p>Existe uma busca crescente por energia limpa e renov?vel, que deve ser acompanhada pelo desenvolvimento de dispositivos eficientes de armazenamento de energia. Um dispositivo de grande interesse ? o capacitor eletroqu?mico, especificamente, os pseudocapacitores que utilizam uma variedade de materiais como PANI ou ?xidos met?licos como eletrodos. A PANI ? um pol?mero intrinsecamente condutor que pode ser sintetizada pela rota qu?mica ou eletroqu?mica. A primeira ? a mais adequada para se obter um pol?mero com alto peso molecular, o que ? primordial para a aplica??o sugerida. Nesse intuito, este trabalho apresenta os resultados de estudos das varia??es das concentra??es do agente dopante, agente oxidante e temperatura de
s?ntese qu?mica da PANI para futuras aplica??es eletroqu?micas. Os materiais obtidos foram caracterizados por MEV, espectroscopia de UV-vis, FTIR, voltametria c?clica (-0,2 a 1,2 V), carga/descarga galvanost?tica e imped?ncia eletroqu?mica. As caracteriza??es estruturais e ?pticas comprovaram a obten??o do pol?mero no estado condutor para todas as condi??es de
s?nteses. As an?lises morfol?gicas revelaram a diminui??o do tamanho de gr?nulos conforme o aumento da concentra??o de ?cido sulf?rico (agente dopante). Enquanto que a segunda vari?vel de
s?ntese mostrou tamanhos menores de gr?nulos na concentra??o de 2,5 mol.L-1 de PSA (agente oxidante). J? a varia??o de temperatura apresentou gr?nulos na superf?cie de nofibras nas temperaturas de
s?ntese de -5 e 0?C, em contrapartida, ocorreu a forma??o de estruturas na forma de placas em 5?C. Os estudos por voltametria c?clica mostraram diferentes respostas para cada condi??o de
s?ntese, tendo potenciais dos estados redox da PANI de acordo com a literatura e aus?ncia de picos redox em outras condi??es, em sua maioria, observou-se processos at? a velocidade de varredura de 25 mV.

p>The growing search for clean and renewable energy should be accompanied by the development of efficient energy storage devices. A device of great interest is the electrochemical capacitor, specifically the pseudocapacitors that use a variety of materials such as PANI or metal oxides as electrodes. PANI is an intrinsically conductive polymer that can be synthesized by the chemical or electrochemical route. p>A utiliza??o mais ampla de fontes renov?veis acarretou em uma nova gera??o de dispositivos eletroqu?micos com alto desempenho em armazenamento e convers?o de energia como supercapacitores eletroqu?micos, c?lulas a combust?vel e pilhas port?teis. Os ?xidos met?licos de baixo custo, como NiO, CoO2, Ni(OH)2, MnO2, bem como pol?meros condutores como polianilina,
s?o estudados como materiais de eletrodo para capacitores eletroqu?micos com intuito de superar o alto custo associado aos demais materiais. Para esta finalidade, o di?xido de mangan?
s ? um material atrativo para estudos cient?ficos devido ao seu baixo custo, baixa toxicidade, menor agressividade ao meio ambiental e por apresentar boa capacit?ncia te?rica. No presente trabalho o MnO2 foi eletrodepositado utilizando a t?cnica de voltametria c?clica aplicando uma faixa de potencial an?dico de 0 a 1,5 V e como substrato foram usadas placas tit?nio eletropolidas. Al?m de eletrodepositar o material, esta t?cnica foi utilizada para estudar o eletrodo obtido na faixa de potencial de 0 a 0,8 V. Essa t?cnica ? extremamente importante por fornecer informa??es sobre a natureza eletroqu?mica das esp?cies analisadas, tais como, processos redox, capacit?ncia, ciclabilidade etc. O processo de eletropolimento do substrato foi analisado usando a t?cnica de microscopia de for?a at?mica a partir da qual constatou-se o melhor substrato para receber a camada de filme do ?xido. A an?lise por microscopia eletr?nica de varredura, possibilitou observar altera??es na mofolgia dos materiais ? medida que havia mudan?as na concentra??o e composi??o das solu??es precursoras, sendo que os filmes dos eletrodos que n?o continha o eletr?lito suporte apresentaram maior uniformidade. A an?lise por difra??o de raios x, mostrou a fase cristalina do MnO2, bem como, a dist?ncia interplanar dos picos observados. Os estudos eletroqu?micos foram realizados pela t?cnica de voltametria c?clica, a qual mostrou os perfis capacitivos do eletrodo investigado e por meio de carga-descarga galvanost?tica, foi poss?vel calcular a capacit?ncia espec?fica que atingiu n?veis de 375 F.g-1. A espectroscopia de imped?ncia eletroqu?mica permitiu estudar as propriedades el?tricas do material, assim como a estabilidade c?clica do material. Contudo, os resultados obtidos indicam a import?ncia do estudo de par?metros como a varia??o da concentra??o da solu??o precursora e do eletr?lito suporte, permitindo obter resultados de capacit?ncia espec?fica do filme eletrodepositado de MnO2, cujo valores superam o reportado em algumas literaturas.

