▼ Mammalian reproduction is mediated by the hypothalamic- pituitary-gonadal axis. The gonadotropin-releasing hormone (GnRH) neuron is the final central output of the hypothalamic portion of this axis, and the proper development and function of this neuronal type is required for normal fertility. Several genes have been identified as necessary for GnRH neuron development and function, including the homeodomain protein Six6, a homologue of Drosophila optix. Mice lacking the Six6 gene have several previously identified phenotypes—predominantly phenotypes of the eye and retina – and additional phenotypes are described here. Male and female Six6-/- mice have impaired fertility due, in part, to significantly reduced numbers of GnRH neurons relative to wild-type littermate control animals. Additional phenotypes identified in the Six6-/- mice were circadian- and suprachiasmatic nucleus- (SCN-) related. Six6-/- mice displayed a variety of circadian behavioral activity patterns, with some mice showing periodicity during constant darkness, and other mice completely lacking circadian periodicity even in entrainment photoperiods. However, regardless of behavioral phenotype or the previously identified optic nerve phenotype, Six6-/- mice lacked suprachiasmatic nuclei as indicated by immunostaining for prototypical SCN markers and observation of hypothalamic morphology. The circadian clock controls many aspects of mammalian physiology, including reproduction in many species. Inactivation of the circadian clock in mice, via knock-out of Bmal leads to infertility, though the mechanisms underlying this infertility remained incompletely understood. Using transgenic, physiological, and behavioral techniques, the reproductive phenotype of Bmal-/- mice was investigated. Tissue-specific inactivation of Bmal in all neurons or in GnRH neurons did not appreciably affect fertility, though neuronal Bmal inactivation resulted in a shortening of the free-running circadian period. Female Bmal-/- mice were unable to respond to estrogen stimulation with an appropriate LH surge. Male Bmal-/- mice showed severely impaired mating behavior, and even Bmal-/- mice given testosterone replacement exogenously did not show normal mating behavior

▼ Hypoxic insult during early pregnancy can give rise to neuronal disorders of the newborn, such as autism, schizophrenia and epilepsy. Of these disorders, the underlying similarity is cortical dysplasia, in which the neuronal layers are disrupted due to abnormal neuronal migration. The infliction of hypoxia during the critical period of neuronal development and migration can cause abnormalities which can propagate into these migration defects observed after birth. The purpose of this dissertation is to investigate the effect of hypoxia on early neuronal development, and the role of lysophosphatidic acid receptor-1 (LPA₁) in this pathological process. LPA₁ is one of 6 G-protein-coupled- receptors (GPCRs) which are activated by a bioactive lipid, LPA. It is highly enriched in the ventricular zone of the cortex during development, suggesting its importance in neural progenitor cells (NPCs). Previously, LPA treatment of ex vivo cortices at embryonic age 13.5, a period of neurogenesis and early neuronal migration, was reported to induce ectopic displacement of mitotic NPCs. Strikingly, in this study, a similar displacement of mitotic NPCs, together with delayed early post-mitotic neuronal migration and N-cadherin disruption, was observed in cortices exposed to hypoxia, which was obliterated in LPA₁ null cortices. This indicates that LPA₁ is an essential mediator of hypoxia in the developing cortex. Through the use of pharmacological inhibitors, it was demonstrated that hypoxia signals through LPA₁ through Gi and subsequent downstream effectors. Importantly, a novel mechanism of how hypoxia activates LPA₁ was discovered: Hypoxia inhibits GRK2, a GPCR kinase which associates with LPA₁ to cause sequestration, hence promoting LPA₁ activity. LPA signaling has been implicated in various stages of vascular development, but blood vessel integrity in these receptor knockout mice have not been investigated in depth. In a separate study, it was observed that LPA₁/ LPA₂ double null mice exhibit an increased blood vessel leakage and blood vessel density specifically in the cortex during the period around birth persisting into early postnatal life. Also, these cortical endothelial cells appear to be unhealthy and electron-light. These striking phenotypes could shed light on the role of LPA signaling in blood brain barrier formation in the brain

