▼ Baylis-Hillman reactions using various aromatic aldehydes, activated alkenes and catalysts have been used to: - access an extensive range of poly-heterocyclic products;explore chemoselectivity; and optimise reaction efficiency. Chromone-3-carbaldehydes and chromone-2-carbaldehydes, prepared via Vielsmeier-Haack and Kostanecki-Robinson methodology, respectively, have been used as Baylis-Hillman substrates with four different catalysts, viz., 1,4-diazabicyclo[2.2.2]octane (DABCO), 3-hydroxyquinuclidine (3-HQ), imidazole and N’,N’,N’,N’- tetramethylpropanediamine (TMPDA), and with methyl vinyl ketone (MVK), methyl acrylate, cyclic enones (2-cyclohexen-1-one, 2-cyclopenten-1-one and chromones) as activated alkenes. Reactions of the chromone- -carbaldehydes with MVK afforded dimeric Baylis-Hillman adducts when catalyzed by DABCO but when the same reactions were repeated using 3-HQ as catalyst, the dimeric products were accompanied by tricyclic Baylis-Hillman adducts. Use of excess MVK, however, led to mixtures of the normal Baylis-Hillman adducts and the tricyclic adducts – interestingly, with the apparent absence of the dimeric products. While reactions of chromone-3-carbaldehydes with methyl acrylate afforded the normal Baylis-Hillman adducts, the chromone-2- carbaldehydes produced, instead, rearrangement products, consistent with an earlier, single observation. Reactions of 2-nitrobenzaldehydes with cyclic enones using imidazole as catalyst afforded the normal Baylis-Hillman adducts, reductive cyclisation of the 2-cyclohexen-1- one and 2-cyclopenten-1-one adducts, using acetic acid and iron powder, afforded the corresponding quinoline erivatives. Treatment of cyclic enones with pyridine-2-carbaldehydes and quinoline-2-carbaldehydes using TMPDA as catalyst generally gave the expected Baylis-Hillman adducts. However, indolizine derivatives were isolated directly from Baylis-Hillman reactions involving pyridine-2-carbaldehydes and 2-cyclohexen-1-one. The remaining Baylis-Hillman adducts were cyclized to the corresponding indolizines by treatment with acetic anhydride both under reflux and under microwave-assisted conditions, the latter approach providing remarkably rapid and efficient access to the polycyclic products. Computer modelling studies have been conducted on selected polycyclic products at the Molecular Mechanics (MM), Quantum Mechanical (QM) and Density Functional (DFT) levels. The theoretical results have been used to calculate UV, IR and NMR absorption data, which have been compared, in turn, with the experimental spectroscopic data. ▼ The first part of the research described in this thesis involves the development of a new methodology for the synthesis of N-hydroxy pyrrolidines, starting from carbohydrates as building blocks. The products were identified as possible synthons for the stereocontrolled synthesis of isosteric analogues of polyhydroxylated indolizidine alkaloids. The consecutive reduction and cyclisation of selectively protected 5-0-mesyl hexose O-(tert-butyldiphenylsilyl) oximes to afford chiral N-hydroxy pyrrolidines is discussed. The mechanism involves a cascade of neighbouring group participation steps by the O-benzoyl protecting groups. This protocol gave rise to novel chiral N-hydroxy pyrrolidines in good overall yield. The choice of leaving group as well as a labile oxime protecting group proved to be of great import in the outcome of the cyclisation reactions. The second part of the research concerns the ongoing development in our laboratories of the synthesis of analogues of biologically active compounds. In this regard, we were interested in synthesising the aza analogues of f3-C-nucleosides and f3-C-glycosides. Our strategy involves the synthesis of a D-ribose derived chiral cyclic nitrone as the key synthon. A facile route towards cyclic nitrones was developed starting from suitably protected hemiacetals of Dribofuranose. Readily available tri-O-benzyl-D-ribofuranose was allowed to react with hydroxylamine hydrochloride to afford an acyclic oxime. Selective silylation followed by iodonation