▼ Integrated nanowire electrodes that permit direct, sensitive and rapid electrochemical based detection of chemical and biological species are a powerful emerging class of sensor devices. As critical dimensions of the electrodes enter the nanoscale, radial analyte diffusion profiles to the electrode dominate with a corresponding enhancement in mass transport, steady-state sigmoidal voltammograms, low depletion of target molecules and faster analysis. To optimise these sensors it is necessary to fully understand the factors that influence performance limits including: electrode geometry, electrode dimensions, electrode separation distances (within nanowire arrays) and diffusional mass transport. Therefore, in this thesis, theoretical simulations of analyte diffusion occurring at a variety of electrode designs were undertaken using Comsol Multiphysics®. Sensor devices were fabricated and corresponding experiments were performed to challenge simulation results. Two approaches for the fabrication and integration of metal nanowire electrodes are presented: Template Electrodeposition and Electron-Beam Lithography. These approaches allow for the fabrication of nanowires which may be subsequently integrated at silicon chip substrates to form fully functional electrochemical devices. Simulated and experimental results were found to be in excellent agreement validating the simulation model. The electrochemical characteristics exhibited by nanowire electrodes fabricated by electronbeam lithography were directly compared against electrochemical performance of a commercial ultra-microdisc electrode. Steady-state cyclic voltammograms in ferrocenemonocarboxylic acid at single ultra-microdisc electrodes were observed at low to medium scan rates (≤ 500 mV.s-1). At nanowires, steady-state responses were observed at ultra-high scan rates (up to 50,000 mV.s-1), thus allowing for much faster analysis (20 ms). Approaches for elucidating faradaic signal without the requirement for background subtraction were also developed. Furthermore, diffusional process occurring at arrays with increasing inter-electrode distance and increasing number of nanowires were explored. Diffusion profiles existing at nanowire arrays were simulated with Comsol Multiphysics®. A range of scan rates were modelled, and experiments were undertaken at 5,000 mV.s-1 since this allows rapid data capture required for, e.g., biomedical, environmental and pharmaceutical diagnostic applications.

▼ Advances in the application of bipolar electrodes (BPEs) for screening of electrocatalysts, localized activation of a single conductive electrode, the optical tracking of single particles interacting with an active electrode, and the introduction of microwires in paper-based analytical devices are described. In an original proof of concept study arrays of BPEs were used to determine the relative activity of model nanoparticle systems for the oxygen reduction reaction (ORR) by a simple optical readout: the electrodissolution of Ag microbands. The number of bands that dissolved during the screening procedure determined the relative activity of the materials. These screening results for model nanoparticle systems were related to traditional electrochemical experiments and showed a strong correlation. Building on that initial study, the BPE platform for screening ORR electrocatalyst candidates was improved so that more materials could be evaluated simultaneously by increasing the density of electrodes in the array, controlled compositional variations were prepared with the implementation of piezodispensing, and a different reporter, Cr, replaced Ag at the BPE anodes which reduced the risk of contamination and improved reliability of screening experiments. Further studies into the versatility of the screening platform have been carried out using non-noble metal systems for the hydrogen evolution reaction (HER), which has a long history of interest for electrochemists. A single conductive electrode material can be made to act as an array of electrodes by confining it at the intersection of two orthogonal microfluidic channels. By manipulating the direction and magnitude of the electric field in the device, faradaic reactions can be selectively localized on the BPE. An approach for optically tracking individual, insulating microparticles interacting with an active UME has been achieved. This approach brings new insight and understanding of single particle electrochemical studies. Finally, a method for incorporating microwires and mesh electrodes into paper-based electroanalytical devices is reported. This has many advantages over traditional screen-printed carbon electrodes that are traditionally used in paper-based devices.

▼ The two major research areas in this thesis are electrochemical As (III) determination and H2O2 reduction in aqueous solution. Although a variety of nanomaterials have been developed, there is still room for new materials and greater understanding of current material classes that may lead to higher sensitivity, lower limit of detection (LoD) and acceptable selectivity. These two research areas are linked by the fact that that they both rely on the careful preparation of well characterised nanomaterials for electrochemical applications, which is the central theme of this thesis. Each of the research areas and their main results are presented below. The first result presented is the electrodeposition of gold nanoparticles (AuNPs) on a gold substrate for Arsenic (III) detection. To obtain the best As (III) analysis sensitivity, a series of conditions were optimized. Also, two most commonly used electroanalysis techniques, square wave anodic stripping voltammetry (SWASV) and amperometric i-t plots were compared, indicating that SWASV leads to a better sensitivity of 0.124 µA ppb-1 and the lower limit of detection (LoD) of 0.244 ppb. Cu (II) interference studies revealed acceptable anti-interference ability at lower concentrations of Cu (II) (below 50 ppb). Next, a series of Au nanospikes and dendrites were electrodeposited with either an inorganic (Pb2+) or organic (cysteine) growth directing agent for different times to obtain varied morphology. These structures were compared with gold nanoparticles of three different shapes (Octahedral, Cubic and Rhombic Dodecahedral) for detection of As (III) by SWASV. The sensitivity and LoD was dependent on the surface crystallographic orientations and the morphology, with superior sensitivity confirmed with a maximum amount of Au (111) facets on the surface for the nanoparticles. The study confirms that electrodeposition parameters or nanoparticle synthesis methods for Au surfaces in arsenic sensing needs to be carefully controlled, to either maximise the (111) facets, or minimise the steps on a polycrystalline Au surface, and that inorganic and organic shape directing agents used for electrodeposition will have differing effects. A new material for As (III) detection was prepared based on ceria cubes decorated with manganese oxide nanoparticles (Mn2O3/CeO2 nanocubes), and used to modify a Au electrode for analysis of As (III) in aqueous solution. This modified electrode displayed improved sensitivity than either oxide on their own, indicating a synergistic effect. The improved sensitivity could be ascribed to the enhanced As (III) adsorption ability of the Mn2O3/CeO2 nanocube during electrochemical pre-concentration, combined with the well-known As (0) deposition and stripping qualities of the gold substrate. Also, a graphene oxide framework (GOF) material was constructed and used to modify a gold substrate. ▼ CVD diamond possesses certain attractive electrochemical properties inter alia low background current, broad potential window, chemical inertness and resistance to electrocorrosion and fouling. As a consequence its use in various areas of electrochemistry, such as electrochemical sensing, wastewater treatment and electrocatalysis is being explored. Unfortunately, alongside these attractive features, bare CVD diamond electrodes, in common with all other electrode materials, cannot be effectively applied in all electrochemical systems of interest, since for example it may not display useful electrochemical activity for the redox process of interest. In these circumstances it may be possible to modify the electrode by addition of other chemical species to the surface, to introduce the relevant activity. One of the main aims of this thesis was therefore to investigate the properties of certain chemical modifications to the diamond electrode surface. A second aim was also to explore for the first time the use of a practically useful form of single crystal diamond, so-called heteroepitaxial diamond, in electrochemistry. The diamond electrodes used were boron-doped material grown by chemical vapour deposition. A range of electrochemical methods, including especially cyclic voltammetry, square-wave voltammetry, impedance spectroscopy and scanning electrochemical microscopy, were used to characterise electrode properties. Other physical methods employed included scanning electron and atomic force microscopy, X-ray photoelectron spectroscopy and dynamic light scattering techniques. The electrochemical properties of heteroepitaxial single crystal diamond were explored and compared to polycrystalline counterparts. The single crystal diamond electrode was found to show superior properties in terms of wide potential window, low background current and homogeneous activity across the electrode surface, coupled with resistance to fouling. Heterogenous electron transfer rate constants were found to be lower than normally found on polycrystalline diamond; this was attributed to reduced density of states and absence of functional groups. An electrochemical route to the preparation of diamond electrodes, modified by
[email protected] core-shell particles was demonstrated. It was observed that these electrode modifiers were far less susceptible to poisoning than bare Pt nanoparticles when used in the electrochemical oxidation of methanol. It was also shown that diamond electrodes with these core-shell particles deposited on them, displayed useful activity for the electrochemical oxidation of nitric oxide. The presence of the PrOx layer was shown to impart useful selectivity against the oxidation of interfering compounds such as nitrite and ascorbic acid, without the loss of sensitivity which normally occurs if nafion coatings are used instead. ▼ Electrochemical analysis using mercury electrodes has been a prominent methodology since the beginning of voltammetry, however, due to its toxic nature their use is being slowly phased out in favour of environmentally friendlier alternatives. In this thesis, carbon electrodes feature heavily as a means to provide a cheaper and non-toxic approach to voltammmetric sensing. Pulse techniques have been used for the sensitive detection of commonly studied analytes, including antimony and iron. A simple fabrication of a vibrating electrode is presented by modifying a commercially available vibrating toothbrush with platinum foil as a means to enhance mass transport for electroanalytical work. Finally, pH determination at carbon electrodes is investigated. Carbon electrodes have been proposed as a simple sensor for pH determination by exploiting the pH sensitive nature of surface quinone groups intrinsic within carbon edge-plane like sites. Using this approach, both EPPG and GC electrodes are suggested as new materials for pH determination and can be used in aqueous solutions over the pH range 1.0 to 13.0, as a cheap and simple alternative to the classic glass electrode.

