▼ CD46 is a ubiquitously expressed transmembrane protein in humans with a role in immune homeostasis. Originally identified as a complement regulator, CD46 has since been regarded a receptor for several pathogens and most recently, described as a T cell costimulatory molecule. Its coligation with CD3 and consequent cleavage from the T cell surface serves as a costimulatory stimulus for T cell activation. In addition, in the presence of IL-2, CD46 induces Tr1 cell differentiation which is characterised by low IFN-γ and high IL-10 secretion. CD3/CD46-induced Tr1 differentiation is defective in patients with MS, rheumatoid arthritis and asthma, highlighting the need to investigate the mechanisms involved in the regulation of the CD46 pathway. CD46 is a highly glycosylated protein with three N-glycosylation sites in the short consensus repeats and multiple O-glycosylation sites in the STP region. Previous data from the lab have shown that CD3 activation causes a change in CD46 glycosylation. Herein, I convey that this change is more pronounced in memory than naive CD4+ T cells and is mainly due to changes in CD46 O-glycosylation. Furthermore, these changes are required for the T cell responses triggered by CD46 costimulation including T cell activation and Tr1 differentiation. Interestingly, CD46 is recruited to the immunological synapse formed between a T cell and an antigen presenting cell and I illustrate that the STP region is needed for this also. These data suggest that the glycosylation status of CD46 regulates its function. In MS, vitamin D deficiency is considered to be a significant risk factor and many patients take vitamin D supplement to help manage their condition. Herein, I report that treatment of healthy and MS CD4+ T cells with vitamin D does not prevent T cell activation but it decreases adhesion molecule expression. Moreover, vitamin D supplementation in MS enhances CD46 cleavage. Therefore, vitamin D also plays a role in the regulation of the CD46 pathway and it would be interesting to investigate whether vitamin D affects CD46 glycosylation. During my MSc, I showed that a recombinant protein derived from adenovirus serotype 35 (which naturally binds CD46) known as Ad35K++ controls the CD46 pathway in CD4+ T cells. Lymphoma cells treated with Ad35K++ in combination with monoclonal antibody therapy rituximab have demonstrated increased sensitivity to rituximab and prove that virus-derived recombinant proteins that target CD46 have therapeutic potential. Considering the key role of CD46 as a T cell costimulatory molecule, I have investigated the effects of Ad35K++ on the CD46 pathway following its use in vivo and confirm CD46 is still cleaved from the cell surface and the cells still become activated. Overall these results provide insight into the mechanisms involved in the regulation of the CD46 pathway and highlight how it can be manipulated for therapeutic use.

▼ Many proteins of eukaryotic cells are known to be O-glycosylated. Glycoproteins with heavily O-glycosylated mucin domains provide important biological functions in a cell: i.e., protection from pathogens, cell-to-cell adhesion and intracellular protein trafficking. Mucin-type O-glycosylation occurs in the Golgi complex and begins with the transfer of GalNAc from UDP-GalNAc onto Ser/Thr residues of polypeptides. This step is catalyzed by a large family (20) called N-a-acetylgalactosaminyl transferases (ppGalNAc-T’s) and forms the GalNAc-a-O-Ser/Thr product. Subsequent elongation is performed by specific glycosyltransferases, producing a variety of glycans. Family members have been classified into peptide- and glycopeptide-preferring subfamilies, although both subfamilies possess variable activities against glycopeptide substrates. Structurally, 19 isoforms contain a C-terminal catalytic domain linked via a flexible linker to an N-terminal ricin-like lectin domain. The (glyco)peptide substrate specificities of the ppGalNAc-T transferases and the roles of the catalytic and lectin domains in glycopeptide glycosylation still remain largely unknown. Based on the systematic random peptide approach created by the Gerken Lab, I have determined the glycopeptide substrate specificities of several ppGalNAc-T isoforms. A series of (glyco)peptides were created in order to specifically probe the functions of the catalytic and lectin domains in terms of neighboring (1-5 residues) and remote prior glycosylation (6-17 residues) from an acceptor site, respectfully. Using several glycopeptide-preferring isoforms, glycosylation was observed from -4, -3, -1 and +1 relative to a neighboring GalNAc-O-Thr, which I attributed to specific GalNAc-O-Thr binding at the catalytic domain. The other series of glycopeptides contained a GalNAc-O-Thr near