▼ Some research suggests that marijuana use facilitates an anti-inflammatory response, yet the relationship between marijuana use and inflammation, as measured by C-reactive protein (
CRP), remains poorly understood. The purpose of the present study was to examine the association between recency of marijuana use and serum C-reactive protein levels in a nationally representative sample of adults. It was hypothesized that marijuana use would be associated with lower
CRP levels, regardless of recency of use. A secondary data analysis was conducted via data from Waves III and IV (N=13,166) of the National Longitudinal Study of Adolescent to Adult Health. Marijuana use during the past 30 days was assessed in Wave III, and marijuana use during the past 30 days and past year was assessed at Wave IV.
CRP was dichotomized with a cutpoint of 3 mg/L. Chi-square analyses examined associations between sociodemographic and health-related variables and
CRP. Binary logistic regression analyses examined the association between marijuana use at Waves III and IV and
CRP levels at Wave IV. Past 30 day marijuana use was reported by 23.5% and 17.7% of participants at Wave III and Wave IV respectively, and 23.6% of participants reported past year marijuana use during Wave IV. Marijuana use was associated with lower
CRP levels in bivariate analyses. However, these associations attenuated after adjusting for sociodemographic and health-related covariates.

CRP or were considered old field. Switchgrass in the
CRP plots appeared to have a higher soluble protein concentration later in the summer than in the old field plot during 2009 and 2010. There was no difference in DPPH reduction between systems for either year, but there was a difference between sample times, with later samples having higher reducing power. Mean chlorophyll concentration in old field was significantly lower than the
CRP later in the summer. In a companion growth chamber study under water stressed conditions, plants grown in soil collected from the
CRP site had lower protein concentration and lower reducing power than those grown in soil from the old field site, but no difference was observed in chlorophyll concentration between the sites. Overall, plants grown in
CRP soil did better in the field and worse in the growth chamber, probably due to a lack of heat stress under field conditions.

▼ Hepatocellular carcinoma (HCC) is one of the most common malignancy and known as country disease in Taiwan. Hepatoma-derived growth factor (HDGF), a mitogen for various types of cells, plays an important role in liver carcinogenesis. HDGF overexpression is associated with tumor progression and poor survival outcome in many types of cancer including lung cancer, melanoma and HCC. C-reactive protein (
CRP) is a acute phase protein, belongs to a group of unique serum protein family and is mainly secreted by the liver. The serum
CRP level rises rapidly by up to 1,000 times during acute or chronic inflammatory conditions such as infection and cardiovascular diseases. However, the role of
CRP in liver carcinogenesis remains unclear. By using a panel of hepatoma cell lines, immunoblot analysis showed that
CRP is highly expressed in all hepatoma cells regardless of their differentiation status. Exogenous
CRP significantly stimulated the proliferation, wound healing, invasion and colonies formation of hepatoma cells. Besides,
CRP increased
CRP and its receptor CD64 expression in hepatoma cells. Interestingly,
CRP also enhanced the expression of HDGF/nucleolin axis in hepatoma cells. Finally,
CRP enhanced Akt phosphorylation and the expression of NFκB members. Because the role of HDGF in liver carcinogenesis and Akt/NFκB activation has been recently established, it seems plausible that
CRP may mediate the malignant transformation of hepatoma cells through activation of HDGF/nucleolin axis.

▼ Members of the genus Mycobacterium respire using two terminal respiratory oxidases: a cytochrome bd-type menaquinol oxidase (encoded by cydABDC) or an aa3-type menaquinol-cytochrome c oxidase. During the transition to hypoxic (< 0.1% air saturation) conditions, cytochrome bd is induced in Mycobacterium smegmatis but the molecular mechanism governing this induction remains unknown. Analysis of the proposed cydABDC operon in M. smegmatis under hypoxic conditions revealed that cydAB and cydDC were not polycistronic. A transposon-mediated random mutagenesis was performed and revealed genes involved in redox homeostasis and defense against oxidase stress including KatG and PruC were required for cydAB operon expression. DNA-binding experiments were performed using a cydA promoter as target DNA and a DNA-binding histone-like protein (Hlp) was identified as protein bound to the cydAB promoter region. A cAMP receptor protein (
CRP)-binding site (GTGAN6CCACA) was identified in the regulatory region of the cydAB operon and mutations in this sequence caused a significant reduction in cydA-lacZ expression.
The M. smegmatis mc2155 genome has two copies of
CRP [Msmeg_0539 (Crp1) and Msmeg_6189 (Crp2)]. Phylogenetic analysis suggested that Crp1 is uniquely present in fast-growing mycobacteria, whilst Crp2 is closely related to
CRP from other slow- growing mycobacteria. In addition, only Crp1 but not Crp2 could complement a Δ
crp mutant of the Gram-negative bacterium, Pectobacterium atrosepticum. To dissect the physiological role of each
Crp protein, transcriptional profiling studies using deletion mutants were conducted. Microarray analysis of the crp1 deletion mutant revealed 239 genes were differentially regulated including those involved in carbon catabolism and energy metabolism. However, a crp2-deletion mutant could not be obtained suggesting that this gene is essential for growth in M. smegmatis. Therefore, crp2 was conditionally expressed and transcriptional profiling studies were performed. Microarray analysis of the crp2 overexpression strain showed 58 genes were differentially regulated including genes from many biological processes. Identification of the
CRP promoter consensus in M. smegmatis revealed that both Crp1 and Crp2 recognise the same consensus sequence yet the regulon of Crp1 is distinct from that of Crp2. Taken together, these data demonstrate that each
Crp could be activated in response to unique signals. The mechanism of how each
Crp is activated remains to be elucidated.

▼ Chronic kidney disease (CKD) rates continue to rise in the United States. Cardiovascular disease (CVD) accounts for half of all deaths in patients with end stage renal disease. C-reactive protein (CRP) is an indicator of inflammation and is a recognized predictor of CVD. Recent evidence supports an inverse relationship between dietary fiber and CRP in healthy populations and populations with chronic disease, but its association with inflammation in CKD has not been thoroughly explored. Dietary fiber is generally restricted in dialysis patients. It is unknown if dietary fiber could play a role in reducing CRP in CKD patients.

such as N(epsilon)-lysine acetylation, are known to alter the
behavior of transcriptional regulators in eukaryotes, but very
little is known about the consequences of acetylation on
transcriptional regulation in bacteria. Here, I provide evidence
that a global transcriptional regulator of carbon metabolism, cAMP
Receptor Protein (CRP), promotes both enzymatic and non-enzymatic
lysine acetylation in E. coli. Non-enzymatic lysine acetylation
occurs when cells ferment acetate, such as during growth on high
concentrations of glucose. Intriguingly, CRP can be
non-enzymatically acetylated on several lysines, including lysine
100 (K100). I provide evidence that neutralization of the K100
positive charge, as would occur upon K100 acetylation, has a dual
effect on CRP activity. First, K100 neutralization decreases CRP
activity at some Class II promoters. This decreased activity likely
results from disruption of the interaction between Activating
Region 2 (AR2) of CRP and the RNA polymerase alpha subunit
N-terminal domain. Second, K100 neutralization increases the CRP
half-life, leading to increased CRP steady state levels. Due to
increased steady state levels, CRP activity is increased at some
Class I promoters, in which CRP does not require AR2. Taken
together, I propose that CRP promotes global acetylation, including
CRP K100 acetylation, when cells are grown on glucose by positively
regulating non-enzymatic acetylation. A consequence of K100
acetylation may be inverse regulation of Class II and Class I
promoters under these conditions. This mechanism may help regulate
carbon flux though central
metabolism.

