▼ Steindachneridion parahybae (Siluriformes) é um bagre de água doce, endêmico da Bacia do Rio Paraíba do Sul (Brasil), e seriamente ameaçado de extinção. Fêmeas de S. parahybae quando criadas em cativeiro apresentam uma falha na maturação final, ovulação e desova. Este trabalho teve como objetivo analisar o eixo hipotálamo-hipófise-gônadas, responsável pelo controle neuroendócrino da reprodução de forma a aumentar o conhecimento deste sistema em animais de cativeiro. Fêmeas adultas foram amostradas mensalmente (exceto nos meses de inverno) entre janeiro/2008 e março/2009 na piscicultura da Companhia Energética de São Paulo. As análises macroscópicas e microscópicas dos ovários permitiram classificar o desenvolvimento ovariano em três estádios de maturação: pré-vitelogênico (crescimento primário), vitelogênico (crescimento secundário) e regressão; e cinco fases de desenvolvimento oocitário foram identificadas: oogônia, oócito perinucleolar, oócito alvéolo cortical, oócito vitelogênico e oócito atrésico. Após a indução à reprodução artificial, a maturação final foi alcançada e os folículos pós-ovulatórios foram identificados. Em S. parahybae, o período reprodutivo foi evidente entre novembro e fevereiro e o desenvolvimento oocitário foi do tipo sincrônio em grupo, sugerindo-se que esta espécie apresenta múltiplas desovas durante este período. Os perfis plasmáticos de 17Beta-estradiol (E2) e testosterona (T) foram fundamentais no desenvolvimento oocitário, e a produção destes esteróides sexuais parece não ser afetada no bloqueio da reprodução em cativeiro. Entretanto, as concentrações de 17Alfa-hidroxiprogesterona (17Alfa-OHP), durante o ciclo reprodutivo e após a indução à reprodução, sugerem que a falha reprodutiva de S. parahybae esteja relacionada com uma disfunção nos progestágenos, principalmente na conversão do 17Alfa-OHP em 17alfa,20Beta-dihydroxy-4-pregnen-3-one (Maturation-Inducing Steroid, MIS), este último considerado como hormônio da maturação final e ovulação em teleósteos. O andrógeno 11-cetotestosterona (11-KT) apresentou maior concentração no estádio vitelogênico e nas fêmeas induzidas à reprodução, sugerindo um envolvimento deste andrógeno na reprodução de S. parahybae. No entanto, o sítio de síntese e ação, assim como, as possíveis funções de 11-KT ainda permanecem pouco investigadas em fêmeas. No sistema encefálico foram caracterizadas duas formas do hormônio-liberador de gonadotropinas (GnRH), catfish GnRH (cfGnRH) e chicken-II GnRH (cGnRH-II). cfGnRH foi identificado em toda região ventral do telencéfalo e em várias regiões ventrais do diencéfalo, e o cGnRH-II foi observado na região do tegumento do cérebro médio, próximo ao terceiro ventrículo. O cfGnRH está intimamente relacionado com a modulação da atividade da hipófise, e ao contrario, o cGnRH-II (que não inerva a hipófise), provavelmente está relacionado com a neuromodulação e/ou comportamento reprodutivo em S. parahybae. Notre étude a porté sur le diagnostic de la présence des perturbateurs endocriniens dans le golfe de Gabès (Tunisie), leurs effets sur la reproduction d’une espèce bioindicatrice de pollution, la palourde R. decussatus ainsi que leurs mécanismes d’action. Pour atteindre ces objectifs, nous avons fait appel à plusieurs outils méthodologiques, des expositions in vivo (effluents urbains, effluents industriels, 17β-œstradiol) et des prélèvements in situ afin d’évaluer le risque encouru par l’écosystème du golfe de Gabès tout entier en utilisant une approche multiparamétrique basée sur différents niveaux de l’organisation biologique (allant du niveau biochimique jusqu’au niveau populationnel). Nous avons mis en évidence la présence des composés reprotoxiques tout au long de la côte Sud-Est de la Tunisie. Les résultats obtenus ont révélé une corrélation entre la contamination chimique (cadmium, activités œstrogéniques et androgéniques au niveau du sédiment) et certains paramètres biologiques liés à la reproduction (diamètre ovocytaire, vitellogénine, glycogène) et un site très divergent de tous les autres sites (El hofra). Cette station d’El hofra semble être touchée par une perturbation endocrinienne liée probablement à une synergie entre la contamination par le Cd et la présence des composés à activité œstrogénique et androgénique dans le milieu. En parallèle à cette étude in situ, trois expérimentations au laboratoire ont été réalisées. Les résultats ont montré que les effluents de la station d’épuration de Sfax (cocktail de contaminants) sont capables d’affecter la reproduction de la palourde en induisant la synthèse de la vitellogénine et stimulant la reproduction chez les deux sexes. L’exposition des palourdes à un contaminant pur, le 17β-œstradiol, a révélé qu’il peut contribuer à expliquer les perturbations du cycle reproducteur observées dans l’expérience précédente. Cette hormone naturelle a un effet et sans doute un rôle dans la physiologie de la palourde. Dans le but d’étudier son mécanisme d’action via le récepteur aux œstrogènes (RE), nous avons cloné une partie de l’ADNc codant pour les RE. Cependant, l’analyse de la réponse transcriptionnelle n’a pas montré de changement significatif dans l'expression du gène aussi bien chez les mâles que chez les femelles suite à une exposition aux œstrogènes (17β-œstradiol, effluents urbains et industriels).

