Branchiuran crustaceans are fairly common parasites of freshwater fishes in Africa and are presently represented by not more than 200 described species belonging to four genera, namely: Argulus Miiller,1785; Dolops Audouin,1837; Chonopeltis Thiele,1900; and Dipteropeltis Calman,1912. Chonopeltis is, however, the only one of these genera which is endemic to Africa and species of this genus have been reported from a number of different freshwater fish hosts and localities throughout the Ethiopic region. Although 13 species of Chonopeltis have hitherto been described, lack of morphological detail and morphometric data in taxonomic and subsequent descriptions, has led to confusion and much controversy as to, for instance, the phylogenetic origin, identity and function of various structures. Consequently, the validity of the taxonomic identity or status and affinities of the various species are questionable. Lack of important morphological information has also caused a considerable number of gaps and deficiencies in our present understanding of various aspects of ecological and pathogenetic significance. In the present study, a comprehensive investigation on various aspects of the morphology, anatomy and histology of adult Chonopeltis specimens was conducted, using not only light microscopy, but also scanning electron microscopy (SEM), histological microtomizing and staining techniques and graphic reconstruction. This investigation is based on several specimens of Chonopeltis victori Avenant-Oldewage,1991 collected during surveys of freshwater fish parasites in 1990 and 1991 from four different fish hosts, namely: Labeo rosae S teindachner,1894; L. congoro Peters, 1852; L. ruddi Boulenger,1907 ; and Barbus marequensis A. Smith,1841, sampled at various localities in the Olifants River in the Kruger National Park, as well as several C. australis Boxshal1,1976 specimens found on L. capensis (A. Smith,1841) and L. umbratus (A. Smith,1841), sampled in Boskop Dam near Potchefstroom. By making use of the mentioned methods of investigation a more complete and reliable morphological description of the distinctive external structures, as well as the internal structures of the cephalic shield alae and especially the reproductive system of these parasites could be given. With the aid of semi-thin (2am) serial sections and graphic reconstruction the various components of the reproductive systems could clearly be identified and the mutual relations and histomorphology of these components be described, discussed and compared with those of other members of the Branchiura. In an attempt to determine the true taxonomic position and relationships of the recognised species of Chonopeltis, the original descriptions, morphometric data, geographical distribution, hosts and affinities are compared, summarised and discussed. The results of the present study reveal a horseshoe-shaped furrow dorsally that separates the alae from the cephalon. ▼ Cystic ovarian disease (COD) is an anovulatory condition in cattle that afflicts between 6-18% of all dairy cows in the US. Ovulation is dependent on the plasminogen activator (PA) family of proteases and protease inhibitor for proteolysis, culminating in follicular rupture. Failure of the follicle to ovulate suggests an aberration in proteolysis in cystic follicles. Polycystic ovarian disease in humans is phenotypically similar to COD is associated with elevated plasma PAI-1. PAI-1 regulates the proteolytic activity responsible for ovulation. Vitamin E supplementation reduces plasma PAI-1 and reduces the incidence of COD. Four experiments were conducted to elucidate the role of the plasminogen activator system in the etiology of COD and the ability of vitamin E supplementation to modulate the PA system in cattle. In experiment one, blood samples were collected from lactating dairy cows upon diagnosis of a follicular cyst at least 3.0 cm in diameter by a licensed veterinarian. Cows were classified based upon their history of follicular cysts. Tissue-type plasminogen activator (tPA) and plasminogen activator inhibitor-1 (PAI-1) concentrations and activities were measured using commercially available ELISA and chromogenic reagents. No differences were detected between cystic and non-cystic dairy cows. In experiment two, six non-lactating beef cows were supplemented with 2750 IU of α-tocopherol every four days for 24 days. Blood samples were collected every two days and assayed for tPA and PAI-1 concentrations and activities. Supplementation with α- tocopherol decreased the ratio of PAI-1 to tPA activities adjusted to the Day 0 measurement (P=0.097). In experiment three, mRNA was isolated from follicular cysts and preovulatory follicles and relative quantitative RT-PCR was performed for tPA, urokinase-type plasminogen activator (uPA), urokinase-type plasminogen activator receptor (uPAR), PAI-1, and β-actin. uPA expression was reduced (P<0.1) and uPAR expression was greater (P<0.1) in follicular cysts compared to preovulatory follicles. Lastly, in experiment four DNA was extracted from