▼ A lei 9.787/1999, tamb?m conhecida como Lei dos Gen?ricos, possibilitou que ind?strias farmac?uticas nacionais produzissem medicamentos com patentes expiradas. A Valsartana, subst?ncia desenvolvida pela Ciba-Geigy e comercializada pela Novartis Pharmaceutical Company com nome Diovan?, se tornou atrativa para ind?stria nacional ap?s a lei dos gen?ricos pela sua importante aplica??o no controle da hipertens?o arterial. Esse f?rmaco ? um inibidor da angiotensina e seus receptores, com classifica??o biofarmac?utica de classe II. Durante a produ??o de subst?ncias, incluindo medicamentos, h? possibilidade da ocorr?ncia do polimorfismo, que consiste na mudan?a do h?bito cristalino do material. Nos f?rmacos este fen?meno impacta nas propriedades f?sico-qu?micas com consequ?ncia direta na biodisponibilidade. V?rias s?o as condi??es que podem causar essa transi??o cristalina, fatores que v?o da pr?pria linha de produ??o, transporte e armazenamento do medicamento. Desta forma a Food and Drug Administration estabeleceu uma s?rie de t?cnicas anal?tica para identifica??o de polimorfismo em f?rmacos. Neste trabalho foi empregada a t?cnica de espectroscopia Raman associada a ferramentas quimiom?tricas, tais como m?todos de regress?o multivariada (PLS e iPLS) para qualifica??o e quantifica??o de polimorfos de Valsartana. Foram utilizadas valsartana amorfa (comercialmente utilizada) e forma E (produzida na presen?a de ?gua/etanol), que tiveram suas estruturas comprovadas atrav?s da difra??o de raios X, an?lise termogravim?trica e calorimetria diferencial de varredura. As amostras com a mistura das duas formas de valsartana foram qualificadas e quantificadas na aus?ncia e presen?a des excipientes, por espectroscopia Raman associada a ferramenta quimiom?trica de calibra??o multivariada ? PLS e iPLS. O modelo de calibra??o obtido via regress?o iPLS com os pr?-tratamenos da primeira derivada Savitzky Golay e varia??o normal padr?o apresentaram elevada capacidade preditiva para novas amostras, comprovadas pelos baixos valores de erro m?dio quadr?tico de valida??o cruzada e erro m?dio quadr?tico de predi??o, para o princ?pio ativo sem excipiente. O modelo de calibra??o para o princ?pio ativo junto aos excipientes tamb?m apresentou boa qualidade, evidenciado pelos baixos valores de erro m?dio quadr?tico de valida??o cruzada e erro m?dio quadr?tico de predi??o, comprovando sua capacidade de previs?o de novas amostras, mesmo na presen?a dos excipientes. confiabilidade na an?lise de f?rmacos, baixo custo e versatilidade, podendo ser utilizada por ind?strias farmac?uticas e ?rg?os regulamentadores no controle de qualidade de f?rmacos.

▼ This work has sought to identify the relationship between management's mobilization styles and the productivity of the sales force in a pharmaceutical company. Sales Managers from large pharmaceutical companies were studied. The individual mobilization style of each manager was identified through the M.A.R.E.?? Motivational Guidelines and afterwards correlated to the marks given for productivity to his/her team as supplied by their individual companies, in order to identify the existing relationship between the results of the teams and the mobilization style of each of their leaders. It was noticed during the study that there was a great difference in behavior as regards the challenges faced by the team members on a day-to-day basis and how much the human factor was responsible for the results obtained.
In order to adequately reflect on the leadership theme, this study was based on authors such as Bass, Bennis, Burns, Machiavelli, Nannus and Spector. As regards the motivational aspects, the authors chosen for reflection were Alderfer, Herzberg, Maslow, McClelland, McGregor, Mitchel, Sinha, Vroom, Zoltners and Zoltners. The Mobilization Styles identified were based on contributions made by Coda and Ricco as well as studies carried out by Erich Fromm. The use of performance indicators has facilitated the identification of the results of each team for comparison to the Mobilization Styles. This factor determines the importance of the use of standard criteria so as to have an appropriate measurement of the people evaluated as well as give fairness to the process and guarantee credibility in the system to those involved. The study shows that certain mobilization styles are associated to favorable sales results, whereas other styles are not as favorable to a better performance of the teams. The study associates better productivity to the Competitive style and demonstrates that the Collaborative style is not conducive to adequate results for the sales teams of pharmaceutical products. The results obtained suggest towards a continuity of this study, initially in institutions working in the same field as the one studied here, so as to verify the data identified. Afterwards, attempting to corroborate the relationship between Managers and Mobilization Styles in the sales areas of companies in other segments, and finally searching for this relationship in other areas of the company.

▼ This dissertation discusses perceptions and opinions of generic industry executives about the development of this market in Brazil through a qualitative survey with in depth interviews. Stakeholders?? competitive forces were also studied in generic industry environment. There is lack of academic information about generic drug industry worldwide. It was made ten interviews with executives in the generic industry in a period of three months study - from August through October 2005 - with the objective of analyzing their perceptions regarding the importance of distributor, pharmacy, physician, patient and government. The main criteria to select the interviewers were based on the strategic positions they have in the companies. Some of the conclusions of this study were that government develops an important role in divulging generic drugs to people. Pharmacy and distributor were indicated as barriers to grow this market due to the sales practices they develop in the stores. Physician plays an important role as a stakeholder in this industry in order to popularize the use of generic drug in Brazil.

