A leishmaniose � uma doen�a parasit�ria causada por protozo�rios intracelulares do g�nero Leishmania transmitidos por picadas de f�meas de fleb�tomo infetados. A doen�a existe em tr�s tipos: visceral, cut�nea e mucocut�nea. A leishmaniose visceral atinge os �rg�os viscerais e � usualmente fatal no prazo de dois anos caso n�o seja tratada. A leishmaniose cut�nea � a forma mais prevalente, causando �lceras na pele que se podem curar espontaneamente. Na leishmaniose mucocut�nea o parasita invade as membranas mucosas do trato respirat�rio superior causando a sua destrui��o. A distribui��o desta doen�a tem-se expandido e o n�mero de casos reportados tem vindo a aumentar exponencialmente. � prevalente em 98 pa�ses e por ano ocorrem aproximadamente 1.3 milh�es de novos casos: 300 000 s�o viscerais e um milh�o s�o cut�neos ou mucocut�neos. O n�mero de mortes estimado para a leishmaniose cut�nea pode ir de 20 000 a 50 000 anualmente. Esta doen�a afeta maioritariamente as popula��es pobres e � considerada uma doen�a tropical negligenciada. O sistema imunit�rio � um fator importante na elimina��o do parasita. Embora a via cl�ssica do sistema complemento seja ativada por Leishmania, � a via alternativa que amplifica a destrui��o de parasitas. Contudo, o parasita desenvolveu estrat�gias para se evadir � atividade do sistema imunit�rio, encontrando-se descrito um n�mero significativo de fatores de virul�ncia que atuam contornando a resposta imune do hospedeiro. Um desses fatores � a metaloprotease de zinco gp63. O presente estudo tem como objetivo analisar a intera��o do parasita com os eritr�citos humanos, sabendo que uma vez introduzido na derme do hospedeiro o parasita para sobreviver tem de ser rapidamente fagocitado. O estudo dos perfis proteicos totais dos extratos de tr�s esp�cies cut�neas de Leishmania demonstrou a poss�vel presen�a de diversas prote�nas, incluindo uma banda compat�vel com a gp63 que tamb�m � segregada para o meio de cultura, tal como previamente descrito. O estudo tamb�m confirma a atividade enzim�tica da banda compat�vel com a gp63. A atividade desta enzima parece estar diretamente relacionado com a concentra��o utilizada para sensibilizar as membranas. Adicionalmente, foi constatado que a atividade hemol�tica est� dependente da presen�a dos extratos do parasita adjuvada por fatores do complemento. No seu conjunto estes resultados indicam que Leishmania usa as suas enzimas proteol�ticas, sobretudo a abundante gp63 para promover a lise do eritr�cito na presen�a de complemento assegurando o suplemento de ferro, uma vez que estes parasitas n�o s�o capazes de gerar o grupo heme. Este estudo tamb�m contribui para a compreens�o das intera��es entre o hospedeiro e o parasita, podendo vir a ser importante na identifica��o de alvos terap�uticos e profil�ticos, assim como na melhoria das metodologias de diagn�stico.

Thats hard knowledge hás got as objective to represent social beffaviors of teachers and Biologys students. Talking abguf the Caatingas biom, to know whats the real subsidies weve got (possible) to maintain biom to survive it. Beyond, that weve research if our biological science universitiy (UFRPE) curriculum, its talking about that to Licenciatures and Bacheroletes, courses. To obten its objectives, we have got some qualitatives propositions. Collecting some information about it. With twenty pupils and eight biologys theachers, as na interview. We made questionary where we can observe questions about evocation techinic, methodologies, to identify the social representations though that. The conclusions that we have was licenciatures and thechers have center of matter objectives of Caatingas climates and vegetations characteristics. And wheres economics, etinics, and social characters? Thats our biological science studies about it region. Its so superficial tems are approached biom. So that, weve intend to ask new knowledges to reforce our questions about Caatingas phisycal, etinics, economics and socials behaviors thats our suggestions in order to contribuition and conservation to its onhy one biom in the world.

