▼ Natural radioisotopes, Th, Pa, Pb, Po and Be, have been used as important proxies in oceanographic investigations for decades. Biopolymers, produced by both phytoplankton and bacteria, play an important role in the scavenging of these radionuclides, thus regulating the particulate organic carbon (POC)/isotope ratios in different oceanographic regions, which have been used to calculate POC flux.
To study the binding mechanisms of colloidal organic material to these radionuclides, marine colloids isolated by cross-flow ultrafiltration from seawater, together with dissolved exopolymeric substances (EPS) produced by laboratory cultured diatoms were collected. By separating, identifying and characterizing radiolabeled organic carriers, the most efficient binding of many of these nuclides likely occurs to acid polysaccharide- and Fe-related protein-containing biomolecules.
To differentiate between uptake to amorphous silica or associated biopolymers, radionuclides were incubated with diatom cultures of Phaeodactylum tricornutum. Partition coefficients (K_(d)) of radionuclides to different fractions of diatom cells, and the role of diatom related biopolymers were examined. More than 50% of selected radionuclides were bound to all-inclusive biopolymer fractions, and logK_(d) values of radioisotopes to whole diatom cells with or without silica frustules were similar, but logK_(d) values for cleaned silica frustules were orders of magnitude lower. Results from isoelectric focusing showed the most efficient binding sites occurring in acid-polysaccharides and iron-binding proteins. As revealed by 2D HSQC NMR spectra, the biopolymers in the HF insoluble fraction were mainly composed of carboxyl-rich, aliphatic-phosphoproteins.
The concentrations of potential carrier phases for radionuclides and their K_(d) values were determined for particles collected by sediment traps deployed at the Oceanic Flux Program site off Bermuda. Chemical considerations, as well as factor analysis and correlations of logK_(d) values with chemical parameters, indicate that hydroxamate-siderophores (HS) are major classes of biopolymers that have a role in binding Po and Pa. MnO_(2) and FeO_(2), whose presence is closely related to that of HS, are also involved in binding of Pa and Po. The carbonate and biogenic silica phases are identified to be important in predicting removal and fractionation of Th and Be in the ocean.

▼ Dysregulation of dopaminergic homeostasis has been established as the primary source of numerous neurological disorders including Parkinson’s and drug addiction. A tonic increase of dopamine (DA) in the nucleus accumbens is required for associating everyday events and behaviors with rewards. Yet many addictive exogenous compounds such as amphetamine (AMPH) and cocaine (
COC) produce a much greater augmentation of synaptic DA levels that are linked to euphoria and a shift in behavior towards drug seeking. The protein responsible for maintaining extracellular levels of DA is the dopamine transporter (DAT). It is primarily located in the perisynaptic area at terminals of pre-synaptic neurons where its main function is to sequester DA from the extracellular space and to transport it back into the cell, a process that is electrogenic. AMPH and
COC directly interact with DAT and alter its ionic currents. Not much is known about the effect of psychostimulant-induced DAT currents on neuronal excitability and neurotransmitter release. We use synthetic chemistry, molecular biology, and biophysics in heterologous expression systems to decipher the actions of drugs of abuse on DAT. Furthermore we demonstrate drug-induced DAT currents can activate Ca
2+ channels associated with dopaminergic excitability. Lastly, we focused on investigating drug effects on excitability in a human midbrain dopaminergic cell line. Understanding how psychostimulants interact with DAT to produce the dysfunctional states of the transporter may facilitate the development of unique therapeutic strategies to treat psychostimulant dependence.

▼ L’objet de ce travail est l’étude de la biocompatibilité in vitro des matériaux à base de COC, afin d’évaluer leur potentiel de biomatériaux implantables.Cette évaluation est réalisée par le suivi de plusieurs paramètres : viabilité cellulaire/cytotoxicité, évaluation des phénomènes de stress oxydant, inflammatoires et hémocompatibilité. Une relation entre ces réponses biologiques et les propriétés physico chimiques des matériaux étudiés a été appréhendée.Les résultats montrent une bonne biocompatibilité des films testés avec un impact significatif de la présence des additifs (anti-oxydant et lubrifiant) sur les paramètres biologiques et physico-chimiques évalués.L’effet simulé du vieillissement biologique de ces matériaux sur leur biocompatibilité et leurs caractères physico-chimiques a également été étudié. Des conditions de pH et d’oxydation extrêmes, ainsi que le contact avec des macrophages pendant 1 mois, ont un effet sur la surface et sur l’interaction des films de COC avec l’environnement biologique sans compromettre leur biocompatbilité. La présence d’additifs a également eu un impact sur ces modifications.En suivant une logique de management du risque, la systématisation de l’ensemble des méthodes développées a permis d’obtenir une approche simplifiée et validée au sein du laboratoire, applicable à l’ensemble des matériaux naturels ou synthétiques susceptibles d’être utilisés dans la fabrication des DM implantables
Abstract : The purpose of this work is the study of the in vitro biocompatibility of COC-based materials in order to evaluate their potential as implantable biomaterials.This evaluation is carried out by monitoring several parameters: cell viability / cytotoxicity, evaluation of oxidative stress, inflammatory reactions and hemocompatibility. A relationship between these biological responses and physicochemical properties of the studied materials has been apprehended.Results show a good biocompatibility of the tested films with a significant impact of the presence of additives (anti-oxidant and lubricant) on the evaluated biological and physicochemical parameters.The simulated effect of biological aging of these materials on their biocompatibility and physico-chemical characteristics has also been studied. Extreme pH and oxidation conditions, as well as contact with macrophages during 1 month, affect the surface and interaction of COC films with the biological environment without compromising their biocompatibility. The presence of additives also had an impact on these changes.Following a risk management logic, the systematization of the developed methods within the laboratory made it possible to obtain a simplified and validated approach, applicable to all natural or synthetic materials that could be used for manufacturing implantable medical devices.

