▼ Blood behaves as a shear-thinning non-Newtonian fluid where its viscosity varies due to both the deformability and aggregation of RBCs with the interaction with macro-molecules in blood plasma. The elevated whole blood viscosity (WBV), which indicates the increased frictional resistance between a moving blood and stationary vessel walls, has been suggested as one of the major determinants or risk factors of atherosclerosis diseases (i.e., cardiovascular diseases, stroke, and peripheral arterial diseases etc.) and microvascular disorders (i.e., diabetic retinopathy, nephrophathy, and neuropathy etc.) by causing both the endothelial injury of vessel walls and poor perfusion at capillaries.In order to investigate the shear-thinning non-Newtonian behavior of blood in regards to the effects of increased wall shear stress and impaired oxygen delivery on various diseases that might be caused by hyperviscosity, the present study was focused on the studies of rheological properties of blood by examining the WBV profiles over a pathologically wide range of shear rates using a scanning capillary tube viscometer (SCTV) and their improvements using high-voltage plasma discharge.Firstly, a new hematocrit-correction model using the Casson model was proposed to correct the measured WBVs of different blood samples with different hematocrits to a standard hematocrit of 45 %, a process which is needed to compare the effect of intrinsic rheological properties or other determinants on blood viscosity for different blood samples. Without the measurement of plasma viscosity, the new model showed about 4 to 6 times more accurate and less deviations than the conventional Matrai's model.Secondly, a new method of measuring the electric conductivity of whole blood was introduced for the purpose of hematocrit determination, demonstrating a simple but accurate hematocrit measurement by employing a low-frequency squarewave voltage signal in a conductance cell, without the usual error associated with the sedimentation of erythrocytes.Thirdly, a new physical treatment method with the application of highvoltage plasma discharges (i.e. DBD and corona discharge) followed by filtration of the coagulated particles was proposed. The results indicated that WBV could be reduced by 9.1 % and 17.7 % for systolic blood viscosity (SBV) and diastolic blood viscosity (DBV), respectively, from the baseline values when DBD-treated blood plasma was filtered prior to mixing with red blood cells. When treated with the corona discharge for 60 pulses, DBV and LDL concentration dropped by 30.1 % and 31.5 %, respectively, from the baseline values.Lastly, a new opaque standard viscosity fluid (SVF) was proposed using maltose with 55 % of concentration to replicate a shear-thinning non-Newtonian behavior of blood for different shear rates. The produced viscosity profiles from three different levels of SVFs provided low-, medium-, and high-standard viscosity fluids that can be used for the performance test of any blood viscometers over a wide range of shear rates. ▼ Heterosexual transmission of HIV-1 via the female genital tract is the leading route of HIV infection in sub-Saharan Africa. Viruses then traffic between the cervical compartment and blood ensuring pervasive infection. Previous studies have however reported the existence of genetically diverse viral populations in various tissue types, each evolving under separate selective pressures within a single individual, though it is still unclear how compartmentalization dynamics change over acute and chronic infection in the absence of ARVs. To better characterize intrahost evolution and the movement of viruses between different anatomical tissue types, statistical and phylogenetic methods were used to reconstruct temporal dynamics between blood plasma and cervico-vaginal lavage (CVL) derived HIV-1 subtype C gp120 envelope sequences. A total of 206 cervical and 253 blood plasma sequences obtained from four