▼ Previous scholarship on the effect of
candidate race on voter evaluations and behavior finds that black
candidates are disadvantaged. With the election of the first
African-American president, do voters still view black candidates
as less qualified and more liberal? Prior research may not be
current for the 21st century, and despite a broad literature that
focuses on the role of race in American Politics, growing interest
in Latino politics tends to focus on Latino voters rather than
evaluations of Latino candidates. Both literatures also overlook
how voters use both race and ethnicity cues along with
candidate
partisanship. This dissertation explores the
effect of
candidate race and ethnicity on voter evaluations and
behavior. Cue Consistency Theory develops hypotheses about how
voters will react to these cues in isolation as well as how they
interact with
candidate partisanship. Original survey experiments
were conducted to inquire how
candidate race affect voter
evaluations of the candidate’s ideology, issue position, competence
and qualifications, as well as voter behavior.
Overall, the results of the non-partisan context show that
Democratic respondents inferred Democratic partisanship from black
and Latino candidates, and were particularly favorable in their
evaluations and support of these candidates. In a partisan context,
race and ethnicity does not have an additive effect, that is black
and Latino Democrats are not perceived as more liberal than an
identical white Democrat. Likewise, on most measures of ideology
and issue position, black and Latino Republicans were not perceived
as less conservative than an identical white Republican. In
general, race and ethnicity does not negatively affect perceptions
of competence and qualifications, nor does it affect turnout and
candidate support. On some measures of
respondents’ attitudes, Republican respondents were less favorable
towards black and Latino candidates. Specifically, in a
non-partisan context, Republicans found the black
candidate less
qualified, and those with cold to neutral feelings towards blacks
were less likely to support that
candidate. In the partisan
context, the Latino conservative Republican was particularly
disadvantaged; being perceived as more liberal on taxing and
spending, as well as less capable of balancing the budget and
working with leaders in business and industry.

▼ In this dissertation I examine the influence of party organizations on candidates in congressional primaries. I concentrate on the magnitude of party influence and the differential effect of parties with respect to candidate background.
Specifically, I test the degree to which candidates with non-political credentials for office (self-financers, celebrities, and members of political families) are more or less advantaged by the presence of institutionally weak parties. I also test
hypotheses related to the effect of party institutionalization on candidates, political organizations, donors, and primary voters. Parties are analyzed both as autonomous actors and as rules-bearing structures. An analysis of commonly employed measures
of party institutionalization suggests that there is no one indicator of party "strength",and raises questions about the use of surveys of executive directors and chairs of state parties. Instead, I employ a number of independent indicators of party
institutionalization. Statistical analysis suggests that party institutionalization has only a modest effect on the process of candidate emergence, with a limited ability to reduce the number of primary candidates or encourage elected officials to run
for the House. Environmental factors such as the presence of an incumbent and the partisan predisposition of the district play a dominant role. Consistent with previous work, candidates with experience in elected office are advantaged in fund-raising and
winning votes. However, party organizations are active in congressional primaries, openly supporting candidates in the primary more often than any other organizational actor. Candidates who have received party support have more success in earning
donations and votes than other candidates. Democratic and Republican primary dynamics display substantial differences. The two parties are more likely to attract candidates that resemble their party’s voter base, and there are distinctions in the
influence of candidate background and party institutionalization on fund-raising and organizational support. Strong Democratic institutions appear to advantage elected officials and long-time party activists, while stronger Republican parties advantage
candidates with elite backgrounds.

▼ The main objective of this study was to identify
genes, DNA variants and plasma metabolites associated with residual
feed intake (RFI) in beef cattle. In the first study, a total of
117 SNPs were selected and genotyped in 531 steers at the
University of Alberta. The population was split into a discovery
and validation population and multiple marker association analyses
were performed in the discovery, validation and combined
populations using ASReml. Twenty two SNPs were associated (P
< 0.05) with RFI in the discovery population and 7 (of the
22) were also significant (P < 0.05) in the validation
population. Twenty five SNPs were associated with RFI (P <
0.05) in the pooled population. A gene network analysis indicated
that the biological processes associated with the significant genes
included lipid, glucose, protein and steroid metabolism, growth,
energy utilization, and regulation of DNA transcription and
translation. The second study was an association analysis using the
117 SNPs and indicated that 7 were associated with various carcass
quality traits (p ≤ 0.005) in the same population. A third
association analysis was performed using steers at the University
of Guelph, as the discovery population, to identify blood
metabolites associated with RFI. Blood samples were collected at 3
periods with period 1, 2 and 3 corresponding to week 2, 6 and 9
into the feeding period respectively. Two, ten and three
metabolites were significantly associated with RFI (P <
0.05) in period 1, 2 and 3 and accounted for 36%, 74% and 52% of
the variation respectively. A validation analysis was performed
using steers at the University of Alberta as the validation
population. The results indicated that 3 metabolites were
significantly associated with RFI in both discovery and validation
populations accounting for 32.8% of the variation in the validation
population. A metabolic network analysis indicated that the
biological pathways associated with the metabolites included AMPK
signaling, growth hormone signaling, lipid and energy metabolism
and cholesterol metabolism. The genes, metabolites, biological
networks and the biological pathways help contribute to a better
understanding of the physiological processes influencing RFI and
carcass quality in beef cattle.

