▼ Local exhaust (LE) ventilation is a ventilation technique where contaminated air is locally extracted close to the contaminant source usually with the purpose to reduce the exposure of workers to dust, fumes or vapour, which can be hazardous to their health. The performance of a LE installation depends however on many influential factors, and there is not yet an international standardized way to test LE constructions. The present study is the natural continuation of some previous studies at the University of Gävle that aimed at contributing to the establishment of such tests. The study entails full scale experimental measurements that include 3-D air velocity measurements and tracer gas tests in a controlled air turbulence environment generated through physical movements of a vertical, human-sized cylinder. These measurements were focused on human exposure, which was analysed by means of a seated human simulator for different configurations in which the exhaust flow rate, turbulence level, the exhaust hood arrangement and the measuring/injecting distance varied. The use of a sonic 3-D anemometer, that yielded both magnitude and direction of the air movement, proved very useful in analysing the generated air turbulence. As a measure of the LE performance, PNV value (Percentage of Negative Velocities) was used. This measure represents the percentage of time when the air flow at the measuring point in front of the exhaust hood is directed away from the nozzle, i.e. when the velocity component in the direction towards the exhaust hood opening is negative. Regarding the results obtained, in an otherwise undisturbed environment, measurement data showed that the natural convection from the human simulator sitting in front of the LE introduces some disturbances of the air flow in the suction region, proportional to the exhaust flow rate. However, when additional turbulence was generated through the controlled movements of the human-sized cylinder, thus creating a controlled turbulence setting, natural human convection leaded to a lower percentage of negative velocities (PNV) in comparison with the case in which human simulator was not present, especially for low exhaust air flow rates and when the exhaust hood was raised from the table. The tracer gas tests implied injection of a neutrally buoyant tracer gas through a perforated sphere placed in front of the exhaust hood. The amount of tracer gas that escaped from the suction flow was measured both in the room air and in the breathing zone. The first measurements yielded a sensitive method for measuring the capture efficiency (CE) of the exhaust hood. The CE is the percentage of injected tracer gas that is directly captured by the exhaust hood. This parameter showed that although the convection flow generated by the human simulator leads to low PNV values, it seems that the tracer gas is not actually being captured, but trapped in that convection flow. Centuries of metalliferous mining activities have resulted in a legacy of contamination throughout the world. Unremediated mine wastes and tailings, as well as contaminated soils, water and sediments, represent ongoing sources of environmental degradation and human exposure, long after mine closure and abandonment. Despite global concern over these contaminant sources, there remain uncertainties surrounding the nature of human exposure to mine wastes and their toxicologically relevant characteristics. As urbanisation expands into areas proximal to abandoned mine sites, an understanding of the human-contaminant interface at this boundary is critical for assessing the potential health risks. This thesis addresses this knowledge gap by investigating the importance of particle size as a factor governing the distribution of metals and metalloids in historical gold mine wastes in regional Victoria, Australia, with an emphasis on arsenic as a contaminant of potential concern. By characterising those particle size fractions that are relevant to dust mobilisation and human exposure, this thesis examines the human-contaminant interface using a multi-pathway approach. In particular, this thesis focuses on the potential for exposure via inhalation of mine waste particulates. The outcomes of the studies presented in this body of work demonstrate that historical gold mine wastes in regional Victoria represent a source of readily ingestible and inhalable particulates characterised by extremely elevated levels of arsenic (and other contaminants) well above their bulk (in situ) concentrations. Although lung bioaccessibility testing and mineralogical analyses revealed that most of the arsenic in inhalable dust has been naturally immobilised, the lung-soluble fraction should be considered when undertaking risk assessments for chronic exposure. This thesis provides a framework for the development of targeted management strategies for unremediated historical gold mining wastes in regional Victoria. The findings suggest there is a need for environmental regulations to shift from generic guideline values to exposure-specific guidelines that more accurately reflect the human health risks posed by historical mine sites. This thesis has emphasised the notion that in order for remedial action to accurately match the level of risk, the sourcepathway- receptor linkage must be evaluated using a systematic size-resolved approach.