whole blood obtained from cystic and normal dairy cows and the promoter region of the PAI-1 gene was sequenced. More Jersey cows with COD possessed a four basepair deletion polymorphism than normal cows (P<0.1) and more Jerseys possessed the deletion polymorphism than Holsteins (P<0.01). ▼ Background Human papilloma virus (HPV) has been associated to conjunctival squamous cell carcinoma (CSCC) and has been documented in studies originating from different parts of the world. In Zambia, eye cancers are on the increase and between 2005 and 2009 they were ranked number three (3) among the top five (5) cancers of which conjunctival squamous cell carcinoma (CSCC) (78%) was the commonest. In this study, the aim was to establish the prevalence and association of HPV infection with CSCC at the University Teaching Hospital (UTH) and Lusaka Adventist Eye Hospital, Zambia. Materials and Methods This was a cross section study conducted at UTH and Lusaka Adventist Eye Hospital, Zambia for a period of three (3) months. A convenient sampling method was used. Conjunctival biopsies were collected from patients aged over 20 years with clinically suspected CSCC and met an inclusion criteria making the sample of 37 participants. Histopathology and polymerase chain reaction assay (PCR) analysis of 37 specimens of conjunctival lesions was undertaken. Tissues confirmed as CSCC (21) were grouped as cases and non-CSCC tissues (16) as controls. Findings were analysed using statistical software SPSS for windows version 18.0. Univariate analysis of variables which were CSCC, HPV, and HIV was conducted to determine the distribution patterns of HPV and HIV. Odds ratios (ORs) and 95% confidence intervals (CIs) were computed stratifying by age, sex, human papilloma virus and HIV status. Results This study revealed that, 59.5% of the conjunctival tissues were malignant of which 95.5% were CSCC and 4.5% were Kaposi’s sarcoma. Most of the participants 57% were HIV positive. 71.4% of patients with CSCC were HIV positive and HPV DNA was detected in 71.4% of CSCC tissues. HPV DNA was detected in 52.4% HIV+ patients and in 62.5% HIV- patients. Conclusion The study showed a high prevalence of HPV infection (71.4%) in CSCC. HPV and HIV infections were both associated with conjunctival SCC. Though HPV infection was found in patients with HIV, there were no significant differences in the results between HIV positive and HIV negative patients.

▼ During differentiation, the testes of Tenebrio molitor have been found to exhibit increases in biosynthetic capacity reflected in alterations in testicular protein and RNA. This biochemical differentiation was influenced by endogenous and/or exogenous hormones. The testes underwent dramatic increases in size and weight during the prepupal stage that were continued through later developmental stages. Histological analysis revealed that the maturation process of the germ cells to produce spermatozoa proceeded from the distal end of the follicles and toward the basal region to form a "differentiation wave." Spermatozoa were found in the prepupal testes. The underlying biochemical machinery of the developmental process was found to be accelerated in manufacturing different elements for germ cell differentiation at certain stages and particularly when the endogenous level of ecdysterone rose during the late prepupal and at mid-pupal stages. Gradual increases in testicular protein and RNA content were observed during the prepupal stage. The observed increases were more dramatic for both protein and RNA content in the pupal stage. The testicular protein and RNA content reached their maximum levels between days 4 and 7 of the pupal stage as did the rate of 3H-leucine incorporation. During the adult stage, the biosynthetic processes for producing protein and RNA were apparently reduced following the first few days after adult emergence. The protein products of the mealworm testes were shown by gel electrophoresis to be many and diverse. The 27 protein products were of various molecular weights, ranging from 12,000 to 127,000 daltons. These products were present at different ages of development and persisted for various times indicating that some of these proteins may be necessary for the formation of specific germ cell types. In addition, a variety of these testicular protein components incorporated leucine at measurable levels throughout development, particularly during the pupal stage. It was ascertained that the rate of incorporation of radioactive leucine into TCA -precipitable testicular protein was not affected by the administration of exogenous juvenile hormone alone (JHI, 1 µg/animal) during the pupal stage. However, the administration of exogenous ecdysterone (0. 