at C-5 (with inversion of configuration) furnished the cyclisation precursor. Anhydrous TBAF-mediated desilylation and subsequent intramolecular nucleophilic attack afforded a cyclic nitrone in excellent yield. Following the same protocol, 2,3-isopropylidene-5- 0-trityl-D-ribofuranose was converted into the corresponding nitrone. The 1,3-dipolar cycloaddition reaction of a nitrone to an alkene is an extremely powerful synthetic method for the creation of complex heterocyclic structures. The reaction of the Dribose derived nitrones with a,(3-unsaturated carbonyl compounds furnished the corresponding cycloadducts in good diastereomeric excess. The exo-product was isolated as the major isomer in each case. The reaction of a variety of carbon nucleophiles, including a Grignard reagent, with the nitrones led to the formation of interesting p-C-glycoside analogues. One of the carbohydrate nitrones was also converted into its thymine C-nucleoside analogue. This work clearly shows that the construction of chiral cyclic nitrones from D-ribose derivatives is an extremely efficient and simple procedure. The final part of the work described in this thesis involve the construction of CD-ring analogues of the natural metabolite, streptonigrin. The use of metalation and palladium catalysed cross-coupling reactions were investigated for the synthesis of highly functionalised biaryls. Pharmacological research is essential for the advancement of treatment therapies to combat diseases that plague mankind. Pyrimidines have been a subject under investigation by medicinal chemists for many years due to their interesting pharmacological properties. In previous studies, pyrimidines and their derivatives have been reported to have antimicrobial, anti-inflammatory, antimalarial, analgesic, and antitumour activities amongst other biological activities. Although there has been a significant amount of research carried out on these heterocycles, there will always be a continuous need for the discovery of novel synthetic drugs which have a higher degree of potency and fewer side effects. Hence, this study was undertaken to determine the pharmacological activities of eight novel 1, 4 dihydropyrimidine analogues (DHPM 1 – 8), that have been synthesized in our laboratory. The dihydropyrimidines were synthesized and characterized and thereafter evaluated for in vitro antimicrobial, antioxidant, anti-inflammatory, cytotoxicity and apoptotic activities. The compounds also underwent a safety study. Antimicrobial activity was evaluated using the disk diffusion assay; compounds displaying superior activity were subjected to further analysis to establish the minimum inhibitory concentration. Overall compounds DHPM 7 and 8 showed the best antibacterial activity against Gram positive bacteria. The minimum inhibitory concentration (MIC) for DHPM 7 against the Gram positive organisms (B.cereus, S.aureus and B.coagulans) was 0.75 µg/mL; however DHPM 7 had a MIC of 0.37 µg/mL against M. luteus. DHPM 8 displayed an MIC of 0.75 µg/mL against B.cereus, S.aureus, M.luteus, S.faecalis and B.coagulans. Antioxidant activity was assessed using the DPPH method. DHPM 2 showed outstanding free radical scavenging capacity of 90.63% at a concentration of 1 mg/mL. The DHPM 1 - 8 were analysed for their lipoxygenase inhibitory activity. Excellent inhibition ranging from 59.37 ± 0.6 to 81.19 ± 0.94% was demonstrated. The inhibitory activity was elucidated by a molecular docking study against the lipoxygenase enzyme (PDB code = 3V99) using the MOE 2013.08 and Leadit 2.1.2 software and high affinities were demonstrated. DHPM 1 - 8 were tested for cytotoxic activity against two human cancer cell lines, MCF-7 and UACC-62 by means of the MTT assay. It was observed for the MCF-7 cell line, DHPM 1, 4, 6, 7 and 8 displayed cytotoxicity above 89% at 50 µg/mL. The DHPMs at 50 µg/mL were noted to be very effective against the Melanoma cell line with DHPM 2 having a cytotoxicity value of 82.62% and DHPM 1, 4, 5, 6, 7 and 8 exhibiting cytotoxicity greater than 96%. ▼ This investigation has