▼ The transposase-derived transcription factors FAR-RED ELONGATED HYPOCOTYL 3 (FHY3) and FAR-RED IMPAIRED RESPONSE 1 (FAR1) hold multifunctional roles in A. thaliana, such as in resetting the circadian clock, flowering and chloroplast division. This thesis aimed to investigate the cause of extensive leaf-lesion formation observed in fhy3 far1 mutants, reminiscent of the programmed cell death (PCD) associated with plant defence responses. Global gene expression analysis suggested a constitutively activated defence response in fhy3 far1 mutants causing the mutant's leaf-lesions. However, defence marker gene expression associated with Salicylic Acid (SA)- and Jasmonic Acid (JA)/Ethylene (ET)-signalling, the two groups of signalling compounds that mediate defence response, was downregulated at the same time. Analysis of transcriptional changes upon biotic challenges suggested that this was the result of negative regulatory feedback in fhy3 far1 mutants. A microarray analysis of key differentially expressed genes in fhy3 far1 mutants revealed an upregulation of the pro-PCD gene CYSTEINE-RICH RECEPTOR-LIKE PROTEIN KINASE 13 (CRK13) and a downregulation of the PCD-inhibiting METACASPASE 2 (MC2). The function of CRK13 and MC2 in PCD suggested that their misregulation in fhy3 far1 mutants could be part of the primary reason for increased leaf-lesion formation in double mutant plants. Transcriptional analysis of responses to biotic challenges in MC2 and CRK13 misexpression lines, however, revealed little similarity to the responses seen in fhy3 far1 mutants. A constitutively activated defence response infers alterations of the associated microbiota. Initial investigations indicated that the fhy3 far1 mutant plant's phenotypical changes of extended leaf-lesion formation will be very likely to affect the phyllosphere, which prompted a next generation sequencing investigation of the phyllospheric microbiota. Its composition showed greatly increased species richness and a more even spectrum of abundances of bacterial species in fhy3 far1, a pattern that is commonly associated with continuing environmental disruption. The WT phyllosphere showed a stronger domination by a few species that were associated with the production of antimicrobials. In addition, the fungal community in fhy3 far1 mutant plants contained more pathogenic fungi in comparison to more saprotrophic fungi in WT. These findings support the suggestion that pathogen-protective properties of beneficial microbes in the phyllosphere are a community characteristic linked to microbial competition.

▼ The United Nations Charter at Article 2(4) specifically calls on all members of the United Nations to refrain in their international relations from the threat or use of force. Article 51 of the Charter, however, permits individual or collective use of force in exercise of the right to self-defence and only so in response to an armed attack. All other resort to use of force is subject to authorisation from and management by the Security Council. The Council determines whether measures for the use of force are appropriate in each circumstance as it is charged with the primary responsibility for the maintenance of international peace and security. However, what appears to be a clear delineation on when an individual State or group of States may resort to the use of force in international law has been blurred by state practice. Actual state practice, exhibited by the States’ interpretation of Article 51 of the Charter and the use of force in purported exercise of the right to self-defence, indicates that States are operating outside the provisions of the Charter. The use of force in this regard is therefore, illegal. It is, however, appreciated that the modern State has to contend with threats that were not contemplated by the Charter in 1945. These include technological advancements in modern warfare and communication, as well as the increase of States interests beyond their territorial boundaries and the significant danger posed by attacks initiated by non-state actors. Some of the actions taken by the States have in retrospect shown to have been justified and necessary at the time for the preservation of State security. Thus, although the action taken may itself have been illegal to the extent that it offends the express provisions xxiii of the Charter, it was nevertheless necessary and justified and therefore, legitimate. Consequently, States now agitate for a wider scope of the right to self-defence; beyond the provisions of Article 51 of the Charter. This agitation is manifest in the arguments set forth by States in justifying their resort to the use of force. This paper addresses the issue whether or not there is need to re-articulate the provisions of Article 51 with a view of taking into account contemporary state practice in the purported exercise of the right to self-defence. Chapter