▼ Ce travail de thèse avait pour objectif d’étudier différents vieillissements des copolymères d’éthylène et de norbornène (ENC), utilisés comme conditionnement de produits pharmaceutiques. Grâce à la stratégie analytique adoptée qui a fait appel à différentes techniques de caractérisation, telles que les techniques séparatives comme la chromatographie d’exclusion stérique, la chromatographie liquide haute performance à polarité de phases inversée, les techniques spectrales dont la spectroscopie infra rouge à transformée de Fourier et la spectroscopie UV, les techniques d’analyse thermique à travers l’analyse thermogravimétrie et la calorimétrie différentielle à balayage, puis d’une étude de toxicité des produits de dégradations, nous avons pu mettre en évidence différents types de modifications dans le volume du matériau après vieillissement. La modification principale dans la masse du matériau, observée à la dose réglementaire de stérilisation (25 kGy), est la scission des chaînes du polymère qui s’accompagne de la création de composés de basses masses molaires, donc de migrants potentiels risquant d’influencer la sécurité d’emploi des ENC. Puis pour des doses élevées de rayonnement (150 kGy) et pendant 500h d’exposition UV, on a la réticulation des chaînes.La présence de l’additif (l’antioxydant phénolique l’Irganox 1010®) empêche la création des CBMM après vieillissements. Cependant, en absence d’additif, les vieillissements génèrent de nouveaux CBMM.Toutefois, l’étude de toxicité montre une certaine toxicité à 150 kGy du grade ENC
The aim of this thesis work was to study different ages of copolymers of ethylene and norbornene (ENC), used as packaging of pharmaceutical products. Thanks to the analytical strategy adopted using different characterization techniques, such as separation techniques such as size exclusion chromatography, reverse phase high performance liquid chromatography, spectral techniques including infrared spectroscopy transforming of Fourier and UV spectroscopy, thermal analysis techniques through thermogravimetric analysis and differential scanning calorimetry, and then a toxicity study of degradation products, we were able to highlight different types of modifications in the volume of the material after aging. The main modification in the bulk of the material, observed at the prescribed sterilization dose (25 kGy), is the cleavage of the polymer chains, which is accompanied by the creation of compounds with low molar masses, and therefore potential migrants, which are likely to influence ENC job security. Then for high doses of radiation (150 kGy) and for 500h UV exposure, there is the crosslinking of the chains. The presence of the additive (the phenolic antioxidant Irganox 1010®) prevents the creation of MBMM after aging. However, in the absence of an additive, aging generates new CBMMs. However, the toxicity study shows some toxicity at 150 kGy of the ENC.

La partie bibliographique situe d'abord l'intérêt destechniques électrocinétiques en microsystème. Ensuite, l'état de l'art sur l'utilisation des monolithes dans cestechniques séparatives est présenté en portant une attention particulière à la description du processus dephotopolymérisation. La fonctionnalisation de surface du copolymère d'oléfine cyclique (COC) est ensuitedétaillée afin d'envisager l'ancrage du monolithe aux parois du canal.La partie expérimentale a porté tout d'abord sur le développement et la caractérisation de deux monolithesacrylates dédiés à la chromatographie à polarités des phases inversées. Le travail expérimental s'est ensuiteorienté vers le développement d’un nouveau procédé permettant la synthèse et l’ancrage du monolithe dansles microcanaux en COC, ceci en une seule étape et à partir des photoinitiateurs de type I. Ce protocolepermet l'obtention d'efficacités intéressantes qui restent cependant inférieures à celles obtenues en capillaire.Des adaptations des conditions d'irradiation ont alors été apportées pour la synthèse de ces monolithes àl’intérieur des microsystèmes afin d'obtenir des efficacités de l’ordre de 250 000 plateaux/m. Pour terminer,les domaines d'applications et la longévité du dispositif ont été illustrés par la séparation de différentesfamilles de solutés (PAH, mycotoxines, catécholamines, acides aminés, amines biogènes) et l'analysed'échantillons réels tels que les vins (pour déterminer la teneur en certaines amines biogènes : histamine ettyramine).
This manuscript is dedicated to the synthesis of organic monolithic stationary phases in plasticelectrochromatographic microchips. The bibliography part shows the benefit of electrokinetic techniques formicrosystems. After an overview on the use of monoliths in separation techniques, the manuscript is focusedon the description of photopolymerization process. Finally, a detailed review of the different protocols offunctionnalization of cyclic olefin copolymer (COC) surface to allow anchoring of the monolith to thechannel walls is produced.The experimental study firstly deals with the development and characterization of two acrylate monolithsdedicated to the reversed phase mode in chromatography. Then, the experimental part focus on a new onestepmethod for the simultaneous synthesis and anchoring of organic monolith inside COC microchipchannel, thanks to the use of type I photoinitiators. This protocol improves efficiencies which neverthelessstay below those obtained in capillary. Further, the adjustment of the irradiation conditions allows to reachup to 250 000 plates/m. Un biocapteur est un outil alliant un bioélément, réagissant spécifiquement avec une cible biologique, et un support physique permettant la transduction du signal pour le rendre mesurable. Les travaux de thèse suivants décrivent la préparation d’un transducteur polymérique, le copolymère d’oléfines cycliques (COC), par greffage de sels de diazonium en surface. La modification de l’isatine par couplage pallado-catalysé compose la partie principale des travaux de synthèse organique effectués. Les méthodes de couplages peptidiques en surface et techniques d’accroche des isatines modifiées sur les surfaces des transducteurs (or ou COC) sont également décrites dans ce manuscrit. Pour finir, l’évaluation des effets biologiques des isatines modifiées et des biocapteurs conçus a été effectuée sur Pseudomonas Aeruginosa et sur d’autres bactéries.
Pseudomonas Aeruginosa is an opportunistic pathogen which can adapt its behavior to the present molecules in its surrounding. It increases in virulence when it detects natriuretics peptides in its environment. Isatin is a molecule which can block this effect. To determine on which receptor isatin acts on, the conception of a biosensor have been conducted. A biosensor is a tool combining a bioelement, reacting specifically with a biological target, and a physical support allowing the transduction of the signal to make it measurable. The following thesis describes the preparation of a polymeric transducer, the cyclic olefin copolymer (COC), by diazonium salts surface grafting. Isatin modification by palladium-catalysed coupling reaction represents the major part of the organic synthesis carried out. Surface peptide coupling methods and techniques to link the modified isatins to the surfaces of transducers (gold or COC) are also described in this dissertation. To finish, biological effects of the modified isatins and designed biosensors have been evaluated on Pseudomonas Aeruginosa and others bacteria.

