▼ This thesis is primarily concerned with the theory that the writings of Seneca the Younger display an array of stylistic choices theretofore unprecedented in the production of philosophical works in Latin, as well as that, in so doing, Seneca is able to cultivate an approach to Latin literature that is uniquely Roman in character. By using two of the “dialogues” of Seneca—De otio and De breuitate uitae—as representative of his prose works, particularly those philosophical in nature, I analyze the author’s specific use of language in order to highlight and to detail those methods which he employs in an effort to appeal to singularly Roman sensibilities as opposed to the cultural menagerie Imperial Rome had become. Through careful philological investigation dedicated to understanding the source of contention between certain concepts and Senecan ideology, I come to conclude that Seneca’s approach to writing fits perfectly not only the Stoic platform to which he is primarily devoted, but also the sociopolitical climate of his day as he diligently attempts to communicate with his Roman audience on a level that they can truly comprehend.

Melatonin is an important modulator of several physiological and behavioural processes, and it influences the function of many different tissues. Melatonin has effective antioxidative properties, but some of its actions in mammals are also mediated through the MT1 and MT2 melatonin receptors. Antioxidative properties are seen especially when the melatonin concentration is high (≥ nM), and melatonin's affinity for its receptors appears at lower concentrations (pM).
Recently, the involvement of melatonin in protecting the heart against cardiac diseases, including myocardial infarction (MI), has been brought out. MI alters the structure and function of myocardium, attenuating for example cardiac contractility by affecting the amount and function of the essential Ca2+ handling proteins, dihydropyridine receptor (DHPR), ryanodine receptor (RyR2) and sarco-endoplasmic reticulum (SR) Ca2+-ATPase2 (SERCA2). MI also evokes many adaptive responses in organisms, such as elevated production of atrial and brain natriuretic peptides (ANP and BNP).
In this thesis, the expression of MT1 and MT2 receptor mRNAs was investigated in several rat tissues. Furthermore, the effect of MI and exogenous melatonin on the rat endogenous melatonin and on the expression of cardiac MT1, MT2, DHPR, RyR2 and SERCA2 proteins was evaluated. The concentrations of ANP and BNP were also measured after post-MI melatonin administration.
The results show the expression of MT1 and/or MT2 receptor mRNAs in the hypothalamus, retina, small intestine, liver and heart, which indicates that at least some effects of melatonin could be mediated through the receptors in these tissues. Melatonin synthesis in the pineal gland increased rapidly in response to MI, supporting an important role of endogenous melatonin in protecting the heart after MI. Furthermore, exogenous melatonin altered the mRNA expression of DHPR, RyR2 and SERCA2 after MI, suggesting that melatonin might contribute to the post-infarction cardiac contractile function. The results also revealed a novel, positive relationship between melatonin and ANP, and thereby bring out one more possible way of melatonin to protect the heart against MI-induced injuries.
Taken together, the present thesis (i) supports the notion that melatonin is an important endogenous protective agent of the organism, and (ii) extends our knowledge of melatonin's post-infarction cardioprotective actions.

Melatoniini on tehokas antioksidantti, mutta jotkut sen vaikutuksista välittyvät myös MT1 ja MT2 melatoniini reseptorien kautta. Antioksidatiiviset vaikutukset tulevat esiin erityisesti silloin, kun melatoniinin pitoisuus on korkea (≥ nM). Alhaisemmilla pitoisuuksilla (pM) on puolestaan havaittavissa melatoniinin sitoutuminen reseptoreihinsa.

