A aspiração folicular transvaginal é um método não invasivo de obter oócitos de animais vivos. Devido à quase inexistência de estudos relacionados à técnica, na espécie eqüina ainda não se conhece bem seus efeitos sobre a fertilidade. Para avaliar o efeito da aspiração folicular sobre a fertilidade de éguas no ciclo subseqüente, foram realizados dois experimentos utilizando um ultra-som de 5,0MHz com transdutor setorial. No primeiro experimento, quinze éguas Crioulas serviram como controle e não foram aspiradas e outras quinze foram distribuídas em três grupos, de acordo com o tamanho do folículo aspirado: 25-29mm (n=4; Grupo 1); 30-34mm (n=6; Grupo2) ou >35mm (n=5; Grupo 3), todas na fase do estro. No segundo experimento, vinte e cinco éguas foram aspiradas durante o diestro e trinta e uma serviram como éguas-controle, não sendo aspiradas. O número de folículos aspirados por ovário variou de quatro a oito. Todos os folículos visualizados no monitor foram aspirados. A escolha do dia da aspiração foi determinada quando as éguas apresentaram acima de quatro folículos com diâmetro superior à 5mm em cada ovário. No primeiro experimento, as taxas de prenhez foram: 75,0% (Grupo 1), 83,3% (Grupo 2), 60,0% (Grupo 3) e 73,3% (Grupo 4 - Controle). No Experimento II, o grupo de éguas com folículos aspirados apresentou porcentagem de gestação de 76,0% e o grupo controle 77,4%. Nos dois experimentos, a taxa de prenhez das éguas controle e das que foram aspiradas foi similar (P>0,05). Esta pesquisa indica que a fertilidade das éguas no primeiro ciclo pós-aspiração não foi afetada pela técnica.

Transvaginal ultrasound-guided follicle aspiration is a non-invasive procedure for oocyte recovery from live animals. In equine medicine there are a few studies about this technique, so there is lack of information. Therefore, to evaluate the effect of follicular aspiration on subsequent fertility in mares, two experiments were conducted using an ultrasound with a setorial probe (5.0MHz). On the first experiment, fifteen Criollo mares were not aspirated and fifteen were assigned to one of three groups according to the diameter of the aspirated follicle: 25-29mm (n=4; Group 1); 30-34mm (n=6; Group 2) or >35mm (n=5; Group 3), all of them in oestrus. On the second experiment, the follicular aspiration was performed in twenty five mares during dioestrus and thirty one were used as control mares. The number of follicles aspirated was between 4 and 8 and all visualized follicles were aspirated. Follicle aspiration was performed when at least four follicles (>5mm) were present on both ovaries. In the Experiment I, the pregnancy rates were 75.0% (Group 1), 83.3% (Group 2), 60.0% (Group 3), and 73.3% (Group 4 - Control). In the Experiment II, pregnancy rates were 76.0% in aspirated group and 77.4% in control group. On both experiments, pregnancy rates were similar in treated and control mares (P>0.05). So far the study indicated that next cycle fertility was not affected by the follicular aspiration.

▼ Purpose: The purpose of the present study is to investigate the capability of the combination of Learning Vector Quantizer (LVQ) Νeural Νetworks (NNs) in the discrimination of benign from malignant thyroid lesions.Patients and Method: The study was performed on Liquid Based Cytology (LBC) specimens taken by FNA and stained by Papanikolaou technique. From the cytological images, using a custom image analysis system, certain features, describing the size, shape and texture of approximately 100 nuclei per case, have been extracted. These features were used to classify each individual nucleus by an LVQ NN. In the sequel, the nucleus classification results for each case were used to classify each individual case by a second cascaded LVQ NN. The cases were distributed according to the histological diagnosis as follows: 165 cases were classified as goiter, 2 as nodular hyperplasias, 61 as Hashimoto thyroiditis, 3 as non specific thyroiditis, 21 as adenomatoid nodules, 3 as oxyphic adenomas and 4 cases as follicular adenomas. Αdditionally, 62 cases of papilary carcinoma, 9 of medullary carcinoma, 2 of anaplastic carcinoma and 3 of follicular carcinoma, were included.The data of about 50% of the cases from each class, were used for the training of two LVQ classifiers and the remaining data were used to test their performance. The proposed system was used to discriminate into the individual cellular level and subsequently into the individual patient level, between benign from malignant nuclei or cases.Results: The application of the LVQ NNs system allows successful discrimination between benign and malignant cell nuclei and lesions (overall accuracy 94,1% and 100% respectively).Conclusions: The results indicate that the use of neural networks combined with image morphometry may offer useful information on the potential of malignancy of thyroid lesions and could improve the diagnostic accuracy of FNA of the thyroid gland, especially in cases of follicular neoplasms classified as suspicious for malignancy and in cases of oncocytic tumors.

