▼ Using a
combination of qualitative and quantitative methods, this study identify the
challenges of and opportunities from Access to ICTs from a gender perspective on
selected residents of Arada Sub-city of Addis Ababa Administration. To this end, the
study investigated the situation regarding access to computer and internet, frequency
of using them, experience in using these technologies and place of use. The study
involved 140 (122 for questionnaire and 18 for interview) participants who were
selected from residents of Arada sub-city using convenience and purposive sampling
techniques. The result of the study has indicated that ICT is affected by different sociocultural
and economic status and there is a gender gap in having access to ICT as a
reflection of the gender disparity appears in the society. The study also find out
challenges in having access to computer and internet technology the majority of the
research respondents have indicated that access, experience, frequency, place and
purpose of using computer and internet technology is affected by gender, educational
attainment, income and age. Women have less access to computer and internet
technology due to many barriers, women have less economical power than the of
male, have less computer skill, have lots of responsibilities at home, shortage of time,
less connected to technologies, unawareness on ICT’s relevance to their life and not
know the benefit out of these technologies are the major findings from the survey and
the interview. In addition the participants notify that ICT’s bring a new opportunity in
their in their in their educational, social and professional life.

▼ In this thesis I will talk about the
combination problem of panpsychism and some possible solutions to it. A reductive approach (Dennett 1993, Churchland 2013) toward a theory of consciousness has not yielded satisfactory results because it fails to address the hard problem of consciousness (Chalmers 1995). A nonreductive approach acknowledges the hard problem and claims that consciousness is a property of the universe. Panpsychism is one such nonreductive approach to consciousness. However, a major roadblock for panpsychism is the
combination problem. My thesis will discuss the
combination problem and two possible solutions to it called the phenomenal bonding relation (Goff 2016) and the Space-Time-Qualia Complex (Skrbina 2011). I conclude that Skrbina’s Space-Time-Qualia Complex answers the
combination problem in a much better way than the phenomenal bonding solution and that is why it is an avenue worth exploring.

▼ Cisplatin is the first and most widely used platinum-based chemotherapy drug and is the cornerstone agent in treating a broad spectrum of cancers, including ovarian cancer, testicular cancer, cervical cancer, bladder cancer, lung cancer, head and neck cancer, lymphoma, and brain tumors. It is one of the few curative anti-cancer agents; however, its clinical application is often limited by severe toxic side effects and resistance possessed by some cancers.
Our group has recently, through the femtomedicine approach, unraveled a new molecular mechanism of Cisplatin. It has been found that Cisplatin is extremely effective for the dissociative electron transfer (DET) reaction with weakly-bound electrons to produce reactive radicals that cause DNA strand breaks, apoptosis, and final clonogenic cell kill.
Based on this DET mechanism, it is proposed that Cisplatin may be administered in combination with a biological electron donor to enhance its chemotherapeutic efficacy. We have tried a few combinations of Cisplatin and electron-donating compounds. In this thesis, we show results of the combination: Cisplatin and Rhodamine-B, the one with the greatest potential as demonstrated in both in vitro assays and in vivo xenograft mouse cancer models. This thesis begins with an introduction to cancer and cancer therapies in Chapter 1, where the theory, objective, and scope of this thesis are introduced. Chapter 2 focuses on some new understandings of Cisplatin-induced DNA damage. In vitro and in vivo experiment results on the effectiveness of the combination of Cisplatin and Rhodamine-B are presented in Chapter 3 and Chapter 4, respectively. In Chapter 5, time-resolved femtosecond laser spectroscopic studies on the reaction between Cisplatin and Rhodamine-B are shown. Chapter 6 as the last Chapter summarizes results obtained in this project and proposes some possible future research.
In vitro experiments confirm the potential of this proposed combination of Cisplatin and Rhodamine-B to treat cancer. From cell survival tests, by applying MTT assays and clonogenic assays, it has been shown that our proposed combination significantly enhances the cell-killing efficacy in cancer cells; but surprisingly, not in normal cells. Besides, plasmid DNA gel electrophoresis and γ-H2AX staining in treated cells indicate that more double-strand breaks can be induced using our combination, compared to Cisplatin only. In addition, measurements on Caspase 3/7 activation and Annexin V-FITC labeling flow cytometry experiments clearly show a significant enhancement in the population of apoptotic cells using our combination. To further verify the effectiveness of the combination of Cisplatin and Rhodamine-B, in vivo xenograft mouse models have been developed. Our combination greatly enhances the tumor growth inhibition and even tumor shrinkage in three different mouse models. ▼ Introductie In dit onderzoek is het verband tussen persoonlijkheid en conflict in broer/zus-relaties onderzocht en of er een verschil is tussen homogene en heterogene paren. Verwacht werd dat positieve persoonlijkheidskenmerken (vriendelijkheid, emotionele stabiliteit, zorgvuldigheid en openheid voor ideeën) zouden samenhangen met minder conflict. Meer negatieve of ambigue persoonlijkheidkenmerken (extraversie) zouden samenhangen met meer conflict. Methode Er hebben 886 brugklassers (N=427 jongens en N=459 meisjes), vragenlijsten ingevuld met vragen over hun persoonlijkheid (QBF) en over de mate van conflict (SRQ) in hun broer/zus-relaties. Resultaten De vijf persoonlijkheidskenmerken verklaarden samen 8,5% van het conflict in broer/zus-relaties. Tegen de verwachting in hing extraversie niet significant samen met conflict. Vriendelijkheid, zorgvuldigheid en emotionele stabiliteit bleken alle drie persoonlijkheidskenmerken die, volgens de verwachtingen, een negatief verband hadden met conflict. Tegen de verwachting in bleek openheid van ideeën positief samen te hangen met conflict. Om te onderzoeken of er een verschil is tussen homogene en heterogene broer/zus-paren zijn er interactie-effecten toegevoegd aan de regressieanalyse. Tegen de verwachting in werd het verband tussen persoonlijkheidskenmerken en conflict niet significant gemodereerd door seksecombinatie. Discussie Verschillende verklaringen worden gegeven voor het vinden van de resultaten. ▼ In this study, we examined the cytotoxic effects of
combination therapy with zoledronic acid (ZOL) and gemcitabine (GEM) on pancreatic cancer cells in vitro and in vivo. Four human pancreatic cancer cell lines were treated with ZOL, GEM or a
combination of both, and the effects of the respective drug regimens on cell proliferation, invasion and matrix metalloproteinase (MMP) expression were examined. A pancreatic cancer cell line was also intrasplenically or orthotopically implanted into athymic mice and the effects of these drugs on tumor metastasis and growth in vivo were evaluated by histologic and immunohistochemical analyses.
Combination treatment with low doses of ZOL and GEM efficiently inhibited the proliferation (P < 0.001) and invasion (P < 0.001) of pancreatic cancer cells in vitro. Western blotting assay revealed that MMP-2 and MMP-9 expression levels were decreased after ZOL treatment. In vivo, combined treatment significantly inhibited tumor growth (P < 0.05) and the development of liver metastasis (P < 0.05). These data revealed that, when used in
combination, ZOL and GEM have significant antitumor, anti-metastatic and anti-angiogenic effects on pancreatic cancer cells. In the present study, we first reported the significance of the
combination treatment of ZOL and GEM in pancreatic cancer using an in vivo model. These data show promise for the future application of this drug regimen in patients with pancreatic cancer.