Este estudo teve como objetivo a análise das representações sociais de professores e licenciandos em Biologia, sobre o bioma Caatinga, para verificar se elas contêm subsídios que poderão contribuir para a conservação e sustentabilidade desse bioma. Além disso, analisamos se esses aspectos estão contemplados nos currículos dos cursos de Licenciatura e Bacharelado em Ciências Biológicas da Universidade Federal Rural de Pernambuco. Para atingir os objetivos propostos, adotamos uma abordagem qualitativa, tendo como sujeitos vinte licenciandos e oito professores desses cursos. Utilizamos como referencial metodológico a técnica de evocação, para identificar as representações sociais, através de questionário. A análise dos resultados permitiu verificar que as representações sociais dos professores e licenciandos estão centradas nas características do clima e da vegetação da Caatinga. Faltam os aspectos sociais, econômicos e éticos do bioma Caatinga, que poderiam apontar para a conservação e a sustentabilidade dessa região. Concluímos que os aspectos encontrados nessas representações refletem os conteúdos dos programas curriculares do curso de Licenciatura em Ciências Biológicas, que citam muito superficialmente temas referentes a esse bioma. Dessa forma, sugerimos a reformulação dos programas curriculares das disciplinas e a realização de novas pesquisas sobre as características físicas, biológicas, sociais, econômicas e éticas do bioma Caatinga, a fim contribuir para a conservação e sustentabilidade desse bioma único no mundo.

▼ The unpredictable rainfall patterns increased the demand for early maturing maize cultivars. The two most limiting production constraints in maize are drought and low nitrogen. In most national breeding programs in Southern Africa there are no early maturing testers for use in the early phases of breeding. This study was conducted with a view to validate the suitability of CML509/CML505 as a tester for the identification of early maturing inbred lines with a potential in the development of drought and low nitrogen resistant hybrid cultivars. Fifty inbred lines were crossed to this and other two testers, CML312/CML442 and CML395/CML444, for comparison. The research assessed the relative importance of general and specific combining ability effects (GCA and SCA) for grain yield (GY), days to anthesis (AD), anthesis-silking interval (ASI),plant height (PH), ear-height (EH), ears per plant (EPP), root-lodging (RL) and husk cover (HC), of the fifty inbred lines. The three-way hybrids were evaluated under low nitrogen, drought and optimum conditions.The testers grouped the lines into different heterotic groups with CML509/CML505 proving to be a good early maturing single cross tester for heterotic group A after it managed to group most of the lines that were originally in group A into the same group again. The inbred lines were grouped into heterotic groups by the three testers using the SCA effects for GY. A total of ten lines were grouped into an unidentified group after they exhibited negative SCA effects with a group A and group B tester respectively. General combining ability and SCA analysis showed that additive genetic variance was more important for grain yield and days to anthesis, a proxy parameter for maturity. This has an implication on the breeding strategy in that the parents with good GCA can be crossed and early testing of genotypes becomes more effective and promising hybrids can be identified and selected based on their prediction from GCA effects. The tester, CML505/CML509, showed good GCA effects for most of the traits. The line LF47 had the best GCA effect for GY (0.60 t ha"1) and LF49 had the poorest GCA effect for GY (-0.62 t ha1). The mean days to anthesis of the crosses with CML509/CML505 were 61.8 days and mean grain yield was 8.9 t ha"1 under, optimum conditions. CML509/CML505 also showed fairly good heterosis with most of the lines under both drought and low nitrogen conditions.

Sinais de eletrocardiograma (ECG) podem ser utilizados tanto para acompanhamento quanto diagn?stico de doen?as card?acas, o que depende de infraestruturas de armazenamento e transmiss?o. Entretanto, como mem?ria e banda de transmiss?o s?o recursos finitos, torna-se necess?rio comprimir dados de ECG. Al?m disso, com rela??o ao monitoramento ? dist?ncia, espera-se que o hardware utilizado seja simples e barato. O presente trabalho ataca os problemas mencionados e prop?e uma metodologia de compress?o de sinais de ECG, onde experimentos com uma implementa??o desse paradigma mostraram que tal abordagem ? poss?vel e tamb?m eficaz, desde que alinhada ?s condi??es de cada caso espec?fico. Este esquema ? baseado em t?cnicas de codifica??o distribu?da de sinais, cuja principal caracter?stica consiste em realizar a maior parte da explora??o estat?stica desse tipo de processo em decodificadores e assim permitir um processo de codifica??o mais simples, o que tem o potencial de facilitar a sua implementa??o em dispositivos embarcados, como o Holter de ECG.