O Profissional de Educa??o F?sica (PEF) vai contribuir com a promo??o principalmente na sua inser??o no escopo de pr?ticas do N?cleo Ampliando de Sa?de da Fam?lia ? Aten??o B?sica (NASF-AB), atrav?s de a??es de aconselhamento e divulga??o de pr?ticas corporais e atividades f?sicas. O presente estudo tem como objetivo analisar as a??es de Promo??o da Sa?de realizadas pelo Profissional de Educa??o F?sica no NASF-AB no Estado do Amazonas. Trata se de um estudo de natureza descritiva, do tipo qualitativo. A amostra foi por conveni?ncia, totalizando 33 profissionais, sendo 11 PEF, 11 Agentes Comunit?rios de Sa?de (ACS) e 11 Enfermeiras (os) que atuam nos NASF-AB dos munic?pios de Autazes, Iranduba, Itacoatiara e Nova Olinda do Norte. Foi utilizado como instrumento um question?rio semiestruturado, com perguntas abertas e fechadas para o alcance das inten??es do estudo. Para an?lise dos dados foi utilizado os pressupostos da an?lise do discurso. Os resultados encontrados mostram que a caminhada, a gin?stica, o alongamento e circuito funcional s?o as a??es mais frequentes dos profissionais. Tamb?m foi encontradas pr?ticas como a shantala com gr?vidas e beb?s. Encontramos nos discursos dos profissionais participantes, referentes as contribui??es da atua??o do PEF, que a atividade f?sica colabora para a minimiza??o dos sintomas das doen?as, principalmente as doen?as cr?nicas n?o transmiss?veis. Em rela??o aos elementos culturais, os PEF evidenciam a dan?a. Nos discursos n?o ? percebido a diferencia??o entre os termos Pr?ticas Corporais e Atividade F?sica, por parte dos PEF, sendo utilizado mais o segundo termo. Embora analisados de uma forma global e n?o particularmente cada munic?pio, ? visto com pouca nitidez, os princ?pios da Promo??o da Sa?de impl?citos nos discursos apurados.
The National Health Promotion Policy (PNPS), created in 2006, institutionalized as Health Promotion actions in the Unified Health System. The Physical Education Professional (PEF) will contribute to the promotion mainly in its insertion in the scope of practices of the Extended Family Health Center - Primary Care (NASF-AB), through counseling and dissemination of corporate practices and practical activities. This study aims to analyze the Health Promotion actions carried out by the Physical Education Professional at NASF-AB in the State of Amazonas. This is a descriptive, qualitative study. A sample was made for convenience, totaling 33 professionals, 11 PEF, 11 Community Health Agents (ACS) and 11 Nurses who work at NASF-AB in the municipalities of Autazes, Iranduba, Itacoatiara and Nova Olinda do Norte. It was used as a semi-structured questionnaire instrument, with open and closed questions to reach the intentions of the study. For data analysis, the assumptions of discourse analysis were used. Em virtude disso a Organiza??o Mundial de Sa?de declarou como emerg?ncia global e elaborou estrat?gias para cont?-la, destaca-se neste contexto a Aten??o Prim?ria ? Sa?de, pois no Brasil ? a ordenadora da rede de sa?de e desempenha a??es primordiais para controle da tuberculose. O presente estudo teve como objetivo avaliar o desempenho dos servi?os de Aten??o Prim?ria ? Sa?de no controle da tuberculose em Manaus, na perspectiva dos enfermeiros. Estudo descritivo de car?ter avaliativo com abordagem quantitativa. Elegeu-se o munic?pio de Manaus devido ao seu destaque nacional como capital de maior incid?ncia de casos. A coleta ocorreu entre os meses de dezembro de 2018 a maio de 2019, nos servi?os de Aten??o Prim?ria ? Sa?de (Unidades B?sicas de Sa?de e Unidades B?sicas de Sa?de da Fam?lia). A popula??o foi composta por 173 enfermeiros que atuavam nesses servi?os. Durante a coleta foi utilizado o instrumento ?Primary Care Assessement Tool? adaptado e validado para avaliar a aten??o ? tuberculose, o qual cont?m 96 quest?es espec?ficas para avalia??o do desempenho dos servi?os de sa?de da Aten??o Prim?ria ? Sa?de sob a ?tica dos profissionais de sa?de e inclui quest?es referentes ?s dimens?es/atributos da Aten??o Prim?ria ? Sa?de: Porta de entrada, Acesso (subdividido em Acesso ao diagn?stico e Acesso ao tratamento), V?nculo (Longitunalidade), Elenco de servi?os (Integralidade), Coordena??o, Enfoque na fam?lia, Orienta??o para a comunidade e Forma??o profissional (Compet?ncia cultural). A an?lise, interpreta??o e apresenta??o dos dados foi feita atrav?s de estat?stica descritiva (medidas de tend?ncia central, dispers?o e vari?ncia, frequ?ncias absolutas e relativas, conforme orienta??es do instrumento utilizado. O taxa ? de Cronbach foi usado para verificar a confiabilidade e consist?ncia interna do instrumento (?=0,86). A partir desses resultados encontrados podemos concluir que na perspectiva dos enfermeiros a maioria das dimens?es apresentam resultados ?Satisfat?rios?. Apesar disso, h? fragilidades como:dificuldades no acesso, na oferta de servi?os e integra??o com a comunidade; que precisam ser trabalhadas para oferecer um servi?o de melhor qualidade ao usu?rio com tuberculose, e a partir disso, considerar uma real mudan?a do cen?rio atual local.
Tuberculosis remains a major global health problem, the incidence of which remains high at global, national, regional and local levels. de Sa?de (SUS), em Manaus. A abordagem da Psicologia da Sa?de, de orienta??o
comunit?ria, destaca como eixos norteadores a atua??o territorial, multiprofissional, e os
processos biopsicossociais implicados nas condi??es de sa?de/doen?a, e na integralidade do
cuidado a sa?de. Os objetivos principais foram: a) investigar a pr?tica de cuidados ? sa?de de
uma UBSF, em Manaus, focada nas situa??es de visita domiciliar enquanto a??o priorit?ria
para sujeitos/usu?rios destacando os aspectos psicossociais nessa a??o de cuidados; b)
Analisar a pr?tica das visitas domiciliares realizadas com aqueles sujeitos com DM2 em
comorbidade com HAS, inseridos no programa HIPERDIA; c) Estudar dois exemplos
significativos de usu?rios em experi?ncias de sofrimento/adoecimento de DM2 e HAS em
cuidados domiciliares. O m?todo utilizado foi o da observa??o participante que possibilitou a
implica??o da pesquisadora no cotidiano do campo investigativo (UBSF). A an?lise dos
resultados, de caracter?stica descritivo-explorat?ria, mostrou precariedades nas condi??es de
vida dos usu?rios no territ?rio adscrito. Dos vinte e um sujeitos/usu?rios visitados, dezesseis
estavam relacionados ao programa HIPERDIA. Assim, nove visitas a sujeitos com DM 2, tr?s
com DM 2 em comorbidade com HAS e outros tr?s com Hipertens?o. As cinco restantes
diziam respeito a outros processos de adoecimento. De maneira geral, esses atendimentos
tendiam a seguir uma l?gica mais voltada ao modelo biom?dico, com algumas nuances
sociais. Entretanto, na fala dos usu?rios visitados percebia-se problem?ticas psicossociais
impl?citas, muitas vezes, pouco trabalhadas na rela??o de cuidados e ainda n?o
adequadamente articuladas com a sintomatologia biom?dica. Os estudos de caso
exemplificaram de forma espec?fica essa din?mica de cuidados, revelando tamb?m uma
rela??o emp?tica entre os profissionais e usu?rios. Al?m de colocar em evidencia que a rede
de cuidados familiar, quando colaborativa, contribu?a com a melhora da qualidade de vida do
usu?rio e de seu autocuidado. Nas considera??es finais enfatizamos a import?ncia da
psicologia da sa?de como contribui??o cient?fica para a APS e a pr?tica das visitas
domiciliares na UBSF.
The study object of this research was the practice of a home visit, a priority, for the care of
subjects / users with DM 2 in Primary Health Care in the Unified Health System (SUS), in