the C- or N- terminus of the substrate to address the directionality preferences of the lectin domain. Results with several peptide- and glycopeptide-preferring isoforms revealed preferences that varied among transferase isoform, where some preferred a C-terminally placed GalNAc-O-Thr, or a N-terminally placed GalNAc-O-Thr and others equally preferred the C-/N- terminally placed GalNAc-O-Thr. These directionality preferences are due to the GalNAc-O-Thr interactions at the lectin domain. Results of these studies revealed for the first time the site-specific glycopeptide glycosylation preferences by some ppGalNAc-T’s and has demonstrated that both domains of the ppGalNAc-T’s have specialized and unique functions that work in concert to control and order mucin-type O-glycosylation. ▼ William Grant Still (1895-1978) was one of the most prolific African-American composers of the 20th Century. In fact during his career, Still composed over 150 musical works. He was the first African-American to conduct a major American symphony orchestra, hear his own Symphony performed by a leading orchestra, have his opera produced by a major opera company, and view his opera broadcast on a national television network. As a result of these and many other musical accomplishments, Still has been titled, "Dean of African-American Composers." Still spent the majority of his life composing and arranging compositions that featured his unique style of incorporating Western harmonies with African-American blues. Although known primarily for his orchestral works, Still's vocal works have been integrated into the core repertoire of many college, university, and professional ensembles. The purpose of this study was to arrange five works by Still for brass quintet including Memphis Man (1923), Bayou Home (1944), Mississippi (1948), Elegy (1963), and All That I Am (1965). A secondary purpose was to present a biographical sketch of Still. A descriptive analysis about each of the arrangements for brass quintet was included along with the actual settings. Included in the document are performance editions of the five works. The vocal works, Memphis Man, Bayou Home, and Mississippi were created from versions of songs in William Grant Still, An Art Song Collection compiled by Celeste Anne Headlee with lyrics by Verna Arvey. Elegy was taken from William Grant Still: An Organ Collection and is the only work in the collection written for organ. All That I Am was transcribed from an edition for solo voice and piano edited by Celeste Anne Headlee. The spirituals included in this study that date from several different periods of his career are representative of many of his other compositions because they do not include extensive ornamentation or embellishment found in works by other composers. Still created passionate melodic lines and straightforward harmonies to communicate inherent emotion and powerful impact to the audience. In these settings, the melodies have been left unaltered.

Les alliages intermétalliques réfractaires à base de niobium sont considérés comme ayant un bon potentiel pour les applications à haute température grâce à un bon compromis entre une bonne résistance à haute température et une bonne ductilité à température ambiante. En outre, cette famille d'alliages présente également un point de fusion élevé et une faible densité. De ce fait, ils sont de bons candidats pour les applications dans les aubes de turbine à basse pression pour une plage de température comprise entre 800 et 1000 ° C. Le but de cette étude est d'étudier les changements de composition chimique, en particulier la précipitation de phase O-Ti2AlNb et les effets d'addition de silicium sur les microstructures et les propriétés mécaniques à haute température. L’étude commence par une première prospective sur les alliages montrant le plus de potentiel pour les applications visées. Les alliages étudiés doivent présenter un bon équilibre entre ductilité induite par le titane qui empêche la précipitation de la phase d-Nb3Al fragile, et une quantité élevée de niobium pour maintenir de bonnes propriétés mécaniques à haute température. Les alliages étudiés sont dopés avec 1 %at Si pour améliorer la résistance aux températures élevées et maintenir une ductilité acceptable à température ambiante. Nous nous intéressons également à l’influence des teneurs en aluminium et silicium sur les transformations de phase. Le système est renforcé par mise en ordre A2 ? B2, par effet de solution solide ou par durcissement structural de la phase O-Ti2AlNb. La cinétique de précipitation de cette phase O-Ti2AlNb est dépendante de la composition chimique. En particulier l’ajout de silicium permet un élargissement du domaine de précipitation et accélère sa cinétique de précipitation.