▼ A dearth of knowledge exists regarding occupational glyphosate exposures among amenity horticulturalists. Glyphosate is the highest volume herbicide used globally, and has recently been classified by the International Agency for Research on Cancer as a 2A 'probably carcinogenic to humans’. The objective of this study was to characterise glyphosate exposures and identify exposure determinants among amenity horticulturists. Human biomonitoring assessments were completed by collecting spot urine samples from workers. In parallel, a dermal and inadvertent ingestion study was completed by collecting wipe samples of the worker’s hands and perioral region and by analysing samples of worker gloves. Samples were analysed using liquid chromatography tandem mass spectrometry (LC-MS/MS). Linear mixed effect regression models were constructed to evaluate determinants of dermal, inadvertent ingestion and total glyphosate exposure. A total of 205 urine samples and 351 wipe and glove samples were collected and analysed for glyphosate across 69 work tasks involving the glyphosate based pesticide products. Glyphosate concentrations ranged from below the limit of quantification to 10.66 μg L−1; the human biological half-life was estimated to be 5 ½ to 10 hours. Peak exposure levels were identified in urine samples collected up to three hours after cessation of the pesticide task. A forward built mixed effect model including sampling time, participant age and application type explained 62% of the variability in worker hand glyphosate concentrations. In the inadvertent ingestion exposure model the determinants hand contamination, the frequency of hand to mouth and surrounding area contact and sampling time explained 50% of the variability in perioral concentration. Combined hand and perioral region concentrations explained 40% of the variability in the urinary glyphosate concentrations (μg L-1). Occupational glyphosate exposures among amenity horticulturalists are higher than expected from environmental exposure alone and comparable with agricultural studies. ▼ Environmental xenoestrogens (EEs) are chemicals that when they enter the body, the body responds to them as it would to endogenous estrogens. Humans are exposed to these chemicals on a daily basis via natural components, additives and contaminants in food and water, through the use of pharmaceuticals and personal care products such as sunscreens, lotions and toothpaste. Exposure to EEs is thought to result in adverse effects on humans such as decreased fertility, increased susceptibility to hormone-sensitive cancers, deformities of the male genitalia and precocious puberty in females. The critical window of exposure is thought to be early fetal development, when tissues are rapidly differentiating under the control of endogenous estrogens. However, there is limited data in the literature on human fetal exposure to EEs. The first objective of this study was to assess human fetal exposure to a suite of 35 EEs by analysis of paired samples of amniotic fluid and maternal urine were collected from 32 New Zealand women between 14 and 20 weeks gestation. The analytical chemistry methods required for this study were developed and validated. The results demonstrate that fetal exposure is highly correlated with maternal exposure. This study is the first to report maternal urine levels of two UV filters and amniotic fluid levels of parabens, UV filters and triclosan. A model based on simple additivity of effect was developed that combined the measured concentrations with literature data on relative estrogenic potency to assess the magnitude of the estrogen signal that may be attributed to the EEs. This model suggests that the fetus may experience an estrogen signal due to the measured EEs that could be as large as the endogenous estrogen signal. A second objective was to use computational docking to study the interactions of the EEs with the human estrogen receptor (hER) protein. The docking studies show that the rigid endogenous ligand, 17β-estradiol (E2) interacts with the hER to produce a single, well-defined complex with the receptor and the flexible EEs produce multiple, distinct energy-equivalent complexes. EEs are not able to interact with the binding cavity to stabilise the rigid hER-E2-like topology of the complex. As a result, the hER-EE complexes can be thought of as more pliable or ‘floppy’ and thus able to respond to the cell context in multiple ways, leading to variations in gene expression in different target tissues. These multiple pathways may explain the range of physiological responses attributed to exposure that depend on the timing of exposure and the sex of the individual exposed.

▼ Background: Arsenic (As) is a naturally-occurring element with known toxicant effects. The primary exposure pathway is through ingestion, but the overall contribution of food versus water and the impact of specific dietary nutrients on urinary As excretion is not well understood. Methods: Secondary analyses of laboratory results from food, water and urine samples, questionnaire and anthropometric data, and dietary records were performed on four study populations: the National Health Exposure Assessment Survey (NHEXAS)-Arizona, Arizona Border Survey (ABS), the Arizona sub-group of the Binational Arsenic Exposure Survey (BAsES), and the 2003-2004 National Health and Nutrition Examination Survey (NHANES). Dietary As intake was measured in duplicate food samples and/or modeled from dietary records for each population using the U.S. Total Diet Study (TDS) arsenic residue database and a published market basket survey. Urinary total As, As⁵, As³, monomethylarsonic acid (MMA), and dimethylarsinic acid (DMA) were analyzed, and sum of species As was calculated as the sum of As⁵, As³, MMA and DMA. Regression analyses modeled the relation between urinary As biomarkers (total, sum of species, MMA:sum of species, and DMA:MMA) and dietary As, adjusted for drinking and cooking water As intake, current smoking, sex, age, ethnicity, body mass index, and nutrient intake. Results: Modeled dietary As based on TDS mean As residue data greatly underestimated exposure as compared with measured As in duplicate diet samples and estimates based on other residue data. Dietary As was a significant predictor of urinary total As in all four populations, of sum of species As in both BAsES and NHANES, and of %MMA and DMA:MMA in NHANES. Dietary protein intake was associated with decreased sum of species As in both BAsES and NHANES, but dietary folate was not. Conclusions: Dietary As contributes a markedly greater proportion of total ingested As and is a better predictor of urinary As than water As intake in the U.S. Among subjects who did not consume seafood, total As exposure from food and water exceeded the provisional tolerable daily intake of 2.1 µg/kg body weight/day in 3-15% of these study populations. Increased protein intake may mitigate the effects of As.