▼ Osteosarcoma is the most predominant primary bone cancer among pediatric patients. The pathogenesis and mechanism of osteosarcoma, however, as yet remains unclear. This study investigated the hypothesis that Ca2+/calmodulin-dependent protein kinase II (CaMK II), activated by Ca2+ and its intracellular receptor, calmodulin, plays a critical role in the growth of human osteosarcoma cells. The inhibition of CaMK II with its pharmacologic antagonist KN-93 was used to investigate the role of CaMK II in the growth of human osteosarcoma cells because our preliminary data showed that =-CaMK II is expressed in several human osteosarcoma cell lines. To examine the in vivo effects of KN-93 on human osteosarcoma cells, MG-63 cells and Saos-2 cells were subcutaneously inoculated in 5-week-old male athymic nude mice. The mice were then treated with either KN-93 or phosphate buffered saline every other day for 6 weeks. The tumor cells were also inoculated in the tibia of the mice at the same time when mice received subcutaneous inoculation. The final total volume of subcutaneous tumors of KN-93-treated group was 40% smaller in MG-63 cells and 41 % smaller in Saos-2 cells than that of each control group at sacrifice. Supporting these findings, micro-computerized tomography of mouse tibia where MG-63 cells were grown provided another line of evidence that pathologic bone formation by osteosarcoma cells was decreased by KN-93 treatment. Further, immunohistochemistry staining on these samples demonstrated that the expression of p21 was significantly increased while that of p-Rb was decreased in the KN-93-treated group. These results suggest that p21-involving signaling cascade mediated by CaMK II may be one of the potential mechanisms controlling the growth of human osteosarcoma cells, which will warrant further investigations aiming at a novel target to treat human osteosarcomas.

▼ In our present day, the term “chivalry” is understood to define the gentlemanly
conduct of a man with respects to a female counterpart. In the fifteenth century, the term
“chivalry” is applied in a broader sense, referring to the inter-dynamics of a grouping of
knights. “Chivalry” thus examines the bachelorhood of the knight and the magnanimous
and egalitarian nature of his position.
Italians have been aware of Arthurian legends since at least the eleventh century.
In fifteenth century Northern Italy, chivalric literature enjoyed refreshed interest due to
an outpouring of Italian and French translations.
Borso d’Este (1413-1471), although illegitimate, was the second sovereign son of
Niccolò III d’Este of Ferrara. Borso’s illegitimacy and lifestyle made it advantageous for
him to embody a knightly persona. In doing so, he employed a variety of artistic campaigns that hyperbolized his pedigree. This thesis focuses on one of these campaigns, the
Hall of the Months, in the Palazzo Schifanoia, an Este hunting retreat on the outskirts of
Ferrara.
Francesco del Cossa and Cosmè Tura painted the Hall of the Months. The program for the fresco cycle is complex. The twelve calendar months run clockwise around
the room. Horizontally, the hall is divided into three registers, from bottom to top: earth,
corresponding zodiacal signs, and corresponding deities. Considering the 1:1 scaling and expansive proportions of the Hall of the Months
(24 m long, 11 m long, and 7.5 m high), the earthly tier illusionistically places viewers
outdoors and serve as a reminder of Borso’s place among sovereigns. Themes of civic
harmony abated any fear of discord or aggression.
Previous scholarship has tended to focus on the astrological aspects of the Hall of
the Months. However, its chivalric imagery has either been neglected or not fully explored. Thus, this thesis focuses only on the “earthly” register, whose chivalric imagery
promulgates Borso’s carefully crafted persona. By providing a new focus of interest for
the Hall of the Months, I hope to open the field to further research about Borso’s biography as well as the importance of chivalric literature in fifteenth-century Italy and in fifteenth-century Italian art.