▼ 3D printing has revolutionised traditional manufacturing methods, opening up and distributing design and production of low cost, custom objects to virtually anyone. Tailoring of print material and part geometry allows for the benefits of this technology to reach multiple engineering and scientific fields, given appropriate design. A multidisciplinary approach concerning development of new print materials and methods was undertaken with the aim of further expansion and application of 3D printing towards electrochemical applications. Specific requirements of materials used in this domain, such as conductivity and chemical stability, led to development of functional printable carbon composites, compatible with consumer grade 3D printers. This allows facile production of cheap, reusable, disposable, electrodes for analytical applications, demonstrating heavy metal detection in aqueous media and allowing further tailoring to specific applications to be easily implemented. A new method for printing of cellulose solutions was developed, with post processing of printed parts resulting in biocompatible, porous, conductive structures. When used as electrodes in microbial fuel cells, improved power and current output over traditionally used carbon cloth electrodes was achieved. Other developments resulting from this work applicable to other fields include a novel trajectory generation method based on exponential functions which can be applied to practically any robotic system, as well as improvements to the production process of metal alloy filaments for 3D printing of metallic components.

L’étude est organisée en trois parties majeures: la synthèse de zéolithes micro/mesoporeux et leur application potentielle dans l’industrie pétrochimique, l’étude théorique de mécanismes réactionnels sur des zeolites microporeux, et le design de métaux mesoporeux avec une chiralité intrinsèque de leur surface interne. Ces matériaux poreux montrent des propriétés excellentes, notamment pour des applications potentielles en catalyse et comme interfaces chirales.
In the present work, the elaboration, characterisation and applications of differentporous materials have been studied. Porous materials are divided into three categoriesdepending on the porous cavity size, namely microporous materials (pore diameter < 2nm), mesoporous materials (2 nm < pore diameter < 50 nm) and macroporous materials(pore diameter > 50 nm). The thesis work is organized in three major parts: the synthesisof hierarchical micro/mesoporous zeolites and their potential application for thepetrochemical industry, the theoretical study of reaction mechanisms on microporouszeolite and the design of mesoporous metals with intrinsic chirality at their inner surface.The hierarchical micro/mesoporous zeolite, composed of microporous andmesoporous features, has been prepared using carbon-silica (C/SiO2) composites derivedfrom a pyrolysis of hydrocarbon gas on silica gel. Our findings demonstrate that not onlythe presence of a high surface area and porosity, but also an improved efficiency of thesematerials for many petrochemical processes such as n-butene isomerization, nhexadecanecatalytic cracking and hydrocracking. The novel synthetic method is expectedto be generalized for other types of zeolites, and is considered to be a promising methodfor creating hierarchical micro/mesoporous zeolites for potential catalytic applications,especially in the petrochemical industry.In addition to the study of practical catalytic aspects, a theoretical approach hasbeen used to investigate potential reaction mechanisms such as the selective isomerizationof 1-butene into isobutene. More specifically, the monomolecular skeletal isomerizationof 1-butene into isobutene on H-FER zeolite was theoretically studied by using theONIOM approach. This process was found to involve the transformation of adsorbed 1-butene through 2-butoxide, isobutoxide, and tert-butyl cation intermediates. The ratedeterminingstep is the conversion of isobutoxide into isobutene, in which the reactionhas to proceed through the primary isobutyl cation transition state. The shape selectivitydue to the “nano-confinement” effect of the zeolite framework strongly affects theadsorption, the stability of alkoxide species and carbenium ion, as well as the skeletalisomerization mechanism of 1-butene.Moreover, the microporous and mesoporous zeolite, the generation of chiralmesoporous metal and its enantioselective recognition properties have been studied.Molecular imprinting (MI) is a major…