▼ In an attempt to maintain production levels during times of depressed pricing, some are exploring the practice of refracturing already hydraulically fractured wells currently within their operating portfolio. This study investigates the last two years of data of the refracturing program that was initiated by Devon Energy Corporation in the Barnett Shale play.
The purpose of this study was to identify the wells refractured in the Barnett Shale from publicly available data and completion filings. Data was collected from Drillinginfo.com, IHS.com, and FracFocus.com for these wells. Estimated ultimate recovery for these wells were generated with the aid of a reservoir engineering software program. Cost estimates were performed for each well based on comparable industry expenditures and completion methodology assumptions. All data collected and subsequent calculations were compiled into a database compatible for upload into a statistical analysis software program. Ordinary least squares regressions were performed within a statistical software program with the following objectives:
Validate some published conclusions that suggest reservoir quality indicates refracture potential;
Identify
candidate characteristics with the best internal rate of return potential; and
Investigate whether a correlational relationship exists when dummy variables are assigned to the use of chemical diverters.
The findings suggest a positive correlation between reservoir quality and refracture initial production potential. When combining the production history with this study’s completion and cost assumptions, the vertical and directional wells are ranked as more attractive refracture candidates in terms of internal rate of return. Chemical diverters showed no correlational relationship with refracture initial production values.

▼ La polyarthrite rhumatoïde (PR) et la maladie coronaire (MC) sont deux maladies touchant l'adulte comportant une susceptibilité génétique et partageant plusieurs chapitres physiopathologiques. Nous avons recherché des gènes de susceptibilité à la PR en employant une approche gène-candidat dans des loci suggerés par les données d'études de criblage du genome. Rhumatoid arthritis (RA) and coronary artery disease (CAD) are two adult diseases involving some genetic susceptibility genes and sharing many physiopathogenic chapters. We tried to find some RA susceptibility genes by a candidate-gene approach located in linked loci. We performed an original study (Genescaf) to approach some CAD susceptibility genes.

▼ Opportunistic Routing (OR) is an effective and enhanced routing scheme for wireless multihop environment. OR is an approach that selects a certain number of best forwarders (candidates) at each hop by taking the advantage of the broadcast nature of the wireless medium to reach the destination. When a set of candidates receive the packet, they coordinate with each other to figure out which one has to forward the packet toward the destination. Most of the research in this area has been done in mesh networks where nodes do not have mobility.
In this survey, we propose a new OR protocol for mobile ad hoc scenarios called as Enhanced Mobility-based Opportunistic Routing (EMOR) protocol. To deal with the node mobility, we have proposed a new metric which considers the following: geographical position of the candidates; the link delivery probability to reach them; the number of neighboring nodes of candidates; and the predicted position of nodes using the motion vector of the nodes. We have compared EMOR with five other well-known routing protocols in terms of delivery ratio, end-to-end delay, and expected number of transmissions from source to the destination. Our simulation results show that proposed protocol improves delivery ratio and number of expected transmission in terms of different type of mobility models.

▼ This thesis studies women politicians’ journey along the path from candidates to elected representatives in party list proportional representation (PR list) systems. While past literature provides sufficient evidence that more women are elected in proportional electoral systems than in majority / plurality systems, there is limited research explaining the differences in women’s representation across varying types of PR list systems. This thesis aims to fill that gap, by focusing primarily on the election of women across preferential (open and ordered list systems) and non-preferential (closed list systems) PR list voting systems. Moreover, unlike the vast majority of previous research, which has relied on aggregate level data only, this research investigates the election of women at the individual candidate level. Such an approach allows the present thesis to consider, next to traditional aggregate level predictors, how party gatekeepers and the news media may either support or hinder women in progressing from candidates to elected representatives. Since the focus is set on the 2009 European Parliament elections, this thesis investigates the process of electing women cross-nationally. The results show that female candidates have a higher likelihood of being elected in non-preferential closed list voting systems than in preferential ordered list voting systems. The results suggest that this is the case because, first, party gatekeepers in ordered list systems place women in less viable electoral list positions than party gatekeepers in closed list systems; second, media cover female candidates less in ordered list systems compared to closed list systems; and finally, female candidates in ordered list systems fail to make up their less competitive starting position with preference votes because preference votes in ordered list systems do not significantly alter the initial party list rankings.

▼ A shotgun genome sequencing project was undertaken
in the expectation that access to the entire protein coding
potential of E. Ruminantium (Welgevonden) will facilitate the
identification of vaccine
candidate genes against heartwater. The
1,516,355 bp sequence is predicted to encode 888 proteins and 41
stable RNA species. The most prominent feature is the large number
of tandemly repeated and duplicated sequences, some of continuously
variable copy number. These repeats have mediated numerous
translocation and inversion events and seem to be responsible for
the generation of both new full and partial protein coding
sequences. There are 32 predicted pseudogenes, most of which are
truncated fragments of genes associated with repeats. Of the 13
members of the order Rickettsiales compared in this study, E.
Ruminantium has the lowest coding capacity (62%), lowest GC content
(27.5%), but the highest proportion of repetitive sequences, which
comprise 8.5% of the genome. Metabolic reconstruction of E.
Ruminantium revealed the metabolic and biosynthetic capabilities
typical of an obligate intracellular organism. We identified a
number of genes unique to E. Ruminantium, most of which are not
functionally characterised in any organism, and those shared with
12 other members of the Rickettsiales. Bioinformatic tools were
used to identify possible vaccine candidates from the annotated
genome sequence. The protective properties of seven open reading
frames (ORFs), which induced cellular immune responses in vitro,
were tested in vivo Only 20% survival was obtained in sheep
immunised with a DNA formulation consisting of three ORFs. We found
that the levels of peripheral blood mononuclear cell proliferation
and interferon-gamma (IFN-γ) production did not correlate with each
other, nor with the levels of protection, suggesting that the
current assays are just not reliable and that IFN-γ expression
alone is not an indicator of protection. Therefore more cytokines
and different assays will have to be investigated to define in
detail what constitutes a protective immune response against E.
Ruminantium infection. However, the data generated from the genome
sequence will continue to facilitate novel approaches to study the
organism and to develop an efficacious vaccine against
heartwater.

