▼ Douglas-fir utility poles are routinely air seasoned before treatment with chemical preservatives. Basidiomycetous fungi invade these poles and may cause strength loss during air seasoning or later if treatment temperatures are insufficient to kill the decay fungi. The purpose of this study was to develop a rapid test to asses the ability of these fungi to degrade wood. Two tests measuring wood toughness, the first mechanical property affected by decay fungi, were chosen. The breaking radius test measures the radius of curvature a specimen can withstand without failing. The impact bending test measures the amount of energy required to cause failure as a pendulum arm strikes the specimen. Variables affecting these tests such as specimen size, grain angle, moisture content and length of incubation with a suspect decay fungus were investigated. Methodologies were designed to reduce variability and give rapid results with each test. Douglas-fir test specimens (1.7mm x 9.4mm x 5.1cm) were incubated for 4 weeks with basidiomycetes isolated from air seasoning Douglas-fir poles. Sixteen specimens per fungal isolate were sampled oven dry for breaking radius and 12 were sampled above the fiber saturation point for impact bending. Between one and 16 isolates per species were tested. Twenty-three fungal species were ranked according to decay capacity and this information was related to isolation frequency. Two brown rot fungi, Poria placenta and Poria carbonica, degrade wood rapidly and are common in air seasoning Douglas-fir. Several white rot fungi including Haematostereum sanguinolentum and Peniohpora spp. are common in Douglas-fir poles, but much less damaging to the wood. Monokaryons and dikaryons of 15 species were compared by decay capacity and no consistent relationship was found. Isolates of each species were also compared to asses within species variability. Differences were found within species with high decay capacities. Large numbers of basidiomycetes invade air seasoning Douglas-fir poles, but most cause little strength loss. This information can be used in assessing when poles or other wood products may be at risk of strength loss and in need of chemical treatment. Advisors/Committee Members: Corden, Malcolm E. ▼ Os representantes da classe Agaricomycetes (filo Basidiomycota) são caracterizados por desenvolverem basidiomas onde produzem basídios e basidiosporos. Grande parte dos representantes deste grupo degrada componentes da madeira, sendo assim chamados de lignolíticos ou lignocelulolíticos, mas também agem como parasitas obrigatórios ou facultativos e como simbiontes. Para ampliar o conhecimento sobre a diversidade taxonômica e ecológica desse grupo de fungos do sertão de Pernambuco, foram realizadas seis coletas em duas áreas em estágio severo e moderado de desertificação em Cabrobó [Barro Branco (CBB) e Fazenda Mosquito (CFM), respectivamente] e um brejo de altitude em Triunfo [Sítio Carro Quebrado (SCQ)], totalizando 18 expedições. Para avaliar a diversidade e as relações desse grupo de fungos com os hospedeiros nas áreas de estudo, foram utilizados testes de χ2 e ANOSIM. No presente tudo, foram coletados 256 espécimes correspondendo a 47 espécies de Agaricomycetes lignocelulolíticos (12 em CBB, 19 em CFM e 41 em SCQ). Dentre estas espécies, há 31 novos registros para o estado de Pernambuco, região Nordeste, Caatinga de Pernambuco, bioma Caatinga, Brasil e América do Sul. Após 18 visitas a campo, as curvas cumulativas de espécies não se estabilizaram, indicando que mais coletas são necessárias. De acordo com a ANOSIM, há diferença significativa na composição de espécies entre as duas áreas de caatinga e brejo de altitude, assim como para abundância de acordo com o teste de χ2, sendo coletado um maior número de espécimes em SCQ. Entretanto, de acordo com este teste, não há diferença na riqueza entre as áreas. Não há diferença também na composição e na riqueza de espécies entre os períodos de coleta (sazonalidade), de acordo com a ANOSIM e com o teste de χ2, respectivamente. Já a abundância de espécies nessas áreas foi diferente quando utilizado o teste de χ2, sendo coletados mais espécimes na estação seca. A tendência em ocorrer em substratos mortos foi observada para Agaricomycetes lignocelulolíticos em todas as áreas de estudo. Entretanto, em áreas de caatinga sob processos de desertificação, os táxons de Hymenochaetales tendem a acompanhar a