treatment naïve women enrolled in the CAPRISA Acute Infection study cohort in South Africa were evaluated for evidence of genotypic and phenotypic differences between viral populations from each tissue type up to 3.6 years post-infection. Evidence for tissue-specific differences in genetic diversity, V-loop length variation, codon-based selection, co-receptor usage, hypermutation, recombination and potential N-linked glycosylation (PNLG) site accumulation were investigated. Of the four participants studied, two anonymously identified as CAP270 and CAP217 showed evidence of infection with a single HIV-1 variant, whereas CAP177 and CAP261 showed evidence of infection by more than one variant. As a result, genetic diversity, PNLGs accumulation and the number of detectable recombination events along the gp120 env region were lowest in the former patients and highest in the latter. Overall, genetic diversity increased over the course of infection in all participants and correlated significantly with viral load measurements from the blood plasma in one of the four participants tested (i.e. CAP177). Employing a structured coalescent model approach, rates of viral migration between anatomical tissue types on time-measured genealogies were also estimated. No persistent evidence for the existence of separate viral populations in the cervix and blood plasma was found in any of the participants and instead, sequences generally clustered together by time point on Bayesian Maximum Clade Credibility (MCC) trees. Clades that were monophyletic by tissue type comprised mostly of low diversity or monotypic sequences from the same time point, consistent with bursts of viral replication. Tissue-specific monophyletic clades also generally contained few sequences and were interspersed among sequences from both tissue-types. Neste trabalho estudou-se a purificação de fator VIII de coagulação (FVIII) e da Proteína C (PC) por cromatografia. Em coluna ANX Sepharose FF como primeira etapa de purificação do plasma permite a eluição do FVIII e da PC com bom fator de purificação, mas não a separação destas. Propomos a separação de FVIII e PC empregando cromatografia de afinidade a metal (IMAC) empregando Cu2+, Ni2+, Zn2+, Co2+ e Fe3+ e dessorção empregando imidazol, cloreto de amônio e variação de pH. Em colunas de Fe3+ e Ni2+ as proteínas praticamente não se ligaram à resina. Em IMAC-Co2+, a PC não é adsorvida pela resina enquanto o FVIII pode ser eluído com imidazol 100 mM. Em IMAC-Cu2+ a PC eluiu com imidazol 10mM e o FVIII com 200mM. Não foi possível eluir as proteínas nem com NH4Cl 1M nem quando o pH foi abaixado até 4,0. Em IMAC-Zn2+ a PC não é adsorvida e o FVIII eluiu com imidazol 200mM ou NH4Cl 1M. Diminuindo-se o pH, a recuperação da atividade do FVIII foi baixa. Concluímos que IMAC-Co2+ apresentou os melhores rendimentos e melhores fatores de purificação para as 2 protéinas.

In this work purification methods for the coagulation factor VIII (FVIII) and Protein C (PC) by chromatography was studied. The use of ANX Sepharose FF column as the first purification step allows the elution of FVIII and PC with good purification factors, but not its separation. We propose the separation of FVIII and PC using immobilized metal ion affinity chromatography (IMAC) using Cu2+, Ni2+, Zn2+, Co2+ and Fe3+ and desorption employing imidazole, ammonium chloride and pH variation. In the columns containing Fe3+ and Ni2+ metal ions, proteins did not adsorb to the resin. In IMAC-Co2+, PC was not adsorbed to the resin, while FVIII could be eluted with 100 mM imidazole. In IMAC-Cu2+ PC could be eluted with 10 mM imidazole and FVIII with 200 mM. It was not possible to elute the proteins from IMAC-Cu2+ column with either 1M NH4Cl or when pH was descreased to 4,0. In IMAC-Zn2+, PC was not adsorbed and FVIII could be eluted with 200 mM imidazole or 1 M NH4Cl. We conclude that IMAC-Co2+ presented the best yield and purification factors for the 2 proteins.