▼ Personality factors are increasingly being recognised as valuable predictors of psychopathology. Neuroticism has been a focus of epidemiologically-oriented personality research, and has been associated with stress, anxiety and depression. The concurrent relationship between these phenotypes and stress invites exploration of genetic regulation of the neural stress pathway to elucidate aetiological overlap. The present study investigated the association of two genes, CRHR1 and HSD11B1, both involved in regulating cortisol functioning, with neurotic personality traits and measures of psychological wellbeing. Nominal associations were found across the traits with genetic variants, and three SNPs in HSD11B1 significantly predicted pathological anxiety levels in a population sample. These findings highlight a novel association between HSD11B1 and anxiety. Replication of this effect in a larger sample would help further elucidate the specificity of this effect to anxiety disorder.

▼ The genetic component of mammalian male fertility is complex and involves
thousands of genes. The majority of these genes are distributed on autosomes and the X
chromosome, while a small number are located on the Y chromosome. Human and
mouse studies demonstrate that the most critical Y-linked male fertility genes are present
in multiple copies, show testis-specific expression and are different between species.
In the equine industry, where stallions are selected according to pedigrees and
athletic abilities but not for reproductive performance, reduced fertility of many breeding
stallions is a recognized problem. Therefore, the aim of the present research was to
acquire comprehensive information about the organization of the horse Y chromosome
(ECAY), identify Y-linked genes and investigate potential
candidate genes regulating
stallion fertility.
To achieve theses goals, a direct cDNA (complementary DNA) selection
procedure was used to isolate Y-linked genes from horse testes and 29 Y-specific genes
were identified. All 29 genes were mapped to ECAY and their sequences were used to further expand the existing map. Copy number analysis identified 15 multicopy genes of
which 9 were novel transcripts. Gene expression analysis on a panel of selected body
tissues showed that some ECAY genes are expressed exclusively in testes while others
show ubiquitous or intermediate expression. Quantitative Real-Time PCR using primers
for 9 testis-specific multicopy genes revealed 5 genes with statistically significant
differential expression in testis of normal fertile stallions and stallions with impaired
fertility. Gene copy number analysis showed that the average copy number of 4 such
genes was decreased in subfertile/infertile stallions compared to normal animals.
Taken together, this research generated the first comprehensive physical gene
map for the horse Y chromosome and identified a number of
candidate genes for stallion
fertility. The findings essentially expand our knowledge about Y chromosome genes in
horses, open a new avenue for investigating the potential role of ECAY genes in stallion
fertility which contribute to the development of molecular tools for the assessment of
fertility in stallions.

▼ This project focuses on political candidates’ image and issue information, informed by the fields of communication, political science, and psychology. Recognizing the field’s heavy use of cross-sectional data, I conducted an experiment to test the effects of manipulated trait-issue combinations on overall
candidate evaluation. Stimuli were presented in the form of political campaign tweets to maximize external validity. Results indicate that pairing a trait from one party and an issue from the other party may be more effective than simply engaging in trait or issue ownership. They also find that appeals to qualification-based traits may require evidence to be effective, while appeals to personality-based traits may be enough on their own. By utilizing a novel approach to test effects of character and issue information, this study illuminated limitations of this approach as well as several directions forward for future research.

▼ Recent global flu pandemic highlights the obvious need for a vaccine against the influenza virus that is both inexpensive to produce and can be produced fast enough for development on a large scale. The influenza virus coat protein hemagglutinin (HA) is a major surface antigen and has emerged as a good candidate antigen for subunit vaccine development. Previous attempts to express HA from the A/Sichuan/2/87 H3N2 influenza subtype in plastids achieved HA gene transcription but no protein accumulation, suggesting that the HA protein may be unstable in the plastid. The aim of this work is to investigate alternative strategies that might achieve HA antigen accumulation in plastids. Two series of gene constructs were made. The first series of constructs coded for the first 14 amino acids of GFP and RBCL N-terminally fused to the full length HA, a full length GFP N-terminally fused to HA and a full length GFP N-terminally fused to HA1. The second series of gene constructs coded for two candidate influenza virus hemagglutinin epitopes, HA91-108 and HA 307-319 fused singly as N and C terminal fusions and as double terminal fusions to full length GFP. A total of ten gene constructs were introduced into the chloroplast genome by biolistic bombardment. Transformants were generated and confirmed through molecular analyses. Transplastomic lines generated with the 14aaRBCL/HA and 14aaGFP/HA constructs accumulated a recombinant protein that was smaller than expected. Further analysis is required to confirm that these proteins are indeed truncated HA fusion proteins. No transformed plant lines were generated with the full length GFP/HA and GFP/HA1 constructs suggesting that the tobacco plastid is not a useful system for producing full-length flu virus HA and HA1 proteins suitable for vaccine development. Expression and stability of
recombinant HA91-108/GFP and GFP/HA307-319 fusion proteins was clearly demonstrated showing that conserved influenza virus HA epitopes can be expressed in plant chloroplasts. ▼ This study applies the frameworks of Krathwohl, Bloom,
and Masia (1964), Smith and Ragan (2004), and one school of
education’s Conceptual Framework as a means to develop a measure of
teacher
candidate dispositions towards diversity on the Performance
Assessment for California Teachers (PACT). The purpose of this
study was to determine the extent to which the PACT contains
evidence to assess valuing diversity, using a theoretically
grounded original rubric designed based upon program-identified
candidate proficiencies, and the PACT’s developmental perspective.
Using secondary data from Secondary Math (N=10), Secondary Science
(N=11) and Elementary Math (N=19) submissions, this mixed methods
study collected and analyzed assessor comments and feedback in
addition to numerical scores comparing
candidate performance on
existing PACT rubrics with the new Valuing Diversity rubric. The
study revealed that the rubric was a useful valid instrument for
identifying valuing diversity in the PACT. While assessors found
evidence to assign a score based on the elements of the rubric,
they also found that the candidates scored lower on the Valuing
Diversity rubric than on the related rubrics that were also
analyzed. Programmatic recommendations including a curriculum
audit, and targeted professional development, as well as
recommended future research are presented. Although further
research is necessary, this study has contributed to the empirical
research on assessing dispositions. The combination of learning
theory, an affective taxonomy, and expected
candidate proficiencies
provided a lens with which to examine candidates attitudes, values,
and beliefs as they influenced the application of knowledge and
skills in their guided practice classrooms.

