▼ Phytosterols (PS) have become a recent popular medication alternative for treatment of hypercholesterolemia and have proven effective. Whether PS decrease risk of cardiovascular disease through other mechanisms, such as lipoprotein particle size and inflammatory markers, remains unclear. PURPOSE: The primary aim of this study was to examine the effects of two forms of PS in milk on lipoprotein particle size, inflammatory markers and
fat-
soluble vitamins. METHODS: Twenty subjects (13 males, 7 females; age; 55±6.1 years, height; 169±10 cm, weight; 77.9±16.9 kg, BMI; 27.3) consumed 16oz of cow’s milk daily for 12 weeks. The three sequential four-week phases consisted of 2% cow’s milk, 2 grams/day of ‘unaided’ PS in skim milk, and 2 grams/day of ‘aided’ triglyceride recrystallized PS (TRP) in

▼ In this work two voltammetric methods were investigated to study the interaction between metals, Cd and Pb, and fat-soluble vitamins (retinol palmitate VA; cholecalciferol VD; α-tocopherol acetate VE; filloquinona - VK). Due to hydrophobic characteristic of these vitamins, the voltammetric measurements were conducted in non aqueous media. In the first method (Method I) acetone was used was used as organic media, tetrabutylamonium iodide (ITBA) 0.2 mol L-1 as electrolyte and an electrode modified reference with double liquid junction - Ag/AgCl/LiCl saturated in ethanol/ITBA 0.02 mol L-1 in N,N - dimetylformamide (DMF). In the second method (Method II) ethanol was used as organic media, acetate buffer pH the 4.6 (in ethanol) as electrolyte and an electrode modified reference with double liquid junction - Ag/AgCl/LiCl saturated in ethanol/acetate buffer pH 4,6 (in ethanol). During the method development the behavior of Cd and Pb in studied conditions was evaluated. Each vitamin was studied individually. Method I was not the more adequate to study the interaction metal-vitamin because in most of the case the vitamin additions caused an increase on the voltammetric signals. Method II presented reduction of the Cd and Pb signals and the anodic shift of peak potentials in presence of the vitamins. The reduction of the signals increased in the order: VK <VD <VE <VA. High peak reduction was observed for Pb with the VA, VE and VD and for Cd with the VK. Measurements of alternating current voltammetry were also investigated to study the adsorption of vitamins on the electrode surface. Comparing the results obtained by the anodic stripping voltammetry with the results by alternated current, it was observed that the variations on the voltammetric signals are related predominantly with to adsorptions of VA and VD while to VE and VK the effect of adsorption was not predominant.
No presente trabalho dois métodos voltamétricos foram investigados para o estudo da interação entre os metais, Cd e Pb, e vitaminas lipossolúveis (palmitato de retinol VA; colecalciferol VD; acetato de α-tocoferol VE; filoquinona - VK). Devido à característica hidrofóbica destas vitaminas, a voltametria foi conduzida em meio orgânico. No primeiro método (Método I) utilizou-se acetona como meio, iodeto de tetrabutilamônio (ITBA) 0,2 mol L-1 como eletrólito e um eletrodo de referência modificado de dupla junção líquida Ag/AgCl/LiCl saturado em etanol/ITBA 0,02 mol L-1 em N,N dimetilformamida (DMF). No segundo método (Método II) utilizouse etanol como meio, o tampão acetato pH 4,6 (em etanol) e o eletrodo de referência modificado de dupla junção líquida Ag/AgCl/LiCl saturado em etanol/Tampão acetato pH 4,6 (em etanol). ▼ High multivitamin (10-fold AIN-93G levels, HV) intake by Wistar rats during pregnancy results in increased male offspring's body weight and food intake and characteristics of the metabolic syndrome. The hypothesis of this research is that fat soluble vitamins, primarily due to vitamin A, are partially responsible for this effect through modification of development of brain pathways regulating food intake and reward in utero and post-weaning. Three studies were conducted. In study 1, dams were fed AIN-93G diets containing the recommended vitamins (RV) or RV with 10-fold fat soluble vitamins (HFS). In study 2, the dams were fed RV, RV with 10-fold vitamin A (HA) or HV diet with only the recommended vitamin A diets. Male offspring were weaned to a high fat diet for 14 weeks. In study 3, dams were fed the HV diet, while their male offspring were weaned to RV, HV or HA diets for 29 weeks. Study 1 showed that the HFS diet does not reproduce the effects of the HV gestational diet on body weight or food intake of the offspring, but affected development of their higher hedonic regulatory pathways associated with food preference. In study 2, HA intake alone in gestational diets had long-lasting programming effects on the dopaminergic system and decreased sucrose preference but not food intake in the offspring. In study 3, HA in the post-weaning diet reduced the effect of the HV dam's diet on the offspring and modified gene expression in food intake and reward pathways. In conclusion, fat soluble vitamins, explain little of the effect of the HV diet consumed during gestation on the male offspring phenotype, but primarily, due to vitamin A, are important modulators of brain pathways regulating reward both in utero and post-weaning.