▼ The Conservation Reserve Program was first established by the Food Security Act of 1985, with the primary purpose of preventing damage to highly erodible soils, made worse by intensified farming practices. Since its inception, the
CRP has grown to become the largest federal, private-land retirement program in the United States with an approximately $2 billion annual budget. The Agricultural Act of 2014 mandates a reduction in the enrollment cap from 32 million acres to 24 million acres by 2018. The reduction coupled with the higher than average commodity prices makes the decision of whether or not to reenroll in to the
CRP a difficult one for producers with expiring land. Expiring lands will equal almost 2 million acres in the year 2015, and approximately 7.1 million acres over the life of the farm bill.
This research will develop a computer based decision aid that incorporates all major aspects of the decision when considering enrollment in the
CRP, to be used by landowners and producers. These considerations include eligibility, the Environmental Benefits Index (EBI), the producers bid level, and evaluation of practice options within the
CRP and options outside the
CRP. The goals of this research are to produce a computer web based decision aid, as described above, with specific emphasis on a few key outputs. These include a probability of acceptance measure, the Net Present Value of all options available to the landowner/manager, and a measure of what that landowner’s/manager’s optimal bid is.
The methodology of this study relies on the use of simulation using the excel add-in Simetar. Users of the program are allowed to enter details about the land being considered for
CRP enrollment that the decision aid then uses to test scenarios for the land. Five hundred randomized point estimates are generated and compiled as a cumulative distribution function comparison chart, stoplight chart, and several other customized output representations for the user’s consideration.
This thesis details a test of an Agriculture and Food Policy Center representative farm. The results, given the input data taken from the farm, suggest that the least risky option for this farm with the highest net returns is to enroll the land in
CRP. It is important to note that this is simply a test of one farm and not a recommendation for all producers. Also, the results could be changed given different assumptions about the producer’s goals.
The study concludes that including risk in the decision making process regarding
CRP enrollment is a critical factor when determining the most financially rewarding result. Including riskiness in the decision making process warrants the use of a decision tool, like the one presented in this thesis.

▼ Background – Several previous studies indicated conflicting findings on parameters of
Metabolic Syndrome and
CRP in cigarette smokers. The aim of this study was to determine
whether there is difference in levels of parameters of Metabolic Syndrome and
CRP in
cigarette smokers and non- smokers in Adama, Ethiopia.
Methods – subjects were 99 healthy male individuals from age 15 to 60 years. The subjects
were divided into two groups of 50 smokers and 49 non- smokers. BMI, blood pressure
plasma levels of total cholesterol, triglycerides, HDL, and
CRP were measured and LDL
cholesterol was estimated using the Friedewald formula.
Results – Overall subjects’ plasma levels of total cholesterol, triglyceride, LDL, HDL and
serum
CRP were significantly elevated in smokers. No significant difference was found in
BMI, systolic and diastolic blood pressure between cigarette smokers and non-smokers. In
smokers alcohol drinking was significantly correlated with increased plasma HDL, systolic
and diastolic blood pressure while khat chewing was correlated with increased plasma total
cholesterol.
Conclusions – The results showed no difference in BMI between smokers and non smokers
and also there was no significant difference in blood pressure in smokers and non smokers.
Cigarette smoking is correlated with increased total cholesterol, TG, LDL, HDL and
CRP.
Among smokers blood pressure, total cholesterol and HDL were significantly higher in those
whose alcohol consumption was higher.

▼ A témaválasztásom azért esett a szepszisre, mivel az intenzív osztályokon, sőt még kórházi viszonylatban is vezető halálokként szerepel az egész világon. Ezt mi sem mutatja jobban, mint az az elrettentő adat, hogy a betegek átlagos halálozási aránya 28-50% között mozog. A mortalitási mutatókon felül az is súlyosbítja a helyzetet, hogy az intenzív osztályok egyik legmunkaigényesebb és legköltségesebb betegségcsoportját alkotja. A szepszis történetének, diagnózisának és terápiájának rövid ismertetésével indul a dolgozat. Ezt követően szó lesz a szepszis markereiről, kiemelten a PCT-ről illetve a
CRP-ről és a jelenlegi irodalmi állásfoglalásról ezen markerek tekintetében. Ezután következik a vizsgálatunk, mely a miskolci Borsod-Abaúj-Zemplén Megyei Kórház és Egyetemi Oktató Kórház Központi Aneszteziológiai és Intenzív Terápiás Osztályának betegadatait dolgozza fel egy fél éves távlatban.

▼ Dolgozatomban elsősorban a baktérium fertőzésekkel foglalkoztam, ami a leggyakoribb, legnagyobb jelentőségű újszülöttkori fertőzéseket okozzák. A neonatalis halálozást hozzávetőlegesen 23,4%-ban újszülöttkori fertőzés okozza. A fertőzés és a sepsis kimutatásának gold standardja a haemokultúra, de korlátozott a szenzitivitása, gyakran negatív az eredménye. A C reaktív protein (
CRP) a legalaposabban tanulmányozott és széleskörűen elérhető laboratóriumi teszt az újszülöttkori fertőzés diagnózisában. Munkám során a Debreceni Egyetem Neonatológiai Tanszékén 2016.01.01.-03.31. között született érett újszülöttek antibiotikumos terápiájának gyakorlatát vizsgáltam meg zárójelentések alapján. A DE KK Neonatológiai Tanszékén az újszülöttek nagy biztonságban vannak. A rizikófaktorok megfelelnek a nemzetközi irodalomnak. Az antibiotikum terápiás ellátás megfelel a nemzetközi protokollnak.