▼ Information processing in the brain depends upon the interactions of diverse signaling units, neurons, which vary in their circuit positions, biophysical and biochemical properties, and electrophysiological phenotypes. These distinguishing properties enable neurons to serve different computational and functional roles. For instance, the fast-spiking inhibitory subclasss, which synchronizes its firing at fast frequencies and makes divergent and convergent contacts onto the persimomatic compartments of its targets, is hypothesized to play an important role in regulating the timing and probability of spike output and thus the flow of information through the brain (reviewed in Chapter 1). However, the functional consequences of synchronized inhibition will depend on the integrative properties and circuit locations of the recipient neurons. Using a combination of in-vitro whole cell electrophysiology, modeling, and model-neuron hybrids (dynamic clamp), we determined how fast synchronized inhibition interacts with integrative properties of a variety of neuronal types to regulate the rate (Chapter 2) and timing (Chapter 3) of spike generation. We find that a neuron's intrinsic physiology substantially affects the ability of fast synchronized inhibition to control neuronal responsiveness. In addition, we demonstrate how the relevant physiology can be flexibly altered by contextual and neuromodulatory factors. The results of these experiments suggest that synchronized fast-spiking activity can differentially affect various circuit elements and each element's responsiveness can be adjusted on a range of time scales to suit the cortex's changing computational requirements. Because the brain is one of the most energetically expensive organs in the body with action potential generation accounting for significant portion of the energy usage, we hypothesized that differences in neuronal properties may also serve to minimize energy consumption subject to functional constraints. Again using electrophysiology, modeling, and dynamic clamp, we compared the energy needed to produce action potentials singly and in trains for a wide range of channel densities and kinetic parameters, and examined which combinations of parameters maximized spiking function while minimizing energetic cost (Chapter 4). We found evidence supporting our hypothesis in a wide range of neurons from several species. We conclude that neuronal biophysics are tuned to perform cost-effective functions

▼ This dissertation has three objectives : (1) assess agreement of injury data reported in two passive surveillance systems at the Marine Corps Recruit Depot, San Diego; (2) examine self-reported smoking as a risk factor for overuse injury among Marine Corps recruits; and (3) identify self-reported risk factors for poor training outcomes among Navy recruits. Five measurements of agreement were computed separately for sprains/strains, fractures/stress fractures, and all injuries between the two surveillance systems (objective 1). Recruits completed a baseline risk factor survey and were followed for overuse injury outcomes (objective 2), or graduation status (objective 3). Results : Objective (1) The data sets have a high proportion of negative agreement and low proportion of positive agreement, especially for fractures /stress fractures. The proportion of overall agreement between the two surveillance systems was extremely high for sprains/strains, and for fractures/stress fractures, and moderate for all injuries. Objective (2) Adjusted proportional hazard regression did not find a significant association with respect to smoking and overuse injury in either male or female Marine Corps recruits. Objective (3) Male Navy recruits who did not run or jog at least 1 month before basic training or had a previous lower limb injury without complete recovery, and females reporting the same or less physical activity compared to their same age counterparts, were more likely to have a poor training outcome. Conclusions : Objective (1) The large offsite surveillance system is better suited to provide historical baseline and post prevention injury rates than the locally maintained surveillance system, and therefore provide a better comparative estimate of the magnitude of each injury type after interventions are made. The local surveillance system is better suited to provide meaningful injury specific severity and exposure data needed to identify the impact and mechanism of injuries, information that can be used to introduce preventive measures in the field. Objective (2) Smoking does not appear to be an independent risk factor for overuse injury in either male or female Marine Corps recruits. Objective (3) An important first step in decreasing poor training outcomes in Navy recruits is to encourage incoming recruits to participate in physical activity, and take steps to identify and rehabilitate recruits who are not completely healed from a lower limb musculoskeletal injury before reporting to basic training