Niobium based refractory intermetallic alloys are considered as having great potentials for high temperature applications based on a good balance of high temperature strength and low-temperature damage tolerance. Moreover this family of alloys also exhibits a high melting point and a low density; hence they are good candidates for low pressure turbine blade applications over the temperature range of 800–1000 °C. The aim of this study is to investigate the chemical composition changes, especially the O-Ti2AlNb phase precipitation and the silicon addition effects on the microstructures and high temperature mechanical properties. This study starts with a first prospective on the chemical compositions of alloys showing the highest potential for applications wanted. The studied alloys show a good balance between the ductility at room temperature induced by titanium which stopped the fragile intermetallic d-Nb3Al precipitation; and a high niobium content to maintain good high temperature mechanical properties. ▼ Mucin type O-glycosylation is one of the most common and diverse protein modifications in humans. The process is initiated by a large family of N-acetylgalatosamine transferases (GalNAc-T1 to 20), that transfer the GalNAc sugar onto specific serine (Ser) and threonine (Thr) acceptor residues on target proteins. The underlying molecular mechanisms that govern acceptor Thr/Ser site selection by the GalNAc-T are not well understood. Mutations and abnormal expression of individual GalNAc-T isoenzymes lead to alterations in site-specific O-glycosylation, which have been associated with many cancers and metabolic syndromes.Previous substrate specificity studies have shown that the peptide sequence and the prior O-GalNAc glycosylation on a substrate dictate the acceptor site selection by GalNAc-Ts. Structurally, nearly all GalNAc-Ts contain a catalytic domain and a lectin domain connected by a flexible linker. The enzyme activity is enhanced by the lectin domain recognizing remote prior glycosylation (>5 residues away from the acceptor) in an N-or C-terminal direction, and a small number of transferases show rate enhancement by the catalytic domain recognizing neighboring prior glycosylation (<5 residues away). In this thesis we have begun to show how the acceptor site preferences of GalNAc-T isoenzymes are fine-tuned by a combination of multiple enzyme-substrate interactions, guided by enzyme domain specificities that ultimately results in a defined pattern of O-glycosylation.In chapter 1, we show that GalNAc-Ts have intrinsic preference for acceptor Thr residues over Ser residues independent of the flanking peptide sequences. Interestingly the Thr/Ser rate ratio varies between GalNAc-T subfamilies thus adding to the increasing dimension of GalNAc-T substrate specificity. In chapter 2, we show that GalNAc-T2’s preference for both N- and C-terminal remote prior glycosylation is primarily based on its short flexible linker that provides rotational and translational capacity to the lectin domain. This work exemplifies how a structural feature, very distant from the catalytic domain active site, is capable of tuning the activity and specificity of a GalNAc-T.In chapter 3 and 4, kinetic studies reveal that GalNAc-T4 and -T12 isoenzymes, which modify densely glycosylated substrates, are more active against di-glycopeptide substrates with two prior glycosylated O-GalNAc sites, compared to mono-glycopeptides. Specifically, GalNAc-T4’s highest preference is for the lectin domain recognizing remote glycosylation, whereas GalANc-T12’s preference is for the catalytic domain recognizing neighboring glycosylation. Este trabalho objetivou identificar quais pol?ticas educacionais inclusivas legitimam a escolariza??o do educando com autismo e que contribui??o os documentos internacionais e nacionais trouxeram ? consolida??o dessas pol?ticas, a pesquisa se desenvolveu por meio da Linha 4: Educa??o Especial e Inclus?o no Contexto Amaz?nico, pertencente ao Programa de P?s-Gradua??o em Educa??o da Universidade Federal do Amazonas, com bolsa de estudo da FAPEAM. O estudo prop?s uma busca no Minist?rio da Educa??o com o objetivo de conhecer documentos e diretrizes endere?adas ? condu??o metodol?gica do processo de escolariza??o desses indiv?duos. Para alcan?armos os objetivos propostos, optou-se pela pesquisa qualitativa, com enfoque dial?tico, nossa caminhada metodol?gica deu-se com an?lise documental, bibliogr?fica e pesquisa de campo. Para coletar os dados foi utilizado um roteiro de entrevista semiestruturada como sugere Bardin (2011), nos serviram como objeto de an?lise os documentos internacionais, nacionais, estaduais e as pol?ticas educacionais inclusivas que versam sobre o direito ? escolariza??o dos indiv?duos com autismo. No que compreende a educa??o especial e inclusiva, utilizamos como aporte te?rico os autores Pietro (2018), Garcia (2017), Baptista (2015), Matos (2012; 2013), Bobbio (2004), Bueno (2004), Saviani (2008), Alves (2011), Carneiro (2007), Bosa (2002), Orr? (2016; 2019), Schmidt (2013), Whitman (2015), Cunha (2016; 2017), Mantoan (2006; 2014), Capellini (2004), Ropoli (2010), Costa (2016), Mazzotta (1996), Manzini (2015) entre outros. Na an?lise de conte?do, confrontamos os dados da pesquisa, tendo como eixos tem?ticos norteadores a rela??o entre o professor do ensino comum e o professor do atendimento educacional especializado que atua nas salas de recursos multifuncionais e o posicionamento dos gestores sobre as pol?ticas educacionais inclusivas e seu impacto no contexto regional amaz?nico, al?m de tem?ticas que envolveram recursos did?ticos e a participa??o da fam?lia. O resultado da pesquisa nos