[EN] The use of biomimetic media such as serum albumins (SAs), a1-acid glycloproteins (AAGs), cyclodextrins (CDs) and sodium dodecyl sulfate (SDS) micelles is combined with photophysical techniques (fluorescence (F) and laser flash photolysis (LFP)) to investigate the issue of photosensitization by drugs or drug derivatives containing a triciclyc heteroaromatic chromophore, namely carbazole (CBZ) or phenothiazine (FTZ). The photobehavior of the methyl ester of carprofen (CPFMe) in the presence of bovine serum albumin (BSA) has been addressed first. The decay of triplet excited state of CPFMe in the presence of BSA at 1:2 (at 430 nm), needed two monoexponential terms and thus two triplet lifetimes for a satisfactory fitting, which revealed binding to two different sites within the protein. The shorter value of triplet lifetime was ascribed to CPFMe present in site I, based on the quenching of 3CPFMe* by the tryptophan (Trp) residue located in this pocket, through an electron transfer mechanism. No significant differences were found between (S)- and (R)-CPFMe. Within bovine a1-acid glycloprotein (BAAG), only one binding site was observed for both CPFMe enantiomers. Stereodifferentiation was observed in the triplet lifetimes in [email protected] complexes, with the shorter lifetimes values for the (S)-enantiomer. CPFMe photobinding to BAAG was detected by following the changes in fluorescence of [email protected] mixtures before and after irradiation. The process was more efficient for (S)-CPFMe. The proposed mechanism involves a reductive photodehalogenation from the triplet of (S)-CPFMe, giving rise to covalent adduct (S)-CBZMe-BAAG. As a way to circumvent the unfavorable thermodynamics of homolytic C-Cl cleavage through the triplet excited state, it has been proposed that the actual operating mechanism involves formation of triplet excimers. To deepen into the mechanism of CPFMe dehalogenation, two diastereomeric dyads based on CPF (CPF-CPF and CPF-CBZ) have been synthesized and irradiated. The self-quenching of the triplet excited states in both dyads is much faster than in CPFMe, and is related to formation of charge transfer species, clearly disfavored in non-polar solvents (THF). The trends observed in the triplet lifetimes, as well as the solvent effects on photoreactivity, are in full agreement with the mechanistic picture of a photoreductive dechlorination as operating mechanism in CPF-based systems. The photochemistry of the neuroleptic drug cyamemazine (CMZ) has been investigated in biomimetic media. The encapsulation process has been followed by fluorescence, as a hypsochromic shift of the band, concomitant with enhanced quantum yields and lifetimes. LFP revealed important changes associated with encapsulation, specifically more selective generation of the triplet excited state, which turns to be longer-lived. Light-induced oxidation of CMZ afforded the corresponding radical cation, which was trapped by oxygen to afford the N,S-dioxide. Introducción: Tramadol y carprofeno son fármacos ampliamente usados en medicina veterinaria para el tratamiento del dolor agudo y crónico por su eficacia, potencia y seguridad. Debido a la escasa evidencia científica acerca del tipo de interacción farmacológica que presentan en su co-administración, este estudio intento evaluar la naturaleza de la interacción usando el análisis isobolográfico de la administración conjunta de tramadol y carprofeno en 3 modelos de dolor agudo experimental en ratones (test de las contorsiones, Test de la formalina del miembro posterior y test de la plancha caliente). Material y métodos: Se seleccionaron ratones machos cepa CF/1, fueron aleatorizados en 3 grupos, con un n=24 para cada grupo. A cada grupo se le realizó uno de los test algesiométricos: Grupo 1: Test de las contorsiones abdominales, Grupo 2: Test de la formalina y Grupo 3: Test de la plancha caliente. A cada grupo se le administró 1:1 de sus ED50 de tramadol y carprofeno. El comportamiento del animal se consideró como signo de dolor siendo especifico para cada test. La antinocicepción se expresó como promedio ± su error estándar (EEM) del Emax o sus respectivos intervalos de confianza del 95%, los valores se analizaron por medio del programa Pharm Tools Pro, version 1.27, The Mc Cary Group, USA. Fue un estudio controlado y doble ciego para la administración de fármacos y el análisis estadístico. Resultados: En los 3 análisis isobolográficos se demostró que la co-administración de las DE50 de tramadol y carprofeno produce una interacción farmacológica antinociceptiva de tipo sinérgica la que es confirmada por sus respectivos índices de interacción. Discusión: Estos resultados demostrarían que la co-administración de carpofeno y tramadol producen un efecto analgésico sinérgico, que es concordante con la teoría de la analgesia multimodal, al poseer cada fármaco distintos mecanismos de acción ambos fármacos se potencian. Además, la literatura describe ampliamente este tipo de interacción, al combinar un AINE (principalmente selectivo COX2) y un opioide, actuando estos en los distintos blancos de la fisiología del dolor. A identificação da dor animal é um aspecto com cada vez maior importância na produção animal, assim como o é minimizar os processos dolorosos ao longo de todos os procedimentos de produção. Quando não for possível eliminar esses estímulos dolorosos, deve ser dada prioridade à devida analgesia. Um desses procedimentos dolorosos é a cirurgia, neste caso, a cirurgia correctiva de deslocamento de abomaso à esquerda. Como não é possível eliminar os estímulos dolorosos resultantes da cirurgia, o que nos resta é proceder à devida analgesia pós-cirúrgica. O objectivo deste trabalho foi perceber se a administração de carprofeno possui maior duração de analgesia do que a flunixina meglumina no maneio de dor pós-cirúrgico na correcção de Deslocamento de Abomaso à Esquerda (DAE). O carprofeno, tal como a flunixina meglumina, é um anti-inflamatório não esteróide, mas é menos utilizado na analgesia pós-cirúrgica devido ao seu custo, apesar de possuir um tempo de semi-vida superior ao da flunixina meglumina. O estudo foi realizado em 26 vacas de raça Holstein Frísia submetidas a correcção cirúrgica de DAE, que foram divididas em dois grupos: Grupo F (14 animais submetidos à administração de flunixina meglumina) e o Grupo C (12 animais submetidos à administração de carprofeno). A analgesia pós-cirúrgica foi avaliada utilizando parâmetros produtivos (produção leiteira nos 8 dias após a cirurgia) e utilizando parâmetros fisiológicos (corpos cetónicos por mensuração de β-hidroxibutiratos sanguíneos no dia 1, 2 e 4). Concluiu-se que não existem diferenças os entre fármacos na analgesia pós-cirúrgica na correcção de DAE, e que para obter resultados mais fidedignos mais estudo serão necessários no futuro.

ABSTRACT - Post-surgical analgesia adjustments left-side displacement of the abomasum - The identification of animal pain is an aspect with growing importance in animal production, as well as minimize the painful processes across all production procedures. When it´s not possible the elimination of the painful stimuli, the analgesia must be prioritized. One of this painful procedures is surgery, in this case, the corrective surgery of the left displaced abomasums (LDA). It is appropriate to perform postoperative analgesia since the elimination of painful stimuli resulting from surgery cannot be done. The objective of this work was to understand if the administration of carprofen has a longer duration of analgesia than the administration of flunixin meglumine in the management of postoperative pain in surgical correction of LDA. Carprofen, as flunixin meglumine, is an anti-inflammatory non esteriod, but it’s less is used, for postoperative analgesia, because of its cost, despite having a half-life greater than flunixin meglumine.
