The use of alternative explanations for biological phenomena –particularly the ones referring to human existence – is widespread. Focusing on the concept of death, the present doctoral thesis aims to explore natural and supernatural explanations across the life span. The present thesis consists of four successive studies. Each study aims to provide answers to different research questions concerning the use of natural and supernatural explanations of death across the life span as well as the possible relation of different explanatory frameworks with emotional, cognitive and social factors. More specifically, the 1st Study explores natural and supernatural explanations of death in children and adolescents aged 10 to 14 years (N=127), in relation to loneliness and mother – child attachment. The 2nd Study focuses on the use of natural and supernatural explanation in adults aged 25 to 65 years (N=175) in relation to religiosity, existential anxiety, depression, anxiety and stress. The 3rd Study aims to explore informant’s influence on adults’ (aged between 25 and 65 years, N=540) acceptance of natural and supernatural explanations of death. Finally, the 4th Study focuses on the natural and supernatural explanations of death in adults aged 25 and 35 years (N=86) in relation to executive functions. Research results suggest that the use of supernatural explanations is widespread among pre-adolescents, adolescents as well as adults. Regarding the adults using supernatural explanations of death, there seem to be differentiations concerning the functions that they attribute to dead agents. Moreover, supernatural explanations of death seem to co-exist with emotional factors, such as anxiety over mother abandonment, secure and anxious attachment with God, existential anxiety, depression and stress. Furthermore, the level of inhibitory control is related with the use of explanatory frameworks. Finally, the information concerning the informant’s knowledge as well as the level of religiosity exert influence on the judgements about the trustworthiness of natural and supernatural explanations. The existence and frequent use of supernatural explanations of death, despite the acquisition of the biological concept of death, suggest that the acquisition of scientific knowledge isn’t always a sufficient condition that leads to the replacement of alternative explanations with the scientific ones. On the contrary, concerning existential issues, apart from cognitive factors, emotional and social factors are, also, possibly related with the choice of explanatory framework.

▼ This thesis aims to argue for the explanatory autonomy of the biological sciences. According to many philosophers and scientists, the biological sciences do not have their own explanatory autonomy because they either can be reduced to other “hard” sciences such as physics and chemistry, or cannot really explain phenomena since they do not have laws of nature. To maintain the explanatory autonomy of the biological sciences, I first argue against one influential form of reductionism, i.e., explanatory reductionism, by showing that explanation in the biological sciences can be achieved without reduction. Then, I demonstrate that the biological sciences do not have laws of nature. Instead, I suggest that it is usually biological models that do the explanatory work. Given the fact that biological models usually do the explanatory work, we still need a story of how biological models can be explanatory. However, to understand how a biological model can explain phenomena in the world, we first need to understand what the model-world relationship is. To this end, a holistic view of the model-world relationship is developed by looking closely at scientific practice. Based on this, a holistic account of model explanation is proposed. The basic idea behind the holistic account of model explanation is that, for a model to be explanatory, it must answer two kinds of questions: counterfactual dependence questions (corresponding to Woodward’s what-if-things-had-been-different questions, “w-questions” for short) that concern the model itself, and hypothetical questions that concern the relationship between the model and its target system. Furthermore, I suggest that the reason a biological model can answer these two kinds of questions is due to the fact that: (a) a model is a structure, that is, a set of dependence relationships that can be employed to answer w-questions, and that (b) the holistic fit between the model and its target warrants the hypothetical inference from the model to its target and thus helps to answer the second kind of question.