▼ Nous avons développé un module de detection de bactérie base sur le suivi électrochimique d’une PCR. Le système a été realisé en Copolymère d’Oléfin Cyclique (
COC). Il s’agit ici de présenter une nouvelle approche pour concevoir des systèmes de despistage d’infection nosocomiale en quelques heures. INTRODUCTION Avec le développement des flux migratoires de population et l’apparition de résistance aux antibiotiques, les infections nosocomiales sont devenues un enjeu de santé majeur. Les besoins en systèmes de diagnostique accessibles financièrement augmentent considérablement. L’approche qui consiste à intégrer entièrement une chaîne d’analyse commence à avoir les faveurs des systèmes de santé et en particulier de l’Organisation Mondiale de la Santé (OMS). Une compagnie nord américaine Cepheid a par ailleurs déjà développé un système automatisé de détection de germes basé sur l’identification de l’ADN par PCR. Cependant le coût élevé des technologies ralentit sa démocratisation malgré un apport considérable pour les tests cliniques de routine. Ici nous nous proposons une stratégie d’analyse qui permet de maîtriser les drastiquement les coûts de consommable. Tout d’abord tout le système est réalisé en
COC, un thermoplastique qui peut être façonné à très grande échelle pour quelques dizines de centimes l’unité. [1]. Ensuite le système de détection, implique un suivi électrochimique qui remplace avantageusement les plateformes optiques traditionnellement coûteuses. Si la première cible de ce système est le dépistage de maladie nosocomiale, il apparaît aussi qu’il est transposable à d’autres domaines où la détection basée sur une PCR peut apporter des solutions. THEORIE L’échantillon, un écouvillon nasal est prélevé sur le patient. Ensuite les bactéries sont lysées grâce à l’utilisation de détergent. Une fois les parois bactériennes détruites, l’ADN est extrait sur phase solide par l’intermédiaire de billes magnétiques de type « charge switch » assemblées en colonnes [2]. Après lavage l’ADN est élué, puis amplifié par PCR en temps réel. Durant cette phase l’amplification de l’ADN est suivie grâce à l’utilisation d’un intercalant redox. Ce principe de détection fait l’objet d’un brevet [3] et nous présentons ici sa première implémentation dans un système de type laboratoire sur puce. Le complexe redox qui sert d’intercalant est détecté par voltammétrie à onde carrée (en anglais : Square Wave Voltammetry SQWV). Ce complexe interagit avec l’ADN double brin par intercalation pendant la phase d’élongation, ainsi il n’est plus disponible pour l’échange d’électrons avec les électrodes de détection. En conséquence, on observe une chute du niveau de courant mesuré pendant la SQWV qui correspond à l’augmentation de la quantité d’ADN. ▼ Les méthodes habituelles de diagnostic du cancer sont invasives et coûteuses et permettent rarement de dépister la maladie à un stade précoce. L'apparition de biomarqueurs de cancer a été mise en évidence à un stade précoce de la maladie, bien avant le développement des symptômes. Ces marqueurs sont encore peu utilisés, notamment du fait de leur présence en faible quantité chez des individus sains, mais aussi de leur manque de spécificité pris individuellement. De nos jours, les analyses cliniques pour le diagnostic ou le suivi de certains paramètres sont effectués par division des prélèvements et mesures séparées de la concentration des différents analytes cliniquement pertinents. Dans ce contexte, le développement de dispositifs permettant la détection simultanée de ces marqueurs dans les fluides biologiques représente un véritable défi. Par ailleurs, la miniaturisation des dispositifs de mesure et leur intégration sur puce ouvrent la voie vers des unités d'analyse automatisables, transportables au chevet des patients et répondant aux besoins des milieux hospitaliers en termes d'analyses rapides, sur de faibles volumes d'échantillons, à bas coût et haut débit, sensibles et spécifiques. L’objectif de cette thèse est de développer des méthodes permettant l'immobilisation localisée de ligands sélectifs basés sur la reconnaissance moléculaire (aptamères) sur différents matériaux de microsystèmes pour la détection simultanée de plusieurs biomarqueurs. Les aptamères sont une nouvelle classe de molécules d'ADN/ARN synthétiques, reconnus pour leur haute affinité et leur excellente spécificité pour une cible sélectionnée. Ils présentent de plus l'intérêt d'être manipulables à température ambiante, et facilement regénérable après dénaturation. Malgré ces avantages, très peu de publications portent sur les technologies à base d'aptamères pour l'extraction sélective et l'analyse de traces de biomarqueurs en dispositif microfluidique. La création de telles zones de confinement permet d'assurer la surconcentration des analytes cibles pour une meilleure performance de la détection.
Common methods of cancer diagnosis are invasive, expensive and rarely allow early cancer diagnosis. The discovery of cancer biomarkers present in the early stage of the disease, well before the development of symptoms make them interesting candidate for early cancer diagnosis. These markers are still not widely used, particularly because of their low concentration in healthy patient samples, but also because of their lack of specificity taken individually. Nowadays, clinical analyses for the diagnosis are performed by dividing samples and separate measurements of the concentration of the various clinically relevant analytes. ▼ After 40 years of research, in vitro systems for mammalian embryo production produce lower quality embryos than those derived from in vivo sources. Recent reports have demonstrated that in vitro bovine oocyte maturation systems benefit from the addition of oocyte secreted factors, specifically Bone Morphogenic Protein 15 (BMP15) from heterologous sources. However, known amino acid sequence variation and species-specific patterns of post-translational glycosylation lead us to hypothesize that utilization of bovine-specific oocyte secreted factors would be more beneficial than the observed effects of heterologous factors.
To test this hypothesis, wild type, bovine BMP15 was cloned using reverse transcriptase PCR from RNA obtained from bovine ovarian tissue. For improved detection and purification of the biologically active recombinant protein, a FLAG tag peptide sequence (Asp-Tyr-Lys-Asp-Asp-Asp-Asp-Lys) was incorporated into the wild type BMP15 gene by PCR. This modified protein was cloned into the pCDNA 3 mammalian expression vector. HEK-293 (human embryonic kidney 293) and FBK (fetal bovine kidney) cell lines were transfected via electroporation and then selected to homogeneity.
Collection and purification of rbFL-BMP15 from conditioned medium was accomplished by incubation with anti-FLAG affinity gel and the use of 3X FLAG peptide for elution. Peptides of 15.4 kDa and 17 kDa were noted from the human HEK-293 transfected cell line, while in contrast, bovine FBK cells produced a single 17 kDa protein. Bioactivity and BMP receptor signaling specificity were ascertained using in vitro treatment of HeLa cells and Western blotting for the BMP signaling molecule phosphorylated-SMAD 1/5. Inhibition of this signaling cascade using dorsomorphin, a selective bone morphogenic protein receptor I inhibitor, demonstrated the purified proteins served as BMP15-like agonists.
To examine the impact of our purified, bovine-specific peptides on oocyte maturation, cumulus oocyte complexes were in vitro matured for 24 hours in the presence of 100 ng/ml recombinant human BMP15 or rbFL-BMP15 from human or bovine cell lines. Real time quantitative PCR analysis of BMP15 stimulated genes, PTGS2 and TSG6, revealed statistically significant increases in transcript level for treatment with human BMP15 by a Dunnett’s test (p<0.05). In this report, however, we failed to detect a significant affect of rbFL-BMP15 on the gene expression of in vitro mature cumulus oocyte complexes at 24 hours with 100 ng/ml rbFL-BMP15. Future studies including differing time points and concentrations, along with the addition of GDF9 to form a possible heterodimer should be investigated for the possibility of improving bovine oocyte in vitro maturation.