▼ O Óxido Nítrico (NO) faz parte de uma família de radicais livres que está envolvida em várias funções do organismo como controle cardiovascular, homeostase, formação óssea, neurotransmissão e funções imunológicas. Tem sido descrito que o NO atuaria na resposta de defesa do hospedeiro frente à infecção dos tecidos orais. Por outro lado, tem sido descrito também que quantidades excessivas de NO podem contribuir para a destruição tecidual na periodontite. Além destes aspectos muito importantes na boca, o NO é uma das moléculas de maior importância para a saúde humana, pois a sua liberação pelo endotélio dos vasos é um componente fundamental para o controle da pressão arterial. Já foi demonstrado que bactérias comensais da boca seriam responsáveis pela produção de nitritos que, no estômago, seriam convertidos a NO e este absorvido. Como as quantidades de nitrito na saliva são cerca de mil vezes maiores do que aquelas detectadas no sangue total, acredita-se que o nitrito da saliva possa ter repercussão no NO do sangue total e possivelmente possa também ter algum efeito na regulação de eventos sistêmicos, como a própria pressão arterial. O número de trabalhos sobre NO na saliva encontrados na literatura científica de língua inglesa não soma 2 dezenas, e pela importância do assunto, faz-se necessário conhecer em detalhes as concentrações de nitrito nos diferentes tipos de saliva, no sangue total, eritrócitos e plasma em pacientes com doença periodontal e controles, antes e depois do tratamento. As hipóteses deste estudo são: 1- Que as concentrações de nitrito sejam menores na saliva total de indivíduos com doença periodontal e que estas aumentem após o tratamento periodontal; 2- Que haveria o mesmo perfil de variação nas concentrações de nitrito no sangue total, eritróticos e plasma vista na saliva total. No presente estudo avaliaram-se as concentrações de NO na saliva total, saliva sumandibular/sublingual, parotídea, no sangue total, eritrócitos e plasma de pacientes controle e pacientes com doença periodontal crônica, com coletas antes e 3 meses após o tratamento periodontal não cirúrgico. A seguir testou-se se eram significativas as diferenças entre os valores obtidos antes e após o tratamento e foram comparados os perfis dos resultados de nitrito na saliva e sangue total. Foram realizadas as dosagens de NO pelo método de análise de quimiluminescência. Os resultados obtidos mostraram que em todos os parâmetros clínicos houve diferença estatisticamente significante antes e após o tratamento periodontal (DPA e DPD), nos grupos controle antes e depois do tratamento (CA e CD) e nos controles antes e doentes antes do tratamento (CA e DPA).Sangramento a sondagem: (DPA versus DPD) p<0,0001, (CA versus CD) p=0,0005 , (CA versus DPA) p<0,0001. ▼ Visando-se determinar seqüências de níveis de plasma sanguíneo em dietas adicionadas ou não de antibióticos para leitões desmamados aos 21 dias de idade, foram realizados dois experimentos onde foram utilizados, no total, 288 leitões. O delineamento experimental utilizado foi de blocos casualizados, com seis tratamentos e seis repetições, sendo quatro animais por unidade experimental. No primeiro período pós desmame dos 22 aos 28 dias, os níveis de plasma sanguíneo usados foram de 0,0%; 3,0% e 6,0% e os mesmos níveis foram usados no segundo período (dos 28 aos 35 dias), porém, combinados de forma a constituir as seqüências de plasma sangüíneo de 0,0 0,0%; 3,0 0,0%; 3,0 3,0%, 6,0 -0,0%; 6,0 3,0% e 6,0 6,0%. Dos 35 aos 42 dias de idade, os leitões de todos os tratamentos receberam uma mesma dieta, isenta de plasma sanguíneo. No experimento 1, durante o período de 22 a 28 dias verificou-se efeito linear (P<0,05) dos níveis de inclusão de plasma sanguíneo na dieta sobre o consumo de ração médio diário, ganho de peso médio diário e conversão alimentar para os animais que receberam dietas contendo níveis de 6,0% de plasma sanguíneo. Não foi verificado efeito linear (P>0,05) nas seqüencias de níveis de inclusão de plasma sanguíneo na dieta sobre o consumo de ração médio diário, ganho de peso médio diário e conversão alimentar, para o período de 22 a 28 dias e para o período de 22 a 49. Não se observou efeito linear (P>0,05) das sequências de inclusão de plasma sanguíneo sobre a altura de vilosidades intestinais (AV) e sobre a unidade formadora de colônias (UFC,g/ml) dos animais desmamados aos 21 dias de idade. No entanto, foi observado efeito linear (P>0,05) da seqüencia de inclusão de plasma na profundidade das criptas do duodeno. Observou efeito linear (P<0,05) das sequências de inclusão de plasma sanguíneo sobre a unidade formadora de colônias (UFC,g/ml).O nível de inclusão de plasma sanguíneo na dieta