▼ Dietary flaxseed has cardioprotective effects that may be achieved through its rich content of the omega-3 fatty acid, alpha linolenic acid (ALA). We investigated the effects of dietary flaxseed both with and without an atherogenic cholesterol-enriched diet to determine the effects of dietary flaxseed on the expression of the adipose cytokines leptin and adiponectin.
Rabbits were fed one of four diets: a regular (RG) diet, or a regular diet with added 0.5% cholesterol (CH), or 10% ground flaxseed (FX), or both (CF) for 8 weeks. Levels of leptin and adiponectin expression were assessed by RT-PCR in visceral adipose tissue. Consumption of flaxseed significantly increased plasma and adipose levels of ALA. Leptin, but not adiponectin, mRNA expression was lower in CH animals and was elevated in CF animals. Changes in leptin expression were strongly and positively correlated with adipose ALA levels and inversely correlated with levels of en face atherosclerosis. Our data demonstrate that the type of fat in the diet as well as its caloric content can specifically influence leptin expression. The findings support the hypothesis that the beneficial cardiovascular effects associated with flaxseed consumption may be related to a change in leptin expression.

▼ BACKGROUND: To date, no studies have evaluated if moderate and/or vigorous PA physical activity (mv-PA) mitigates the adverse effect of a pro-inflammatory diet. METHODS: We conducted a cross-sectional analysis using data from the National Health and Nutrition Examination Survey (NHANES) and constructed the Dietary Inflammatory Index (DII) score. Expenditure of mv-PA was obtained from two sources. Multivariate Logistic Regression was performed to calculate Odds Ratios (OR) and 95% Confidence Intervals (CIs). RESULTS: Participants with a DII score in the top quartile were 33% more likely to exhibit elevated
CRP than persons with a DII score in the lowest quartile (referent). This estimate did not vary appreciably when either mv-MET-mins/wk or mv-PA.Mnt.mins/wk were added to the model. When examining the independent effect of self-reported leisure-time mv-PA, those who expended the recommended 1000 or more mv-MET-mins/wk were less likely to have elevated
CRP compared to the referent group of < 250 mv-MET-mins/wk. Likewise, with accelerometer-assessed PA, the topmost quartile of mv-PA minutes per week was associated with lower
CRP compared to the referent quartile. These estimates did not vary appreciably when the DII score was added to the model. CONCLUSIONS: Both a pro-inflammatory diet and mv-PA appear to be independently associated with a chronic low-grade inflammatory state.

▼ This project is concerned with method development in mass spectrometry (MS)-based proteomics in order to find putative biomarkers for early pregnancy ofdomesticated dogs. It is of importance for dog breeders to know whether the dogsbecome pregnant post-mating. Unlike humans, dogs are not known to possess aspecific hormone that indicates fetal development; therefore other biomarkers mustbe investigated. The approach of choice in this project was to look at proteins throughMS-based proteomics. For this purpose, serum samples from 11 pregnant dogs (case,different breeds) and 7 non-pregnant dogs (control, all beagle dogs) were sampledbefore-hand at the Swedish University of Agricultural Sciences. Each dog wassampled Day 1, Day 8, Day 15, Day 22 and Day 29 after optimal mating. Twodifferent proteomics approaches were conducted: Bottom-up (“Shotgun”) proteomicsand targeted proteomics (“targeted analysis”). In this study, Label-free Quantification(LFQ) was employed, which is a relative quantitative technique. The massspectrometer of choice was the Quadrupole-Orbitrap QExactive plus massspectrometer coupled to a nano-Ultra Performance Liquid Chromatography (UPLC).Method optimization was done with respect to concentration of samples prior to MSanalysis, as well as different LC-gradients. From shotgun screening experiments, itwas possible to identify 252 proteins. Ultimately, 9 proteins were investigated usingtargeted final analysis: CRP, SERPINC1, CP, PROS1, SERPING1, A2M, AGP,SERPINA1 and HP. For targeted final analysis, 21 peptides were considered.Calibration curves were constructed using 8 of the 21 targeted peptides; 1 peptide perprotein, except for HP which had 2 peptides per protein. The SERPINA1 and CPproteins had no appropriate peptides for targeted final analysis and were thusexcluded. It was confirmed that CRP was up-regulated in case dogs compared tocontrol dogs. The other investigatedproteins showed no significant signs of regulation. In order to improve the results; itwould be desirable to include more dogs in the study which would benefit thestatistics of protein regulation. However, the use of isotopic labeled standards andemployment of a Parallel Reaction Monitoring (PRM) method should be prioritizedfor obtaining absolute quantitative data.

▼ Purpose: To review the data of ESR and CRP for screening, diagnosing, and prognosticating adults who are co-infected with HIV and MTB. Additionally, we review the same data to assess MTB treatment response in the same patients.
Methods: We conducted a retrospective study of patients with suspected smear negative pulmonary tuberculosis at King Edward VII hospital in Durban, South Africa from October 2007 to November 2009. Sputum culture for M. tuberculosis was the gold-standard diagnostic test for active pulmonary TB. Results were stratified by HIV-infection and CD4 count. Participants were over 18 years of age, had any 2 of 4 tuberculosis symptoms, (cough, fever, night sweats and loss of weight) for more than two weeks. Data from 90 patients was for ESR and CRP at baseline, after 2 weeks and monthly up to six months during treatment for MTB. Multiple statistical models including McNemar’s test, and paired Student’s t-tests as well as regression models were used to analyze the data using SAS version 9.2.
Results: ESR and CRP had high diagnostic sensitivity for culture-positive TB. Therefore, a normal result may be valuable to “rule out” active disease. We showed that a combination of (ESR 100 or CRP 10) has a sensitivity of 100% and a specificity of 23% among culture confirmed TB patients. When comparing ESR and CRP between culture confirmed TB patients and those who tested negative for TB, at baseline ESR and CRP were both elevated. However, both ESR and CRP levels decreased during the course of treatment.
Conclusions: ESR and CRP had high diagnostic sensitivity for culture-positive TB. Therefore, a normal result may be valuable to “rule out” active disease. ESR and CRP which are elevated at baseline among HIV/TB patients can potentially be valuable biomarkers to monitor response to therapy.