▼ Cardiac tissue engineering has the potential to develop regenerative therapies for damaged myocardium, but due to the complex structure and composition of the tissue in both healthy and diseased states, a clinical solution has not been realized. Human embryonic stem cells (hESCs) and human induced pluripotent stem cells (hiPSCs) are self- renewing, pluripotent cell sources that can potentially generate an unlimited number of cardiomyocytes (CMs). However, the clinical application of these cells is hampered by the limited knowledge of 1) the long-term ability of transplanted cells to maintain a therapeutic effect and 2) the eventual fate of these still very immature cells. Identifying the biochemical and physical factors that govern cardiac maturation and understanding how they influence this process will be imperative to propel this field forward. This dissertation describes the influence of endothelial cells on major aspects of hiPSC- derived CM physiology and their functional maturation. To date, neither repopulation of lost myocardial mass nor functional recovery of cardiac infarct regions has been demonstrated after transplantation of human pluripotent stem cell (hPSC)-derived CMs. Strikingly, even after long- term incubation, transplanted CMs remain morphologically and physiologically immature. To this end, we postulate the importance of endothelial influences on CM maturation based on their intimate association from the formation of the early heart tube through adulthood. In the subsequent chapters, we demonstrate the maturative influences of co- culture hESC-derived CMs with human umbilical vein endothelial cells (HUVECs) on the electrophysiology and gene expression profiles towards a more mature phenotype. Moreover, we investigate the specific effect of endothelial paracrine factors on Ca²⁺-handling machinery by evaluating changes in kinetic parameters and pharmacological sensitivity. Ultimately, we demonstrate that treatment of hiPSC-derived CMs with endothelial conditioned media increases 3Hz pacing ability, Ca²⁺ transient kinetics, and sensitivity to pharmacological agents that affect the functionality of Ca²⁺ reuptake and initiation of the Ca²⁺ transient. We argue that the functional maturation of hPSC-derived CMs and elucidating the mechanisms that govern these processes will significantly contribute to the therapeutic efficacy and safety of cell-based therapies for myocardial repair

▼ Tumor metastasis is a complex, multi-step process initially depending on local invasion and followed by intravasation, survival in the circulation, extravasation and colonization of a distant site. Many studies have shown that EGFR and Src signaling and its cooperation with integrins are involved throughout metastasis. However, the mechanisms by which cross-talk between RTK signaling and integrins contribute to metastasis are not well-described. EGFR signaling promotes carcinoma cell metastasis that is dependent on activation of both Src kinase and integrin [alpha]v[beta]5, but little is known about the downstream effectors contributing to EGFR-mediated metastasis. Therefore, the studies in this dissertation proposed to investigate novel downstream effectors of metastasis dependent on EGFR, Src, and integrin [alpha]v[beta]5. Lead candidate effector molecules were systematically identified using an in silico strategy and validated in a well-described chick CAM model of EGF-induced tumor cell metastasis. This approach revealed a requirement for MUC1 in spontaneous metastasis to the lungs but not primary tumor formation. MUC1 contributes to the mucous barrier protecting epithelial cells from the environment and participates in intracellular signaling cascades as a substrate for EGFR and Src. Interestingly, both extracellular and intracellular domains of MUC1 have been linked to tumor progression and metastasis. Expression of mutant MUC1 constructs revealed that the cytoplasmic domain is required for EGF-induced cell migration mediated by integrin [alpha]v[beta]5 and promotes metastasis without enhancing primary tumor growth. In biochemical and cell biological approaches, EGFR signaling enhanced MUC1 cleavage, nuclear localization and transcription of genes associated with metastasis. Genetic and pharmacological strategies revealed a requirement for Src in EGF-induced MUC1 cleavage and nuclear translocation, which were associated with sponteanous metastasis in a murine orthotopic pancreatic tumor model. Therefore, the findings described in this dissertation define a critical role for MUC1 in tumor metastasis regulating by cooperative signaling between EGFR, Src, and integrin [alpha]v[beta]5