simvastatin, a potent lipophilic statin, and the mechanism of its effect on prostate cancer cells is studied in vitro and in vivo. Results show promising effects of simvastatin on human prostate cancer proliferation, colony formation and migration, in
both androgen-dependent and –independent disease. Effects were tested in vivo and showed a reduction of tumor size in simvastatin treated nude mice compared to controls. Mechanism of effect of simvastatin on cancer cell micrometastasis was also studied
using ECIS, gene arrays, adhesion assays immunocytochemistry and western blotting and showed its effect on integrin αvβ3 activation and consequently adhesion, as well as impairment of tumor secreted factor effect, in addition to stabilization of
endothelial barrier. In this research we also studied the effect of combination on proliferation and migration in vitro as well as on tumor growth in vivo and showed promising effect with a highly significant reduction of proliferation, migration and
tumor growth compared to controls. In conclusion, our pre-clinical research shows beneficial effect of simvastatin alone or in combination with docetaxel in the management of prostate cancer and prevention of micrometastasis.

▼ In a typical functional brain imaging experiment, scientists aim to map the specifi c areas of the brain that are activated while subjects perform a designated task (which may be cognitive, motor, or other). For comparison purposes
(comparing patients vs. controls, females vs. males, as some examples), combining the brain maps from the subjects in an efficient way becomes imperative so that we can get an overall picture of activity for each group. We use statistical tests that have
been developed historically for combining independent sources of information to create maps for each group of subjects in a neuroimaging study. These statistical tests follow two basic approaches - combining p-values and meta-analysis. Through these
methods we aim to draw conclusions about the behavioral pattern of two or more groups with respect to each other. We also want to compare the performance of the diff erent methods. Group comparisons have been done in the past using 'group maps" for each
population through a fi xed e ffects model or a random eff ects model. This dissertation explores some pre-existing statistical combination techniques used in combining and inter- preting 'group maps". We will use parametric and non-parametric approaches
to compare between two or more populations. While combining and comparing brains, there are two aspects that arise - spatial and statistical. We will only focus on the latter aspect. We will assume that the voxels of the brain are independent of each
other. However, as we conduct various tests to compare group maps at each voxel, to minimize false positives i.e. voxels declared active when they are not, we will threshold at each voxel. In this dissertation we will explore thresholding through false
discovery rate, permutation tests and bootstrapping and we compare these methods to draw a conclusion about which one would be apt to use.

▼ This interpretive exploratory case study investigated the challenges and barriers to the multi-grade context in general, as well as to Information and Communication Technology (ICT) implementation in a multi-grade context. Possible solutions based on teacher perceptions were generated inductively within a three-level typology. The research took place in three phases. Pre-training data were gathered in the first phase, using an open-ended questionnaire (all participants: n=20); personal interviews (n=9); and two focus group interviews, which consisted of three participants per focus group. Training on the Intel® Teach ICT programme and the provision of laptops (one per teacher) constituted the second phase. The third phase included post-training data generation using an open-ended questionnaire (all participants) and two focus group interviews, which consisted of three participants per focus group, to elicit teacher perceptions of the training programme and their use of the laptops in the classroom after the training. The findings suggest that first order (school or meso level) challenges pertaining to multi-grade teaching relate to the lack of resources; curriculum challenges; learner related challenges; isolation, impact on communication and teaching; time constraints; and a work overload as a result of the multiple roles. Second order challenges (self or micro level) related to the perceived uncaring attitude of the Department of Basic Education that left teachers feeling hurt and neglected; the unpreparedness for the multi-grade context which demotivated teachers; and the negative perceptions the teachers had about their learners. Third order challenges (system level, beyond the school level or micro level) were related to the lack of Departmental support from officials; the lack of curriculum training for the multi-grade context; and the lack of support on various fronts. Regarding ICT related challenges, the findings suggest that first order barriers related to the lack of suitable infrastructure for ICT implementation; lack of peer support; lack of access to appropriate hardware and software; and lack of time. Second order barriers related to the negative beliefs in the self; negative perceptions about the learners’ ability to use the laptop; and negative beliefs related to teaching and learning. The third order barriers were related to the lack of support and assistance from the Department of Basic Education for ICT implementation. The findings suggest that the participants viewed the provisioning of enabling programmes for parents as an important solution to first order challenges, and a change in mindset as the most important solution for second order problems. Third order solutions included a “multi-grade Renaissance” towards a new model for multi-grade teaching; re-thinking the curriculum requirements regarding multi-grade teaching by the Department of Basic Education; increased support and training from the Department; incentives to teach with ICT in the multi-grade context; encouraging further research…