possibilitou compreender a inclus?o dos alunos com autismo nas escolas da SEDUC em Manaus, bem como fomentar pesquisas envolvendo as tem?ticas do autismo e escolariza??o atrav?s das pol?ticas educacionais inclusivas em nosso estado. A crescente utiliza��o de tecnologias de telecomunica��es pela sociedade tem levado a que a ind�stria desenvolva infraestruturas que suportem esse crescimento. Os arm�rios exteriores de telecomunica��es s�o um elemento importante na rede de telecomunica��es, permitindo albergar e proteger os equipamentos necess�rios ao funcionamento da rede das condi��es adversas. Este g�nero de arm�rios dever� ter um correto desempenho t�rmico para garantir uma temperatura inferior a 55 �C, limite para o funcionamento dos equipamentos, e evitar um consumo demasiado elevado de eletricidade. Este estudo prop�e-se caracterizar o desempenho t�rmico do arm�rio com recurso estudos num�ricos realizados com o DesignBuilder/EnergyPlus. O desenvolvimento do modelo num�rico � acompanhado de estudos experimentais, utilizando um arm�rio real em funcionamento, de forma a garantir que o modelo � v�lido e que permite obter resultados fi�veis. Entre os principais resultados, pode-se observar que a utiliza��o de ventila��o mec�nica � eficaz na extra��o de calor do interior do arm�rio, por�m, existe um limite para o qual o aumento do valor de caudal de ar n�o se traduz, de forma significativa, em diminui��o da temperatura interior. As propriedades radiantes dos materiais utilizados na constru��o do arm�rio influenciam significativamente o comportamento t�rmico do arm�rio, sendo que, os materiais met�licos apresentam melhor desempenho que os n�o met�licos, dentro dos casos estudados. A localiza��o geogr�fica da EBT tamb�m � um fator fulcral para o comportamento t�rmico do arm�rio. Os resultados permitem elaborar, de forma quantitativa, uma caracteriza��o t�rmica do arm�rio e melhorar o perfil de utiliza��o do mesmo. Analisando os diversos gr�ficos relativos aos ensaios num�ricos efetuados torna-se poss�vel analisar de que forma os v�rios fatores afetam a temperatura no interior do arm�rio. Atrav�s de mais estudos seria poss�vel elaborar uma caracteriza��o mais completa, sendo poss�vel caracterizar mais casos.

The growing use of telecommunication technologies by society has led the industry to develop infrastructure to support this growth. The telecommunications outdoor cabinets are an important element in the telecommunications network, allowing host and protect the equipment needed to run the network in adverse conditions. The enclosure must have a proper thermal performance to ensure a temperature below 55 � C, limit for the operation of equipment, and avoid an excessive consumption of electricity. This study aims to characterize the thermal performance of the cabinet using numerical studies with DesignBuilder/EnergyPlus. The development of the numerical model is followed by experimental studies using a real cabinet in operation, to ensure that the model is valid and allows to obtain reliable results. Neste trabalho propomos a utiliza??o de redes neurais convolucionais para segmenta??o de ?reas desmatadas na regi?o do sul da Amaz?nia Legal em imagens de sat?lite Landsat-8. Para realiza??o deste trabalho foi constru?do um banco de imagens mosaico, composto por amostras de ?reas desmatadas e ?reas de floresta extra?das das imagens de sat?lite Landsat- 8, identificando as ?reas desmatadas atrav?s da utiliza??o dos dados de desmatamento do projeto PRODES do INPE. As arquiteturas de redes convolucionais utilizadas foram as propostas no trabalho de Serr?o et al. (2020) e de Miyagawa et al. (2018). O treinamento das redes foi realizado com 32 ?pocas utilizando m?todos de otimiza??o SGDM, RMSProp e ADAM e os m?todos de regulariza??o L2 e Dropout. Combinando as tr?s arquiteturas com esses m?todos, totalizaram 36 simula??es. Para avaliar o desempenho das arquiteturas para segmenta??o das ?reas desmatadas, permitindo uma compara??o entre os modelos, foi escolhida a m?trica acur?cia. Ap?s a avalia??o do desempenho dos modelos no conjunto de valida??o, seis deles foram selecionados para serem avaliados com o conjunto de teste. O modelo que apresentou o melhor resultado, uma acur?cia de 99.97%, foi o que utilizou a seguinte combina??o: CNN2 + RMSProp + Dropout. O resultado deste trabalho foi comparado com os resultados dos trabalhos de Ortega et al. (2019), Adarme et al. (2020) e De Bem et al. (2020) tendo obtido resultados superiores ?queles obtidos por esses autores. In this work, we propose the evaluation of convolutional neural networks architectures to segment deforested areas in the southern region of the Brazilian Legal Amazon, using Landsat-8 satellite images. In order to carry out this work, a mosaic image data set was elaborated, consisting of samples of deforested areas and forest areas extracted from the Landsat-8 satellite images. The deforested areas were identified through the use of deforestation data from the PRODES project at INPE. The architectures of convolutional networks used in our research were those proposed in the work of Serr?o et al. (2020) and Miyagawa et al. (2018). The training of the networks was carried out over 32 epochs using SGDM, RMSProp and ADAM optimization methods and the L2 and Dropout regularization methods. The combining of the three architectures with these methods, resulting in 36 simulations. To measure the performance of the architectures for segmented deforested region, allowing a comparison between the models, the accuracy metric was chosen. After evaluating the performance of the models in the validation set, six of them were selected to be evaluated with the test set. The