▼ Recent global flu pandemic highlights the obvious need for a vaccine against the influenza virus that is both inexpensive to produce and can be produced fast enough for development on a large scale. The influenza virus coat protein hemagglutinin (HA) is a major surface antigen and has emerged as a good candidate antigen for subunit vaccine development. Previous attempts to express HA from the A/Sichuan/2/87 H3N2 influenza subtype in plastids achieved HA gene transcription but no protein accumulation, suggesting that the HA protein may be unstable in the plastid. The aim of this work is to investigate alternative strategies that might achieve HA antigen accumulation in plastids. Two series of gene constructs were made. The first series of constructs coded for the first 14 amino acids of GFP and RBCL N-terminally fused to the full length HA, a full length GFP N-terminally fused to HA and a full length GFP N-terminally fused to HA1. The second series of gene constructs coded for two candidate influenza virus hemagglutinin epitopes, HA91-108 and HA 307-319 fused singly as N and C terminal fusions and as double terminal fusions to full length GFP. A total of ten gene constructs were introduced into the chloroplast genome by biolistic bombardment. Transformants were generated and confirmed through molecular analyses. Transplastomic lines generated with the 14aaRBCL/HA and 14aaGFP/HA constructs accumulated a recombinant protein that was smaller than expected. Further analysis is required to confirm that these proteins are indeed truncated HA fusion proteins. No transformed plant lines were generated with the full length GFP/HA and GFP/HA1 constructs suggesting that the tobacco plastid is not a useful system for producing full-length flu virus HA and HA1 proteins suitable for vaccine development. Expression and stability of
recombinant HA91-108/GFP and GFP/HA307-319 fusion proteins was clearly demonstrated showing that conserved influenza virus HA epitopes can be expressed in plant chloroplasts. No transgene transcripts could be detected in transplastomic lines generated with the GFP/HA91-108, HA307-319/GFP or HA307-319/GFP/HA91-108 gene constructs. No transformed lines were generated with the HA91-108/GFP/HA307-319 construct. ▼ Scholars of American politics have long observed that we live in an era of "candidate-centered" politics. Since around the 1960s, it seems as though candidates have relied less on parties and party organizations to conduct their campaigns
and have increasingly built and utilized their own campaign organizations, turning the parties primarily for fundraising purposes. While many point to the electoral reforms of the Progressive Era (e.g., the Australian ballot and the direct primary) as
being the catalyst for the decline of parties, those reforms were largely adopted by the states by the 1910s. Thus, there remains a significant lag between the adoption of the supposed cause (Progressive reforms) and the effect (candidate-centered
politics). With this project, I seek to explain the rise of candidate-centered politics in U.S. House elections as a function of institutional structures, economic development, and technological innovation. Specifically, the format of ballot used in a
given district is found not only to have direct effects on the degree of candidate centeredness, but the format also structures the effects of other variables. Rising incomes and the related rise in campaign spending also appear to have driven candidate
centeredness, though these effects are largely limited to those districts using one particular ballot format. Finally, television is found not to be the great liberator of candidates some scholars have argued; rather, its effects are limited and
indirect. I demonstrate that elections in the U.S. evolved in the context of an electoral environment that was changing on numerous fronts.

▼ Ketosis is a severe metabolic disease caused by negative energy balance (NEB) that leads to decreased milk production. Thus, the main objectives were to investigate literature information on the genetic background of NEB, subclinical and clinical ketosis through a systematic review and meta-analysis and to identify genomic regions associated with subclinical and clinical ketosis in first and later lactations, and also for subclinical ketosis in two periods in early lactation (5 to 21 DIM and 22 to 45 DIM) by performing genome wide association studies (GWAS). The weighted single-step GBLUP (wssGBLUP) method was used for GWAS to identify genetic regions associated with the traits. The genes identified within the top-20 genomic windows were used for functional analyses. Biological understanding of this metabolic disease will help enhance accuracy of selection in the Canadian dairy cattle breeding programs and help with the development of biomarkers for early diagnosis and prevention of ketosis.