disponibilidade de substrato, ocorrendo preferencialmente em árvores vivas. ▼ O presente trabalho investigou a dinâmica da comunidade de fungos poroides em remanescentes de Floresta Atlântica de Pernambuco, baseado na hipótese de que fatores bióticos e abióticos influenciam significativamente seus padrões estruturais. O estudo ecológico foi conduzido em três fragmentos em diferentes estágios de regeneração da RPPN Frei Caneca: Barragem das Moças, Caranha e Serra do Quengo. Em cada fragmento foram estabelecidas unidades amostrais, compostas por três transectos, visitadas durante 12 meses entre 2012 e 2013. Os fungos encontrados ao longo dos transectos foram levados para laboratório, analisados macro e microscopicamente e identificados. Espécies interessantes ou novas para a ciência foram analisadas também com métodos de biologia molecular. As variações na composição, riqueza e abundância das espécies foram investigadas ao longo dos fragmentos e do tempo em relação aos fatores climáticos: “precipitação”, “temperatura” e “umidade relativa do ar” e aos fatores do substrato: “pH”, “dureza”, “umidade relativa do tronco”, “temperatura da madeira”, “volume” e “área superficial”. As diferenças encontradas foram testadas através do teste de qui-quadrado e a ANOSIM, utilizando o índice de Bray-Curtis. A influência do substrato foi testada através da análise de correspondência retificada, utilizando o coeficiente de correlação de Spearman. Além disso, foi calculado o índice de Shannon-Wiener para medir a diversidade dos fragmentos e o coeficiente de correlação de Pearson para medir o grau da correlação entre seus valores. Duas ordens, nove famílias, 45 gêneros e 74 espécies foram encontradas. Destas, 17 foram coletadas pela primeira vez na Floresta Atlântica de Pernambuco, representando novos registros para a América do Sul, Brasil e Nordeste. Os fragmentos diferiram em relação ao tempo de regeneração (idade), tamanho, histórico de perturbações e grau de isolamento, porém não significativamente em relação à composição de espécies da comunidade fúngica. A mata com maior histórico de perturbação foi também a que apresentou maior número de espécies fúngicas. A distribuição dos fungos foi influenciada principalmente pelos fatores “fragmento” e “umidade relativa do ar”. A diversidade de fungos foi diferente entre fragmentos, mas não entre meses. Todavia, a abundância foi maior durante a estação chuvosa. A maioria das espécies ocorreu em troncos de 10 a 30m² e com até 1m³. Contudo, os resultados de qui-quadrado não foram significativos nem em relação à área, nem em relação ao volume. O pH foi a única variável com correlação significativa entre fatores do substrato analisados, . A maioria das espécies ocorreu em pH de 3 a 5, mas a acidez encontrada no substrato pode ter sido decorrente da presença dos basidiomas e, consequentemente, do processo natural de decomposição da madeira. ▼ Lactarius is an important component of the ectomycorrhizal community in cold-dominated arctic and disjunct alpine habitats where it associates primarily with the woody shrubs Betula, Dryas, and Salix. Little is known of the alpine fungi in the central and southern (Montana, Wyoming, Colorado) Rocky Mountain alpine zone (elevation 3,000-3,900 m) of North America. The goal of this study was to examine the genus Lactarius and here at least six species from subgenera Russlularia and Piperites are confirmed above treeline through molecular phylogenetic analyses of ITS and rpb2 DNA in conjunction with detailed morphological examination. All (except two putative new species) appear to have broad intercontinental distributions in North America and Eurasia according to molecular comparison with type material, and collections from Europe, Greenland, Scandinavia, Svalbard, and Alaska. Rocky Mountain alpine collections of L. lanceolatus and the type from Alaska form a well-supported clade paraphyletic with respect to well-supported clades consisting of L. aurantiacus and several North American subalpine taxa. Rocky Mountain alpine collections of L. nanus, L. glyciosmus, L. repraesentaneus, and L. salicis-reticulatae all form well-supported clades with material from European type localities and other arctic-alpine material; although some clades contain nested (L. hysginoides within L. nanus) and possibly cryptic species (L. aff. salicis-reticulatae from Colorado). The well-known arctic-alpine L. pseudouvidus/L. brunneoviolaceus