model that presented the best result, with an accuracy of 99.97%, was the one that used the following combination: CNN2 + RMSProp + Dropout. Avec les récentes réformes, dont la loi Hôpital, Patient, Santé et Territoires (HPST) de Juin 2009, l’hôpital s’est transformé non seulement dans sa forme organisationnelle par la mise en oeuvre des pôles mais aussi dans son rapport à son environnement. Cette organisation polaire vise à rechercher une performance par une approche médico-économique. Ainsi les dispositifs de gestion ont été mis en place et cherchent à rendre compte de cette performance. Dans ce contexte, comment peuvent travailler des professionnels de champs aussi différents que celui de la gestion et celui du soin ? La gestion sous-entend une optimisation des moyens, alors que le service public défend en priorité des valeurs d’utilité sociale et d’attention aux usagers. Les acteurs du soin ont une forte identification professionnelle à cette mission d’utilité sociale. Dans ce travail, nous proposons d’identifier en quoi les décalages culturels au sein des pôles hospitaliers, participent à une perte de sens et de reconnaissance des agents à leur travail. Ces écarts culturels seront appréciés plus précisément au travers de l’appropriation des outils de gestion. Cette étude se base sur des entretiens réalisés au sein de différents hôpitaux (CHU et CH) et de secteur Médecine, Chirurgie, Obstétrique (MCO) et psychiatrie. Elle révèle lanécessité de pratiques managériales innovantes tournées notamment vers la gestion interculturelle afin que chaque acteur de ces nouvelles structures hospitalières se sente compris et intégré dans cette dynamique. L’étude proposée peut constituer un retour d’expérience intéressant pour mieux comprendre et améliorer l’utilisation des nouveaux outils de gestion. Des préconisations sont proposées et peuvent constituer des pistes de réflexion afin de répondre aux défis actuels de l’hôpital public.

With the latest reforms, among which the Hospital, Patient, Healthcare and Territory (HPST) Law, the French Hospital has changed not only the form of its organisation through the implementation of a polar organisation, but also its environmental concern. This polar organisation aims to strive for performance and efficiency via an managerial and medical approach. Thus, some management tools have been set up aimed in particular at giving feedback on such performance. In this context, how can professionals from such distinct fields as management and healthcare work? Management implies the optimization of resources when the public sector above all upholds values of social usefulness and patient care. Healthcare actors feel very much involved in this mission of social usefulness. In this work we propose to identify how cultural gaps within hospital poles contribute to a loss of meaning and acknowledgment among healthcare agents. Those cultural gaps will be examined more particularly through management tool appropriation. A habita??o social de baixa renda sofre atualmente no Brasil um grande d?ficit medido em fun??o da demanda populacional em crescimento, sendo que, atrav?s das pol?ticas p?blicas habitacionais n?o se consegue dar solu??o eficiente em termos de qualidade, tempo de execu??o e facilidades construtivas em conson?ncia com o meio ambiente e os n?veis necess?rios para o correto desempenho da qualidade de vida. Neste contexto, novos sistemas construtivos surgem como solu??es no mercado comercial, como ? o caso dos pain?is sandu?ches pr?-moldados aplicados a moradias de baixo custo. Estes sistemas consideram o planejamento, dimensionamento e detalhamento dos mesmos, sendo um dos motivos de inclus?o dentro desta pesquisa. Baseado nessa problem?tica, este trabalho tem como objetivo desenvolver e caracterizar pain?is sandu?ches constitu?dos por um n?cleo de EPS e faces de ferrocimento. Para atingir esse objetivo, inicialmente foi verificado a utiliza??o de dois tipos de refor?o (malhas met?licas) nas faces de ferrocimento. Com a melhor configura??o do refor?o, produziu-se o painel sandu?che, este ?ltimo foi caracterizado por ensaio mec?nico (flex?o) e ensaio f?sico (t?rmico). Ainda foi projetada uma habita??o de baixo custo em conson?ncia com as carater?sticas bioclim?ticas da cidade de Manaus, na qual ser?o implementados os pain?is sandu?ches como sistema de veda??o e divis?ria.

Brazil is currently suffering a large deficit in social housing, which can be measured based on the population growth, and on the to public housing policies through which it is not possible to provide efficient solutions in terms of quality, execution time and construction methods in line with environmentally friendly approaches and the necessary levels required to achieve a good quality of life. In the light of this, new construction systems have been emerging in the market as solutions, such as precast sandwich panels applied to low-cost housing. This system considers planning, dimensioning and detailing, and for that reason it was included in this research. Based on the problem previously stated, the objective of this work is to develop and characterize sandwich panels made of an expanded polystyrene (EPS) core and ferrocement facings. To accomplish this objective, two types of reinforcement (metallic meshes) for the ferrocement facings were evaluated. With the best reinforcement arrangement, the sandwich panel was produced, and it was characterized by means of mechanical testing (bending) and physical testing (thermal). Moreover, a low-cost housing was designed in line with the bioclimatic characteristics of Manaus, a city in which sandwich panels will be implemented for external and internal walls.