▼ Research into persistence and attrition of graduate
students points to a variety of institutional barriers, individual
characteristics, and personal circumstances that contribute to the
decision to complete a program or abandon graduate study.
Specifically, what is not known is how students who spend a great
deal of time in the dissertation phase marshal their resources to
successfully complete the degree. This study was intended to help
understand the lived experiences of successful doctoral candidates
and describe how doctoral candidates who lingered for an extended
period in the dissertation phase of their degree program
successfully complete their candidacy. The purpose of this study
was to examine and describe the lived experiences of three doctoral
candidates who earned PhD degrees from Colleges of Education at
Midwestern research universities and spent five or more years to
successfully complete their dissertations. Of specific interest was
how the doctoral candidates made sense of the experiences and made
progress to complete their dissertations. This study sought to
answer the question: How do long-time-to-doctorate (LTTD)
candidates describe the experiences associated with an extended,
successful dissertation process?
The Interpretative
Phenomenological Analysis (IPA) methodological framework of this
study guided sampling and participant selection, the research site,
data collection methods, interviews, data management, and data
analysis. Three LTTD candidates were interviewed and provided their
experiences from the time of successfully completing their
preliminary examinations to the time they deposited their
dissertations. Additional sources of data included a timeline of
the dissertation process completed by each participant and the
acknowledgements section of two of the participant’s deposited
dissertations. These three sources of data provided rich and unique
perspectives with similarity in experiences across the
participants’ narratives. IPA methods were employed to analyze data
including pre-analysis, initial notation, development of emerging
themes, identifying gaps, and identifying connections across
themes. Although they assumed they were well-prepared for a PhD
program, the participants described being surprised by how they
struggled throughout their dissertation processes, and in some
cases getting stuck in liminality. These times of stuckness and the
depth of the liminality they experienced added time to their
completion process, in part contributing to their long time to
doctoral degree. The participants described strategies and supports
they employed to survive the dissertation journey, and in some
cases suggested that they wished they had used those strategies
sooner. Finally, all of the participants detailed how important
completion of the PhD program was to their personal and
professional identities, and the way they made meaning about their
experiences with faculty involved in the degree completion and
identity formation process involved with becoming doctorate.

The record-setting number of women on the ballot translated into a new high-water mark for the descriptive representation of women in Congress. While it is clear that female candidates were more successful in getting on the ballot during this election cycle than ever before, what remains unclear is how voters perceived the diverse traits these candidates brought to their campaigns and how those traits impacted voter evaluations. In this dissertation, I build on the existing literature on gender stereotypes and white female candidates to explore this question.

▼ While much attention has been paid to newspaper endorsements and their effects on vote choice, little attention has been paid to the endorsement process and the factors that editorial boards use when assessing candidates. This study develops
a very basic theory and tests hypotheses that suggest editorial boards consider a number of candidate level factors during the endorsement process. Editorial behavior in the 2008 Republican and Democratic presidential nomination campaigns was the focus
of this examination. Logistic regression analysis of the 2008 data suggests that both Democratic and Republican candidates trailing in national polls are less likely to accrue editorial support. For both models, ideological congruence between candidate
and newspaper is the best explanatory variable for editorial choice. The more a candidate and a newspaper board agree on salient issues, the more likely that candidate will receive the newspaper’s endorsement. These findings suggest editorial choice is
based on certain identifiable factors relating to the support levels and issue stances of the candidate. In other words, newspaper endorsements are generally quite predictable events.

Language as an Identification Resource in Secondary English Teacher Preparation:
An Analysis of Discourses

The topic of the research presented here was teacher educators’ and teacher candidates’ talk as an identification resource in the coursework of an undergraduate and Master’s level secondary English teacher preparation program. Two research questions framed this study: What identity constructions of English teacher are available in the discourses of secondary English teacher preparation? How are the discourses and available identifications transformed through language use in course meetings? Participants in the study were two English teacher educators, twenty teacher candidates enrolled in two secondary English methods courses at a mid-sized, private, urban university, and five teacher candidates who agreed to participate in interviews and one focus group. Qualitative methods for data collection and analysis were used including semi-structured interviews, classroom observations with video- and audio-recording, constant comparative analysis, and discourse analysis. Findings were that participants’ talk was connected to prominent Discourses in the fields of English education and teacher education. Talk in the courses created specific identification opportunities for teacher candidates. Teacher candidates accepted, rejected, and transformed the available identifications through specific language use and genres of talk. Findings from this study have implications for structures and practices in teacher education and contribute to theory building of how teacher candidates become teachers who identify, or not, with particular conceptions of English teacher.

