細菌のheat shock proteins(HSPs)は,細胞内で分子シャペロンとして働くのみならず,細胞外に分泌され接着分子として働くことや,宿主免疫細胞のレセプターに結合し,様々な免疫反応を引き起こすことも近年の研究で明らかにされている.今回,Prevotella intermedia(P.intermedia)が菌体外多糖を産生してバイオフィルムを形成する際のHSPsの役割について検討するため,まず,リコンビナントGroEL(rGroEL)を用いて家兎抗GroEL抗体を作製し,P.intermedia GroELが菌体外に放出されていることをウェスタンブロッティングにて確認した.バイオフィルム形成に及ぼす影響については,rGroEL,リコンビナントDnaK(rDnaK)タンパクを用いて,crystal violet microplate assayで検討した.また,P.intermediaのバイオフィルム形成調節遺伝子を明らかにするため,バイオフィルム形成株と非形成株間で,GS-Junior system次世代シークエンサーを用いた変異解析もあわせて行った.特異抗体を用いたウェスタンブロッティングの結果,P.intermedia strain 17の培養上清にはGroELが含まれることが確認された.バイオフィルムアッセイでは,培地に加えたrGroEL,rDnaKは,バイオフィルムを形成するstrain 17のバイオフィルム形成を完全に抑制した.逆に,バイオフィルムを形成しないstrain 17-2とATCC 25611は,これらのHSPs添加培地中で明瞭なバイオフィルム形成性を示した.rGroEL,rDnaKでコートしたプレート上では,strain 17-2のみが凝集塊をつくってバイオフィルムを形成し,strain 17,ATCC 25611はバイオフィルムを形成しながった.Strain 17-2のゲノム配列の変異解析を行った結果,chromosome I上のTaql-like C-terminal specificity domain proteinとアノテーションされたPIN 17_0430遺伝子と,glucose/galactose transporter遺伝子(PIN 17_A 1569)に変異が確認された.今回の研究では,P.intemediaのGroEL,DnaKがstrain 17のバイオフィルム形成を抑制し,バイオフィルム非形成株のポリスチレンプレートへの接着を促進したことから,P.intermediaが外部環境に定着し,バイオフィルム形成を開始する際に重要な役割を果たしていることが示唆された.バイオフィルム非形成株と親株のゲノムマッピングでは,2つの遺伝子で変異が確認された.これら遺伝子の詳しい機能は不明であるが,P.intermediaバイオフィルム形成に関与するものと考えられる.