A presente pesquisa aborda a tem?tica do uso de res?duos cer?micos provenientes da ind?stria de tijolos cer?micos da regi?o metropolitana de Manaus, como material pozol?nicos e agente mitigador para a rea??o ?lcali-agregado. As adi??es reativas s?o eficientes em inibir ou reduzir as rea??es expansivas resultantes da combina??o dos ?lcalis do cimento com agregados potencialmente reativos. Afim de avaliar o uso de res?duos cer?micos como adi??o pozol?nica e redutor de rea??es expansivas em materiais ciment?cios foram realizadas an?lises qu?micas, f?sicas e mineral?gicas, deste modo, buscando-se determinar o seu ?ndice de atividade pozol?nica, bem como avaliar a poss?vel a??o mitigadora da rea??o ?lcali-agregado. A partir dos resultados obtidos com a moagem controlada e a caracteriza??o qu?mica e estrutural, atrav?s das t?cnicas de Fluoresc?ncia de raios-x (FRX), Difra??o de raios-x (DRX), Termogravimetria (TG), Derivada Termigravim?trica (DTG), An?lise T?rmica Diferencial (DTA) e Espectroscopia de Infravermelho por Transformada de Fourier (FTIR), al?m disso, avaliou-se o potencial pozol?nico das adi??es por teor de hidr?xido de c?lcio fixado por Chapelle.Os resultados mostram que o RC atendeu aos requisitos qu?mico-f?sicos para pozolana. Os ensaios de petrografia e de determina??o da mitiga??o de barras de barras de argamassa com o uso de 50% do res?duo cer?mico indicaram baixa reatividade. This research addresses the issue of the use of ceramic waste from the ceramic brick industry in the metropolitan region of Manaus, as pozzolanic material and mitigating agent for the alkali-aggregate reaction. Reactive additions are efficient in inhibiting or reducing expansive reactions resulting from the combination of cement alkalis with potentially reactive aggregates. In order to evaluate the use of ceramic waste as a pozzolanic addition and reducing expansive reactions in cementitious materials, chemical, physical and mineralogical analyzes were carried out, thus seeking to determine its pozzolanic activity index, as well as to evaluate the possible mitigating action of alkali-aggregate reaction. From the results obtained with controlled grinding and chemical and structural characterization, using the techniques of X-ray fluorescence (FRX), X-ray diffraction (DRX), Thermogravimetry (TG), Thermigravimetric derivative (DTG), Analysis Differential Thermal (DTA) and Fourier Transform Infrared Spectroscopy (FTIR), in addition, the pozzolanic potential of additions by Chapelle's fixed calcium hydroxide content was evaluated. The results show that the RC met the physical-chemical requirements for pozzolana. The petrography and mitigation determination tests for mortar bars using 50% of the ceramic residue indicated low reactivity. V Sloveniji je že dlje časa samooskrba s hrano pomembna tema, ki je na nizki ravni. Posebej nizko je ta stopnja vidna pri zelenjavi, zato smo se odločili pristopiti k analizi problematike. V raziskavi smo k problematiki pristopili »od spodaj navzgor« in sicer z namenom, da najdemo empirične rešitve, ki kmetovalcem omogočajo pogoje, s katerimi bi dvignili nizko samooskrbo. V ta namen smo razvili 5 matematičnih modelov, s katerimi se bo analizirala problematika samooskrbe. 1. model je namenjen analizi ekonomske upravičenosti v raziskavo vključenih kultur, kot tudi posameznih sort in hibridov. Rezultati le-tega predstavljajo vhodne podatke za naslednje faze razvoja modelov. Rezultati analize modela 1 kažejo ekonomsko upravičenost vseh analiziranih zelenjadnic. V 2. fazi smo razvili 2 modelni orodji. S prvim ocenjujemo potrebne količine zelenjadnic za zagotovitev oskrbe po zelenjavi za določen teden v letu, v naboru razpoložljivih kultur. Drugo modelno orodje 2. faze pa izmed razpoložljivih kultur in njihovih hibridov/sort oceni potrebne površine, s čimer zadovoljimo želene potrebe po oskrbi z zelenjavo. 3. faza je namenjena izboru ustrezne metode ciljnega programiranja, na podlagi katere bi lahko ocenili ustrezne sorte/hibride. Za oceno problema smo uporabili 3 metode ciljnega programiranja (tehtano ciljno programiranje, prioritetno ciljno programiranje in Čebiševo ciljno programiranje). Kot najustreznejše se je pokazalo tehtano ciljno programiranje, ki je doseglo nekoliko slabše rezultate v primerjavi s prioritetnim ciljnim programiranjem, ki pa je med izračunom doseglo večje število napak. Kot najslabše se je pokazalo Čebiševo ciljno programiranje. V fazi 4 je sledilo načrtovanje kolobarja znotraj posameznega koledarskega leta. S pomočjo razvitega orodja smo sestavili 538 letnih scenarijev, ki jih potem v fazi 5 združimo v 183 5-letnih kolobarjev. Rezultati kažejo, da na 874,66 ha obdelovalnih površin zagotavljamo popolno samooskrbo z zelenjavo za območje Pomurja od 1. maja do 13. novembra. V nadaljevanju raziskave na osnovi s. w. o. t. analize stanja trga na organizacijske strani pridelave in prodaje predlagamo ukrepe, ki stimulirajo pridelavo. Prva je uvedba indeksa izkoriščenosti njivskih površin kot ukrepa, s katerim bi lahko stimulirali pridelavo preko neposrednih plačil, ki so drugače razdeljena. Drugi ukrep predstavlja povezovanje pridelovalcev. Tretji ukrep je organiziranje skupnega prostora, kjer se srečata ponudba s strani pridelovalcev in povpraševanje s strani odkupa. Kot ključen ukrep pa je seveda ureditev cen.