▼ Ovulation is fundamentally crucial to the reproductive success of all mammals. Despite this fact there remain major knowledge gaps in our understanding of how the Luteinizing Hormone (LH) surge, which initiates ovulation, controls this process. There have been numerous theories regarding this phenomenon, yet the underlying mechanisms involved remain relatively unknown. In this thesis I sought to elucidate mechanisms involved in ovulation, with a particular focus on the role played by the expanded cumulus oocyte complex (
COC). Specifically, I investigate whether the cumulus cells and their associated matrix following expansion could contribute actively to its own extrusion from the ovarian follicle during ovulation. I developed a novel hypothesis whereby the cumulus cells transition to an adhesive, motile and invasive cell phenotype in response to an ovulatory stimulus, hCG an analog of LH. I investigate whether the cumulus cells from expanded COCs are capable of cell adhesion to various extracellular matrices found in the follicle wall, and whether this is dependent upon hormonal stimulation by comparison to cumulus cells from unexpanded COCs, not receiving such stimulation. Further, I investigate whether the cumulus oocyte complex is capable of transitioning to a migratory cell phenotype. I tested this with established methods used in the study of cancer cell metastasis. I determine whether this phenotype is firstly dependent on an ovulatory stimulus, and whether it is cumulus cell specific. I attempt to elucidate the molecular mechanisms involved by investigating expression of the well-characterised CD44 cell migration pathway in COCs, during an ovulation time-course. I then use specific antagonists to this pathway, to inhibit cell migration. The final step in our hypothesis involves the investigation of the invasive capacity of the expanded
COC. I analyse whether the expanded COCs are capable of degrading an extracellular matrix barrier during migration assays, and I compare this ability to characterised invasive and non-invasive breast cancer cell lines. I also investigate possible mechanisms involved in the invasive phenotype by inhibiting the matrix metalloprotease system, proposed to play an important role in the degradation of the follicle wall during follicle rupture, and by examining the Adamts1 null mouse, as Adamts1 is a protease shown to be crucial during ovulation. This thesis demonstrates novel and exciting properties of the cumulus oocyte complex during ovulation; offering new insight into our understanding of this complex process. It shows that the oocyte and its surrounding cumulus cells are not merely a passive entity, as previously thought, but rather may play an active role during this vital reproductive process.

Diabetes Mellitus is no more a condition of the affluence but a fast-rising non-contagious disease of global importance which still remains a leading cause of indisposition and death. Currently about half a billion people are suffering from this condition globally. Research has it that 8 out of 10 adults are living with Diabetes in sub-Saharan countries. Ghana over the years has a prevalence rate of diabetes patients to be 9% making it quite disturbing.
Although there has been an existing care among diabetics and their health providers, there is the need to know the extent of continuity of care between diabetics and their health providers.
Objective: The aim of this study is to determine the extent of continuity (Relational, Longitudinal, Flexible and Team) and satisfaction of care between diabetic patients and their care providers.
Methodology
This study was facility-based cross-sectional study which was conducted among diabetics in in Greater Accra Region. Data was collected via the use of a pre-tested structured questionnaire. Information on socio-demographic characteristics and four dimensions of continuity of care and patients’ satisfaction was obtained using ODK software and exported to Stata 15 for analysis. Means and standard deviations were determined for continuous variables. Pearson chi-square test was used to determine the association between dependent and independent variables. Multiple logistic regression was used to determine the strength of association of factors associated with relational continuity of care. Reported p-values < 0.05. Results
The highest extent of continuity of care was team continuity (mean=0.9) followed by relational and flexibility continuity of care (mean=0.8). Longitudinal continuity of care was the least experienced by patients (mean=0.5). Most patients (98.3%) were satisfied with diabetes care they received from health care providers. Factors associated with high relational continuity of care: high flexible continuity of care and low longitudinal continuity
Conclusion
Experience of continuity of care among diabetics is high excluding longitudinal continuity of care. High relational continuity of care translated into high satisfaction of diabetes care received from health care providers. Good adherence to medication and high team continuity of care positively influenced attainment of high relational continuity of care. Further research is needed in order to understand the extent of longitudinal continuity and its effect on relational continuity

▼ Background: The use of contraceptives has become prevalent among women in Ethiopia. Oral
contraceptive use has been suggested to trigger changes in glucose metabolism, energy
expenditure, blood pressure and body weight, among various population and ethnic groups.
Objective: To elucidate the pattern of fasting blood sugar (FBS), blood pressure (BP) and body
mass index (BMI) among combined oral contraceptive (
COC) pills users, taking into account
other confounding factors including socioeconomic conditions, physical activity and nutritional
status.
Methods: Institution based cross-sectional study design was employed. A total of 110 healthy
women using
COC pills were recruited as cases. Another 110 healthy women not using any
hormonal contraceptives were recruited as matched controls. A study was conducted between
October 2018 and January 2019. Fasting Capillary blood samples were collected from the study
participants for the estimation of FBS level. Systolic and diastolic blood pressures were
measured. Body weight and height were measured to compute BMI. Data obtained was entered
and analyzed using IBM SPSS version 23 software packages.
The results: Fasting blood sugar level in
COC users was significantly increased compared to
controls (88.55±7.89 vs. 86.00±9.85 in users and controls respectively, P=0.036). There was a
significant difference in mean arterial blood pressure (MAP) of
COC users compared to controls
(88.2±8.48 vs. 86.0±6.74 respectively, P=0.039). The mean weight and BMI of
COC users were
increased significantly (P =0.04 and P =0.004, respectively). Changes in mean FBS and weight
of
COC users were independent (P=0.27 and P=0.46) to the duration of use of
COC (3-12, 13-24
and ≥25 months). On the other hand, changes in mean BMI and MAP of
COC users were
dependent on the duration of use (P=0.03 and P=0.000 respectively).
Conclusions: Overall,
COC use appears to cause 2.9% increase in blood glucose level, 2.5%
increase in mean arterial blood pressure and 3.9% increase body mass index among users.

▼ Chain of custody in the forest sector is very dependent on effectively tracking products though the supply chain and manufacturing processes, including sawmilling, drying, planing, and pulp processes. The effectiveness of a tracking system is largely influenced by the characteristics of the process it works within, the nature of the tracking system being used, and increasingly by the penetration of technologically advanced material tracking methods into on-the-ground CoC practices. A variety of CoC systems that exist in the global marketplace accommodate the use of advanced materials tracking systems as a tool for their their implementation. These advances not only offer the potential to reinforce the traceability of products in inventory, but also promote maintenance of their certified status between the various organizations that exist along the wood product supply chain, and ultimately to the end consumer.
In the past, a number of different product tracking methods have been used, all of which suffer certain shortcomings in the challenging environment of the forest industry, and the extremely complex nature of Chain of Custody tracking. This work explores the development of a novel material tracking method using the innate IR signatures of polymeric compounds, varied by compound and concentration to develop millions of potential combinations, and consequently millions of unique identities. The combined variation of multiple IR peak frequencies and magnitudes provide the conceptual basis of a chemical barcode system, named "Chemi-Code", to be explored.
To prove the validity of this concept, a series of polymers were assayed for suitability in such a system by tracking their IR response stability in the presence of solar UV radiation and over time. As well, the feasibility of varying polymer concentration, and subsequently identification of concentrations by DRIFT spectroscopy was explored by constructing response curves between polymer concentration and peak absorbance, and assessing associated error.
Seven polymers were identified and assessed. Only two of these polymers were found to be sufficiently stable for use in the context of the forest industry, and of those two, only one behaved in a manner that would allow peak absorbance to be used as an identifiable chemical variable. Although only one of the seven polymers was suitable for use in the "Chemi-Code" system, the study did demonstrate the validity of the concept by showing that polymers can be exploited for use in such a system.