▼ A cromatografia a líquido de alta eficiência e a voltametria de onda quadrada foram estudadas e comparadas estatisticamente para a detecção do contaminante emergente metilparabeno (MePa). Os parabenos vêm sendo amplamente utilizados como conservantes em cosméticos, alimentos e remédios. É crescente o número de estudos que visam à detecção dos parabenos em virtude dos males causados à saúde humana, devido às suas características de serem carcinogênicos e interferentes endócrinos. Para determinação cromatográfica, foi desenvolvido um método analítico que utilizou filtração à vácuo seguida de análise cromatográfica acoplada a um detector UV. O eletrodo de diamante dopado com boro (BDD) foi utilizado para determinação eletroanalítica de MePa pela técnica voltamétrica. Para a análise cromatográfica, foi observada boa linearidade (0,999) na faixa de 0,5 a 30 mgL-1, com limite de detecção e quantificação de 0,49 mgL-1 e 0,73 mgL-1, respectivamente, para amostra de água natural superficial e para a amostra de cosmético 1,26 mgL-1 e 1,88 mgL-1, respectivamente. A dependência da corrente pelo pH e os parâmetros eletroquímicos (frequência, amplitude e step) foram estudados para obtenção de condições experimentais ótimas na determinação de MePa. Foi observado que o eletrólito suporte ideal é o tampão PBS 0,1 molL-1 (pH 7,5). Linearidade adequada (0,998), na mesma faixa de trabalho, com limite de detecção e quantificação de 0,43 mgL-1 e 1,44 mgL-1, respectivamente, foram encontrados para a técnica voltamétrica. Os métodos utilizados em ambas as técnicas foram validados em uma amostra de água natural superficial, coletada em região pouco impactada, e em uma amostra de cosmético. Utilizando o método cromatográfico, foram obtidos intervalos de recuperação entre 95,6% e 101,6% com RSD inferior a 4,8% (n = 3) e entre 78,0% e 99,8% com RSD inferior a 2,2% (n = 3) para amostras de água natural superficial e de cosmético, respectivamente. Na determinação eletroanalítica, soluções fortificadas daquelas amostras mostraram intervalo de recuperação entre 92,6% e 106,4%, com um desvio-padrão relativo (RSD) inferior a 9,76% (n = 3), para amostra de água superficial natural, e entre 90,0% e 112,6% com RSD inferior a 7,4% (n = 3), para amostra de cosmético. Os resultados das detecções cromatográfica e eletroanalítica foram comparados e analisados estatisticamente segundo o método de Miller e Miller. Através do teste t de Student pareado, demonstrou-se que a metodologia eletroanalítica é capaz de determinar MePa com boa precisão tal como a técnica padrão de detecção, a cromatografia, na faixa de trabalho de 1,0 a 30 mgL-1.

▼ Abstract: Electrochemical devices can be used for a fast, inexpensive and sensitive detection of several analytes, such as biological molecules for diseases diagnostics. The versatility of these devices allows their use in wide range of electrocatalytic processes such as energy conversion, including oxygen evolution reaction. In this sense, graphene-based materials are a remarkable platform for electrocatalysis, due to their impressive mechanical properties, chemical stability, high conductivity and high surface area. Reduced graphene oxide (rGO) has been highlighted due to the presence of oxygenated functional groups in its structure, making it hydrophilic and favors the formation of composites. Thus, when combined with materials such as Prussian blue and Ni-Fe (oxy)hydroxides, reduced graphene oxide composites present synergistic properties, due to its fast electron transfer kinetics and the stabilization of the eletrocatalysts. In this thesis, we studied the properties of conventional and 3D-printed electrodes modified with graphene and its composites in electrochemical devices. Firstly, we focused on photochemical one-pot synthesis and characterization of reduced graphene oxide/Prussian blue (PB) composites, which presented a controllable shape and size of PB. Then, the performance of bare and modified glassy carbon electrodes with the composites were investigated towards sensing of hydrogen peroxide and simultaneous detection of ascorbic acid, dopamine and uric acid. Posteriorly, a new substrate for sensing of dopamine was proposed, based on graphene/polylatic acid (PLA) composite filaments. Thus, from the filaments, 3D-printed electrodes were obtained and their electrochemical properties were modulated by an electrochemical pre-treatment. From the pre-treatment, graphene sheets were exposed to the surface of the electrodes, increasing the response towards dopamine. Finally, we have reported the use of the activated 3D-printed electrodes as support for electrocatalysts of Ni-Fe (oxy)hydroxides for the oxygen evolution reaction

▼ Nano- and microscale motors have been studied intensively over the past decade and are of much current interest because of their potential applications in the electronic and biomedical fields. While research on molecular motors and artificial muscles has resulted in a number of breakthroughs, one of the most promising recent directions has been the discovery of free-moving catalytic motor systems. Catalytic motors have the potential to operate with a variety of possible fuel-catalyst combinations. In this thesis we report mechanistic studies of catalytic motors that use hydrogen peroxide and other fuels, as well as the observation of pairwise motor interactions in fuel-containing solutions.
In the mechanistic study, we developed a mixed potential model for the hydrogen peroxide bimetallic nanorod system. This analysis can correctly predict the relative speed and direction of different metal pairings. We applied the same method to the hydrazine fuel system and catalytic micropumps, and those experiments supported a similar electrokinetic mechanism for those systems also. Control experiments with catalytic nanorods that had low electronic conductivity also supported the electrokinetic mechanism.
To expand the application of catalytic nanomotor systems and to further understand scaling effects we fabricated bimetallic catalytic motors by lithographic methods. These experiments showed that catalytic motors still function in the 10µm size range and that electrokinetic propulsion is still dominant over inertial mechanisms. Some differences in behavior were observed that may be attributed to artifacts of the fabrication process, such as poor electrical contact between components of the bimetallic motors and poisoning of catalytic surfaces. Lithographically fabricated chiral bimetallic designs led to successful fabrication of rotary motors, which are interesting for the observation of pairwise motor interactions and collective phenomena..
One of the interesting extensions of catalytic rotor research is the possible interactions and dynamic assemblies brought about by microscale rotating objects, which should operate on different principles compared to centimeter-scale rotary motor systems. We designed and fabricated fast-spinning catalytic rotors based on bimetallic nanorod designs, and our observations have shown various types of interactions between spinning rotors in close proximity, which may be due to a number of different effect. We then constructed a magnetic rotational system involving ferromagnetic nanorods and compared them with the catalytic micro-rotors. Interactions between rotors were again observed, and the potential exists for the formation of dynamic ordered matrices.

▼ Current trends in analytical chemistry aim at developing simpler and more affordable sensing devices. Electrochromic materials enable the fabrication of electrochemical sensors that exploit colour changes as a means to reduce instrumentation requirements. Although many chemical compounds used in
electroanalysis exhibit electrochromic properties, the inclusion of these materials in the construction of electrochromic analytical sensors has begun to emerge only recently.
This thesis focuses on the development of a new type of analytical devices relying on the use of electrochromic materials, reducing to a minimum the required components, and removing the need for silicon-based electronics.
First, the working principle of self-powered electrochromic sensors is presented, demonstrating how with the appropriate geometry it is possible to obtain a sensing platform capable of providing a quantitative readout of the analyte concentration. The experimental data obtained are combined with the use of numerical simulations to develop a mathematical model that helps to have a further understanding of the behaviour of the device. Moreover, these simulations were used as a design tool to improve the performance of future electrochromic devices.
New materials in the form of screen-printing pastes have also been formulated to substitute certain components of the device such as the electrochromic material or the electrolyte. These materials were used in the last part of the thesis to manufacture a fully printed electrochromic sensor able to provide quantitative results without the aid of an external signaling device.