▼ Tutkielman teoriaan pohjautuvana kantavana oletuksena on, että poliittisesta luottamustoimesta saatava palkkio on aina yhteiskunnan valinta. Luottamustoimen arvostus määrittää sen tavoiteltavuuden ja vaikuttaa polkuriippuvuuden kautta tulevien päätöksentekijöiden pätevyyteen. Tutkielman teoreettinen osuus pohjautuu pääosin Poutvaaran ja Takalon (2007) ja Messnerin ja Polbornin (2004) citizen-candidate- malleihin, joista ensimmäinen käsittelee vaaleja suurten (luku kolme), ja jälkimmäinen pienten vaalien näkökulmasta (luku neljä). Luvun kolme mallissa poliitikkojen saamat palkkiot ja kampanjointikustannukset ovat suuria. Luvussa neljä tilanne on päinvastoin, koska kansalaisten oletetaan tuntevan ehdokkaat. Luvun viisi empiirisessä osuudessa mallien tuloksia ja rakenteita on sovellettu kyselytutkimukseen (aineisto n=385), joka tehtiin Suomen ylioppilaskunnissa toimiville opiskelijapoliitikoille ja ylioppilaskunta-aktiiveille kevätlukukaudella 2009. Kyselyn tutkittavana perusjoukkona ovat Suomen ylioppilaskunnissa toimivat opiskelijapoliitikot, joiden ehdokkuuspäätökseen vaikuttavia tekijöitä (palkkioita, vaihtoehtoiskustannuksia ja kampanjointikustannuksia opiskelijapolitiikasta) sekä suhtautumista (ammatti)poliitikkoihin tarkastellaan lukujen 2-4 teorian oletusten suhteen. Teorian ja empiirisen aineiston yhteensovittaminen osoittautuu hyvin haastavaksi. Luvussa viisi havaitaan, että ehdokkuuspäätökseen vaikuttavat tekijät ovat moninaiset ja vaihtelevat etenkin opiskelija-aloittain, ja että näitä tekijöitä tulee tarkastella pitkällä aikavälillä. Yksittäisistä ehdokkuuspäätökseen vaikuttavista tekijöistä riskillä yksilön opiskeluajan pidentymisestä on ehdokkuuspäätöksen suhteen selvästi vaihtoehtoiskustannuksia lisäävä vaikutus. Opiskelija-aloittain vaihtoehtoiskustannukset ehdokkuudesta ovat tulosten mukaan pääsääntöisesti suurimmat etenkin lääketieteen, farmasian, eläinlääketieteen ja matemaattis-luonnontieteen opiskelijoilla. Toisaalta myös ehdokkuuspäätökseen positiivisesti vaikuttavat tekijät eli siitä saatavat taloudelliset, sosiaaliset ja psykologiset palkkiot ja oman luottamustoimen arvostus, ja luottamustoimen tuottamat positiiviset ulkoisvaikutukset ovat näissä ryhmissä pienimmät. Siksi myös näiden opiskelija-alojen joukossa on keskimääräistä huomattavasti vähemmän hallitsevia opiskelijapolitiikkoja. Yleisesti ottaen suurimpia hyötyjä opiskelijat kokevat saavansa ylioppilaskunnassa ja opiskelijapolitiikasta toimimisesta erityisesti sosiaalisen pääoman ja suhdeverkoston laajentumisen suhteen, joista koetaan olevan hyötyjä myös työelämän suhteen. Opiskelija-aloista yhteiskuntatieteiden (paitsi kansantalous ja taloustiede) ja osa humanististen alojen opiskelijoista kokee saavansa muita enemmän hyötyjä opiskelijapolitiikasta. ▼ Antarctic desert soil ecosystems are predominately comprised of prokaryotes, which have developed unique ecological functions to cope with the extreme environmental conditions experienced in Antarctica. While Antarctic soils have exhibited diverse microbial community structures, including the presence of rare bacterial lineages, the ecological functions or genomic capacities of the microbial dark matter in this environment has remained largely unexplored.Mitchell Peninsula and Robinson Ridge are polar desert sites located in the Windmill Islands, East Antarctica, which are very low in carbon and nitrogen. Here, PCR amplicon 454 pyrosequencing targeting the bacterial SSU rRNA genes revealed both sites to encompass a microbial community “hotspot” comprised of a high relative abundance of
candidate phyla WPS-2 (9.3%) and AD3 (5.1%), as well as uncultured Chloroflexi and Actinobacteria. In addition, the abundance of Cyanobacteria, the primary carbon and nitrogen fixer in many environments, including Antarctica, was extremely low (average 0.35%).Shotgun metagenomics and differential coverage binning was used to recover 23 draft genomes from Robinson Ridge, including for the first time, two
candidate division WPS-2 and three AD3 draft genomes. While Cyanobacteria abundance was confirmed to be low, metagenomic analysis revealed that 45% of the draft genomes recovered were carrying a novel type IE RuBisCO, indicative of carbon fixation. With no bacterial chlorophyll or rhodopsin identified, a dark carbon fixation process reliant on the oxidation of atmospheric H2 and CO was discovered. This process occurs through the use of the novel 1E RuBisCO, as well as specialised high affinity type 1h/5 [NiFe]-hydrogenases and carbon monoxide dehydrogenases, all of which were also widely distributed in the Robinson Ridge metagenome. In contrast to the major carbon acquisition pathway identified, no nitrogen fixation genes were present, yet denitrification capacity was widely detected in the draft genomes. As denitrification would lead to the loss of nitrogen from the ecosystem, the balance of nitrogen in Windmill Islands region needs further investigation.An inconsistency in the classification of WPS-2 was identified across the major SSU rRNA gene databases. WPS-2 in Greengenes was comprised of two bacterial phyla: WPS-2 and SHA-109, while WPS-2 in RDP was actually sequences from Planctomycetes. Phylogenetic analysis revealed WPS-2 to be comprised of two sub-clusters that has a close relationship to phylum Chloroflexi. I propose these sub-clusters to be composed of either facultative autotrophs (sub-cluster I) or obligative heterotrophs (sub-cluster II). Genomic inference and fluorescence in situ hybridisation showed in Antarctic soils, WPS-2 bacteria exist as small diderm cocci, with diameters ranging between 0.6-1.2 m.Here, I propose that the microbial community in the extremely carbon-limited soils of Mitchell Peninsula and Robinson Ridge have developed a unique dark carbon fixation process based on the use of a highly dependable…