group of violet-staining species appears to be a complex possibly containing additional species. North American material originally described as part of this group, is well-separated phylogenetically and represents a putative new species (L. pallidomarginatus Barge & C.L. Cripps ad int.) so far confined to the Southern Rockies with shrub Salix. The monophyly of the violet-staining section Uvidi is supported. Lactarius lanceolatus, L. nanus, and L. salicis-reticulatae appear largely restricted to arctic-alpine habitats across their broad range, where they associate with dwarf and shrub Salix. Lactarius glyciosmus and L. repraesentaneus are not restricted to arctic-alpine areas and occur with Betula and krummholz Picea (possibly also Salix), respectively in the Rocky Mountains; the latter is also known with Betula in broader parts of its range. ▼ I. Psilocybe cubensis was cultured successfully in two media. Medium A was devised by Catalfomo and Tyler and Medium B was a modification of a medium which has been used for ergot alkaloid production by Claviceps purpurea. Only when the fungus was kept on Sabouraud agar plates.did it subsequently produce psilocybin when transferred to liquid media. A quantitative time-course study of psilocybin production in the two media was carried out. Maximal production appeared on the fifth day. The activities of an acid phosphatase, acting on psilocybin, were measured from mycelia grown in the two media. Enzyme activity from the A culture was very high and a blue color caused by oxidation of psilocin formed in five minutes. The effect of adding L-tryptophan on alkaloid production as well as the fate of tryptophan-C¹⁴ was also investigated. Tryptophan stimulated significantly psilocybin production in the very beginning in the B medium. The degradation of tryptophan was different in the two media. It was converted to kynurenine and anthranilic acid in A medium and to tryptamine in tryptophan added B medium (B' medium). Radioactive D,L-tryptophan side chain labeled, gave labeled psilocin and psilocybin. Potassium deficiency decreased psilocybin production while a potassium supplement had no effect. The fungus did not produce polyacetylenic compounds in the medium but ergosterol was detected as a major acetate derived metabolite when the fungus was kept on MYP agar plates and transferred subsequently to liquid media. Psilocin has very slight antibiotic activity against Candida albicans whereas psilocybin has none. II. Eight species of Lentinus and Lentinellus were investigated for the occurrence of a phenol-O-methyltransferase. Only Lentinus lepideus and Lentinus pbnderbsus showed enzyme activity in both light and dark conditions. The specificity of the enzyme for a number of substrates was also examined. Of six compounds tested, methyl p-coumarate, methyl caffeate and methyl ferulate.served-as substrates. The products of enzymic activity were identified-by radioautography.

The biochemical composition of fungi can vary with climatic conditions and cultivation substrate. Several species have shown biochemical activities, such as Pycnoporus sanguineus, which is highlighted by orange to red color of their basidiocarps. Although the antimicrobial activity verified in various fields such as pharmaceutical, industrial and agricultural, there is no method for growing basidiocarps described in the literature. This work aimed to develop a substrate with Eucalyptus sp. for the cultivation of P. sanguineus and production of basidiocarps. There were two assays, and in vitro and in formulated substrate, developed in the Unioeste, Campus of Marechal CÃndido Rondon. We used sawdust of Eucalyptus sp. Separate in two particle sizes: less than 500 microns (G1) and between 500 and 841 microns (G2). In the in vitro assay were used four isolates of P. sanguineus (Ps04, Ps08, PS13 and PS14), grown in Petri dishes containing culture medium CBA plus sawdust in the proportions of zero, 1, 5, 10 and 15% for both sizes. In each dish was peaked a 0.5 cm disc containing mycelium and kept in dark at 25Â2ÂC, until the first treatment to reach the edge of the Petri dish. Was evaluated the diameter of colony, growth rate of mycelium, the fresh weight of mycelium and production of the pigment cinnabarin. The experiment was DIC in 4x2x5 factorial design, four isolates of P. sanguineus, two particle size of sawdust of Eucalyptus sp. and five concentrations of