▼ Rabies post exposure prophylaxis (PEP) includes administration of both vaccine and anti-rabies immunoglobulin. Rabies immunoglobulins are expensive and available in limited quantities. There is an inherent variability in quality and specificity between batches of immunoglobulin preparations and possibility of contamination with known or unknown pathogens. To provide a cost-effective and safe replacement for currently used human rabies immunoglobulin (HRIG) human anti-rabies monoclonal antibodies (huMAbs) were developed. The present study evaluated the overall prophylactic efficacy of SO57 and SOJB MAbs individually, in a mouse model when used in combination with rabies vaccine and compared with conventional HRIG. When the protective effect of antibodies was examined in mice challenged with CVS-N2C virus, a dose-dependent survivorship response was observed in animals treated with antibodies and no animal survived in the control group. At the highest dose of antibody given 80% survivorship was recorded for SO57 and SOJB antibodies where as only 50% survived with HRIG. Serum half-lives were found to be approximately 16 days for SO57 (IgG1); 11 days for SOJB (IgG3) and 8 days for HRIG (polyclonal). Furthermore, we investigated the potential interference of antibodies on vaccine mediated-immunity. When compared to the control group which was given only vaccine shots, the groups of animals given both antibody and vaccine developed lower virus neutralizing antibody (VNA) titers. The VNA titers appear to be a function of dose of antibody given and serum half-life of each antibody. The interference, as correlated with VNA titers developed in mice, is comparable to HRIG for both SO57 and SOJB MAbs. When we investigated more on the induction of interference by antibodies on vaccine –mediated immunity with combinations of (i) antibody and different concentrations of vaccine (ii) administration of antibody and vaccine at different time intervals followed by intracerebral (i.c.) challenge all the groups of mice recorded higher survival rates. But the survival rates for SO57 and SOJB were comparable to that of HRIG. Unlike in the previous studies of mouse monoclonal antibodies, we observed that passively administered human monoclonal antibodies and HRIG protects mice against intra cerebral challenge. The protective effect at the point of i.c. challenge may be a synergistic effect of passive and active immunities and account for higher survival among groups. Together this data suggests that human anti-rabies monoclonal antibodies (huMAbs) viz: SO57 and SOJB have potential to replace the currently used human rabies immunoglobulin (HRIG) for rabies post exposure prophylaxis (PEP) regimen. ▼ Adverse health effects from exposure to air pollution, although at present only partly understood, are a global challenge and of widespread concern. Quantifying human exposure to air pollutants is challenging, as ambient concentrations of air pollutants at potentially harmful levels are ubiquitous and subject to high spatial and temporal variability. At the same time, individuals have their very own unique activity-patterns. Hence exposure results from intertwined relationships between environmental and human systems add complexity to the assessment process. It is essential to develop a deeper understanding of individual exposure pathways and situations occurring in people’s everyday lives. This is important especially with regard to exposure and health impact assessment which provide the basis for public health advice and policy development. This thesis describes the development and application of a personal monitoring method to assess exposure to fine particulate matter in a variety of microenvironments. Tools and methods applied are tested with respect to feasibility, intrusiveness, performance and potential for future applications. The development of the method focuses on the application in everyday environments and situations in an attempt to capture as much of the total exposure as possible, across a complete set of microenvironments. Seventeen volunteers took part in the pilot study, collected data and provided feedback on methodology and tools applied. The low-cost particle counter applied showed good agreement with reference instruments when studied in two different environments. Based on the assessment of the two instruments functions to derive particle mass concentration from the original particle number counts have been defined. The application of the devices and tools received positive feedback from the volunteers. Limitations are mainly related to the non-weatherproof design of the particle counter. The collection of time-activity patterns with GPS and time-activity diaries is challenging and requires careful processing. Resulting personal exposure profiles highlight the influence of individual activities and contextual factors. Highest concentrations were measured in indoor environments where people also spent the majority of time. Differences between transport modes as well as between urban and rural areas were identified.

▼ Airborne particles are everywhere around us, and have always been. Particles generated by human activities has increased drastically since industrialization, and several epidemiological studies have shown that inhaled particles can cause adverse health effects. The concern about health effects have during the last decade shifted towards fine and ultrafine particles, not least due to the emerging field of nanotechnology. Of special interest are the particles to which we are exposed indoors – in the industrialized part of the world we spend around 90% of our time indoors (at home and at workplaces/schools). Particles generated in these environments often cause intense peaks in concentration, and are often consequences of our own activities. Especially combustion/thermal processes (such as welding, frying, burning candles etc.) cause peaks in number concentration, often more than an order of magnitude higher than ambient concentrations. We have conducted time-resolved particle measurements in several homes which confirm this. From these measurements, we have been able to show just how much occupants' activities affect the indoor concentration of ultrafine particles compared to outdoor concentrations. We have also estimated e.g. total integrated daily exposure. Exposure and emission measurements have also been conducted at a carbon nanotube producing facility, and a method for counting particles containing carbon nanotubes has been suggested and validated. Why certain particles are more dangerous than others is often investigated in animal exposure studies, where exposure levels are unrealistically high. For several reasons, the results of such studies are not simple to translate to the human system. To increase our understanding of which particle properties can cause effects in humans, a methodology for conducting human exposure studies have been developed and validated. In a controlled chamber we have exposed human test subjects to normal concentrations of common particle types; candle smoke, particles from terpene–ozone reactions and welding fume. Together with medical expertise, we have been looking for effects of these exposures. By using non-invasive tests (e.g. urine and blood samples and ECG) biochemical markers of exposure, and changes in heart rate variability (HRV) have been studied. A significant increase in the high frequency domain of the HRV during exopsure for candle smoke was found.

