (protocole de Stupp), le pronostic des patients reste sombre avec un taux de survie à 5 ans inférieur à 10 %. La présence de cellules souches cancéreuses (CSC) favorise le maintien de la tumeur puis l’échappement tumoral responsable de la rechute des patients. Il est donc nécessaire d’identifier de nouvelles cibles thérapeutiques pour améliorer la prise en charge de ces patients. Dans ce contexte, l’immunothérapie, stratégie utilisant le système immunitaire pour traiter le cancer, représente une voie prometteuse.Nous identifions ici le OAcGD2 comme un nouvel antigène tumoral du GBM et a partir de biopsie tumorale provenant d’une cohorte de 37 patients. L’utilisation de primoculture de GBM nous permet de plus de démontrer que ce marqueur est également exprimé par les cellules souches de GBM. Nous apportons la preuve de concept de l’immunothérapie du GBM à l’aide d’anticorps monoclonaux anti-OAcGD2 in vitro et in vivo. A côté de la cytotoxicté immunologique, nous identifions un nouveau mécanisme de mort programmé, l’oncose, mis en jeu par les AcM anti-OAcGD2 permettant de sensibiliser ces cellules aux agents de chimiothérapie comme le témolozomide. L’utilisation de l’immunothérapie ciblant le marqueur gangliosidique tumoral OAcGD2 pourrait ainsi permettre d’améliorer l’efficacité de la thérapeutique actuelle (protocole de Stupp) du GBM

Glioblastoma multiforme (GBM) is the most common and agressive primary brain tumors in adults. Despite the concomitant use of surgery with radiotherapy and chemotherapy, the prognosis of patients remains extremely low. The presence of cancer stem cells (CSC) promotes the maintenance of the tumor and then the tumor escape responsible for the relapse of the patients. Therefore it is necessary to identify new therapeutic targets to improve the management of these patients. In the past few decades, immunotherapy represents an important part of treating certain type of cancer. It uses the immune system to treat cancer. Here, we found that O-acetyl GD2 (OAcGD2) is expressed in surgically resected human glioblastoma tissue. In addition, we demonstrated that 8B6 monoclonal antibody specific for OAcGD2 could effectively inhibit glioblastoma cell proliferation in vitro and in vivo. Mostly, we found that OAcGD2 was expressed on the GBM stem cells. We also observed that mAb 8B6 promoted the elimination of GBM via a oncosis-like mechanism. Moreover, this mechanism of programmed cell death induced by anti-OAcGD2 mAbs, sensitizes GBM cells and CSCs to chemotherapy agents such as temolozomide (TMZ). Taken together, these results indicate that O-acetylated GD2 represents a novel antigen for immunotherapeutic-based treatment of high-grade gliomas, and that anti-OAcGD2 mAbs combined with TMZ could enhance therapeutic response in GBM.

La O-GlcNAcylation est une modification post-traductionnelle appartenant au groupe des glycosylations. C’est une modification dynamique, analogue à la phosphorylation, dont la réversibilité est contrôlée par un couple d’enzymes, la O-GlcNAc transférase (OGT) et la O-GlcNAcase (OGA). Bien que les fonctions de la O-GlcNAcylation aient été étudiées à différents niveaux de régulation cellulaire, aucune étude n’a porté jusqu’ici sur sa localisation potentielle au niveau des radeaux lipidiques (rafts). Or, il a été récemment démontré que l’élévation des niveaux de O-GlcNAcylation participait activement au phénomène de résistance à l’insuline et un nombre important de preuves a également été apporté sur le rôle critique des microdomaines lipidiques dans ce phénomène de résistance. On sait également depuis peu qu’après stimulation par l'insuline, l’OGT est recrutée à la membrane plasmique par l'intermédiaire de son domaine PPO (PIP-binding activity of OGT), et que cette relocalisation inactive la voie de signalisation PI3-kinase/Akt par antagonisme vis-à-vis de la phosphorylation. Ces trois observations suggèrent que l’OGT puisse constituer un relai essentiel permettant de transmettre l’information portée par des stimuli externes au travers de la membrane plasmique, et plus particulièrement des rafts, par le biais d’une régulation des voies de signalisation. Au cours de ma thèse, nous avons adapté une technique permettant de purifier les microdomaines lipidiques, en procédant par une extraction par le détergent CHAPSO suivi d'une étape d'ultracentrifugation. Nos