Our study aimed to assess the presence of endocrine disrupting compounds in the Gulf of Gabes area (Tunisia), their effects on the reproductive potential of the marine bivalve, Ruditapes decussatus as well as elucidating their mechanisms of action. Polybrominated diphenyl ethers (PBDEs) are widely used flame retardants suspected to act as endocrine disruptors. We tested the commercial PBDE mixture DE-71 and its in vivo metabolites for estrogenic activity. MCF-7 breast cancer cells culture, ERE-luciferase gene expression, 3H-β-estradiol displacement from recombinant ERα, and ovariectomized (OVX) mice served as bioassays. Although DE-71 did not bind ERα, it was able to increase MCF-7 cell proliferation and this was prevented by the antiestrogen fulvestrant. DE-71 co-treatment reduced the effect of estradiol in MCF-7 cells. In the OVX mouse (BALB/c) 3-day assay, DE-71 administered alone had no effect on uterine or vaginal tissues but when administered subcutaneously potentiated estradiol’s effect on uterine weight in a dose-dependent manner. DE-71 administered SQ to BALB/c mice for 34 days slightly increased uterine epithelial height (UEH), vaginal epithelial thickness (VET) and mammary ductal lumen area, and attenuated the estradiol-induced increase in UEH; these effects were not seen in C57BL/6 mice. DE-71 increased liver weight in BALB/c, C57BL/6 and estrogen receptor-alpha knockout (ERαKO) mice. Liver cytochrome P450 1A (CYP1A) and CYP2B activities increased 2.5-fold and 7-fold respectively when DE-71 was administered PO, but only CYP2B increased (5-fold) after SQ treatment. Six OH-PBDE metabolites were found in mice after 34-day DE-71 treatment and all were able to bind recombinant ERα. Para-hydroxylated metabolites displayed a 10- to 30-fold higher affinity for ERα compared to ortho-hydroxylated PBDEs. Para-OH-PBDEs induced ERE-luciferase and produced an additive effect when coadministered with β-estradiol. DE-71 was also additive with β-estradiol. At high concentrations (≥ 5x10-5 M), ortho-OH-PBDEs were antiestrogenic in the ERE-luciferase assay. In conclusion, DE-71 behaves as a weak estrogen in both MCF-7 breast cancer cells and ovariectomized adult mice. Mice strain, treatment route and duration determined if DE-71 was estrogenic. BALB/c mice are more susceptible to DE-71 effects in estrogen target tissues than C57BL/6 mice. DE-71 increased liver weight, 5%-51% depending on mouse strain and treatment regime, independently of ERα. The observations that the DE-71 mixture does not displace 3H-β-estradiol from ERα while the hydroxylated metabolites do, suggest that the cellular and tissue effects were due to a metabolic activation of individual congeners.