▼ Suboptimal diet is a major public health concern, responsible for ~10 million death/year globally associated with nutrition, plus additional deaths associated with environmental emissions from food production. Informing consumer choices is crucial and would require to consistently combine latest epidemiological evidence on the impact of diet and pollution, with life cycle assessment (LCA) of food systems to analyze relevant trade-offs. This dissertation aims to address four critical challenges for assessing the life cycle impact and benefits of food systems on human health: (1) The failure to capture both environmental impacts and nutritional effects of foods consistently. (2) The lack of nutritional assessment metrics that evaluate the performance of individual food items based on health burden. (3) The overly simplified assessment of impacts of particulate matter (PM2.5) on human health, which do not consider spatial variation in exposure, nor evidence for non-linear exposure-response. (4) The need for a consistent approach to evaluate multi-ingredient mixed dishes, a central component in modern diets. Chapter 2 developed a novel Combined Nutritional and Environmental Life Cycle Assessment (CONE-LCA) framework that evaluates and compares in parallel the environmental and nutritional effects of foods or diets in a common metric, disability adjusted life years (DALYs). A proof-of-concept case study indicated that nutritional health net benefits of adding a serving of milk to the average U.S. diet exceeded environmental impacts and highlighted the need for considering nutrition as a new LCA impact category. Chapter 3 operationalized the nutritional approach by establishing the Health Nutritional Index (HENI). This health burden-based nutritional index quantifies the health burden of one food serving in minutes of healthy life lost or gained, using epidemiological evidence for a comprehensive set of 16 dietary risks. Application to ~7,000 food items in the U.S. diet revealed substantial variability in HENI scores between and within food categories, thus the importance of informed choices at the level of individual food items. In Chapter 4, we developed spatially-explicit intake fractions for ground-level PM2.5, NH3, SO2, and NOx emissions in the contiguous U.S. for agriculture and other relevant sectors. Using a non-linear exposure-response function and state-specific burden data, we developed the corresponding characterization factors considering a marginal and an average slope. Spatial estimates varied by three orders of magnitude, sector-specific estimates by a factor of four, and the average slope doubled estimates compared to marginal. This work stressed the importance of spatially-explicit and sector-specific estimates in LCA. ▼ Background: Pre- exposure prophylaxis (PrEP) is a biomedical intervention that was approved by the Federal Drug Administration (FDA) in 2012 to reduce the chances of acquiring HIV (human immunodeficiency virus) by 90% or more. Numerous studies have been conducted worldwide analyzing different risk populations and showing the efficacy of PrEP by reducing transmission of HIV. Currently, the state of Michigan has no evidence on who is taking PrEP, which facilities are prescribing or referring PrEP, or what PrEP activities are taking place in a clinical setting. Local health departments (LHDs) are key players in screening individuals who are at risk for getting STDs, which serve as a population of those who could also get infected with HIV. Methods: In 2017, the Michigan Department of Health and Human Services (MDHHS) developed a survey to assess LHDs capacity, processes, and technical assistance regarding PrEP. Prior to this survey there has been one other survey about PrEP but was only distributed to private physicians in Michigan. The current survey was sent to 45 LHDs, with responses from medical directors, health officers, and STD clinic management. The survey was designed to assess three components for each LHDs: current PrEP activities, strengths and weakness of the activities, and potential resources or tools that could aid clinics in future PrEP implementation. Results: The results reveal that of the 59 respondents, the most frequent respondents were registered nurses, which comprised sixty-seven percent (67%) of the sample. In addition, of the 45 LHDs in the state, 42 (93%) provided responses. According to the three groups of respondents, 12% of clinics were prescribing PrEP, 52% were referring PrEP, and 37% were not doing any activities around PrEP. Clinics who are currently prescribing PrEP want to continue their PrEP activities and monitor PrEP uptake. However, 64% of clinics indicated they were undecided or not likely to implement PrEP in the future. All clinics expressed interest in receiving more guidance from the state health department and an increase of education and resources for staff and patients. Conclusion: Considering this was a baseline survey, the response rate was quite high for LHDs throughout Michigan. The survey indicated what PrEP activities LHDs were engaging in and what they would like to do in the future. About half of the respondents stated they are not doing any activities around PrEP, which can call for more action in those clinics. However, all respondents expressed interest in receiving more information and resources on how to expand PrEP activities in their clinic. ▼ ABSTRACT Over two decades ago, the European Union developed and started implementing the Extended Producer Responsibility (EPR) policy to prevent household hazardous waste (HHW) pollution and its environmental and human health risks. Because of the increase in population, rapid urbanization, and the amount of importing hazardous waste in developing countries, appropriate solid waste management and disposal are needed especially for HHW treatment. As a developing country, Mongolia does not have specific regulations and treatment requirements for HHW. Unlike developed countries, the mandatory EPR policy run by government will be necessary for Mongolia because small and family-owned businesses are not willing to handle waste voluntarily due to the cost of collection, recycling, and final disposal. Moreover, a Producer Responsibility Organization (PRO) will face difficulties if advanced recycling fees and end-of-life