A usabilidade de próteses avançadas pode ser melhorada através do aumento do controlo motor sobre a prótese e da resposta sensitiva do paciente. Como a topografia da superfície desempenha um papel importante, ao nível da nanoescala, no crescimento e condução dos axónios, o dispositivo nanotopográfico foi construído com o objetivo de estudar a resposta axonal de neurónios periféricos a diferentes tamanhos de nanogradeamentos. O design inovador deste dispositivo incorpora: (1) câmaras microfluídicas para isolamento axonal; (2) canais fisicamente modificados com vários relevos nos nanogradeamentos; e (3) uma abordagem de bifurcação baseada na nanotopografia. A linha celular F11 (neurónios sensitivos nociceptivos) foi escolhida como modelo para analisar a resposta celular no dispositivo nanotopográfico. Materiais e técnicas otimizadas foram usados para assegurar a qualidade do dispositivo: os canais foram moldados em PDMS e os nanogradeamentos impressos na superfície de COC. Em paralelo, colorações e ensaios bioquímicos foram realizados para avaliar as condições ótimas de cultura das F11, nomeadamente os ensaios de ADN, ATP, e LIVE/DEAD.
As potenciais aplicações deste dispositivo nanotopográfico foram apresentadas. A Neurofluídica é um campo em recente desenvolvimento que visa construir dispositivos microfluídicos para a investigação neurobiológica. O dispositivo nanotopográfico tem potencial para ser útil nesta área emergente, fornecendo informação acerca das preferências topográficas de cada tipo de célula. Com base nisto, poderia ser possível obter-se a segregação física de subtipos de neurónios. Em resposta às altas e crescentes taxas de amputações, estes avanços poderiam ser úteis no desenvolvimento de interfaces neuro-protéticas altamente sofisticadas.
The usability of advanced prostheses can be improved by increasing the degree of motor control over the prosthesis and sensitive feedback to the patient. As nanoscale surface topographies play an important role in axonal outgrowth and guidance, a nanotopographic device was constructed for studying the axonal response of peripheral neurons to different nanograting sizes. This innovative device design incorporates: (1) microfluidic chambers for axonal isolation; (2) physically modified channels with several nanograting ridges; and (3) a bifurcating nanotopography-based approach. The F11 cell line (nociceptive sensory neurons) was chosen as model to analyse cell response in the nanotopographic device. Optimal materials and techniques were used to assure the quality of the device: Channels were molded on PDMS and nanogratings were printed on the COC surface. In parallel, staining and biochemical assays were performed to evaluate the optimal F11 culture conditions, such as DNA, ATP, and LIVE/DEAD assays.

▼ It is well established that conditions experienced in utero by the developing fetus can elicit permanent effects on the post natal period. Although not as well understood, a growing body of research also suggests that this can also occur in response to peri-conceptional insult. Glucosamine (GlcN) is a popular dietary supplement that is also used experimentally as a hyperglycaemic mimetic. The work contained in this thesis tests the hypothesis that pre and peri-conceptional exposure to GlcN has adverse effects on reproductive outcomes in the mouse. Preliminary experiments (Chapter 2) confirmed that the inclusion of GlcN into the in vitro maturation (IVM) media used for mouse cumulus oocyte complex (
COC) maturation, reduced oocyte developmental potential. Subsequent experiments (Chapter 3) demonstrated that the inhibition of O-linked glycosylation of unknown proteins reversed the effects of GlcN. It was also shown that GlcN exposure during IVM altered Pentose phosphate pathway (PPP) activity within the oocyte. As predicted, preliminary in vivo experiments performed in Chapter 4 showed that maternal, peri-conceptional GlcN administration compromised fetal development. This was seen by a decreased mean implantation rate and litter size as well as an increase in the proportion of fetal resorptions on gestational day 18 (d18), and provided the impetus to examine the in vivo effects of GlcN exposure more carefully. It was subsequently hypothesized that these adverse effects would be heightened if given to mice with overweight-induced metabolic pathologies. In contrast to Chapter 4 outcomes, GlcN elicited no effects on d18 implantation, resorption or litter size parameters, but did reduce fetal weight. Furthermore, birth defects were higher in mice given GlcN and maintained on a low fat (LF) diet. An additional cohort of mice was allowed to give birth, and offspring were assessed for 16 weeks. There was an unexpectedly high death rate in the offspring of mice maintained on a high fat (HF) diet but not given GlcN, therefore preventing optimally controlled post natal analyses to occur. Of the remaining mice, a number of physiological differences were detected within GlcN-exposed groups. Since the principle difference between mice in Chapters 4 and 5 was maternal age, an addition experiment investigating the effects of peri-conceptional GlcN exposure in 8 week and 16 week old mice was undertaken (Chapter 6). Consistent with previous results, GlcN treatment reduced mean implantation rate and litter size only in 8 week old mice and reduced fetal weigh and length solely in 16 week old mice. Increased birth defects were also detected in the HF group given GlcN. Collectively these results provide important insights into the importance of optimal conditions during the peri-conceptional period to facilitate successful subsequent development. ▼ The aim of this study was to gain an insight into how teachers work practically with the Communicative Oriented Curriculum in English and what factors that could influence the teachers’ methodology. The investigation partially replicated a study by Kırkgöz (2008). The study was conducted at three state elementary schools in Turkey by using multidimensional qualitative research procedures, including classroom observations and interviews. Results demonstrated that all participants showed attributes identified to an Eclectic-Oriented teaching approach placing them in the middle of a continuum from Transmission to Interpretation-Oriented teachers. The results were later compared with the original study displaying both a satisfying and unsatisfying outcome depending on a qualitative or quantitative comparison. Findings also indicated that a holistic perspective must be considered in order to interpret and understand the results.