▼ In the Kress Gallery of the Birmingham Museum of Art is a painted panel of the
Journey of the Queen of Sheba, created by the Florentine workshop of Apollonio di
Giovanni and Marco del Buono in the mid-fifteenth century. Its companion panel, The
Meeting of Solomon and the Queen of Sheba, is housed in the collection of the Museum
of Fine Arts, Boston. Together, these two artworks once formed the fronts of a pair of
elegant marriage chests, often referred to as cassoni.
Although these panels have been reproduced and briefly discussed in several
publications, they have never been given a thorough analysis. My thesis project fills this
gap with an in-depth study of these two panels, replacing them as much as possible into
their historic context in order to comprehend their original role and significance. In the
process, I have not only compiled previous research on my topic, but also forged ahead to
discover the original patron of these chests, who was previously unknown.
In my thesis, I begin with the social context of marriage cassoni in fifteenthcentury
Florence, considering the role that these chests once played in both the public
rituals of marriage and the private domestic culture of the home. Next, I survey the
workshop in which these chests were produced: the various domestic products it created,
the stylistic development evident in surviving works, and the process by which the
elegant marriage cassoni were crafted. I then consider the iconography of the Queen of Sheba, tracing her story’s evolution through history from the time of King Solomon
through the mid-fifteenth century, and exploring the ways in which the panels in
Birmingham and Boston weave together the Queen’s traditional iconography with
contemporary artworks and events. Finally, I investigate the issue of patronage.
Beginning with my identification of the Pitti coat of arms in The Meeting of Solomon and
the Queen of Sheba, I consider possible candidates for the patron of this cassone pair. I
determine that, in all likelihood, it was Luca Pitti who commissioned these domestic
artworks for his daughter’s marriage to Francesco Segni in 1459.

CCh were significantly smaller. Our evidence indicates distinct
isoform functions with PMCA1 involved in overall
Ca2+-clearance, while PMCA4 is essential for
the [
Ca2+]i and contractile responses to
CCh. We hypothesized that a localized increase of
[
Ca2+]i in the absence of PMCA4 may inhibit
the CCh-receptor-mediated function. CCh-induced contractility after
treatments with iberotoxin (IBTX,
Ca2+
sensitive K
+ channels
(BK
+Ca) blocker) and
2-aminoethoxydiphenyl borate (2-APB, capacitative
Ca2+ entry (CCE) blocker) did not eliminate
the difference between WT and Pmca4
-/-,
indicating that neither BK+
Ca channels nor CCE were altered.
However, the L-type
Ca2+-channel (Cav1.2)
blocker nifedipine exhibited significantly less inhibition in
Pmca4
-/- than in WT and Cav1.2 activators
Bay K 8644 and KCl significantly increased the force in
Pmca4
-/- and restored the force to the same
level as in WT. Western blot indicated the expression of Cav1.2 was
not changed. Our data indicates the activities of Cav1.2 and
ACh-receptor mediated signal transduction may be disrupted by
localized increases in [
Ca2+]i due to the
absence of PMCA4 in Pmca4
-/-.
Immunocytochemistry of PMCA and Cav1.2 indicates that PMCA1 and
PMCA4 isoform have different distribution pattern, with PMCA1
uniformly distributed while PMCA4 punctated, yet, PMCA4 always
associated with Cav1.2.

▼ The mother is a prominent figure in the art of the seventeenth-century Dutch Republic. Artists used the mother to show virtues that should be taught to children. It was her example that not only educated her children in how to act, but also ensured that the Dutch society would be moral. This thesis examines how painters used the image of the mother to represent the
ideal and to prompt the mothers to reach this virtue. It will explore some of the numerous
images of seventeenth-century Dutch mothers and will examine how these images reflect
the spiritual and moral beliefs of society. It will investigate why these images were so popular and why leading painters such as Pieter de Hooch, Nicolaes Maes, and Jan Steen produced so many of these images. By examining three types of images, the mother as physical nourisher, the mother as educator, the mother as disciplinarian, this thesis will prove that Dutch society not only ardently adhered to these principles, but also praised the women who exemplified them. It begins by focusing on images that deal with the mother providing nourishment for her child and will present an overview of the importance of the nursing mother within society and address the religious implications placed on the mother who refused to nurse. It will explore how the mother becomes the life sustainer of society. Next, it will show how it remains the mother’s responsibility to provide proper nourishment for her children as they grow. The mother’s role as educator will then be examined. This chapter will address the church’s view on education and why such tasks were considered important. Finally,
the mother will be shown in the role of disciplinarian by looking at images of instruction
by Dutch mothers. These images will uncover not only what behaviors were deemed
acceptable within society but also why it was the mother’s responsibility to correct the child. By examining the mother in these three roles, this thesis will show how she became a representation of the ideal mother meant to inspire the Dutch women.