▼ This thesis concerns several advances in electroanalytical chemistry which are separated into four parts: the electrochemical investigation of diffusional behaviour, the mechanistic and kinetic study of electrochemistry with room temperature ionic liquids (RTILs), the study of weakly-supported electrochemistry and a comparison of the Butler-Volmer and Marcus-Hush kinetic theories of electron transfer. A study of the diffusional behaviour of electroactive species is essential for further studies, especially in the case when electrochemistry is complicated through ion-pairing interactions between the electroactive species and other electrolytes. In Part II of this thesis, the possibility of the ferricenium ion-paired with perchlorate and hexauorophosphate in acetonitrile was discussed firstly employing chronoamperometric technique. Afterwards, the hexaammineruthenium III/II couple supported by chloride, nitrate and sulfate respectively was studied by a similar method. In order to avoid unwanted ion-pairing effects, room temperature ionic liquids can be applied as solvent, which provide high conductivity by their own ionic nature so that experiments can be conducted without adding additional supporting ions. Because of RTILs have distinctive properties, for example, high viscosity, high conductivity and ionic nature, electrochemistry could be greatly changed compared to those in conventional solvents. Part III of this thesis gives a detailed description of this topic. First, a study of the reduction of 1,4-benzonquinone in 1-ethyl-3-methylimidazolium bis(triuoromethanesulfonyl)imide is presented to show the new mechanistic insight into comproportionation in a electrochemical process. Second, a discussion of the oxidation of hydroquinone in the same RTIL is introduced to suggest a possible ECE scheme which was never reported before. The interest of weakly supported electrochemistry is also well-established, which not only provides another alternative strategy to avert ion-pairings but also offers more physical insights into electrochemical processes. Quantitative methods analysing voltammetries without an excess amount of supporting electrolyte are developed by introducing a migration term into the mass transport equation. In Part IV, new mechanistic insights into the reduction of 2-nitrobromobenzene and the dimerisation of 2,6-diphenylpyrylium in acetonitrile were provide by using weakly-supported cyclic voltammetry. Also, pulse techniques was also adopted to investigate the reduction of cobaltocenium and cobalt(III) sepulchrate, giving an alternative way for electrochemical analysis. A major application of electroanalytical chemistry is investigating electrochemical kinetics. Two kinetic models mostly concerned by electrochemists are Butler-Volmer and Marcus-Hush formalisms. ▼ The experimental work discussed in this thesis explores the effects of metal modification on the electroanalytical ability of boron doped diamond electrodes. Boron doped diamond (BDD) electrodes have found increased application to electroanalysis in the past two decades, yet relatively little of the literature is focused on metal, nano and microparticle modification of the substrate. In this thesis three metals have been used to modify the BDD electrode; bismuth, antimony and nickel. Bismuth and antimony nanoparticle modified BDD electrodes were directly compared to unmodified BDD and a bulk bismuth electrodes in the determination of trace levels of cadmium and lead using anodic stripping voltammetry. In both instances, the modified electrode allowed for the simultaneous determination of each analyte that was otherwise unattainable at the unmodified BDD electrode. The nickel modified BDD (Ni-BDD) electrode was used in the determination of organic analytes, namely glucose, methanol, ethanol and glycerol. The nickel nano and microparticle electrodes gave the characteristic Ni(OH)₂/NiOOH redox couple in alkali pH, the oxidised form of which (NiOOH) catalysed the oxidation of the organic analytes. The chapter on glucose sensing with the Ni-BDD electrode is preceded by an extensive literature review on the advances of non-enzymatic glucose sensing, and the application of catalytic metals and nanomaterials in this field. Throughout the course of this DPhil, there has been a collaborative project between Asylum Research and myself within the Compton group to develop a commercial electrochemical atomic force microscope (EC-AFM) cell. The aim was to produce an adaptable EC-AFM cell capable of dynamic electrochemical experiments whilst simultaneously or instantaneously acquiring an AFM image of the modified surface, in-situ. This project was successful, and the EC-AFM cell has contributed to a number of chapters in this thesis, and has now been commercialised.

▼ No presente trabalho apresentou-se o emprego de sensores e de biossensores eletroquÃmicos para a determinaÃÃo dos pesticidas endossulfam (EDS), tiabendazol (TBZ) e cloridrato de formetanato (FMT) em amostras de cana-de-aÃÃcar, tomate, manga, uva e Ãguas naturais. AlÃm disso, informaÃÃes acerca do mecanismo redox dos pesticidas EDS, TBZ e FMT tambÃm foram apresentadas. Os sensores consistiram em eletrodos de gota pendente de mercÃrio (HMDE, do inglÃs Hanging Mercury Drop Electrode), de diamante dopado com boro (EDDB) e de carbono vÃtreo modificado com nanotubos de carbono de paredes mÃltiplas funcionalizados e ftalocianina de cobalto (CV/NTCPMF-FcCo-Naf.0,5%). Para os biossensores, a enzima Lacase (Lac) foi imobilizada pelo mÃtodo de ligaÃÃes cruzadas sobre ouro previamente modificado com nanopartÃculas de ouro (Au/NpAu/Lac-Glu.2%) e sobre eletrodo modificado com NTCPMF-FcCo (CV/NTCPMF-FcCo-Naf.0,5%/Lac-Glu.2%). A reduÃÃo do EDS foi regida por processo quase-reversÃvel. TBZ sofreu oxidaÃÃo irreversÃvel sobre EDDB e CV/NTCPMF-FcCo-Naf.0,5%. FMT sobre EDDB apresentou oxidaÃÃo irreversÃvel do tipo quÃmico-eletroquÃmico. No entanto, mecanismo eletroquÃmico-quÃmico-eletroquÃmico foi observado para FMT sobre CV/NTCPMF-FcCo-Naf.0,5%, totalizando quatro processos de oxidaÃÃo (um irreversÃvel, dois quase-reversÃveis e um reversÃvel). Os parâmetros otimizados na fabricação da tinta foram a composição de 60% grafite e 40% poliuretana (m/m), a natureza e quantidade de solvente, dimensão do conjunto impresso, quantidade de demãos aplicadas no processo de \"screen-printing\", espessura da máscara adesiva e o tipo de isolante que define os contatos elétricos e a área ativa do eletrodo. Também foi proposto e desenvolvido um procedimento de ativação da superfície do eletrodo, que consiste em uma compressão do compósito, seguido de tratamento eletroquímico da superfície do eletrodo de trabalho, que se mostraram fundamentais para o desempenho adequado do sensor. Os resultados desses estudos inciais foram avaliados usando K3[Fe(CN)6], como sonda eletroquímica. Em seguida, o EIGPU foi avaliado quanto ao seu potencial para determinação de analitos de interesse farmacológico, usando como modelo o paracetamola (APAP) e suas misturasb com cafeína (CAF) em amostras de fármacos (LOD=0,81a, 0,84b(APAP), 1,6b(CAF) µmol L-1); ambiental: Zn2+, Pb2+, Cu2+ e Hg2+ simultaneamente em amostras de etanol combustível (LOD=300, 65, 30, 46 nmol L-1, respectivamente) e biológico: epinefrina (EP) em amostras artificiais de fluido cérebro-espinal sintético (LOD=42,8 e 768 nmol L-1, para DPV e SWV, respectivamente). Para isso, foram usadas Voltametria Cíclica (CV), Voltametria de Pulso Diferencial (DPV), Voltametria de Pulso Diferencial de Redissolução Anódica (DPASV) e Voltametria de Onda Quadrada (SWV). Na determinação dos analitos de interesse ambiental, os eletrodos impressos foram modificados com sílica SBA-15, antes e após organofuncionalização com a espécie benzotiazol-2-tiol. Os compósitos modificados foram preparados substituindo quantidades correspondentes de pó de grafite de modo a obter eletrodos com 5% (m/m) do modificador. Antes de realizar as análises, foram feitas as otimizações dos parâmetros envolvidos em cada técnica escolhida e obtidas as curvas analíticas. Em seguida, o EIGPU foi utilizado na determinação dos analitos nas respectivas matrizes. A quantidade dos analitos presentes nas amostras foi determinada pelo método de adição de padrão, em triplicata. Os resultados foram comparados com aqueles obtidos por Cromatografia Líquida de Alta Eficiência (HPLC) e Espectroscopia de Absorção Atômica (AAS), para as análises farmacêuticas e ambientais, respectivamente.