▼ For five generations, Lauver Family Farms has been
founded upon faith, family, and farming near Rockwell City, IA. It
is these core principles and beliefs that drive everyday actions
through conservation minded decisions, community involvement, and a
passion for the land. Presently, the farm is operated by
Grandfather Don Lauver, Father Kevin Lauver, and sons Andrew and
Jacob Lauver. The Lauver Family Farm was originally purchased in
1942 by Joseph Gordon, who at his peak held 700 acres in his name.
In 1945 Glen and Viola Lauver purchased what is now Lauver Family
Farms, located on the Des Moines Lobe land region of Iowa. The Des
Moines Lobe is a glacial lobe encompassing rich, heavy soils with
high organic matter, requiring dredge ditches and tiling in many
areas.
Through a commitment to conservation, corn and soybean
acres are rotated annually. With regard to corn cultivation and
planting practices, soybean stubble is field cultivated once,
followed by planting. On soybean ground, the corn stalks are disk
ripped, and then field cultivated twice before planting soybeans.
The goal is to minimize trips through the field by exhibiting these
conservation tillage practices. If land has much slope or erosion
potential, then it is only disked and then planted. Currently, the
farm is comprised of 400 acres of row crops and 50 acres of
wetland, 30 acres on the Home Farm and 20 acres on the Obye Farm,
enrolled in the Conservation Reserve Program in 2002.
Kevin and
Don Lauver, the primary decision makers, requested an analysis of
the environmental and economic impact of the Conservation Reserve
Program on the farm. By taking acres out of production for at least
10 to 15 years that perennially drown due to often wet soil
conditions, they will be able to utilize the Conservation Reserve
Program as a risk management tool. Now, Lauver Family Farms is
faced with a decision to determine if a 10 or 15 year enrollment in
the Conservation Reserve Program has the greatest economic and
environmental return, since the current enrollment expires in 2016.
Procedures and methods were established to meet the purpose of
this thesis to determine which option was the most profitable
long-term for the operation. The purpose includes evaluating the
sources of data relevant to Lauver Family Farms decision by
utilizing decision tools to make a collective decision on the
future of the farmland and opportunity costs analyzed.
Lauver
Family Farms’ objective for this project was to determine how the
Conservation Reserve Program provides a return on the investment of
the decision to re-enroll, or even enroll more acres in the
program. This analysis will be used each time an enrollment
decision must be made, and will be of significant importance as
sons Andrew and Jacob Lauver make management decisions in the years
to come.

▼ Background: The role of CRP and IL-6 as a marker in the prognosis of asymptomatic TAA patients has not been well established. Therefore we evaluated a possible association between serum CRP and IL-6 and aneurysm dimension in this group of patients. Methods and Results: Serum CRP and IL-6 were determined and aneurismal size was measured in 26 patients with TAA. Mean (SD) CRP and IL-6 were 0,58 (1,07) mg/dl and 7,47(17,78) pg/ml respectively. Serum CRP and IL-6 did not correlate with aneurismal dimension. Moreover there was significant correlation between values of the ratio above 0,8 and aneurismal dimension (r=0,78485). Secondly there is also significant association smoking, age group of 69-80 years old and dyslipidemia and aneurysm dimension (p=0,0023, p=0,0607 and p=0,0704 respectively). Conclusions: This report shows that values of the ratio CRP/IL-6 above 0,8 are associated with aneurismal dimensions. The clinical significance of this finding is early to be established. Further studies with larger groups of patients with longer follow-up are needed to evaluate the usefulness of the serum CRP and IL-6 as markers of the disease progression.
Σκοπός: Ο ρόλος των CRP και IL-6 ορού στην ανευρυσματική νόσο της θωρακικής αορτής σε ασυμπτωματικούς ασθενείς ως δεικτών πρόγνωσης και εξέλιξης της νόσου δεν ερευνάται επαρκώς μέσα από την παγκόσμια βιβλιογραφία. Μέσα από αυτή την εργασία προσπαθούμε να αξιολογήσουμε μία πιθανή συσχέτιση ανάμεσα στις CRP και IL-6 ορού και το μέγεθος του ανευρύσματος σε αυτή την ομάδα ασθενών. Μέθοδος και Αποτελέσματα: Σε 26 ασυμπτωματικούς ασθενείς με ανεύρυσμα της θωρακικής αορτής, προσδιορίστηκαν οι τιμές των CRP και IL-6 ορού και μετρήθηκε και το μέγεθος του ανευρύσματος. Οι μέσες τιμές (τυπική απόκλιση) των CRP και IL-6 ορού είναι 0,58 (1,07) mg/dl και 7,47 (17.78) pg/ml αντίστοιχα. Από τα αποτελέσματα φαίνεται ότι δεν προκύπτει κανένας συσχετισμός ανάμεσα στις τιμές των CRP και IL-6 ορού και το μέγεθος του ανευρύσματος. Σημαντικό εύρημα είναι ότι σε τιμές του λόγου CRP/IL-6 άνω του 0.8 προκύπτει σαφής και δυνατός γραμμικός συσχετισμός (r=0,78485) ανάμεσα στις τιμές αυτές και το μέγεθος του ανευρύσματος. Επιπλέον προκύπτει σημαντική σχέση ανάμεσα στο κάπνισμα, την ηλικιακή κατηγορία 69-80 έτη και την υπερλιπιδαιμία και το μέγεθος του ανευρύσματος (p= 0.0023, p=0,0607 και p=0,0704 αντίστοιχα). Συμπέρασμα: Σ’ αυτή την εργασία, οι τιμές του λόγου CRP/IL-6 που είναι άνω του 0,8 σχετίζονται με το μέγεθος του ανευρύσματος. Η κλινική σημασία αυτού του ευρήματος είναι νωρίς για να καθοριστεί. Περισσότερες μελέτες χρειάζονται να γίνουν με μεγαλύτερα δείγματα πληθυσμού και με μεγαλύτερο χρόνο παρακολούθησης για να εκτιμηθεί επαρκώς η χρησιμότητα των CRP και IL-6 ορού ως δεικτών πρόγνωσης και εξέλιξης της ανευρυσματικής νόσου.

▼ The pheromone-mediated circuitry that governs bioluminescence in Vibrio fischeri is well understood; however, less is known about the environmental conditions that influence pheromone and light production. The environment has been shown to
have a profound effect on luminescence in V. fischeri, as cells in symbiotic association with the Hawaiian bobtail squid, Euprymna scolopes, are ~1000-fold brighter than non-symbiotic cells despite reaching similar cell densities. In this dissertation, I
show that luminescence is governed by a complex regulatory web and that certain environmental conditions mediate the regulation of pheromone circuitry. My first goal was to identify and characterize previously unidentified regulators that control
luminescence in V. fischeri strain ES114. I helped develop and employed a transposon mutagenesis system to discover novel negative regulators of luminescence. In this study, I characterized twenty-eight independent luminescence-up mutants with insertions
in 14 loci. This work revealed that environmental conditions such as inorganic phosphate and Mg2+ concentrations are integrated into the regulation of the pheromone-dependent lux system. Furthermore, I showed that competition between the LuxI- and
AinS-generated pheromones is an important and density-dependent factor in the level of light produced by V. fischeri ES114 cells, such that C8-HSL inhibits luminescence in dense populations. The second goal of this work was to clarify the role of cAMP
receptor protein (CRP) in luminescence. Attempts to study the effects of glucose on V. fischeri luminescence have been contradictory and inconclusive, possibly due to strain-specific effects. I confirmed that both cAMP and glucose modulate light
production in ES114, which is consistent with CRP regulation of luminescence. Previous researchers proposed a model wherein CRP regulates transcription of luxR, which encodes a pheromone-dependent activator known to induce luminescence. The data reported
in this dissertation indicates that CRP is involved in the expression of not only luxR but also ainS. Using qRT-PCR and purified CRP, I show that CRP significantly increases transcript levels of both luxR and ainS, and that the CRP protein binds the
upstream promoter regions of both genes. Taken together, this dissertation illustrates and clarifies how environmentally responsive regulators are integrated with pheromone-mediated circuitry in the regulation of V. fischeri
bioluminescence.