Cytosolic Ca2+,
[Ca2+]I , has a key role in intracellular
signalling during excitation-contraction coupling (E-C coupling) in cardiac myocytes. The
sarcoplasmic reticulum (SR) is a main intracellular Ca2+ store
and the Na+-Ca2+ exchanger (NaCaX) is
a major mechanism to extrude Ca2+ for balancing the
Ca2+ influx via L-type Ca2+ channels
during excitation. Furthermore, [Ca2+]I
also affects the configuration of the action potential (AP). The present study, by
combination of animal experiments and computer simulations, investigated the roles of
[Ca2+]I, SR and NaCaX in cardiac
myocytes, in Ca2+-induced Ca2+
release (CICR) and in modulation
of APs. The following were studied: (I) the stretch-induced effects on rat atrium and the
role of [Ca2+]I in modulation of AP; (II)
the role of the SR in modulation in rat atrium by stretch; (III) the role of NaCaX; (IV)
the role of [Ca2+]I in modulation of
action potential duration (APD) in myocytes with short and long action potential duration.
In isolated rat atrial preparations, the physiological or moderate stretch stimulus
caused two- phasic rise of developed contraction, rapid and slow phases, accompanied with
slow increments of [Ca2+]I and
prolongations of action potentials durations in continuous recordings. In sustained stretch,
the APD and [Ca2+]I were all increased
significantly when intra-atrial pressure increased from 1 to 3 mmHg. In computer
simulations, employing a rat atrial model (RA model), it was found that stretch-activated
channels and increased Tn C affinity for Ca2+ alone could not
produce the changes in the
experiments. Only after both mechanisms applied to model cells, the main experimental
findings could be mimicked (I). The prolongation of APD induced by stretch in rat atrial
preparations was reversed after depleting the Ca2+ content of
the SR by application of the
SR functional inhibitors, ryanodine, thapsigargin and caffeine (II). In the computer
simulation using modified guinea pig ventricular model, the Ca2+
entry via the reversal of NaCaX was found to be accounting 25% of the total activator
Ca2+ for triggering Ca2+ release
from the SR during normal excitation. This contribution increases with elevated
[Na+]i (III). In a guinea pig ventricular
model (GPV model) and a RA model were employed for investigating the regulation of APD by
[Ca2+]I-dependent membrane currents.
Increased SR Ca2+ content produced an elevated
[Ca2+]I in both model cells, leading to
prolongation of APD in the RA model but shortening in the GPV model. Increased
[Ca2+]I enhances the NaCaX current in the
same scale in both models, but inhibits L-type Ca current much more in the GPV model than
the RA model (IV).
In conclusion, (I) Stretch-induced [Ca2+]I
increase prolongs the rat atrial AP by enhancing the NaCaX inward current. Stretch-activated
channels (SACs) and increased affinity of TnC for Ca2+ are

Chronic overload of the heart is the major cause of left ventricular hypertrophy (LVH) and eventually heart failure. It is generally accepted that autocrine/paracrine factors, such as angiotensin II (Ang II) and endothelin-1 (ET-1) contribute to the development of LVH. Cardiac hypertrophy and failure are characterized by attenuated responsiveness to β- adrenergic stimulation and accumulation of collagenous material to the left ventricular wall. The present study aimed to characterize the roles of ET-1 and Ang II in the regulation of cardiac function. The role of the plasmamembrane Ca2+-ATPase (PMCA) in ET-1 induced cardiac responses and the role of type XV collagen in cardiac function were also studied.
Both ET-1 infusion and mechanical loading were able to induce positive inotropic effect and induction of early response genes in isolated perfused hearts. ET-1 also induced strong vasoconstriction. Cardiomyocyte-specific PMCA overexpression inhibited the ET-1 induced hypertrophic response, while inotropic response remained unaltered. ET-1 was found to induce release of adrenomedullin (AM), a potent vasorelaxing and inotropic peptide. Infusion of AM antagonized the vasoconstrictive effect of ET-1 independently of nitric oxide. In hypertrophied rat hearts ET-1 was found to contribute significantly to the Frank-Starling response, a fundamental mechanism regulating contractile performance of the heart. In mice hearts, ET-1 was found to play a dual role in load induced elevation of contractile strength: ETA receptors mediated an increase, while ETB receptors mediated an inhibitory effect on contrcatile force. Ang II was not contributing to the contractile response to load in either rat or mice hearts. Blunted response to β-adrenergic stimulus and increased vulnerability as a result of exercise was observed in mice lacking collagen XV.
In conclusion, the present results underscore the importance of the local factors, especially ET-1, in regulation of cardiac function, not only in terms of hypertrophic but also in terms of contractile response to load. The results also suggest a role for PMCA in regulation of cardiac function. Lack of type XV collagen was found to result in cardiac dysfunction with many features similar to those of early heart failure.