▼ Norén, A. 2020. The Reign of Akhenaten: The Inhabitants of Tell el-Amarna through a Religious Perspective. The purpose of the study is to demonstrate the complex religious climate of Tell el-Amarna, the capital city of ancient Egypt in the 18th dynasty, during the reign of Amenhotep IV/Akhenaten (1353–1336 B.C.E.). These complex elements refer to the devotion of pharaoh and his new-established ideology of Atenism and the simultaneously ongoing worship of traditional gods of which many were actively persecuted by the state during this time. The conclusions of this study presents a general picture of the lives of the inhabitants of el-Amarna by drawing on examples of religious objects which have been excavated from the city and what these may suggest about the religious beliefs of the people. However, the study does not focus primarily on the distribution of objects or social stratification. The sources which have been employed in this study consists of books and articles published by Barry J. Kemp and Anna Stevens, among others. The primary source material by Kemp and Stevens comprises archaeological theories and methods which in this study have been employed in an attempt to bring the material one step further, namely to analyse the religious climate at el-Amarna through the perspective of its inhabitants and place the material evidence in relation to the ideologies of Akhenaten.

Syftet med studien är att redogöra för det komplexa religiösa klimatet i Tell el-Amarna, som var huvudstaden i det antika Egypten under den 18nde dynastin, under Amenhotep IV/Akhenatens regeringstid (1353–1336 f.v.t.). Med komplexa element menas den hängivenhet som uttrycktes inför farao och hans nyetablerade ideologi som kallas Atenism samtidigt som det existerade en fortsatt tro på de traditionella gudarna som aktivt förföljdes och raderades av staten under den här tiden. Slutsatserna i den här studien presenterar en generell bild av livet för invånarna i el-Amarna genom att återge exempel på religiösa objekt som har hittats i staden och vad dessa må berätta gällande det religiösa utövandet hos människorna som bodde här. Studien fokuserar inte primärt på spridningen eller mängden av objekt och heller inte på social stratifikation. De källor som använts i den här studien består av böcker och artiklar som publicerats av Barry J. Kemp och Anna Stevens samt andra forskare. Det primära källmaterialet av Kemp och Stevens utgörs av arkeologiska teorier och metoder som används i den här studien med avsikten att ta materialet ett steg längre genom att analysera det religiösa klimatet i el-Amarna ur invånarnas perspektiv och sätta de materiella lämningarna i relation till Akhenatens ideologier.

▼ Environmental interest, regulation changes, and costs have motivated the oil and gas industry to begin recycling produced water in high concentrations during new well stimulation. Accordingly, the potential impact of this practice on production should be investigated. In this thesis paper, tests were conducted to determine whether Marcellus produced water would cause incompatibilities in the
Utica and Point Pleasant formations. A multivariate statistical analysis was then completed using a historical dataset of over 300 Marcellus wells to measure the effect of produced water used in stimulation had on well production. The results indicate that recycling produced water in high proportions, even from the Marcellus, should have no measurable impact on the productivity of
Utica and Point Pleasant wells. This conclusion supports the use of recycled water not only to comply with regulations and address environmental concerns, but also as a method to reduce water management costs by at least 40 percent.

Utica Shale across the Appalachian Basin in Bradford County, Pennsylvania. Concepts documented in this thesis can be used for other shale-gas systems. Unlike most shale-gas system studies, S-wave modes were used in addition to P-wave data in this study to better characterize the
Utica Shale. Fast S-converted shear (P-SV1) and slow S-converted-shear (P-SV2) volumes provide new seismic imaging options for shale-gas studies and enable expanded seismic attributes that can be used to characterize shale-gas systems.

▼ The early Late Ordovician interval, known as the
Utica-Point Pleasant play, in southeastern Ohio is poorly constrained due to limited data sets. A previous study by the Appalachian Basin Oil and Natural Gas Research Consortium and partners evaluated the interval across the Appalachian Basin using five cores. For this study, unpublished data from well Farley 1305-H, located in Washington County, Ohio, was used to better evaluate the
Utica-Point Pleasant play in southeastern Ohio. The data include geophysical logs, Rock-Eval pyrolysis, mineralogy, triaxial testing, gas desorption, and porosity data. The Point Pleasant Formation, in regards to well Farley 1305-H and proximal area, has sufficient TOC (avg. 2.4 wt. %), gas-filled porosity (avg. 4.75 %), mineralogy (normalized clay value of 32%), and geomechanical properties to be a successful unconventional play. Geophysical logs correlate the formation's high organic content and porosity. Calculations of % Ro values (avg. 1.6) are parallel to gas composition data, which indicates that a good portion of gas in place is wet and dry. The
Utica Shale was determined to be a poor unconventional reservoir in regards to well Farley 1305-H, lacking conducive TOC [less than] 1 wt. %), gas-filled porosity ([less than] 2 %), mineralogy (normalized clay value of 49 %), and geomechanical properties.