Election to provide opportunities and pipelines for political participation, related to the development of political system and the implementation of democratic politics, the regime is peaceful, orderly transfer of tools, is also an important democratic process, which is a democratic country to maintain political legitimacy of the main Methods, but also non-democratic countries in transition towards a democratic system of government an important mechanism, and voting is the most popular political participation of citizens, the most common, is also close to the general public most likely a form of participation.
The study use the visit information of Taiwan elections and democratization investigation in 2006 that relating to Kaohsiung Mayor election survey data, in order to investigate factors influencing the choice of the voters from the social psychology study ways to survey the Michigan Model in party Identification,
candidate evaluation, campaign issues and other variables in the voting model of the importance. Finally, we attempt to identify influencing the choice of Kaohsiung people to vote deciding factor.
This study showed that, the five social background of voters (including the gender, age, ethnic group, educational level, families income) and party Identification,
candidate evaluation, campaign issues, ethnic identity, these five variables and four intermediate variables were significantly associated. Particularly, the ethnic identity is a very important effect and these four intermediary variables also have very significant influence for choice of the voters voting.
Polynomial logistic regression was used to establish the simple and complete model in order to check variables and intermediary variables between the relations of voter choice. In simple model was found that party Identification, the five
candidate evaluation and the three campaign issues affect the voters choice definitely, especially the party Identification, the relatively reliable in the
candidate evaluation and the remedial for the Love River, the completion of Singuang Ferry Wharf in the campaign issues, but to understand that ethnic identity with regard to the voters intention was not significant affects. In complete model showed that the nine variables (including the age, ethnic group, educational level, families income and the party identification, more ability to work, to understand that the people demand, relatively reliable, the most enthusiastic service to the people, remedial for the Love River, the completion of Singuang Ferry Wharf and the incident of walking fees) affect the voting choice of the voters. In particular, age, party identification, have more ability to work, relatively reliable, remedial for the Love River, the completion of Singuang Ferry Wharf were the most important explanatory variables and have significant relevance to the election with the support of the mayor candidates.

▼ This research explores whether âparty identification,
candidate orientation, ethnic identification and issue orientation,â the four intermediate variables of election, are correlated to the results of Taipei mayor elections in 2002 and 2006. The data in this research are mainly based on the interview broadly carried out to collect facts for âthe study on democratization through electionâ executed by Professors Huang Chi and Huang Siu-Twen in 2002 and 2005. It is with these data that this research compares the voting behavior of the voters in the two mayor elections in an attempt to find out the deviation of election for further studies.
Each individual voter has of his own a variety of social backgrounds which inevitably form to some extent a connection with each of the four intermediate variables. In examination of the connection just mentioned, it is obvious that of all the voterâs backgrounds the place of birth is probably the most potential and influential, and has a significant connection with each of the four factors. However, an evaluation of the connection between the voting preference and the four factors indicates that the four factors are apparently convincing in justification of the results of the second Taipei mayor election in 2006. In addition, a detailed analysis of the connections between voterâs social backgrounds and his voting preference shows that the voterâs place of birth plays an important role in making his voting preference. Other social backgrounds except the voterâs educational level are slightly related to the voting preference in the Taipei mayor election in 2006.
Based on the analysis mentioned above, it is understood that there are many variables which will influence the voting preference. Of all the intermediate variables, however, the four factors of party identification
candidate orientation ethnic identification and issue orientation are perhaps most crucial and decisive. By means of the analysis and evaluation of the four factors functioning in the recent elections, it is discovered that the people here in Taiwan are relatively better educated and have made a tremendous progress in democracy. It seems that Taiwan has already developed into a nation of two-party politics.

▼ In recent years, negative campaigning has been found in most Taiwanâs elections, which almost became one of the corrupt customs that are deeply ingrained in electoral culture. Although candidates who are defamed usually sternly deny that or strongly rebut to rumors by clarifying the facts, they still may be overwhelmed by the malicious attacks, even eventually went into mudsling. It doesnât only encourage low quality elections, but also bring dishonor to the spirit and value of democracy. This study proposes strategies of humorous rebuttal and applies it to advertising as a countermeasure to negative image advertising with an aim to protect the images of candidates and bury the hatchet by the power of witty utterance. Specifically, the purpose of this study is to compare the advertising effects among different types of rebuttal advertising and examine the moderation of the gender of responding
candidate and different types of negative advertising.
A 2 (types of negative advertising: mocking attack vs. linking attack)x 4 (types of rebuttal advertising: traditional rebuttal-positive response vs. traditional rebuttal -negative response vs. humorous rebuttal- self-deprecation vs. humorous rebuttal- other-deprecation) x 2 (responding
candidate gender: male vs. female) between-subjects design experiment was conducted to test seven hypotheses and one research question. The results indicate that (1) using humorous rebuttal leads to better advertising attitudes and better votersâ responses toward the responding candidates, while using traditional rebuttal can undermine opponentsâ images and votersâ attitudes toward the opponents more effectively; (2) male (vs. female) candidates can strengthen the positive effectiveness of humorous rebuttal (vs. traditional rebuttal) on advertising attitudes and responses toward the responding candidates, while by using traditional rebuttal, only female candidates can effectively injury opponentsâ images and make voters have poor responses to them; (3) in terms of sub-strategies of traditional rebuttal, female (vs. male) candidates can strengthen the positive effectiveness of positive (vs. negative) response on advertising attitudes and votersâ responses toward the responding candidates; (4) in terms of sub-strategies of humor rebuttal, the effectiveness of humorous self-deprecation and other-deprecation advertising used by male candidates are near the same, but it is more proper for female candidates to use self-deprecation other than other-deprecation advertisements; and (5) mocking attack (vs. linking attack) will strengthen the positive effectiveness of humorous rebuttal (vs. traditional rebuttal) used by candidates and weaken the positive effectiveness of positive (vs. negative ) response on advertising attitudes and votersâ responses toward the responding candidates, while in the case of linking attack, opponentsâ image and votersâ attitudes toward them can only be injured by using traditional rebuttal.