▼ This thesis describes studies that investigated 1) the effects of courtship on the neuroendocrine system of female rough-skinned newts, Taricha granulosa, and 2) whether the observed courtship-induced neuroendocrine changes affected female sexual receptivity. Sexual behaviors of female T. granulosa changed dramatically during courtship. Initially, females exhibited unreceptive behaviors to a courting male. After several hours of courtship, females displayed receptive behaviors, and males responded by attempting to transfer a sperm cap. Once courtship, insemination and post-insemination behaviors were complete, the female no longer became receptive to future courtship. Both telencephalic irLHRH and plasma estradiol concentrations changed as females became receptive. Early in courtship, when females were unreceptive, irLHRH concentrations in the telencephalon were elevated, but by the time females became receptive irLHRH concentrations were low. The change in telencephalic irLHRH concentration occurred in the nervus terminalis, a little understood cranial nerve that may influence reproduction. Plasma estradiol levels were low at courtship initiation, but were elevated by the time females became receptive. The observations, that irLHRH and estradiol concentrations were associated with changes in female sexual behaviors and that these hormones influence female sexual receptivity in other vertebrates, suggested that courtship by a male activates female sexual behavior by stimulating endogenous changes in LHRH and estradiol. Increasing or decreasing plasma estradiol concentrations or blocking the action of this steroid in the brain were found to have no effect on female receptivity. Also, injecting LHRH or LHRH analogs into brain was found to have no effect on female receptivity. Progesterone implantation inhibited receptivity suggesting that this steroid may be responsible for the post-insemination decrease in female sexual behavior. These results suggest that although courtship induces physiological changes in female T. granulosa, the changes in LHRH and estradiol may not be influencing receptivity. ▼ This thesis considers the heterogeneity of human chorionic gonadotrophin (hCG) in vivo. This was studied in terms of the presence of free alpha hCG subunit, free beta hCG subunit, and metabolites, which accompany dimer hCG secretion in both pregnancy and malignancy. Also, the microheterogeneity of hCG arising from variations in its carbohydrate content was studied to establish whether it changes systematically during pregnancy or in malignancy. The use of highly specific monoclonal antibodies enabled hCG, its free subunits, and metabolites, to be accurately monitored in maternal serum and urine samples, both from the earliest stages of blastocyst implantations until delivery, and in trophoblastic and non-gestational malignancy. These profiles were confirmed by gel chromatography. The changes in hCG heterogeneity were also correlated with a detailed immunohistochemical study of placental tissue. From these studies it was possible to develop a proposal for the physiological mechanisms regulating hCG production. It is proposed that hCG synthesis is regulated through the differentiation of cytotrophoblasts into syncytiotrophoblasts. The mechanism of control involves limited translation of the genes for alpha and beta hCG subunits in cytotrophoblasts and in differentiating intermediate cells. Translation of the hCG genes ceases following the fusion of differentiating cytotrophoblasts into syncytiotrophoblasts, and the cellular machinery of syncytiotrophoblasts transcribes messenger RNA (mRNA) accumulated during differentiation. Thus, the total concentration and relative balances of mRNA for the hCG subunits in syncytiotrophoblasts are dependent on the rate and number of differentiating cytotrophoblasts. This proposal enabled the production of hCG and its subunits to be correlated with the changing histology of the placenta in pregnancy, and with unusual histological changes occurring in malignancy. The clinical role of hCG heterogeneity was investigated. Although dimer hCG was the predominant species secreted in pregnancy and malignancy, hCG heterogeneity provided information about both complications of pregnancy and malignancy. Examples of this included the following: increased free beta subunit to hCG ratios in early pregnancy in patients with subclinical abortions or abnormal pregnancies; increased levels of urinary immunoreactive hCG fragments in pre-eclamptic patients with renal complications; increased ratio of free beta subunit to hCG in patients with previous trophoblastic disease, signalling the proliferation of potentially malignant cell lines. The use of chromofocusing columns