▼ Sorghum and cowpea contain structurally distinct flavonoids that may provide complementary bioactive properties. This study investigated the interactive effects of combining sorghum and cowpea flavonoids on inflammation and reactive oxygen species (ROS) generation in non-malignant colon myofibroblasts (CCD-18Co). The effect of sorghum-cowpea flavonoid combinations on ATP binding cassette (ABC) transporter proteins in Caco-2 cell model was also investigated.
The White sorghum-white cowpea extract combinations at (0.1 μg/mL) synergistically (1.5-2 times) attenuated LPS-induced ROS generation as well as LPS-induced inflammation in non-cancer CCD-18Co cells by inhibiting the activation of NF-κB and related downstream targets such as IL-6, IL-8 and TNF-α at both mRNA and protein levels.
Combination of pure apigenin (dominant flavonoid in white sorghum) and quercetin (prevalent flavonoid in white cowpea) demonstrated similar enhanced action (1.5-2 times) in protecting the CCD-18Co cells against ROS generation and inflammation at 0.1 μM, indicating the flavones and flavonols in sorghum-cowpea mixtures may be responsible for the observed synergistic effect. The magnitude of synergy was strongly dependent on the relative ratios of sorghum-cowpea and apigenin-quercetin mixtures, which suggests different mechanisms of action for the flavones and flavonols.
The White sorghum-white cowpea as well as apigenin-quercetin combinations synergistically modified the activity of efflux transporters in Caco-2 cell monolayers by inhibiting the expression of apically located ABC transporters including BCRP, MRP2, MRP3 and MDR1 at both mRNA and protein levels. These results suggest that synergistic down-regulation of efflux transporter proteins could be a mechanism for enhanced action sorghum-cowpea
combination against inflammation.
Overall findings from these studies suggest that consuming sorghum and cowpea together may favorably influence health outcomes beyond what can be predicted from individual commodity. This is important because cereal and legumes are usually consumed together; strategic cereal and legume combinations might be formulated for optimized benefits in chronic disease prevention.
Further in vivo studies with sorghum-cowpea diets are required to assess the physiological effects of sorghum-cowpea flavonoids mixtures on biomarkers of inflammation and chronic disease.

▼ Skeleton representations are a fundamental way of representing a variety of solid models. They are particularly important for representing certain biological models and are often key to visualizing such data. Several methods exist for extracting skeletal models from 3D data sets. Unfortunately, there is usually not a single correct definition for what makes a good skeleton, and different methods will produce different skeletal models from a given input. Furthermore, for many scanned data sets, there also is inherent noise and loss of data in the scanning process that can reduce ability to identify a skeleton.
In this document, I propose a method for combining multiple algorithms' skeleton results into a single composite skeletal model. This model leverages various aspects of the geometric and topological information contained in the different input skeletal models to form a single result that may limit the error introduced by particular inputs by means of a confidence function. Using such an uncertainty based model, one can better understand, refine, and de-noise/simplify the skeletal structure. The following pages describe methods for forming this composite model and also examples of applying it to some real-world data sets.

▼ Listeria monocytogenes is a foodborne pathogenic bacterium responsible for ~500
deaths and a financial burden of ~$2.3 billion each year in the United States. Though a zero
tolerance policy is enforced with regard to its detection in cooked ready-to-eat foods, additional
preemptive control alternatives are required for certain products. Among these alternatives are
strategies permitting the usage of food antimicrobial combinations to control the pathogen.
Research on antimicrobial combinations can provide insight into more efficient control of the
pathogen, but is currently lacking.
The purpose of this study was to evaluate the in vitro inhibition of L. monocytogenes
exposed to the antimicrobials e-Poly-L-Lysine (EPL), lauric arginate ester (LAE), and sodium
lactate (SL) at pH 7.3, octanoic acid (OCT) at pH 5.0, and nisin (NIS) and acidic calcium sulfate
(ACS) at both pH 5.0 and 7.3. A broth dilution assay was used to determine single antimicrobial
minimum inhibitory and bactericidal concentrations for L. monocytogenes Scott A, 310, NADC
2783, and NADC 2045. Optical density differences (delta<0.05 at 630 nm) were used to denote
inhibition. Concentrations producing population decreases of greater than or equal to 3.0 log10 CFU/ml after incubation
were considered bactericidal.
Inhibition resulting from combinations of antimicrobials (NIS+ACS, EPL+ACS,
SL+ACS, NIS+LAE, OCT+ACS, and OCT+NIS) was assessed using a checkerboard assay, and fractional inhibitory concentrations (FIC) were determined. FIC values were plotted on
isobolograms and were used to create FIC indices (FICI). Isobologram curvature was used to
classify combinations as synergistic, additive, or antagonistic. From FIC indices, interactions
were defined as antagonistic (FICI >1.0), additive (FICI =1.0), or synergistic (FICI <1.0).
Strain-dependent factors had a bearing on MIC and MBC values for NIS and EPL. At
pH 7.3, NIS+ACS displayed synergistic inhibition, NIS+LAE and EPL+ACS demonstrated
additive-type interactions, and the SL+ACS pairing was unable to be defined. At pH 5.0,
interpretation of the OCT+NIS interaction also presented challenges, while the OCT+ACS
combination resulted in synergistic behavior.
Additional studies are needed to validate in vitro findings on surfaces of ready-to-eat
meats. Future in vivo studies should investigate the ability of synergistic combinations
(NIS+ACS and OCT+ACS) to control the pathogen. Better characterizations of inhibitory
mechanisms should also be performed.