▼ Music has been widely used in human anaesthesia, due to low cost, ease and safety. Additionally, its beneficial effect on physical and biochemical parameters, analgesia, and minimization of adverse effects of anesthetics and analgesics, makes perioperative exposure to music an attractive complementary tool during anaesthesia.The aim of this research was to study the effect of music, provided during the perioperative period, on factors associated with anaesthesia and analgesia in dogs exposed to nociceptive stimuli under experimental and clinical conditions. In the first part, 20 Beagles were applied to identical nociceptive stimuli: 3 groups were formed, each of which consisted of the same 20 dogs. The first group listened to Chopin Music (CM), the second to Mozart Music (MM) and the third was the No-Music (NM) group. Dogs were premedicated with acepromazine (0.03 mg/kg) and butorphanol (0.1 mg/kg) intramuscularly; anaesthesia was induced with propofol and maintained with isoflurane while fentanyl (1 μg/kg) was provided intravenously as rescue analgesia. Anesthetic depth was monitored by use of the bispectral index (BIS), and autonomic nervous system responses (HR, BP, RR) were used to monitor the level of analgesia. Furthermore, measurements of Substance-P concentration in the dogs' serum were carried out in order to objectively evaluate pain during the intraoperative period. In the second part of the study, 24 female dogs, undergoing ovariohysterectomy, were divided into two groups of 12. Dogs of the first group were exposed to Mozart Music (MM), while those of the second were not exposed to any musical stimulus during the perioperative period (NM). The experimental design was almost identical to the first part. The BIS was not used, however the parameters evaluated were the same as in the first part of the study.In the first part of the study, the groups of dogs exposed to music achieved higher levels of sedation, required less propofol, isoflurane and fentanyl and exhibited less pronounced changes in BIS values (p <0.05), in relation to the group not exposed to music. In the second part of the study, the dogs of the MM group exhibited less fluctuations in arterial pressure than those the NM group (p<0.05). Finally, a statistically significant effect of time on the alterations of Substance-P concentrations was observed, regardless of exposure to music or not.Classical music provided during the perioperative period in laboratory dogs, results, when compared to dogs not exposed to music, in higher levels of sedation, lower consumption of propofol, isoflurane and fentanyl and less alterations in BIS values. Also, exposure to Mozart's music resulted in less fluctuations of substance P concentrations. However, these positive effects of classical music were apparent in light but not deeper anaesthesia in dogs subjected to ovariohysterectomy, probably due to dose-dependent suppression of auditory function caused by isoflurane. s-operatÃrio foi avaliado o consumo de analgÃsicos em 6 e 24 horas, alÃm da incidÃncia de nÃuseas, vÃmitos e prurido. Le ipotesi fino ad ora elaborate, nonostante siano lontane dal potersi definire conclusive, prevedono l'apprezzamento del margine di solvibilità relativo al rischio di tasso d'interesse (IRR) assumendo che la struttura per scadenza sia oggetto di shift paralleli della curva. Noti i limiti dell'approccio deterministico ed in forza dei principi fino ad ora consolidati, il presente elaborato si propone l'obiettivo di quantificare il requisito di capitale di una compagnia operante nel ramo vita, a fronte dell'IRR, ipotizzando che la dinamica dei tassi sia governata da un processo stocastico nella forma del modello Cox Ingersoll e Ross (
CIR). Le simulazioni sono state sviluppate sugli equilibri patrimoniali di una teorica compagnia, al fine di apprezzare in maniera asettica il contributo dell'impostazione promossa.

p>Nowadays, the financial guarantees system of insurance market is being interested by a Community reform process (Solvency II project). Even if the current hypothesis are far to be definitive, the present guidelines state that the Solvency Capital Requirement (SCR) related to Interest Rate Risk (IRR) has to be quantified assuming deterministic shocks to the yield curve. The aim of the thesis is to improve the assessment of SCR connected to IRR, calculating interest rates according to Cox, Ingersoll and Ross (
cir) stochastic model. Simulations are developed on the asset liability equilibria of a theoretical life insurance company, in order to better appreciate the SCR algebra sensitivity to changes in
CIR model parameters.

p>High entropy alloys are a new class of materials. This dissertation deals with high entropy alloys synthesized utilizing mechanical alloy. Elementary powders (Co, Cr, Cu, Fe, and Ni) are ground in planetary and high-energy ball mills, in both, with process control agent (stearic acid and toluene). Elementary powders are taken out at particular time intervals for the phase analysis through an X-ray diffractometer. The particle morphology was scrutinized by Scanning electron microscopy. The phase analysis through an X-ray diffractometer reveals the formation of a face-centered cubic (FCC) phase. The decrease in the size of the crystallite is observed, by the Rietveld method, with the increase of the milling time. When analyzing the process variables, we found that the combination of the high energy ball mill and toluene as the process control agent was the best result. The high entropy alloy was obtained in 5 hours, and toluene prevented contamination of the CoCrCuNiFe alloy both by the action of oxygen present in the atmosphere and by the materials that make up the tungsten carbide vial set with balls.

▼ The growing demand on enantiomerically pure compounds has stimulated the interest for the development of methodologies to obtain these compounds. Among them, asymmetric catalysis is one of the most employed tools. In this technic, the choice of the chiral ligand is fundamental to obtain high levels of activity and enantioselectivity. In this context, this thesis is focused on the synthesis of several families of highly modular chiral ligands from readily available starting materials. Particularly, we worked on the synthesis of
P-oxazoline (
P= phosphine, phosphinite, phosphite, phosphoroamidite),
P-other N-donor groups (
P= phosphite, phosphoroamidite, phosphonite and N= thiazole, sulfoximine, hydrazone, amine, pyridine),
P-thioether (
P= phosphine, phosphinite, phosphite) and a family of
P*-stereogenic phosphine-phosphite ligands. These ligands have been applied in the Rh- and Ir-catalyzed hydrogenation of functionalized and minimally functionalized olefins, Pd-catalyzed allylic substitution reaction and Pd-catalyzed decarboxylative protonation. Furthermore, in some cases, DFT studies in combination with experimental ones have been performed to better understand the origin of the obtained enantioselectivities or in order to guide the ligand optimization.

