Os tumores hep�ticos s�o a segunda causa de morte relacionada com cancro em seres humanos. J� no c�o, o carcinoma hepatocelular � raro mas o progn�stico � igualmente mau. Os crit�rios histopatol�gicos para o diagn�stico do carcinoma hepatocelular n�o est�o bem estabelecidos, especialmente no caso dos tumores bem diferenciados, podendo-se tornar estes casos um desafio. Por esse motivo, o papel da reticulina no diagn�stico de carcinomas hepatocelulares na Medicina Humana j� � utilizado de forma relativamente recorrente. O presente trabalho teve como objetivo determinar qual a utilidade da reticulina no diagn�stico do carcinoma hepatocelular num modelo de murganho e qual a sua aplica��o � esp�cie canina. Avaliou-se de forma qualitativa e quantitativa a capacidade da reticulina para diferenciar os tumores dos focos de displasia e f�gado saud�vel em murganhos. No c�o foi comparado o padr�o de reticulina apresentado por carcinomas hepatocelulares, hiperplasias nodulares e cirrose. Verificou-se que no murganho as fibras coradas a negro est�o empurradas para a periferia do tumor, comprimidas de encontro ao tecido adjacente, formando uma rede compacta que permite estabelecer limites bastante precisos do anterior. No c�o, os padr�es apresentados nas mais variadas les�es assemelham-se ao que est� descrito na literatura em Medicina Humana. A reticulina pode assim ser um importante aux�lio na diferencia��o do carcinoma hepatocelular de les�es benignas, devendo no entanto ser utilizada com cautela e suportar o diagn�stico feito pela hematoxilina e eosina.

Hepatic tumors are the second leading cause of cancer-related death in humans. In the dog, hepatocellular carcinoma is rare but the prognosis is equally bad. Histopathological criteria for the diagnosis of hepatocellular carcinoma are not well established. For this reason, the role of reticulin in the diagnosis of this tumor in human medicine has been gaining ground. The aim of the present study was to determine the usefulness of reticulin in the diagnosis of hepatocellular carcinoma in a mouse model and its application to the canine species. The ability of reticulin to differentiate tumors from foci of dysplasia and healthy liver in mice was qualitatively and quantitatively evaluated. In the dog the reticulin pattern presented by hepatocellular carcinomas, nodular hyperplasia and cirrhosis was compared. It was noted that in the mouse the fibers are pushed to the periphery of the tumor, compressed against the adjacent tissue, forming a compact net that allows establishing quite precise limits of the carcinomas. In the dog, the patterns presented in the most varied lesions resemble what is described in the literature in human medicine. Reticulin may thus be an important aid in the differentiation of hepatocellular carcinoma from benign lesions, but should be used with caution and support the diagnosis made by hematoxilin and eosin.

A laminite � a doen�a mais grave do d�gito do equino, afetando 34% dos cavalos e comumente � necess�ria a eutan�sia. A venografia digital prediz o deslocamento da falange distal atrav�s da avalia��o da perfus�o vascular. � uma ferramenta muito valiosa que pode ser utilizada no diagn�stico, tratamento, monitoriza��o e progn�stico da laminite. O objetivo do presente estudo consiste em reportar os dados cl�nicos observados no primeiro venograma de todos os casos cl�nicos, descrever um caso cl�nico de um cavalo com laminite onde foram realizados v�rios venogramas com o objetivo de diagnosticar, monitorizar, estabelecer um tratamento e progn�stico e inferir a import�ncia do venograma digital no diagn�stico de laminite. Para tal, recorreu-se a uma amostra de 7 �guas previamente diagnosticadas com laminite e submetidas aos mesmo procedimento de venografia digital no Hospital Equino Weems e Stephens. Desta forma foi poss�vel avaliar as altera��es encontradas em diferentes �reas venogr�ficas e a atribui��o a um significado cl�nico. Este estudo permitiu reconhecer a import�ncia dos venogramas em casos cl�nicos com laminite, uma vez que � medida que existem altera��es no aparelho suspens�rio do casco � poss�vel detetar precocemente essas altera��es com a venografia digital sendo crucial no progn�stico dos cavalos afetados.