been geared towards several aspects of chromone chemistry. Selected 2-(N,N-dimethylarnino)chromones have been synthesized via 2-hydroxyacetophenone boron difluoride complex intermediates, and potentiometric analysis of these compounds in ethanolwater has been used to determine the influence of substituents on their basicity. The pKa values have been found to lie within a narrow range (1.92 - 2.52), and the observed substituent effects have been rationalized with the aid of semi-empirical and ab initio molecular orbital calculations. An efficient route has been developed for the synthesis of the naturally-occurring chromone, "granulosin" [7,8-(methylenedioxy)-2-propylchromone], and several C-2 side chain analogues in good yields, by condensing 2'-hydroxy-3',4'-(methylenedioxy)acetophenone with a range of ethyl carboxylate esters. These compounds show significant cytotoxic activity against the brine shrimp, Artemia salina, and two of them, the 2-ethyl and 2-benzyl derivatives also show 100% activity as pesticides on Beet army worms (BAW). Another naturally-occurring chromone derivative, 5-hydroxy-2-isopropyl-7-methoxychromone, and four C-2 side chain analogues have been prepared in moderate yields. These compounds also show significant cytotoxic activity against the brine shrimp, Artemia salina, and it is apparent that the presence of the hydroxyl group at C-5 is critical for such activity. The electronimpact mass spectra of both series of chromone derivatives have been investigated, permitting the elucidation of characteristic fragmentation patterns. In work directed towards the synthesis of potential HIV -1 protease inhibitors, five novel chromone-containing analogues of the clinically useful drug, ritonavir, have been synthesized. The design strategy has involved the coupling of substituted chromone-2- carboxylic acids with a specially prepared, hydroxyethylene dipeptide isostere to afford ritonavir analogues containing chromone termini. An interactive docking procedure has been used to explore the docking of ritonavir and the novel chromone-containing analogues into ' the active site of the enzyme, and has indicated the capacity of the ritonavir analogues to form hydrogen-bonds with the HJV-l enzyme receptor. Various substituted cbromone-3-carbaldehydes, which have been synthesized from the corresponding o-hyclroxyacetophenones using Vilsmeier-Haack methodology, have been examined as substrates for Morita-Baylis-Hillman reactions, using 3-hyclroxyquinuclidine as the catalyst and arcylonitrile and methyl acrylate as the activated alkenes. Optimization of the reaction conditions has permitted efficient conversion of the cbromone-3-carbaldehydes to the Morita-Baylis-Hillman products and, in some cases, dimeric products, within 24 h. Heating of the Morita-Baylis-Hillman products, arising from reactions with methyl acrylate, at 80 ºC for 3 h in the presence of DABCO as catalyst, has been shown to effect transformation to the corresponding dimers in good yield. ▼ 7,7'-Dihydroxy-8,8'-biquinolyl (azaBINOL) was prepared from 7-hydroxyquinoline via N,N-dimethyl O-quinol-7-yl carbamate by directed ortho-metallation followed by FeCl₃-mediated oxidative coupling of the 8-lithioquinoline intermediate. Saponification of the resulting dicarbamate provided crystalline (±)-azaBINOL in 56% overall yield. 6,6'-Bis(dimethylaminocarbonyl)-7,7'-dihydroxy-8,8´-biquinolyl was prepared from the same dicarbamate by a double anionic-Fries rearrangement in 43% yield. 7,7'-Bis(diethylamino-carbonyloxy)-6,6'-diiodo-8,8´-biquinolyl was synthesized from 7-(diethylaminocarbonyloxy)-8-iodoquinoline in 54% yield by a tandem halogen-dance/oxidative coupling process. The diiodide was converted to 6,6'-bis[(methylamino)-sulfonyl]-7,7'-dihydroxy-8,8´-biquinolyl by a three-step sequence in 91% overall yield. Resolution of (±)-azaBINOL into its atropisomeric enantiomorphs was accomplished in two ways: (a) via chromatographic (HPLC) separation of diastereomeric bismenthylcarbonates generated from the racemic diol and homochiral menthylchloroformate, and (b) via hydrolytic enzymatic kinetic resolution of the (±)-azaBINOL dipentanoate ester derivative with bovine pancreas acetone powder. Method (b) was superior and afforded the unreacted (+)-(aR)-diester (46%, ≥99%ee) accompanied by (–)-(aS)-azaBINOL (30%, 92%ee after recrystallization). 