This thesis evaluates and extends work done in the Journal of Cheminformatics article "Virtual screening of bioassay data" by Amanda Schierz (2009) and the datasets it provides. Schierz was able to locate several primary datasets in PubChem, a repository of bioassay data, and their corresponding secondary screening results. Bioassay data is very imbalanced in the sense that only a few substances will show an effect on the target protein and, hence, can possibly be developed into medicines. Schierz experimented with several data mining tools in the software package WEKA using misclassification costs to improve results and found that the optimum misclassification cost was dependent on which data mining tool was used. The purpose of this study is to see if there are other factors that also affect the optimum misclassification cost. The C5.0 data mining tool in IBM SPSS Modeler was used and is similar to the algorithm called MetaCost J48 in WEKA. Using Modeler, C5.0, gave much better results than MetaCost J48 and in three to six minutes instead of the 60 minutes taken by the WEKA algorithm. The optimum misclassification cost was very different. For Modeler it occurred at cost 469 with a true positive rate of 92% and a false positive rate of 4.76%. For MetaCost J48 it occurred at cost 1500 with a true positive rate of 54%. The false positive rate was not given for J48 but the best WEKA algorithm (CSC SMO) had a true positive rate of 76.92% and a false positive rate of 11.91%. Experiments within C5.0 showed that how the parameters were set affected the results. For example, changing the pruning severity from the default of 75% to 25% changed the optimum misclassification cost from 42 with a true positive rate of 85% and a false positive rate of 1.11% to an optimum misclassification cost of 469 with a true positive rate of 92% and a false positive rate of 4.76%. Changing the number of predictors also affected the optimum misclassification cost.

▼ A detailed understanding of the molecular mechanisms whereby molecules of RNA can catalyze important reactions such as phosphoryl transfer is fundamental to biology, and of high significance in the development of new biomedical technology. This thesis describes the testing, application and development of quantum models that advance our understanding of the mechanisms of RNA catalysis. Molecular simulations of catalytic mechanisms of RNA require the use of fast, accurate approximate quantum mechanical (QM) models. These models, however, were not necessarily designed and parameterized for biocatalysis. In order to assess the degree to which commonly used approximate QM models are appropriate for biocatalysis applications, a series of models has been tested against a wide range of data sets, including new datasets particularly relevant for RNA catalysis, and compared with high-level benchmark calculations. Results provide new insight into the strengths and weaknesses of these methods, and help to guide next generation model development. We note that both NDDO and SCC-DFTB based QM models fail dramatically in their ability to adequately describe the conformational landscape of DNA and RNA sugar rings. In order to overcome this problem, an empirical sugar pucker energy term has been introduced via multi-dimensional B-spline interpolation of a potential energy surface correction. The corrected semiempirical models closely reproduce the ab initio puckering profiles as well as the barrier of an RNA transesterification model reaction. In addition, a series of RNA transesterification model reactions with various leaving groups have been studied with density-functional calculations in solution to investigate linear free energy relationships (LFERs) and their connection to transition state structure and bonding. These relations can be used to aid in the interpretation of experimental data for non-catalytic and catalytic mechanisms. A driving force in this research has been the development of software infrastructure for scientific computation, including new interfaces to other computational chemistry software, libraries to retrieve information, convert format and apply potentials, and tools for data analysis and visualization.

▼ The recent years have witnessed growing interest in the power of scientific visualization for simulation-based neuroscience. The research presented in this thesis develops methods to generate physically-realistic visualizations of neocortical models reconstructed by the Blue Brain Project, a pioneering endeavor that uses a simulation-based approach to build large-scale, biologically-detailed digital reconstructions of neocortical microcircuitry. The project has established a domain-specific framework to visualize hybrid representations of these neocortical models, which it uses to evaluate their structure, composition and dynamics. The current framework offers naive, optical emission-only models that oversimplify light-tissue interaction and ignore other computationally expensive phenomena such as absorption, scattering and fluorescence. The methods presented in this dissertation overcome these limitations, making it possible to visualize neocortical models on a physically-plausible basis, taking into account the intrinsic optical properties of cortical tissue and the spectroscopic characteristics of its fluorescent structures. This requires rigorous optical models that accurately simulate light interaction with cortical tissue, and accurate volumetric models of the tissue itself which reflect its optical properties during simulation. We present an efficient framework for creating high fidelity large-scale volumetric models of neocortical microcircuitry that govern the way in which light is distributed in cortical tissue. These models are reconstructed in two steps: first, we build piecewise watertight mesh models of neocortical neurons from their morphological skeletons. Then, we apply solid voxelization to the meshes to build volumetric models. We also present two novel optical models for simulating light interaction with fluorescent