▼ The genetic code of most organisms was evolved to encode 20 amino acids. Although the ability to encode 20 amino acids provides the basis to translate proteins necessary for life, researchers are also limited to these 20 amino acids for conventional site-directed mutagenesis. The ability to encode unnatural amino acids provides researchers the ability to circumvent the limitation imposed by the genetic code. Genetically encoding unnatural amino acids provides researchers the means to not only mimic naturally occurring posttranslational modifications but also the ability to encode amino acids with new physical or chemical properties to study biological processes. The incorporation of unnatural amino acids into proteins had been developed in Escherichia coli and also in yeast. We have developed a methodology to genetically incorporate unnatural amino acids in mammalian cells in response to an amber codon (UAG). The incorporation of unnatural amino acids is high in E. coli and yeast, but the incorporation in mammalian cells is relatively low. In addition to developing the system to incorporate unnatural amino acids in mammalian cells, we have also improved suppression efficiencies by modifying the synthetase and unnatural amino acid. To incorporate unnatural amino acids in response to an amber codon, the tRNA anticodon is mutated from a GUA to a CUA. We were able to show that engineering the anticodon-binding domain of the synthetase could enhance the recognition of the tRNA and thus increased suppression efficiencies. Furthermore, by masking the carboxyl group of the amino acid by an ester group, we were able to increase the bioavailability of an unnatural amino acid to further increase suppression efficiencies. Most evolved synthetases aminoacylate unnatural amino acids that are structurally similar to the native substrate of the wild- type synthetase. We were able to adapt Methanosarcina mazei pyrrolysine synthetase (PylRS) to charge a considerable disparate amino acid from its native substrate, O-methyl-L-tyrosine. In addition, the X-ray crystal structure was solved for the evolved PylRS complexed with O-methyl-L-tyrosine at 1.75Å. This multifaceted approach provides the basis to engineer the PylRS to incorporate a significantly diverse selection of unnatural amino acids than previously anticipated

▼ Vaccine-based immunotherapy holds promise as an effective treatment of metastatic malignancies with potential for long-term remission. The goal is to elicit Th1-type immune responses that utilize cytotoxic T lymphocytes to generate a protective antitumor response. Aluminum salts (alum), which generate predominantly Th2-type immune responses, are the sole adjuvants approved for use in the majority of countries worldwide. Thus, for significant advancement of cancer immunotherapy, there is a great need for the development of new immunostimulatory adjuvants that will induce Th1-type immune responses. This dissertation documents the research and development of several peptides that act as Th1-type adjuvants. These peptides are derived from, or based upon, an endogenous immunostimulatory protein, HMGB1. Most focus is placed on one particular peptide, named Hp91. The dissertation starts with an evaluation of the Th1 adjuvant properties of Hp91, and is followed by a study of the characteristics and mechanism of action. The remainder of the dissertation focuses on the research to find, as well as engineer, more potent adjuvant derivates based upon Hp91. Examples include shortened and mutated derivates, peptide dimers, and encapsulation within nanoparticles. A summary of the findings are presented here. The dissertation author shows that Hp91 potentiates in vivo cellular immune responses to peptide and protein antigen. Hp91 promotes the in vivo production of the Th1 cytokines, as well as antigen- specific activation of CD8+ CTLs. Hp91-induced secretion of IL-6 is MyD88- and TLR4-dependent, is mediated through p38 MAPK and NF[kappa]B, and signaling is dependent on clathrin- and dynamin-mediated endocytosis of Hp91. Additionally, Hp91 activates the MyD88-independent signaling pathway. The author demonstrates that the C- terminal domain of Hp91, named UC1018, is responsible for the adjuvant activity. UC1018 generates significant antitumor responses when used as adjuvant in a prophylactic cancer vaccine. Additionally, several Hp91 mutants showed enhanced immunostimulatory activity. The findings presented in this dissertation are significant because they identify and characterize novel, Th1-type synthetic peptide adjuvants that may allow development of new prophylactic and potentially therapeutic treatments of cancer. The results warrant continued development of these peptide adjuvants and may serve as a guide for the future engineering of synthetic Th1-type peptide adjuvants