▼ The accurate and precise quantification of fat-soluble vitamins (FSVs), specifically vitamins A, D and E, has proved to be a challenge for clinical laboratories. This challenge is partially the result of the nature of FSV molecules and the limitations in techniques commonly used in their analysis. Consequently, there is a variation in the results obtained through different techniques or by different clinical laboratories. As a result, it has been difficult to reach agreement on the recommended levels or reference intervals of these vitamins. The current project encompassed the development of a state-of-the-art analytical method for the analysis of vitamins A (retinol), D (25-hydroxyvitamin D [25-OHD]) and E (α-tocopherol) in blood using liquid chromatography-tandem mass spectrometry (LC-MS/MS), which is a highly sensitive and specific quantification technique. Four studies were then performed to explore several scientific knowledge gaps related to vitamin measurement using the developed FSV quantification methods. The Current translational clinical research introduced novel and precise simultaneous measurement methods for blood FSV analytes, which were applicability to clinical sampling trials. This thesis also provides the first study exploring the stability FSVs simultaneously in routine sample spectrum utilising a precise LC-MS/MS method. In addition, stability of each analytes is justified based on calculation of the acceptable clinical limits, which reflect biological variation as well as method imprecision. This project also highlighted problematic issue related to trueness and reliable traceability of commercial calibrators to high order references and that interrupts effort of method harmonisation and patient result comparability. Furthermore, the current thesis provides with first study validated the measurement of 25-OHD3 and retinol in UCB diluted plasma. Lastly, this thesis reports the first study exploring status of five FSV analytes in two Australian populations at different latitudes using simultaneous measurement LC-MS/MS method. Furthermore, this is the first work examined the correlation between the blood FSV levels in two Australian populations.

▼ Η βαριατρική χειρουργική είναι η μόνη διαδικασία που μπορεί να εφαρμοσθεί προκειμένου να επιτευχθεί μόνιμη απώλεια βάρους στην πλειονότητα των ασθενών με υπερ-νοσογόνο παχυσαρκία (130). Οι βαριατρικές χειρουργικές επεμβάσεις ορίζονται ως περιοριστικές, δυσαπορροφητικές ή περιοριστικές και δυσαπορροφητικές ταυτόχρονα. Οι περιοριστικού τύπου χειρουργικές επεμβάσεις έχουν συνδεθεί με αυξημένα ποσοστά αποτυχίας όσον αφορά στην απώλεια βάρους στους υπερ-παχύσαρκους ασθενείς, με αποτέλεσμα να παρατηρείται προοδευτική αύξηση της εφαρμογής βαριατρικών εγχειρήσεων δυσαπορροφητικού τύπου σε όλο τον κόσμο για αυτήν την υπο-ομάδα παχυσαρκίας (100, 108, 130, 131).
