▼ Transfemoral amputees suffer the loss of the knee and ankle joints, as well as partial or complete loss of many of the lower extremity muscle groups involved in ambulation. Recent advances in lower limb prostheses have involved the design of active, powered prosthetic knee and ankle-foot components capable of generating knee and ankle torques similar to that of normal gait. The associated onboard motors, conditioning/processing, and battery units of these active components result in increased mass of the respective prosthesis. While not an issue during stance, this increased mass of the prosthesis affects swing. The goal of this study is to develop and validate mathematical models of the transfemoral residual limb and prosthesis, expand these models to include an active ankle-foot, and investigate counter-mass magnitude(s) and location(s) via model optimization that might improve kinematic symmetry during swing. Single- (thigh only, shank only) and multi-segment (combined thigh and shank) optimization of counter-mass magnitudes and locations indicated that a 2.0 kg counter-mass added 8 cm distal and 10 cm posterior to the distal end of knee unit within the shank segment approximated knee kinematics of able-bodied subjects. This location, however, induced artificial hip torques that reduced hip flexion during swing. While such a counter-mass location and magnitude demonstrated theoretical potential, this location is not clinically realistic; mass can only be added within the prosthesis, distal to the residual limb. Clinically realistic counter-masses must also keep the total prosthetic mass to less than 5 kg; greater mass requires supplemental prosthetic suspension, would likely increase energy expenditure during ambulation, and contribute to increased likelihood of fatigue even with active prosthetic components. The ability to simulate the effects of active prosthetic components inclusive of varying placement of battery and signal conditioning units may advance the design of active prostheses that will minimize kinematic asymmetry and result in greater patient acceptance. L'œuvre de Martín Kohan (Buenos Aires, 1967), écrivain prolifique à la notoriété grandissante au sein de la littérature argentine contemporaine, est l'objet de ce travail de recherche. Nous abordons le corpus – constitué par l'essentiel de l'œuvre : huit romans sur les neuf publiés à ce jour, La pérdida de Laura (1993), El informe. San Martín o el otro cruce de los Andes (1997), Los cautivos. El exilio de Echeverría (2000), Dos veces junio (2002), Segundos afuera (2005), Museo de la Revolución (2006), Ciencias morales (2007), Cuentas pendientes (2010); et deux recueils de nouvelles, Muero contento (1994), et Una pena extraordinaria (1998), principalement – sous l'angle de l'antagonisme, motif essentiel de l'univers fictionnel de Kohan, non seulement sur le plan de la diégèse mais également sur celui de la forme. Ce travail part de l'observation d'une constante au sein d'un corpus pourtant très varié : un rapport dichotomique, fait d'incompréhension et parfois de violence, entre la culture populaire ou la culture de masse, et la culture lettrée. La première partie de la thèse en propose une analyse systématique dans les cinq romans qui abordent explicitement ce type de conflit sous les modalités les plus diverses. La deuxième partie explore d'autres procédés qui tendent à confirmer le rôle structurant de l'antagonisme au sein de l'œuvre : si la mise à mal de l'union amoureuse semble exprimer la fatalité du malentendu, la dévalorisation des personnages du « milieu » et de la figure du consensus d'une part, et l'aspect revigorant des conflits menés à leur dernière extrémité d'autre part, révèlent l'aspect positif et particulièrement dynamique de l'antagonisme. La dernière partie se consacre aux implications de ce motif, omniprésent au point de représenter une nécessité : sur le plan politique – car sans être réaliste, la littérature de Kohan met en action la révolution marxiste en élaborant des figures de contre-pouvoir – ; sur le plan littéraire, où l'antagonisme montre toute sa fécondité. Le rapport hypertextuel, établi notamment avec des textes qui font autorité dans la littérature nationale, en est une expression essentielle ; mais la dichotomie est aussi une composante qui se manifeste à tous les niveaux de l'écriture, de l'unité minimale du mot à l'architecture complexe des romans. À la lutte entre personnages fait écho celle – défi formel le plus ambitieux – qui se joue entre le fond et la forme, source d'inépuisable ambivalence qui constitue l'une des spécificités les plus intéressantes de cet écrivain brillant.