Modelos probabil?sticos de t?picos s?o modelos estat?sticos capazes de identificar t?picos em uma cole??o de texto. Eles s?o amplamente aplicados em tarefas relacionadas ? ?rea de Processamento de Linguagem Natural, uma vez que capturam com sucesso rela??es latentes por meio da an?lise de dados n?o rotulados. Entretanto, solu??es anal?ticas para a infer?ncia Bayesiana desses modelos s?o geralmente intrat?veis, dificultando a proposta de modelos probabil?sticos que sejam mais expressivos. Neste cen?rio, os Autocodificadores Variacionais (ACVs), m?todos que empregam uma rede de infer?ncia baseada em redes neurais respons?vel por estimar a distribui??o a posteriori, tornaram-se uma alternativa promissora para inferir distribui??es de t?picos em cole??es de texto. Estes modelos, contudo, tamb?m introduzem novos desafios, tal como a necessidade de distribui??es cont?nuas e reparametriz?veis que podem n?o se ajustar ?s distribui??es reais dos t?picos. Al?m disso, redes de infer?ncia tendem a apresentar um problema conhecido como colapso de componentes, onde apenas alguns t?picos contendo poucos termos correlacionados s?o efetivamente extra?dos. Para tentar evitar estes problemas, prop?em-se dois novos m?todos de t?picos. O primeiro (GSDTM) ? baseado em uma distribui??o cont?nua pseudocateg?rica denominada Gumbel-Softmax, capaz de gerar amostras aproximadamente categ?ricas, enquanto o segundo (LMDTM) adota uma mistura de distribui??es Normais-log?sticas, que pode ser adequada em cen?rios onde a distribui??o dos dados ? complexa. Apresenta-se tamb?m um estudo sobre o impacto que diferentes escolhas de modelagem t?m sobre os t?picos gerados, observando um compromisso entre coer?ncia dos t?picos e a qualidade do modelo gerador. Por meio de experimentos usando duas cole??es de dados de refer?ncia, tr?s m?tricas distintas de avalia??o quantitativa e uma inspe??o qualitativa, mostra-se que o modelo GSDTM supera de forma significativa os modelos de t?picos considerados estado da arte em grande parte dos cen?rios de teste, em termos de coer?ncia m?dia de t?picos e perplexidade.

Probabilistic topic models are statistical models which are able to identify topics on textual data. They are widely applied in many tasks related to Natural Language Processing due to their effective use of unlabeled data to capture latent relations. Analytical solutions for Bayesian inference of such models, however, are usually intractable, hindering the proposition of highly expressive text models. In this scenario, Variational Auto-Encoders (VAEs), where an artificial neural-based inference network is used to approximate the posterior distribution, became a promising alternative for inferring latent topic distributions of text documents. These models, however, also pose new challenges such as the requirement of continuous and reparameterizable distributions which may not fit so well the true latent topic distributions. O presente relatório de estágio pretende descrever todas as atividades que acompanhei e realizei durante os 6 meses de estágio curricular, realizado no âmbito do curso de Mestrado em Análises Clínicas, da Faculdade de Farmácia da Universidade de Lisboa. O estágio foi realizado no laboratório de análises clínicas Nova Era-Luz e durante o decorrer do mesmo, acompanhei as valências de Bioquímica, Hematologia e Imunologia. Inicialmente permaneci na secção de Bioquímica onde são realizadas análises principalmente em amostras de soro (Olympus AU400 e MiniCap) e urina (Aution Max AX4280 – determinação de parâmetros químicos). A secção de hematologia é a que engloba um maior número de técnicas manuais e de aparelhos automáticos. Os aparelhos automáticos presentes nesta secção são o Sysmex XT2000i (realização de hemogramas); o VesMatic 30/30 Plus (determinação da velocidade de sedimentação); o Adams A1C HA8160 (doseamento da hemoglobina glicada); o Hb Gold (electroforese de hemoglobinas); e o Sysmex Ca500 (estudo da coagulação sanguínea). Por fim, passei pela secção de imunologia onde as análises são realizadas principalmente em dois aparelhos, o Advia Centaur e o VIDAS. O relatório encontra-se divido pelas áreas referidas e em cada uma delas apresento um fundamento teórico da técnica, princípios e ainda uma explicação dos resultados obtidos em cada um dos aparelhos presentes nas várias secções bem como das técnicas manuais.