Παρά την απώλεια βάρους που μπορεί να επιτευχθεί με αυτές τις χειρουργικές επεμβάσεις, οι μετεγχειρητικές ανεπάρκειες σε πρωτεΐνη, σίδηρο, βιταμίνες, άλατα και λιποδιαλυτές βιταμίνες αποτελούν σύνηθες φαινόμενο και απαιτούν χορήγηση συμπληρωμάτων προκειμένου να διατηρηθούν σε φυσιολογικά επίπεδα. Οι ανεπάρκειες αυτές οφείλονται στη δυσαπορρόφηση που προκύπτει από τη χειρουργική παράκαμψη τμημάτων του γαστρεντερικού σωλήνα, όπου τα διάφορα διατροφικά συστατικά απορροφούνται (118).
Αν και αναμενόμενες, οι μεταβολικές ανωμαλίες πολύ συχνά δε διαγιγνώσκονται ή υποεκτιμούνται. Τα βιβλιογραφικά δεδομένα που αναφέρονται στις διατροφικές ανεπάρκειες που παρατηρούνται μετά τις βαριατρικές εγχειρήσεις είναι περιορισμένα, και απαρτίζονται από αναφορές σε μεμονωμένα περιστατικά ασθενών (124-126), από επισκοπήσεις αποτελεσμάτων παλαιότερων ερευνών (118, 132) και από περιστασιακές προοπτικές μελέτες (118, 131). Αν και υπάρχουν αρκετές πληροφορίες για διατροφικά συστατικά, όπως είναι η πρωτεϊνη, ο σίδηρος, η βιταμίνη Β12, το φυλλικό οξύ και το ασβέστιο (118, 131), τα στοιχεία που παρατίθενται για τις λιποδιαλυτές βιταμίνες είναι ελάχιστα και ελλιπή. Αυτό οφείλεται σε σημαντικό βαθμό στη δυσκολία απομόνωσης και προσδιορισμού των λιποδιαλυτών βιταμινών από τον ορό του αίματος, διαδικασία αρκετά επίπονη, δαπανηρή και χρονοβόρα. Οι περισσότεροι χειρούργοι προσπαθούν να ελέγξουν με έμμεσο τρόπο τα επίπεδα των λιποδιαλυτών βιταμινών, όπως για παράδειγμα με παρακολούθηση των τιμών της PTH, άνοδος της οποίας συνηγορεί σε πιθανή ελάττωση των επιπέδων της βιταμίνης D3 (133), ή με παρακολούθηση του χρόνου προθρομβίνης των ασθενών ανά τακτά χρονικά διαστήματα, αύξηση του οποίου μπορεί να υποδεικνύει την ελάττωση των επιπέδων της βιταμίνης Κ (134). Στις περισσότερες περιπτώσεις, η ανεπάρκεια των λιποδιαλυτών βιταμινών γίνεται αντιληπτή μόνο από τις κλινικές της εκδηλώσεις ( 124, 125, 135).

▼ Abstract: The Cystic Fibrosis (FC) is the illness of bigger prevalence amongst the incident hereditary illnesses in the caucasian subjects. The illness is gradual and presents as clinical manifestation to the chronic respiratory blockage, masculine infertility and deficiency of profit of weight for the damage to the exocrine pancreas. The majority of the patients has high electrolyte levels in the sweat The present work consisted of the importance of the precocious diagnosis of the nutrition repercussions on the evolution and prognostic of the illness, therefore it evaluated the nutritional profile of the micronutrients (
fat-
soluble vitamins and oligoelements) in patients with Cystic Fibrosis taken care of in the clinic of Cystic Fibrosis of the Hospital of the Clinics of the UNICAMP. We studied 86 patients and 34 healthy volunteers in the control group. Blood samples were collected for measurement of serum biochemical parameters for enzymatic colorimetric methods.