▼ Alcohol is the most widely used psychoactive drug (Crocq, 2003; Global Drug Survey, 2018), responsible for approximately 3.3 million deaths each year (World Health Organization, 2014), and a large global burden of disease, injury, and economic cost (Rehm, Mathers, et al., 2009; World Health Organization, 2015). The global pervasiveness of alcohol consumption is partly fuelled by extensive alcohol marketing efforts that occur in the absence of effective communication about alcohol-related harms. Given the fact that the alcohol industry marketing practices are beverage-specific, and that alcohol is sold and consumed in the form of beer, wine, spirits, and mixed drinks, there is a need for beverage-specific counter-marketing campaigns (mass media anti-alcohol campaigns and alcohol warning labels) to help balance the effects of beverage-specific pro-alcohol marketing messages. Existing alcohol counter-marketing campaigns are scarce and limited in scope. These campaigns have increased consumer awareness and knowledge of the health risks (Andrews, 1995; MacKinnon, Nohre, Pentz, & Stacy, 2000; Wilkinson & Room, 2009), but have not changed drinking behaviour (Anderson, Chisholm, & Fuhr, 2009; Greenfield, Graves, & Kaskutas, 1993; MacKinnon & Nohre, 2006; MacKinnon et al., 2000; Scholes-Balog, Heerde, & Hemphill, 2012; Stockley, 2001; Wakefield, Loken, & Hornik, 2010; Wilkinson & Room, 2009). Conversely, tobacco counter-marketing campaigns have increased public knowledge about tobacco's harmful effects, and decreased smoking prevalence (Azagba & Sharaf, 2013; Farrelly, Davis, Haviland, Messeri, & Healton, 2005; Hammond, Fong, McNeill, Borland, & Cummings, 2006; Kennedy et al., 2012; Wakefield et al., 2010). Lessons from the tobacco experience suggest the need to: (a) identify the full range of alcohol-related harms, including the unquantifiable ones, and (b) conduct formative research to more clearly understand the target audiences of counter-marketing campaigns. This thesis addressed these research needs through three separate research studies. Study 1 (Chapter 2) argues that lessons from the tobacco experience suggest that alcohol counter-marketing campaigns that communicate the full range of harms may be more effective than the currently existing ones. The current lack of focus on the full range of alcohol-related harms is possibly due to the absence of a comprehensive study examining the full range of harms. Previous reviews are mainly systematic reviews that were aimed at quantifying the harms associated with alcohol use. Consequently, the harms that could not be quantified were generally ignored. Since previous systematic reviews do not provide a clear picture of the full range of harms, and are insufficient from a health communication perspective, there arose a need for conducting an integrative review to address the existing limitations in the literature. ▼ This research broadly investigates mediated discourse and knowledge construction among media outlets commonly identified as "traditional" and "new." Specifically, this research represents a case study of fracking in the Bakken shale region of North Dakota. Using qualitative, interpretive methods this dissertation considers what knowledge(s) are constructed, upheld, and silenced in mediated representations of fracking in the Bakken.This dissertation draws upon a poststructural definition of discourse, which views knowledge and meaning as constructed realities, rather than Real in an objective sense (Castree, 2001, 2014; Foucault, 2010/1972; Hall, 1997). Although the power and resources required to produce discourse is unequal, taken-for-granted ways of thinking and doing are nevertheless always open to challenge from relatively less powerful sources. This is consistent with Foucault’s (1995/1975) conception of power as circulatory and disciplinary, rather than oppressive.Data for this research come from a mix of “traditional” and "new" media sources. Some scholars argue that these distinctions become less important in a converged mediascape (Jenkins, 2006). Nevertheless, this research proceeds from the position that (1) the productive norms of traditional and new media could result in distinct knowledges and claims to truth, and furthermore (2) current research continues to distinguish between the productive norms and types of knowledge constructed by traditional and new media (e.g. Geiger and Lampinen, 2014; Kim, 2015). Scholars argue that traditional media represent objective accounts of events, whose texts are undeniably powerful shapers of knowledge, and disseminated by a professional caste culturally sanctioned to report on events, i.e. journalists (e.g. Gerhards and Schafer, 2010; Lockwood, 2011). Alternately, scholars note that new media, e.g. blogs and social media aggregators, present the lay public and under-represented organizations with productive avenues to challenge the dominant discourses put forth by commercial, mainstream traditional media outlets (e.g. Atkinson and Rosati, 2009; Kaye and Johnson, 2011; Parker and Song, 2009) Scholars are currently split regarding new media’s potential to substantially offset established traditional media power (e.g. Tang and Yang, 2011; van Dijck, 2009, 2012).This research takes place within multiple environmental contexts: the United States recent increase in oil and gas development, oil companies’ global expansion of “unconventional” extraction practices such as hydraulic fracturing, and global climate change. Given our dependence on myriad forms of media for knowledge of these events, this project’s implications project beyond a regional case study of fracking in North Dakota. ▼ La confrontation du théâtre et de la liturgie est un lieu commun de la pensée. Il est un motif rhétorique récurrent chez les