▼ Le ruxolitinib, un inhibiteur de JAK1/2, est actuellement utilisé pour le traitement des syndromes myéloprolifératifs et montre des propriétés anti-inflammatoires. Cependant, les mécanismes cellulaires et moléculaires à l’origine de ses effets restent peu élucidés. Au travers de ces travaux, les effets régulateurs du ruxolitinib sur l'expression de marqueurs de l'inflammation et de protéines de détoxication des médicaments ont été évalués in vitro dans différents types cellulaires. Dans les macrophages humains en culture primaire, le ruxolitinib réprime l’expression de cytokines inflammatoires secondairement induites par la signalisation de l’IFNβ, en réponse au LPS. Similairement, le ruxolitinib inhibe l’expression de protéines de la phase aiguë induites dans les hépatocytes humains en culture primaire et la lignée cellulaire HepaRG. Cet inhibiteur réprime notamment l’expression de la CRP secondairement induite par la signalisation IL6, en réponse au LPS. Aussi, il permet de restaurer l’expression et l’activité de protéines hépatique de détoxication des médicaments réprimées par un stimulus inflammatoire. Dans l’ensemble de ces études, le ruxolitinib montre une efficacité à des concentrations atteintes chez les patients. Ces travaux montrent tout d’abord que par ses propriétés anti-inflammatoires retrouvées dans différents modèles, le ruxolitinib pourrait se montrer intéressant pour le traitement d’un large panel de pathologies inflammatoires. De plus, son utilisation en clinique pourrait nécessiter d’adapter les doses de médicaments associés dont le métabolisme est susceptible d’être modulé par une diminution de l’inflammation par cet inhibiteur de JAK.
Ruxolitinib is a Janus kinase (JAK) 1/2 inhibitor currently used for the treatment of myeloproliferative neoplasms. It shows anti-inflammatory properties, but the involved molecular and cellular mechanisms remain poorly understood. The regulatory effects of ruxolitinib on the expression of inflammatory markers and drug-detoxifying proteins were investigated in different cellular models through these studies. In human macrophages, ruxolitinib represses the expression of inflammatory cytokines secondarily induced by the IFNβ signalling pathway, in response to LPS. Similarly, ruxolitinib inhibits the expression of acute phase proteins induced in human hepatocytes and in the HepaRG cell line. It also notably represses the expression of CRP secondarily induced by the IL6 signalling pathway, in response to LPS. Moreover, ruxolitinib restores the expression and activity of hepatic drug-detoxifying proteins inhibited in inflammatory conditions. In these studies, ruxolitinib is shown to be efficient at concentrations which can be reached in patients. Altogether, these results show that, firstly, as its anti-inflammatory properties are shown throughout different cellular models, ruxolitinib could be interesting for the treatment of various inflammatory pathologies. ▼ Zerebrale Erkrankungen, wie Schädelhirntrauma (SHT) und Subarachnoidalblutung (SAB) sind mit einer hohen Morbidität und Mortalität vergesellschaftet und stellen eine ernsthafte medizinische und ökonomische Herausforderung dar. Grundlage für die Entwicklung neuer effektiver Therapieansätze ist das Verständnis der pathophysiologischen Mechanismen dieser Krankheiten. Das Entstehen eines vasogenen Hirnödems ist eine schwere Komplikation nach SHT und SAB und beruht u.a. auf einem Öffnen der Bluthirnschranke (BHS). Ein möglicher zu Grunde liegender Mechanismus könnte die Aktivierung der Myosin-leichte-Kette-Kinase (MLCK) sein, was man therapeutisch unterbinden könnte.rnIn der vorliegenden Studie wurde in zwei unterschiedlichen experimentellen, zerebralen Schadensmodellen der Einfluss des kontraktilen Apparates auf die BHS Störung untersucht. In dem Schadensmodell des SHT sind die Hauptergebnisse: 1.) die Myosin-leichte-Kette-Kinase (MLCK) wird durch das induzierte Schädelhirntrauma hochreguliert. 2.) eine pharmakologische MLCK Inhibition stabilisiert die BHS, senkt den ICP und das Hirnödem nach experimentellen SHT. 3.) die MLCK Inhibition führte nicht zu einer Verbesserung des Hirnschadens, der neurologischen Funktion oder der zerebralen Inflammation 24 Stunden nach SHT, obwohl angenommen wird, dass die Entstehung eines Hirnödems den sekundären Hirnschaden vergrößert. In einer weitern Studie wurde untersucht, durch welchen Signalweg dieser zugrunde liegende Mechanismus aktiviert wird. In einem in-vitro BHS Model konnte gezeigt werden, dass C-reaktives Protein (CRP) über die Bindung an Fcγ-Rezeptoren den kontraktilen Apparat aktiviert und somit zu einem Öffnen der BHS führt. Obwohl der CRP Plasmaspiegel nach experimentellen SHT ansteigt, kommt es nicht zu einer Verringerungrndes Hirnwassergehaltes in FcγR-/- Mäusen. Die Entstehung des vasogenen Hirnödems wird im murinen CCI Model somit nicht über den Fcγ-Rezeptor vermittelt. Die in-vitro gezeigte Fcγ vermittelte Öffnung der BHS konnte in-vivo in dieser Studie nicht reproduziert werden. Mit der vorliegenden Studie kann nicht ausgeschlossen werden, dass CRP über einen Fcγ unabhängigen Mechanismus eine Öffnung der BHS vermittelt. Jedoch deuten die Daten daraufhin, das CRP im murinen CCI Model eine untergeordnete Rolle spielt. Die FcγR-/- Mäuse zeigten allerdings ein deutlich reduziertes Kontusionsvolumen und eine reduzierte Mikroglia Aktivierung, was darauf hindeutet, dass FcγR eine wesentliche Rolle bei der zerebralen Inflammation spielen.rnIn dem Schadensmodell der experimentellen SAB konnte gezeigt werden, dass die Inhibition der MLCK die Folgen einer SAB mindert. Sie führt zu einer Senkung des Hirnödems, des intrakraniellen Drucks und Verbesserung der neurologischen Erholung nach experimenteller SAB. ▼ Les bactéries sont en permanence confrontées à des changements d'environnements. La régulation transcriptionnelle joue alors un rôle majeur dans l'adaptation des bactéries. En particulier, la bactérie phytopathogène D. dadantii s'est récemment adaptée à l'hôte végétal. Elle produit en particulier des pectate lyases (Pel) qui dégradent la pectine, ciment des parois végétales, et jouent un rôle majeur dans le développement de la maladie. Les gènes pelD et pelE, malgré la forte divergence dans leur expression, sont issus d'un transfert horizontal suivi d'une duplication récente. La question de l'intégration de ces gènes avec les régulations préexistantes s'est alors posée.Dans un premier temps, les mécanismes moléculaires détaillés de la régulation de pelD ont été étudiés. Il a été montré que cette régulation s'appuie sur un promoteur divergent de forte affinité pour l'ARN polymérase mais de faible efficacité pour la transcription et sur un arrangement stratégique de quatre sites de fixation de répresseur FIS et deux sites de l'activateur CRP. Tous ces éléments interagissent entre eux pour produire une régulation fine de l'expression de pelD. L'origine de la divergence régulatrice entre les paralogues pelD et pelE a par la suite été explorée. De manière surprenante, la divergence entre ces deux gènes et leur sélection s'appuie presque exclusivement sur un décalage de la position du promoteur de pelE (« TSS turnover ») qui l'a transformé en initiateur de la dégradation de la pectine. Ce mécanisme très fréquent chez les eucaryotes pluricellulaires (homme, drosophile, souris…) n'avait jamais encore été décrit chez les bactéries.A travers l'étude des promoteurs pelD et pelE de D. dadantii, de nouveaux mécanismes renforçant l'importance de la régulation transcriptionnelle dans les processus adaptatifs ont ainsi été découverts
Bacteria face frequent environmental changes. Transcriptional regulation plays a major role in the adaptation to these changes. In particular, the phytopathogen bacteria Dickeya have recently adapted to vegetal hosts. They produce Pecate lyases (Pel), among others, to degrade pectin in plant cell walls, which is necessary for disease development. The pelD and pelE genes, despite the strong divergence in their expression, originate from a horizontal gene transfer followed by a recent duplication. This raises the question of their integration into the preexisting regulatory networks.Detailed molecular mechanisms of the transcriptional regulation of pelD were studied first. It was shown that this regulation relies on a high-affinity but low transcription efficiency divergent promoter and a strategic arrangement of four FIS repressor binding sites and two CRP activator binding sites. ▼ L’étude des marqueurs biologiques dans la broncho-pneumopathie chronique obstructive (BPCO) et l'asthme, deux maladies respiratoires chroniques affectant des millions de personnes dans le monde, pourrait améliorer leur diagnostic, leur traitement et leur prévention.Cette thèse comprend deux parties. La première visait à évaluer l'association entre un marqueur spécifique des poumons, la protéine surfactant D (SP-D) sérique, et la BPCO, et à trouver un seuil de SP-D capable de discriminer les patients BPCO des témoins. Elle a été réalisée dans le cadre d’une étude cas-témoin au Liban incluant des patients BPCO (n=90), des asthmatiques (n=124) et des témoins (n=180). La deuxième partie visait à évaluer les associations chez les adultes des marqueurs de l’inflammation systémique (protéine C-réactive ultra-sensible, hs-CRP (n=252), et des cytokines (n=283)) et des marqueurs de dommages dus au stress oxydant (8-isoprostanes 8-IsoPs (n=258) du condensat de l’air exhalé) avec les phénotypes de l’asthme.Elle a été réalisée dans le cadre de l'étude épidémiologique longitudinale Française des facteurs génétiques et environnementaux de l'asthme (EGEA).Les résultats ont montré que les niveaux de SP-D sériques étaient associés positivement avec la BPCO et des seuils des niveaux de SP-D chez ces patients ont été identifiés avec d'excellentes valeurs discriminantes. Dans EGEA, aucune association n'a été trouvée entre les niveaux de hs-CRP sériques et le contrôle de l’asthme. Des profils de cytokines sériques (identifiés par analyse en composante principale) avec des niveaux élevés d’interleukine(IL)-1Ra et d’IL-10 ont été associés avec moins de crises d'asthme et un risque plus faible d'un mauvais contrôle de l'asthme sept ans plus tard. Les résultats des analyses préliminaires sur les associations entre les niveaux de 8-IsoPs et les phénotypes de l'asthme sont également présentés.Globalement, ces résultats ont montré l'utilité d'étudier les marqueurs biologiques en lien avec la BPCO et l'asthme.
Studying the biological markers in chronic obstructive pulmonary disease (COPD) and asthma, two chronic respiratory diseases affecting millions of individuals around the world, could improve their diagnosis, their treatment and their prevention.This thesis includes two parts. ▼ Master’s thesis in the first part focuses on the effectiveness of communication in the selection of reference nodes during the anchor-free localization in WSN. In this part two anchor-free localization algorithms AFL and
CRP are explored. Knowledge learned on the selection of reference nodes and communication complexities of these algorithms are used in the second part. The second part is aimed at eliminating weaknesses in communication and subsequently proposed a new algorithm for selection of references. The principle of the newly proposed algorithm CASRU (Centralized Algorithm for Selection of Reference Nodes) is based on the principle of
CRP. CASRU algorithm builds logical tree structure and then filters the communication through it. New algorithm achieves much greater success in the selection in randomly distributed networks while reducing the amount of communication load.