▼ O ácido cis-11-metil-2-dodecenóico (DSF), substância utilizada como autoindutor nos sistemas de Quorum Sensing da bactéria Xanthomonas axonopodis pv. citri, foi utilizado como um modelo de estudos para a análise de moléculas secretadas por populações celulares utilizando técnicas eletroanalíticas. Cultivos celulares provenientes de três linhagens diferentes da bactéria Xanthomonas axonopodis pv. citri foram utilizados para os estudos. Uma linhagem selvagem (WT), que produzia DSF em quantidades naturalmente observadas, e duas linhagens geneticamente modificadas. Em uma linhagem (ΔC) a modificação genética resultava em elevada produção de DSF, para a segunda (ΔF) o resultado era a anulação da produção do autoindutor. As principais técnicas analíticas utilizadas foram: voltametria cíclica, eletroforese capilar e cromatografia líquida (com detecção UV-Vis e acoplada a espectrometria de massas), sendo a última para efeitos de comparação. Procedimentos de extração líquido-líquido com solventes orgânicos foram realizados como etapas de pré-tratamento de amostras, visando diminuir a quantidade de interferentes e realizar uma pré-concentração do analito. Três metodologias principais foram adotadas. Uma primeira voltada para a diferenciação dos cultivos celulares, analisando-se ou os sobrenadantes provenientes dos cultivos ou os extratos resultantes dos pré-tratamentos desses sobrenadantes, tendo como principal foco a identificação de um sinal diferencial entre os cultivos que correspondesse às quantidades de DSF, ou seja, inexistente para a linhagem ΔF, de alta intensidade para linhagem ΔC e de intensidade intermediária para a linhagem WT. A segunda metodologia foi baseada no uso de uma molécula modelo para realizar experimentos e obter dados como parâmetros físico-químicos, o ácido láurico (ácido dodecanóico) foi escolhido por sua semelhança estrutural e facilidade de obtenção, curvas de calibração e experimentos de separação de moléculas com estrutura semelhante foram realizados com a técnica de eletroforese capilar. Uma terceira metodologia foi desenvolvida utilizando o padrão do ácido cis-11-metil-2-dodecenóico (DSF), curvas de calibração foram obtidas com o padrão isolado apresentando ótimas linearidades, tanto com a técnica de eletroforese capilar quanto com cromatografia líquida com detecção UV-Vis, experimentos de adição de padrão às amostras também foram executados. Os experimentos realizados com as amostras não apresentaram os resultados esperados, os problemas foram associados principalmente à quantidade de interferentes na amostra e a baixa concentração do analito nos cultivos, sendo que os métodos de extração realizados não foram suficientes para solucioná-los. ▼ Genetics tests and assays have enormous scope of applications in biotechnology and medicine. Implementation of these series of monotonous biochemical reaction steps in microfluidic devices has lead to benefits in time, flexibility of operation, and cost of the reagents and systems. The main objective of this thesis is the development of components and related technologies for the realization of an integrated microfluidic system for these assays. Some of the critical technologies required are: (a) a detector to convey information from the molecular level to user level, (b) microvalves for control and isolation of reagents in the microchip and (c) micropumps for the transport of nanovolumes of the reagents across the chip. All the three critical areas have been analyzed and developed. An inline electrochemical detector, capable of detecting 14.5 amol of electroactive Ferrocene carboxylic acid (FeAc), has been developed. It uses a novel electrochemical cell and supporting electronics design to eliminate the problem of gas evolution. FeAc was also tagged to calf thymus DNA sample and the DNA detected. A novel electrokinetic pumping mechanism has also been developed, which uses scaling in microsystems to eliminate pressure flow and allow electrokinetic flow, also eliminating gas evolution normally associated with electrokinetic pumping by using the non-linearities in the system. Pumping velocities of 10 mum/s have been achieved. An inline microvalve using Paraffin as the microactuation material has been developed. Burst pressures of 23 psig have been demonstrated. The valve is found to fully close at power as low as 35 mW. While the devices presented here are stand-alone devices, the direction of research has been highly influenced by the goal of incorporating these devices into a complete genetic analysis system. Hence the fabrication techniques used are all low temperature processes; the materials are compatible, plastic-based, and the devices easily integrable.

▼ Il y a un intérêt croissant concernant la découverte de nouvelles méthodes commercialement viables pour réaliser des analyses spectro-électrochimiques — combinant des techniques électrochimiques et spectrales. Pour ce faire, nous avons préparé un matériau d'électrode transparent et conducteur, l’oxyde d’indium dopé à l’étain. Nous avons utilisé la technique d’électrofilage conduisant à la formation de fibres très fines avec une surface spécifique élevée. Ces électrodes ont ensuite été recouvertes d'une couche supplémentaire de silice poreuse et fonctionnalisée pour maximiser la surface spécifique et introduire des propriétés de détection supplémentaires. Le dispositif a été utilisé dans la détection du bleu de méthylène qui est un colorant industriel mais également un polluant environnemental. il a été mis en évidence qu’avec l'utilisation d'une telle électrode, il était possible de détecter des concentrations inférieures aux niveaux environnementaux nocifs. Enfin, les électrodes fonctionnalisées ont également été utilisées avec succès pour générer une luminescence plus intense et plus stable, ce qui ouvre de nouvelles perspectives pour la conception de capteurs spectroélectrochimiques
There is a growing interest in finding new and commercially viable methods of performing a spectroelectrochemical analysis which combines electrochemical and spectral techniques. For this purpose, an electrode material that is transparent and conductive needs to be prepared. In this work, such electrode was prepared by electrospinning which is a technique capable of forming very thin fibers with high surface area. Those electrodes were also covered with additional layer of porous and functionalized silica to maximize the surface area and introduced additional sensing properties. This material was used in the detection of methylene blue which is an industrial dye and an environmental pollutant. It was found that using such electrode it was possible to detect concentrations that are smaller than the harmful environmental levels. Finally, the layers were also used with success to generate luminescence which is opening new prospects for the design of spectroelectrochemical sensors

▼ The main purpose of this work - to
investigate patterns of electrochemical oxidation of ascorbic acid
on polyaniline and poly(N-methylaniline) modified electrodes, in
order to develop ascorbate-sensitive sensors. A detailed study of
various factors affecting the aniline and N-methylaniline
electrochemical polymerization and the resulting properties of PANI
and PNMA layers was carried out for this purpose. Comparative study
of modified electrodes in solutions of different acidity was
performed and it was shown that PNMA had a better redox activity in
slightly acidic and neutral solutions compared to polyaniline. The
nature of amperometric response of modified electrodes to ascorbate
was investigated and autocatalytic mechanism of ascorbate
electrooxidation on PANI modified electrode was suggested. Using
PANI and PNMA modified electrodes, prototypes of amperometric
ascorbate sensors have been developed and their comparative studies
were carried out.