pères de l’Église pour définir a contrario et par surenchère le bon ethos du chrétien à l’Église. Ce tour de pensée ecclésiastique, typique de la synthèse augustinienne de la rhétorique antique et du christianisme, n’est pas seulement un héritage livresque au XVIIe siècle. Il est particulièrement pertinent à la vue des enjeux auxquels est confrontée l’Église catholique : elle doit répondre aux accusations protestantes, qui traitaient la messe de farce ; le théâtre renouvelé de l’antique se rétablit grâce au soutien du pouvoir, se sédentarise et devient un divertissement régulier. Cette banalité nouvelle fait de la Comédie, aux yeux des augustiniens, le lieu d’une « représentation vive » et continuelle des passions du monde, particulièrement de l’amour et de l’honneur : le théâtre apparaît comme une liturgie inversée. Là où les pratiques de piété sont censées amoindrir les passions et nourrir la foi, le théâtre excite les passions et étouffe l’esprit de prière. La querelle de la moralité au théâtre montre non seulement une concurrence morale, mais aussi psychique et affective. Les deux représentations prétendent susciter la présence d’esprit et « toucher » le cœur, voire lui « imprimer des mouvements ». La messe est qualifiée de « représentation vive du sacrifice de la croix », pendant laquelle le fidèle doit se remémorer vivement le sacrifice christique et sa signification grâce à une lecture allégorique, et se l’appliquer à lui-même. Par la considération et l’accomplissement de cérémonies, par la vocalisation des psaumes, le fidèle est invité à produire des « actes » du cœur pour s’unir à Jésus-Christ. Ce rapport au texte comme trace à suivre, et ce rapport au corps et à la voix comme media pour s’auto-exciter, expliquent pourquoi les comédiens professionnels sont condamnés par les dévots : ils excitent en eux les passions contraires à l’Esprit saint, ils rappellent des sentiments qu’un pénitent ne pourrait pas se remémorer sans « horreur ». La « représentation » est alors conçue comme un effort de remémoration.Le rétablissement du théâtre à l’antique nécessitait un discours pour en éclairer les visées et en légitimer l’existence dans une société chrétienne et monarchique. Traduire la mimesis aristotélicienne par « représentation » plutôt que par « imitation » rendait le théâtre beaucoup plus proche de la liturgie et lui ajoutait les connotations de vue, de présence et de mémoire. Le débat entre plaire et instruire est un débat entre théâtre-divertissement et théâtre-cérémonie. Incomber au théâtre la fonction d’instruire, c’était le rapprocher d’une prédication et de la messe, car instruire, signifiait instruire chrétiennement. L’échec de sanctification du théâtre des années 1640 fit conclure à une incompatibilité du théâtre avec la folie et la modestie chrétienne, mais la possibilité d’une instruction civique par le théâtre émerge à la fin du siècle. ▼ The quest to achieve high resolution in ion mobility spectrometry (IMS) has continued to challenge scientist and engineers in the field of separation science. The low resolution presently attainable in IMS has continued to negatively impact its utility and acceptance. Until now, efforts to improve the resolution have mainly focused on better instrumentation and detection methods. However, since the resolution of IMS is diffusion limited, it makes sense to address this limitation in order to attain high resolution. This dissertation presents a new IMS technique, which utilizes a high electric field and opposing high gas flow velocity with the aim to improve resolution. This approach essentially reduces the residence time of ions in the analyzer. This new technique is called "counter-flow ion mobility analysis" (CIMA). Theoretical modeling of this new technique predicted that a resolution of over 1000 is possible, which is over one order of magnitude better than that of conventional IMS techniques currently used. A wind tunnel was designed and constructed to produce a plug gas flow profile that is needed for CIMA. The test region of the wind tunnel was used as the CIMA analyzer region and was constructed from power circuit boards, PCBs, (top and bottom walls) and conductive plastic side walls. An inclined electric field was created by applying suitable voltages to multiple electrode traces on the PCBs. This inclined field, when resolved into its x- and y-components, was used to oppose the counter-gas flow and transport the ions to the detector, respectively. The results obtained did not show an improvement over conventional IMS techniques because of a limitation in the voltage that could be applied to the analyzer region. However, the results predict that high resolution is possible if (1) the ratio of the electric fields in the horizontal (x direction) to the vertical (y direction) is within the range of 2 – 0.5, (2) very high electric field and high gas flow velocities are applied, and (3) wall effects in the counter-flow gas profile are eliminated. While the resolution obtained using the present instrumentation is far from what was predicted, the foundation for ultimately achieving high resolution has been laid. The use of a wind tunnel has made the instrumentation possible. As far as the author knows, this is the first time a wind tunnel has been used in chemical measurement instrumentation. Chapter 5 of this dissertation, reports a method developed for predicting the reduced mobility constants, of chemical compounds. This method uses a purely statistical regression analysis for a wide range of compounds which is different from similar methods that use a neural network. The calculated value for this method was 87.4% when calculated values were plotted against experimental K0 values, which was close to the value for the neural network method (i.e., 88.7%).