5 µg/animal) to pupal Tenebrio resulted in an increase in the rate of radioactive leucine incorporation into TCA -precipitable testicular proteins, particularly during the first six days after pupal ecdysis. The amount of ecdysterone injected appeared to stimulate the production of the same testicular protein components that were present during normal pupal development. Injection of a higher dose of ecdysterone (1.5 µg/animal) during some of the pupal ages appeared to alter the testicular differentiation program by enhancing the incorporation of leucine into not only the age -specific testicular protein components but also into new protein components which did not normally appear at these specific ages. ▼ DNA sequence data from male reproductive genes in numerous taxa have shown that these genes typically evolve more rapidly than other genes, often as a result of directional selection. In the genus Drosophila, the rapidly evolving male accessory gland protein genes (Acps) of melanogaster subgroup flies have contributed to this observation. Acps are small proteins that are transferred to females during mating as a major component of the seminal fluid and are considered agents of chemical communication between the sexes. Acps are known to contribute to normal ovulation and sperm storage, as well as increase oviposition rates and reduce female receptivity. Thus, Acps are considered likely targets of directional selection because of their potential roles in postcopulatory sexual selection and antagonistic coevolution between the sexes. Outside of melanogaster subgroup Acps, little is known about the evolutionary biology of male reproductive genes in Drosophila. For example, the male testis contains a richly diverse transcriptome but no studies have explored the evolutionary dynamics of a large set of testis-expressed genes. If clear differences in the evolutionary dynamics of different classes of male reproductive genes exist, empirical documentation of these differences will help identify the specific evolutionary forces at work. Additionally, mating systems differ between Drosophila species, potentially affecting the evolutionary dynamics of Acps across lineages. Comparative analyses of Acps from species with different mating systems are needed to address this issue. Finally, if Acps are generally rapidly evolving in Drosophila species, comparative analyses of orthology and Acp gene loss/gain are needed to determine how Acps respond to persistent directional selection across lineages. The data presented here aim to address these questions. Included are polymorphism and divergence data from 56 genes of Drosophila arizonae and D. mojavensis, repleta group species with mating systems that differ dramatically from melanogaster subgroup flies. The sample includes 19 Acps, 31 testis-expressed genes, and six more evenly expressed genes. Comparative genomics analyses of D. melanogaster-D. mojavensis male reproductive genes and D. melanogaster-D. pseudoobscura Acps are also presented to address questions of functional conservation across lineages. ▼ The paired preputial glands of the albino mouse, Mus musculus, (Swiss strain) were studied with regard to histology, physiology and basic histochemistry. For the histological study of the development and subsequent aging of the preputial gland, the glands of male and female mice of different ages, ranging from newborn to 12 months old, were used. In order to study the effects of androgens and estrogens upon the macroscopic and microscopic structure of the preputial gland, white Swiss mice were divided into four major groups (according to age and status) each of which were, in turn, divided into subgroups a, b, c, and d for treatment; mice in subgroups a and b received daily subcutaneous injections of testosterone propionate and of estradiol benzoate respectively; mice in subgroup creceived daily subcutaneous injections of the vehicle, sesame oil; mice in subgroup d served as controls. There were, of course, male and female mice in each grouping. Routine histochemical tests for lipids and proteins were carried out on the excised preputial glands of weanling and of young adult mice, of both sexes, in order to localize and to identify the secretory products of the acinar cells. Histologically and physiologically (in terms of response to androgens and estrogens) the preputial glands of the mouse are similar, in most respects, to those of the rat as described by other authors. However, there are apparent differences in the microscopic anatomy and in the histochemistry of these homologous organs; firstly, the presence of an intimate association of the mouse preputial gland with a central hair follicle which persists throughout the lifetime of the animal and which is not found in the rat gland; secondly, the absence of perinuclear proteinaceous granules in the acinar cells of the mouse preputial gland which are present in the acinar cells of the rat preputial gland, in addition to lipid droplets. Lipids, primarily in the form of droplets of neutral fats are, to my knowledge, the only secretory products of the mouse preputial gland. Developmentally, histologically, and histochemically, the preputial gland of the mouse is an excessively developed sebaceous gland and unlike the rat preputial gland which cannot be categorized so simply, belongs to the "monocrine" variety of holocrine gland.