▼ The gene MECP2 is a profound mediator of both brain structure and disease. While it is known that MECP2 causes 90% of Rett Syndrome patients in women, its role in determining both brain structure in normal populations and in the etiology of autism in boys is largely unknown. This dissertation focuses on the integration of genetic and brain imaging data, with mass-computing and statistics, to further understand the role that this crucial CNS gene plays on the outcome of human brain volume and morphology in humans. The first study examined common variation in MECP2 derived from genotyping in an ethnically homogeneous population of 300 Norwegians in various disease states from the TOP study. A highly significant association was detected, in males only, between variation in MECP2 and brain structure measures including cortical surface area and overall brain volume, along with subcortical structures. The cortical surface area association replicated in an independent sample from the ADNI study, and several specific cortical regions were significant in both studies. This finding led to the next-generation deep sequencing of MECP2 in autistic boys, in the search for both exonic and regulatory mutations that may cause or modify autism. A synonomous exonic variant and 9 3'UTR variants, undetected in controls, were discovered. Haplotype association combining data from AGRE, the Wellderly Study, and HapMap showed enrichment of a rare haplotype in autistics and a possibly protective haplotype enriched in controls. Many novel single nucleotide and indel variants were discovered, some in putative functional elements. The final study attempts to link genetic sequence data with brain imaging data, using construction of genetic variables for input into morphology analysis, and a sequence-based multivariate distance matrix regression method (MDMR) for in silico functional characterization of the many variants detected. The rare enriched haplotype, possessed by nine autism patients, showed significant morphology differences, usually lateral to medial ingrowth, in the hippocampus, thalamus, cerebellum, and pallidum. The protective haplotype showed lateral hippocampal outgrowth. Sequence- based association picked up two potential functional indels in the promoter region that were associated with multiple brain measures. It is the hope of this author that this work has contributed to an understanding of the etiology of autism, and provided a method to use next- generation sequencing data to uncover variants that affect any potential phenotype examined

▼ Prolonged pulmonary vasoconstriction and vascular remodeling cause idiopathic pulmonary arterial hypertension (IPAH). Pulmonary vasoconstriction is caused by pulmonary artery smooth muscle cell (PASMC) contraction, and enhanced proliferation of PASMC contributes to vascular remodeling. An increase in cytosolic Ca²⁺ concentration ([Ca²⁺ cyt) in PASMC triggers both contraction and proliferation. Inhibition of Ca²⁺ influx completely inhibits prolonged pulmonary vasoconstriction and stops PASMC proliferation. Store-operated Ca²⁺ entry (SOCE) is an important mechanism that mediates Ca²⁺ influx in PASMC and most other cell types. We previously reported that SOCE was significantly enhanced in PASMC exposed to hypoxia and in PASMC from IPAH patients. Stromal interaction molecule (STIM1/2) and Orai proteins (Orai1/2/ 3) were identified recently as essential components that mediate SOCE. We aim to examine whether the expression levels of STIM and Orai are altered in IPAH-PASMC compared to control PASMC, and whether these putative changes in expression level are responsible for the enhanced SOCE and proliferation observed in IPAH-PASMC. The protein expression level of STIM2 was increased in IPAH-PASMC, whereas STIM1 protein expression was decreased. Hypoxia (4% O₂, 5% CO₂, balance N₂) enhanced the protein expression level of STIM2, but not STIM1, in rat PASMC after 48 hours. In IPAH-PASMC knockdown of STIM2 decreased SOCE and proliferation, while knockdown of STIM2 in control PASMC had no effect on either SOCE or proliferation. Overexpression of STIM2 in control PASMC failed to enhance SOCE or proliferation. These data indicate that enhanced protein expression of STIM2 is necessary, but not sufficient, for enhanced SOCE and proliferation of IPAH-PASMC. Orai2 protein expression level was increased in IPAH-PASMC. Orai1 and Orai3 protein expression levels were not statistically significantly altered. Furthermore, 48 hours of hypoxia significantly increased protein expression of Orai1 and Orai2, but not Orai3. Overexpression of Orai2 alone in PASMC did not enhance SOCE. Co-overexpression of both STIM2 and Orai2, however, did augment SOCE in PASMC. In conclusion, upregulated protein expression of STIM2 is necessary, but insufficient, for augmented SOCE and enhanced proliferation in PASMC from IPAH patients. Increased expression of both STIM2 and Orai2 is sufficient to enhance SOCE in normal PASMC