elektrocheminės oksidacijos ant elektrodų, modifikuotų polianilinu
ir poli(N-metilanilinu), dėsningumus, siekiant sukurti jautrius
askorbatui jutiklius. Ištirta įvairių faktorių įtaka anilino ir
N-metilanilino elektrocheminei polimerizacijai bei gautų PANI ir
PNMA sluoksnių savybėms. Atlikti palyginamieji modifikuotų
elektrodų tyrimai skirtingo pH tirpaluose ir parodyta, kad PNMA
lyginant su polianilinu pasižymi geresniu aktyvumu silpnai
rūgščiuose ir neutraliuose tirpaluose. Ištirtas modifikuotų
elektrodų amperometrinio atsako į askorbatą pobūdis, ir pasiūlytas
autokatalizinis askorbato elektrooksidacijos mechanizmas ant PANI
modifikuoto elektrodo. Panaudojus PANI ir PNMA modifikuotus
elektrodus, sukurti amperometrinių askorbato jutiklių prototipai ir
atlikti jų palyginamieji tyrimai.

elektrodus, sukurti amperometrinių askorbato jutiklių prototipai ir
atlikti jų palyginamieji tyrimai.
The main purpose of this work - to
investigate patterns of electrochemical oxidation of ascorbic acid
on polyaniline and poly(N-methylaniline) modified electrodes, in
order to develop ascorbate-sensitive sensors. A detailed study of
various factors affecting the aniline and N-methylaniline
electrochemical polymerization and the resulting properties of PANI
and PNMA layers was carried out for this purpose. Comparative study
of modified electrodes in solutions of different acidity was
performed and it was shown that PNMA had a better redox activity in
slightly acidic and neutral solutions compared to polyaniline. The
nature of amperometric response of modified electrodes to ascorbate
was investigated and autocatalytic mechanism of ascorbate
electrooxidation on PANI modified electrode was suggested. Using
PANI and PNMA modified electrodes, prototypes of amperometric
ascorbate sensors have been developed and their comparative studies
were carried out.

▼ Graphene, as a recently discovered carbon allotrope, possesses with it many outstanding properties ranging from high electrical conductivity to great mechanical strength. Single layer graphene can be prepared by mechanical cleavage of graphite or by a more sophisticated method, CVD. However, the scale-up process for these preparation techniques is still unconvincing. Solution-processed graphene from exfoliation of graphite oxide on the other hand provides an alternative prospect resulting in the formation of graphene nanoplatelets (GNPs), which can be readily manipulated to tailor-suit various application demands. The main aim of the thesis is to explore the possibility and availability of this versatile method to produce graphene nanoplatelet and its composites with good all-round performance in energy and bioanalytical applications. A range of physical and chemical characterisation techniques were utilised including SEM, TEM, AFM, XPS, XRD, DLS, FTIR, Raman and UV-Vis spectroscopy in order to investigate the structural and chemical information of the graphene-based materials prepared. Functionalisation of graphene oxide with polyelectrolyte polymer could facilitate deposition of platinum nanoparticles in the formation of Pt-GNPs composites. The resultant composite was employed for bioanalytical application in the detection of an important neurotransmitter, glutamate, based on glutamate oxidase enzyme. The performance of Pt-GNPs based glutamate sensor exhibited enhanced sensitivity and prolonged stability compared to the sensors based on Pt decorated diamond or glassy carbon electrodes. The significant interfering effect from concomitant electrochemically active biological compounds associated with Pt-GNPs electrode however could be alleviated via opting for Prussian blue deposited GNPs electrode instead. The oppositely charged Pt-GNPs due to different functionalising polymers were also subject to self-assembly, which was enabled by the electrostatic interaction of the opposite charges of Pt-GNPs. The self-assembled film showed enhanced mechanical stability than the conventional drop-casted film and provided reasonably good activity towards oxidation of hydrogen peroxide. Three-component composite of graphene, nanodiamond and polyaniline was prepared via in-situ polymerisation for usage as an electrode material in electrochemical capacitors ("supercapacitors"). The addition of graphene was shown to significantly enhance specific capacitance while nanodiamond could improve the stability of the electrode by strengthening the polymer core. Another approach to produce a supercapacitor was via electrodeposition of nickel and cobalt hydroxides on graphene oxide film corporated with bicarbonate salt. The film was then subject to thermal reduction of GO and expansion of graphene layers within the film was observed. This leavening process enhanced the surface area of graphene film and thus the higher specific capacitance was obtained. ▼ The present dissertation was focused on the topic of chemical sensors for rapid detection of toxic substances, an investigation that was made for first time in the literature. For this purpose, biosensors using printed circuits composed from graphene microelectrodes using stabilized lipid membranes were constructed and developed.A number of particular methodologies were used to embody various receptors such as enzymes (urease and cholesterol oxidase) and an antibody (antibody D-dimer) in the sensors.Various physicochemical and instrumental methods of analysis, eg., differential scanning calorimetry (DSC), scanning electron microscopy (SEM), Raman spectrophotometry, etc., were used to locate the position of the receptor (eg. antibody, enzyme, etc.) into the stabilized lipid membrane of the sensor and the mechanism of signal generation.The response of the sensor to various potentially toxic substrates eg. urea (urease immobilization), cholesterol (cholesterol oxidase immobilization) and D-dimer (immobilizing antibody D-dimer) and its stability with time were studied.For example, the response of urea biosensor towards various urea concentrations was found to have high sensitivity (ca. 70 mV/ concentration decade) over a range of urea concentrations between 1 Χ 10−6 Μ - 1 Χ 10−3 Μ. The response of cholesterol biosensor towards various concentrations of the substrate were also found to be highly sensitive (ca. 64 mV/ concentration decade) over a concentration range between 1 Χ 10−6 Μ - 1 Χ 10−3 Μ; this sensitivity is larger than those provided up to date in the literature. Finally the response of the D-dimer immunosensor was found satisfactory over a wide concentration range of analyte (10-6 μg/ mL - 10-3 μg/ mL) with a sensitivity of ca. 59 mV/ concentration decade with a rapid response of 15 s.The selectivity of the sensor construction method and on how to limit interferences were studied so that the sensor could be applied in real samples. The conditions for satisfactory stability and reproducibility of the chemical sensors were thoroughly studied and their construction into nanoscale was investigated.The results of the implementation of the constructed devices in real samples were satisfactory and show the possibility of construction of portable chemical sensors for the detection of toxicants for first time in the literature with many advantages, which can be used in the future as alternative systems instead of the time consuming, costly and complex chromatographic methods of analysis.