protein kinase II (CaMKII) is a multifunctional enzyme that is
activated by increases in intracellular
Ca
2+ . CaMKII is expressed in all tissues but
is most abundant in the brain. Diversity in subunit composition and
Ca 2+ sensitivity permit the coordination of
multiple cellular functions including muscle contraction, synaptic
vesicle release, proliferation and differentiation. The factors
allowing CaMKII to mediate a broad spectrum of cellular processes
with such precision are just beginning to be understood. Proximity
of
Ca 2+ signals and the CaMKII activator
Calmodulin (CaM), as well as CaMKII subunit composition influence
the processes that CaMKII will mediate. As with other kinases,
intracellular localization is thought to be a major factor in
substrate selection. This dissertation presents two perspectives on
CaMKII function. First, in light of the diversity and complexity of
CaMKII function, investigation of compartmentalized CaMKII will
provide more insight into the physiological condtions under which
it operates. Targeted CaMKII genes also have the potential to be
therapeutic agents. For example, CaMKII activates a chloride
conductance at the plasma membrane of secretory epithelia. Plasma
membrane targeted CaMKII may provide stimulation of this chloride
channel that is adequate compensation for the defects in fluid
secretion characteristic of cystic fibrosis (CF). CF is a lethal
hereditary disease characterized by the accumulation of thick mucus
secretions from polarized epithelial cells lining the airways and
gastrointestinal tract, which impede organ function. The
feasibility of targeting CaMKII to specific subcellular locations
was investigated in HeLa cells with three CaMKII gene constructs
each with a different plasma membrane targeting motif. The data
show that plasma membrane targeting of CaMKII is feasible when the
targeting motif is attached to the N-terminus of the protein, but
not the C-terminus. The second part of this dissertation examines
the role of CaMKII at the G2/M transition of the cell cycle.
Previous reports show that CaMKII is required for progression from
G2 into mitosis. Expression of autonomous CaMKII prevents this
transition. Cell lines that display stable expression of an
autonomous mutant CaMKII (CaMKIIT286D), reveal experience a delayed
mitosis. It is likely that this delay is the result of autonomous
CaMKII activation of a cell cycle checkpoint protein that impedes
mitosis.