fees from consumers fund the organization, since the country`s GDP per capita in 2014 was only US $11,508.98 (Trading economics). Therefore, under the EPR program, cooperation between a municipality and PROs is necessary and mutually beneficial, especially in waste collection and transporting. This is especially important because of the potential health risks resulting from incineration of HHW. Due to open burning fluorescent bulbs in the central waste facility, it is estimated that 69,637kg Hg/yr could be released into the atmosphere directly. Human health risks from mercury exposure, using a traditional risk characterization procedure, showed potential neurological concerns (hazard quotient: 14.42, exposure time: 12hr/d) for children who live within 500m of this municipal waste incineration. In contrast, renal effects due to lead exposure (82.28kg Pb/yr) from lead-acid vehicle battery recycling factories showed a low health risk for children. To prevent this children`s neurological effect implementation of the mandatory EPR program for HHW such as automobile batteries, fluorescent bulbs, and used tires is necessary in developing countries because most of MSW facility are designed poorly to capture emissions of hazardous substances. ▼ Children residing in legacy mining towns are potentially disproportionately exposed to metal(loid)s via ingestion (of food, water, and incidental soil), and dust inhalation, thus, increasing their risk of exposure when engaging in school or home gardening or playing outside in non-garden, playground soil. The purpose of this citizen-science study was to assess preschool children’s potential metal(loid) exposure through these routes. Parents and teachers were trained to properly collect water, soil, and vegetable samples from their preschools throughout Nevada County, California. Arsenic (As), cadmium (Cd), and lead (Pb) concentrations in garden irrigation sources did not exceed the U.S. EPA Safe Drinking Water Act and Lead and Copper Rule’s maximum contaminant and action levels. In general, As and Pb concentration from garden and playground soil samples exceeded the U.S. EPA Regional Screening Level (RSL), CalEPA’s California Human Health Screening Level (CHHSL) and California Department of Toxic Substances Control Screening Level (DTSC-SL). In contrast, all Cd concentrations were below these recommended screening levels. Dust samples (< 10 μm diameter) were generated from surface garden and playground soil collected at the preschools. Soil and dust samples were then analyzed by in-vitro bioaccessibility assays using synthetic lung and gastric fluids to estimate the bioaccessible fraction of As, Cd, and Pb in the body. Metal(loid) exposure via homegrown produce revealed that lettuce, carrot, and cabbage grown in the preschool gardens accumulated a higher concentration of metal(loid) than those store-bought nation-wide (U.S. FDA Total Diet Study, 2006-2013). None of the vegetables exceeded the respective recommendation maximum levels for Cd and Pb set by the World Health Organization Codex Alimentarius Commission. Lastly, a dietary assessment consisting of a 24-hour recall and food frequency questionnaire was administered to determine ingestion rates of foods shown to accumulate these contaminants. The results of this study suggest that consumption of preschool-grown foods and incidental soil ingestion were major contributors to preschool-aged children’s exposure to As, Cd, and Pb. This comprehensive environmental monitoring and exposure assessment that includes child-specific dietary data and the average daily doses of contaminants through ingestion of soil, water, and vegetable and inhalation of dust informs future risk assessment and public health interventions related to childhood exposure to these metal(loid)s. ▼ The use of pesticides has caused pollution of surface and groundwater, soil and air across the world. Many pesticides cause health effects in humans. General populations are exposed to some degree and diet is reported to be a main source of exposure. It is important to study exposure and exposure-effect relationships in occupational groups and in general populations. For such studies, good methods to monitor exposure, such as human biomonitoring are needed. However, for many pesticides, there is a lack of knowledge on exposure biomarkers and analytical methods to measure exposure. Thus, there is a need for validated biomarkers of exposure and analytical methods.In this thesis, new methods were developed and validated to analyze the fungicides thiabendazole (TBZ), imazalil (IMZ) and pyrimethanil (PYM) and their exposure biomarkers in urine using LC-MS/MS. Further, an LC-MS/MS method for analysis of glyphosate (GLY) and one of its metabolite in urine was established. The analytical methods were sensitive enough to measure a wide range of concentrations of exposure biomarkers in populations and showed good precisions.Human experimental exposures (oral and/or dermal) with TBZ, IMZ, PYM and GLY were performed with 2-3 volunteers to investigate basic toxicokinetics. The excretion of biomarkers in urine was rapid with short half-lives for all the four pesticides. The biomarkers of IMZ, TBZ and PYM were found conjugated with glucuronides and sulfates. The concentrations of biomarkers varied quantitatively with the degree of exposure.The analytical methods were applied to biomonitor the exposure in occupationally exposed groups and in general populations. The measured biomarkers in population groups in Sweden reflect concentrations which seem to be far below the excreted concentrations after an intake of a dose equivalent to a dose half or equal to the ADIs. The diet of the general population could be a possible source of exposure. The measured exposure biomarkers of IMZ in greenhouse workers and of PYM in orchardists were higher than the general population and reflected concentrations that were sometimes close to those following an exposure at a dose half or equal to the ADIs. In conclusion, the analytical methods performed well and can be applied in biomonitoring studies. The identified biomarkers of all four pesticides were related to the exposures and the human experiments facilitated validation of the biomarkers. The short urinary excretion half-lives require a well-planned sampling strategy.