Soluble vitamins were determined after their isolation by fast protein liquid chromatography (High Protein Liquid Chromatograpy-HPLC).We observe that for the analysis of the serum levels of

soluble vitamins in human blood samples using reverse-phase high performance liquid chromatography coupled with photodiode array detection. The biochemical forms of
fat-
soluble vitamins analyzed include retinol, cholecalciferol, calcidiol, α-tocopherol, γ-tocopherol, and phylloquinone. This method is novel in the use of prostaglandin B1 as an internal standard for quantification and utilizes a simple method of extraction without derivatization or saponification. The method had a high degree of reproducibility as evidenced by coefficients of variation 1.7% and 2.5% of the mean for retinal and cholecalciferol respectively.

▼ A solid-phase extraction (SPE) method was developed for separation and quantification of vitamin A (retinol, beta-carotene, retinyl-palmitate, retinyl-acetate), vitamin D (D2, D3), vitamin E (alpha,beta,gamma,delta tocopherols, tocopheryl-acetate), and vitamin K (K1, K2, K3) from different food matrices. The polarities of all fourteen
fat-
soluble vitamins (FSVs) were determined and chemical properties of many different organic solvents were studied. Quantification was conducted using liquid chromatography-mass spectrometry (LC-MS). All fourteen
fat-
soluble vitamins were found to be non-polar in nature; beta-carotene was found to be the most non-polar vitamin in the group, followed by vitamin esters. Tocopherols, ketones (
vitamins K1, K2, K3), and alcohols (
vitamins D2, D3, and retinol) were found to be relatively more polar. All fourteen forms were
soluble in dimethyl sulfoxide (DMSO) and were thus extracted with DMSO and methanol from food matrices.
To determine the recoveries of known concentrations of vitamin standards methanol and water were found to be suitable polar solvents for conditioning of the SPE column. Hexane was used for elution. This methodology was further applied to separate fourteen forms of FSVs from food matrices. 80-100% recoveries of FSVs were observed after SPE of provitamin samples (containing 2-3 ingredients). Poor recoveries of FSVs were observed after SPE of complex food samples such as multivitamin capsules, vitamin fortified drink mixes, and chewable nutrition tablets, which consisted of many different ingredients with varying polarities. The presence of other ingredients interfered with the SPE process leading to the elution of other compounds in the hexane solution along with FSVs.
Repeated measures ANOVA compared means of total FSVs after SPE and total FSVs from food labels. No significant differences were observed in the means (p < 0.05). A strong reliability (alpha>0.9) was observed between the three SPE extractions from each food sample. This SPE methodology can therefore be used for consistent and efficient separation of FSVs from products which do not have compounds that interfere with the extraction process.

▼ The objectives of the study were to define the dose-response relation of the amount of added oil and: 1) the absorption of carotenoids, phylloquinone and tocopherols in salad vegetables and 2) the absorption of retinyl palmitate formed from the ingested provitamin A carotenoids, ¦Á- and ¦Â-carotene. Women (n = 12) each consumed 5 salads containing equivalent amounts of carrot, cherry tomato, romaine lettuce and spinach. The salads with salad dressings containing 0, 2, 4, 8 or 32 g tocopherol-stripped soybean oil were ingested in random order separated by ¡Ý 2 weeks. Blood samples were collected at baseline and 2, 3.5, 5, 7, and 9.5 h postprandially. Chylomicron fractions were extracted and analyzed by HPLC with coulometric array electrochemical detection. When the salads were ingested with 0 g oil, there was negligible absorption of ¦Á- and ¦Â-carotenes, lutein, lycopene, phylloquinone, retinyl palmitate, ¦Á- and ¦Ã-tocopherols. For ¦Á- and ¦Â-carotenes, lycopene, retinyl palmitate, and ¦Á- and ¦Ã-tocopherols, absorption was increased with each amount of oil compared with 0 g oil (P <0.05). Starting from 4 g oil, all analytes (¦Á-carotene, ¦Â-carotene, lutein, trans-lycopene, vitamin A, ¦Á-tocopherol, ¦Ã-tocopherol, and vitamin K1) showed significant increases in absorption compared with 0 g oil salad dressing. The absorption of each carotenoid and fat-soluble vitamin was highest with 32 g ingested oil (P < 0.002).