▼ Micro-/nanoimprint lithography is a high-throughput, high-resolution, and low-cost mass production fabrication process often used for creating microfluidic devices and optical components. In the imprint lithography process, a surface pattern of a stamp is replicated into a material by mechanical contact and three dimensional material displacements. These stamps are exposed to high pressures and temperatures and need to be able to withstand these circumstances in order to be durable. Diamond is potentially the ideal surface material for an imprint lithography stamp, since it has a high hardness, a high thermal conductivity coefficient, a low thermal expansion coefficient, it is chemically inert and highly wear resistant. Up until now, only molds made completely out of diamond have been used for imprint lithography. These molds were fabricated using single crystal or polished Chemical Vapor Deposited (CVD) diamond, which were then micro-structured by focused ion beam, reactive ion etching or e-beam lithography. Unfortunately, the availability of large area, single crystal diamond is very limited, and therefore extremely costly. On the other hand, diamond synthesis by chemical vapor deposition provides the possibility to deposit polycrystalline diamond films on areas up to tens of cm2. However, it is a rather slow process where typical growth rates are about 1 μm/hour, and thus production of full diamond stamps is time consuming and expensive. In this thesis, two new methods for the fabrication of CVD diamond imprint lithography molds have been developed. In the first method, a layer of 0.5 μm CVD diamond is deposited on micro-structured silicon. The second method makes use of porous silicon templates through which diamond can be grown, resulting in micro-structured diamond molds. Since polishing micro-structured diamond layers is not possible, the molds were coated with an anti-adhesion layer in order to facilitate release of the mold after imprinting. Both methods proved to be suited for imprinting into a cyclic olefin copolymer developed by TOPAS (grade 6013). With the first method, imprint dimensions of 1 μm with a depth of 350 nm were realized, and imprint dimensions of 2.5 μm with a depth of 2 μm were realized with the second fabrication method. These new approaches greatly reduce complexity of the fabrication process for durable stamps, and thereby the costs involved in creating imprint lithography stamps.

▼ A portable thermocycler for polymerase chain reaction
(PCR) using an infrared-based heating method has been developed and
applied for the DNA amplification in polymer biochips. In the
thermocycler, an infrared lamp as a heating source and a
thermocouple as a temperature sensor have been adopted. This
experiment has evolved a precise temperature control system without
the use of an on-chip integrated temperature sensor or heater,
which allows us further flexible design of lab-on-a-chips in low
cost. A plastic microchip has added advantage of low cost batch
fabrication techniques, but its PCR has been considered as one of
the difficult tasks due to its low heat transfer coefficient. The
microchip is fabricated using Cyclic-Olefin Copolymer (
COC) and
Polycarbonate (PC) material. To prevent protein adsorption in the
channels, Bovine Serum Albumin (BSA) is used in the reaction
mixture. This method for DNA amplification shows reasonably good
results for PCR in PC chips.

Le cycle global d’analysecomprend généralement plusieurs étapes de séparations chimiques longues, manuelles etdifficiles à mettre en oeuvre en raison de leur confinement en boite à gants. Il apparaîtaujourd’hui nécessaire de proposer des solutions innovantes et viables dans le butd’automatiser ces étapes mais aussi de réduire le volume de déchets radioactifs en fin de cycleanalytique. Une solution est alors la conception de plateformes analytiques miniaturiséesautomatisées et jetables.L’objectif de cette thèse est de concevoir un système miniaturisé alternatif à lapremière étape actuelle d’analyse des combustibles usés séparant, par chromatographied’échange d’ions, l’Uranium et le Plutonium des autres éléments constituant l’échantillon. Cesystème doit permettre à la fois de conserver les performances analytiques du processusactuel, de réduire drastiquement l’exposition des expérimentateurs par l’automatisation, ainsique le volume de déchets produits en fin de cycle analytique. Ainsi, la séparation a étéimplantée sur un microsystème jetable en plastique (COC), au design spécialement adapté àl’automatisation : un lab-on-CD.Le prototype développé intègre une micro-colonne séparative d’échange d’anionsremplie d’un polymère monolithique dont la synthèse in-situ ainsi que la fonctionnalisation desurface ont été optimisées spécifiquement pour la séparation voulue. Le développement duprotocole de séparation adapté à ces micro-colonnes a été réalisé à l’aide d’un outil desimulation de l’élution des différents éléments d’intérêts. Cet outil permet de prévoir lagéométrie de la colonne (section et longueur) afin d’obtenir, en fonction de l’échantillon, desfractions de collecte de l’Uranium et du Plutonium pures.Finalement, le prototype actuel est capable de conduire simultanément 4 séparationsde façon automatisée et permet de réduire à la fois le nombre de manipulations, le tempsd’analyse mais aussi de diviser approximativement par 1000 le volume des déchets liquidesgénérés.
The chemical analysis of spent nuclear fuels is essential to design future nuclear fuelscycle and reprocessing methods but also for waste management. ▼ The advancement in smart technologies for organic conducting polymers as flexible substrates in LEDs, PVs and solid state lighting necessitates the development of ultra-high barrier films to protect the devices from moisture and oxygen. The current encapsulation methodology of using layers of plastics and inorganic oxides has several deficiencies. Alternatively, the use of single layer of polymer nanocomposites is a promising substitute for these inorganic based encapsulation layers. The use of polymer materials have the advantage of flexibility, active electrodes printability and easy to make the devices for large area applications. The nano-fillers with high aspect ratio as nanocomposites ingredient in polymers reinforces its mechanical strength and also acts as a scavenging material for moisture and increases the residence time and/or for the penetrating moisture in the film.
Chapter 1 gives the basic overview in the field of barrier technology films and coatings from polymers and inorganic oxide as either mono/multi layer hermetic encapsulation methods. The understanding of both chemistry and physics behind the moisture permeation and its interaction with the film material was discussed. The inclusion of functional nano-fillers as moisture trapping agents in the film provide better device protection achieved. The methods and instruments to measure such ultra-low permeation within the films are discussed. Finally, the advantage of polymer based nanocomposites for low-permeable films with existing materials are briefly discussed in this chapter.
In this thesis, we employed both thermoplastic and thermoset polymer nanocomposites as encapsulation layer for device sealing. The use of ion-containing polymers (ionomers) as a sealant layer was also studied.
Chapter 2 presents the detailed experimental procedures with materials and methods used in this thesis along with the synthesis methodologies to make films from the polymer.
In chapter 3, we used cyclic olefin copolymer
COC (copolymer of ethylene and norbornene) as an encapsulation layer with silica and layered silicate nano-fillers. The compatibility between hydrophilic silica and hydrophobic
COC was achieved by maleic anhydride grafted PE with anchoring on
COC as a compatibilizer and then silica filler was added to make the nanocomposite films. FTIR spectroscopy confirms the bond formation of silica with
COC/MA-g-PE. The mechanical (tensile and DMA) and thermal studies (DSC) suggested that there is an improvement observed when adding silica/silicate layers in the polymer matrix with increased tensile strength, storage modulus and Tg. The calcium degradation test show enhanced
performance towards moisture impermeation in the film.
Chapter 4 deals with the synthesis of PVB based nanocomposite film with silica/layered silicate as nanofillers in the base matrix with varying degree of acetalization in the film. ▼ The primary objective of this work is to investigate the communication capabilities of short-range millimeter-wave (mm-wave) communication among Network-on-Chip (NoC) based multi-core processors integrated on a substrate board. To address the demand for high-performance multi-chip computing systems, the present work studies the transmission coefficients between the on-chip antennas system for both intra- and inter-chip communication. It addresses techniques for enhancing transmission by using antenna arrays for beamforming. It also explores new and creative solutions to minimize the adverse effects of silicon on electromagnetic wave propagation using artificial magnetic conductors (AMC). The following summarizes the work performed and future work.
Intra- and inter-chip transmission between wireless interconnects implemented as antennas on-chip (AoC), in a wire-bonded chip package are studied 30GHz and 60 GHz. The simulations are performed in ANSYS HFSS, which is based on the finite element method (FEM), to study the transmission and to analyze the electric field distribution. Simulation results have been validated with fabricated antennas at 30 GHz arranged in different orientations on silicon dies that can communicate with inter-chip transmission coefficients ranging from -45dB to -60dB while sustaining bandwidths up to 7GHz. The fabricated antennas show a shift in the resonant frequency to 25GHz. This shift is attributed to the Ground-Signal-Ground (GSG) probes used for measurement and to the Short-Open-Load (SOLT) calibration which has anomalies at millimeter-wave frequencies. Using measurements, a large-scale log-normal channel model is derived which can be used for system-level architecture design. Further, at 60 GHz densely packed multilayer copper wires in NoCs have been modeled to study their impact on the wireless transmission between antennas for both intra- and inter-chip links and are shown to be equivalent to copper sheets. It is seen that the antenna radiation efficiency reduces in the presence of these densely packed wires placed close to the antenna elements. Using this model, the reduction of inter-chip transmission is seen to be about 20dB as compared to a system with no wires. Lastly, the transmission characteristics of the antennas resonating at 60GHz in a flip-chip packaging environment are also presented.