▼ Im Zentrum dieser Arbeit steht ein einzelner Cluster aus
Inositol-1,4,5-trisphosphat (IP3) Rezeptorkanälen (IP3Rs). Die fundamentale
Bedeutung eines solchen Clusters beruht auf seiner Funktion in der
intrazellulären Signalweiterleitung. Er setzt Ca2+ aus intrazellulären
Speichern wie dem endoplasmatischen Retikulum (ER) frei, wodurch eine Vielzahl
von Prozessen reguliert wird. Dabei spielen sowohl hoch lokalisierte
Elementarereignisse, sogenannte Puffs, als auch Ca2+ Wellen eine entscheidende
Rolle. Während ein Ca2+ Puff nur einen einzelnen Cluster involviert,
erstrecken sich Ca2+ Wellen auf Teile der Zelle oder sogar die gesamte Zelle.
Wellen werden nur durch das koordinierte Zusammenspiel von mehreren Puffs
ausgelöst. Daher stellt ein einzelner IP3R Cluster den gundlegenden Baustein
für die durch IP3 vermittelte Ca2+ Dynamik dar.
Im ersten Teil der Arbeit haben wir Ströme und Konzentrationsprofile
simuliert, wie sie bei der Freisetzung von Ca2+ aus dem ER auftreten. Ein
Cluster wurde als ein leitendes Loch in der Membran des ER mit einem
Durchmesser von 6-36 nm beschrieben. Das endoplasmatische Retikulum wurde als
eine Scheibe mit einem Radius von 1-12μm und einer inneren Höhe von 28nm
modeliert. Wir haben die Abhängigkeit der Ströme von der Ca2+ Konzentration im
ER, wie sie in Versuchen mit Lipid-Doppelschichten gemessen worden ist, an die
zelluläre Geometrie angepaßt. Die simulierten Ströme stimmen sehr gut mit
Messungen der Signalmasse in Xenopus Oozyten überein. Die Abhängigkeit der
Ströme von der Ca2+ Konzentration im ER stellte sich als linear heraus.
Weiterhin ergab sich, daß die Ströme ungefähr proportional zu der Wurzel aus
der Anzahl der offenen Kanäle in einem Cluster sind. Die zytosolischen Ca2+
Konzentrationen an einem offenen Cluster erreichten Werte von 25-170μM. Der
Anstieg der Ca2+ Konzentration in einer Entfernung von einigen Mikrometern von
einem offenen Cluster war im Bereich von einigen Nanomolar. Die Ströme

Um die Konsequenzen zu untersuchen, die sich aus der stark lokalisierten Ca2+
Freisetzung in einem deterministischen Modell ergeben, haben wir einen neuen
Ansatz zur Beschreibung der Reaktions-Diffusions-Dynamik zwischen
diffundierenden Molekülen und räumlich fixierten Reaktionspartnern entwickelt.
Die immobilen Reaktanten bildeten Cluster, die aus nur wenigen Molekülen
bestehen. Diese Moleküle nahmen nur dann an der Reaktion teil, wenn sie
aktiviert wurden. Wir haben diese Aktivierungsdynamik auf die zeitabhängige
Ausdehnung eines aktiven Gebietes innerhalb des Clusters abgebildet. Eine
lineare Stabilitätsanalyse zeigte, daß die Diffusion die Stabilität der

▼ L-type Ca2+channels are protein complexes that play a critical role in the
regulation of muscle contractility. Gating as well as regulation of voltage-
dependent Ca2+channels is for a large part determined by the interaction
between ion-conducting a 1 and b subunits.
An aim of this study was to analyze modulation of the smooth-muscle
Ca2+channel a 1C-b subunit by the accessory b 2a subunit at single-channel
level. In addition, the functional role of the a 1-b interaction was studied
using an synthetic peptide (AID-peptide) that corresponds to the sequence of
the a 1 interaction domain (AID). Ca2+ channels derived by coexpression of a
1C-b and b 2a subunits in HEK cells exhibited a significantly higher open
probability (Po) than a 1C-b channels. High Po gating of a 1C-b·b 2a channels
was associated with promotion of long-lasting openings (t >10ms). Binding
experiments with the AID-peptide showed a specific affinity to the native b 2
subunit. Administration of the AID-peptide to the cytoplasmic side of inside-
out patches induced a rapid decline of Po of a 1C-b·b 2 channels. The AID-
peptide did not modify a 1C-b channel function in absence of expressed b 2a
subunit, and the scrambled control peptide failed to affect a 1C-b ·b 2a
channels. The results demonstrate that the b 2 subunit controls fast gating of
a 1C-b channels, and suggest the a 1-b interaction domain as a possible target
of modulation of the channel.
Recently it was shown that ahnak, a 700 kDa phosphoprotein, is linked to
cardiac L-type Ca2+ channel via the b 2 subunit by coprecipitation
experiments. So far, the molecular details of this interaction has not been
studied. Herein, the interaction was defined between b 2 subunit and ahnak.
Overlay binding assays as well as sedimantation equilibrium analysis with
glutathione-S-transferase fusion proteins revealed specific, high-affinity b
2a binding for the most C-terminal region of ahnak (ahnak-C2, aa 5262-5643).
Within the b 2a protein, two binding sites for ahnak-C2 with differing
affinities (Kd1=53 nM, Kd2=212 nM) were determined. The high affinity site
localizes in the C-terminal (aa 195-606, Kd=55 nM) b 2a portion, while the
binding site of lower affinity resides in the N-terminal (aa 1-200, Kd=328 nM)
portion of b 2a subunit. Protein-protein interaction between ahnak-C2 and b
subunits was corroborated by in vitro pull-down assay with native Ca2+
channels, since ahnak-C2 affinity beads recovered cardiac b 2 subunits and
skeletal muscle b 1a subunits together with the corresponding a 1C and a 1S
proteins. These results underline the significance of b subunits in tethering
the a 1 subunit of L-type Ca2+ channels to ahnak.