Melanoma is an aggressive form of cancer characterised by poor prognosis and resistance to treatment. Despite recent advances in melanoma treatment, patient outcomes continue to be underwhelming. After these new therapies showed initial promise; resistance and poor duration of response have limited their effectiveness as monotherapies. We are currently at the forefront of a wave of research into the underlying mechanisms of melanoma characteristics. It is now understood that for effective treatments to be developed, we must have a greater understanding of the genetic mechanisms of melanoma’s treatment resistance and abnormally aggressive phenotype. Ultraviolet radiation (UVR) has continually been intrinsically linked to melanoma development. In addition to this, resistance to traditional DNA-damaging chemotherapy drugs such as cisplatin remains a hallmark of melanoma cells. Nucleotide excision repair (NER) orchestrates the repair of helix distorting DNA damage, induced by both ultraviolet radiation (UVR) and cisplatin. There is evidence that the global genome repair (GGR) arm of NER is dysfunctional in melanoma and it is known to have limited induction in melanoma cell lines after cisplatin treatment. To further investigate the links between NER and melanoma, we examined the expression of multiple key genes involved in the NER pathway function and regulation, in a cohort of melanoma tumours. The expression of these transcripts was then compared to clinical parameters. In addition to this, expression of GGR damage recognition protein, XPC, was also quantified in melanoma tumours.

▼ The use of drug combinations (DCs) in cancer therapy can prevent the development of drug resistance and decrease the severity and number of side effects. Synthetic lethality (SL), a genetic interaction wherein two nonessential genes cause cell death when knocked out simultaneously, has been suggested as a method of identifying novel DCs. A combination of two drugs that mimic genetic knockout may cause cellular death through a synthetic lethal pathway. Because SL can be context-specific, it may be possible to find DCs that target SL pairs in tumours while leaving healthy cells unscathed.
However, elucidating all synthetic lethal pairs in humans would take more than 200 million experiments in a single biological context – an unmanageably large search space. It is thus necessary to develop computational methods to predict human SL.
In this thesis, we develop connectivity homology, a novel measure of network similarity that allows for the comparison of interspecies protein-protein interaction networks. We then use this principle to develop Species-INdependent TRAnslation (SINaTRA), an algorithm that allows us to predict SL between species using protein-protein interaction networks. We validate it by predicting SL in S. pombe from S. cerevisiae, then generate over 100 million SINaTRA scores for putative human SL pairs. We use these data to predict new areas of cancer combination therapy, and then test fifteen of these predictions across several cell lines. Finally, in order to better understand synergy, we develop DAVISS (Data-driven Assessment of Variability In Synergy Scores), a novel way to statistically evaluate the significance of a drug interaction.

▼ This thesis looks at conceptual combination, in particular it investigates how noun noun compounds are interpreted. Several themes run throughout the work. Real compounds (e.g. coat hanger, crab apple) are compared to novel ones (e.g. banjo cactus, zip violin). Also, compounds are examined in each of the possible permutations of artefacts (A) (e.g. coat, banjo) and natural kinds (N) (e.g. crab, cactus), (AA, AN, NA and NN).Experiments 1 - 4 examine noncompositionality in noun noun compounds. Possible sources of noncompositionality are investigated using both feature listing and feature rating tasks. Although some differences were found, results were similar between different types of compound, evidence of noncompositionality being found in each. The results also confirm that most of the meaning of a noun noim compound is derived from the second constituent (noun2).Experiments 5 and 6 look at two different types of compoimd interpretation - slot filling and property mapping. In experiment 5, slot filling is found to be the preferred interpretation type overall, but property mapping is more common in compounds composed of two natural kinds (NN). Experiment 6 examines possible factors influencing the choice between slot filling and property mapping interpretations. It was found that constituent similarity plays an important role, and also that this interacts with whether or not the constituents have important properties which clash. Experiment 7 looks at compound identification. Results suggest that the first constituent (nounl) may be critical in such tasks. Experiment 8 compares the importance of nounl and noun2 in determining the type of interpretation given to a compound. Neither position is found to be more influential than the other, although relational information does seem to be associated with specific nouns in each position. Throughout the thesis findings are related to current theories of conceptual combination, such as prototype models, the concept specialisation model and theories of compound interpretation by analogy.

▼ Large arrays of coherently-combined Quasi-Continuous-Wave (QCW) fibre lasers have been identified as a plausible solution to creating high-repetition rate (~1 kHz), high-wall-plug efficiency lasers capable of driving a wakefield for accelerator applications. The coherent combination of four near-identical Photonic Crystal Fibre (PCF) lasers to produce laser pulses with pulse energies of 5.7 mJ and peak powers of 22 GW has been successfully demonstrated by other groups. Extensions of the four PCF experiment also demonstrated the coherent combination of 8 photonic crystal fibres to attain pulse energies of 12 mJ and peak powers of 35 GW. However, these PCFs were near-identical in length, core size, fibre structure and dopant concentration. Work done in Oxford focused on the coherent combination of fibre lasers which were unmatched in length, core size, internal structure and dopant concentration. The aim was to identify the point at which the fibre differences caused the combining efficiency to degrade. Potentially, this information would be used to identify the tolerance levels required for the large-scale manufacture of the fibres needed to construct a facility-scale laser system for laser-wakefield acceleration. This thesis covers the coherent combination of mismatched PCFs, a scenario which could occur during the construction of a facility-scale terawatt laser system. Here, hundreds or potentially thousands of PCFs could be purchased from different manufacturers, each of whom use their own methods of manufacture and tolerances. An investigation into the challenges associated with combining two highly-mismatched PCFs was performed, with the PCFs chosen to be very different to exaggerate any effects which could degrade their coherent combination. This work resulted in the successful coherent combination of these highly-mismatched PCFs, with a measured degree of linear polarisation of 85.9 ± 0.5 % and a fraction of available beam power of 93.0 ± 0.5 %. Also, supporting work investigated the difference between simulation and measurement in explaining the observed degree of linear-polarisation of the combined-beam.