Laminitis is the most serious disease of the equine digit, affecting 34% of horses and euthanasia is commonly requerid. The digital venography predicts the displacement of the distal phanlanx through the evaluation of the vascular perfusion. It is a very valuable tool that can be used in the diagnosis, treatment, monitoring and prognosis of laminitis. The objective of the presente study was to report the clinical data observed in the first venogram of all cases, to describe a clinical case of a horse with laminitis where several venograms were performed with the purpose of diagnosing, monitoring, establishing a treatment and prognosis and inferring the importante of the digital venogram in the diagnosis of laminitis. For this, a sample of 7 mares previously diagnosed with laminitis and submitted to the same digital venography procedure were used at the Weems and Stephens Equine Hospital. In this way it was possible to evaluate the alteration found in different venographic areas and the attribution to a clinical significance. This study allowed to recognize the importance of the venograms in clinical cases with laminitis, since as there are alterations in suspensory apparatus of the distal phalanx it is possible to detect these alterations early with the digital venography being crucial in the prognosis of the affected horses.

The term sexually transmitted infections (STIs) includes a number of syndromes caused by pathogens that can be acquired through sexual activity with a high incidence in recent years. Under the concept of STI are grouped both classical venereal diseases (urethritis, syphilis, chancroid and lymphogranuloma venereum) and others with different ways of transmission (hepatitis B and C, MSM intestinal amebiasis, infection with human immunodeficiency virus (HIV) and some parasites). Encompass the most important pathogens Neisseria gonorrhoeae, Chlamydia trachomatis, Treponema pallidum, Trichomonas vaginalis, HIV and other emerging organisms like Mycoplasma genitalium, Mycoplasma hominis, Ureaplasma urealyticum and Ureaplasma parvum. The correct diagnosis of STIs is important for public health problem and because the complications that can occur in patients undiagnosed. In clinical practice often STIs have a syndromeic diagnosis, sometiems by the lack of sensitive of the diagnostic tests employed or by the lack of these tools in some laboratories. The Thesis has three main objectives that have been addressed in three different articles. The first objective is to evaluate an avidity technique to identifying recent infection by HIV 1 using an automated chemiluminescence immunoassay (QLIA). Recent Infection Testing Algorithm (RITA) is a generic name for a variety of laboratory techniques that distinguish the recent HIV acquisition. The study indicates that the test is a reliable method to identify recent HIV-1 infection and could be used in a RITA algorithm to estimate the incidence of HIV-1 infection in the population. The second objective of the Thesis was to describe the use of an automated technical VITROS Syphilis TPA (treponemal QLIA assay) for diagnosis of syphilis compared to other enzymeimmunoassays, as well as the classic reaginic test, and its use as a first step in the new reverse algorithm comparing with the classic Rapid Plasma Reagin (RPR) with subsequent confirmation with Treponema Pallidum hemagglutination assay (TPHA). The easy use of this automatic system can be implemented as a screening test in laboratories with high workload due to its high negative predictive value. To complete the study of STIs, the third aim of the Thesis was to study and evaluate a new biology molecular technique using PCR multiplex with different types of samples from selected populations previously studied by other techniques. This method allows the detection of seven major pathogens of STI (Neisseria gonorrhoeae, Chlamydia trachomatis, Trichomonas vaginalis, Ureaplasma urealyticum, Ureaplasma parvum, Mycoplasma hominis and Mycoplasma genitalium) from a single sample. The data obtained were compared with the conventional methods performed in our laboratory. The sensitivity, specificity and predictive values obtained were highly significant, allowing the adoption of this technique for the diagnosis of STIs.