▼ In bovine oocytes, the resumption of meiosis is characterized by the
breakdown of the germinal vesicle (GVBD). When cumulus-oocyte complexes
(COCs) are cultured in-vitro in the presence of gonadotropins, GVBD is characterized
by an initial inhibitory phase, which is followed by an acceleration in the rate of
GVBD. An initial transcriptional event is required for gonadotropin-induced in-vitro
maturation. The objectives of this thesis were: 1) to define the time course required
for transcriptional initiation in bovine cumulus oocyte complexes (COCs); 2) to
determine the pattern of expression for Nr4A1 and Egr1 mRNAs in bovine COCs;
and 3) to reduce the expression of Nr4A1 mRNA expression to determine its effect on
oocyte maturation.
Bovine COCs were cultured in the presence follicle stimulating hormone
(FSH) alone or FSH with the transcriptional inhibitor, 5,6-dichloro-1-B-Dribofuranosylbenzamidazole
(DRB), in order to refine the time course required for
transcription initiation and to determine the pattern of expression for Nr4A1 and Egr1
mRNAs. All experiments contained a control group of COCs that were cultured for
the entire duration in the presence of DRB. By adding DRB at 0, 30, 60, 90, 120,
150, or 180 minutes after the initiation of culture, it was determined that gene
transcription required for GVBD occurs between 0 and 60 minutes after the start of
culture. Analysis of COCs cultured for 0, 30, 60, 90 or 180 minutes demonstrated
that Nr4A1 mRNA levels increased significantly (P<0.05) at 30 minutes after the start
of culture, which is consistent with the time of transcription initiation required for
GVBD. In contrast, Egr1 mRNA levels did not significantly differ throughout the
culture period.
Small interfering RNAs (siRNAs) designed from the sequence for Nr4A1
were used to reduce Nr4A1 mRNA expression and determine the effects of Nr4A1
mRNAs on GVBD in bovine COCs. Expression of Nr4A1 mRNA decreased in
abundance in treatment groups containing siRNAs specific to Nr4A1 (siNr4A1) with
the greatest decrease in expression occurring in the 25nM and 50nM siNr4A1
treatments. As expected, fewer COCs underwent GVBD when cultured in the
presence of DRB at 9 and 20 hours as compared to COCs cultured in FSH alone.
Additionally, no significant differences were observed between the FSH and nonspecific
siRNA (siNS) treatment groups in the proportion of COCs undergoing
GVBD at either 9 or 20 hours of culture. Fewer COCs cultured in the presence of
50nM siNr4A1 underwent GVBD by 9 hours of culture as compared to those cultured
in FSH alone. The percentage of COCs that underwent GVBD did not differ between
the siNr4A1 and FSH control treatments at 20 hours. The expression of Nr4A1
mRNA at 30 minutes after the start of culture did not differ with FSH, siNr4A1, or
siNS treatments.
In summary, gene transcription required for GVBD in bovine COCs occurs
within 0 to 60 minutes of culture. Nr4A1 mRNAs are present in bovine COCs and
these mRNA levels increase significantly after 30 minutes of culture. Furthermore,

▼ Ovulation requires sequential molecular events and structural remodelling in the ovarian follicle for the successful release of a mature oocyte into the oviduct. Critical to this process is progesterone receptor (PGR), a transcription factor highly yet transiently expressed in granulosa cells (GC) of preovulatory follicles and abundant in the oviduct. Progesterone receptor knockout (PRKO) mice are anovulatory, with a specific and complete defect in follicle rupture. Therefore, this model was used to examine the critical molecular and biochemical mechanisms necessary for successful ovulation. Progesterone is known to affect oviductal cells in vitro, but how PGR regulates oviductal structure and function is poorly understood. A systematic evaluation of ovarian and oviductal morphology during the periovulatory period revealed no structural defects in PRKO mice relative to heterozygous (PR+/-) littermates. However, in response to the LH surge/hCG treatment, ovulation only occurred in PR+/- ovaries, with numerous corpora lutea observed and cumulus oocyte complexes (COCs) in oviducts, while PRKO ovaries did not ovulate and showed entrapped COCs within large, luteinising follicles. Transplantation of PRKO ovaries into wild-type mice (PRWT) did not rescue the infertility phenotype. Therefore, although PGR is expressed in other tissues, ovarian PGR is essential for ovulation. Further experiments identified PGR-regulated processes at multiple levels. In whole ovaries 10 h post-hCG, inflammatory genes were disrupted in PRKO mice, including cytokines, endothelial adhesion factors, vasoconstrictors, T-cell antigens, and the prostaglandin synthase, Ptgs2. In GCs and COCs isolated 8 h post-hCG, microarray analyses identified 296 and 44 differentially expressed genes respectively between PRKO and PR+/- samples. Gene ontology analysis revealed associations with the processes of proteolysis, vascular remodelling/angiogenesis, inflammatory responses, cell adhesion, migration and invasion. The latter three processes were characterised in periovulatory COCs using in vitro assays and were shown to be transiently activated, peaking at ovulation then declining dramatically in COCs collected from the oviduct immediately post-ovulation. However, periovulatory PRKO and PR+/- COCs showed similar rates of adhesion, migration and invasion in the presence of collagen I. Upregulation of the chemokine receptor, Cxcr4, by LH/hCG via PGR in both GCs and COCs was validated by RT-PCR and immunohistochemistry. Mitochondrial membrane potential was altered in PRKO oocytes compared to PR+/- and therefore their developmental potential may be reduced. Further, a bioassay measuring retention of prostaglandin (PGE2) within the matrix of expanded COCs suggested that the matrix integrity of PRKO COCs may be compromised. ▼ In this thesis Graphit-iC™, an amorphous carbon coating developed by Teer Coatings Ltd. was modified and deposited onto CoCr and WHMWPE substrates in order to improve the wear properties. It was identified that depositing a hard coating onto soft substrate would cause high stresses and lead to coating delamination. Consequently the polyethylene substrates were ion implanted with nitrogen to reduce the hardness differential at the substrate-coating boundary. The coating was characterised using a pin on disc method in order to determine wear and friction. Hardness and fatigue was characterised using nano-indentation and the coating adhesion was measured using scratch testing. Application of the coatings resulted in a significant reduction in wear. Wear factors as low as 3.65x10¯18m³/Nm were achieved for coated CoCr substrates compared to 3.53x10¯15m³/Nm reported in the literature for uncoated CoCr. The coating resulted in friction coefficients between 0.12 and 0.19 with hardness ranging from 6.65 and 15.63GPa. Similarly coating UHMWPE resulted in a reduction in the wear factor to less than 9.6x10¯17m³/Nm. It was concluded that the deposition of amorphous carbon coatings can improve wear of hip joint prostheses, although consideration must be made for the adhesion of the coating to the substrate so that it does not contribute to an early failure of the device. Improved adhesion can be achieved by reducing the hardness differential between the coating and adhesion, either through softening the coating or by using interlayers.