▼ Canine Parvovirus remains a leading cause of
enteritis in dogs in South Africa and many other countries despite
the wide availability of effective vaccines. The virus does not
affect all dogs equally and the course of the disease depends on
the age, immune status and breed of the puppies as well as the
viral dose, route of exposure and the virulence of the strain.
Although aggressive supportive treatment can be successful, the
treatment and convalescent periods may be prolonged and
consequently expensive and the mortality rate relatively high,
causing many clients to forego treatment and elect for euthanasia
of their pet. Acute phase proteins (APP) are proteins that change
in concentration by at least 25% in animals subjected to external
or internal inflammatory challenges, such as infection,
inflammation or surgical trauma. Increased concentrations are
associated with poor outcome in certain diseases. C-reactive
protein (
CRP) is the most sensitive APP in dogs. Its normal
physiological concentration is low but increases rapidly with
inflammation or tissue destruction. Due to the fact that
CRP has a
relatively short half life in serum (6-8 hours) and a high response
in diseased animals, it can be used as a valid measure of a
systemic response to an initiating stimulus at the time of blood
sampling. By taking serial measurements, objective information
about the extent of the ongoing lesions in the patient can be
obtained and therefore may be used as a prognostic indicator. The
objective of this prospective observational study was to evaluate
the association of serum
CRP concentrations in puppies suffering
from canine parvoviral enteritis with morbidity and mortality, and
to determine the usefulness of
CRP to predict duration of
hospitalisation time. Seventy-nine client owned puppies naturally
infected with canine parvovirus were included. Parvovirus infection
was diagnosed on electron microscopic examination of faeces from
the puppies.
CRP was measured using an automated human C-Reactive
Protein Turbidimetric Immunoassay (TIA), which has been validated
for use in dogs. Serum
CRP measurements were performed at
admission, twice daily for the first 48 hours, then once daily
until death or discharge. There was a positive association between
odds of mortality and
CRP concentration on admission, as well as 12
and 24 hours after admission (P=0.04,P=0.005 and P=0.003,
respectively). Survival time was negatively associated with