▼ This dissertation examines the forecast model selection problem in economics in both theoretical and empirical settings. The forecast model selection problem is that there often exists a menu of different suitable models to forecast the same economic variable of interest. The theoretical portion of this dissertation considers agents who face this problem in two distinct scenarios. The first scenario considers the case where agents possess a menu of different forecast techniques which includes rational expectations but where the selection of rational expectations is costly. The assumptions that are necessary to include rational expectations as a choice are characterized and the equilibrium dynamics of a model under the appropriate assumptions is studied and shown to exhibit chaotic dynamics. The second scenario considers agents who possess a menu of econometric forecast models and examines the equilibrium outcomes when agents combine the different forecasts using strategies suggested by the forecasting literature. The equilibrium outcomes under these forecasting assumptions are shown to exhibit time-varying volatility and endogenous structural breaks, which are common features of macroeconomic data.
The empirical portion of the dissertation proposes a new dynamic
combination strategy for the forecast model selection problem to forecast inflation. The procedure builds on recent research on inflation persistence in the U.S. and on explanations for the efficacy of simple
combination strategies, often referred to as the forecast
combination puzzle. The new
combination strategy is shown to forecast well in real-time out-of-sample forecasting exercises.

▼ [ES] Los tratamientos en los que se utilizan Polímeros Terapéuticos ofrecen numerosas ventajas en comparación con los tratamientos convencionales y otros enfoques de nanomedicina. Entre estas ventajas se puede destacar la especificidad para cruzar ciertas barreras biológicas y su capacidad de acumulación pasiva en tumores. Además, la conjugación de fármacos a polímeros ofrece ventajas adicionales tales como una farmacocinética mejorada, multivalencia, co-entrega de fármacos en la proporción deseada y liberación/activación específica en el sitio de acción requerido a través de la aplicación de enlaces polímero-fármaco que responden a estímulos fisiológicos. Uno de los tipos más importantes de polímeros que se utiliza para la administración de fármacos pertenecen a los polielectrolitos polipeptídicos. Su uso se debe principalmente, a su biocompatibilidad, biodegradabilidad, multivalencia y versatilidad estructural, así como a la plasticidad sintética en la modificación de cadenas laterales.
La aplicación conjunta de ciencia de polielectrolitos con otras ramas de la química es muy prometedora; sin embargo, aún permanece en un estadío temparano en su desarrollo. Esto es debido a que, el control del autoensamblaje de polielectrolitos sigue siendo una tarea complicada y la investigación en esta área puede resultar muy laboriosa a la hora de encontrar sistemas biocompatibles más avanzados con un único perfil de acción y por supuesto, se abre un nuevo campo de estudio sobre las nuevas propiedades desconocidas de estos. Este tema es novedoso por la posibilidad de realizar diferentes estudios de combinación de polielectrolitos con residuos supramoleculares y representa el estudio de nuevas arquitecturas potencialmente mas complicadas.
En la presente tesis doctoral se estudiarán el desarrollo de sistemas de administración de fármacos basados en polielectrolitos supramoleculares con un alto grado de control sobre las propiedades fisicoquímicas, centrándose principalmente en el control de la forma y el tamaño. Se han estudiado en profundidad varias familias de poliglutamatos de forma estrella con núcleos de diferente hidrofobicidad para determinar cómo la estructura del núcleo y la longitud de la cadena de polielectrolitos afectan el mecanismo de autoensamblaje.
Una vez que se definieron estas correlaciones, se seleccionaron los candidatos más prometedores para la preparación de dos sistemas de transportede fármacos que consisten en partículas esféricas o en forma de cilindro. Finalmente, también se realizó la conjugación de varios fármacos (fasudil y dinaciclib) como agentes únicos o en combinación a través de diferentes enlaces biodegradables. Las propiedades fisicoquímicas y la actividad in vitro de los conjugados se estudiaron en profundidad y actualmente se están llevando a cabo experimentos in vivo en un modelo de cáncer de mama metastásico triple negativo ortotópico preclínicamente relevante, con los conjugados previamente seleccionados.

combination is a cognitive process that produces complex concepts (e.g., adjective-noun pairs) from simple concepts. The Selective Modification Model (SMM; Smith, Osherson, Rips, & Keane, 1988) postulates that simple adjective-noun combinations (e.g., red apple) are understood by the modifier red selecting the colour attribute of the head noun apple. Theories of conceptual
combination have not extended to fulfill our understanding of how complex adjective-noun pairs (e.g., empty dream) are processed. This exploratory study had two main objectives: to determine which semantic variables best captured the processing of complex adjective-noun pairs and to examine the semantic effects of conceptual
combination to extend current theories. Adjective-noun combinations were manipulated based on subjective ratings (i.e., concreteness and plausibility; see the preliminary study) or objective measures (i.e., age of acquisition and semantic distance) and compared. Two hundred and ninety-three participants were randomly assigned to complete one of three computerized tasks that differentially engaged semantic processing from shallow to deep, including the non-pronounceable double lexical decision task (Experiment 1), the pronounceable double lexical decision task (Experiment 2), and the meaningfulness task (Experiment 3). Across all tasks, the subjective model outperformed the objective model in reaction time and accuracy analyses. Adjective-noun processing was facilitated by concrete, early acquired head nouns, as well as adjective-noun pairs that were rated as plausible and situated close in semantic space. Interestingly, adjectives paired with abstract head nouns were difficult to process across tasks regardless of how plausible the pair was. In conclusion, semantic variables rated by participants are valuable and may better capture how the mental lexicon is organized and accessed, and further research should pursue innovative ways of examining how abstract head nouns are processed to incorporate into existing theories.