▼ V doktorski dizertaciji smo raziskovali uporabo ogljikovega dioksida pri visokih tlakih za impregnacijo maščobo-topnih vitaminov. Za impregnacijo smo izbrali vitamina K3 in D3. Predvsem vitamin D3 je izjemno občutljiva substanca, zato zahteva izjemno pozornost. Za impregnacijo obeh vitaminov smo uporabili polisaharidne arogele. Aerogeli so porozni materiali z izjemnimi lastnostmi. Zaradi visokih poroznosti imajo velike specifične površine in nizke gostote. Če te lastnosti združimo še z biokomatibilnostjo ter biorazgradljivostjo polisaharidov, dobimo idealne materiale za farmacevtske aplikacije.
Cilj doktorske dizertacije je bil impregnacija vitaminov v polisaharidne aerogele, da bi jih s tem dodatno zaščitili ter tako povišali njihovo stabilnost. Delo je razdeljeno v dva večja dela. V prvem delu smo uporabili superkritični ogljikov dioksid za impregnacijo vitamina K3 in vitamina D3. Vitamin K3 smo uporabili kot modelno substanco, naš glavni cilj pa je bil impregnacija vitamina D3. Raziskali smo termodinamske in kinetične lastnosti obeh vitaminov. Dobljene vrednosti smo razložili z uporabo že razvitih kinetičnih modelov. V prvem delu dizertacije smo določili tudi strukturne lastnosti vzorcev ter preverili sproščanje vitamina in vitro.
V drugem delu doktorske dizertacije smo preiskovali impregnacijo vitamina D3 z ogljikovim dioksidom. Tokrat smo uporabili ogljikov dioksid pri različnih pogojih, od superkritičnega do tekočega ogljikovega dioksida. Preiskovali smo kinetično obnašanje pri različnih pogojih ter določili stopnjo degradacije vitamina glede na izbrane pogoje. Z različnimi karakterizacijskimi metodami smo določil vpliv tako superkritičnega kot tekočega ogljikovega dioksida na končne strukturne lastnosti aerogelov. V tem delu dizertacije smo prav tako izvedli sproščanje vitamina D3 in vitro, izvedli pa smo tudi stabilnostno študijo za potrditev povišane stabilnosti vitamina D3 ob impregnaciji v aerogele.
This doctoral dissertation examines utilization of carbon dioxide as solvent for high pressure impregnation of fat-soluble vitamins, in particular vitamin K3 and vitamin D3. Chosen vitamins, especially vitamin D3, are handling challenging since they are highly sensitive substances. Polysaccharide alginate aerogels were used as vitamins’ carriers. Aerogels are airy materials with outstanding properties such as high porosity, high specific surface area and low density. Combined with the biocompatible and biodegradable polysaccharides, they fulfil pharmaceutical demands for ideal candidates.
General idea of this dissertation was to achieve protection and prolonged stability of sensitive vitamins against environments by impregnating them into polysaccharide aerogels. The work is divided in main two parts. In the first part, carbon dioxide was used in its supercritical state. The impregnation experiments were conducted with both vitamin K3 and vitamin D3. ▼ Supercritical fluid extraction (SFE) is an alternative to traditional extractions with organic solvents. SFE consists of removing the analyte(s) from the matrix, solubilizing them, moving the analyte(s) into the bulk fluid, and sweeping the fluid containing the analyte(s) out of the extraction vessel.
As the fluid leaves the extraction vessel, decompression of the fluid occurs, changing its volume and temperature which can lead to analyte loss.
This work focussed on the trapping process with the restrictor immersed in a liquid after SFE. Experiments compared the effects of trapping parameters on the collection efficiencies of
fat-
soluble vitamins of similar polarities and structures. The most important variable was the selection of collection solvent and its physical properties, such as viscosity, surface tension and density were found to be important.
Additionally, adding a modifier to the collection solvent in an attempt to change its physical properties and influence collection efficiencies for a polarity test mix was studied. Addition of a modifier can improve collection efficiencies and allow higher collection temperature to be used, but the modifier did not increase trapping recoveries to the extent that collection pressurization did.