studies in Financial Economics. The first chapter focuses on the
predictive power of the implied correlation index on the future
S&
P 500 Index returns. The second chapter investigates a
nonlinear contemporary relationship between stock returns and oil
price changes. The last chapter discusses the relationship between
impact trading costs and a number of market factors that affect the
costs. In the first chapter, I investigate the predictive power of
the implied correlation index on the future
S&
P 500 Index
returns. This new index was launched by Chicago Board Option
Exchange following 2007-2008 financial crisis. As it is derived
from the
S&
P 500 Index option price and the option prices of
the largest 50
S&
P Index stocks, it is widely regarded by
market participants as a gauge of average expected future stock
return correlation. Because of its role in measuring systematic
risks, any changes in this index may provide useful information
about the future market movements. Motivated by this index'
s
forward-looking characteristic, I propose a linear regression where
the future
S&
P 500 Index multi-period return is regressed on a
number of controls such as the current period changes of
S&
P
index and the implied correlation index etc. I use weekly data and
three different sample splits for in-sample estimation and
out-of-sample performance evaluation. I find that the implied
correlation index is informative for the period 2007-2009 in
predicting the
S&
P 500 Index returns of 28 to 39 weeks. My
model consistently outperforms the random walk model using the
Superior Predictive Ability test. This implied correlation index is
also useful in predicting the
S&
P index future multi-week
returns for the period 2009-2011 and a longer time span from
2007-2011. I also do a test for the Efficient Market Hypothesis by
incorporating the implied volatility index in the regression. There
is no evidence supporting the view that the market is efficient for
those time periods. In the second chapter, I estimate a nonlinear
contemporary relationship between stock returns and oil price
shocks. Previous studies on this issue suffer a number of
limitations. For example, they do not control the factors
potentially driving the economy and the oil market simultaneously.
Although, Kilian and Park (2009) does a good job in identifying the
relationship by distinguishing different oil market shocks, they
use a linear regression framework and do not address the
contemporary relationship. Considering the different impacts of
different-size oil shocks on stock returns, I propose a two-step
estimation procedure for identifying their relationship. In the
first step, I follow Kilian and Park'
s methodology, i.e. a
structural vector autoregression, to estimate the demand-specific
oil shocks. During the second step, I use a nonparametric quantile
regression to estimate the relationship between stock returns and
the estimated exogenous oil price shocks. This way, I can control

▼ The growing interest in obtaining enantiomerically pure compounds in obtaining enantiomerically pure compounds has led to a significant development in the field of asymmetric catalysis. In this context, this thesis is focused on the synthesis of several families of highly modular chiral ligands from readily available compounds. Specifically, we worked on the synthesis of thioether-phosphite ligands, phosphite-pyridine, phosphite-triazole and hidroaximide and thioamide ligands. They all have in common that are solid, stable and therefore easy to handle. These ligands have been applied in the Rh- and Ir-catalyzed hydrogenation of functionalized and unfunctionalized olefins, in the Ru- and Rh-catalyzed asymmetric transfer hydrogenation of ketones, in Pd-catalyzed allylic substitution reactions and in the Ni-catalyzed addition of organoalumininum to aldehydes. Moreover, in some cases DFT studies have been performed to speed up the optimization of ligands. Hence, a variety of chiral chemical compounds of high synthetic interest (i. e. alcohols, functionalized and non-functionalized alkanes, substituted allyl) lhigh enantioselectivities were achieved and in some cases the products were obtained in their enantiomerically pure form(> 99% ee).

▼ ATPDases ou ATP-difosfohidrolases são enzimas que clivam o ATP e o ADP a AMP e Pi e estão envolvidos em inibição da agregação plaquetária. No parasita Schistosoma mansoni nosso grupo identificou e clonou o gene da ATPDase1, e a proteína foi localizada na superfície do tegumento. Recentemente, clonamos o gene da ATPDase2 usando a informação do banco de dados de ESTs de
S. mansoni e imunolocalizamos a sua proteína também no tegumento. ATPDase2 foi encontrada em ambas as membranas do tegumento basal e apical juntamente com a ATPDase1, entretanto ATPDase2 somente foi encontrada no espaço sincicial do tegumento. A presença de ambas as enzimas sobre a superfície externa do tegumento sugere um maior papel sobre a regulação de abundância de nucleotídeos. Análise da expressão de ambos os genes foram realizadas por RT- PCR em tempo real usando RNA de ovos, miracídeo, cercária, esquistossômulo e verme adulto. Os resultados mostraram que o gene da ATPDase1 foi mais expresso em ovos (7 vezes), adulto (6 vezes), cercária (3,5 vezes) e esquistossômulo (1,5 vezes) quando comparado ao miracídio, que foi tomado como referência. O gene da ATPDase2 foi mais expresso em ovos (16 vezes), cercária (11 vezes), miracídio (7 vezes) e verme adulto (2 vezes) quando comparado a esquistossômulo, mostrando que ambos os genes são modulados ao longo de seus estágios de ciclo de vida . Para maior caracterização destas enzimas, elas foram expressas heterologamente na levedura Pichia pastoris como proteínas de fusão com cauda de 6 histidinas e as proteínas recombinantes foram purificadas por cromatografia de afinidade com resina de Ni-NTA. As ATPDases recombinantes foram obtidas de forma ativa e medições de atividade enzimática foram realizadas. ATPDase1 - mostrou atividades ATPásica e ADPásica em torno de 650 e 160 nmoles Pi.min -1. mg-1 , respectivamente. ATPDase2 teve atividades ATPásica e ADPásica na faixa de 1050 e 250 nmoles Pi.min-1.mg-1 , respectivamente. Adicionalmente, atividades UTPásica e UDPásica também foram encontradas nestas enzimas. Estudos de dicroísmo circular com estas duas enzimas elucidaram suas estruturas secundárias. Com isto, ATPDase1 (S66 to Q507 ) teve alfa-hélice (7 %), folha-beta (45 %) e estrutura randômica (48 %), e a ATPDase2 (N83 a K564 ) mostrou conter alfa-hélice (14 %), folha-beta (33 %) e estrutura randômica (53 %). s mostramos que a ATPDase2 é secretada pelo parasita no meio, de forma similar como descrito para as ATP-difosfohidrolases humanas CD39L2 e CD39L4. s pétrophysiques et élastiques des calcaires urgoniens de Provence. Une approche intégrée et multi-échelle est proposée pour caractériser les propriété
s pétrophysiques et élastiques des carbonates. Cette étude est basée sur des mesures de vitesse d’ondes
P (Vp) et
S (Vs) à l’échelle du laboratoire (centimètre–décimètre) et du terrain (mètre–décamètre). En laboratoire, les Vp et Vs ainsi que l’anisotropie sont mesurées sur des plugs et sur des carottes, en utilisant différentes fréquences centrales ultrasonores. Sur le terrain, l’approche consiste à mesurer les Vp et Vs entre deux puits distant de 2 m sur une profondeur de 14 m. Les mesures sont ensuite interprétées en fonction de la géologie observée aux échelles macro- et microscopiques. Les principaux résultats montrent que les Vp et Vs moyennes sont indépendantes de l’échelle de mesure, car elles sont dictées par la porosité. L’anisotropie causée par les fractures (15%) et les hétérogénéité
s (5%) se manifestent par une variabilité autour des vitesses moyennes. L’approche adoptée ici a permis de définir les interactions entre les propriété
s de la matrice, les hétérogénéité
s, les fractures et les propriété
s élastiques des roches carbonatées. Elle a montré que les propriété
s élastiques tout comme les structures géologiques varient en fonction de l’échelle.