▼ This thesis attempts to study the subjective recognition of candidates in the mind of voters for whom the images of the candidates come up to a vital dimension in their decision making. In the 2014 election of Penghu magistrate, senior voters cast their ballets according to their identification with political parties. Senior citizens with preference for political parties tend to stay with their political inclination. While most voters place priority in personal performance rather than party recommendation, the devastation of performance both at the level of local and central government becomes the decisive factor in ballet casting.
I intend to conduct this study to clarify the relation between individual
candidate images and the decision in vote casting out of 432 valid questionnaires on eligible citizens household registering in Penghu. My discussion of
candidate images include identification with political parties, personal qualities, positions in public issues and their retrospective evaluation. I intend to apply descriptive statistics, Chi-square Test, and analysis of variance to examine the social background of the voters and the association with their decision in ballet casting, in order to scrutinise the key factors of their victories in elections.
My study indicates that all variance of the image of the candidates are all key factors in votersâ decision making, hence all four factors in discussion behold high positive correlations. All in all, those who are above 60, with education below junior high, with distinct preference to either pan-blue or pan-green camp, with stronger ties with political parties, are inclined to electing candidates nominated by their parties. Voters with conspicuous preference to parties tend to acclaim personal qualities of their advocates while male and pan-green electors are liable to maintain lower retrospective evaluation. Voters with explicit preference are prone to approval of
candidate images. Thatâs also where my study reveals greater differences in the retrospective evaluation. The winning
candidate, Kuang-fu Chen (DPP) prevails in most categories of comparison while Kun-shung Su (KMT) shows no less competition in these items. The decisive factor is attributed to the impotence of governmental performance at centre and local level and the overall sentiment of disappointment in the general public.
To conclude, most voters are prone to decisions over personal qualities instead of party nomination in this election of magistrate. Such tendency manifests votersâ choice over stagnant and restricted preference to political parties and suggests a breakthrough in rising above bipolar confrontations thus forecasts a rational perspective in their objective speculations.

▼ Botulinum neurotoxins (BoNTs) produced by
Clostridium botulinum are the causative agents of botulism and
represents a family of seven structurally similar but antigenically
different serotypes (A to G). The BoNTs are expressed in C.
botulinum as a single polypeptide chain and then
posttranslationally nicked, forming a di-chain polypeptide chain
consisting of a 100-kDa heavy chain and a 50-kDa light chain held
together by a disulfide bond. Topologically, the neurotoxins are
composed of three domains, a binding domain (HC), a translocation
domain (HN) and a catalytic domain. The BoNTs act preferentially on
cholinergic nerve endings in both humans and animals and thus
produce a flaccid paralysis that may result in death. In southern
Africa, BoNT types C and D have been associated with botulism in
cattle. To combat the disease, a bivalent vaccine consisting of
formalin-inactivated type C and D holotoxins is currently
available, and although it is efficacious, several concerns
regarding its production has been raised, most notably its cost.
The development of efficacious recombinant subunit vaccines may
provide a means whereby many of the production problems may be
eliminated or minimized. Consequently, the aim of this
investigation was to produce a recombinant botulinum neurotoxin
serotype D binding domain [BoNT/D(HC)] vaccine
candidate for
preventing BoNT/D intoxication. Towards this end, the gene fragment
for the heavy chain (HC) of the BoNT produced by the C. botulinum
type D vaccine strain D-50 was amplified, cloned in Escherichia
coli and characterized by nucleotide sequence analyses. An
alignment of the deduced amino acid sequence with that of
characterized clostridial type C and D neurotoxins demonstrated
that the heavy chains are composed of highly conserved domains
interceded with tracts of amino acids exhibiting little overall
relatedness, although considerable identity between the components
ofa specific pair is apparent in certain of the regions. The
deduced amino acid sequence exhibited 99, 66 and 73% identity with
the reported amino acid sequences of BoNT/D-SA, BoNT/D and BoNT/C1,
respectively. Attempts at expressing the native gene sequence for
the HC from BoNT/D-50 in Brevibacillus brevis 47-5Q were
unsuccessful. This may have been due to differences in codon bias
between the heterologous gene and B. brevis. Consequently, a
completely synthetic codonoptimized gene encoding the HC of
BoNT/D-SA was constructed and expressed using a B. brevis 47-5Q
mutant as expression host, obtained on mutagenesis with
N-methyl-N’-nitro-Nnitrosoguanidine (NTG). Extracellular expression
of the 48-kDa recombinant protein was verified by Western blot
analyses with anti-BoNT/D antibodies. The recombinant BoNT/DSA(HC)
protein was purified from the culture supernatant and used to
vaccinate mice, after which their survival against challenge with
active toxin was evaluated. Mice given two subcutaneous
vaccinations were protected against intraperitoneal administration
of 4 X 102 mouse lethal dosages (MLDs) of 16S BoNT/D-50 toxin.…