▼ Background: Exposure of non-sustainable practices put MNCs in bad light, hurting their brand image. Therefore, it is of great importance to ensure supplier sustainability upwards the chain. Vast focus has been put on the first-tier suppliers. However, there are few studies that have investigated the issue of second-tier supplier sustainability, and there are many real life examples of firms’ who failed to ensure second-tier supplier sustainability. Hence, the need for further investigation within the subject. Purpose: The purpose of this study was to investigate how two Scandinavian-based MNCs ensure second-tier supplier sustainability within their supply chain. Methodology: This thesis is conducted with an inductive approach, collecting qualitative data through semi-structured interviews and archival research, at two independent companies. Thus, a multiple case study. Empirical findings were analyzed through a template analysis. Based on the theoretical framework and formulated research questions, categories have been derived, constructing the outline for our empirical findings and analysis. Conclusions: (I) Social and cultural distance are major determinants settling which second-tier suppliers being regarded as critical. Conclusively, these two factors are the major determinants identifying whom to prioritize. Furthermore, IT is recognized as a vital and beneficial tool to utilize when mapping the network of second-tier suppliers. (II) In order to achieve second-tier supplier compliance of CoC, “the grey area approach” is identified as the used approach by investigated companies. Further, long-term relationships with first-tier suppliers are a major key in order to ensure second-tier sustainability in long-term perspective. (III) Last but not least, first-tier supplier network investigation is regarded prior to choosing a first-tier supplier. However, it is nothing decisive since other factors are more important in the process.

▼ La caractérisation juste et reproductible en isotopie et en concentration des radioéléments est l’une des thématiques essentielles des laboratoires d’analyse dans le domaine du nucléaire. Afin de minimiser les temps de manipulation en boite à gants des personnels et la production de déchets radioactifs liés à l’analyse de combustibles nucléaires, il est nécessaire de proposer des solutions efficaces et innovantes. Depuis quelques années, la miniaturisation des systèmes séparatifs constitue l’un des axes de développement majeurs de la chimie analytique et ces microsystèmes constituent certainement une des solutions pour répondre aux exigences de l’analyse nucléaire. Ce travail a pour objectif la conception d’une plateforme analytique miniaturisée et à usage unique, dédiée a la séparation des lanthanides, issus des combustibles usés, en amont de leur analyse par spectrométrie de masse. Destiné à remplacer une étape de séparation chromatographique au centre d’un processus analytique de trois étapes, le nouveau protocole basé sur l’isotachophorèse (ITP) doit satisfaire un cahier des charges précis. Les propriétés de complexation des lanthanides ont d’abord été exploitées afin d’obtenir avec un agent chélatant unique et rigoureusement sélectionné, l’acide 2-hydroxy-2-methylbutyrique (HMBA), la sélectivité intra période nécessaire à leur séparation complète par ITP. Basées sur des modèles théoriques existants, des études complémentaires, notamment des paramètres influençant la résolution, ont permis l’amélioration des performances globales du système ainsi que son dimensionnement. The accurate and reproducible characterization of radioactive solutions in isotope composition and concentration is an essential topic for analytical laboratories in the nuclear field. In order to reduce manipulation time in glove box and production of contaminated wastes, it is necessary to propose innovative and efficient solutions for these analyses. Since few years, microchips are a major field of development in analytical chemistry and those devices could provide a solution which fits the needs of nuclear industry. ▼ L’objectif de ce travail de thèse était d’étudier l’effet de la stérilisation par électrons accélérés sur les copolymères d’oléfines cycliques (COC), utilisés comme conditionnement de produits pharmaceutiques, ainsi que son impact sur les interactions avec des formulations pharmaceutiques. Grâce à la méthodologie analytique adoptée qui a fait appel à différentes techniques de caractérisation, telles que la chromatographie d’exclusion stérique, la chromatographie liquide haute performance à polarité de phases inversée, la spectroscopie infra rouge à transformée de Fourier, la microscopie à force atomique et les mesures d’angles de contact, nous avons pu mettre en évidence différents types de modifications dans le volume et sur la surface du matériau après stérilisation ainsi qu’après vieillissement. La modification principale du dans la masse du matériau, observée à la dose réglementaire de stérilisation (25 kGy), est la scission des chaînes du polymère, qui s’accompagne de la création de composés de faible masse molaire, donc de migrants potentiels risquant d’influencer la sécurité d’emploi des COC. En effet, certains de ces composés ont été retrouvés avec une concentration relativement importante dans les solutions de mise en contact avec les COC stérilisés, et notamment en solution aqueuse. Toutefois, l’étude préliminaire de toxicité a montré l’absence de cytotoxicité des extractibles obtenus à la dose de la stérilisation.Les modifications relatives à la surface des COC radio-stérilisés sont, quant à elles, de deux natures : physique avec une augmentation de la rugosité de surface et chimique avec la formation de produits d’oxydation polaires ; ces deux types de modifications conduisent à l’augmentation de la mouillabilité de surface. Cependant dans certains cas, notamment après vieillissement, ces modifications sont relativement faibles, même à des doses supérieures à celle préconisée pour la stérilisation, ce qui peut être corrélé à l’absence de l’effet de l’irradiation sur le comportement des COC vis-à-vis des solutions médicamenteuses. En effet, aucune variation de la sorption des principes actifs choisis n’a été montrée entre les COC irradiés et non irradiés vieillis.
The aim of this work was to study the effect of electron beam radio-sterilization on cyclo olefins copolymers (COC) used as pharmaceutical storage materials, as well as to investigate its impact on the interaction with pharmaceuticals formulations.