p>Carbonate reservoirs are also exploited for water production, geothermal energy, and carbon geological storage. Their Geophysical characterization remains challenging because of complex diagenetic history. This work offers new insights into the characterization of petrophysical, and elastic properties of the Urgonian limestones in the Provence region. An integrated multi-scale approach is proposed to characterize carbonate rocks petrophysical and elastic properties. This study relies on
P- and
S-wave velocity (Vp and Vs) measurements carried out at laboratory (centimeter–decimeter) and field (meter–decameter) scales. Laboratory scale Vp, Vs, and anisotropy are measured on plugs and cores, while on the field they are measured between two boreholes (crosshole) over a distance of 2 m and 14 m depth. The measurements are then compared to the geology from the macro- to the microscopic scale. The main results show that the average Vp and Vs are porosity related, and are independent from scale. Anisotropy caused by fractures (15%) and heterogeneities (5%) is responsible for variations around the mean velocities. The approach adopted during this work has enabled to scope out the interplay between matrix properties, heterogeneity, fracturing, and elastic properties in carbonate rocks. It has shown that the elastic properties evolve with scale as well as the geological structures.

▼ Supramolecular chemistry has become a rapidly
growing field since the Nobel Prize was awarded to the work of
Lehn, Pedersen, and Cram in 1987 for their pioneering work on crown
ethers and non-covalent host-guest interactions. Metal-ligand
interactions represent a particularly useful and well-studied means
of preparing supramolecular polymers from telechelic building
blocks. This present work focuses on utilizing main-chain
metallo-supramolecular polymers or small molecules, based on
2,6-bis(1’-methylbenzimidazolyly)pyridine (Mebip) ligands, to
create materials aimed at applications such as light healable
polymers, or the detection of chemical warfare
agents. With the objective to develop new
organic/inorganic hybrid materials, which combine good mechanical
properties and other attractive properties with ease of processing,
new classes of metallo-supramolecular polymers were explored. The
general design approach merged the structure of known polymer
systems, for example semiconducting polymers (poly(
p-phenylene
ethynylene)
s) or thermally stable hydrocarbons (poly(
p-xylyene)
s),
with the advantages of a dynamic (reversible) polymerization
process, to allow access to materials that are otherwise difficult
to process. The new telechelic metallo-supramolecular polymers
could be readily solution-processed and represent illustrative
examples for metallo-supramolecular polymers with good mechanical
properties, while maintaining the properties of parent
polymer. Polymers with the ability to repair
themselves after sustaining damage are poised to extend the
lifetime of materials used in a wide range of applications.
Reported here is the development of metallo-supramolecular polymers
that can be mended through exposure to light. Upon exposure to
ultraviolet light, the metal-ligand motifs are electronically
excited and the absorbed energy is converted into heat, causing
reversible disengagement of the metal-ligand motif and a temporary
reduction in the molecular weight. As a result, defects heal
quickly, with high efficiency, and if needed repeatedly.
A modular sensory system, for the detection of
organophosphates, which utilizes a multi-metal/multi-ligand
approach is presented. The sensing scheme relies on fluorescent
sensor complexes of Mebip ligands and selected metal ions.
Detection of organophosphates is accomplished through competitive
coordination of the analyte with the metal. Mixing and matching
consciously designed ligands and carefully selected metal ions
allows the formation of sensor complexes, which allow
organophosphate detection with high selectivity and
sensitivity.

p>This study investigated the biological activity of etanolic extract of leaves and stem of Mimosa debilis (leguminosae). The eluate of etanolic extract (hexan, chloroform, etil acetate, acetone and methanol) were obtained for chromatography in silica gel column. The test of substance class revealed the presence of terpenes and steroids. The antibacterial activity were investigated against the gram positive microorganisms
S. pneumoniae and
S. aureus and the gram negative microganisms E. coli and
P. aeruginosa in diffusion in disc test and dilution broth test. The diffusion in disc test was made with etanolic extract and eluate. Its was vewrified the formation of an inhibition zone only when the discs were impregned with 2,7 mg the extract per disc. All the extracts, except the metanolic eluate revealed positive results against the gram positive microorganisms. There was no formation of inhibition zone, in the used concentration, in the gram negative microorganisms culture. Dilution broth tests were made only with extracts that revealed the bests results in diffusion disc test. The eluate of etila acetate was choosen to verify the inhibition of growth in
S. pneumoniae and
S. aureus. The quantification was obtained for optical density. The results showed that the evaluate of leaves and stems on M debilis inhibited sigificatively the growth of
S. pneumoniae and
P. aeruginosa.