▼ The effects of energy balance on hormonal patterns and recovered oocytes were evaluated in 20 lactating Holstein cows during two trial periods (fall/spring). Cows were randomly selected and assigned to one of two dietary treatments formulated so that cows consumed 3.6% BW (HE- 1.78 mcal/kg; n=6 in fall, n=5 in spring) and 3.2% BW (LE-1.52 mcal/kg; n= 5 in fall, n=4 in spring). Body weight and body condition score (BCS) were recorded prior to parturition and weekly throughout the fall trial. Ultrasound guided transvaginal follicular aspirations were conducted twice weekly between d 30 and 100 of lactation. Follicle size and number were recorded. Follicular fluid (FF) was aspirated from the largest follicle, and serum samples were collected for hormone assay (IGF-1; estradiol (E2); progesterone (P4, serum ); LH and FSH). Oocytes were collected and graded based upon cumulus density and ooplasm homogeneity, then fertilized and cultured in vitro. Milk yield averaged 41.64 Â± .3 kg/d (mean Â± SE) for HE and 32.8 Â± .3 kg/d for LE. There was a significant cubic day postpartum by treatment interaction for milk yield. Dry matter intake and BW treatment by week interactions were significant for the cubic and linear components, respectively. Oocyte numbers increased linearly from d 30 to 100 postpartum. HE cows produced more good + oocytes (1.5 Â± .2 ) than LE cows (1.4 Â± .1). Follicles less than or equal to 5 mm predominated throughout the study (6.4 Â± 3.0). However, greater numbers of follicles 10 to 14 mm and greater than or equal to 15 mm were found in the fall (1.98 Â± .08 and .50 Â± .06) than spring (1.11 Â± .3 and .23 Â± .07). Follicular fluid IGF-1 was higher in HE (2.3 Â± .2 ng/ml) than in LE cows (1.6 Â± .2 ng/ml). Mean basal serum FSH concentration was lower at 28 d postpartum (173 Â± 8 pg/ml) compared to later (521 Â± 25 at d 60 and 650 Â± 25 pg/ml at d 110). Serum P4 peaked at 35 d postpartum, with HE cows having 1 ng/ml higher P4 than LE cows. Current concepts for the manipulations of farm animals suggest that beyond the fulfillment of the primary scope of assisted reproduction - which in this case is the collection of oocytes of good quality from ewe-lambs - the welfare and the future reproductive performance of the donors should remain unaffected. Using as a model lambs of Karagouniko breed, the present thesis was mainly focused on the study of: (1) the follicular development at particular ages in ewe-lambs, (2) the ovarian response and oocyte recovery rate after singe or repeated laparoscopic follicular aspiration (OPU) at different ages, (3) the modification of the ovarian stimulatory protocols, (4) the meiotic competence of oocytes collected after single or repeated OPU, (5) the effects of follicular aspiration and ovarian stimulation in winter- or autumn-born lambs on their onset of puberty, body growth and future reproductive performance and (6) the feasibility to predict the lambs with good follicular development based on oestradiol concentration in blood samples collected during the stimulatory treatment. The results indicate that: (1) normal follicular development of Karagouniko lambs does not substantially differ from that of other breeds, (2) follicular development after ovarian stimulation using fixed FSH dose decreases with the age of donor-lamb. However, ovarian response is relatively stable when the FSH dose is adjusted to the body weight of lambs, irrespectively to the lambs’ age. (3) adjusting the administered gonadotrophin dose to lamb’s body weight, the number and the quality of collected oocytes can be much improved, (4) at the 3rd and 4th month of age the collected oocytes appear to be incapable for in vitro maturation. This did not stand true when an additional OPU preceded the one performed at the aforementioned ages, (5) both the hormonal treatments and the OPU technique - applied even consecutively - do not affect the onset of puberty, the body growth and the reproductive activity irrespectively of the season of birth and that (6) assessment of oestradiol concentration 12 hours before OPU is a safe criterion to predict good oocyte donor-lambs, although poor donors can be precluded as early as 12 hours after the start of the stimulatory treatment.