structures in low- and highly-scattering volumes. These optical models are integrated into a high level framework that simulates the imaging pipelines of transmitted light brightfield, widefield epi-fluorescence and light-sheet fluorescence microscopes. It is based on the principles of geometric optics and Monte Carlo ray tracing. Afterwards, we exploit this framework to render in silico realistic microscopic images resembling those created by the actual instruments. In conclusion, we generalize the concept of physically-based simulation of brain imaging modalities and review the computational models required to simulate fMRI. The purpose of this thesis is to analyze the scientific realism of Ernan McMullin and explore its Aristotelian roots. In this respect I present here the views of McMullin on the purpose of natural sciences from the perspective of the historical progress made by scientific activity: from prediction and the Aristotelian demonstrative ideal to Theoretical science and explanation. Moreover, I undertake to examine the issue of retroduction, which forms the nucleus of McMullin’s philosophical thought. This has lead me to an in-depth exploration of the views of Aristotle; I have taken into account not only what he writes in his treatises on logic, but also his cognitive system, as manifested mainly in De Anima and in various of his other works. I argue that the threefold concept epagoge-nous-phantasia as elaborated by Aristotle contains the element of a creative, logical movement, the so-called ‘mental leap’ or ephodos, as he describes it, necessary for the mind’s inductive progress towards the grasp of the universal. Ι suggest, then, that precisely this idea of ‘mental leap’ has its counterpart in McMullin’s theory of retroduction, in which the philosopher states that the inference drawn from the observed fact back to the unknown cause presupposes the operation of the imagination. Moreover, I maintain that the retroductive strategy that McMullin adopts has its roots in the Aristotelian cognitive system. Despite the fact that Aristotle did not actually recommend any retroductive strategy, he did offer those conceptual schemata (syllogismos, epagoge, nous, phantasia-phantasma, apagoge, dia ti esti) which laid the foundations for the arguments McMullin has developed in favour of scientific realism. I thus attempt a reconstruction of McMullin’s retroductive strategy, in the belief that it is grounded in the following argument: if we accept that the method of ampliative retroduction is able to justify the theoretical assertions concerning the non-observable, and that the explanatory success of a theory is an element in this ampliative retroduction, then we have to accept that the explanatory success of theories can justify these theoretical assertions. According to McMullin we can, indeed, make this assertion, provided, however, that we shift the question from truth to reality, i.e. to the field of the ontology of the non-observable.

Στο πλαίσιο αυτό σταχυολογούμε τις θέσεις του E. McMullin για τον σκοπό των φυσικών επιστημών υπό το πρίσμα της ιστορικής πορείας που διαγράφει το επιστημονικό εγχείρημα: από την πρόβλεψη και το Αριστοτελικό αποδεικτικό ιδεώδες έως τη Θεωρητική (Θ-) επιστήμη και την εξήγηση. ▼ The rise in money associated with intercollegiate athletics has created a high-pressure environment for both coaches and players. Today’s intercollegiate coaches and players compete in a situation, whereby the sole value of success is the team’s won-loss record. To date, the coaching research has been found to contain a plethora of studies investigating various characteristics of the coach/player relationship (Chelladuai, 1984; Horne & Carron, 1985; Gould, 1988; Black & Weiss, 1992; Coté, Salmela, Trudel, Baria, & Russell, 1995; Bloom, Durand-Bush, & Salmela, 1997). However, there is a dearth of research investigating the effects of explanatory style on coaching techniques. The current study investigated the influence explanatory style (the manner in which an individual explains his/her surroundings, experiences, or outlook towards the world around him/her) has on the coaching techniques of Division I intercollegiate golf coaches. The most common terms associated with the classification of an individual’s explanatory style are optimism and pessimism (Seligman, 1991). The present study used a mixed-model methodology. Individual optimism and pessimism levels were obtained from Division I Golf coaches (N=8) and golfers (N=40) during a three-day Division I intercollegiate golf tournament, through the administration of the Life Orientation Test-Revised (LOT-R) (Scheier, Carver, & Bridges, 1994). Qualitative data were obtained through standardized open-ended interviews with five of the eight coaches based on their LOT-R scores. The researcher incorporated an interpretational qualitative analysis of the interview data. The results from the quantitative portion of the study were compared and contrasted (triangulation) with the results from the qualitative portion of the study. These results were used to investigate the effects of optimism and pessimism on the coaching techniques of intercollegiate golfers. The results of this mixed-model research design indicated a weak positive relationship between the players’ self-reported explanatory style and the coaches’ perceived players’ explanatory style (r = .32, p< .05). A weak positive relationship was also found between the coaches’ self-reported explanatory style and the players’ perceived coaches’ explanatory style (r = .37, p < .05). Further qualitative analysis indicated that coaches implement the technique of redirecting their less optimistic player’s thought and self-talk processes after a less successful round of competitive golf. In addition, the coaches believed their players might lack a realistic understanding of their own explanatory style. Lastly, the results indicated the coaches attempt to engage in self-awareness training techniques as a means of changing pessimistic explanatory style. Advisors/Committee Members: Billy Hawkins.