Electrocardiographic (ECG) signals can be used for both disease monitoring and diagnosis, which is also dependent on storage and transmission infrastructures. Nonetheless, as memory and transmission bandwidth are finite resources, it is necessary to compress ECG data. Additionally, with respect to remote monitoring, it is interesting that the target hardware be simple and affordable. The present work tackles the mentioned problems and proposes a ECG compression methodology, where experiments with a real implementation showed that such an approach is feasible and also effective, as long as aligned with an application?s specific conditions. This scheme is based on distributed source coding techniques, whose main feature is to perform most of the associated statistical exploration in decoders, while allowing a simple encoding process, which has the potential to favor its implementation in embedded systems, such as ECG holter devices.

▼ Yield reductions due to competition of weeds with crop plants can be extremely important. Early weed competition can be as important in reducing crop yield as weed competition for the entire season. The objectives of this study were (1) to determine the weed control achieved by narrow row-cropping patterns, (2) to measure the competitive effect achieved with narrow row-cropping patterns, with low photosynthetic capacity crop plants or with high photosynthetic capacity plants, in competition with high photosynthetic capacity weeds, (3) to determine at what time weed control becomes important and (4) to determine if the time at which competition begins could be determined by change in weight, sugar content, and/or nitrogen content of the commercial crop. Also, two greenhouse studies were made to evaluate the differential uptake of nitrogen by two competiting species, corn (Zea mays L.) and pigweed (Amaranthus rectroflexus L.), and to evaluate the effect of competition for soil and light between two broadleaf species, snap beans and pigweed. Yields of bush snap beans (Phaseolus vulgaris L.), corn (Zea mays L.), and onion (Allium cepa L.) were all decreased significantly by the presence of weed competition throughout the entire growing season. Reduction in total area occupied by an individual crop plant resulted in decreased effects on vegetative and reproductive parts due to weed competition and increased the effect of crop plant-to-plant competition. Early weed control was important in all crops. In 1969, yield reductions in corn and beans allowed, to compete with weeds for a period of five to six weeks after crop emergence were equal to those from plots with weed competition for the entire season. In 1970, corn required two weeks of cultivation after emergence and snap beans required three weeks of cultivation after emergence to decrease losses due to weed competition. Weed weight yields were significantly reduced in snap beans at the 5 x 5 inch spacing when compared to the 15 x 5 and 35 x 5 inch spacings. As measured by leaf fresh weight, competition was determined to begin between snap beans and redroot pigweed at 36 to 44 days after crop emergence at crop plant spacings of 35 x 5 and 15 x 5 inches. Significant weed competition did not develop after 51 days at crop plant spacings of 5 x 5 inches. Competition between corn and redroot developed 41 days after emergence for all plant spacings. This study indicated the time at which competition between crop and weeds could be determined by leaf fresh weight measurements or by measuring the leaf area of the crop plants. At a fertilizer rate of 77-101-64 pounds of N-P-K per acre, corn made greater gains in total green weight than did pigweed in the greenhouse. However, with the further addition of 300 pounds of ammonium nitrate per acre, pigweed continued to make gains while corn growth was unaffected. ▼ Corn (Zea mays L.) has been a large part of American agriculture dating back to 1000 BC when flint corn arrived from Mexico, followed by dent corn in 1500 AD. Over the years, the improvement in these cultivars with better breeding methods and genetics in combination with crop management factors has led to ever-greater corn yields. These crop management factors include: hybrid, planting population, nitrogen (N) fertility, additional nutrient fertility, and foliar protection; and used together can increase grain yield by more than 100 bushels/acre. Nitrogen has the largest impact on corn productivity and one of the biggest decisions farmers make every year is determining their N fertility program. For these reasons, the objective of this research was to determine how N management practices can be implemented to improve corn productivity, and the thesis focused on two research areas: Utilizing Sub-Surface Drip Irrigation to Supply In-Season N and its Effect on Corn Productivity Nitrogen applied preplant helped to maximize the yield potential and increase kernel number by promoting a strong start to the growing season. Supplying N in-season through fertigation