▼ I present an application of a high-resolution subsurface imaging technique known as “full-waveform inversion” (FWI) to a vibroseis seismic dataset from eastern Ohio, USA. The data were collected over a crooked line with rough topography, 3.5 km maximum offsets, and no significant frequency content below 12 Hz. These parameters present challenges to obtaining quality images from FWI. The use of a preconditioner–the ‘scaled-Sobolev preconditioner’ (SSP - Zuberi and Pratt, 2017)–on the gradient of the misfit functional was key to obtaining low wavenumbers without discarding high wavenumbers. The results represent the first successful application of FWI with the SSP to a field dataset, with a high-resolution image that generally matches the trends of the Big Injun sand and Berea sandstone layers at the survey location. The novel FWI results confirm the absence of small scale structure (including the lack of visible faults) in the first 0.66 km.

▼ Hydraulic fracturing and shale gas development have
led to an abundance of domestic natural gas production, and the
environmental impacts of these processes are under investigation.
Previous studies in regions where the Marcellus Shale has been
developed have shown that methane (CH4) derived from natural gas
was present in drinking water wells near gas wells, likely due to
well casing failures. Here we present a 28-month time series of
groundwater CH4 concentration, CH4 stable isotope composition (d13C
and d2H), pH, and specific electrical conductivity through a period
of increasing shale gas extraction from the
Utica Shale of eastern
Ohio. The study period corresponded with an increase in the number
of active Carroll County gas wells from 3 in late 2011 to 354 in
2015. CH4 was detected in all groundwater wells, with
concentrations ranging from below 0.2 µg L-1 to above 25 mg L-1.
d13C-CH4 and d2H-CH4 measurements averaged -65 ±11 ‰ (n=78) and
-180 ±61 ‰ (n=85), respectively, indicating a biogenic CH4 source
origin. Radiocarbon dating of four dissolved CH4 samples indicates
that coal formations are the source of elevated CH4. We found no
positive relationship between CH4 concentration in groundwater and
proximity to active gas well sites, and we found no significant
change in CH4 concentration, isotopic composition of CH4, pH, or
conductivity in water wells during the study period. Ongoing
monitoring of private drinking water wells is critical to ensuring
residents are not exposed to harmful levels of natural gas or other
fracking contaminants.

Utica Shale/Point Pleasant Formation system has recently become a highly developed unconventional target for oil and natural gas production, leading to an increased desire for knowledge of the controls on deposition of this system. Precambrian basement features have long been known to affect deposition of older strata near these features across Ohio, but the effects of far field tectonics is not fully agreed upon. Precambrian faults and lineaments are known to exist and have been mapped, but are thought to have ceased their influence on deposition by the time of the Knox unconformity during the Cambrian. In the case of the Ordovician
Utica Shale, Point Pleasant Formation, Trenton Limestone, and the Black River Group, many believe that the deposition of these strata were not affected by Precambrian faults/lineaments or other basement features, rather their main influence was changes in sea level due to basin loading, climatic changes, and localized uplift/subsidence. The objective of this study was to determine if the deposition of these younger strata were influenced by Precambrian faults and basement features. This was done by analyzing a group of oil and gas wells in Lorain, Cuyahoga Lake, Geauga, Ashtabula, Trumbull, Mahoning, Stark, Portage, Summit, Wayne, Ashland, and Medina counties in northeastern Ohio. Electric well logs from these wells were ii added to Petra, a geological mapping program, and the tops of the
Utica Shale, Point Pleasant Formation, Trenton Limestone, and the Black River Group were picked. These tops were then contoured into four structure maps and 4 isopach thickness maps. These maps were analyzed with known Precambrian faults and lineaments, along with known Precambrian basement features, to determine if these Precambrian influences affected the deposition of these Ordovician strata. It is known that high spatial resolution structure maps show contour offsets across known and inferred structural features. It is also known that high spatial resolution isopach maps show thinning and thickening across known and inferred structural features. The purpose of this study was to take these methods and apply them to the
Utica Shale/Point Pleasant Formation system in order to determine structural influence from the Precambrian basement on Ordovician strata in northeastern Ohio.

▼ The Gorgon is a creature described as terrible in ancient literature. It was depicted with glaring eyes, tusks and a hanging tongue and was a part of Greek antiquity from Archaic to Roman Period. The Gorgon motif has frequently been adorned on different materials. The reason as to why such a creature was depicted has been a subject of interest in earlier studies. The Gorgon motif has been elaborately studied in combination with buildings, armours and vases. A gap of knowledge that is still to be filled is a deeper examination of the Gorgon motifs on coins, which is the inspiration for this study in which the main aim is to approach an understanding of what function the Gorgon motif could have had on Archaic Greek coins. The study is based on a collection of 42 Archaic coins from Athens and Neapolis in Macedon. Through Panofsky's theory of iconography the material is analyzed and discussed via a series of sub-questions; ‘Did the Gorgon motifs differ depending on the location?’, ‘What combination of features appear on the coins?’, ‘To what extent was the Gorgon myth linked to the locations that used the motif and what other myths were used on coins during the same period? ’, ‘Is there a link between the use of Gorgon motifs on coins and on other material objects?’ The paper measures the possible explanations of the Gorgon motif with archaeological finds and ancient texts dealing with the Gorgon, many of which point to the fact that the Gorgon’s function generally served a purpose as an apotropaic symbol. Its function as a possible amulet is investigated using previous research that studies the symbolic significance of the Gorgon, as well as tracing its background and examination of the Gorgon myth to find possible connections with other mythical creatures.