▼ Acute phase proteins (APP) are ideal biomarkers for
inflammation due to their stability, relative ease of assay and
apparent relation between their concentration and the extent of the
insult to tissue. C-reactive protein (
CRP) is a positive major APP
in dogs and can be used as a predictive marker for risk of disease
and to monitor the response to treatment. Increased concentrations
in certain diseases are associated with poor outcome. This
cross-sectional, observational study of 75 dogs naturally infected
with Babesia rossi, a cause of virulent canine babesiosis, was
designed to examine the association of
CRP concentration at
admission and the magnitude of
CRP change 24 hours after admission
with outcome. Dogs were excluded if there was evidence of
concurrent inflammatory diseases at the time of admission,
infection with subtypes other than B. rossi, concurrent Ehrlichia
canis infections or euthanasia for reasons other than poor
prognosis. Diagnosis was confirmed by polymerase chain reaction and
reverse line blot.
CRP concentrations were determined by an
automated human
CRP Turbidometric Immunoassay (TIA), previously
validated for use in dogs (Bayer
CRP TIA, Newbury, UK), on serum
samples collected by jugular venipuncture on admission, prior to
any therapy, and thereafter daily until discharge or death. There
was no significant difference in admission
CRP concentration
between survivors (n = 57; median = 97.4 mg/l; mean ± SD = 107.5 ±
49.5), and non-survivors (n = 11; median = 101.4 mg/l; mean ± SD =
122.1 ± 64.6) (p = 0.39). After elimination of non-significant
predictors, a multiple exact logistic regression model for
predicting mortality contained glucose and
CRP. Mortality was
associated with decreased glucose levels (p = 0.0002) and increased
CRP levels (p = 0.045) on admission. Multiple regression analysis
failed to show a significant relationship between admission
CRP
concentration and number of days of hospitalization in the
survivors, adjusting for age and sex (p = 0.65). No significance
was found in the relationship between the magnitude of change in
CRP concentration 24 hours after admission, and the number of days
of hospitalization in survivors, (p = 0.34). Using an admission
CRP
concentration cut-off of 60 mg/l, survival proportions between the
two groups were no different (p = 0.34) and when applied to the
group of dogs that survived, it was not associated with length of
hospitalization (p = 0.25). In corroboration with previous reports
glucose was identified as a major prognostic marker for mortality,
but additionally the pro-inflammatory marker
CRP was identified as
a significant co-prognosticator. Copyright

▼ Pollinators are critical for the proper functioning of natural ecosystems and they benefit or are required by many agricultural crops. Despite their importance, there is little information available to enhance restoration efforts or manage habitats to benefit pollinators, especially for native species. My study was conceived to determine the abundance, richness and diet breadth of native invertebrate pollinators using native and restored prairie grasslands in the prairie pothole region (PPR) of eastern North Dakota. I compared native pollinators on native grasslands managed by the U.S. Fish and Wildlife Service (FWS) and on Conservation Reserve Program (
CRP) grasslands because they collectively constitute the largest land area of potential pollinator habitat in the United States PPR. Using vane traps and collecting pollinators visiting individual flowers, I evaluated change in species abundance, richness and pollinator/plant interactions biweekly from May to September in 2012 and 2013. I did not detect a significant difference in native pollinator abundance or richness between native and restored prairie grasslands. However, I did detect a decrease in native pollinator and a higher species turnover in
CRP grasslands relative to my native grassland sites in 2013. Pollinator richness and abundance changed among seasons and locations, likely due to diverse pollinator taxa responding to temporal and spatial variations in the floral community. In total, I observed 283 interactions corresponding to 55 plant taxa and 31 native bee taxa resulting in 177 unique links. Interactions and links were significantly higher on native grasslands, likely reflecting the higher diversity of blooming forbs in native prairie grasslands relative to
CRP. Native bees were caught mostly on native plants in native grasslands and on introduced plant species on my
CRP sites. My study suggests that pollinator abundance and richness in restored grasslands was similar to that in native grasslands. However, the floral composition varied by grassland type and
CRP grasslands may not be as effective at providing stable pollinator habitat. My analyses of pollen loads of individual pollinators identified 53 plant species that could be added to seed mixtures to diversify native and restored sites for pollinators. Similarly, identifying management disturbances for native grasslands and mature
CRP restorations would increase the diversity of plant species important to native pollinators to enhance the overall value to a diverse pollinator community.