p>O presente estudo investigou atividade biológica de extrato etanólico de folhas e caules de Mimosa debilis (Leguminosae). Os eluatos do extrato etanólico (hexano, clorofórmio, acetato de etila, acetona e metanol) foram obtidos por meio de cromatografia em coluna com sílica gel. Testes de classe de substância revelaram presença de terpenos e esteróides. Atividades antibacterianas foram investigadas contra os microorganismos gram-positivos Streptococcus pneumoniae e Staphylococcus aureus e os microorganismos gram-negativos Escherichia coli e Pseudomonas aeruginosa em testes de difusão em disco e teste de diluição em caldo. O teste de difusão em disco foi realizado com extrato etanólico e todos os eluatos. Verificou-se a formação de zona de inibição somente quando os discos foram impregnados com 2,7 mg de extrato. Todos os extratos com exceção do eluato metanólico revelaram resultados positivos contra os microorganismos gram-positivos. Não houve formação de zona de inibição, nas concentrações utilizadas, em culturas de microorganismos gram-negativos. Teste de diluição em caldo foi realizado apenas com o extrato que revelou melhores resultados no teste de difusão em disco. Neste contexto, o eluato de acetato de etila foi eleito para verificação da inibição de crescimento de
S. pneumoniae e
S. aureus. A quantificação foi obtida por meio da medida de densidade óptica. Os resultados mostraram que o eluato EtOAc folhas de M. debilis inibiu significativamente (
p 0,05; teste-t e teste Tukey) o crescimento de
S. pneumoniae e
S. aureus.

▼ Over the last three decades, the medical community
has seen a drastic increase in the number of
antibiotic resistant
infections, particularly caused by the ESKAPE (Enterococcus
faecium,
Staphylococcus aureus, Klebsiella penumoniae,
Acinetobacter baumannii, Pseudomonas
aeruginosa and Enterobacter
spp.) pathogens. While all six species are known to cause an array
of human disease, I have been primarily concerned with bacteremia
and skin and soft tissue
infections associated with A. baumannii,
S. aureus, and P. aeruginosa. Bacteremia and skin
based infections
have the highest mortality rates and exhibit physiological
differences compared
to laboratory culture conditions. Indeed, the
antibiotic resistant nature of the ESKAPE
pathogens, combined with
the dwindling antibiotic pipeline, has resulted in a need for new
therapeutic options, thus my research has focused on the
development of three new antimicrobial
therapies. First, a topical
ointment containing mupirocin and neomycin has been shown to have
a
wide spectrum of activity, with efficacy against S. aureus, A.
baumannii, and P. aeruginosa, and
has been shown to clear
colonization of multi-drug resistant S. aureus. A second topical
ointment
containing compounds found to be effective against
Pseudomonas biofilms also demonstrates
efficacy against a murine
wound model infected with A. baumannii, P. aeruginosa, and S.
aureus. The third therapy was the identification and
characterization of two compounds with
activity as efflux pump
inhibitors for A. baumannii and P. aeruginosa that could be used
as
adjunctive therapy in combination with conventional
antibiotics. Seven genes that are likely to be
the targets of
these compounds have been identified, allowing for an early model
of their cellular
mechanism of action for future testing. All
together the three newly described therapeutic
candidates we have
developed may provide new strategies for the therapeutic
intervention of
extremely antibiotic resistant bacterial
species.

▼ <p>M.A. p><p>This study is a critical assessment and analysis of Xhosa poetry that addresses social and political concerns with particular reference to the poetry of Qangule (1970), Mtuze (1982, 1990) and Xozwa (1994 ). Chapter one introduces the study by presenting its aim, scope, method of research, theory, definition of concepts and significance of the study. Chapter 2 focuses on the theoretical framework of the study. It discusses the theory of Marxism, which is used as a basis of this study, by explaining the concept of Marxism, its historical background, assumptions, methodology and Marxism and poetry. Chapter 3 deals with social concerns that are addressed by Mtuze (1982 and 1990) and Xozwa (1994 ). These social concerns are divorce and alcohol usage. The discussion in this chapter focuses on how divorce affects members of the family concerned, especially children of divorced parents. It also looks at how alcohol is used in society. While the discussion in this chapter acknowledges moderate use of alcohol, it focuses more on how this substance may also be abused. It looks at how alcohol abuse affects individuals and society economically and how dangerous it is to the health and lives of people. Chapter 4 focuses on political concerns as addressed by Qangule (1970) and Mtuze (1982). It assesses and analyses poems that address the oppression of black people during the apartheid regime in South Africa. Concerns like citizenship and deprivation of freedom, labour conditions and language are discussed in this chapter. Chapter 5 is a concluding statement which gives a summary of the study, evaluation of Xhosa poetry with regard to addressing the concerns discussed in the study and recommendations regarding future research.p>