further increased corn productivity by promoting N accumulation and helped extend the growing season by maintaining leaf photosynthetic activity. The combination of preplant N and in-season N led to the greatest yields among the treatments by not only increasing kernel number, but also by producing heavier kernels as a result of limiting crop stress and supplying the mineral nutrition to the crop as needed. Combining Hydra-Hume with Side-Dress N Applications to Improve N use and Productivity of Corn Split applications of N, in addition to the different methods of split application (Y-drop or coulter) were compared to applying all the N fertilizer preplant to determine which method or timing had the greatest impact on corn productivity. The split applications of N tended to yield slightly higher than when all N was applied preplant. However, the greatest yields were achieved by split-applying N with a V6 side-dress coulter of 28% urea ammonium nitrate (UAN) down the center of the row in combination with the liquid formulation of a humic acid called Hydra-Hume. ▼ Delaying some of the N supplied to the corn crop is considered a way to increase N uptake and yield and to limit N losses, but the length of the delay may increase the risk of yield loss. We conducted a 3-year experiment at Urbana, Illinois in which half of the N (112 kg N ha-1) was injected as UAN at planting and the other half or all (224 kg N ha-1) of the N was dribbled UAN next to the row, at each of eight stages ranging from V3 to R3. In corn following soybean, when half of the N was applied at planting, the other half could be applied as late as R2 without significant yield loss, and application at R3 produced 92% of yield that the treatment with the second application at V3 did. Delaying application of all 224 kg N ha-1 through V15 did not decrease yield significantly, but yield declined by 12, 19 and 37% with application at VT, R2 and R3, respectively. In corn following corn, delaying the second half of the N to R3 produced 96% of the yield that the second application at V3 did, and none of the delayed timings lowered yields significantly. Delaying all the N to V9 did not significantly decrease yield, but yield thereafter decreased as the delay increased, from 11% less at V12 to 42% less at R3. Without N at planting, N needed to be applied by mid-vegetative stages to prevent yield loss, but when half the N was applied at planting, there was surprisingly little yield loss from delaying application until past pollination. SPAD readings showed that the split-N treatments were able to recover their healthy leaf color no matter how late N was applied, but with no N at planting, plants could not regain their full leaf color (or yield) if N was applied later than mid-vegetative stages. Our results indicate that even a lengthy delay in application of N can result in full yields, especially when half of the N is applied early. This application window is shortened if all of the N is delayed, in which case application should be made before plants reach mid-vegetative growth stages. ▼ Maize (Zea mays L.) is one of the most important commercial crops in the world as well as an important model organism for basic research in plant biology. The shoot architecture of maize is primarily determined by apical and axillary meristems, specialized groups of stem cells that are responsible for producing branches, lateral organs, and stems. Thus the maintenance and initiation of meristems can directly affect maize reproductive potential and yield. A major goal of my research thesis was to understand the role of different genetic, hormonal and environmental factors in regulating maize shoot growth to shed light on the molecular mechanisms underlying maize architecture. In the first chapter of my thesis, I contributed to the characterization of the role of auxin signaling in regulating maize architecture by studying two semi-dominant mutants defective in the early stages of reproductive organogenesis, Barren inflorescence1 and Barren inflorescence4 (Bif1 and Bif4). BIF1 and BIF4 encode AUXIN/INDOLE-3-ACETIC ACID (Aux/IAA) proteins, important negative regulators of the auxin signaling pathway. By in situ hybridizations and yeast-2-hybrid screens, I showed that many maize activating AUXIN RESPONSE FACTORs (ARFs) interact with both BIF1 and BIF4 and could potentially play a role in regulating maize inflorescence development. As part of this study, we provided evidence that BARREN STALK1 (BA1), a basic helix-loop-helix (bHLH) transcriptional regulator essential for maize axillary meristem initiation (Gallavotti et al., 2004), is a potential target of auxin signaling. This work has been published in PNAS in 2015 and I was a co-first author (Galli et al., 2015). In the second chapter of my thesis, we provided new insights into the interplay between inflorescence development and mineral nutrition. Boron is a fundamental micronutrient for plant growth. Previous studies in our lab showed that ROTTEN