one of the paper introduces the problem. It states the hypotheses underlying the research, sets forth the research questions sought to be answered and identifies the theoretical framework from which the paper stems. It also outlines in summary the views of different scholars on the right to self-defence in international law. Chapter two is the theoretical chapter. It analyses the evolution and scope of the right to self-defence in international law and examines in detail the said right under customary international law and treaty law. The Afsluitdijk (Enclosure Dam) is a main dam that forms the fundamental part of the larger Zuiderzee Works that separates the Zuiderzee, a salt water inlet of the North Sea, and turning it into the fresh water lake of the IJsselmeer. It protects the central Netherlands from the effects of the North Sea. It was constructed between 1927 and 1933, and runs from Den Oever on Wieringen to the village of Zurich in Friesland. It is 32 km long and 90 meters wide, rising to 7.25 meters above sea-level, with an incline of 25% on each side. At the location along the Afsluitdijk where the final water passage was closed (‘De Vlieter’), a lookout tower known as ‘The Monument’ was built in 1933. It was presented by the Zuiderzee Works Construction Company and designed by architect W.M. Dudok. The site has a statue of a “Steenzetter”, and also marks the inscription ‘A living nation builds for its future’. A statue of Cornelis Lely has also been erected near Den Oever on the Barrier Dam itself. The Afsluitdijk with The Monument are currently managed by the Ministry of Infrastructure and the Environment (“The client”). There is a restaurant at The Monument where the client has sublet it for private operator to manage it. With the anticipation of increase storm intensity and sea level rise in the future, the client had previously engaged a consultant to analyze the existing design of Afsluitdijk for a design storm of 1:10,000 years. According to the client, the result of the study concluded that the standard cross-section of Afsluitdijk is not sufficient to withstand the design storm of 1:10,000 years, hence, modifications of it are required. The study also found that during the design storm, there is a significant overtopping over the dike. The client is concerned about the significant overtopping found from the previous study may have adverse impact on the non-standard cross-section of the dike where The Monument is located. Therefore, the client has requested our design team to look into the dike section at The Monument location.

▼ Read a recent Defence White Paper of any number of countries or look at the range of foreign relations one country has with others in the contemporary age, and one is likely to come across the term 'defence diplomacy'. The traditional function of armed forces has been to prepare for and undertake the use of force. As part of this role, armed forces have cooperated with those of other nations to enhance security by countering or deterring potential enemies. In the post-Cold War era however, a new form of defence relations has emerged; in contrast to, yet supplementing their traditional role, armed forces have been employed in building cooperative relationships between former and potential future foes. This shift is explained by the concept of defence diplomacy; the concept identified by Andrew Cottey and Anthony Forster that armed forces have a peacetime role in pursuit of broader foreign and security policy goals. Recognising that defence cooperation activities have a long history, Cottey and Forster differentiated defence diplomacy between 'old', meaning those traditional defence cooperation activities aimed at allies and friendly states, and 'new', meaning defence cooperation aimed at potential or former enemies. New Zealand, like many other countries, has used the term since the 1990s to describe those aspects of the diplomatic relationship, specifically peacetime cooperative activities, performed by the Ministry of Defence and the New Zealand Defence Force with the armed forces of other nations. This thesis explores the origins of defence diplomacy and the adoption of the concept by New Zealand. It looks at the way in which New Zealand has developed and managed its 'old' defence diplomacy in Asia through examining the example of the Five Power Defence Arrangements. It then explores New Zealand’s 'new' defence diplomacy with what are considered here as “non-like-minded” states such as China, Viet Nam and Indonesia. Through these three case studies, the thesis examines key dilemmas and problems of defence diplomacy that have arisen in the development of these key relationships. The thesis then concludes with an analysis of New Zealand’s defence diplomacy according to the framework established by Cottey and Forster. Advisors/Committee Members: Capie, David, Ayson, Robert.