▼ Mixed parasitic infections are common in many parts of the world. Plasmodium falciparum and Schistosoma mansoni are co-endemic parasitic infections with broad global distribution. However, little is known about how concurrent infections affect the immunity to and/or pathogenesis of each other. Thus, this study was undertaken to examine the parasitological, histopathological and hematological conditions that occur in Schistosoma mansoni-Plasmodium berghei co-infected mice. Mice infected with
S. mansoni cercariae were divided into two groups, which were super-infected at weeks 4 and 7 post-infection with
P. berghei. Giemsa stained blood specimens, hematoxyline and eosin stained liver and brain tissues were used for the analysis of parasitaemia, liver granuloma and cerebral sequestration of malaria parasites, respectively. In addition to these, hemoglobin level, percentage weight loss, eggs per gram of liver and survivability of
P. berghei infected mice were used as parameters for the comparison of co-infected and mono-infected groups. Sampling of hemoglobin and weight measurements were done at days 3, 5, 7, 9 and 11 post-
P. berghei infection. Sampling of blood for malaria parasitaemia was done at days 5, 7, 9 and 11 post-
P. berghei infection. Independent t-test analysis of the results showed that co-infection of mice with
P. berghei, 7 weeks post-
S. mansoni infection resulted in significant (
P<0.05) increase in malaria parasitaemia, severe loss of hemoglobin and weight, lower number of schistosome eggs per gram of liver, and decrease in granuloma size and number. In addition, reduction of malaria parasite sequestration in the brain with resultant increase in the life span of the infected mice was associated with co-infection. However, there was insignificant difference during co-infection of mice with
P. berghei 4 weeks post-
S. mansoni infection. The overall interplay between these two infections may be explained immunologically, with malaria infection down-regulating delayed hypersensitivity reaction, which would lead to reduction in granuloma formation around schistosome eggs. Similarly, chronic schistosome infection may modulate host immunopathological responses that would lead to cerebral sequestration. Taken together, the study showed schistosome and malaria co-infections to profoundly affect each other. These findings can have implications in treatment and in the efficacy of vaccines against schistosomiasis and malaria in co-endemic localities.

p>In this Work Project, I evaluate the relevance of the debate around fair value’
s Level II and Level III assets and liabilities. The literature outlines some qualitative shortcomings of this valuation technique. However, it seems important to understand how those assets and liabilities affect the balance sheet from a quantitative point of view. The data collected indicate that 10% of the balance sheets of Standard & Poor’
s 500 companies is evaluated using fair value measurement. My analysis reveals that the market prices are positively associated with fair value. It shows they are highly and positively sensible to Level III assets, suggesting that shortcomings outlined in the literature concerning the poor reliance in those inputs are not translated in the market behavior. The market even reacts in the opposite way as it approves the use of Level III inputs.