de leitões desmamados aos 21 dias de idade é de 6,0% para o período de 22 aos 28 dias de idade dos animais e a seqüência de níveis de plasma sangüíneo para os períodos consecutivos de 22 a 35 dias de idade é de 6,0% e 3,0% em dietas com antibiótico, houve efeito sinérgico do plasma sangüíneo com antibiótico. No experimento dois, onde as dietas foram isentas de antibiótico, para o período de 22 a 28 dias, foi verificado efeito linear (P<0,05) dos níveis de inclusão de plasma sanguíneo na dieta sobre o consumo de ração médio diário, ganho de peso médio diário e conversão alimentar a medida que o plasma sanguíneo foi adicionado na dieta.Não foi observado efeito (P>0,05) das seqüências de níveis de plasma sanguíneo sobre o consumo de ração médio diário, ganho de peso médio diário e conversão alimentar para o período de 22 a 35 dias e para o período de 22a 49 dias. Não se observou efeito (P>0,05) sobre a altura de vilosidades e profundidade de criptas. Dois experimentos foram conduzidos para avaliar diferentes sequências de níveis de plasma sangüíneo nas dietas de leitões desmamados aos 28 dias de idade. No primeiro experimento, foi utilizado delineamento inteiramente casualizado com seis tratamentos e oito repetições de três animais. No segundo experimento, utilizou-se delineamento em blocos ao acaso com seis tratamentos e oito repetições de três animais cada. O peso inicial foi utilizado como critério para constituição dos blocos. Na primeira semana após o desmame, os tratamentos consistiram em três níveis de plasma sangüíneo bovino (0,0; 2,5 e 5,0%). Na segunda semana, foram usados os mesmos níveis de plasma, porém, combinados de forma a constituir as sequências 0,0 - 0,0%; 2,5 - 0,0%; 2,5 - 2,5%, 5,0 - 0,0%; 5,0 - 2,5% e 5,0 5,0%. Os dois experimentos tiveram as mesmas sequências de níveis de plasma, sendo que no primeiro experimento, as dietas foram isentas de antibiótico. No experimento um, os leitões que receberam 2,5% de plasma na dieta, apresentaram maior peso final. O maior nível de plasma (5,0%), aumentou o ganho de peso e o consumo de ração dos animais. Houve aumento na contagem linfocitária e de hematócrito dos leitões que receberam plasma na dieta. Os animais que receberam dietas com plasma sangüíneo apresentaram maior número de Bifidobacterium e Lactobacillus no cólon proximal. No experimento dois, o peso final, o ganho de peso e o consumo de ração foram maiores, nos animais que receberam o maior nível de plasma na dieta. A população bacteriana foi diminuída a medida que se aumentou o nível do alimento. Conclui-se que a sequência de plasma sangüíneo a ser usada em dietas sem antibiótico para leitões, nos períodos de 28 a 35 e 36 a 42 dias de idade, é de 2,5% na primeira semana, com retirada a partir dos 35 dias de idade e de 5,0% na primeira semana, com retirada a partir dos 35 dias, quando se usa antibiótico. Conclui-se, ainda que, existe sinergismo do plasma sangüíneo com o antibiótico.

Two experiments were conducted to evaluate different sequences of levels of blood plasma in diets of pigs weaned at 28 days of age. In the first experiment, we used a randomized completely design with six treatments and eight replications of three animals. In the second experiment, we used a randomized block design with six treatments and eight replicates of three animals each. The initial weight was used as a criterion to form blocks. In the fisrt week after weaning, the treatments consisted of three levels of bovine blood plasma (0.0; 2.5 and 5.0%). In the second week, were used the same plasma levels, however, combined to form sequences 0.0 to 0.0%, 2.5 - 0.0%, 2.5 - 2.5%, 5.0 - 0.0%; 5.0 - 2.5% and 5.0 - 5.0%. Both experiments had the same sequences plasma levels, whereas in the first experiment, diets were free of antibiotic. In the one experiment, the piglets received plasma diet 2.5% showed higher final weight. The highest level of plasma (5.0%), increased weight gain and feed intake of animals. ▼ Transfusing the right blood to the right patient is the intention of blood transfusion in many health care organizations. Blood transfusion in Ethiopia takes place in hospitals, but collecting of blood from donors takes place in the National Blood Bank, which is under the Ethiopia Red Cross Society. However, still hospitals can collect and transfuse for urgent and regional cases. Currently, there is a gap of expertise among health professionals involve in blood transfusion, such as shortage of knowledge, which blood type for whom. Developing knowledge-based system to support professionals involve in blood transfusion helps to solve the problem, and is found to be feasible to develop a knowledge-based system within the area of blood