The present report will describe all the activities that I have performed during my 6 months internship as a master student of clinical analysis at the Faculty of Pharmacy of the University of Lisbon. I worked as an intern at Nova Era-Luz laboratory and I did several procedures in three specific areas: Biochemistry, Hematology and Immunology. At the beginning, in the biochemistry section, I started by doing serum (Olympus AU400 e MiniCap) and urine (Aution Max AX4280 – chemical parameters) analyses. Hematology department is the one that has a major number of manual techniques and automatic devices which include Sysmex XT2000i (CBC); VesMatic 30/30 Plus (sedimentation rate); Adams A1C HA8160 (glycated hemoglobin assay); Hb Gold (hemoglobin electrophoresis) and Sysmex Ca500 (blood coagulation assay). Lately, in the immunology section I took contact with the two principle devices that are used for this type of analysis: Advia Centaur and VIDAS. So, this report is divided in the three sections mentioned above and in which one of them I present a theoretical foundation of the technique, principles and an explanation of the obtained results either by device or manual technique.

Introdu??o: A dor lombar pode atingir at? 80% da popula??o mundial, sendo a principal causa de afastamento laboral. A fisioterapia utiliza de recursos e t?cnicas para tratar a dor lombar, dentre elas a Osteopatia. Pertencente ?s terapias manuais, a Osteopatia pode auxiliar no tratamento da dar lombar cr?nica inespec?fica atrav?s de t?cnicas de manipula??o vertebral. Objetivo: Investigar os efeitos de duas t?cnicas manipulativas da Osteopatia, alta velocidade e baixa amplitude e m?sculo energia, em trabalhadores feirantes com dor lombar cr?nica inespec?fica. M?todos: Ensaio cl?nico randomizado, duplo cego. A amostra foi composta por 30 trabalhadores feirantes da cidade de Manaus-AM, apresentando dor lombar cr?nica inespec?fica, selecionados aleatoriamente para um ?nico dia de interven??o. Foram aplicados os question?rios de anamnese, incapacidade funcional (Roland-Morris), qualidade de vida (SF-36) e medos e cren?as (FABQ-Phys e FABQ-Work) para an?lise descritiva. Para a compara??o dos efeitos pr? e p?s-interven??o foram aplicados a escala de intensidade da dor (NPRS), o teste de Schober para mobilidade e o dinam?metro isocin?tico Biodex-3 para avaliar o pico de torque isom?trico e isocin?tico a 60 e 120?/s para flexores e extensores do tronco. Ap?s a avalia??o, os sujeitos foram randomizados em 3 grupos de interven??o: Grupo Alta Velocidade e Baixa Amplitude (G1); Grupo M?sculo Energia (G2); e Grupo Placebo (G3). Todos foram reavaliados imediatamente ap?s a interven??o para as vari?veis de dor, mobilidade e pico de torque. Resultados: Ambos os grupos (G1 e G2) que sofreram interven??o apresentaram redu??o significativa da intensidade da dor (p<0,01; p=0,02, respectivamente) e aumento da mobilidade ap?s interven??o (p<0,01; p=0,01, respectivamente), sendo que o G1 apresentou melhora significativa quando comparado com o G3 para dor (p=0,033) e mobilidade (p<0,01) e com o G2 para mobilidade (p<0,01). As vari?veis relacionadas ao PT n?o apresentaram diferen?a significativa em nenhum dos grupos. A raz?o flex?o-extens?o (F/E) dos PT isom?trico e isocin?tico a 60?/s dos m?sculos do tronco apresentou valores abaixo das refer?ncias (0,49; 0,63, respectivamente), demonstrando um desequil?brio muscular, com for?a superior de extensores do que flexores. Conclus?o: A t?cnica de AVBA e a t?cnica de ME foram capazes de reduzir a dor e aumentar a mobilidade da coluna lombar imediatamente ap?s uma interven??o em trabalhadores feirantes com dor lombar cr?nica inespec?fica, sendo que a t?cnica de AVBA apresentou resultados superiores quando comparado com a t?cnica de ME para a vari?vel de mobilidade.