We acquaint with the whole spectrum of services suitable for distant management of data centre from distant operating of supply till the distant brace, substitution of monitor, keyboard and mouse. ▼ Listeria monocytogenes est une bactérie pathogène responsable de la listériose chez l’homme. Cette bactérie est capable d’envahir et de se répliquer dans des cellules phagocytaires et non-phagocytaires de l’hôte. De fait, Listeria interagit et interfère avec de nombreux composants cellulaires au cours de sa réplication. Mon projet de thèse s’est focalisé sur les altérations du protéome des cellules infectées par Listeria. Je me suis en particulier intéressé à l’activation de protéases de l’hôte et aux dégradations de certains facteurs cellulaires en réponse à l’infection.Une analyse protéomique de cellules humaines traitées avec la Listeriolysine O (LLO), une toxine secrétée par Listeria formant des pores dans les membranes cellulaires, nous a permis d’identifier plusieurs dizaines de facteurs cellulaires dégradés en réponse à l’exposition à cette toxine.Nous avons pu confirmer la dégradation de ces protéines dans le contexte d’une infection par Listeria in vitro (dans un modèle de cellules en culture) et in vivo (dans un modèle animal). Nous anticipons que la dégradation de ces protéines, impliquées dans différentes fonctions cellulaires, joue un rôle au cours de l’infection par Listeria en modifiant la physiologie des cellules hôte.Nous avons en parallèle mis en évidence que Listeria interfère avec le fonctionnement des lysosomes. Nous avons montré que la LLO déstabilise les lysosomes des cellules hôtes et induit leur perméabilisation. Ceci permet la libération de certaines protéases lysosomales, telles que les cathepsines, dans le cytosol des cellules hôte. Ces cathepsines, une fois libérées, pourraient alors cliver différentes protéines cytosoliques et ainsi modifier la physiologie des cellules infectées.L’ensemble de mes travaux a permis de montrer que Listeria interfère avec le fonctionnement de nombreuses protéases de l’hôte et induit la dégradation de plusieurs facteurs cellulaires. Ces cibles dégradées constituent une nouvelle classe de protéines dont nous pouvons maintenant tester le rôle potentiel dans l’infection. Mes travaux ont d’autre part permis de montrer que d’autres toxines bactériennes, similaires à la LLO, induisent également des altérations du protéome des cellules hôtes. Ceci suggère que les mécanismes identifiés avec Listeria sont conservés entre différentes classes de pathogènes bactériens.
Listeria monocytogenes is a foodborne pathogen responsible for human listeriosis which invade and replicate in both phagocytic and non-phagocytic cells. Listeria intracellular life cycle involves interference with host cell components. My PhD project aims to characterize host cell proteome modifications in cells infected by Listeria. I focused my research on host proteases activation and the degradation of host cell proteins in response to infection. ▼ My study centered on a bioindicator songbird, the Louisiana Waterthrush (
Parkesia motacilla), hereafter waterthrush, an organism that co-occurs in both forested and aquatic habitat across the aquatic-terrestrial interface. This enabled the opportunity to quantify demographic, spatial, and epigenetic (i.e., DNA methylation) responses in a highly forested watershed of the Central Appalachians, the areas that have undergone the most rapid transformations over the last decade from unconventional shale gas development and activities. I organized my dissertation into 4 parts (Part 1: Introduction, Part 2: Louisiana Waterthrush Demography, Part 3: Spatial Assessment of Louisiana Waterthrush Foraging, Part 4: Louisiana Waterthrush Molecular Ecology) including 6 chapters that indicate multiple biotic and abiotic factors interacted with or were altered by shale gas development resulting in atypical, negative disturbances that drove a steep decline in a waterthrush population in West Virginia.
Part 1 includes Chapter 1 and is an introduction to my dissertation. I introduce the reader to the rationale for my study, the focal species, research objectives, and the study area. I also mention some limitations to my study that can be considered in any future research endeavors.
Part 2 comprises Chapters 2–3 which are a comprehensive examination of demographic parameters over a six-year period (2009–2011, 2013–2015). In Chapter 2, I examined demographic response to shale gas development for nest abandonment, nest survival, nest productivity, a source-sink threshold, riparian habitat quality, and territory density and length. Nest productivity was lower in areas disturbed by shale gas where a source–sink threshold suggested these areas were more at risk of being sink habitat. Overall results suggest a decline in waterthrush site quality as shale gas development increased. In Chapter 3, I focused on first-year return rates (site fidelity), site fidelity factors, and apparent survival. I related natal fidelity and pairing rates to territory density, and also compared of breeding attempts between return and non-returning females with and without territory shale gas disturbance. The study identified potential conflicts between factors that influence adult survival and site fidelity that may affect long-term population persistence.
Part 3 includes Chapters 4–5 and focuses on utilizing and accounting for spatial properties intrinsic to stream ecosystems to make informed decisions regarding waterthrush foraging. Chapter 4 was a follow-up to a waterthrush aquatic prey study at our site in 2011 that suggested shale gas development negatively affected waterthrush demography from alterations in their aquatic prey at a watershed scale. During 2013–2014, I quantified waterthrush demographic response and nest survival in relation to potential changes in its aquatic prey due to shale gas development. ▼ Despite the money and efforts put forth to control obesity in the United States, it remains a serious problem. This research project investigated the correlates of American adult overweight and obesity in the Mohawk Valley region in upstate New York, seeking to understand how cultural models of healthy foods, individuals' eating habits, and the social relationships of people contribute to increased body mass. The project drew on cultural consensus analysis (Romney, Weller, and Batchelder 1986) and the related theory of cultural consonance (Dressler 2005) in an attempt to link culture (beliefs about healthiness of foods) to behavior (food intake), and in turn link these to the body (the biological variable of weight). The inclusion of social network data allowed for examination of potential links between network structural or attribute variables and BMI. In total, 139 individuals were interviewed.