▼ O trabalho analisou, com base em indicadores bibliométricos, a internacionalização da produção científica em Ciências Biológicas da Universidade Federal do Rio Grande do Sul (UFRGS) publicada na forma de artigos no período de 2000 a 2011 em periódicos indexados no Science Citation Index da Web of Science (WoS). A internacionalização foi examinada sob três dimensões principais: difusão, colaboração e impacto internacional. Fazem parte da análise os artigos indexados em 33 categorias de assunto da WoS selecionadas para a pesquisa e que contêm pelo menos um autor vinculado à UFRGS no campo Address dos registros bibliográficos. Os 5.168 artigos analisados revelam a atividade e o crescimento significativo da produção científica no período, com prevalência de algumas áreas e de autores a elas associados. Os indicadores de difusão internacional demonstram forte preferência dos autores vinculados às Ciências Biológicas da UFRGS pelos periódicos estrangeiros, que representam 96,5% dos títulos utilizados para publicação e procedem especialmente dos EUA, da Inglaterra e da Holanda. O uso do idioma inglês foi elevado e ocorreu em 94,8% das publicações. A publicação dos artigos ocorreu em 37 países, incluindo o Brasil, e a proporção de artigos veiculados em periódicos estrangeiros foi de 81,9%. Os indicadores de colaboração internacional revelam a predominância da coautoria, registrada em 99,2% das publicações. A colaboração internacional ocorreu em 1.158 artigos, que correspondem a 22,4% do total, mostrando-se inferior aos índices nacionais verificados em estudos anteriores, mas próxima das médias globais de coautoria identificadas nas últimas décadas. Os artigos publicados em colaboração internacional foram produzidos com autores de 84 países, além do Brasil. EUA e Argentina foram os principais países colaboradores, seguidos de Alemanha, França e Reino Unido. As principais instituições estrangeiras colaboradoras foram Universidad de Buenos Aires, University of California, Harvard University, University of North Carolina e University of Texas. Em relação ao nível de colaboração, prevalece a colaboração bilateral, embora a colaboração multilateral também indique a participação das Ciências Biológicas da UFRGS nas pesquisas da chamada Big Science. Os indicadores de impacto internacional revelam um conjunto significativo de documentos que citaram a produção científica em Ciências Biológicas da UFRGS no período de 2000 a 2013, formado prioritariamente por artigos publicados em periódicos estrangeiros, utilizando o idioma inglês. ▼ This thesis focuses on the synthesis of functional materials composed of ligands that possess a methylene hinge – a flexible link between two coordinating groups – and the capacity to form a six-membered chelate ring with transition metals that moderates structural flexibility. A significant portion of this work characterises and explores the properties of a class of functional materials called metal-organic frameworks (MOFs). By utilising methylene-hinged ligands, MOFs are designed that contain intrinsic flexibility and consequently interesting guest exchange and gas adsorption properties. The first methylene-hinged ligand described, di-2-pyrazinylmethane, exhibited an exceptionally rich coordination chemistry with silver(I). Appropriate conditions led to the crystallisation of three isostructural 3-D silver(I) MOFs with large 1-D channels. Although these materials lacked permanent porosity, they possessed the ability to undergo guest-induced structural contraction and expansion. This remarkable solid-state behaviour was related to the chemistry of the guest. The theme of structural flexibility is continued in subsequent chapters which describe the structural flexibility and gas adsorption properties of several MOF materials composed of Cu(II), Co(II), Ni(II), or Mn(II) and the ligand bis(4-(4-carboxyphenyl)-1H-pyrazolyl)methane or bis(4-(4-carboxyphenyl)-3,5-dimethyl-1H-pyrazolyl)methane. Some of the work reported in chapter 3 that deserves particular attention is the exceptional CO₂/N₂ selectivity of a Cu(II) MOF. This material undergoes post-synthetic structural contraction to achieve pore dimensions capable of discriminating between CO₂ and N₂ based on their respective kinetic diameters. Another noteworthy material is described in chapter 4; a flexible Mn(II) MOF capable of quantitative post-synthetic metal binding with preservation of single-crystallinity. The framework flexibility of this material allowed elucidation of the primary coordination sphere of the grafted metal ion and subsequent solid-state transformations by X-ray crystallography. A large portion of this thesis demonstrates and emphasises the positive impact of structural flexibility in MOFs as functional host materials. Chapters 5 and 6 describe methylene-hinged