▼ How best to model humans for accurate SAR estimates remains an open question. Much work has been done over the years, attempting to quantify which and how various anatomic parameters such as tissue morphology, size, dielectric properties and relative location, affect energy absorption; and how to incorporate those parameters into models that provide conservative SAR estimates protecting all individuals. Since the available computational models are either highly complex realistic models or simple single tissue models, this issue is not easily explored, since the identified parameters are not easily varied within the models. Attempts have been made in this thesis to overcome these limitations by creating a unique alternative model of the human head termed Geometry Head, incorporating a reduced set of tissues in a semi-homogeneous, simplified geometry for which the key parameters may be varied parametrically. Data was gathered from literature regarding what variations exist in human populations for tissue parameters, and used to populate the model. The computational model was validated using simplified physical models of human heads, custom made for this purpose. The Geometry Head model was used to systematically test what relationships exist between several anatomic variations in adult humans and consequent energy absorption due to RF exposure at a single frequency of 900 MHz, namely: cranial thickness; skin thickness; head size; and dielectric properties of tissues. Overall, results suggest that SAR predictions could not have been anticipated using a priori reasoning, as location and magnitude of SAR maxima were seen to alter in unexpected and irregular ways with variations in tissue parameters. A compromise model like the one proposed here, which provides a point of diminishing returns between the high complexity of the multi-tissue models and the single-tissue homogeneous model, allows increased granularity for determination of SAR maxima in sensitive tissues. Results of this work provide some inroads into establishing the level of complexity necessary for simulating human heads for radiofrequency exposure compliance calculations. Relationships between magnitude and location of regions of high SAR in the head and variables listed above have been quantified and qualified in this thesis. The model proposed here is available for public use upon request. It can easily be amended to incorporate more or fewer tissues with varying levels of complexity; augmented to better resemble anatomy of children's heads; or used in its current state to further explore relationships between anatomic variations and absorption at the same frequency used here, or other RF frequencies.

▼ La biosurveillance humaine permet aujourd’hui d’évaluer notre exposition aux produits chimiques par la mesure soit des substances elles-mêmes, soit de leurs métabolites ou marqueurs d'effets sur la santé, à partir des fluides corporels ou des tissus. Les informations recueillies dans le cadre d’enquêtes épidémiologiques renseignent sur l'exposition humaine et constituent des bases précieuses dans la recherche des relations exposition-réponse chez les humains. Les travaux de cette thèse s’inscrivent dans le cadre de l’enquête transversale IMePoGe conduite entre 2008-2010 dans la région Nord–Pas-de-Calais (au nord de la France), incluant 2000 résidents adultes âgés de 20 à 59 ans, et visant à quantifier les niveaux d’imprégnation de la population à 14 métaux et métalloïdes (aluminium, antimoine, arsenic total, béryllium, cadmium, cobalt, chrome, mercure, manganèse, nickel, plomb, thallium, vanadium, zinc) choisis pour leurs effets toxiques et la fréquence de l’exposition professionnelle et environnementale. Les objectifs spécifiques de cette thèse étaient, tout en portant une attention particulière au plomb et au cadmium, deux métaux néphrotoxiques connus dans la littérature : i) d’établir la distribution de l’imprégnation aux métaux et métalloïdes dans la population du Nord de la France et de comparer le niveau régional d’imprégnation avec les données nationales et internationales ; ii) d’identifier les facteurs majeurs de variation de l’imprégnation et les sources d’exposition au plomb et au cadmium dans la population générale ; iii) d’étudier la relation entre la variation du débit de filtration glomérulaire et les niveaux d’imprégnation aux métaux. Globalement, les concentrations sanguines et urinaires de la plupart des métaux et métalloïdes étaient plus élevées que celles rapportées dans l’enquête nationale nutrition santé conduite sur la même période dans la population française, à l’exception du vanadium urinaire et du plomb sanguin. La plombémie moyenne régionale (moyenne géométrique) était de 18,8 μg/L. De nombreuses sources d’exposition au plomb existaient dans la population et étaient à la fois d’origine professionnelle, environnementale et alimentaire. Pour ce qui est du cadmium, le tabagisme se présentait comme la principale source d’exposition récente ou chronique au métal : la cadmiémie moyenne, reflet de