▼ In nineteenth-century America a new approach to treating insanity was adopted. This approach was called moral therapy. Physicians who practiced moral therapy tried to create an ideal, curative environment for the disordered mind through landscape and architecture but fell short because reality demanded they use restraints and medicine to limit the behavior of their patients. This paper and accompanying online exhibit explores the conflict between moral therapy's utopian setting and its dystopian reality from the 1830s through the 1870s. By 1880 most American asylums had become centers for custodial rather than curative care but an enduring legacy of moral therapy has continued to the present day.

▼ Les cellules humaines sont soumises à des stress induisant des cassures double-brin de l’ADN principalement réparées par la voie NHEJ, où la kinase DNA-PKcs joue un rôle central. L’activité de DNA-PKcs, régulée par de nombreuses phosphorylations, est cruciale pour le maintien de l’intégrité génomique. Plus récemment, il a été montré que cette protéine était également modifiée par l’O-GlcNAcylation dans la lignée COS7. Sachant l’équilibre existant entre phosphorylation et O-GlcNAcylation, nous avons étudié le rôle de cette nouvelle MPT dans la régulation de l’activité de DNA- PKcs. Nous avons montré que DNA-PKcs est O-GlcNAcylée dans les cellules HeLa. Puis nous avons montré que la modulation de l’O-GlcNAcylation de DNA-PKcs impacte son autophosphorylation en Ser2056, suggérant l’existence d’une balance O-GlcNAcylation /phosphorylation, ainsi que la capacité des cellules à réparer les DSBs par la voie NHEJ. De plus, nos résultats nous laissent envisager que cette modification puisse jouer un rôle dans la stabilité de la protéine. DNA-PKcs est une cible potentielle dans les stratégies de lutte contre le cancer. Nous avons étudié l’impact d’un composé sur DNA-PKcs. Cette molécule provoque une réduction de la quantité et de l’activité de DNA-PKcs, impliquant son ubiquitinylation et sa dégradation par le protéasome et menant à une sensibilisation des cellules à un traitement génotoxique. Dans ce contexte, nous avons développé une puce à anticorps pour évaluer le profil phosphoprotéique des voies de réparation de l’ADN et ainsi évaluer l’effet d’inhibiteurs de DNA-PKcs. L’ensemble de ces résultats contribuent à une meilleure compréhension de la régulation de DNA-PKcs.
Human cells are subjected to stresses inducing DNA double-strand breaks mainly repaired by the NHEJ pathway, where the kinase DNA-PKcs plays a central role. The activity of DNA-PKcs, regulated by numerous phosphorylations, is crucial for the maintenance of genomic integrity. More recently, it has been shown that this protein is also modified by O-GlcNAcylation in the COS7 cell line. Knowing the balance between phosphorylation and O-GlcNAcylation, we studied the role of this new PTM in the regulation of DNA-PKcs activity. We have shown that DNA-PKcs is O-GlcNAcylated in HeLa cells. We then showed that the modulation of DNA-PKcs O-GlcNAcylation affects its autophosphorylation on Ser2056, suggesting an O-GlcNAcylation/phosphorylation balance, as well as the ability of cells to repair DSBs by NHEJ pathway. Moreover, our results allow us to consider that this modification may play a role in protein stability. DNA-PKcs is a potential target in anticancer strategies. We studied the impact of a chemical compound on DNA-PKcs activity. This molecule causes a reduction in the amount and activity of DNA-PKcs, through its ubiquitinylation and its degradation by the proteasome and leading to sensitization of the cells to genotoxic treatment. o health discourses stem from historical and social notions of biological, cultural, and racial inferiority. Popular U.S. newspapers pay scant attention to Latina/
o health concerns and often inaccurately portray Latinas/os as undeserving foreigners that continue to drain social services such as health care. A content analysis of 291 New York Times, Los Angeles Times, Chicago Tribune, and Houston Chronicle newspaper articles (2006-2010) reveals that Latina/
o health discourses are grounded in a racialized medical narrative that justifies and sustains white racial oppression. Systemic racism and the white racial frame are utilized as theoretical frameworks to better understand how mainstream newspapers construct the medical racialization of Latinas/os and contribute to health disparities, unequal access to health services, and inadequate health care. The findings reveal that Latina/
o health issues concerning high costs, population increase, and political marginality, influence anti-Latina/
o legislation, sustain prevailing racism, and create exclusionary health practices. Fundamentally the anti-Latina/
o sentiment presented in the newspapers and disseminated throughout society equates to the denial of resources, the denial of health care, and thus the denial of life. Challenging racist Latina/
o perceptions is an important area of social science and anti-racism research. Ultimately, without a healthy Latina/
o workforce, the economy could not sustain itself and society would be susceptible to economic, social, and political collapse.

▼ L'insuffisance cardiaque (IC) touche 1-2% des adultes dans les pays développés. On distingue classiquement 2 formes d’IC : l’IC chronique, dont la mise en place est lente, et l’IC aigue dont l’apparition est rapide ou due à l'aggravation des symptômes et/ou des signes de l’IC. Quelles soient chroniques ou aigues, certaines formes d'IC n'ont pas de traitement efficace. L'insuffisance cardiaque à fraction d'éjection préservée (ICFEp) devient la forme prédominante d'IC chronique mais la compréhension de sa physiopathologie reste encore incomplète. Un modèle de rat transgénique sur-exprimant le récepteur β3- adrénergique humain au niveau endothélial a été développé dans l'équipe. Comme les patients, ce modèle développe une ICFEp avec l’âge qui se manifeste notamment par une dysfonction diastolique et une intolérance au stress. Dans le cadre de ma thèse et sur la base d'une étude transcriptomique, j'ai évalué les altérations du cycle du calcium, du métabolisme ainsi que l'apparition de fibrose et d'inflammation. Le choc septique est considéré comme une IC aigue particulièrement violente. Le deuxième axe de ma thèse a été de développer des modèles pertinents de choc septique et d’évaluer le potentiel thérapeutique d’une modification post-traductionnelle : la O-GlcNAcylation, déjà connue pour ses effets bénéfiques dans des formes d’IC aigues. Ainsi, une augmentation des niveaux de l'O-GlcNAc, grâce à un agent pharmacologique, le NButGT, améliore la fonction cardiovasculaire et la survie des animaux en choc septique.
Heart failure (HF) affects 1-2% of adults in developed countries. There are two distinct forms of HF: chronic HF, with a slow development, and acute HF that appears rapidly or is due to an aggravation of the symptoms and/or signs of HF. For some forms of HF, acute or chronic, no effective treatment exists. Heart failure with preserved ejection fraction (HFpEF) is becoming the predominant form of chronic HF but the understanding of its pathophysiology remains incomplete. A transgenic rat model over-expressing the human endothelial β3-adrenergic receptor has been developed in the team. As in patients, this model develops HFpEF with age, and is characterized by diastolic dysfunction and stress intolerance. During my thesis and based on transcriptome data, I evaluated alterations of the calcium cycle, metabolism as well as the appearance of fibrosis and inflammation. Septic shock is considered to be a form of severe acute HF. The second axis of my thesis was to develop relevant models of septic shock and to evaluate the therapeutic potential of a post-translational modification: O-GlcNAcylation, already known for its beneficial effects in acute HF forms. Indeed, an increase in levels of O-GlcNAc, through a pharmacological agent, NButGT, improved cardiovascular function and survival of animals in septic shock.