compounds from the viewpoint of synthetic precursors to more complex ligands that contain a fused pyrazino[2,3-b]indolizine or indolizino[2,3-b]quinoxaline core. The synthesis, metal complexes, and corresponding solid-state structures of a series of these ligands are described. L’utilisation des éléments et produits du corps humain à des fins scientifiques implique, aux termes du Code de la santé publique, plusieurs types d’opérations tels que la conservation, le transfert et la cession. Ces opérations permettent une circulation entre chercheurs nécessaire à la mise en commun et au partage de leurs connaissances et savoir-faire au bénéfice du développement de nouvelles thérapies. Si l’encadrement juridique de cette utilisation se résume à un contrôle de police administrative, le législateur fait également référence à des opérations évoquant des pratiques contractuelles (transfert et cession). Or, le code civil fait obstacle, par des dispositions qu’il qualifie d’ordre public, à ce que les éléments et produits du corps humain fassent l’objet d’un droit patrimonial. Cette apparente contradiction conduit à s’interroger sur la nature juridique des éléments et produits détachés du corps humain à des fins scientifiques, éléments et produits qualifiés dans le domaine de la recherche de « matériel biologique humain ». L’emploi de cette notion incite à clarifier le régime juridique applicable en vue à la fois de renforcer la protection de la personne « prélevée » et de garantir la sécurité juridique de l’utilisation du matériel biologique humain.

According to the Public Health Code, the use in a scientific purpose of human body components and products implies many operations such as storage, transferring and selling. These operations enable a movement of the components and products, between researchers, which is necessary to the sharing of knowledge and know-how in order to develop new drugs. If the legal framework of this use can be summarized to an administrative control, the lawmaker refers as well to some operations like contractual practices such as transfer agreement. Nevertheless, the Civil Code obstruct, by public policy rules, the components and products to be subject to a right of ownership. This apparent contradiction led to wonder about the legal nature of the components and products detached from the human body for a scientific use. Such components or products are termed in the research field as « human biological material ». The use of this notion incite to clarify the legal system applicable in order to both strengthen the defense of the person from whom the human biological material is taken and the use of this human biological material.

▼ Multi-core, multi-processor, and Graphics Processing Unit (GPU) computer architectures pose significant challenges with respect to the efficient exploitation of parallelism for large-scale, scientific computing simulations. For example, a simulation of the human tonsil at the cellular level involves the computation of the motion and interaction of millions of cells over extended periods of time. Also, the simulation of Radiative Heat Transfer (RHT) effects by the Photon Monte Carlo (PMC) method is an extremely computationally demanding problem. The PMC method is example of the Monte Carlo simulation methodâ ¯an approach extensively used in wide of application areas. Although the basic algorithmic framework of these Monte Carlo methods is simple, they can be extremely computationally intensive. Therefore, an efficient parallel realization of these simulations depends on a careful analysis of the nature these problems and the development of an appropriate software framework. The overarching goal of this dissertation is develop and understand what the appropriate parallel programming model should be to exploit these disparate architectures, both from the metric of efficiency, as well as from a software engineering perspective. In this dissertation we examine these issues through a performance study of PathSim2, a software framework for the simulation of large-scale biological systems, using two different parallel architecturesâ ¯distributed and shared memory. First, a message-passing implementation of a multiple germinal center simulation by PathSim2 is developed and analyzed for distributed memory architectures. Second, a germinal center simulation is implemented on shared memory architecture with two parallelization strategies based on Pthreads and OpenMP. Finally, we present work targeting a complete hybrid, parallel computing architecture. With this work we develop and analyze a software framework for generic Monte Carlo simulations implemented on multiple, distributed memory nodes consisting of a multi-core architecture with attached GPUs. This simulation framework is divided into two asynchronous