▼ Advanced maternal age has been shown to influence follicular and luteal dynamics in bovine ovary resulting in reduced fertility. The overall objective of the four studies presented in this thesis is to identify the maternal age-associated transcriptional changes in granulosa cells of the dominant follicles during follicle development. In the first study, mRNA expression levels of housekeeping genes were measured by real–time quantitative PCR (RT-qPCR) in granulosa cells of dominant follicles and FSH-stimulated follicles to select and validate suitable reference genes for relative gene expression analyses during maternal and follicular aging. Stability of six reference genes (GAPDH, ACTB, EIF2B2, UBE2D2, SF3A1 and RNF20) was analyzed using GeNorm, DeltaCT and NormFinder programs and comprehensive ranking order was determined based on these programs. Geometric mean of multiple genes (UBE2D2, EIF2B2, GAPDH and SF3A1) was more appropriate reference control than individual genes for the comparison of relative gene expression among dominant and FSH-stimulated follicles during maternal and/or follicular aging studies. In the second study, maternal age-associated changes in the transcriptome of granulosa cells recovered at the time of selection of the dominant follicle from aged (n=3) and young cows (n=3) were determined by EmbryoGENE bovine oligo-microarrays (EMBV3, Agilent Technology). The mRNA expression of five transcripts (CYP19A1, PCNA, GJA1, TPM2, and VNN1) was confirmed in a different set of granulosa cell samples by RT-qPCR to validate microarray data. A total of 169 genes/isoforms were differentially expressed (≥ 2-fold-change; P ≤ 0.05) in aged cows vs. young cows. These transcripts revealed inefficient 1) control of gonadotropins, and gonadotropin-induced changes in the cytoskeleton and extracellular matrix, 2) lipid metabolism and steroidogenesis 3) cell proliferation, cell cycle control and intercellular communication, and 4) higher oxidative stress responses in aged cows vs. young cows. In the third study, changes in the transcriptome of granulosa cells of the preovulatory follicle 24 h after LH treatment from aged (n= 3) and young (n=3) were determined. A total of 1340 genes were expressed differentially (≥ 2-fold change; P ≤ 0.05) in aged cows vs. young cows. The mRNA expression of five transcripts (RGS2, PTGS2, TNFAIP6, VNN1, NR5A2 and GADD45B) was confirmed in a different set of granulosa cell samples to validate microarray data. These transcripts were related to delayed 1) response to LH treatment 2) cellular differentiation and luteinization and 3) progesterone synthesis. Intra-follicle levels of progesterone were lower (P < 0.05) in aged cows compared to young and mid-aged cows. The fourth study compared the aged-associated changes in the transcriptome of granulosa cells during follicle development from the time of dominant follicle selection to preovulatory stage (24 h after LH). ▼ Delayed childbearing age, increased environmental pollutants accompanying modern life style, and an increase in inbreeding due to high demand for milk and meat yield have resulted in the decline in human and bovine reproduction respectively. Anti-Mullerian hormone (AMH) is expressed starting from 7-8 weeks of gestation in males and 20 weeks of gestation in females, in up to 100x higher concentration in males. AMH has been described as a non-invasive and accurate marker of ovarian quantitative fertility parameters in both humans and cattle. However, it is still unknown how AMH levels correlate to different fertility parameters in different cattle breeds; can AMH levels in the mother be used as a fetal sex marker during pregnancy in cows? In humans, can AMH collected in the single follicular fluid (mono FF), as well as granulosa cell (GC) AMH receptor 2 (AMHR2) be used as an embryo developmental potential marker? Here we present evidence of the higher levels of systemic and follicular AMH in Zebu compared to European cattle, corresponding to higher levels of AFC and oocytes. Cows pregnant with a male fetus, as compared to a female fetus, were observed to have a significantly different change in plasma AMH between day 35 and 135 during gestation. The placenta and cotyledon were found to express AMHR2 between day 38 and 80, however not significantly different between pregnancies with an opposite sex fetus. In the human study, systemic and average mono FF AMH, and average mono GC AMHR2 expression were significantly negatively correlated with the patient’s blastocyst rate. When patients were divided into normal and high groups based on the their systemic AMH levels, the following was observed: normal systemic AMH level patients had a lower average FF AMH level and a higher blastocyst rate compared to the higher systemic AMH level patients. Also, in normal systemic AMH patients, blastocysts corresponded to follicles with a lower level of AMH, while in high systemic AMH patients blastocysts corresponded to follicles with a higher level of AMH.