Testausohjeiden tarkoitus on toimia yleisesti kattavina ohjeina tyypillisimmille projektitoimituksille. Laitteiston perusteellinen testaus on olennainen osa Valmetin projektin toimitusprosessia. Testausohjeilla pyritään tehostamaan laitteiston testausprosessia, ja varmistamaan turvallisuus.
Testaus tapahtuu kahdessa vaiheessa. Laitteistolle tehdään ensin tehdaskoestus Valmetin toimitiloissa. Laitteiston tehtaaseen asennuksen jälkeen suoritetaan käyttöönottotestaus. Tämä opinnäytetyö sisältää testausohjeet projektin molempiin vaiheisiin. Testausohjeiden laatimiseksi seurattiin usean projektin tehdaskoestusta ja yhden projektin käyttöönottotestausta.
Tuotetut testausohjeet ovat salassapidettäviä, ja ne jäävät Valmetin käyttöön. Opinnäytetyö ei sisällä testausohjeita. Tämän opinnäytetyön julkaistavassa raporttiosiossa käsitellään testausohjeiden tekoprosessi ja testausohjeiden sisältö yleisesti. Työhön kuuluu myös sähkötyöturvallisuuden huomioiminen tehdaskoestuksessa ja käyttöönotossa. Sähkötyöturvallisuuden varmistaminen huomioitiin testausohjeissa, ja sähkötyöturvallisuutta käsitellään raporttiosiossa. EU:n konedirektiivin ja siihen liittyvien standardien vaatimukset ovat myös osa tätä työtä. Direktiivin vaatimukset ohjasivat testausohjeiden laatimista, ja opinnäytetyöhön sisältyy direktiivissä käsitelty vikatilanneanalyysi.
The topic of this thesis is Testing manuals for I/O interfaces and devices connected to fieldbus. Thesis was produced for Valmet Automation. Hardware of automation systems delivered by Valmet consists of I/O interfaces, I/O devices and devices connected to fieldbus. The purpose for testing manuals is to serve as universally comprehensive instructions for typical projects delivered by Valmet. Thorough testing of the hardware is essential part of the project delivery process of Valmet. Testing manuals aim at optimizing and securing the testing process of the hardware.
Testing is done in two parts. At first, factory acceptance testing is done in facility of Valmet. Site acceptance testing is done after hardware has been installed in factory. This thesis includes testing manuals for the both phases of testing. Factory acceptance testing of several projects and site acceptance testing of one project were observed to produce testing manuals.
Produced testing manuals are confidential and remain in the use of Valmet. Process of producing testing manuals and general content is included in published report of this thesis. Part of thesis was also electrical work safety in factory acceptance testing and deployment. Ensuring electrical work safety was acknowledged in testing manuals and electrical work safety is covered in report. ▼ The search for a ‘historical Moses’ is one which has been debated for several centuries. In spite of copious archaeological finds in Egypt and other parts of the ancient Near East, no material remains have been found to substantiate the Exodus story. Mythological stories from the ancient Near East bear striking similarities to the Moses narrative found in the Hebrew Bible. The inconsistencies found in the Hebrew Bible further hamper the attempt to find a historical Moses, instead Moses is found only in tradition. Taking these issues into consideration, other possibilities need to be investigated. This dissertation places Moses growing up in the court of Akhenaten an 18th Dynasty Egyptian Pharaoh in the middle of the 14th century BCE. The study investigates the possibility of Moses as a son of Akhenaten with Nefertiti as a stepmother, his own biological mother, a Mitannian princess, having died giving birth to Moses. Several similarities between Akhenaten and Moses are discussed throughout the study. The study moves into the 21st century with the groundbreaking discovery of DNA, which provides new conclusions which before were only debated.