The occurrence of a methylation reaction of decanoic acid during the SFE and collection processes, using a methanol modified fluid or collection solvent was investigated. The majority of the reaction occurred during the collection process and the degree of methylation was found to be dependent on temperature, but not on static or dynamic extraction time. When no additional acidic catalyst other than carbon dioxide in the presence of water was present, conversion was limited to about 2%, but was quantitative with an added acidic catalyst.
The last portion of this work involved the application of the SFE process to the extraction and analysis of extractable material in eight hardwood and softwood pulp samples. Grinding the samples increased extractable fatty acid methyl esters (FAMEs) by ten-fold, and in-situ derivatizations resulted in higher FAME recoveries than derivatization after SFE. Liquid trapping enhanced recoveries of lower FAMEs when compared to tandem (solid/liquid) trapping. In-situ acetylations sometimes yielded acetylated glucoses. Large differences in FAMEs concentrations were seen for hardwood samples, but lesser differences were seen for the softwood pulp samples.

▼ The objective of the present study was to investigate the effect of the dietary supplementation of two levels (1.25 and 2.5% of the concentrate feed) of a natural zeolite, clinoptilolite, on the health status and performance of dairy cows. Fifty-two clinically healthy Holstein cows were used in the experiment. They were randomly assigned to one of three groups according to their age, parity and date of parturition. The first two groups were served as the experimental groups (Groups A and B) and the third (Group C) served as the controls as follows: Group A: was comprised of 17 cows that were fed a concentrate supplemented with 1.25% clinoptilolite. Group B: was comprised also of 17 cows receiving a concentrate with 2.5% clinoptilolite. Group C: was comprised of 18 cows that were fed the basal ration that did not contain any clinoptilolite. During the dry period all cows were receiving 20 kg corn silage, 2 kg molasses and 2 kg concentrates per day, whereas in the lactation they were receiving 30 kg corn silage, 2 kg molasses and 300 g concentrates per litter of milk produced per day. The concentrates offered to the cows of the three groups were formulated to be isonitrogenous, isoenergetics and have the same content in macroelements. The zeolitic tuff used contained approximately 92% clinoptilolite and its cation exchange capacity was 220 meq 100g-1. The starting day of the experiment was set one month before the expected day of calving (250th day of gestation). At that point two blood samples from the jugular vein were obtained from each animal, the first without anticoagulant and the second with anticoagulant. In the serum of the first blood sample was determined the concentrations of: β-hydroxyboutyric acid (β-ΗΒΑ), acetoacetic acid (AcAc), sorbitol dehydrogenase (SDH), γ-glutamyltransferase (γ-GT), glucose, blood urea nitrogen (BUN), total proteins, calcium (Ca), inorganic phosphorus (P), magnesium (Mg), potassium (K), sodium (Na), vitamin E (Vit. E), vitamin A (Vit. A) and β-carotene. In the second sample were determined the packed cell volume, the haemoglobin concentration and the white blood cell count.
Το αντικείμενο της συγκεκριμένης πειραματικής έρευνας, είναι η διερεύνηση της επίδρασης που έχει στην υγεία και τις αποδόσεις των γαλακτοπαραγωγών αγελάδων, η μακροχρόνια προσθήκη (335-365 ημέρες) στο σιτηρέσιο τους, ενός φυσικού ζεόλιθου, του κλινοπτιλόλιθου. Χρησιμοποιήθηκαν συνολικά 52 αγελάδες φυλής Holstein, που εξετάστηκαν πριν την έναρξη του πειραματισμού και βρέθηκαν υγιείς. Τα ζώα χωρίστηκαν σε 3 ομάδες, από τις οποίες οι 2 πρώτες αποτέλεσαν τις ομάδες στων οποίων στο σιτηρέσιο προστέθηκε κλινοπτιλόλιθος (Α και Β) και η τελευταία την ομάδα των μαρτύρων (Γ).