expériences ont montré pour la première fois la localisation de l’OGT et de protéines O-GlcNAcylées, parmi lesquelles nous avons identifié la chaîne bêta du récepteur de l'insuline et Hsp70, au niveau des microdomaines lipidiques. Le recrutement de ces éléments est contrôlé par l’insuline: nous avons en effet observé une association progressive de la glycosyltransférase aux microdomaines lipidiques après stimulation par cette hormone. Nous avons démontré que, en réponse à l'insuline, l’OGT est recrutée au niveau des lipides rafts, ce qui suggère une régulation négative de la voie PI3-Kinase/Akt par O-GlcNAcylation, au niveau des microdomaines lipidiques. Cette étude est la première suggérant une relation directe entre la O-GlcNAcylation des protéines associées aux rafts et la régulation de la signalisation cellulaire par l'insuline. En marge de ses travaux, nous avons mis en évidence l'existence de la O-GlcNAcylation chez les deux parasites apicomplexes Toxoplasma gondii et Plasmodium falciparum. Une approche biochimique et bioinformatique a également permis de prouver l'existence propre de l'OGT chez le toxoplasme.

▼ The purpose of this study was to investigate the increasing importance of person-organisation fit in organisations and how this might be attained during the recruitment and selection process. It achieved this aim by examining Schneider’s (1987) attraction-selection-attrition model within a graduate recruitment context. Three stakeholder perspectives were studied – job seekers, recruiters and employees – with each perspective assigned to one element of the model. The study consisted of three distinct parts. First, an online survey was completed by 137 university students to understand the ‘attraction’ element of the model, and to gauge the importance placed on person-organisation fit by job seekers. Second, 26 graduate recruiters were surveyed, and a further seven interviewed, to ascertain the emphasis placed on person-organisation fit during the ‘selection’ phase, and to gain an understanding of how ‘fit’ is assessed. Finally, ‘attrition’ to the firm was explored by surveying graduates and measuring the level of values congruence obtained with their employing organisation. The results revealed that although job seekers do consciously assess the values of the organisations to which they apply, person-organisation fit was not found to be of much importance to them. Instead, self-selection was based upon more tangible elements, such as reputation, of a firm. On the other hand, it was found that person-organisation fit has become a prevailing feature of the graduate selection process, with organisations spending a large percentage of time evaluating a candidate’s ‘fit’ to the firm rather than their ‘fit’ to the specific requirements of the job. Person-organisation fit was typically assessed during in-person interactions, with recruiters evaluating a range of behavioural competencies that related back to the firm’s core values. This study also revealed a low level of values congruence (i.e. person-organisation fit) is actually achieved in the appointment of university graduates. Overall, support for the attraction-selection-attrition framework is not found, as person-organisation fit appears to be neither of great importance to job seekers, nor is a high level of ‘fit’ attained in the recruitment and selection of graduates. ▼ The O-acetylation of the essential cell wall polymer peptidoglycan is a major virulence factor identified in many bacteria, both Gram positive and Gram negative, including Staphylococcus aureus, Bacillus anthracis, Neisseria gonorrhoeae and N. meningitidis. With Gram-negative bacteria, the translocation of acetate from the cytoplasm is thought to be performed by an integral membrane protein, PatA. The acetate is then transferred to peptidoglycan by a peripheral membrane O-acetyltransferase PatB, whereas a single bimodal membrane protein, OatA, appears to catalyze both reactions of the process in Gram-positive bacteria. Only phenotypic evidence existed in support of these pathways because no in vitro biochemical assay was available for their analysis, which reflected the complexities of investigating integral membrane proteins that act on a totally insoluble and heterogeneous substrate such as peptidoglycan. In this thesis, I present the first kinetic and