Los conjugados poliméricos son nanoconstrucciones multicomponente presentes actualmente en clínica como terapia anticancerígena, tanto como agentes únicos, como formando parte de combinaciones. Estos nanoconjugados tienen el potencial de mejorar farmacológicamente el tratamiento de tumores sólidos, debido a una acumulación pasiva en el tumor (efecto ‘EPR’) y a un diferente mecanismo de internalización celular y posterior liberación del fármaco(s). La transferencia de conjugados polímero-proteína a uso clínico rutinario, y el desarrollo clínico de conjugados polímero-fármaco anticancerígeno sitúa a los conjugados poliméricos como una de las primeras clases de nanomedicinas con potencial terapéutico antitumoral ya demostrado. La experiencia obtenida con los primeros nanoconjugados en clínica, ha proporcionado las bases para el desarrollo de polímeros conjugados más sofisticados de segunda generación con propiedades terapéuticas mejoradas. El desarrollo de nuevos portadores poliméricos biodegradables con propiedades mejoradas, la utilización de terapia de combinación o el diseño de conjugados dirigidos a nuevas dianas moleculares son algunas de las aproximaciones a seguir para conseguir conjugados más específicos y efectivos considerados de segunda generación. La propiedad de multivalencia que poseen los polímeros nos permite la conjugación de varios compuestos activos en el mismo esqueleto polimérico, la combinación del modulador activo con un citotóxico, otra sustancia activa o un residuo dirigente puede aumentar marcadamente el valor terapéutico de estas macromoléculas. En este sentido, el punto de partida de la presente propuesta es un nuevo concepto establecido por nosotros en 2005 con el desarrollo de HPMA copolimero-AGM-Dox, basado en la terapia de combinación (terapia endocrina + quimioterapia dentro de la misma matriz polimérica) para el tratamiento de cáncer de mama hormono-dependiente (Vicente et al., 2005). El valor terapéutico de esta nueva estrategia ha sido previamente demostrado en modelos celulares, lo que implicaría una clara aplicación potencial en el tratamiento de tumores hormono-dependientes en mujeres postmenopáusicas.
En el presente proyecto, se plantea corroborar la actividad antitumoral del nanoconjugado de combinación HPMA copolimero-AGM-Dox utilizando modelos in vivo, así como elucidar las bases moleculares responsables de su destacada actividad anticancerígena. La comprensión del mecanismo de acción molecular nos permitirá el diseño futuro de nanoconjugados de combinación con propiedades mejoradas. En una segunda etapa se propone mejorar el valor terapéutico de esta nueva terapia de combinación mediante la utilización de un polímero biodegradable como portador, ácido poli-L-glutámico (PGA), desarrollando un nuevo conjugado de combinación PGA-AGM-Dox y evaluando su actividad antitumoral en modelos in vitro e in vivo. combination of the screw connection. A previous cold-formed steel clip angle test program (which is Phase 1) developed design methods for clip angle. Therefore, the object of this thesis is to further investigate the behavior and design methods of loading-bearing cold-formed steel clip angles under different screw pattern. For each limit state, a test program was conducted to investigate the behavior, strength, and deflection of the clip angle. The test result were compared with previous CFS clip angle design method. Amending existing CFS clip angle method were developed by each of the four limit states studied in this project.

▼ This thesis will defend a new variety of panprotopsychism which does not have a class of so-called combination problems that some popular varieties of panprotopsychism have. Panprotopsychism is the view that conscious entities depend on protophenomenal constituents and that these constituents are fundamental, ubiquitous and neutral. One very popular variety, Constitutive Russellian Panprotopsychism (CRP) has three additional distinctive doctrines. (1) Russellian Monism: protoconsciousness is located in a fundamental constituent’s intrinsic nature. (2) Neutral Monism: protophenomenal properties are neither physical nor phenomenal. (3) Constitution: the right combination of protophenomenal constituents alone is meant to produce consciousness. There are at least three combination problems for CRP. The first is that the conditions for the combination of protoconscious entities, and the nature of these bonds, is a mystery. Consequently, we can conceive of a structurally and protophenomenally duplicate world without any phenomenal properties. This is the Conceivability Problem. Secondly, consciousness seems unified and monadic, which is incompatible with the idea that consciousness has parts. However, if consciousness cannot have parts strong emergence seems necessary, which is undesirable. This is the Composition Problem. Finally, even though microphysical and protophenomenal properties are meant to be isomorphic, the structure of our brains and visual fields are not. This is the Structural Mismatch Problem. I defend Structural Transformational Identity Theory Panprotopsychism (STIP) as a superior alternative. STIP has three distinctive doctrines. (1) Identity Theory Panprotopsychism: fundamental constituents are both simultaneously physical and protophenomenal. (2) Transformational Fusion: multiple fundamental constituents go through intrinsic change to generate a singular simple. (3) Modified Ontic Structural Realism: fundamental constituents, including protophenomenal constituents, are not relata, but relations. Some of these relations are microphysical while others are protophenomenal. My conclusion is that the three combination problems for CRP do not exist for my STIP even though the latter view preserves all of the original intuitions of the former view.

▼ Hypertension is a leading public health problem globally, particularly in low- and middle-income countries. The current approach to pharmacological management of hypertension has limitations and may contribute to poor hypertension control. A strategy of early use of lowdose combination therapy may improve outcomes but there is uncertainty about its potential effects. Methods Epidemiology of hypertension in India and Sri Lanka was described. Systematic reviews and meta-analyses were conducted to quantify the dose-related efficacy and safety of combination BP lowering drugs. Data from qualitative interviews of providers and patients were analysed to explore barriers to use of preventive drugs for CVD and acceptability of combination therapy as a solution to key barriers. Data from a large study in rural India were analysed to model the potential benefits of early use of low-dose triple combination therapy at a population level. Results The prevalence of uncontrolled hypertension among adults in India and Sri Lanka is currently estimated to be >70%. Meta-analyses of RCTs showed that compared to placebo, initial therapy with low-dose dual combination reduced BP by 10.1/5.9 mm Hg, whereas low-dose triple combination, tested as replacement therapy in only one trial, reduced BP by 17.9/9.8 mm Hg. Low-dose triple combination therapy vs. one std. dose monotherapy reduced BP by about 10/4 mm Hg. While there was no clear difference in tolerability between low-dose dual or triple combination therapies compared to placebo, data were sparse. From a patient and provider perspective, complex regimens and cost of drugs are major barriers to use of prescribed drugs. The use of combination therapy to simplify drug regimens and reduce cost was highly acceptable. The protocol for a new trial to address relevant evidence gaps is described. If the anticipated effects of this trial were applied at a population level the strategy of early use of low-dose combination therapy compared to the stepped care therapy can reduce BP by 7.8/3.5 mm Hg and improve BP control by 11%. Conclusion A strategy of early use of low-dose triple combination therapy has large potential to improve hypertension control safely and effectively, and is likely to be cost-effective whilst being acceptable to patients and providers. However, current data are insufficient and the ongoing TRIUMPH study will provide more definitive evidence.

