口腔感染症から分離される細菌には,sucrose非依存性に菌体外マトリックスを産生する株が存在する.これらの細菌の産生する菌体外マトリックスは貪食抵抗因子として働くだけでなく,菌体を周囲の環境から保護し,疾患の慢性化,難治化に重要な役割を果たすことが分かっている.我々はこれまでに数回の根管治療にもかかわらず,持続的に単一の細菌種が分離される難治性根尖性歯周炎の病巣からRothia mucilaginosa DY-18株(DY-18株)を分離し,この菌株が菌体外マトリックスを産生することで治療に抵抗して病巣で長期に生存することを明らかにしてきた.本研究ではDY-18株のバイオフィルム形成について更に詳細に検討し,その遺伝学的な背景を明らかにすることを目的に,浮遊状態の細胞と,バイオフィルム形成状態の細胞の遺伝子発現をマイクロアレイ分析した. 培養菌液の粘度からEPSの産生量を経時的に測定すると,種菌接種30時間後から42時間後まで粘度が著しく上昇していた.また,種菌接種36時間後の振盪培養した浮遊状態と,静置培養したバイオフィルム形成状態の培養菌液の粘度を比較したところ,振盪培養では粘度上昇が認められず,静置培養時とは菌体外マトリックスの産生量に大きな差があることが示された.そこでDY-18株のゲノム情報を基にマイクロアレイをデザインし,浮遊状態とバイオフィルム形成状態の細胞における遺伝子の発現量を測定した.その結果,バイオフィルム形成状態でDNA polymerase Ⅲ subunit beta,signal transduction histidine kinase,molecular chaperoneをコードする遺伝子が有意に発現上昇していることが明らかになった.これらの遺伝子は,DY-18株のバイオフィルム形成に重要な役割を果たしていると考えられる.