▼ In topographic mapping, axons project to appropriate targets based upon their spatial positions, with adjacent cells projecting to adjacent targets. During development, retinal ganglion cells project to the optic tectum to map along the tectal surface based on the position of the originating soma in the retina. Retinotectal topographic map formation has been highly studied, but questions about the mechanisms of mapping along the mediolateral tectal axis remain. In this dissertation, I show that the activities and interactions of two sets of gradients, Wnt3 -Ryk and ephrinB1-EphB, are necessary to form an accurate mediolateral map. During map formation, I show that the front of the tectal Wnt3 gradient shifts laterally between E10 and E12, while the ephrinB1 gradient remains stable, generating a laterally-shifting intersection between these two gradients at which mapping forces are balanced. The timing of this gradient movement corresponds to the similarly timed medial-to-lateral development of retinal axons and interstitial branches in the tectum. By overexpressing Ryk and EphB2 and downregulating Ryk via in ovo electroporation, I demonstrate that Ryk and EphB2 provide opposing mapping forces within retinal interstitial branches, with Ryk repulsing these branches laterally while EphB2 attracts them medially. These mediolateral direction choices occur near branch initiation without later direction reversals, suggesting that branches respond to specific balance points in a time -limited manner, defining the mediolateral map. This concept of a moving series of balance points which drives opposing forces within growth cones to define topographic positions in a time-specific manner comprises the Moving Gradient Model of topographic mapping. Interstitial branches invade the tectal surface to select specific deeper tectal laminae for arbor stabilization and synapse formation after map formation. The mechanisms by which these laminae are selected or targeted are largely unknown. In this dissertation, I show that six Frizzleds are expressed in RGC subsets, while five Wnts appear in specific tectal laminae during map formation and tectal laminar targeting. To study how these Frizzleds may affect laminar targeting, I employ in ovo retinal electroporation and AChR[Beta]2 labeling to characterize small populations of retinal ganglion cells, as well as to overexpress Frizzled1 to observe targeting effects

▼ Kidney development involves complex morphogenetic interactions, some of which are not currently well defined. The morphologic development of the nephron, in specific, is a well described process that is part of a robust literature on renal developmental morphogenesis. It is an aim of the field to elucidate mechanisms controlling the morphogenesis of the developing kidney. To achieve an understanding of renal morphogenetic programs a wealth of microarray data has been collected describing temporal and spatial aspects of the developing kidney in vivo. This data is augmented with microarray data from in vitro systems that recapitulate aspects of renal development. This data has largely been examined in isolation, and to begin addressing renal development from an integrative perspective we collect diverse spatial and temporal microarray data sets and together with the in vitro models begin an integrated analysis. The aim is to provide insight into morphogenetic programs controlling renal development and provide a model for analysis of any complex process for which disparate data exists. The “crossing” of in vivo developmental data by multiple methods provides contextualization to the data sets. Integration of in vivo spatial data with in vitro model microarray data highlighted novel cytokine and growth factor mediated pathways involved in late development of the collecting system and nephron. To specifically analyze the morphogenetic processes controlling renal tubulogenesis in spinal cord-induced metanephric mesenchyme, the model was analyzed from transcriptomic, anatomic, and biochemical perspectives to arrive at a characterization of in vitro nephrogenesis permissive of perturbation. Analysis of multiple nephrogenesis data sets identified regulatory mechanisms in a series of temporal networks. A screen of putative tubulogenesis regulating signaling pathways identified in transcriptomic analyses was performed. PKA activity was found to regulate morphogenesis of the renal vesicle, with sustained activity of the enzyme preventing tubular morphogenesis. Microarray analysis confirmed PKA activity prevented progression of development from the renal vesicle stage of morphogenesis. The Wnt signaling pathway was implicated and confirmed to upregulated by PKA signaling. Inhibition of the Wnt pathway rescued the PKA mediated disruption of tubulogenesis. This systems level analysis highlights novel early and late mechanisms of renal tubular morphogenesis.