▼ Although platinum drugs cisplatin, carboplatin and oxaliplatin are routinely used in the clinic to treat various cancers including testicular, ovarian, head and neck, and colorectal cancers, problems of drug resistance and side effects would limit their use. Thus thousands of cisplatin analogues have been prepared by changing nature of the leaving groups and carrier ligands with the aim of reducing the side effects and widening the spectrum of activity. It has been possible to reduce side effects by changing the nature of the leaving groups e.g. carboplatin that has bicarboxylate leaving group as against chloride in cisplatin is less toxic than cisplatin. Changing the carrier ligands has impacted activity to the compounds and types of cancer that is successfully targeted. For example, oxaliplatin which has DACH (1R, 2R)-(-)-1, 2-diaminecyclohexane) carrier ligand as against ammonia in cisplatin, is active against colorectal cancer while cisplatin and carboplatin are not. Substantial resemblance between chemistry of palladium (II) and platinum (II) has prompted studies of Pd(II) complexes as potential anticancer drugs. Combining drugs with different modes of action can synergize their effects, where a combination therapy comprised of different mechanisms often provide distinct advantages over monotherapy. Much research is currently done on the combination of targeted therapy with phytochemicals. In this study, six novel palladium compounds (NH1, NH2, NH3, NH4, NH5 and NH6) have been synthesized using bulky planaramine ligands: quinoline, benzimidazole, 8-hydroxyquinoline and imidazo(1,2-α)pyridine. Moreover five tumour active phytochemicals: curcumin, EGCG, emetine, patulin and 6-shogaol have been combined with commonly used Pt based drugs as well as designed palladium compounds with the aim of providing a means of overcoming cisplatin resistance in ovarian and colorectal cancers. All of the designed palladiums have been characterized by elemental microanalysis, IR spectroscopy, 1H-NMR spectroscopy, Mass spectroscopy. Moreover, three of the compounds (NH3, NH4 and NH6) have been characterized by single x-ray crystallography. Anticancer activity of the compounds were tested against eight different human cancer cell lines (ovarian cancer: A2780, A2780cisR and A2780ZDO473R; colorectal cancer: HT-29 and CACO-2; breast cancer: MCF-7; cervical cancer: Hela) by MTT reduction assay. NH3 has shown 10 to 96 times greater activity than cisplatin against all tested cell lines while NH1 has shown comparable activity. In binary combination with curcumin and EGCG, NH3 has shown sequence and concentration dependent synergism against ovarian and colorectal cancer models. 6-shogaol and curcumin in combination with NH1 have also shown synergism in many instances. ▼ Albendazole (ABZ) is a benzamidazole derivative that binds to β-tubulin and inhibits the polymerisation of microtubules. ABZ has a remarkable activity against a variety of tumour cell lines including colorectal, liver, and ovarian cancer cells. In pre-clinical models, ABZ has been shown to have both anti-tumour and anti-angiogenic activities. Nevertheless, poor aqueous solubility of ABZ limits its absorption, and as a result, its efficacy. Therefore, the first aim of this project was to improve the aqueous solubility of ABZ. Using a combination of ionisation with acid and complexation with hydroxypropyl-β-cyclodextrin (HPβCD), a relatively high concentration of ABZ in solution was achieved. Comparison of pharmacokinetic profiles of ABZ/HPβCD with a conventional formulation of ABZ in hydroxypropyl methylcellulose (ABZ/HPMC) in nude mice revealed that complexation with HPβCD results in a significantly higher peak plasma concentration (Cmax) and area under the curve (AUC) of ABZ and its metabolites. Moreover, in mice-bearing human colorectal cancer cells, HCT-116, ABZ/HPβCD treatment led to a significant delay in tumour growth with increase in survival of animals as compared with vehicle and ABZ/HPMC treatments.The second aim of this research was to improve the efficacy of ABZ using combination therapy. To this end, the interaction between ABZ and different chemotherapeutic agents were assessed using the Sulforhodamine B assay (SRB) and quantified by median effect analysis method. Among the tested agents, a synergistic anti-proliferative effect was observed with the combination of ABZ and 2-methoxyestradiol (2ME) in HCT-116 and DU145. Of interest, 2ME, a microtubule targeting agent binds to similar colchicine-binding site of β-tubulin as ABZ and inhibits microtubules polymerisation. Synergistic interaction of ABZ and 2ME was accompanied with the activation of extrinsic pathway of apoptosis.In vivo, the combination of low concentration of ABZ with 2ME resulted in an increase in the survival rate of mice-bearing HCT-116 tumours. This effect was accompanied by a decrease in plasma and tumour vascular endothelial growth factor (VEGF) as well as a reduction in microvessel density. In addition, combination therapy led to a significant decrease in proliferation rate of the tumour and an increase in apoptosis. Noticeably, high concentration of ABZ, in combination with 2ME, resulted in an antagonistic effect on tumour growth and survival of the animals. Taken together, the solubility and anti-tumour efficacy of ABZ was highly increased by complexation with HPβCD, leading to the conclusion that the formulation may be suitable for parenteral administration. Moreover, combination of ABZ and 2ME has shown promising results in pre-clinical model. Additionally, the finding that the combination of two microtubule-binding agents that share the same binding site can act synergistically in vitro and in vivo may lead to the development of new therapeutic strategies in cancer treatment.