▼ LIN-12/Notch is a conserved transmembrane receptor that is required during animal development for proper cell-fate decisions and specification. In Caenorhabditis elegans, activation of LIN-12 occurs through binding to ligand expressed by an adjacent cell. This binding event triggers two cleavage steps and results in the release of the LIN-12 intracellular domain [LIN-12(intra)], which translocates to the nucleus to form a ternary complex with two other proteins: LAG-1/Su(H)/Cbf1 and SEL-8/Mastermind/Mastermind-like. This ternary complex will then transcriptionally activate target genes via LAG-1 Binding Sites (LBSs). LAG-1 is the sole DNA-binding component within the complex, and in the absence of LIN-12(intra), can act as a transcriptional repressor. LIN-12 signal transduction can be studied in the C. elegans Vulval Precursor Cells (VPCs), which exhibit precise spatiotemporal patterning regulated by LIN-12 activity. Here, I show that LAG-1 is positively autoregulated by LIN-12 activity in cells where LIN-12 activity is high. Autoregulation is mediated by an enhancer element that contains a cluster of 18 LBSs that are located within a conserved high occupancy target region, which is a span of DNA that is pulled down promiscuously in ChIP-Seq experiments. Mutation of the LBSs abrogates preferential expression mediated by the enhancer in cells with high LIN-12 signal transduction. When the HOT region is deleted from the endogenous lag-1 locus, expression in the VPCs is strongly reduced and no overt Lag phenotype occurs. Instead, cold-sensitive vulval and egg-laying defects, reminiscent of phenotypes seen in lin-12 hypomorphs, are found. Autoregulation of lag-1, therefore, appears to contribute to the robustness of LIN-12 cell fate specification in response to stochastic environmental and genetic perturbations. Under adverse environmental conditions, C. elegans enter a state of diapause in which they form dauer larvae, which are long-lived and stress-resistant. The VPCs of dauer larvae remain developmentally arrested indefinitely until favorable conditions are reintroduced. Experimentally, this arrest can be relieved by depletion of the Forkhead transcription factor DAF-16. I show that expression of the components of the LIN-12/SEL-8/LAG-1 ternary complex are downregulated during the L2d-dauer molt (prior to dauer entry) and that this downregulation is not relieved by DAF-16 depletion. Instead, DAF-16 depletion leads to resumption of LIN-12 signaling and expression of ternary complex only in completely formed dauer larvae. These observations suggest that DAF-16 is required for the maintenance but not the initiation of blocking LIN-12 signaling. The components of the ternary complex are required to effect LIN-12 signaling. This work contributes to better understanding how these components are regulated and how their expression can affect LIN-12 -mediated cell fate decisions.

Following initial infection, the bacterial pathogen ascends from the lower genital tract to the upper genital tract leading to pelvic inflammation and scarring of inflamed tissue. The scarring can lead to ductal obstruction, causing retention of fluid in the upper genital tract and ovaries, termed hydrosalpinx. This inflammation due to Chlamydia infection can lead to infertility. Despite the commonality of the disease and its severe consequences, the molecular pathogenesis and mechanism of pelvic inflammation following Chlamydia infection has yet to be elucidated. We investigated the molecular pathogenesis and its relation to host autophagy here. The ATG5 gene is required for autophagy. Our lab has previously shown via mouse model that deletion of the ATG5 gene in myeloid cells results in a significant increase in upper genital tract inflammation following Chlamydia infection. To further investigate the role of autophagy in Chlamydia pathogenesis, mice with deleted ATG5 gene in epithelial cells were intravaginally infected with Chlamydia. The infection was monitored through weekly vaginal swabbing and calculation of Chlamydia bacterial titers from each swab. After Chlamydia infection cleared, the animals were sacrificed, upper genital tract pathology was observed and cytokine analysis was conducted on the vaginal swabs that were taken throughout the infection. The p62 autophagy protein acts as a scaffold to bring bacterial pathogen to autophagy machinery. In the current study, p62-deficient mice were obtained and infected with Chlamydia. Vaginal swabbing throughout Chlamydia infection allowed us to estimate bacterial titers and assay the host cytokine response to the infection, and finally the upper genital tracts of the animals were removed for observation. We found no significant difference in the pathology, bacterial clearance, or cytokine analysis between epithelial Atg5-deficient animals and wild type animals. This suggests that the Chlamydia bacteria may be employing mechanisms to evade or inhibit host autophagy in epithelial cells to promote its own survival. Additionally, there was no