▼ Two questions apply regarding Australia in World War II: what measures were taken to defend mainland Australia, and were those measures effective in meeting the perceived threat to Australia? Mainland Australia is defined as the mainland, Tasmania and the littoral waters out to 200 nautical miles (excluding New Guinea). Mainland defence is assessed at a systemic level, where the major elements of mainland defence are point defence systems, area defence systems, lines of communication, surveillance and intelligence. The minor elements are production capacity and national will. A qualitative assessment is made in each of six shifts in perceived threat. The fit for purpose test is applied: were the elements of mainland defence at that time appropriate to meet the perceived threat? The outcomes of the analysis follow.At the start of the European war the perceived threats were German attacks on shipping (minor scale) or, at a medium scale, sustained attack on shipping and concurrent heavy raids if a war started with Japan. Mainland defence was partially fit for purpose.When France fell and Italy declared war (mid-1940) there were three possible scales of Japanese attack: bombardment, light raids and invasion. Mainland defence was partially fit for purpose.After the start of the Pacific War, events moved rapidly in the first 100 days. Threats were seen as: air attack; naval bombardment; a sea-borne raid; attack to permanently occupy territory; and invasion of Australia. Mainland defence was not fit for purpose.When MacArthur arrived (March 1942) threats were: attacks in force against Australia, including invasion; attacks against Australian lines of communication. By then mainland defence was partially fit for purpose.During the ‘holding war’ (mid-1942 – mid-1943) the Government held that incursion could not be ruled out. However the services saw the threat as sea bombardment, torpedo attack and air attack. By now mainland defence was fit for purpose, with the exception of Anti-Submarine Warfare.Finally from mid-1943 to mid-1944 there was some residual threat. Mainland defence, despite being reduced, was fit for purpose. From mid-1944 on there was no threat, so the questions do not apply. ▼ This research examines service innovation in a large-scale manufacturing firm. It explores how co-creation, a foundational premise of the emerging field of service science, is operationalised as a competitive strategy. Globally, manufacturing firms, including those in the defence sector resistant to change, are embracing service innovation as source of differentiation and advantage. Central to this approach is the notion of value co-creation. Therefore, this thesis seeks to better understand how a manufacturing company embraces service innovation by exploring: 1) What factors (enablers and barriers) influence co-creation (CoC); and 2) How do firms design collaborative and open innovation (OI) service systems to co-create value through customer collaboration and innovative partnerships with new stakeholders? This thesis adopts an in-depth qualitative inquiry to answer these research questions. It explores changes within organisations, identifies new forms of outreach and engagement with customers, and assesses the value of strategic alliances with other firms. It develops a tailored research methodology and summarises the results of analysis, key findings, and conclusions in a generalizable manner to build on the body of scholarship associated with service innovation. The thesis is based on case studies that help better understand the UK’s largest manufacturing company, BAE, as it servitizes. It draws upon a diverse set of maritime, ground, air, and joint force cases across distinct military domains, weapons platforms, geographies, cultures, and programme scales to provide a comprehensive look at service transformation. It also examines how private and public sector organisations co-create value across a myriad of MoD agencies, military forces, service providers, and suppliers innovatively networked to improve service outcomes. The thesis traces how the focal firm adopts service strategies to meet changing customer demands through more open service design and delivery processes reliant on increased collaboration and integrated systems. The Research Aim of this work is to examine how value is co-created across networks of diverse stakeholders (technical, business, military, and manufacturing) to develop a co-creation framework. Research Objectives are focused on: • the discovery of insights how firms, design more open and collaborative service systems • the identification of drivers, barriers, and enablers to co-creation as part of servitization • the development of a framework, models, and tools to guide service innovation in manufacturing / defence While derived from a defence context, these outputs are deliberately generalizable to contribute to theory and to inform practitioner efforts to support a manufacturing shift away from purely product-based businesses. This work