transfusion. In this study, a knowledge-based system prototype is developed for blood transfusion. This system will minimize the errors committed such as, which blood type should be transfused for patients in need of urgent or non-urgent transfusion requirements. The system registers a promising and encouraging performance result which is highly applicable. Target users are keen that the system is a clinching effort hence, valuable support in blood transfusion is expected of the system. Users need to provide patients profile such as name, gender and age-group before proceeding to the next blood product options. For whole blood transfusion, users need to test blood type compatibility, room temperature test and decision of urgent and non-urgent issues. For the other blood products, red blood cell, Platelet and plasma, selecting the blood type of a patient is done, and then the system will generate the appropriate blood type sequentially. The output of this study is applicable to training of newly employed health professionals in blood transfusion and for teaching aid in hematology courses within the field of medicine. This study is done first by problem identification followed by tacit knowledge extraction by using interview and observation. For explicit knowledge elicitation, document analysis such as manuals and books is done. The acquired knowledge from domain experts and document analysis is modeled using hierarchal knowledge representation. Then the knowledge is represented in production rule (If-Then-Action) and the prototype is developed using SWI-prolog editor environment. The system works using the backward inference mechanism. The system is tested and evaluated by users, and promising result was registered. The system can be applicable to blood transfusion, since it registers 83.3% complete knowledge for the task. ▼ Desde los tiempos de Poiseuille, se han desarrollado variadas técnicas para medir la viscosidad de la sangre. Durante las décadas de los 60’s y 70’s con la aparición de los primeros reómetros las propiedades reológicas de la sangre fueron medidas y se determinó su comportamiento dependiente del gradiente de velocidad. Además se observó que posee un comportamiento pseudoplástico, es decir, que a medida que aumenta su velocidad su viscosidad disminuye. Desde un punto de vista médico, la sangre y su plasma sanguíneo son los fluidos más eficaces para la detección de patologías globales. Estas patologías pueden estar relacionadas con su viscosidad, con las proteínas presentes en el plasma o con las propiedades de sus glóbulos rojos, como su agregación, deformabilidad o la capacidad elástica de su membrana celular. En los últimos años, con el nacimiento de la microfluídica a principio de los 90’s, nuevas técnicas para el diagnóstico de enfermedades se han desarrollado. La ventaja del uso de la microfluídica en el diagnóstico de enfermedades viene dada por: el bajo requerimiento de muestra para realizar la detección, su portabilidad, la facilidad de uso y el bajo costo de su fabricación. El objetivo de esta tesis ha sido el estudio de la interfase fluido-aire, por medio del desarrollo de un dispositivo microfluídico y método sencillo que permite obtener la viscosidad tanto de fluidos newtonianos e.g. plasma sanguíneo y sangre con un error no superior al 10%. Además de ser capaces de observar el comportamiento no-Newtoniano de la sangre, y a su vez, relacionar su viscosidad con características específicas de sus células rojas como la agregación y la flexibilidad de su membrana. Los estudios de sangre se realizaron a distintos hematocritos, distintos días desde la extracción de la muestra y muestras de anemia y alfa-talasemia. La tesis ha sido desarrollada principalmente desde un punto de vista experimental y está separada en 2 partes. La primera contempla los resultados obtenidos en el estudio de frentes de fluidos Newtonianos. La segunda parte se centra en los resultados obtenidos para la sangre y su relación con las propiedades de sus células rojas. ▼ Coagulation resulting from contact activation due to blood-material interactions remains a challenge in the use of blood-contacting medical devices. Our group is investigating the protein-surface interactions influencing material-induced blood plasma coagulation with the goal of contributing towards the rational development of biomaterials with improved hemocompatability. Traditional biochemical theory of contact activation imparts specific activating abilities to negatively-charged hydrophilic surface. However, recent studies have indicated a need to revise this view as activation of blood factor XII (FXII), a contact activation protein, is not specific to negatively-charged