▼ Um sistema completo foi desenvolvido para o novo acoplamento de uma célula eletroquímica em fluxo com um eletroforese capilar equipado com detector condutométrico sem contato (EC-CE-C4D). Este acoplamento compreendeu o design da interface EC-CE, um potenciostato protegido contra fuga de alta tensão, subsistema de controle de fluxo por bombas solenóide e um equipamento CE-C4D adaptado para a aplicação. Um software dedicado para aquisição de dados, controle e operação do sistema foi desenvolvido na ferramenta gráfica de programação LabView. O sistema foi inicialmente aplicado na eletrodeposição de traços dos metais Cd2+, Pb2+, Cu2+, Zn2+ e Tl+, seguida pela redissolução e separação destes por CE, o que resultou em fator de pré-concentração de até 55 vezes para todos os metais, exceto Zn2+. Os metais acumulados foram redissolvidos na matriz original da amostra para permitir a separação e determinação conjunta com cátions majoritários (p. ex. alcalinos e alcalinos terrosos e amônio, se acima do LD). Para amostras cuja matriz possa causar interferência, pouco antes da injeção no CE, foi realizado processo de cleanup pela redissolução dos metais no eletrólito de separação. Experimentos pra separação bidimensional com o potencial de deposição ou redissolução voltamétrica como primeira dimensão e o tempo de eletromigração como segunda dimensão foram propostos e demonstrados. A oxidação eletroquímica de espécies orgânicas na interface EC-CE surgiu como um segundo campo de aplicação para: a) derivatização de analitos neutros tornando-os carregados ou ionizaveis, p. ex., conversão de álcoois alifáticos de cadeia curta em carboxilatos que possam ser separados e quantificados por CE-C4D; b) investigação/monitoramento de processos de eletrodo pela detecção de produtos carregados ou ionizáveis de reação de eletrodo, p. ex., investigação de condições experimentais para eletrooxidação de glicerol objetivando-se a síntese de produtos com maior valor comercial.
A complete instrumental system was developed for the new coupling of an electrochemical flow cell to capillary electrophoresis with contactless conductivity detection (EC-CE-C4D) and comprising an innovative design of the EC-CE interface, a potentiostat circuit protected from high voltage leakages, a solenoid pump based flow management subsystem, and a CE-C4D equipment tailored for this application. Dedicated software for data acquisition, devices control and operation automation was developed in LabVIEW graphical programming tool. The system was first applied to the electrodeposition of traces of Cd2+, Pb2+, Cu2+, Zn2+ and Tl+ , followed by stripping and separation by CE, allowing a 55-fold preconcentration factor in 6 min for all metals but Zn2+. The accumulated metals were stripped into the original sample matrix to allow their separation together with the sample\'s major cations (e.g. alkaline and alkaline earth metals and ammonium, if above LOQ). ▼ O estudo de polímeros condutores desperta um enorme interesse científico e tecnológico devido às diversas aplicações possíveis para esses materiais, como o desenvolvimento de dispositivos eletro-eletrônicos e sensores. Através de processos de dopagem esses polímeros tornam-se eletroativos, possibilitando por meio de técnicas eletroquímicas a determinação de diversas substâncias. Na agropecuária, a utilização intensiva e crescente de produtos químicos como pesticidas e fertilizantes, tem provocado diversas contaminações e agressões, despertando interesse na utilização de sensores para o monitoramento dessas substâncias em tempo real. Os eletrodos modificados com polímeros condutores têm-se apresentado como uma alternativa devido a algumas peculiaridades como alta estabilidade física e química e excelentes possibilidades analíticas devido à versatilidade da polimerização eletroquímica. Este trabalho teve como objetivo o desenvolvimento de novos materiais de eletrodos modificados com polímeros condutores para a determinação eletroanalítica de pesticidas. Os pesticidas estudados foram os herbicidas 2,4-D, bentazon, glifosato, paraquat e o inseticida paration metílico. Foram realizadas medidas voltamétricas e de impedância com eletrodos de carbono vítreo, pasta de carbono e eletrodos modificados com polianilina e polipirrol, visando a obtenção de sensores para a detecção destes pesticidas. Estudos de interação dos pesticidas com os polímeros foram realizados utilizando-se a técnica de espectrofotometria de UV-visível para a determinação da concentração dos pesticidas em estudos cinéticos. Também utilizou-se a espectroscopia de impedância eletroquímica para auxiliar no entendimento das interações entre os pesticidas e os polímeros condutores. Adicionalmente foi aplicada uma metodologia de análise de fluxo descontínuo (BIA), utilizada com sucesso na determinação de metais, para a determinação de pesticidas. Os resultados mostraram uma seletividade nas interações entre os polímeros condutores e os pesticidas. Por meio da resposta voltamétrica dos polímeros condutores, os eletrodos de pasta modificados com polianilina e polipirrol mostraram-se hábeis na detecção dos pesticidas não eletroativos como o 2,4-D, o bentazon e o glifosato, bem como na determinação do paraquat. Estes resultados atribuem aos eletrodos modificados desenvolvidos neste trabalho a característica de sensores de diagnóstico da presença destes pesticidas em solução. A metodologia de BIA na determinação de pesticidas, apresentou inúmeras vantagens do ponto de vista analítico, como a não renovação da superfície de eletrodo, micro-volumes de injeção de amostra, dezenas de análise sem troca do eletrólito suporte, tempos de análise inferiores a um segundo bem como a possibilidade de utilização de dispositivo portátil para análises em campo.

▼ Se han desarrollado métodos analíticos que permiten la caracterización de compuestos bioactivos presentes en el corcho procedente del alcornoque Quercus suber L. Se han aplicado técnicas de HPLC desarrollando nuevos métodos para el análisis de compuestos fenólicos de bajo peso molecular y elagitaninos, así como para el aislamiento y purificación de estos compuestos mediante cromatografía preparativa. Se han propuesto diferentes procesos de extracción de compuestos fenólicos de bajo peso molecular, así como de elagitaninos en muestras de corcho. Por otro parte, se han desarrollado métodos para la determinación del contenido de fenoles totales en aguas de cocido de corcho mediante técnicas electroanalíticas y utilizando diferentes tipos de electrodos de trabajo tales como: carbón vitrificado y electrodos impresos modificados con grafeno. Las técnicas utilizadas han sido voltamperometría de diferencial de pulsos (DPV) y redisolución adsorptiva (AdSDPV). El contenido de fenoles totales de bajo potencial de oxidación se ha determinado expresándolo como equivalentes de ácido gálico o elágico (dependiendo del tipo de electrodo utilizado). Por otra parte, mediante análisis de inyección de flujo, se propone un método para la determinación del contenido total de fenoles en agua de cocido de corcho. Se han comparado los resultados obtenidos mediante DPV, AdSDPV y FIA con detección amperométrica, con aquellos obtenidos del análisis de las mismas muestras utilizando el método espectrofotométrico utilizando el reactivo Folin-Ciocalteau. Se ha encontrado una buena correlación entre los diferentes métodos y el espectrofotométrico, si bien el contenido de fenoles es mayor cuando se hace uso del método espectrofotométrico, dado que éste último es menos selectivo.