parts: (a) a threaded, GPU-accelerated pseudo-random number generator (or producer), and (b) a multi-threaded Monte Carlo application (or consumer). The advantage of this approach is that this software framework can be directly used within any Monte Carlo application code, without requiring application-specific programming of the GPU. We examine this approach through a performance study of the simulation of RHT effects by the PMC method on a hybrid computing architecture. We present a theoretical analysis of our proposed approach, discuss methods to optimize performance based on this analysis, and compare this analysis to experimental results obtained from simulations run on two different hybrid, parallel computing architectures. ▼ Propylene/propane separation is one of the most challenging separations, currently achieved by energy-intensive cryogenic distillation. Despite the great potentials for energy-efficient membrane-based propylene/propane separation processes, no commercial membranes are available due to the limitations (i.e., low selectivity) of current polymeric materials. Zeolitic imidazolate frameworks (ZIFs) are promising membrane materials primarily due to their well-defined ultra-micropores with controllable surface chemistry along with their relatively high thermal/chemical stabilities. In particular, ZIF-8 with the effective aperture size of ~ 4.0 A has been shown very promising for propylene/propane separation. Despite the extensive research on ZIF-8 membranes, only a few of ZIF-8 membranes have displayed good propylene/propane separation performances presumably due to the challenges of controlling the microstructures of polycrystalline membranes. Since the membrane microstructures are greatly influenced by processing techniques, it is critically important to develop new techniques. In this dissertation, three state-of-the-art ZIF membrane synthesis techniques are developed. The first is a one-step in-situ synthesis technique based on the concept of counter diffusion. The technique enabled us to obtain highly propylene selective ZIF-8 membranes in less than a couple of hours with exceptional mechanical strength. Most importantly, due to the nature of the counter-diffusion concept, the new method offered unique opportunities such as healing defective membranes (i.e., poorly-intergrown) aswell as significantly reducing the consumption of costly ligands and organic solvents. The second is a microwave-assisted seeding technique. Using this new seeding technique, we were able to prepare seeded supports with a high packing density in a couple of minutes, which subsequently grown into highly propylene-selective ZIF-8 membranes with an average propylene/propane selectivity of ~40. The last is a heteroepitaxial growth technique. The first well-intergrown membranes of ZIF-67 (Co-substituted ZIF-8) by heteroepitaxially growing ZIF-67 on ZIF-8 seed layers were reported. The ZIF-67 membranes exhibited impressively high propylene/propane separation capabilities. The presence of a methanol co-solvent in the growth solution was critically important to reproducibly prepare high quality ZIF-67 membranes. Furthermore, when the tertiary growth of ZIF-8 layers was applied to the ZIF-67 membranes, the membranes exhibited unprecedentedly high propylene/propane separation factors of ~ 200 possibly due to enhanced grain boundary structure. ▼ Atualmente, um dos desafios da administração pública é atender de maneira eficiente, rápida e satisfatória as demandas crescentes e diferenciadas advindas da sociedade. Os governos têm sido pressionados a apresentar maior eficiência na aplicação do recurso público, maior efetividade nos resultados esperados dos serviços e programas sociais, além de prestar informações e serviços à sociedade de forma transparente e democrática. Porém, tal meta é impossível de ser alcançada sem um processo de informatização das operações internas nos órgãos governamentais. A necessidade de técnicas e ferramentas para auxiliar o projeto e o desenvolvimento ágil de softwares de maior qualidade e em menor tempo é uma das preocupações da Engenharia de Software. Muitos produtos são desenvolvidos em função de artefatos já especificados e implementados, utilizando técnicas de reutilização. Neste contexto, a técnica Linha de Produtos de Software (Software Product Line - LPS) surge como uma proposta de construção e reutilização sistemática de software baseado em um domínio especifico. O objetivo principal desta pesquisa é a instanciação e a automatiza ção de um processo LPS orientado a família de produtos no domínio Sistemas Web de Apoio à Gestão de Fomento de Projetos (SAGF) utilizados por agências estaduais de fomento a projetos (Fundações de Amparo á Pesquisa - FAP). O processo LPS utilizado é baseado na abordagem PLUS (Product Line UML-Based