synthase (iNOS) plays critical roles in inflammation and host
defense. iNOS expression is induced by inflammatory stimuli and
exhibits constant activity once expressed. Hence NO production from
iNOS has been thought to be primarily controlled via enzyme
expression. In this study, we identify an array of novel mechanisms
showing that iNOS is regulated at multiple levels including gene
expression, protein aggregation, and protein degradation.
Calmodulin (CaM) has always been thought as an
iNOS cofactor facilitating electron transfer amid NO syntheses. We
now show that CaM is essential for iNOS induction. CaM inhibition
or knockdown prevented iNOS expressions in macrophages stimulated
by inflammatory mediators. Further studies revealed that CaM acted
through
Ca2+/CaM-dependent kinase II
(CaMKII), which functioned as a cardinal initiator in iNOS gene
transactivation via both LPS-NF-¿¿¿¿¿B and IFN-γ-STAT1 pathways.
Our study also found that iNOS gene transactivation required
cytosolic
Ca2+ elevations. CaM or CaMKII
inhibition was found to prevent iNOS induction in endotoxemic mice
and improved survival rates. These studies extend the role of CaM
from an enzyme cofactor to an essential modulator in iNOS gene
transactivation. Once expressed, biological iNOS
output is determined by the levels of functional enzyme, which is
influenced by protein stability. Our studies found that while
initially existing as a soluble protein, iNOS progressively formed
inactive protein aggregates. Blocking NO production prevented iNOS
aggregation inside cells. iNOS aggresome formation could be
recaptured by exposing cells to exogenous NO. The finding that NO
per se induces iNOS aggregation and inactivation suggests aggresome
formation as a feedback inhibition mechanism in iNOS
regulation. After elucidating NO-mediated iNOS
aggregation in physiological conditions, we then investigated iNOS
protein stability under pathological circumstances upon heat shock
protein 90 (Hsp90) or CaM inhibition. Hsp90 was previously reported
to play an important role in iNOS function and gene expression. We
now find that dissociation of Hsp90 from iNOS led to iNOS
aggregation and these aggregates were cleared by
ubiquitin-proteasome system. Further studies revealed that the SPRY
domain-containing SOCS box protein 2 (SPSB2), but not the
C-terminus of heat shock protein 70-interacting protein (CHIP), was
essential for aggregated iNOS turnover in Hsp90-inhibited cells.
Our studies found that CaM inhibition disrupted CaM-iNOS
interaction, exposing the hydrophobic CaM-binding domain on iNOS
which mediated iNOS aggregation. These studies identified novel
roles of Hsp90 and CaM in modulating iNOS protein
stability. To gain a complete understanding of
iNOS regulation, we investigated the ubiquitination site
responsible for proteasomal degradation of iNOS. The N-terminal