▼ Like all plants, grapevines are host to a plethora of microorganisms. During the winemaking process, the microflora of the grape surface can be transferred into the fermentation environment. While the yeast Saccharomyces cerevisiae is the main driver of alcoholic fermentation during wine production, it has become apparent that other grape and cellarderived microbes can have a significant impact on fermentation and the resulting wine. This impact is manifested in the sensorial differences between wines resulting from inoculated fermentations and spontaneous ‘wild’ fermentations and is attributed to the various secondary metabolites and enzymes these microorganisms produce. Carbohydrate active enzymes can affect the composition of grape juice by altering the extractability and organoleptic properties of many wine compounds. Of these enzymes, glycoside hydrolases mediate hydrolysis of glycosides and are found ubiquitously through nature. Micro-organisms, especially those associated with plants, use glycoside hydrolases to afford access to plant resources by degrading the plant cell wall and similarly soil microbes utilise these enzymes during degradation and composting of plant material. During winemaking, glycoside hydrolases have the potential to affect the breakdown of complex sugars in plant cell walls, which can aid in juice extraction and clarification. Glycoside hydrolases can also mediate the release of volatile flavour and aroma compounds from glycosyl-linked, non-volatile precursors. These enzymes therefore represent an attractive target for biotechnological applications. A combination of metagenomics and synthetic biology were used to explore the enzymatic potential of two grapevine derived environments, an Chardonnay grape must (CGM) and a mixed varietal grape marc (MVGM), with the aim of identifying novel enzymes with potential applications as winemaking adjuncts. The CGM was specifically selected as a source of fungal microbiota native to the grapevine, whilst the grape marc was chosen as a source of plant decomposing microorganisms from a glucose-limited environment. Whole genome sequencing and bioinformatic techniques were used to probe the genomic landscape of these two environments, identify enzymes of interest, assess the types of microbiota present in the microbial communities and where possible assemble microbial genomes. The two environments contained very distinct microbial communities. As expected, the Chardonnay must community comprised a high portion of fungal genomes whereas the MVGM was dominated almost exclusively by bacteria. Five candidate polygalacturonases, with potential applications in wine clarification, and seven β-glucosidases, with potential applications in wine aroma and flavour, were chosen for heterologous expression in Pichia pastoris and Saccharomyces cerevisiae. ▼ Pyroxasulfone is a new pyrazole herbicide that controls weeds by inhibiting very long chain fatty acid synthesis. This mechanism of action places pyroxasulfone in the Weed Science Society of America (WSSA) group 15 or Herbicide Resistant Action Committee (HRAC) group K3 herbicides that include
s-metolachlor and dimethenamid-
p. Dimethenamid-
p and
s-metolachlor are referred to in literature as acetamide, acetanilide, chloroacetamide, or chloroacetanilide herbicides. In this thesis, these two herbicides are referred to as chloroacetamide herbicides based on the HRAC classification. The soil interactions of pyroxasulfone were evaluated and compared to
s-metolachlor and dimethenamid-
p to better understand how pyroxasulfone will behave under various field conditions Pyroxasulfone was compared with these two standard herbicides because of their similar mechanisms of action, use patterns, potential for use in similar cropping systems, and similar weed control spectrums. Sorption coefficients were determined for 25 different soils to evaluate relative differences in binding among pyroxasulfone, dimethenamid-
p, and
s-metolachlor. Across all soil types, the relative order of binding was pyroxasulfone=dimethenamid-
p <
s-metolachlor. Pyroxasulfone and dimethenamid-
p were not statistically different in terms of their binding; however,
s-metolachlor binding was statistically greater than both dimethenamid-
p and pyroxasulfone. For all three herbicide, organic matter was the only soil property which was highly and significantly correlated to herbicide adsorption; all other soil properties correlated with herbicide adsorption could be explained by the correlation of OM and those soil properties. Based on the water solubility of these three herbicides, we expected the order of binding to be dimethenamid-
p <
s-metolachlor < pyroxasulfone. This study displayed the unique characteristics of pyroxasulfone in that it has the lowest water solubility of the three herbicides, yet sorption coefficient values indicate that pyroxasulfone is only loosely adsorbed by soil. Reduced soil binding along with a higher unit of activity makes pyroxasulfone a potent herbicide that provides comparable weed control when applied as low as one-eighth of typical application rates for other chloroacetamide herbicides. Field studies were conducted in 2009 and 2010 at two contrasting field sites to evaluate the dissipation and movement of pyroxasulfone and
s-metolachlor in the top 30 cm of the soil profile. The site at the horticultural farm (HORT) has a Nunn clay loam soil (Argiustoll), whereas the site at the Limited Irrigation Farm (LIRF) has an Olney fine sandy loam soil (Haplargid). Dissipation half-lives (DT50) were the shortest at the Hort farm site with the heavier textured clay loam soil and increased moisture content. The LIRF site with lighter textured sandy loam soil and decreased moisture resulted in extended DT50 values and more variation between replicates for both herbicides. ▼ A radiação tem sido utilizada para buscar o aumento de compostos com atividade antimicrobiana e antioxidante em extratos de plantas. Como Syzygium cumini (L.) Skeels apresenta atividades antioxidante, antimicrobiana, antidiarreica e adstringente já conhecidas, o objetivo deste trabalho foi avaliar os metabólitos secundários encontrados nos extratos irradiados com 60Co nas doses de 6, 8 e 10 kGy de raiz e caule de
S. cumini, analisar as propriedades citotóxica e antioxidante e quantificar a atividade antimicrobiana nos extratos irradiados. Para a análise qualitativa, foi realizada a cromatografia em camada delgada. Foram realizadas atividade hemolítica e anti-hemolítica. Na dosagem de compostos fenólicos foi utilizado o reagente de Folin-Ciocalteau. Para a quantificação dos flavonóides, as misturas extrato-reagente permaneceram por 30 minutos no escuro e a absorbância foi medida em 420 nm. O método do DPPH aplicado à
s concentrações dos extratos irradiados permitiu a avaliação quanto à capacidade do sequestro do radical. No método do radical ABTS, a mistura extrato-solução foi lida na absorbância de 734 nm. A atividade antioxidante total foi avaliada a partir do método do Fosfomolibdênio. Para a quantificação dos taninos, a mistura vanilina-ácido sulfúrico foi adicionada aos extratos com respectivas leituras em 500 nm. A avaliação da atividade antimicrobiana dos extratos foi testada nas concentrações de 2000 a 3,9 μg/mL contra isolados clínicos de
S. aureus e
P. aeruginosa. Os resultados fitoquímicos qualitativos revelaram a presença de flavonóides, derivados cinâmicos, triterpenos, esteróides, mono e sesquiterpenos, cumarinas, proantocianidinas condensadas e leucoantocianidinas nos extratos irradiados. Os melhores valores de atividade hemolítica foram os extratos irradiados na dosagem de 125 μg/mL, que resultou em 0% de hemólise. No caso de atividade anti-hemolítica, os extratos irradiados não apresentaram efeito significativo quando comparados com os extratos controle, contudo alíquotas menores de extrato, principalmente para caule, ocasionaram a melhora do efeito citoprotetor. Os maiores valores de compostos fenólicos foram encontrados nos extratos de raiz irradiados, todavia a irradiação causou a diminuição das concentrações de compostos fenólicos nos extratos de caule. Para flavonóides, o extrato irradiado com 8 kGy de raiz e com 6 kGy de caule obtiveram os melhores resultados quando comparados ao extrato não irradiado, enquanto que os taninos foram enriquecidos no extrato de caule irradiado a 8 kGy. O sequestro de radicais livres, na atividade do radical DPPH, manteve valores próximos de 90% para as concentrações acima de 125 μg/mL. Os resultados do radical ABTS indicaram grande capacidade de sequestro para extrato de raiz irradiado com 10 kGy, confirmado com os resultados do TEAC. Na atividade do fosfomolibdênio, a maior capacidade antioxidante foi verificada para o extrato de caule irradiado com 8 kGy. ▼ Uma ferida é por definição uma perda da continuidade da pele ou tecido mole. Quando o processo fisiológico normal de cicatrização é alterado, estamos perante uma ferida crónica. As feridas crónicas tornaram-se um problema de saúde pública a nível mundial, quando associadas a outras comorbilidades e ao risco aumentado de infeção. Estima-se que cerca de 1 a 2% da população mundial experienciou uma úlcera de perna. As úlceras de perna, tal como as restantes feridas crónicas, quando associadas a infeções bacterianas causam as mais variadas consequências: atraso do processo de cicatrização, alteração na qualidade de vida dos doentes e familiares, aumento do tempo de internamento e em casos mais extremos, causar morte. O objetivo deste trabalho consistiu em determinar o perfil de suscetibilidade dos isolados mais prevalentes em úlceras de perna na comunidade. Foi realizado um estudo retrospetivo, reportando os dados recolhidos no laboratório Avelab entre Janeiro de 2016 a Dezembro de 2018. Este estudo incluiu 398 indivíduos com suspeita de infeção bacteriana em úlceras de perna. Foram realizados exames bacteriológicos de exsudados de úlcera de perna com avaliação microscópica do esfregaço (coloração de Gram) e exame cultural dos exsudados purulentos. O teste de suscetibilidade aos antimicrobianos (TSA) foi realizado pelo método de difusão em disco de Kirby-Bauer. Das 398 amostras analisadas obtiveram-se 325 amostras positivas para infeção e 73 negativas. Relativamente à
s amostras positivas, verificou-se maior prevalência de úlceras de perna no género masculino (53%). A média de idades no género masculino foi de 76, enquanto no género feminino foi de 72. Os microrganismos mais prevalentes e responsáveis pela infeção das úlceras de perna foram
S. aureus (33,5%),
P. aeruginosa (28,7%) e Proteus mirabillis (10%). No estudo de TSA para
S. aureus verificou-se maior suscetibilidade a Trimetropim/Sulfametaxazol, Gentamicina e Tetraciclina; maior resistência foi
verificada a Eritromicina e Penicilina. Em relação a
P. aeruginosa verificou-se que apresentou suscetibilidade para a maioria dos antibióticos testados, com exceção de Ciprofloxacina que apresentou maior número de resistências. A distribuição de úlceras de perna é semelhante entre géneros, surgindo maioritariamente em população idosa. Os perfis de suscetibilidade obtidos para
S. aureus e
P. aeruginosa foram idênticos aos já descritos na literatura, com maior resistência de
S. aureus aos beta-lactâmicos e
P. aeruginosa à
s quinolonas. O uso combinado de antibióticos poderá ser mais eficaz no tratamento de úlceras de perna.