surfaces. The central hypothesis of this dissertation proposes that all material surfaces are potential activators of the intrinsic coagulation cascade, but protein adsorption moderates the molecular interactions of contact activation. Measurements of FXII activation in buffer solutions in the presence of mixed thiol self-assembled monolayers suggest a minimum of FXII activation for surfaces with wettabilities nearing 45 dyne/cm. From direct measurement of kallikrein activation at test surfaces we observed that contact activation reactions involving FXII, high-molecular weight kininogen, and prekallikrein are not specific to negatively-charged hydrophilic surfaces, lending further support for the role of protein adsorption-competition at procoagulant surfaces. Preliminary investigations into contact activation of blood plasma coagulation patterned thiol-modified surfaces suggest spatial chemical heterogeneity influences blood plasma coagulation. In summary, the studies described in this dissertation underscore the need to view all materials as potential activators of the intrinsic coagulation cascade with protein adsorption moderating interactions of contact activation proteins. Šiame darbe nagrinėjami kietazės ekstrakcijos ir skysčių – skysčių ekstrakcijos metodai, bei lyginami jų efektyvumai ekstrahuojant kodeiną iš kraujo plazmos ir šlapimo. Ekstrakcijų efektyvumai buvo nustatinėjami UV spektrometrijos būdu. Darbo tikslas – nustatyti optimalias mėginio ruošimo toksikologinėje analizėje metodikas, tinkamas kodeino išskyrimui iš biologinių terpių – kraujo plazmos, šlapimo, bei palyginti šių metodikų efektyvumą. įvertinus gautus duomenis, galima teigti, kad ekstrahuojant kodeiną iš šlapimo, skysčių – skysčių ekstrakcijos ir kietafazės ekstrakcijos efektyvumai buvo panašūs. Ekstrahuojant kodeiną iš kraujo plazmos – efektyvesnė pasirodė kietafzė ekstrkcija.

In this master’s thesis are examined solid phase extraction and liquid - liquid extraction methods. Codeine has been extracted by using these methotds, efficiencies of extraction methods were determined by UV spectrophotometry. The aim of this this master’s thesis is to determine the optimal sample preparation methodologies for toxicological analysis, codeine proper isolation of biological media - plasma, urine, and to compare the effectiveness of these methods. After assessment of the data, it can be said that the extraction of codeine from urine, efficiencies were similar extracting by using liquid - liquid extraction and solid phase extraction. Codeine is more efficiently extracted from blood plasma by solid phase extraction method than liquid - liquid extraction method.

▼ Smokers incur a sustained free radical load which may increase their vitamin E requirement. However, in the present study this was not apparent from plasma and red blood cells (RBC) vitamin E concentrations which were similar in both smokers and non-smokers. However, RBC from smokers were more susceptible to hydrogen peroxide-stimulated peroxidation than those from non-smokers (p<0.001). Furthermore, plasma concentrations of lipid peroxides, thiobarbituric acid reactive substances and conjugated dienes were also elevated in smokers compared with the non-smokers (p<0.05). These indices of oxidative stress were markedly decreased (p<0.001) in both the smokers, and non-smokers, following consumption of 280 mg dl- tocopherol acetate/day for ten weeks. Plasma and RBC vitamin E concentration increased substantially following supplementation, but the % increase in vitamin E required to improve resistance to in vitro RBC peroxidation was significantly greater in non-smokers (p<0.01). This may reflect an endogenous adaptive response to oxidant stress in RBC of smokers. Erythrocyte vitamin E concentrations increased in a dose dependent manner during 20 weeks of supplementation with either 70,140,560 or 1050mg d – tocopherol per day. In smokers each dose was associated with a significant decrease in susceptibility of erythrocytes to peroxidation (p<0.001). However, red cells of non-smokers on the 1050mg supplement demonstrated an increased susceptibility to peroxidation (p<0.001). Thus, vitamin E may demonstrate prooxidant activity in non-smokers at high and prolonged intakes. Moreover, prolonged supplementation with d – tocopherol in non-smokers induced a decline in plasma ascorbate concentration (p<0.02) in association with an increasing erythrocyte vitamin E uptake (p<0,001). Both smokers and non-smokers may benefit from increased vitamin E intakes, although their requirements may be very different. However pharmacological doses may not be required since it appears that doses as low as 70mg are equally effective.