sawdust added to the culture medium, both with six repetitions. In the assay with formulated substrate, we used two isolates of P. sanguineus (Ps08 and Ps14), grown in polypropylene bags of 28x16 cm, containing sawdust of Eucalyptus sp. and rice bran. At zero, 5 and 20%, particle size in G1 and G2, with humidity of 75% and compacted to 0.5 g mL-1. The substrates remained 30 days in BOD at 25Â2ÂC, then taken to the greenhouse. Two havest of basidiocarps were made, at 90 and 180 days. Where were evaluated diameter, fresh and dry weight, the average number of basidiocarps and production cinnabarin. Was used experimental DIC in 2x2x3 factorial design, with two isolates of P. sanguineus, two particle size of sawdust of Eucalyptus sp. and three concentrations of rice bran added to sawdust, both with six repetitions. Was observed that the sawdust of Eucalyptus sp. is suitable for substrate for the production of basidiocarps of P. sanguineus, which is G1 showed better results only in vitro assays, while on substrate, the two particle sizes showed good results for the production isolates of P. sanguineus. The Ps08, produced more mass of basidiocarps, while the Ps14 showed higher content of cinnabarin in the basidiocarps. It can be concluded that the genetic characteristics of the biological potential of the isolates of P. sanguineus are more related to productive results that the characteristics of substrates used in this research

▼ A biopolpação consiste no biotratamento de madeira por fungos degradadores de lignina como etapa prévia à produção de polpa celulósica. De uma forma geral, as madeiras biotratadas facilitam os processos posteriores de polpação. Entretanto, ensaios anteriores que avaliaram a biopolpação em escala piloto mostraram que é difícil estabelecer os cultivos de basidiomicetos de interesse livre de bolores contaminantes, já que não é fácil controlar perfeitamente a assepsia durante a inoculação. Para contribuir com o avanço do processo de biopolpação, este trabalho avaliou os cultivos mistos de Ceriporiopsis subvermispora e Phanerochaete chrysosporium sobre madeira de Eucalyptus grandis e Eucalyptus urograndis em regimes de incubação em temperaturas variáveis e em condições não assépticas. A estratégia de iniciar os cultivos em temperatura mais elevada (37°C) e depois manter as culturas em temperaturas variáveis de 27°C e 37°C se mostrou eficiente para inibir o crescimento de contaminantes indesejáveis tanto nos cultivos mistos como nos individuais de cada espécie avaliada. Para avaliar o efeito do biotratamento da madeira numa etapa subsequente de polpação quimiotermomecânica (CTMP) foi feito um estudo preliminar de ajuste de variáveis de polpação por refinamento mecânico de cavacos pré-digeridos com licor alcalino contendo sulfito de sódio. Os dados mostraram que é possível simular o rendimento e as características fisico-mecânicas de polpas industriais empregando uma etapa de pré-digestão dos cavacos com 6% de Na2SO3 e 3% de NaOH a 120°C por 2 h, seguida de desfibramento e refino em refinador de discos. As curvas de refino nesse processo CTMP mostraram que os cavacos biotratados deram origem a polpas com 300 mL de Freeness (CSF) consumindo menos energia do que o observado para o refino da madeira controle. As economias de energia chegaram a 60% em alguns casos, porém não foram maximizadas pelo efeito sinérgico do cultivo misto dos dois basidiomicetos em questão. Experimentos adicionais foram desenvolvidos empregando as espécies fúngicas Pycnoporus sanguineus e Trametes versicolor que foram eficientes para competir com contaminantes mesmo a 27 °C. Neste caso, as economias de energia no processo CTMP subsequente somente foram obtidas com tempos mais longos de biotratamento (30 dias) e ainda assim os valores obtidos foram inferiores àqueles obtidos com as madeiras biotratadas por C. subvermispora e P. chrysosporium. Também a combinação das espécies C. subvermispora e Pleurotus ostreatus foi avaliada. Neste caso havia informação prévia da literatura indicando um efeito sinérgico da ação das duas espécies no que diz respeito a secreção da enzima manganês-peroxidase (MnP) e na degradação de lignina. Os resultados corroboraram que os cultivos mistos de C. subvermispora e P.ostreatus proporcionam maior produção de MnP do que os cultivos individuais de cada espécie. Por outro lado, P. ostreatus foi ineficiente para promover a redução do consumo de energia no