▼ The purpose of this quantitative study was to examine the relationship between the Capital Asset Pricing Model’s risk indicator beta and the average monthly returns for stocks in the S&P 100. The problem addressed was that low beta stocks produced higher returns than high beta stocks. The study was conducted using the S&P 100 constituents. The study expanded the research literature regarding the beta anomaly and found a statistically significant result for an association between beta and average monthly returns for stocks in the S&P 100. The study has implications for investors and financial practitioners as to whether beta can still be used as a risk indicator.

Herein, a portfolio selection methodology updating a reasonably diversified selection of competing

P 500 constituents within and across various predefined industry groups and which produced above average long-term returns with minimized downside-risk, is proposed. Within the various predefined groups of stocks, Simugram methods are used to model and optimize on the distribution of returns up to and including a horizon of interest. Improvements to previous methods are focused toward calibrating the sampling distribution based on an empirical dataset within the various groups comprising the investor'
s portfolio, optionally allowing for a varying sampling frequency as dictated by the various group dynamics. By combining within-group optimization alongside with the capability of exiting aggressive long-strategies at seemingly riskier times, focus is on providing more frequent updates on a list of constituents with improved performance in both terms of risk and return.

▼ Ce mémoire a pour objectif de bonifier les connaissances quant à la prévision de la prime de risque du marché des actions au Canada. La méthodologie présentée s’appuie sur l’hypothèse que les variables, permettant de prévoir la prime de risque, caractérisent la conjoncture économique et sont définies comme des variables d’état dans le modèle de Merton (1973). Les deux modèles de prévision utilisé
s peuvent être différencié
s par leur caractère univarié et multivarié. Le second modèle fait usage de l’analyse en composantes principales afin de créer des facteurs capturant la majeure partie de la covariance des groupes de variables. On identifie trois groupes de variables spécifiques; ces groupes se distinguent par leur caractère macroéconomique, technique ou leur lien avec le sentiment du marché. Une innovation est présentée dans la création de cinq séries de données mensuelles afin de refléter le sentiment du marché des actions canadiennes. Une approche globale est employée dans le but de prévoir la prime de risque mensuelle du
S&
P/TSX, puis une approche segmentée répète le même processus pour dix secteurs d’activité
s. Pour chacune de ces deux approches, une série de tests hors échantillon, « out of sample » en anglais est étudiée. Le pouvoir de prévision de la prime de risque des groupes de variables est alors testé en contexte de récession et de période d’expansion. Finalement, le gain d’utilité des modèles est calculé à l’aide d’une simulation de l’allocation d’un portefeuille en fonction des prévisions préétablies.
De façon globale, bien que plus d’indicateurs techniques soient statistiquement significatifs, les variables macroéconomiques semblent mieux performer dans la prévision de la prime de risque du Canada sur la période observée. Le ratio dividende-prix, le ratio de distribution des bénéfices et une prime de crédit semblent dominer les autres variables. L’utilisation d’indicateurs techniques est particulièrement intéressante lors de la prévision sectorielle, le momentum sur douze mois se distinguant de façon notable.

▼ In this study I study the effect of board effectiveness, measured by Board Shareholder Confidence Index (BSCI) on firm performance and risk. I find that there is modest positive relationship between BSCI total scores and firm performance, which is similar to the earlier findings. More importantly, I show a strong relationship between board effectiveness ratings and firm risk. This relationship is tested using a panel regression, a two stage least square and simultaneous equation modelling. The findings are robust to all three econometric techniques. I further explore the relationship between individual sub-scores and find similar results in sub-score analyses. The relationship holds using multiple measures of risk and return, giving us a comprehensive picture of the hypothesized relationship.

▼ The Common Water Snake, Nerodia sipedon sipedon, is one of six subspecies, and the only species of Nerodia to inhabit West Virginia. Because it is abundant and information on the species is lacking within the state, it was used as my study organism. The effectiveness of manual palpation, the most commonly used method in snake diet related studies, was tested against museum dissection by comparing presence and absence of gut contents. Nineteen individuals of 76 manually palpated collected snakes were compared to 6 individuals of 37 dissected snakes using a Pearson chi-square analysis. Results suggested manual palpation is an effective method to obtain diet data, X2 (1, N=135) = 0.7296, p= 0.05. In addition, morphological data were collected from field specimens and used to determine the best predictor of sex, as well as to obtain information on average snout-vent length, total length, and mass of N. s. sipedon in West Virginia. Field notes of cover object use, behavior, and other herpetofauna from the field site were also reported. Data collected from this study will provide information on the effectiveness of manual palpation and information on the natural history of N. s. sipedon in West Virginia.