▼ Negative childhood experiences and adult intimate partner violence may leave individuals vulnerable to poor mental and physical health in adulthood. Victims of emotional abuse and intimate partner violence have a higher risk of developing chronic medical conditions in adult life, including cardiovascular disease. Similarly, adults who lived in poverty as children have higher risk of cardiovascular disease. The immune system releases acute-phase proteins such as C-reactive protein (CRP) not only in response to infections but also in response to exposure to traumatic experiences. CRP has been linked to the incidence and progression of atherosclerosis, a precursor for cardiovascular disease. To understand potential pathways that link emotional abuse, intimate partner violence and exposure to poverty to chronic medical conditions in adulthood, this dissertation first investigates the association between emotional abuse and elevations in CRP. Second it examines the relationship between adult intimate partner violence and CRP. For emotional abuse and intimate partner violence gender interactions are tested. Lastly, the dissertation tests for an effect of poverty status in childhood and/or adulthood on CRP and investigates the extent to which poverty may compound the effects of emotional abuse and intimate partner violence on CRP.This dissertation uses data from the National Longitudinal Study of Adolescent to Adult Health (Add Health), a prospective observational study of a nationally representative sample of U.S adolescents enrolled in grades seven to twelve in the 1994-1995academic year. Funded by the National Institute of Child Health and Development (NICHD), the study is overseen by the Carolina Population Center at the University of North Caroline at Chapel Hill (UNC) (Harris, 2008). Add Health focuses on social factors that influence adolescent and young adult family and peer networks, health behaviors and health status. This dissertation uses data from three of the four waves of data: Wave I data was collected during 1994-1995 when respondents were enrolled in grades seven to twelve; Wave III data was collected from original Wave 1 respondents in 2001-2002 when these young adults were between the ages of 18 and 26 and Wave IV data was again collected from original Wave I respondents were between the ages 24 and 32 now settling into young adulthood. At Wave IV respondents provided blood samples in the form of dried blood spots (DBS), which were later analyzed for a variety of biological markers including C-reactive protein. All analyses for this dissertation used statistical methods adjusted for the complex sampling design of Add Health. Repeated exposure to emotional abuse was significantly associated with CRP in fully adjusted models, although there was no evidence of a gender interaction. Similarly, experiencing chronic intimate partner violence in adulthood was significantly associated with elevated CRP levels in fully adjusted models. ▼ Histone deacetylase 2 (HDAC2) is a critical part of
a co-repressor complex recruited by glucocorticoid receptors on
binding of steroid ligands to suppress inflammatory gene
transcription. Loss of HDAC2 in lungs of chronic obstructive
pulmonary disease (COPD) patients is linked to an impaired response
to glucocorticoids. Cigarette smoke (CS), the main etiological
factor in the pathogenesis of COPD, negatively alters lung HDAC2
activity and protein expression. We recently found that HDAC2, a
phospho-protein, is an early target of CS-induced serine
phosphorylation associated with increased binding of activated
protein kinase CK2α. Cigarette smoke extract (CSE)-induced
phosphorylation of HDAC2 required serine sites S394, S411, S422 and
S424, is dependent on CK2 catalytic subunits and negatively
regulates HDAC2 deacetylase activity. Most importantly however,
HDAC2 phosphorylation is critical for CREB binding protein
(CBP)-HDAC2 interaction and subsequently lysine residue acetylation
of HDAC2.
Loss of HDAC2 catalytic activity in vivo either via
genetic knock-out or in mice with increased susceptibility to
oxidative-stress markedly enhances the lung inflammatory response
with subsequent resistance to steroid therapy following LPS
exposure. Overall, these data suggests that phosphorylation and
acetylation are key factors that control the relative expression
and enzyme activity of HDAC2; a critical factor in the cellular
response to pro-inflammatory stimuli with corticosteroid therapy.

Aronson and Laughter (2015) amongst many others, studied and analyzed over 200 studies on the topic of culturally relevant education (CRE), and found convincing results that speak to the academic outcomes that are consistently reached whenever teachers employ CRE in the classroom (Cammarota, 2014; Covarrubias, 2017; Gay, 2010; Ladson-Billings, 1995; Paris, 2012;
approaches have demonstrated success, they are not being practiced and replicated in many parts of the state and country (evidenced by the persistent achievement gap), and it is not clear as to how administrators and teachers can work together to ensure that CRE practices are discussed, learned, and employed in school settings (Covarrubias, 2017). This mixed methods study will explore ways in which a school can impact teacher ideology and disposition toward prioritizing and supporting culturally responsive practices; using Participatory Action Research methodologies. Research that explores these topics is important, being that educators need to do a better job addressing success (or lack thereof) for students of color in California and in the entire United States.

▼ Objectives Inflammation and glycohemoglobin (HbA1c) are established risk factors for the development of cardiovascular disease. Recent evidence implicates high sensitivity C-reacting protein (hs-CRP), and thus inflammation, in the metabolic syndrome and diabetes mellitus, particularly in women.MethodsWe investigated the synergy of inflammatory markers and menstrual status in the glycaemic control of women. Several inflammatory markers among them hsCRP, IL-1, IL-4 and serum amyloid A (SAA) were measured in 1131 women . We classified these patients according to HbA1c. Traditional risk factors such as age, body mass index, triglycerides, LDL cholesterol, HDL cholesterol, haemoglobin A1c (Hba1c) were also measured at drug free baseline.ResultsUnivariate analysis revealed a significant correlation between HbA1c and hsCRP (Pearson r=0.219, ρ<0.001) or SAA (Pearson r=0.218 ρ<0.001). It also demonstrated a significant correlation of HbA1c with IL-1 (Pearson r=0.22, p<0.001), ΙL 1 (Pearson r=0.201 ρ<0.001) and IL4 (Pearson r=O.267 ρ=<0.001). Multiple linear regression analysis was applied in order to verify the relation of HbA1c to inflammation markers, adjusted for age, plasma creatinine, systolic blood pressure, glucose metabolic disorders, hyperlipidemia and menstrual status. Σκοπός: Η συστηματική φλεγμονή, διαδραματίζει σημαντικό ρόλο στην παθογένεια του σακχαρώδη διαβήτη. Σκοπός της έρευνας είναι η διερεύνηση σχέσης μεταξύ γλυκαιμικού δείκτη και γλυκαιμικού φορτίου και δεικτών φλεγμονής.Μεθοδολογία: Για την επίτευξη του στόχου χρησιμοποιήθηκε αντιπροσωπευτικό δείγμα 1131 γυναικών. Μετρήθηκαν βιοχημικοί δείκτες καθώς και οι δείκτες φλεγμονής IL-1, IL-4, SAA, hsCRP, στο προαναφερθέν δείγμα ατόμων σε συνάρτηση με τις αντίστοιχες τιμές της HbA1c αυτών.Αποτελέσματα: Έχει γίνει μονοπαραγοντική αλλά και πολυπαραγοντική ανάλυση των δεδομένων που δείχνουν την συσχέτιση των ανοσοδεικτών με την HbA 1 c. Στην μονοπαραγοντική ανάλυση διεφάνη η θετική συσχέτιση της γλυκοζυλιωμένης αιμοσφαιρίνης με την hsCRP (Pearson r=0.219, ρ<0.001), το SAA (Pearson r=0.218 ρ<0.001), την ΙL 1 (Pearson r=0.201 ρ<0.001) και την IL4 (Pearson r=O.267 ρ=<0.001). Μετά από διόρθωση για συγχυτικούς παράγοντες όπως της ηλικίας, της κρεατινίνης, την διάρκεια της εμμηνόπαυσης και. της αρτηριακής πίεσης, αποδείχτηκε η ανεξάρτητα θετική στατιστικά σημαντική συσχέτιση για κάθε μοντέλο γραμμικής πολλαπλής παλινδρόμησης του γλυκαιμικού φορτίου με τον SAA (β=0.491 ρ<0.001) τον αριθμός των λευκών αιμοσφαιρίων (β=0.015 ρ=0.001) την ΙL 1 (β=0.148 ρ<0.001) την IL4 (β=0.642 ρ<0.001) όπως επίσης και την hsCRP (β=0.314 ρ<0.001)Συμπεράσματα: Παρατηρήθηκε…