▼ This thesis is concerned with the deposition of nanoparticle films onto boron-doped diamond and tin-doped indium oxide (lTO) surfaces and the characterisation of the films using electron microscopy, powder diffraction methods and quartz crystal microbalance (QCM) data. The redox behaviour of the porous films was examined using cyclic voltammetry in various media to investigate potential electroanalytical applications. TiOz (anatase) mono-layer films were immobilised onto an inert boron-doped diamond substrate. Cyclic voltammetry experiments allowed two distinct steps in the reduction - protonation processes to be identified that are consistent with the formation of Ti(III) surface sites accompanied by the adsorption of protons. Preliminary data for electron transfer processes at the reduced TiOz surface such as the dihydrogen evolution process and the 2 electron - 2 proton reduction of maleic acid to succinic acid are discussed. Novel multi-layer TiOz films were deposited with a variety of organic binder molecules onto ITO substrates. The redox reactivity of Cuz+ with 1,4,7,10- tetraazacyclododecane- 1,4,7, IO-tetrayl- tetrakis (methyl-phosphonic acid) in solution and immobilised on an electrode surface are investigated. The influences of film thickness, scan rate, and pH on the electrochemistry of immobilised pyrroloquinoline quinone was investigated with two possible electron transport processes observed. The thickness of TiOz phytate films was found to change the shape of the resulting cyclic voltammograms dramatically. Computer simulation and impedance spectroscopy allowed insights into the diffusion of electrons to be obtained. 1, 1 ~Ferrocenedimethanol was employed as an adsorbing redox system to study the voltammetric characteristics of carboxymethyl-y-cyclodextrin films and evidence for two distinct binding sites is considered. The apparent transport coefficients for dopamine and Ru(NHJ)6J+ are estimated for TiOz Nafion® films. The electrochemical processes in biphasic electrode systems for the oxidation of water-insoluble N,N-didodecyl-N;N~diethyl-benzene-diamine (DDPD) pure and dissolved in di-(2-ethyl-hexyl)phosphate (HDOP) immersed in aqueous electrolyte media are described. Transfer of the anion from the aqueous electrolyte phase into the organic phase accompanies the oxidation of pure DDPD. In the presence of HOOP, oxidation is accompanied by proton exchange. The electrochemically driven proton exchange process occurs over a wide pH range. Organic microdroplet deposits of OOPD in HDOP at basal plane pyrolytic graphite electrodes are studied using voltammetric techniques and compared to the behaviour of organic microphase deposits in mesoporous Ti02 thin films. Two types of Ti02 thin film electrodes were investigated, (i) a 300-400 nm film on ITa and (ii) a 300-400 nm film on ITa sputter-coated with a 20 nm porous gold layer. ▼ The research presented in this thesis firstly concerns the use of electrochemical techniques to develop approaches to wool processing which have a lower impact on the environment than conventional chemical methods. Wool is a sulfur rich substrate and current methods used in wool processing often rely on sulfur-based reducing agents such as metabisulfite. However, due to increasing concern over the environmental impacts of metabisulfite, alternative methods are of interest. Electrochemical techniques have been applied to the process of wool setting in the presence of thiol setting agents. Wool disulfide bonds are reduced during this process and the thiol setting agent is converted to the disulfide. Efficient conversion of the disulfide back to the thiol setting agent would allow catalytic amounts of thiols to be used in wool setting. The electroreduction of cystine and 2-hydroxyethyl disulfide has been examined at a range of metal and carbon electrodes to find efficient methods of generating the corresponding thiols, cysteine and 2-mercaptoethanol respectively. Gold and silver were identified as the most efficient electrode materials. In industrial wool processing, the use of large-scale metal electrodes is expensive and therefore, high surface area gold and silver nanoparticle electrodes were fabricated by electrochemically depositing the metals onto low-cost carbon substrates. The most efficient electrochemical system for generating the thiol setting agent involved the electroreduction of cystine at the gold nanoparticle electrode and this system was used to successfully demonstrate that wool setting can be achieved using relatively low concentrations of cysteine. Further research was carried out to investigate methods for the controlled preparation of metal nanoparticle electrodes and their utility for detecting hydrogen peroxide was examined. A simple and versatile approach for the preparation of tethered gold nanoparticle assemblies was developed by exploiting electrostatic interactions between citrate-capped gold nanoparticles and amine tether layers attached to carbon surfaces. The nanoparticle assemblies were optimised for the detection of hydrogen peroxide by selecting the size and density of electrostatically assembled nanoparticles. The number of amine functionalities on the surface and the assembly conditions controlled the nanoparticle density. Nanostructured palladium electrodes fabricated using vapour deposition methods to immobilise palladium nanoparticles directly onto carbon substrates were also examined for the electroanalysis of hydrogen peroxide.

▼ O presente trabalho descreve uma forma inédita e simples de construção de eletrodos de ouro (CDtrodos) a partir de discos compactos graváveis (CD-Rs). Os CD-Rs possuem em sua constituição uma película de ouro com uma espessura entre 50 e 100 nm e com área aproximada de 100 cm2. Esta superfície de ouro pode ser facilmente utilizada para construção de eletrodos convencionais usados em células estacionárias ou em células em fluxo. Além disso, a espessura nanométrica do filme de ouro também pode ser utilizada para construção de microeletrodos de banda com áreas inferiores a 10-6 cm2. Devido ao fato do CD-R ser produzido em escala comercial, o custo do mesmo é muito baixo, tornando possível sua utilização na forma de eletrodos descartáveis. Para comprovar a viabilidade da utilização destes filmes na confecção de eletrodos, seu desempenho foi comparado com o de eletrodo de ouro comercial, sendo observado um comportamento análogo. Diferentes maneiras de utilização do disco compacto gravável foram testadas, tanto em amostras sintéticas, como também, em amostras reais. Utilizando o método de redissolução potenciométrica à corrente constante (CCPSA), foram analisadas amostras de mercúrio em água potável e em água do mar, adaptando o CD-R a uma célula de injeção em fluxo. Nestes estudos, o limite de detecção de mercúrio situou-se em 250 ng L-1, utilizando-se tempo de deposição de 300 segundos. Estudos preliminares em células estacionárias levaram a limites de detecção inferiores. Em célula estacionária, foram realizadas análises de cobre em aguardente e em água potável. Em alguns casos, os resultados obtidos com as amostras reais foram comparados com outra técnica (E.A.A.), onde os resultados obtidos foram muito próximos a àqueles obtidos com os eletrodos de ouro. Análises em água de abastecimento, mostraram que em células estacionárias o limite de detecção para cobre situa-se em 30 ng L-1, para tempos de deposição de 600 segundos.
This work describes an original and novel form of construction of gold electrodes (CDtrodes) from recordable CDs (CD-Rs). The CD-Rs contain in its constitution a fine gold layer with a thickness from 50 to 100 nm and an approx. 100 cm2 of area. This surface can be easily used for construction of conventional electrodes for stationary or flow electrochemical cells. Moreover, the nanometer thickness of the gold film can be employed for the construction of microelectrodes with areas smaller than 10-6 cm2. Due to the fact that CD-Rs are produced on commercial scale, the cost of each unit is very low, a feature that enables the use of these electrodes in disposable form. To prove the suitability of these films to make gold electrodes, their performance was compared with the response of a commercial electrode, and a similar behavior was observed.