biochemical analysis of a PG O-acetyltransferase using PatB from N. gonorrhoeae as the model system. The enzyme has specificity for muropeptides that possess tetrapeptide stems on muramoyl residue sand with simple chitooligosaccharides as substrates, rates of reaction increase with increasing degrees of polymerization to 5/6. The data provided here strongly supports a mechanism whereby catalysis occurs by a double-displacement nucleophilic pathway involving a ping-pong bi-bi mechanism that proceeds through a covalent acetyl-serine intermediate. In addition to the catalytic serine, the aspartic acid and histidine residues that complete the catalytic triad were identified. I then applied the tools developed for the analysis of PatB to the Gram-positive O-acetyltransferase, OatA. The preliminary characterization of the catalytic domain of that enzyme detailed in this thesis will form the basis for future high-throughput screening with the goal of identifying OatA inhibitors. This information will be valuable for the identification and development of peptidoglycan O-acetyltransferase inhibitors, which could represent potential leads to novel classes of antibiotics. Together, these data solidify our understanding of the mechanism by which an important bacterial virulence factor is produced and pave the way for future development of novel antimicrobials. ▼ Le cancer colorectal (CCR) est une pathologie dont les causes tiennent en partie d’un style de vie basé sur un régime alimentaire de type occidental et un mode de vie sédentaire. Cela implique par conséquent que le CCR soit associé à d’autres troubles métaboliques tels que l’obésité et le diabète. D’un consensus général on considère que le CCR est une maladie qui est mieux traitée si elle est dépistée relativement tôt ; malheureusement il n’existe toujours pas d’outils de diagnostic suffisamment fiables. A ce propos, différents types de glycosylation ont été proposés comme de potentiels biomarqueurs de cette maladie. Les travaux abordés dans cette thèse ont concerné l'influence de la O-GlcNAcylation (consistant en le transfert d'un seul monosaccharide de N-acétylglucosamine sur des résidus de sérine/thréonine de protéines cytoplasmiques, nucléaires et mitochondriales), sur la glycosylation complexe dans des lignées cellulaires coliques (HT29, HCT116 et CCD841CoN). La O-GlcNAcylation est intimement liée au statut nutritionnel, il est d’ailleurs proposé que son niveau soit plus élevé dans les pathologies métaboliques. Néanmoins il est clairement montré que les cellules et les tumeurs coliques expriment des niveaux plus élevés de O-GlcNAcylation. Plusieurs études ont montré que l’inhibition ou la délétion de la O-GlcNAc transférase (OGT), l’enzyme responsable des processus de O-GlcNAcylation, réduisait l'agressivité ou les propriétés associées au caractère tumoral dans différents modèles de cancer. Quelques études ont également montré que la sous-régulation de l’OGT se répercutait sur les glycosylations complexes : N- et O-glycanes, et glycosphingolipides (GSLs). La O-GlcNAcylation participe à des processus cellulaires clés tels que la transcription et la traduction. Dans une étude préliminaire, nous avons examiné les niveaux de transcrits de quatre-vingt-six glycosyltransférases et glycosidases dans des conditions de siOGT. Le siOGT a permis de constater que plusieurs de ces gènes étaient régulés positivement ou négativement, bien que parmi ces gènes seuls 10 étaient régulés négativement de manière significative. L'influence de l’OGT sur l’expression protéique de plusieurs glycosyltransférases a également été explorée. Nous avons ensuite utilisé plusieurs lectines en microscopie par fluorescence et en cytométrie en flux ainsi que la spectrométrie de masse (glycomique) afin d'élucider les structures complexes des glycanes et leur niveau d’expression dans les cellules coliques traitées par des siOGT. La diminution de O-GlcNAcylation n'a eu qu’un effet modéré sur les N- et O-glycanes globaux. Cependant, l'expression de plusieurs antigènes de surface a été modifiée par le siOGT. La E-cadhérine, un facteur majeur de la transition épithéliale-mésenchymateuse, a été régulée positivement dans les cellules HT29 traitées par le siOGT. De plus, les glycanes complexes portés par la E-cadhérine ont été eux-mêmes modifiés dans les cellules traitées par le siOGT.