in HPLC enabled the changes in hCG microheterogeneity to be evaluated at different stages of pregnancy and malignancy. It is hypothesized that atrazine acts as an endocrine disruptor in amphibians, targeting male reproduction. To determine the impact of environmental atrazine on amphibians, we undertook a comprehensive study utilizing field and laboratory experimentation. In field studies utilizing Rana catesbeiana and R. clamitans melanota, we found evidence that secondary sexual traits are altered by environmental contamination. We also found that testes weights in both bullfrogs and green frogs were reduced at sites with chemical contamination compared to reference sites. At the histological level, renal and testicular dysgenesis were more prevalent at medium-contaminated sites than low or high. Ovotestes occurred most frequently (16.7%) at a site containing atrazine. The frequencies of renal parasites increased, while metacercarial cysts decreased with site contamination. Prevalence of hepatic and renal inflammation was higher in captures from medium and high contaminated sites compared to low. Bullfrog females from high contamination sites had reduced hematocrit than low and medium sites. In our field locations, there was evidence of endocrine disruption in frog populations. However, atrazine did not correlate well with observed effects; methoxychlor and metolachlor were associated with various endpoints. To further identify the role of atrazine in developmental effects, we exposed Xenopus laevis tadpoles to environmental atrazine concentrations (0, 1, 5, 20, 60, 120 ppb) during their larval period. Many endpoints examined presented with non-monotonic responses. Tadpole survival was reduced only at 5 ppb atrazine. At 5 and 60 ppb atrazine, tadpoles reached metamorphosis earlier than controls. Atrazine concentrations 5, 20 and 60 ppb yielded smaller metamorphs than controls, and metamorphs from 5 ppb atrazine also had shorter body lengths, limbs and abdominal girth. Malformations of the spine and limbs were highest at 5 and 20 ppb atrazine. We also observed a lowest observable adverse effect level (LOAEL) of 1 ppb atrazine for testicular dysgenesis, which is within environmental levels and supports concerns regarding the herbicide's ability to hinder amphibian reproductive output. Overall, our research suggests that low environmentally relevant doses of atrazine have the ability to impact wild amphibians by altering endocrine processes.

Les réseaux neuroendocrines qui contrôlent la reproduction sont mis en place pendant le développement sous l’action des stéroïdes sexuels endogènes. Toute perturbation de l’équilibre hormonal pendant ces phases critiques pourrait être à l’origine de troubles de la fonction de reproduction chez l’adulte. Ce travail vise à identifier l’impact d’une exposition développementale à un oestrogène de synthèse, l’éthinyloestradiol (EE2), sur les réseaux neuroendocrines et les conséquences physiologiques et comportementales chez l’adulte et sa descendance. Nos résultats ont montré que l’EE2 induit des perturbations des comportements sexuels chez les mâles et chez les femelles et une modification des réseaux hypothalamiques à GnRH et des neurones à calbindine qui contrôlent la physiologie et les comportements reproducteurs. Nous avons également montré que certains effets de l’EE2 sont transmis jusqu’à la quatrième génération, mettant ainsi en évidence le caractère transgénérationnel de ces perturbations. Ces résultats mettent en évidence la sensibilité des réseaux neuroendocrines aux perturbateurs endocriniens et la nécessité de prendre en compte ces paramètres dans l’évaluation de leurs effets sur la santé et la reproduction.

Neuroendocrine networks controlling reproductive function are established during development by the action of endogenous sex steroids. Any disturbance in the hormone balance during these critical phases may cause several disorders in reproductive function in adulthood. This work aims at identifing the consequences of a developmental exposure to the synthetic pharmaceutical estrogen ethinylestradiol (EE2) on the neuroendocrine and behavioral outcomes of the reproductive function in adult individuals and their offspring. Our findings showed that EE2 induced disturbances of sexual behaviors in males and females and modified the GnRH and the calbindin hypothalamic networks of exposed animals. We also showed that some effects of EE2 were transmitted up to the fourth generation, pointing out the transgenerational character of certain effects. All these results highlight the sensitivity of neuroendocrine networks to endocrine disruptors and the need to consider these parameters in assessing their effects on health and reproduction.