Software Engineering) e enfatiza o uso de padrões e frameworks na definição de um conjunto de sistemas que compartilham similaridades e variabilidades que satisfazem as necessidades específicas do domínio. Para automatizar o processo de LPS são implementadas e integradas a ferramenta Fênix e o framework Titan em um ambiente de geração de aplicação que permite instanciar e gerar aplicações Web para auxiliar a submissão, gestão, monitoramento e avaliação de propostas eletrônicas de projeto a serem avaliadas pelas FAPs. Para implementação do ambiente são utilizadas tecnologias de software livre Java (JSP, Struts e Hibernate), PHP, SVG (Scalable Vector Graphics) e PostgreSQL. Para testar, avaliar e validar o ambiente de geração de aplicações é realizado um estudo de caso para o Sistema Web da Fundação de Apoio ao Desenvolvimento do Ensino, Ci^encia e Tecnologia do Estado de Mato Grosso do Sul (FUNDECT). A evolução tecnológica fez com que as aplicações web estivessem cada vez mais presentes no mercado e surgissem novos padrões arquiteturais, novos dispositivos e novas experiências de utilização das aplicações. Tudo isto com o propósito de satisfazer as exigências que o mercado e os utilizadores finais impõem. Toda esta evolução potenciou o aparecimento de uma nova versão do HTML, o HTML5, a qual está em constante progresso. Não se pode esperar que o HTML5 esteja totalmente concluído para implementar interfaces com esta tecnologia, podendo assim ser considerado um living standard, porque já existem funcionalidades suficientemente maduras e que inclusivamente já são utilizadas em produtos no mercado. Devido ao facto do HTML ser uma linguagem de marcação, torna-se indispensável associar o HTML ao JavaScript, de modo a poder disponibilizar dinamismo e funcionalidade às páginas web. Existiu por isso, tal como o HTML, uma grande evolução relativa à linguagem, mais propriamente às frameworks JavaScript de desenvolvimento web existentes. Devido ao progresso destas tecnologias, esta dissertação pretende analisar as várias funcionalidades disponibilizadas pela tecnologia HTML5 e as frameworks JavaScript existentes no mercado. Para tal, estas tecnologias serão usadas, de forma experimental, num projeto de desenvolvimento de software na empresa onde decorre este trabalho. Os resultados focam-se em identificar as funcionalidades HTML5 implementadas e apresentar uma comparação entre as frameworks JavaScript em estudo, segundo um conjunto específico de critérios. É do interesse da empresa onde este trabalho foi realizado aplicar as funcionalidades HTML5 e frameworks JavaScript, de modo a identificar as vantagens e desvantagens de cada tecnologia para, num futuro próximo, as aplicar em aplicações web. ▼ The theme of this work is the observation and understanding of the effects of pore functionalization on adsorption properties of stable Metal-Organic Frameworks (MOFs). Over the first two sections of this work, sets of materials with representative pore sizes and functional groups are studied for adsorption properties. Observed trends are used to identify the best pore properties achievable via functionalization for adsorption systems. The third section of this work provides perspective on MOF materials and proposes target pore features for an efficient adsorbent for carbon dioxide capture from flue gas. First, the highly stable UiO-66 series of materials was selected for a pure-component adsorption study. The selectivity and capacity for CO2 can be best enhanced with the smallest, most polar functional group, such as an amino group, but significantly enhance water adsorption. Large, non-polar groups do not yield a completely hydrophobic material, but may be useful for humid gas separations as pore filling with water is inhibited. Next, a breakthrough study was conducted using CO2:CH4 and CO2:N2 mixtures on a set of stable MOFs. UiO-66-NH2 and UiO-66-DM, where DM=dimethyl, outperform predictions based on published isotherms and have dynamic CO2:CH4 selectivity comparable to zeolite NaY. UiO-66-DM may be a good candidate for further study due to the combination of partial hydrophobicity and good selectivity. Finally, by combining a review of literature with observations made in this work, a perspective on MOFs as efficient humid gas separation materials is provided. The presence of water vapor prevents use of current high performance adsorbents, but several MOF pore features show promise for these separations. The designable nature of MOFs allows for targeted design of size-matched pores and single-molecule traps which can selectively or cooperatively adsorb CO2 in the presence of water. Also, many MOF materials would be well suited for advanced pressure swing adsorption cycles and engineered sorbents, which enables greater material utilization and system efficiency.