▼ Purpose: The purpose of this study is to evaluate collegiate wrestlers and their state of hydration before and after a Division I collegiate wrestling season. Methods: Eight collegiate wrestlers (mean ± SD; 19.88 ± 1.05 yrs) gave informed consent to participate in the investigation. Measurements were obtained during the middle of the season and after the season. Height and body mass, a urine sample, a venous blood sample and a VAS thirst scale were obtained at each visit. Prior research had indicated that all hydration measures may not give the same answer and was a focal point of this study. Therefore, hydration status was evaluated using two different testing criteria, urine specific gravity (USG) [two methods] via refractometry and plasma osmolality using a biochemical assay. Results: Body mass was not significantly (p = 0.05) different between the two testing time points. Measurements for USG, regardless of methods were not significantly different between visits but plasma osmolality was significantly different with test visit 1 at the beginning of the season, producing significantly higher values than after the season. For USG a slight level of hypohydration existed but plasma osmolality indicated a bit higher level. No changes were seen between perceived level of thirst between the two testing time points. The two methods of measuring USG were highly correlated r > 0.95. However, USG and plasma osmolality were not correlated. Conclusion: The primary findings of this study were that significant differences exist from during the season compared to after season for plasma osmolality which is not reflected by USG despite no body mass differences. Finally, USG and the gold standard measure of hydration, plasma osmolality do not track hydration in a similar manner andare not correlated. Thus, consistent with prior research these two measures should not be considered to be identical in their evaluation of hydration status. Advisors/Committee Members: Kraemer, William (Advisor).

▼ Labrus bergylta (ballan wrasse) have recently emerged as a key resource to aquaculture through proven efficacy in controlling infestations of sea lice (Leclercq et al., 2014a). However, due to complex ecology, and a complete lack of sexual dimorphism gender identification endures as a key restriction to optimising broodstock management therefore male selection and establishing optimal sex ratios is difficult (Talbot et al., 2012). L. bergylta, are noted to demonstrate unusually coloured plasma ranging in hue from green to blue with the haem catabolite biliverdin established as the causal pigment in the majority of cases (Abolins, 1961). As most vertebrates excrete biliverdin, or rapidly metabolise it to prevent toxicity, accumulation to such excess is a phenomenon which merits attention. Notably, correlation between plasma biliverdin and gender has been reported in some Labridae. Although patterns vary between species, the abundance or characteristics were such that sexual identity could be established (Gagnon, 2006). Pigment analysis was therefore proposed as a potential sex-marker in L. bergylta. In the initial experimental phase (Chapter 3), the ultimate aim was to isolate and identify the blue pigment from L. bergylta plasma, and to develop a method of quantification. The initial phase confirmed the target pigment was biliverdin IXα by visible spectroscopy, TLC, HPLC, MSMS, and a series of reactions. Following this, a protocol was developed (Chapter 2) to quantify the pigment. This method was applied accross plasma sampled from four geographically distinct wild populations with established biometrics including age, mass, length, gender and external phenotype. Subsequent analysis revealed that although pigment abundance did not vary relative to ontogeny, and there was no difference in concentration between the binary genders, plasma biliverdin was depleted in individuals undergoing sex change. Although this conclusion was complicated by significant biliverdin variation relative to origin and phenotype, which were interrelated based on relative distributions across populations, further analysis of plasma pigment in related species identified that biliverdin accumulation was associated with protogynous species. Considering the anti-oxidant capacity of biliverdin and other potentially relevant functions, this was indicative of association with the tissue remodelling processes which accompany inversion. During Chapter 3 it was noted that the biliverdin appeared tightly bound to a protein moiety. Based on the hypothesis that the pigment was actively managed and accumulated in L. bergylta plasma by this association, the next phase of experiments (Chapter 4) was an exploration of biliverdin and its binding protein in L. bergylta. The experiments revealed plasma biliverdin comigrated with the protein such that it was depleted from solution at the same rate indicating that all of the pigment was associated.