Parmi les outils existants pour l'évaluation de la qualité des milieux aquatiques, les biomarqueurs permettent de détecter précocement l'exposition et/ou les effets d'une contamination. Toutefois, en raison du manque de connaissance fondamentale de leur régulation au niveau moléculaire, peu de biomarqueurs ont été spécifiquement développés chez les invertébrés, alors que ces derniers représentent 95% de la biodiversité animale. Grâce aux récentes avancées technologiques dans le domaine du séquençage de l'information génétique et protéique, la découverte de nouvelles protéines chez ces organismes est à présent possible. Centré sur l'utilisation d'une espèce d'intérêt en écotoxicologique, l'amphipode d'eau douce Gammarus fossarum, et sur la problématique de la perturbation endocrine, ce travail doctoral a pour objectif de découvrir de nouvelles protéines impliquées dans la reproduction du gammare et de proposer des biomarqueurs spécifiques de reprotoxicité. Par une première approche protéogénomique, un important catalogue de protéines a été généré. Puis, afin d'identifier de nouvelles protéines impliquées dans la reproduction, une comparaison entre les protéomes des tissus reproducteurs mâle et femelle a été réalisée, suivi de l'étude de la dynamique du protéome au cours de différents processus physiologiques : ovogénèse, embryogénèse et spermatogénèse. Enfin, une dernière expérience sur des organismes mâles exposés à différents perturbateurs endocriniens a permis d'identifier différents candidats biomarqueurs de reprotoxicité. Cette étude ouvre la voie à des développements rapides de biomarqueurs spécifiques d'une espèce animale d'intérêt en écotoxicologie

Among the tools for assessing biologic quality of aquatic ecosystems, biomarkers are ideally suited for the early detection of contaminant exposure and/or effects. Yet, because of the lack of fundamental knowledge of their molecular regulation, few specific developments have been carried out on invertebrates even though they account for more than 95% of animal biodiversity. Thanks to recent technological advances in nucleic and proteic information sequencing, discovery of new proteins is now possible for these organisms. Focussed on the use of an ecotoxicologically relevant species, the freshwater amphipod Gammarus fossarum and the problem of endocrine disruption, this doctoral thesis aims to discover new proteins involved in gammarid reproductive function and to propose specific biomarkers of endocrine disruption. With a first proteogenomic approach, an important protein catalogue was generated. Next, to identify proteins involved in gammarid reproduction, male and female reproductive tissue proteomes were compared, followed by the study of proteome dynamics in several physiological processes: oogenesis, embryogenesis and spermatogenesis. Finally, a last experiment on male gammarids challenged with different endocrine disrupter chemicals identified several candidate biomarkers of reprotoxicity. ▼ Four studies were conducted in the llama (Lama glama) evaluating changes in reproductive physiology, vaginal cytology, and growth. Study I. Temporal changes in vaginal cytology were correlated with alterations in serum estradiol -17β and progesterone concentrations. A 2 * 2 factorial study examined the treatment effects of a) concurrent male exposure without copulation and b) collection of vaginal smears for cytologic examination on serum hormone concentrations. Repeated collection of vaginal samples produced: a) an increase in the relative percentage of cornified cells, b) a decrease in the percentage of intermediate cells; and c) no changes in serum estrogen or progesterone concentrations. The presence of a male did not influence serum estrogen, progesterone or vaginal cytology. Study II. The life-span of the corpus luteum was determined following hCG-induced ovulation and sterile or fertile breedings. Peak plasma progesterone concentrations of 8.5 ± .3 ng/ml at 9,8 ± .8 days following either sterile breeding or hCG administration was observed. Luteal regression, began 10.3 ± .3 and 10.8 ± .8 days following sterile breeding or hCG adminstration, respectively, and was completed by day 12-13 post-stimulation. Following a fertile breeding, progesterone increased significantly by 4 days post-breeding and remained above 2 ng/ml in all animals that subsequently delivered a full term cria. Fertile breedings could be differentiated from sterile breedings or hCG induced ovulation by 15 days post-stimulation by differences in progesterone concentration. Study III. The changes in plasma concentrations of progesterone, estradio1-170, total estrogens, T₃, T₄ and cortisol concentrations during pregnancy, parturition and the early postpartum period were determined in experiment III. Progesterone concentrations were significantly elevated by 5 days postbreeding, remained elevated throughout pregnancy until approximately 2 weeks prior to parturition and declined to < 0.5 ng/ml by the day of parturition. Total estrogen (TE) and estradiol-17β (E₂β) concentrations varied between 6-274 pg/ml and 4-114 pg/ml, respectively, during the first 9 months of pregnancy. Mean TE and E₂β concentrations increased between 9 months and the end of pregnancy with peak mean concentrations of 827 ± pg/ml (TE) and 196 ± 