EAR (RTE), a maize boron efflux transporter, is necessary for maize inflorescence development and fertility (Chatterjee et al., 2014). Here we characterized several RTE-like genes in maize, and showed that the close paralogous gene RTE2, which shares a similar expression pattern with RTE, strongly enhances the rte phenotype in boron deficient conditions, resulting in stunted plants with strong vegetative and reproductive defects. This work showed that in soils with poor boron content both transporter proteins, RTE and RTE2, are necessary to support growth and fertility of maize plants. My main contribution to this study, published in Genetics (Chatterjee et al., 2017), was to characterize the expression of different gene family members. In the last chapter of my thesis, my research focused on a novel recessive mutant called needle1 (ndl1). ndl1 is a temperature sensitive mutant with variable phenotypic expressivity, showing several defects in development, the most notable of which is the formation of tassels with reduced number of branches and spikelets. Microorganisms play crucial roles in aquatic environments in determining ecosystemstability and driving the turnover of elements essential to life. Understanding thedistribution and evolution of aquatic microorganisms will help us predict how aquaticecosystems will respond to Global Change, and such understanding can be gained bystudying these processes of the past. In this project, we investigate the evolutionaryrelationship between brackish water bacteria from the Baltic Sea and Caspian Seawith freshwater and marine bacteria, with the goal of understanding how brackishwater bacteria have evolved. 11,276 bacterial metagenome-assembled genomes(MAGs) from seven metagenomic datasets were used to conduct a comparativeanalysis of freshwater, brackish and marine bacteria. When clustering the genomes bypairwise average nucleotide identity (ANI) at the approximate species level (96.5%ANI), the Baltic Sea genomes were more likely to form clusters with the Caspian Seagenomes than with Swedish lakes genomes, even though geographic distancesbetween Swedish lakes and the Baltic Sea are much smaller. Phylogenomic analysisand ancestral state reconstruction showed that approximately half of the brackishMAGs had freshwater ancestors and half had marine ancestors. Phylogeneticdistances were on average shorter to freshwater ancestors, but when subsampling thetree to the same number of freshwater and marine MAG clusters, the distances werenot significantly different. Brackish genomes belonging to Acidimicrobiia,Actinobacteria and Cyanobacteriia tended to originate from freshwater bacteria, whilethose of Alphaproteobacteria and Bacteroidia mainly had evolved from marinebacteria.

▼ The "POWER - PLANT" deals with the rehabilitation of the former Central Power Conversion and Dale Schoeller (Nejedly I and III) in the village Libušín Mine. Will be newly designed for building the power plant technology with progressive fluidized bed boiler to biomass steam turbine with an output of 7 MW and absorption (trigeneration) unit producing heat, cooling and electricity, which also will function as a cooler primary circuits. The source of water for the power plant will not only former mining pit Nejedlý I which is currently flooded drinking water at about 12 ° C. This system uses high efficiency embedded fuel (biomass), which is ultimately required less. A greater proportion of electricity generated and part of the heat will be distributed to the public network. In most areas will be proposed publicly accessible greenhouses of steel-aluminum construction filled ETFE foil forming the heat insulating membrane filled with air. The reason is to create conditions for the cultivation of tropical and subtropical plants. Greenhouses will be connected to the absorption unit power (underground meanders through which water will circulate), the ventilation shaft mine and mine water - therefore it will be possible to manage the conditions of the internal environment of the building without the influence of the season and especially without mounting other technological devices. It will provide for cooperation with the absorption unit for cooling the primary circuit - therefore eliminating the need for cooling towers or fans. The project will build a detached departments of the Faculty of Mechanical Engineering in Prague - Institute of progressive technologies and systems for energy and the Faculty of Agronomy and Natural Resources CULS. The reason is to allow students and scientists actively and in practice mainly participate in the operation and especially the development of the issue. The public in the area besides themselves open to the greenhouses indoor and outdoor thermal swimming. Part of the entrance hall is a bistro and foremost lecture hall. The aim is decentralized botanical-energy complex of buildings that produce electricity, heat, cooling, and biomass, and creating recreational, educational and research conditions.