2,2'-Di-tert-butyl-azaBINOL was prepared in enantioenriched form from scalemic samples of azaBINOL-dipentanoate by addition of t-BuLi followed by saponification in the presence of air. First-order rate constants for the enantiomerization of azaBINOL in H₂O were measured (over 316-366 K) and activation parameters determined as ΔH‡ = 34.0 kcal mol–1 and ΔS‡ = 18.7 cal mol⁻¹ K⁻¹ by Eyring plot analysis. The further functionalized 2,2'-di-tert-butyl-azaBINOL was found to be configurationally labile and showed racemization half-lives of 30.5 h and 1.9 h at 23 °C in MeOH and CHCl₃, respectively. A series of cyclic diethers were prepared from azaBINOLs and dihalides X(CH₂)nX' (n = 1-6) and their pKa values determined by potentiometric titration in MeOH. Biquinolyl basicity was dependent on interannular dihedral angle (IDA; assessed by X-ray crystallography and UV spectroscopy), the methylene diethers (IDA ≤60°) exhibited single pKa points of exceptionally low magnitude (2.7-3.1). 6,6'-Bis[(methylamino)sulfonyl]-7,7'-dihydroxy-8,8´-biquinolyl, resolved via an HPLC based method, catalyzed the enantioselective addition of TMSCN to aldehydes in the absence of additives. Resulting cyanohydrins were obtained in ≥ 90% yield and up to 20%ee. Control experiments established the requirement of sulfonamide functionality, free hydroxyl groups, quinoline nitrogen atoms and the dimeric manifold for catalytic activity. ▼ The tricyclic indole and tetracyclic quinoline heterocyclic systems were chosen from Naresh Kumar (NK) library in terms of their structural similarity to compounds identified from WEHI library screening. It was also of a particular interest to modify the structure of WEHI 0088846 and test the new analogues against the designated cancer cells. A suitable methodology was developed for the preparation of novel dihydropyranoindoles via the Hemetsberger indole synthesis of arylethers, derived from corresponding hydroxybenzaldehydes with activated alkynes, and a thermal cyclisation in high boiling solvents. Biological studies showed that tetracyclic systems had significant cytotoxic activity at higher concentration against the neuroblastoma cancer cells. More importantly, these systems, at the lower concentration, considerably enhanced the SAHA toxicity. Encouragingly, the toxicity of designated systems on the normal human cells was found to be significantly less than the cancer cells. The nucleophilic substitution of methyl hydroxyindole esters with Î±-haloketones yielded the arylether ketones and subsequent cyclisation resulted in tricyclic and tetracyclic furoindoles. The attempts to cyclize the corresponding intermediates in the presence of catalytic amount of acid generated the desired tricyclic and tetracyclic furoindoles. The additional mono cyclized furoindoles with the non-cyclized methyl ketone were arised from methyl 4,6-dihydroxyindole-2-carboxylate. The individual cytotoxic efficency of furoindoles were found to be more favourable than the SAHA enhancement effect against the cancer cells. Interestingly, the non-cyclized intermediate provided the best IC50 value at very low concentration. The first synthetic procedure to prepare the original hit compound WEHI 0088846 was reported and the modifications on the three key regions of compound afforded a range of analogues in good yields. The new hydrazone-hydrazide compounds were found to be cell specific towards the neuroblastoma cells and two derivatives displayed very low micromolar potency against the neuroblastoma cells. The most reactive position of the methyl 5,6-dibenzyloxyindole esters was determined via the Vilsmeier Haack reaction and formylated products were used for the design of novel indole-containing imines. The tested imine analogues were shown to have a significant potential for