processo CTMP subsequente. ▼ A região semi-árida do Brasil, que compreende parte de nove Estados do nordeste, é caracterizada pelo clima quente e seco e um curto período de chuvas (até 1000 mm/ano). A área é dominada por um complexo vegetacional, a Caatinga (floresta tropical sazonalmente seca), que revela várias paisagens compondo o bioma. De modo geral, é caracterizada pela vegetação xerófila de médio a baixo porte, tropófila, semi a caducifólia e espinhosa ou não, onde predominam leguminosas e cactáceas, com bromeliáceas que se destacam na paisagem. Incluindo o trabalho de Pe. Camille Torrend, que foi o primeiro a coletar e estudar os Agaricomycetes lignolíticos, na década de 30, apenas 37 espécies foram referidas para o semi-árido. Com o intuito de contribuir para o conhecimento da diversidade taxonômica e ecologia dos Aphyllophorales no semi-árido brasileiro, a partir de 2006 foram realizadas mais de 20 expedições de campo bem como a revisão taxonômica de materiais de Herbários (ALCB, CEPEC, HUEFS, HUVA, IPA, O e URM) provenientes dos estados de Alagoas, Bahia, Ceará, Paraíba, Pernambuco, Piauí, Rio Grande do Norte e Sergipe. As coletas foram mais intensas e realizadas de forma estratégica no estado de Pernambuco, mais especificamente no Parque Nacional do Catimbau e em Estações Experimentais do IPA (Araripina, Serra Talhada e Caruaru), com o objetivo de coletar não apenas material, mas também informações ecológicas a respeito dos táxons encontrados. Entre coletas e revisões de herbários foram analisados mais de 1000 espécimes, correspondentes a 130 espécies de 14 famílias, distribuídas em sete ordens (Agaricales, Auriculariales, Gloeophyllales, Hymenochaetales, Polyporales, Russulales e Trechisporales). Dichomitus brunneus, D. ochraceus, Lentinus amburanus, Phellinus caatinguensis e Wrightoporia stipitata são descritas como espécies novas. Gloeophyllum carbonarium e Lentinus fasciatus são novos registros para os Neotrópicos. Oito espécies são novos registros para o Brasil e outras 14 para o Nordeste. Das 130 espécies, sete (5.4%) são consideradas causadoras de podridão castanha. As Hymenochaetaceae, de modo geral, apresentaram altos níveis de especialização em plantas nativas da Caatinga no Parque Nacional do Catimbau, das quais Phellinus piptadeniae é considerada um parasita-facultativo frequente ou predominante em Piptadeniae sp. (hostrecurrence) e P. rimosus é um parasita-facultativo restrito a Caesalpinia sp. (host-specificity). Phellinus piptadeniae é uma espécie bem delimitada pela morfologia, ecologia e molecularmente, enquanto P. rimosus representa um complexo taxonômico com mais de uma espécie. ▼ Lignin is the second most abundant biopolymer, and the first most abundant source of phenolic structures, on Earth. Recent research has focused on lignin monomers as replacements for phenolic monomers derived from nonrenewable resources. Biological decomposition and conversion may be one way to accomplish this objective. In this study, the effects of two basidiomycetous fungi (Coriolus versicolor and Trametes gallica) and two actinobacteria strains (Microbacterium sp. and Streptomyces sp.) and their combination on lignocellulose (kenaf) decomposition was evaluated. The results showed that after 8 weeks of incubation up to 34 wt. % of the kenaf biomass was degraded, and the combination of fungi and bacteria was the most efficient. Lignin decomposition accounted for ~ 20 wt. % of the observed biomass reduction, regardless of the culture used. Most of this lignin was present as solubilized oligomers rather than monomers. Only after the monosaccharides were utilized by the microorganisms was the production of laccase, manganese-dependent peroxidase and lignin peroxidase enzymes induced, allowing lignin degradation to commence. The presence of carbohydrates was found to be detrimental for lignin degradation. In a subsequent series of experiments, we targeted the degradation/modification of isolated industrial-kraft lignin while trying to reduce the process time by solubilizing lignin with DMSO to increase lignin availability for enzymes. The addition of 2 vol% DMSO to nutrient free aqueous media increased the lignin solubility up to 70% while the quasi-immobilized fungi (pre-grown on agar) maintained their ability to produce lignolytic enzymes. While biological treatment was done for 6 days, significant modification was already observed in less than 24 hours. The