significant difference in the pathology, bacterial clearance, or cytokine analysis between the p62-deficient animals and wild type animals. Resumo: Apesar de Formicidae constituir um dos grupos animais mais significativos em riqueza de espécies no Brasil, suas relações filogenéticas permanecem controversas. Uma notável fonte de caracteres para análises taxonômicas e filogenéticas é a caracterização espermática e do aparelho reprodutor. Tal caracterização tem se mostrado uma ferramenta na clarificação de questões da biologia reprodutiva e sistemática de diversos grupos de insetos. Desta forma, o presente estudo analisou, através de microscopia de luz e eletrônica de transmissão, a morfologia espermática de quatro espécies do gênero Pachycondyla (Formicidae: Ponerinae), bem como o aparelho reprodutor masculino: P. marginata, P. termitofane comutata, P. harpax e P. striata. A ultraestrutura dos espermatozoides de tais espécies é semelhante à descrita para outros Hymenoptera por apresentar: (1) acrossomo cônico do tipo bicamada; (2) adjunto do centríolo situado entre o núcleo e ambos os derivados mitocondriais e (3) flagelo constituído por dois derivados mitocondriais alongados, dois corpos acessórios triangulares e um axonema de padrão 9+9+2. Entretanto, as mesmas compartilham inúmeras características cujas evidências indicam evoluir como homoplasia via paralelismo, não conferindo, pois, suporte para uma resolução filogenética entre tais espécies e demais clados próximos. Dentre estes caracteres se destacam a organização espacial dos componentes acrossomais, o caráter cromatínico nuclear, a configuração da extremidade da transição núcleo-flagelo, além da condição simétrica do adjunto do centríolo. Além dessas, várias outras características corroboram a definição do estado parafilético do respectivo gênero, tais como o aspecto cromatínico, a condição simétrica do adjunto do centríolo e a organização terminal flagelar, por serem compartilhadas entre determinadas espécies de Pachycondyla e grupos filogeneticamente distantes, evidenciando, pois, as divergências dentro da mesma espécie. Por fim, a análise morfológica do aparelho reprodutor de P. marginata e P. striata evidenciou a existência de um padrão morfológico reprodutivo para Formicidae, uma vez que tais espécies exibiram íntima similaridade às descrições realizadas à respectiva família

Resumo: A obesidade é um grave problema de saúde e entre os tratamentos indicados, destaca-se a farmacoterapia. Diversos medicamentos foram aprovados para este fim, entretanto, alguns apresentaram sérios riscos, inclusive para o sistema reprodutor, e foram retirados do mercado. Sabe-se que os receptores serotoninérgicos, como o 5-HT2C, desempenham um papel na função sexual masculina. A lorcaserina, fármaco agonista seletivo do receptor 5-HT2C, foi liberada recentemente como medicamento anorexígeno. Na literatura não existem dados sobre seus efeitos no sistema reprodutor masculino e na ejaculação. Esse trabalho avaliou, em ratos adultos Wistar, parâmetros reprodutivos, com ênfase sobre a qualidade espermática e mecanismos ejaculatórios, utilizando a lorcaserina como agonista. O estudo foi desenvolvido em etapas. Experimento I, piloto, com 4 grupos experimentais: controle (salina) e tratados com 0,5, 1 e 10 mg/Kg de lorcaserina nos ciclos de luz claro ou escuro, por 14 dias; Experimento II, com 4 grupos: tratados com lorcaserina, 10 mg/Kg por 60 dias e 30 mg/Kg por 30 dias, no ciclo claro, e seus respectivos grupos controle. Experimento III, estudo farmacológico in vivo e in vitro, o qual investigou os efeitos da lorcaserina na ejaculação. No experimento I não foram observadas alterações na maioria dos parâmetros, exceto uma diminuição no ganho de peso corpóreo no grupo tratado com 10 mg/Kg no ciclo claro e uma aceleração no tempo de trânsito dos espermatozoides na região da cabeça/corpo do epidídimo no mesmo grupo no ciclo escuro. No Experimento II houve uma diminuição no ganho de peso e no consumo de ração no grupo exposto à lorcaserina por 30 dias. Este mesmo grupo apresentou diminuição no peso da vesícula seminal cheia e vazia, bem como diminuição no número de espermatozoides e aceleração do tempo de trânsito na região de cabeça/corpo do epidídimo. O grupo exposto a 10 mg/Kg apresentou aumento no peso do fígado, diminuição no número de espermatozoides e aceleração do tempo de trânsito na região da cauda do epidídimo. Não houveram alterações na morfologia e contagem espermática nos testículos, bem como nos níveis de gonadotrofinas. Ambas exposições causaram dano à motilidade espermática e apenas os animais tratados por 60 dias apresentaram aumento na concentração de testosterona plasmática e intratesticular. No Experimento III, a lorcaserina não afetou a contratilidade dos órgãos reprodutores in vitro. In vivo, foi capaz de causar ejaculações completas sem estímulo sensorial no modelo anestesiado e na avaliação de penile grooming, e melhorou o desempenho de animais sexualmente experientes na cópula, diminuindo o limiar ejaculatório.