applies and seeks to contribute to service, strategy, and systems theory. ▼ There is an increasing gap between the progress of technological systems and the successful exploitation of these systems through cyber-attack. Whilst the mechanism and scope of cyberspace is progressing with each passing day, risk factors and the ability to process the required amount of data from cyberspace efficiently are proving to be major obstacles to achieving desired outcomes from cyber operations. This, coupled with the dramatic increase in the numbers of cyber attackers, who are constantly producing new ways of attacking and paralysing cyber systems for political or financial gain, is a critical issue for countries that have linked their major infrastructures with Internet applications. The defensive methods currently applied to counter these evolving attacks are no longer sufficient, due to their preventive and reactive nature. This research has developed a new Active Situational Awareness theoretical model for Active Defence that aims to enhance the agility and quality of cyber situational awareness in organisations in order to counter cyber attacks. Situational Awareness (SA) is a crucial component in every organisation. It helps in the assessment of an immediate situation in relation to the environment. Current SA models adopt a reactive attitude, which responds to events and works in passive manner to any progressing enemy cyber attack. This creates a defensive mind-set and consequently influences the operator to process and utilise knowledge only within the concept of attack prevention. Thus, one can assume that operators will only gather certain knowledge after the occurrence of an attack, instead of actively searching for new intelligence to create new knowledge about the cyber attack before it takes place. This research study introduces a new approach that incorporates an Active Defence posture; namely, a ‘winning attitude’ that conforms to the military stratagems of Sun Tzu, where operators always engage attackers directly in order to create new knowledge in an agile manner by deploying active intelligence-gathering techniques to inform active defence postures in cyberspace. This also allows the system being protected to remain one step ahead of the attackers to ultimately defeat them and thwart any costly attacks. To back these statements, this study issued a survey to 200 cyber defence and security experts in order to collect data on their opinions concerning the current state of Active SA. Structural Equation Modelling (SEM) was then employed to analyse the data gathered from the survey. The results of the analysis revealed significant importance of Active Offensive Intelligence gathering in enhancing Cyber SA. The SEM showed there is a significant impact on SA Agility and Quality from Active Intelligence gathering activities. This exploratory study investigates the research problem (RP) ‘How may DST improve its product innovation process?’ The Department of Defence, Defence Science and Technology (DST) is recognised as a national leader in safeguarding Australia, through the delivery of scientific advice and innovative technology solutions for the Australian Defence Force (ADF) and the wider defence industry community. With an annual budget of over $400 million, DST is Australia’s second largest publicly funded research and development (R&D) organisation. The environment for DST product innovation is characterised by high complexity, large expenditures, product innovations frequently abandoned or failed, and significant resource and operational constraints. Against this background, it could be argued that a detailed study of the DST product innovation process could contribute to both a better theoretical understanding of defence industry product innovation and a practical guide for DST product innovation process and hence improvement. A review of the literature indicates scant research to help defence industry academics and practitioners understand important aspects of defence industry innovation, including inspirations and objectives for defence innovation, stages in the defence industry innovation process, key individuals and groups involved (as well as their roles), key success factors (KSFs) and constraints associated with product innovation processes, as well as criteria to measure defence innovation outcome success. A greater understanding of these defence industry innovation factors may contribute to improved defence innovation outcomes. This study therefore seeks to address the lack of defence industry research in these areas, to develop a better understanding of those factors, which, in turn, may contribute to improving product innovation process and outcomes in DST. The RP for this study, ‘How may DST improve its product innovation process?’, has been developed into the following five research questions (RQs): RQ1: ‘What are the inspirations and objectives for DST product innovation involvement?’ RQ2: ‘What are the stages in the DST product innovation process?’ RQ3: ‘Who are the key individuals and groups involved in the DST product innovation process?’ RQ4: ‘What are the KSFs and constraints that affect the DST product innovation process?’ RQ5: ‘Can a set of criteria to measure a successful DST product innovation be specified?’ This research adopted a qualitative, multi-case study approach to develop inductive, theory-building insights into each of the five RQs and hence RP. Against a background that most DST innovations are failures, informed by literal replication logic, 30 successful DST cases were selected and analysed. ▼ 'Battling with Words' - a battle of communicating new messages of acceptance and empathy in an organisation that desires cultural change. Australia is a diverse nation. The people of Australia speak many different languages, profess different beliefs, follow different social practices, possess different abilities, skills and knowledge and live in different kinds of families. The Australian Department of Defence is responsible for the security and defence of this nationa diverse nation of multicultural and multilingual people. Yet, the demographic makeup of Defence does not reflect the demography of the Australian community which it serves. Compared to the wider community, Defence is an Anglo-Australian, male-dominated organisation. Such a demographic profile is no longer desirable or sustainable. Importantly Defence realises the need to reflect adequately the composition of society particularly if the population is to have confidence in the armed forces (Centre for the Democratic Control of Armed Forces, 2006). Further, a more diverse Defence Force is now considered a capability issue; that is, a force multiplier for mission success (Defence Committee 2012). Therefore a major challenge facing Defence is how to create a workplace culture that shifts the institution away from an exclusive, homogeneous culture to an inclusive heterogeneous one (Silk et.al. 2000).While numerous reports of recent years have tried to address this issue of homogeneity, none have specifically addressed the role language plays in maintaining and perpetuating cultural norms. This thesis puts forward a number of reasons for needing language change, arguing that unless the language practices of the institution change in concert with other social inclusion policy changes, it will be difficult for leadership to 'walk the talk' of change, as demanded by a number of reports (Silk et.al. 2000; McGregor 2011; Defence Committee 2012; Department of Defence 2013b).The thesis demonstrates how language and culture are inextricably linked and investigates how language is used to rally and bond Defence personnel in three main ways: 1) around a limited set of officially endorsed Values of the three uniformed Services, of the Australian Public Service (APS), and of Defence; 2) around a preferred idealised Defence identity presented as iconic in key Defence documents; and 3) around the everyday talk at work which supports and maintains the dominant position of the Anglo-Australian male in Defence culture. A focus on language is the next piece of the mosaic needed to create and maintain a culture of social inclusion. If Defence wants to increase diversity, then it needs to turn its attention and considerable strength to battling with words, so that it can create a Department of Defence characterised by the highest possible degree of sustainable diversity and social inclusion, and ultimately the greatest capability. Įstatyme numatyta, kad būtinosios ginties ribos peržengiamos tuo atveju, kai sunkiai sutrikdoma sveikata, jei gynyba aiškiai neatitiko kėsinimosi pobūdžio ir pavojingumo. Nors teismų praktikoje pripažįstama, kad skirtingos vertybės turi skirtingą apsaugos lygį, tačiau būtinosios ginties atveju, įstatymo leidėjas nereglamentuoja skirtingo žalos, padarytos agresoriui ginant skirtingo reikšmingumo vertybes, dydžio. Darbo tikslas: atsakyti į klausimą – ar atitinka būtinosios ginties sąlygas atvejis, kai ginant turtą agresoriui padaromas sunkus sveikatos sutrikdymas? Darbe išsikelti uždaviniai: 1. Art. 28 of the Criminal Code of the Republic of Lithuania provides for the right to the self-defence that can be exercised when defending own property. It is also provided in the law that the limits of self-defence are exceeded when severe harm is caused to health in cases when the defence is apparently inadequate to the character and dangerousness of an attempt. Though it is recognised by the court practice that different values have different protection level, but in case of self-defence, the legislator has not projected different extent of harm caused to aggressor upon defending values of different importance. Upon qualifying cases of the self-defence, the defendant must sum up the conflict situation, to be aware of the aggressor’s motifs, to know precisely the beginning and the end of the attempt, to be aware whether an attempt is real and obvious, whether the defence will be adequate to the aggressor’s actions, whether