▼ The observation that synthetic siRNA molecules could induce RNAi responses following exogenous addition to human cells opened up an entirely new avenue for human disease intervention. Unfortunately, due a size of 1̃4,000 Da and inherent anionic character from the phosphodiester backbone, synthetic siRNA molecules fail to traverse the cellular membrane and display unfavorable pharmacokinetic patterns in vivo. The challenge has been to create safe and efficient delivery systems in order for siRNA therapeutics to begin to be translated into clinically- relevant settings. To date, the majority of siRNA delivery strategies have relied on nanoparticle formulations. Due to inherent problems of nanoparticle-based delivery systems, our laboratory has sought to design soluble, monomeric siRNA delivery strategies utilizing a class of small cationic peptides termed Peptide Transduction Domains (PTDs). Unfortunately, PTD- siRNA conjugation strategies have failed in enhancing the delivery of siRNA molecules due to PTD-neutralization from the strong anionic charge of the siRNA phosphodiester. In an attempt to circumvent the anionic properties of the siRNA backbone, our laboratory began novel oligonucleotide synthesis routes to mask phosphodiester linkages with bioreversible, phosphotriester modifications. Building upon the S-Acyl- ThioEthyl (SATE) phosphotriester linkage, a cytoplasmic thioesterase sensitive linkage used for small molecule pro -drugs, we have successfully synthesized a collection of phosphodiester-neutral siRNN oligonucleotides. SATE- modified siRNN molecules were shown to be efficiently synthesized and displayed robust RNAi responses in cells in culture following cytoplasmic conversion to RNAi- compatible phosphodiesters. However, creation of PTD-siRNN conjugates with the standard SATE modifications were futile due to the extensive hydrophobicity donated from the SATE modification. In order to circumvent this hydrophobicity barrier, we synthesized a collection of Amino- SATE (N-SATE) phosphotriester siRNN containing terminal primary amines to increase the solubility of PTD- siRNN conjugates. Indeed, N-SATE siRNNs have been successfully synthesized, induce efficient RNAi responses and have succeeded in overcoming hydrophobic issues associated with SATE siRNN oligonucleotides. Due to this increase in solubility, PTD-siRNN conjugation attempts are currently underway to determine the ability of PTD-siRNN to function as a viable siRNA delivery platform

▼ Adult stem cells replenish tissues during normal cellular turnover or injury. A stem cell divides asymmetrically to produce one stem cell (a process called self-renewal), and one daughter cell that initiates differentiation. The mechanisms ensuring the balance of cells are critical for tissue homeostasis. Misregulation of stem cells during disease or aging can lead to impaired tissue maintenance and repair resulting in diminished life quality or death of an individual. Stem cells are located in a microenvironment, or niche, within a tissue. The niche, composed of other cells, ECM, and/or a basement membrane, provides extrinsic cues to stem cells that regulate self- renewal, maintenance, and survival. Therefore, analyses of stem cells should include the niche, to ensure that an observation is physiologically relevant. While in vivo studies of stem cells are critical, complex vertebrate tissues are difficult for 1) identifying stem cells 2) determining niche components 3) deciphering signaling pathways within stem cells, and between stem cells and the niche. Therefore, studies of stem cells within the context of disease or aging would add more hurdles to the research process. This dissertation presents research using the powerful model organism, Drosophila melanogaster, to study the effects of aging on germline stem cell (GSC) behavior in the testis, a well-characterized tissue. Chapter 2 shows that aging results in reduced GSC function due to extrinsic changes to the niche and intrinsic changes to the GSCs. Janus kinase -Signal Transducer and Activator of Transcription, Jak-STAT, signaling that regulates stem cell maintenance, was reduced during aging, and restoration of signaling led to a rescue of GSC number within the niche in aged animals. Male flies are able to replace lost GSCs through reversion (dedifferentiation) of early germline progenitor cells. Chapter 3 shows that GSC numbers are lower during aging possibly due to compromised reversion. Analysis of early cyst cells, one of the components of the niche, shows defects in response to reversion in aged flies. This dissertation summarizes that interactions between the stem cells and their niche dictate tissue health; changes to the niche and stem cells lead to decreased tissue homeostasis during aging