10 pg/ml (E₂β) observed during the last week of pregnancy. E₂β and TE concentrations declined rapidly immediately prepartum. Cortisol concentrations varied between 2.6 and 51.9 ng/ml (14.0 ± 0.5) from conception until 2 weeks prior to parturition when they began to decline. Only a slight increase in glucocorticoid concentrations were observed in association with parturition. Study IV. The growth patterns of the llama from birth to 12 months of age was characterized. Regression analysis revealed a strong positive correlation between log transformed measurements of thoracic circumference and height with body weight. ▼ Abstract: Endocrine disruptors are chemicals that may mimic or antagonize endogenous hormones, altering the critical hormonal balance required for proper health and development. It is well known that exposure of organisms to some hormonally active agents during critical periods of development imprints permanent changes that can be detected later in adulthood. In many animal species these substances reach the fetus via placenta and/or the offspring via the mother's milk. Although first researches have focused on environmental estrogens and antiandrogens, androgenic activity has been described as contaminant in rivers and beef cattle. Previous studies demonstrated that exposure of experimental animals to high levels of androgens during perinatal age causes permanent morphological, biochemical, and functional alterations in several organs and systems. Perinatal exposure of female rodents to exogenous androgens results in both physiological and behavioral masculinization and causes reproductive disruption in male rodents. The aim of this study was to assess the possible effects of an androgenic compound on genital system and reproductive function of animals exposed during critical periods of development. Pregnant female rats were allocated into four experimental groups: control, which received corn oil (vehicle), and treated with testosterone propionate at doses of 0.05mg/kg, 0.1mg/kg and 0.2mg/kg. Treatments were performed subcutaneously, from gestational day 12 until the end of lactation. The female offspring was evaluated for fetal germ cell number, body weight and anogenital distance at post-natal day 1, number of nipples, puberty onset, histology of reproductive organs, hormonal levels, sexual behavior, fertility test, immunohistochemistry of steroid receptors, proliferation/apoptotic index, uterotrophic test and uterine stimulation with estrogens. On male offspring, at adult age and middle-age, histology of testis and epididymis, sperm counts, morphology and motility, hormonal levels, sexual behavior and fertility test were evaluated. Results demonstrated that, in females, the perinatal testosterone propionate exposure caused an alteration in the pattern of uterine steroidal receptors imunostaining in all tested doses, a decreased proliferation/apoptotic index in the uterine tissue at 0.1 and 0.2mg/kg and a greater predisposition from uterus in responding to estrogens stimulation at dose of 0.2mg/kg. However, these alterations were not capable of impairing female sexual differentiation and normal physiology of female genital tract. On male offspring, the perinatal androgenization provoked a reduction in sperm production and reserves at adult age, without altering fertility after natural mating. Bisphenol A (BPA), the monomer of polycarbonate plastics and epoxy resins, can disrupt intrauterine implantation of fertilized ova in mice. This effect is also induced by exposure to chronic stress or high doses of diethylhexyl phthalate (DEHP), a plasticizer found in polyvinyl chloride products. I assessed the potential combinatory effects of BPA and stress on blastocyst implantation, uterine morphology, adhesion protein expression, and urinary hormone levels. Subcutaneous injections of BPA administered from gestation days (GDs) 1–4 paired with a stressor (rat exposure across a grid) reduced the number of implantation sites on GD 6 at a dose where neither BPA nor stress had this effect on their own. Uterine luminal area was increased by BPA when paired with stress. BPA reduced epithelial cadherin (e-cadherin), a uterine adhesion protein, independently from the stressor. Urinary estradiol was significantly increased by BPA relative to controls, regardless of stress. In other experiments, effects of concurrent BPA and DEHP administered were assessed. Inseminated female mice were injected with BPA, DEHP, or BPA + DEHP from GDs 1–4. Implantation measured in uteri on GD 6 was disrupted by a combined dose but not by the individual doses. This dose also decreased the amount of e-cadherin and cadherin-11, another adhesion protein expressed by cells, while cadherin-11 was also affected by BPA alone. In further experiments designed to elucidate the interaction of BPA and DEHP, mice were fed 14C-BPA and injected with varied doses of DEHP, then tissues were excised and measured for radioactivity. When given DEHP, males and cycling and peri implantation females showed increased BPA deposition in reproductive tissues and serum. As people are commonly exposed to both DEHP and BPA through consumer products, it is important to determine their interactions and also to understand how dose-response is affected by other factors such as stress.