l’exposition récente, était de 0,39 μg/L et passait de 0,26 μg/L chez les non-fumeurs à 0,84 μg/L chez les fumeurs ; la cadmiurie moyenne, reflet de l’exposition chronique, était de 0,37 μg/L (0,33 μg/g créatinine) et passait de 0,33 μg/L (0,29 μg/g créatinine) chez les non-fumeurs à 0,46 μg/L (0.37 μg/g créatinine) chez les fumeurs. ▼ Despite its importance for Disaster Risk Management (DRM), the mapping of human distribution and population exposure has lagged behind hazard modeling and mapping. Assessing population exposure to actual or potential disasters can benefit all phases of the disaster management cycle, e.g. risk and impact assessment, mitigation, preparedness (including early warning and evacuation), and response. This assessment requires geo-information on population distribution at a range of spatial and temporal scales, as disasters can strike at any time and with little warning, and affect from local to global areas. This thesis comprises contributions of population distribution modeling to advancing Disaster Risk Management and Reduction efforts by: (i) developing geospatial models that improve population distribution datasets at a range of relevant spatial and temporal scales and resolutions; (ii) applying those data to (real) disaster risk scenarios by combining geospatial population layers with geophysical hazard maps; (iii) using spatial analysis for quantitatively and qualitatively assessing human exposure to specific hazards and levels, for cartographic representations and visualization, and for showing contributions to DRM. We conclude that since impacts of hazards and disasters are place and time dependent, several DRM and Disaster Risk Reduction phases and activities would benefit from relying on more spatially-detailed and time-specific assessments of population exposure, at a range of relevant spatio-temporal scales (local to global). Also, improving population distribution data for human exposure assessment requires addressing challenges present in input data and geospatial modeling. While at local scale in data rich environments more detailed and sophisticated models can be developed with acceptable uncertainty, scaling up such approaches to the global domain requires addressing different challenges, such as limitations in data availability, quality, and concepts in order to maximize the range of uses of population data, especially for supporting ongoing international development agreements. Finally, geospatial information on population distribution constitutes crucial baseline data for risk analysis and DRM across a range of hazards and threats, and investing in improving data benefits population-related analyses by detailing and revealing a sharper picture, with the aim of ‘leaving no one behind’. Cette thèse résume une démarche intégrée visant à évaluer l’exposition humaine et ses variations spatio-temporelles en cas de tsunami dans une zone urbaine littorale en Indonésie, ainsi que la capacité d’évacuation vers des refuges. Ce travail de recherche systématise des méthodes permettant d’estimer la quantité de population présente heure par heure durant n’importe quel jour de la semaine et de l’année, à une échelle très fine, dans une zone urbaine. Il se fonde pour cela sur une hypothèse de rythme de vie contrôlant les activités et donc la distribution de la population. L’heure d’arrivée d’un tsunami étant imprévisible, ces informations sont très importantes pour améliorer les programmes de réduction du risque. Cette démarche permet ainsi de dégager des scénarios types de distribution de la population, utilisés pour ensuite évaluer la capacité d’évacuation de ces populations. Le modèle de simulation dynamique issu de cette recherche permet de mesurer l’accessibilité des zones selon certains scénarios, et de proposer des améliorations pour une meilleure préparation de la protection des civils.

This dissertation summarizes an integrated approach whose aim is to assess the human exposure and its spatial and temporal variations in the event of a tsunami in a costal urban zone of Indonesia, as well as the capacity to join evacuation shelters for populations under threat. This research systematizes methods to estimate the amount of people present hour by hour during any day of the week and the year, at a very fine scale, in an urban area. It uses a hypothesis about a common rhythm of life which controls the activities and therefore the distribution of the Padang inhabitants. Considering that time of a tsunami occurrence is impossible to estimate, this information is very important to improve risk reduction programs. This approach allows in particular identifying various types of scenarios for the distribution of the population that can then be used to evaluate the evacuation capacity of these populations. A dynamic simulation model resulting from this research allows for the measurement of the accessibility of shelters following these scenarios. The analysis of the results suggests improvements for a better preparation on the part of authorities to protect civilians.