the harm caused to the aggressor will not be disproportionate if compared to the harm that aggressor would have caused to the protected value, and finally, in case the court recognises that there have not been grounds for self-defence or that the limits were exceeded – what legal consequences are waiting for the defendant having exercised his right. ▼ This thesis frames a study undertaken to explore key management challenges associated with the Namibian Defence Force (NDF) during the peace support operations (PSOs) mandated by the United Nations Mission in Liberia (UNMIL) in the period 2004 to 2008. The purpose of the study is to conduct an in-depth analysis of the management challenges that confronted the Namibian Defence Force (NDF) during these operations. The Namibian Defence Force (NDF) was involved in a number of UN led peace support operations worldwide, before its participation in the United Nations peace support operations (PSOs) in Liberia over a period of four years (2004 – 2008). During the subsequent internal review it came to light that the Namibian Defence Force experienced a number of management challenges in relation to command, control and communications (C3). The review also revealed that the Namibian Defence Force (NDF) operational requirement capabilities were not well-matched with the United Nations’ standards and were not compatible with the United Nations Standing Operational Procedures (SOPs). The peace support operations (PSOs) is expected to achieve its mandate within such constraints, support the myriad of relief agencies in its theatre of operations and to encounter the challenges which are situational dominance in this modern environment, and they are obstacles in the way to realise the objectives of the mission. The research problem of this study was to explore the challenges associated with Namibia’s peace support operations in Liberia. In this regard, the research had reviewed the reports of problems registered by the Ministry of Defence (MOD) in general, and the Defence Headquarters (DHQs) in particular. The main objective of this research was to establish how management challenges with regard to command, control and communications affected the capability of the Namibian Defence Force (NDF) to fulfil its mandate during the United Nations Mission in Liberia (UNMIL) Peace Support Operations (PSOs). The study was guided by the United Nations (UN) Charter, which was adopted and signed on 26 June 1945. The UN Charter provides the following relevant chapters for this study: Chapter VI is applicable for peacekeeping, Chapter VII relevant for peace enforcement and Chapter VIII regulating for regional peacekeeping operations, (UN, 2012, p. 155). ▼ Bats are speculated to be reservoirs of several emerging viruses including coronaviruses (CoVs) that cause severe acute respiratory syndrome (SARS), Middle-East respiratory syndrome (MERS), porcine epidemic diarrhea and swine acute diarrhea syndrome. These viruses cause significant disease in humans and agricultural animals. MERS-CoV causes serious disease in humans with a thirty-five percent mortality and has evolved proteins that can effectively suppress an innate antiviral response in human cells. Bats that are naturally or experimentally infected with these or similar viruses do not show apparent signs of disease and the molecular mechanisms of protection are not yet known. My doctoral thesis tested the hypothesis that big brown bat cells have unique adaptations in innate antiviral signaling pathways involved in the control of virus replication and coronavirus-induced inflammatory cytokines. To test this hypothesis, we generated the first commercially available North American bat (Eptesicus fuscus; big brown bat) kidney epithelial cell line. Using this cell line, we were able to demonstrate that big brown bat cells have evolved a unique repressor molecule, c-Rel that can effectively suppress double-stranded RNA (poly(I:C)) mediated expression of a key inflammatory cytokine, tumor necrosis factor alpha (TNF). MERS-CoV is thought to have evolved in insectivorous bats before spilling over to camels and eventually to humans. To further our understanding about bat-coronavirus interactions, we demonstrated that big brown bat cells are resistant to MERS-CoV-mediated subversion of antiviral responses. We determined that interferon regulatory factor 3 (IRF3) plays a critical role in controlling MERS-CoV propagation in big brown bat epithelial cells. Indeed, my doctoral thesis has identified two unique adaptations in big brown bat cells that might allow these bats, and probably other species of bats to successfully co-exist with coronaviruses. My thesis supports the hypothesis that bats function as global reservoirs for emerging coronaviruses by providing definitive examples of adaptations that would allow bats to co-exist with these viruses. Future work from my thesis will focus on adapting some of these antiviral strategies in human cells to control coronavirus-mediated disease in humans.