▼ Genistein, the primary phytoestrogen in soy, was investigated for potential adverse effects on the developing female reproductive system with particular focus on the ovary. Mice were treated with genistein at doses that span the range of human exposure including vegetarian mothers during pregnancy and lactation to infants on soy based infant formulas. Neonatal genistein exposure caused the formation of multi-oocyte follicles (MOFs) in the ovary. This effect is mediated by ERβ as mice lacking this receptor do not develop MOFs while mice lacking ERβ do. Further study of genistein's effects on the ovary revealed inhibition of neonatal oocyte nest breakdown; oocytes were still attached by intercellular bridges and the normal progression of apoptosis was attenuated. Mechanistic studies of MOF formation revealed alterations in cell adhesion molecules. In addition, genistein is not unique in its ability to cause ovarian disruption; other environmental estrogens caused MOFs as well as altered cell adhesion molecule expression. Further, these effects appear to be exacerbated by preferential binding to ERβ. Assessment of reproductive function showed that mice treated with genistein (0.5 and 5 mg/kg) showed signs of early reproductive senescence while mice treated with genistein (50 mg/kg) exhibited infertility characterized by fewer, smaller, implantation sites as well as reabsorptions; ovaries from these mice had no corpora lutea. Stimulation with exogenous gonadotropins restored ovulation, suggesting problems with the hypothalamic-gonadal axis. These data taken together demonstrate that neonatal exposure to genistein at environmentally relevant doses causes adverse effects on the developing reproductive system and in particular on the ovary. ▼ Urinary steroids have been studied during early and late pregnancy in domestic horses or sporadic samples at various stages of pregnancy in wild equidae. In our studies, urinary estrone sulfate (E1S) and pregnanediol glucuronide (PdG) were monitored throughout pregnancy in six pony mares by enzyme immunoassay (EIA). Both hormones were corrected by creatinine (Cr) index to compensate for the variation in specific gravity. The mean concentration for ElS, (μg/mg Cr), was .38 ± .03 at d 0, decreased to .17 ± .04 at d 1, and maintained at less than .5 μg/mg Cr until d 30. Although, there was an apparent increase to .80 ± .34 at d 34 (NS, P = .122), the first significant increase was .69 ± .15 at d 46 (P = .0275). Mean concentrations remained relatively stable at this approximate level until d 60. This level was followed by a sustained significant increase observed from d 60 onwards. Mean concentrations of El S increased to 1.11 ± .25, 2.01 ± .45, and 5.48 ± 1.47 at d 64, d 76, and d 86, respectively. Levels of EIS further increased reaching a peak of 143.3 ± 9.51 at d 142 (P = .0006), with maximum for individual mares ranging from d 114 to 170, and also ranging from 115.4 to 286.1 pg/mg. In all cases, maximum concentrations were followed by a gradual decline toward parturition with a more rapid decrease 1 to 3 days before parturition. The first significant decrease following the maximum concentration was 91.40 ± 13.11 (P = .0024) at d 184. Estrone sulfate was 12.1 ± 3.8 one day prepartum and decreased to .4 ± .1 and .1 ± .01 at d 1 and 4 postpartum, respectively. The mean concentrations of PdG (ng/mg Cr) increased from 147 ± 4.3 at d 0 to 50.87 ± .17 (NS, P> .05), 36.8 ± 8.1 (P = .016), and 27.6 ± 7.3 (P = .049) at d 6, 8 and 10, respectively. This increase was followed by a decline and generally the levels fluctuated ranging from 20 to 30 ng/mg Cr until d 80. At d 86, the PdG levels increased to 54.7 ± 11.7 (P = .033). This was followed by a further increase to 141.8 ± 21.4 (P = .0139, compared to d 93) at d 135, then continued to increase to 213.0 ± 25.2 at d 198, and remained at this approximate level until d 303. During the last month of gestation, the mean concentrations of PdG increased from 171.8 ± 9.8 at d 29 prepartum to reach a peak of 388.4 ± 108.6 at d 7 prepartum. Maximum concentrations were followed by a slight decrease to 354.5 ± 84.0 at d 1 prepartum and then decreased to 150.6 ± 23.4 and 39.6 ± 9.3 ng/mg Cr at d 1 and 4 postpartum. In comparing the two hormones, E1S remained baseline followed by a slight increase at d 35, whereas PdG was relatively stable until both hormones increased after d 70 of gestation. This might be related to secretion of both hormones by the fetus and their rapid metabolism by placenta.