▼ Les Oligosaccharides soluble (OS) sont essentiellement générés durant le processus de dégradation des N-glycoprotéines nouvellement synthétisées et mal conformées (ERAD) et durant la voie « turn-over » des glycoprotéines matures. Dans le but de déterminer si la modification post-traductionnelle O-GlcNAc est effectivement impliquée dans le processus de dégradation des N-glycoprotéines, nous avons analysé par spectrométrie de masse les OS provenant des cellules CHO après traitement par l’inhibiteur PUGNAc. Le PUGNAc ou « O-(2-acetamido-2-deoxy-D-glucopyranosylidene) amino-N-phenylcarbanate » est un inhibiteur puissant de l’O-GlcNAcase (OGA) qui catalyse l’hydrolyse du résidu O-GlcNAc des résidus sérine et thréonine des protéines O-GlcNAcylées.L’analyse par spectrométrie de masse révèle l’apparition d’une population d’OS de structures anormaux dans les cellules CHO suite au traitement par le PUGNAc. Cette population a été identifiée comme ayant des structures possédant des résidus GlcNAc au niveau de leur extrémité non-réductrice issues d’une dégradation lysosomale incomplète des glycoprotéines. Contrairement au PUGNAc, le NButGT, un autre inhibiteur de l’OGA, n’aboutit pas à l’apparition de cette population. Ainsi, Nous avons démontré que ces structures s’accumulent exclusivement dans la fraction membranaire conséquence de l’inhibition des β-hexosaminidases lysosomaux par le PUGNAc. Notre étude avait permis, d’un part, de mettre en évidence la capacité du PUGNAc de mimer une maladie de surcharge lysosomale et, d’autre part, de montrer un autre aspect des effets indésirables induits par le PUGNAc et qui nécessite d’être pris en considération lors de l’utilisation de cet inhibiteur.
Free oligosaccharides (fOS) are generated as a result of glycoproteins catabolism that occurs in two principal distinct pathways: the endoplasmic reticulum-associated degradation (ERAD) of misfolded newly synthesized N-glycoproteins and the mature N-glycoproteins turnover pathway. We analyzed fOS by Mass spectrometry in PUGNAc CHO treated cells in order to investigate whether O-GlcNAc modified proteins were involved in N-glycoprotein degradation process. The O-(2-acetamido-2-deoxy-D-glucopyranosylidene) amino-N-phenylcarbanate (PUGNAc) is a potent inhibitor of the O-GlcNAcase (OGA) catalyzing the cleavage of β-O-linked 2-acetamido-2-deoxy-beta-D-glucopyranoside (O-GlcNAc) from serine and threonine residues of post-translationally modified proteins. Mass spectrometry (MS) analysis revealed the appearance of an unusual population of fOS after PUGNAc treatment. The structures representing this population have been identified as containing non-reducing end GlcNAc residues resulting from incomplete lysosomal fOS degradation. Only observed after PUGNAc treatment, the NButGt, another OGA inhibitor, did not lead to the appearance of the population. O:54 antigen from Salmonella enterica. The wb*
O:54 operon (encoding wbbE, wbbF and mnaA) is found on a naturally-occuring plasmid (pWQ799). Previous research suggested only WbbE and WbbF are necessary for
O:54 antigen assembly due to the presence of a chromosomal gene encoding WecB, a functional homolog of MnaA. WecB and MnaA synthesize the essential UDP-ManNAc donor. However, this study determined optimal production of
O:54 in E. coli was dependent on the presence of MnaA. A growth defect was observed in cells producing
O:54 antigen, but unable to ligate it to the lipopolysaccharide acceptor (due to ΔwaaL). Cells recovered growth over time via mutations that turned off (or down)
O:54 antigen biosynthesis. During this study, a cysteine-free version of WbbF that retained function was established, providing a key resource for future topology mapping experiments.

La présence de cellules souches cancéreuses (CSC) favorise le maintien de la tumeur puis l’échappement tumoral responsable de la rechute des patients. Il est donc nécessaire d’identifier de nouvelles cibles thérapeutiques pour améliorer la prise en charge de ces patients. Dans ce contexte, l’immunothérapie, stratégie utilisant le système immunitaire pour traiter le cancer, représente une voie prometteuse.Nous identifions ici le OAcGD2 comme un nouvel antigène tumoral du GBM et a partir de biopsie tumorale provenant d’une cohorte de 37 patients. L’utilisation de primoculture de GBM nous permet de plus de démontrer que ce marqueur est également exprimé par les cellules souches de GBM. Nous apportons la preuve de concept de l’immunothérapie du GBM à l’aide d’anticorps monoclonaux anti-OAcGD2 in vitro et in vivo. A côté de la cytotoxicté immunologique, nous identifions un nouveau mécanisme de mort programmé, l’oncose, mis en jeu par les AcM anti-OAcGD2 permettant de sensibiliser ces cellules aux agents de chimiothérapie comme le témolozomide. L’utilisation de l’immunothérapie ciblant le marqueur gangliosidique tumoral OAcGD2 pourrait ainsi permettre d’améliorer l’efficacité de la thérapeutique actuelle (protocole de Stupp) du GBM
Glioblastoma multiforme (GBM) is the most common and agressive primary brain tumors in adults. Despite the concomitant use of surgery with radiotherapy and chemotherapy, the prognosis of patients remains extremely low. The presence of cancer stem cells (CSC) promotes the maintenance of the tumor and then the tumor escape responsible for the relapse of the patients. Therefore it is necessary to identify new therapeutic targets to improve the management of these patients. In the past few decades, immunotherapy represents an important part of treating certain type of cancer. It uses the immune system to treat cancer. Here, we found that O-acetyl GD2 (OAcGD2) is expressed in surgically resected human glioblastoma tissue. In addition, we demonstrated that 8B6 monoclonal antibody specific for OAcGD2 could effectively inhibit glioblastoma cell proliferation in vitro and in vivo. Mostly, we found that OAcGD2 was expressed on the GBM stem cells. We also observed that mAb 8B6 promoted the elimination of GBM via a oncosis-like mechanism. Moreover, this mechanism of programmed cell death induced by anti-OAcGD2 mAbs, sensitizes GBM cells and CSCs to chemotherapy agents such as temolozomide (TMZ). Taken together, these results indicate that O-acetylated GD2 represents a novel antigen for immunotherapeutic-based treatment of high-grade gliomas, and that anti-OAcGD2 mAbs combined with TMZ could enhance therapeutic response in GBM.

▼ Traditional U.S. historical scholars, as a matter of practice, carelessly push the history of the Mexican American community to the fringes of the collective historical consciousness of this country by selecting the common stories that students of all races learn. This study examines the problem of insufficient and inadequate historical scholarship pertaining to people of Mexican ancestry of the United States. By providing a local history of Mexican Americans in the Sacramento Valley this project provides a challenge to the legacy of historical exclusion by documenting the contributions and experiences of the Mexican American World War II generation and provides an alternative historical perspective to the World War II narrative.