the reduction of Kelly cell viability at a concentration of 10 Î¼M compound alone. This dissertation focuses on the gas phase properties and reactivity of two types of N-heterocyclic compounds: N-heterocyclic carbenes (NHCs) and nucleobase derivatives. Imidazole-2-ylidene and imidazolidin-2-ylidene are stabilized carbenes widely applied in organometallics and organocatalysis. The only structural difference between the two is the degree of unsaturation on their heterocyclic backbone. Imidazole-2-ylidene is unsaturated (denoted as unsatNHCs) while imidazolidin-2-ylidene is saturated (denoted as satNHC). Our studies focused on the proton affinities comparison between unsatNHCs and satNHCs. The results indicate that satNHCs are more basic than their unsaturated counterparts. They are both more basic than the first generation Grubbs catalyst ligand tricyclohexylphosphine. N-Heterocyclic carbenes (NHCs) catalyze Umpolung condensation reactions of carbonyl compounds, including the benzoin condensation and Stetter reaction. These types of reactions have not been examined in the gas phase. We explored the feasibility of examining these reactions in the absence of solvent. Charge-tagged thiazolylidene catalysts were used to track the reactions by mass spectrometry. We found that the addition of the NHC catalysts to a carbonyl compound to form the "Breslow intermediate" can only be accessed by introducing acylsilanes, which takes advantage of Brook rearrangement. The second step addition, with another aldehyde or a Michael acceptor, does not occur under our gas phase conditions. The differential reactivity between the condensed and gas phases was discussed. A light-responsive NHC was to have different electronic properties between its two isomers. We conducted a series of molecular orbital calculations to clarify the electronic structure difference between the two isomers. The DNA glycosylase MutY repairs the DNA damage by removing adenine from mismatched adenine:8-oxoganine base pair. To help understand the mechanism of MutY, gas phase proton affinities and acidities of four MutY substrates were measured. The measurement matches calculated values by our group. The tautomerization of protonated 6-methyl-1-deazaadenine was also discussed. It is hypothesized that nicotinamide adenine dinucleotide (NAD+) can initiate in vitro DNA transcription catalyzed by E.coli RNA polymerase (RNAP). To prove this hypothesis, a mixture of DNA template, E.coli RNAP, CTP and NAD+ were made, aiming to find the 5’-NAD+ modified RNA sequence NAD+pC. An LC-MS method was developed to analyze NAD+pC.

The thesis entitled Synthesis of biologically important functionalized nitrogen containing heterocyclic compounds is divided into ten chapters. Chapter 1 deals with general survey of literature on the importance and synthesis of the nitrogen containing heterocyclic compounds. Chapter 2 gives the objective of the work on synthesis of N-hetero cyclic compounds we planned to synthesize. The chapters 3, 4, 5, 6, 7 and 8 deals with the general survey of literature on the importance and synthesis of a particular group of nitrogen heterocyclic compounds, detailed methodology and experimental procedures followed for the synthesis of those heterocyclic compounds. In chapter 9 and 10 gives the details of the thesis summary and conclusion. Chapter 1: Introduction to N-Heterocycles This chapter describes an overview of available literature on the biological activity. Chapter 2: Aim and Objective This chapter describes an overview of the work on evaluation of N-heterocyclic compounds we chose for the synthesis and importance of green chemistry. Chapter 3: One-pot synthesis of polyhydroquinolines In this chapter we have discussed on the pharmacological importance of polyhydroquinolines and also reviewed on previous synthetic methods. An efficient and straight forward general method has been developed for the synthesis of polyhydroquinolines by simply combining aldehyde, alkyl acetoacetate, and#945;-keto compounds including 5,5-dimethyl-1,3-cyclohexanedione or 1,3-cyclohexanedione