Les plantes médicinales constituent une source inexhaustible de composés (des produits naturels - PN) utilisé en médecine pour la prévention et le traitement de diverses maladies. L'introduction de nouvelles technologies et méthodes dans le domaine de la chimie des produits naturels a permis le développement de méthodes ‘high throughput’ pour la détermination de la composition chimique des extraits de plantes, l'évaluation de leurs propriétés et l'exploration de leur potentiel en tant que candidats médicaments. Dernièrement, la métabolomique, une approche intégrée incorporant les avantages des technologies d'analyse moderne et la puissance de la bioinformatique s’est révélé un outil efficace dans la biologie des systèmes. En particulier, l'application de la métabolomique pour la découverte de nouveaux composés bioactifs constitue un domaine émergent dans la chimie des produits naturels. Dans ce contexte, le genre Acronychia de la famille des Rutaceae a été choisi sur la base de son usage en médecine traditionnelle pour ses propriétés antimicrobienne, antipyrétique, antispasmodique et anti-inflammatoire. Nombre de méthodes chromatographiques modernes, spectrométriques et spectroscopiques sont utilisées pour l'exploration de leur contenu en métabolites suivant trois axes principaux constituant les trois chapitres de cette thèse. En bref, le premier chapitre décrit l’étude phytochimique d’Acronychia pedunculata, l’identification des métabolites secondaires contenus dans cette espèce et l'évaluation de leurs propriétés biologiques. Le deuxième chapitre vise au développement de méthodes analytiques pour l'identification des dimères d’acétophénones (marqueurs chimiotaxonomiques du genre) et aux stratégies utilisées pour la déréplication de ces différents extraits et la caractérisation chimique des composés par UHPLC-HRMSn. Le troisième chapitre se concentre sur l'application de méthodologies métabolomique (RMN et LC-MS) pour l'analyse comparative (entre les différentes espèces, origines, organes), pour des études chimiotaxonomiques (entre les espèces) et pour la corrélation des composés contenus avec une activité pharmacologique.

Medicinal plants constitute an unfailing source of compounds (natural products – NPs) utilised in medicine for the prevention and treatment of various deceases. The introduction of new technologies and methods in the field of natural products chemistry enabled the development of high throughput methodologies for the chemical composition determination of plant extracts, evaluation of their properties and the exploration of their potentials as drug candidates. Lately, metabolomics, an integrated approach incorporating the advantages of modern analytical technologies and the power of bioinformatics has been proven an efficient tool in systems biology. ▼ This thesis examines the historical evolution of racial hierarchies and white racism in American society, and in particular how these ideological frameworks have impacted Black American society. By utilizing a case study approach, in order to fully identify the pervasive nature of white racial and cultural narratives and the ways they have denied Black progress and freedom, the goal is to outline how systemic incorporations of racialized stereotypes within political rhetoric and popular culture have worked to reinforce white racial hierarchies and white cultural paradigms. The three periods examined in this case study are as follows: the Post-Civil War American South, 1960's Civil Rights Era and the modern Hip Hop Generation. Misrepresentations within music, media and sports all too often resemble neo-colonial, paternalistic and racialized myths of the past. While politicians, particularly conservatives, have consistently used racialized messages to fan white fears and gain voter support with reactionary "law and order" rhetoric and by blaming minorities for American socio-economic problems. The criminalization of Blackness in American society is based on white fears, not relative crime rates. Whites, since the collapse of Reconstruction policies in the South, attempted to force Blacks back onto the plantations, railroads and iron mines of the South. Black criminality became the excuse for reinforcing racial hierarchy in American society, convict leasing replaced slavery in the South. Conservative politicians spewed forth racialized rhetoric to disenfranchise Black voters in the South, while lynching and race riots acted as violent methods to reinforce white domination and white racialized notions of Black inferiority. By the 1960s violence became a tool of the agents of the FBI to repress Black Power groups and their attempts to challenge white racial hierarchies in America. However, by the late 60s racism and outright violence became unpopular and new more subtle, more systemic forms of reinforcing racial caste systems and white supremacism in American society were needed. The impacts of deindustrialization, white flight, gerrymandering, rezoning, political marginalization and the elimination of an entire generation of Black leaders needs to be discussed for someone to fully recognize the legacy of white racism. Politicians and popular culture today have come to support racial hierarchies either intentionally or unintentionally by consistently over representing Black criminality and pushing racialized images, that in many ways came eerily similar to Jim Crow representations of Blackness. From Blaxploitation Films to Gangster Rap and Hood Films, this thesis examines how white dominant cultural representations of Black Criminality have become embedded within American popular culture and politics, and how these racialized images and narratives have conditioned American society to accept white supremacist notions of race and crime.