▼ A range of brominated flame retardants (BFRs) have been incorporated into plastics, electronic equipment, foams and textiles. Among the most common of these, polybrominated diphenyl ethers (PBDEs), have come under a great deal of scientific and regulatory scrutiny due to their persistence in the environment, bioaccumulation potential and toxicity. With bans and manufacture withdrawals of PBDEs occurring in many nations over the past 15 years, replacement flame retardants often referred to as “novel” brominated flame retardants (NBFRs) have been employed in consumer goods and products. These compounds are structurally similar to PBDEs and share similar physicochemical properties to their predecessors. Consequently, NBFRs are being detected in environmental and biotic matrices, including humans, with increasing regularity. At present, however, the legislative mechanisms which regulate and restrict the use of PBDEs do not apply to the most common NBFRs. The objective of this thesis has been to investigate the occurrence and fate of PBDEs and NBFRs in Australian indoor and outdoor environments. Central to this work has been the focus of elucidating major pathways of human exposure to the emerging compounds in relation to their banned legacy counterparts. The early stages of this project entailed the development and optimization of extraction and quantitation protocols using selective pressurized liquid extraction (S-PLE) and gas chromatography coupled to tandem mass spectrometry (GC-MS/MS). Using these methods, analysis revealed that both PBDEs and NBFRs are widespread in the surface soils of the city of Melbourne, Australia. These were the first measurements of PBDEs or NBFRs in Australian soils and provided baseline data against which to assess the implications of new health investigation levels (HILs) legislated in 2013. The survey of urban soils revealed a number of key potential sources to soils, including polymer manufacturing and waste disposal processes such as incineration and landfill. Among the land-uses studied however, contamination at electronic and electrical waste (e-waste) recycling facilities was 2- 3 orders of magnitude greater than the levels at other examined point-sources. A follow-up study investigating the spatial distribution of PBDEs and NBFRs in soils around these facilities determined strong evidence that e-waste recycling in Melbourne did in fact have the potential to contaminate land over distances up to 900 m, with concentrations in soil decreasing exponentially with distance from the source. The final aim of this research was to investigate the concentrations of PBDEs and NBFRs in indoor dust from homes, offices and vehicles in Melbourne and estimate human exposure via dust inhalation and ingestion. This study provided the first wide-ranging survey of NBFRs in indoor dust in Australia and offered further evidence of PBDE contamination in indoor environments. As aflatoxinas são substâncias naturais que apresentam efeitos tóxicos aos humanos e são reconhecidamente carcinogênicas. Estas substâncias podem estar presentes na dieta humana ou, em casos específicos, no ar respirado. Desta maneira, a exposição humana às aflatoxinas é objeto de muita preocupação. Uma das maneiras mais eficazes de avaliar a exposição humana as aflatoxinas é através da mensuração da presença de biomarcadores da exposição a estas substâncias em fluidos biológicos. Dentre as possibilidades de biomarcadores de exposição às aflatoxinas tem-se que aflatoxina M1 (AFM1), presente na urina e leite humano, é considerada um biomarcador válido. Assim sendo, o objetivo deste trabalho de pesquisa foi avaliar a presença de AFM1 em amostras de urina provenientes de indivíduos residentes na região urbana e rural da cidade de Piracicaba-SP, assim como, de leite de gestantes de Piracicaba e cidades da região. Nos indivíduos doadores de amostras de urina foi levantado também o padrão de ingestão de alimentos com alto risco de conter aflatoxinas, através da aplicação de inquéritos de freqüência alimentar e recordatórios 24 horas. A análise de AFM1 em urina e leite foi realizada por cromatografia liquida de alta eficiência (CLAE) com detecção por fluorescência. A extração e purificação do extrato foram realizadas com auxílio de colunas de imunoafinidade. No total 69 amostras de urina e 18 de leite foram analisadas. Entre as amostras de urina detectou-se a presença de AFM1 em 54 (78%) das amostras, com concentrações variando de 1,8 até 39,9 pg/mL. Não foi observada diferença estatística entre as concentrações médias detectadas entre urinas de indivíduos da zona urbana e rural, bem como no nível de consumo de produtos de risco. Apesar das concentrações de AFM1 detectadas serem inferiores as concentrações médias reportadas em outros países a freqüência de amostras positivas foi bastante elevada mostrando que as populações estudadas estão sendo expostas às aflatoxinas. Assim, melhores avaliações dos níveis de exposição necessitam ser realizados considerando que a amostragem utilizada foi pontual, pode existir variação de contaminação sazonal com aflatoxinas na dieta e a contaminação é heterogênea dentro no alimento. Não foi observada uma correlação entre o nível do consumo de produtos de risco e as concentrações detectadas em amostras de urina. Apenas uma amostra de leite apresentou contaminação detectada; entretanto, o nível de contaminação estava entre o limite de detecção (LD) e o limite de quantificação (LQ).