and ammonium acetate in the presence of a catalytic amount of Iron(III) fluoride in ethanol at 75-80 °C. The process presented here is operationally simple, environmentally benign and has excellent yields with short reaction times. Furthermore, the catalyst can be recovered conveniently and reused efficiently for multiple runs. Scheme 1. ▼ The reaction of salicylaldehyde with various activated alkenes, viz., methyl vinyl ketone, ethyl vinyl ketone, phenyl vinyl sulfone, phenyl vinylsulfonate, acrolein and acrylonitrile, under Baylis-Hillman conditions, has been found to proceed with the chemoselective formation of chromene derivatives. The reaction conditions have been optimised and chromene derivatives have been obtained in isolated yields up to 87 %. The generality of the reaction, using 1,4-diazabicyclo[2.2.2]octane (DABCO), as the catalyst, and a heterogeneous (chloroform-water) solvent system, has been established using a range of salicylaldehyde derivatives,. including 2-hydroxynaphthaldehyde. The formation of chromene derivatives, under these conditions, has been assumed to proceed via an initial, Baylis-Hillman reaction, followed by cyclisation involving intramolecular conjugate addition, and subsequent dehydration. Evidence supporting this sequence has been obtained from the isolation ofBaylis-Hillman products from reactions involving the use of tertbutylclimethylsilyl-protected salicylaldehyde, 4-hydroxybenzaldehyde and tert-butyl acrylate as substrates. The potential of the ''Baylis-Hillman zwitterion" to participate as a donor species in Michael-type addition reactions has been explored and a series of climeric products has been isolated. The Baylis-Hillman methodology has also been successfully extended to the synthesis of sulfurcontaining heterocyclic systems, and a range of 3-substituted thiochromenes has been obtained in moderate yields, using 2,2'-dithiobenzaldehyde and various activated alkenes in the presence of 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) as catalyst. The electron-impact mass spectra of selected chromene and thiocbromene derivatives have been investigated permitting comparison of the fragmentation of the oxygen- and sulfur-containing analogues. In a study directed at the synthesis of potential HIV -1 protease inhibitors, chromene- and thiocbromene-containing analogues of the clinically useful drug, ritonavir, have been prepared. Thiochromene and chromene derivatives were converted to the corresponding 3 -carboxylic acids and coupled with a specially prepared, hydroxyethylene dipeptide isostere to afford ritonavir analogues containing cbromene and thiochromene termini in ca. 60% yield.

Coumarins are naturally occurring α-benzopyrone derivatives known for their pharmacological properties such as anticoagulant, antimicrobial, anticancer, antioxidant, anti-inflammatory and antiviral properties. The pharmacological, biochemical, and curative applications of coumarins depend on the substitution around the coumarin core structure. In the present study, seven halogenated coumarins CMRN1 - CMRN7 were synthesized and evaluated for mosquito larvicidal, repellancy , and insecticidal activity against Anopheles arabiensis. Furthermore, the antimicrobial properties of compounds CMRN1 - CMRN7 were evaluated by assessing the bacterial and fungicidal activities using the disc diffusion method. The anti-inflammatory properties were evaluated using the 5-lipoxygenase kit assay. The evaluation of the safe use of the compounds was determined using the Brine shrimp lethal test. The potential carcinogenic properties of the studied compounds was done using the Salmonella mutagenicity test. The anti-cancer property of the studied compounds was evaluated against UACC62 (Melanoma), MCF-7 (Breast cancer), and PBMC (Peripheral blood mononuclear) cell lines using of MTT assay. The apoptotic potential of the synthesized coumarin was evaluated against UACC62 (Melanoma) cell by assessing their morphological changes, membrane change, mitochondria membrane potential, and caspase-3 activity using the