▼ This dissertation spans three distinct methodological contributions to the econometric and analytic modeling literature. The first chapter is motivated by the challenge of estimating the impact of a customized multi-treatment coupon campaigns on visit rates for a large panel across a two-year trial. Prior literature has highlighted the necessity for both detecting and mitigating the issue of endogeneity. In response, I first develop a novel robust Wald test for idiosyncratic endogeneity in the presence of individual heterogeneity and a time-varying endogenous regressor of restricted range for a nonlinear, unobserved effects model. Building on a prior working paper, I extend the results to tackle the identification of idiosyncratic endogeneity in the presence of time-constant endogeneity for unbalanced panels. I propose a two-stage estimation procedure that tests for positive covariance between time-varying unobservables and a time-varying, binary endogenous variable that completely controls for the latent, time-constant heterogeneity for count responses. Simulations suggest endogeneity detection for both unbalanced panels scenarios mirrors the balanced panel benchmark closely using metrics of rejection rates, nominal size, and statistical power given proper instrumental variables. A nonlinear, instrumental variables GMM procedure is proposed for parameter estimation given positive endogeneity detection. The second chapter studies the estimation of a nonlinear (generalized additive) mixed effects models for count data containing parametric factor smooths. These models are further extended to estimate Poisson responses generated from both stationary and non-stationary conditionally serially correlated AR (1) processes. Simulation studies are used to verify the accuracy of these models in a range of scenarios. The models are then applied to the problem of estimating a carryover effects of a promotional campaigns after promotions end. The final chapter seeks to extend the forecasting combination model of Granger and Ramanathan by approximating the optimal conditional mean solution with conditional quantiles. Useless forecasters at arbitrary quantiles of the response are discarded via a LASSO penalty. The final combination forecast outperforms the equal weight combination benchmark as well as a variety of other combination models in terms of out-of-sample predictive power and other significant performance metrics at the expense of marginally increased forecast variance.

▼ O processo de internacionalização dos meios de produção e o aumento do volume do comércio mundial de bens, de serviços e de capitais reforçam a necessidade de uma linguagem universal, confiável e comum para o setor empresarial. Esta linguagem está presente nos relatórios contábeis e financeiros. Como as informações contábeis podem variar substancialmente de um país para outro, de acordo com os princípios de contabilidade que os governam, e com as práticas empresariais adotadas, as Normas Internacionais de Contabilidade IASB buscam a convergência das normas contábeis de cada país para as Normas Internacionais de Relatórios Financeiros IFRS. O presente trabalho visa detectar os impactos provenientes das operações de business combinations, nas demonstrações financeiras apresentadas pelas empresas Britânicas do segmento de petróleo, gás e energia que negociam títulos na bolsa de Londres. A pesquisa envolveu uma amostra de 19 empresas no total de 44 organizações, que são listadas na Bolsa de Londres (FTSE). Esse mercado foi escolhido por ele ser membro da União Européia e por esta organização obrigar todas as empresas com ações em suas bolsas a apresentarem suas demonstrações contábeis de acordo com as normas internacionais, a partir de 2005. Mas também, porque as empresas deste segmento fazem parte dos dois principais índices de avaliação da bolsa de Londres, O FTSE 100 e FTSE 250. Como metodologia, utilizou-se a pesquisa documental e a análise de conteúdo. O estudo considerou os seguintes aspectos: forma de cumprimento das IFRS em 2005, uso das exceções previstas pela IFRS 1 e a adoção da IFRS 3 - Business Combination. A pesquisa mostrou que todas as empresas pesquisadas apresentaram as demonstrações contábeis em IFRS em 2005. Houve alterações nos valores do ativo da empresas, a maioria para menor. Somente oito empresas evidenciaram impacto do goodwill nos relatórios contábeis. Todas as empresas informaram que, no balanço de abertura, foi realizada a reavaliação dos ativos pelo teste de Impairment, e não houve mais amortização de goodwill, conforme determinado pelo IASB. O número de empresas pesquisadas foi pequeno em relação à quantidade de empresas com ações negociadas na Bolsa de Valores de Londres, em função da limitação do setor de petróleo, gás e energia. Essas considerações expõem limites, mas mostram que, para as empresas britânicas, o processo de convergência das demonstrações contábeis para a Norma Internacional irá continuar.

▼ ENGLISH ABSTRACT: In traditional medicine, there is a long-standing culture of combining herbal drugs to increase the therapeutic efficacy. The improved medical action is thought to be due to synergistic interactions between different plant bioactive components. The aim of this study was to test the pharmacological interactions in a medicinal plant
combination which consisted of Agathosma crenulata, Dodonaea viscosa and Eucalyptus globulus. The rationale for the analysis of this particular mixture is that it had noteworthy antibacterial activity and exhibited the highest activity out of seven medicinal plant mixtures previously investigated. Using chromatographic analysis, the phytochemistry of the plants was also assessed.
The chloroform: methanol (1:1; v/v) extracts or hydo-distilled essential oils (A. crenulata and E. globulus) were screened individually and in combinations (double and triple plant
combination) for activity against five respiratory pathogens using a microdilution assay. The antimicrobial interactions in combinations were assessed with the fractional inhibitory concentration (FIC) and the isobolograms. The organic extracts generally showed the highest antimicrobial activity with E. globulus having the highest activity with MIC values below 1 mg ml-1 representing noteworthy activity. The overall activity of the aqueous extracts was poor. The essential oil activity of E. globulus was mostly noteworthy (0.5 to 2 mg ml-1) while A. crenulata essential oil displayed moderate activity (1 to 4 mg ml-1).
The ΣFIC values for double combinations (1:1) of A. crenulata with D. viscosa, A. crenulata with E. globulus and D. viscosa with E. globulus were calculated from the minimum inhibitory concentration (MIC) data and the interactions were classified as synergistic, additive, indifferent and antagonistic. The highest synergistic interactions observed were for a 1:1
combination of A. crenulata with E. globulus against K. pneumoniae, S. aureus and B. subtilis with ΣFIC values of 0.07. There was only one incident of antagonism noted in the study for D. viscosa with E. globulus (1:1) against C. neoformans with ΣFIC value of 4.25. The double combinations against selective pathogens (K. pneumoniae, S. aureus and E. coli) were further analysed for interactions using isobolograms. Mostly, the antimicrobial interactions as presented by the isobolograms were congruent with FIC results which further validated the occurrence of relevant antimicrobial interactions in those combinations. The ΣFIC values for triple combinations (1:1:1) revealed mostly synergistic interactions. When the triple combinations were analysed further against certain pathogens based on the predictions of the Design of Experiments software program (MODDE 9.1®), the MIC values remained the same despite the different combinations that were tested