resulting product showed the removal of phenolic monomers and/or their immediate precursors and a significant intramolecular cross-linking among the reaction products. Thus a new path for lignin biotreatment and further utilization was observed leading to the formation of polymers rather than monomers. Our interest therefore shifted to lignin utilization as a biochemically modified macromolecule. The biologically modified lignin was isolated via two different paths: 1) precipitation by acidification followed by washing with water or alcohols, or 2) vacuum evaporation followed by drying. The results of novel detailed chemical analysis of the modified lignin polymers showed that each of the washing steps can be used as a modification process, since each of them produced a slightly different polymer, with varied thermal stability, swelling and buffer capacities. The resulting lignin based polymers turned out to be insoluble in either organic solvents such as DMSO, DMF, NMP, dioxane etc., or in water. ▼ Wood is the largest source of biotic carbon on earth and the principle drivers of its decay are basidiomycete fungi. The biochemical mechanisms of wood decay by basidiomycete fungi are fundamental processes in forest ecosystems that dictate carbon evolution rates, soil organic matter deposition, and overall ecosystem function. These decay mechanisms are also unique in their ability to efficiently convert recalcitrant woody biomass to fermentable sugars and can serve as a biological template for industrial lignocellulose conversion to make renewable biofuels more economical. However, basidiomycete wood decay mechanisms are not fully understood and therefore not replicable in vitro, due in part to a lack of understanding of how decay mechanisms change throughout the progression of decay. In addition, gene transcripts and proteins used to facilitate decay are produced in concert with other biomolecules with non-degradative functions which makes resolution of degradative genes difficult. This dissertation contributes to resolving these problems by describing the temporal progression of decay among several species of wood-degrading basidiomycetes and functionally categorizing genes and secreted proteins involved in mediating interspecific interactions. This was done by first spatially resolving decay into a temporal sequence by growing model brown-rot fungi directionally on thin wood wafers. This system was used to co-localize changes in fungal physiology with chemical changes in wood substrates and the production of fungal metabolites to identify the functional significance of those physiological changes. The same wafer culture design was then used to resolve changes in fungal secretomes between two phylogenetically disparate brown-rot species over the course of decay using proteomics co-localized with lignocellulose-degrading enzyme assays. Interspecific differences were further investigated by comparing decay performance of the same two fungi on a Poales substrate, sorghum bagasse. Sorghum decay rates along with component removal and enzyme assays were monitored during decay to determine the genetic and biochemical basis of substrate preferences of the two species. Temporal alterations to fungal secretomes were compared among several model white and brown-rot fungi as well. Comparative proteomics concurrent with lignocellulose-degrading enzyme assays were used to identify common patterns among both rot types, as well as interspecific variability of decay mechanisms within species. Finally, changes in gene expression, protein secretion, and enzyme activity profiles in response to fungal competitors were described by modifying the thin wood wafer microcosms to incorporate two brown-rot species grown in opposition to one another. Resolution of decay into a sequence revealed a biphasic decay mechanism in brown-rot fungi delineated by early stage, non-hydrolytic pretreatment followed by later stage glycoside hydrolase-mediated saccharification. ▼ In the first of two parts of the investigation, the stimulation of Armiil aria mellea (Fr.) Quél., a common root-rotting fungus, by Aureobastdium pullulans (DeBary) Arnaud was studied. A. pullulans is an ubiquitous imperfect fungus, inhabiting soil and other substrates. It was shown to produce in culture a volatile, heat stable, neutral substance which stimulated the mycelial development, rhizomorph initiation and rhizomorph