Annexin V-PI staining, JC-1, caspase-3 enzyme kits, respectively, on flow cytometer. The results were compared to a known anti-cancer drug, doxorubicin. The results showed that compounds CMRN1, CMRN2, CMRN4, CMRN5 and CMRN7 exerted 100% larval mortality within 24 h of exposure. All halogenated coumarins reversibly knocked down adult mosquitoes but did not kill them after 24 h of exposure. Furthermore, the adulticidal activity of the compounds was considered only mild to moderate. The antimicrobial activity of the synthetized coumarins CMRN1 - CMRN7 were assessed against E. coli, K. pneumoniae, S. marcescens, S. faecalis, B. cereus, B. coagulans, B. stearothermophilus, C. freundii, S. aureus and M. luteus bacteria and three yeast cultures, C. albicans, C. utilis, S. cerevisiae as well as two fungal species, A. flavus and A. niger. Compounds CMRN1 and CMRN2 showed bacterial growth inhibition for all the tested species except K. pneumonia and B. stearothermophilus. Compounds CMRN4 and CMRN7 showed moderate bacterial inhibition against B. cereus, M. luteus and S. aureus. The anti-inflammatory activity of the coumarins analogues showed that 1 mg/mL of the compounds CMRN1, CMRN2, CMRN4 and CMRN5 displayed moderate anti-inflammatory activity when compared to the positive control, 15-lapoxygenase. Piperidine derivatives are essential heterocyclic compounds that have beneficial roles in the medical and commercial sector. They can be isolated from plant material and can be chemically synthesised using simple cost efficient methods. Piperidines and their derivatives are clinically used to prevent postoperative vomiting, facilitate radiological evaluation, correct gastrointestinal function as well as speed up gastric emptying before anaesthesia. Piperidine derivatives also demonstrate a wide spectrum of biological activities which include; antimicrobial, anticancer, anti- TB, anti-HIV, anti-inflammatory, analgesic, anti-influenza, anti-inflammatory and antitumor activity. The properties of piperidine derivatives depend on the nature of the side chains and their orientation. Based on the promising data that demonstrated the synergistic effects of biological agents with piperidine derivatives, the aim of our research is to determine the pharmacological activities, i.e. (i) antimicrobial activity, (ii) anti-inflammatory, (iii) anti-oxidant activity, (iv) cytotoxicity, and (v) biosafety of six piperidine derivatives, PM1 to PM6. All six piperidine derivatives (PM1-PM6) screened for antimicrobial activity exhibit characteristics of varying degrees of microbial inhibition against some Gram-positive and Gram-negative bacteria (B. cereus, B. subtilis, E. coli, S. aureus, Kl. Pneumonia, M. liuteus and P. aurenginosa) with the exception of B. polymixa, S. marcescens and S. faecalis. Certain piperidine derivatives did not demonstrate high inhibition activity towards the fungal strains, with inhibition only shown against four fungal species; A. niger, A. flavus, C. albicans and S. cerevisiae. Thus it is proposed that minor changes could be made to the structure of the compounds so that they can alter the effect that the compounds have on the specific fungi strains. With regard to antioxidant activity it is noted that the concentrations of the test compounds are directly proportional to the percentage of scavenging capacity. In comparison of the piperidine derivatives (PM1-PM6) to Rutin (reference standard), it was illustrated that Rutin displayed the best antioxidant activity. All six piperidine derivatives (PM1-PM6) showed greater than 50% anti-inflammatory activity, whilst the anti-inflammatory reference standard NCGA displayed the greatest activity in comparison to the piperidine derivatives tested. The safety of the piperidine derivatives was tested by assaying cytotoxicity, against melanoma, MCF7 cancer cells and normal fibroblasts as well as Brine shrimp lethality assay. All piperidine derivatives demonstrated high cytotoxicity activity against both cancer cell lines (melanoma and MCF7) and around 50 – 52% cytotoxicity against healthy cells.