elongation of A. mellea. A. pullulans is the only microorganism reported so far to stimulate the growth of A. mellea. The effect of ethanol on the growth of A. mellea was similar to the effect of a cell-free filtrate from a liquid culture of A. pullulans. The cell-free filtrate was shown by gas chromatography to contain ethanol. Ethanol supplied daily as 50 ppm in a glucose-asparagine medium resulted in a ten-fold increase in the number of rhizomorphs produced by A. mellea. The effects of 3-indolylacetic acid, ɣ-(indole-3)-n-butyric acid, the sodium salt of 2,4-dichlorophenoxyacetic acid and tryptophane on the growth of A. mellea were tested, but the stimulatory effect of A. pullulans was not reproduced by them. One rhizomorph tip of A. mellea could develop an extensive rhizomorph system in autoclaved soil if the stimulatory substance produced by A. pullulans was available. In the second part of the investigation the effect of soil moisture and the related effect of some soil-inhabiting Fungi imperfecti on the spread through soil of the root-rotting Basidiomycete Coniopnora puteana (Schum. ex Fr.) Karst.were studied. A soil moisture level of 20 - 25% saturation was satisfactory for the growth of C. puteana in non-sterile soil. At this moisture level C. puteana was able to grow out from a small alder disc inoculum in the centre of a petri dish and invade alder discs at the periphery of the dish. In wetter conditions (50% saturation and higher) it was unable to grow out into the soil. In autoclaved soil the cptimum moisture level for the growth of C. puteana was between 75% and 100% saturation. Small amounts of non-sterile soil were added to autoclaved soil at different moisture levels, with an effect on the growth of C. puteanas similar to that of completely non-sterile soil. Trichoderma viride Pers. ex Fr., a known antibiotic producer, was inoculated in autoclaved soil and produced a greater inhibition of C. puteana in the wetter treatments than in drier ones. The inhibitory effect of Acti-dione (cycloheximide), an antifungal antibiotic active in the pH range 3-5, was examined in non-sterile soil end in autoclaved soil. The same concentration of antibiotic resulted in a greater inhibition of the growth of C. puteana at the higher moisture levels than at the lower ones.

In fungi, sexual reproduction is orchestrated by genomic regions known as mating type loci (MAT), which can be defined by two physically unlinked sex determining regions (tetrapolar mating system) or a single locus (bipolar system). Kwoniella mangroviensis is a saprobic basidiomycetous yeast, belonging to the Kwoniella clade of the order Tremellales, which was described as possessing a bipolar mating-system, similar to the related pathogenic species Cryptococcus neoformans and Cryptococcus gattii of the sister Filobasidiella clade. Studies aimed at elucidating the evolution of the MAT locus of these two Cryptococcus species of clinical importance, targeted several related saprobic species in the Kwoniella and Filobasidiella clades. An evolutionary model ensuing from those studies suggests that the tetrapolar mating-systems found in most species are ancestral and gave rise to the bipolar mating-system of C. neoformans by chromosomal rearrangements and fusion events. The present study comprised strains from the original work describing K. mangroviensis, as well as additional isolates from plant substrates in Europe and Africa. A multilocus sequence typing approach revealed genetic variability among those strains and led to the identification of two novel species closely related to K. mangroviensis: Kwoniella sp. A and Kwoniella sp. B. The mating system of K. mangroviensis and sibling species was further explored by a genetic approach based on sequencing of two MAT genes: STE20 and the divergently transcribed genes SXI1 and SXI2. The results obtained demonstrated tetrapolar mating systems in K. mangroviensis, as well as in Kwoniella sp. A and Kwoniella sp. B. Additionally, the MAT locus structure of K. mangroviensis was unveiled by sequencing a 43 kb genomic region containing the STE20 gene. Twelve genes also present in the MAT loci of related species were identified, and full synteny was found between K. mangroviensis and Cryptococcus heveanensis, a distant member of the Kwoniella clade. These findings provided novel insights into the evolution of MAT loci in basidiomycetous yeasts in the Tremellales.

This work concerns the investigation of the molecular mechanisms of sexual reproduction in fungi and their possible implication for fungal lifestyles (parasitic vs. saprobic) and for the emergence of asexual fungal lineages. The association between pathogenicity and sexuality is well-known in the basidiomycete plant parasite Ustilago maydis (subphylum Ustilaginomycotina), an economically important smut fungus. However, Ustilago species are phylogenetically interspersed with species of the genus Pseudozyma, which are considered saprobic and asexual. In this work, a study focused on genes involved in determining sexual identity (mating type or MAT genes), showed that Pseudozyma prolifica retains full sexual competence and pathogenicity, being therefore indistinguishable from U. maydis. For other Pseudozyma species, molecular analyses of PRF1, a gene that encodes a master regulator of sexual reproduction in U. maydis, showed no substantial evidence of loss of sexual reproduction. However, some clues were also found suggesting that some Pseudozyma species may be evolving towards a saprobic lifestyle. The earliest derived lineage of Basidiomycota (subphylum Pucciniomycotina) includes also important plant pathogens (rust and anther smut fungi) as well as lineages composed solely of saprobic organisms. Among the latter, the red yeasts of the order Sporidiobolales have the advantage of completing their life cycle in culture media, but have remained very little explored concerning the characterization of mating systems, the identification of MAT genes and the evolutionary relationships between sexual and asexual species. A comprehensive analysis of more than 200 strains belonging to 32 species of the Sporidiobolales indicated that asexuality seems to originate frequently from sexual lineages, but does not seem to persist long enough to form truly asexual species devoid of MAT genes. A more in-depth investigation of the red yeasts Rhodosporidium toruloides and Sporidiobolus salmonicolor allowed the identification for the first time in the Pucciniomycotina of the complete set of MAT genes. A detailed and multidisciplinary characterization of the mating system in the latter species yielded surprising results. A novel mating system that differs substantially from the two mating paradigms in basidiomycetes, the bipolar and tetrapolar systems, was brought to light. Given the basal phylogenetic position of the Pucciniomycotina within the Basidiomycota, this new system designated pseudo-bipolar, constitutes a significant contribution to the study of the evolution of MAT systems in fungi.

▼ Phellinus s.l. a été créé par Quélet en 1886. Il comprend actuellement 180 espèces, soit près de la moitié du nombre total d'espèces d'Hymenochaetaceae. Les caractères généralement considérés pour la definition du genre incluent des basidiomes à réaction xanthochroique positive permanente, une trame des tubes jaune à brun, un système d'hyphes dimitique pour l'essentiel avec des hyphes génératives à septa simples (i.e absence de boucles), des hyphes squelettiques brunâtres, et des éléments de type sétoïdes au niveau de l'hyménium ou plus rarement de la trame. Au niveau de leur physiologie (ou de leur biologie nutritionnelle), les Phellinus s.l. sont lignivores, possédant un système enzymatique capable de dégrader préférentiellement la lignine, et également partiellement la cellulose et l'hémicellulose, des composés de la paroi cellulaire du bois, produisant à terme une pourriture blanche. Ils se développent tant sur bois vivant (et sont des parasites parfois économiquement importants) ou sur bois mort. Les Phellinus sont généralement les champignons lignivores les plus divers et les mieux représentés dans les forêts tropicales. Ils participent ainsi activement au maintien des écosystèmes forestiers. Des études récentes, de morphologie fine et le développement d'approches complémentaires, nonmorphologiques, de type biochimique d'abord, génomique ensuite, ont toutefois démontré que la conception généralement admise par la majorité des auteurs modernes (Larsen and Cobb-Poulle 1990, Ryvarden, 1991; Fischer, 1996; Góes-Neto et al, 2001) est largement hétérogène et polyphylétique. En conséquence, de nombreux groupes morphologiques plus cohérents et monophylétiques ont émergés (ou ré-émergés) et ont été reconnus comme des genres satellites indépendants plus ou moins larges selon les cas. Certains de ces genres avaient été reconnus par des auteurs anciens, et ont pu être exhumés de la litérature; pour d'autre groupes des genres ont dus être crées. Citons par ex. les Fomitiporia, Fomitiporella, Fulvifomes, Fuscoporia, Porodaedalea, …. Afin d'élargir les connaissances de ces champignons dans la Région Sud du Brésil, une étude taxonomique a été réalisée à partir de révisions d'herbier et de l'analyse des spécimens collectés en 2010, 2011 et 2013, dans les trois États du sud du Brésil. Les données de la littérature ont également été rassemblées avec l'objectif de fournir un cadre des connaissances actuelles du groupe dans la région étudiée. Plus de 600 spécimens d'Hymenochaetaceae poroïdes ont été analysés. Quarantequatre espèces, distribuées dans neuf genres, ont été identifiées. De ces espèces et d'espèces mentionnées dans la littérature, 26 sont connus pour Paraná, 25 pour Santa Catarina et 35 pour Rio Grande do Sul. Six espèces sont décrites comme nouvelle pour la science.
