▼ La temperatura es un parámetro importante para el desarrollo de la digestión anaeróbica, que es la base de la producción de biogás; puede limitar sectores para aplicar la tecnología, regular la eficiencia, ayudar en la eliminación de patógenos, afectar el tiempo de retención de la materia e incrementar la producción de biogás. Este trabajo se desarrolló para evaluar la dinámica de producción de biogás en función del régimen térmico, estableciendo la relación entre las temperaturas que afectan a un biodigestor tipo chino y proponer zonas en Chile que posean mayor aptitud para aplicar esta tecnología. El biodigestor utilizado con capacidad de 10 m3, se alimentó tres veces por semana con una mezcla de relación 3:1 agua/estiércol (v/v). Se realizaron monitoreos en dos periodos, febrero a julio (2011) y enero a agosto (2012); en este último se instalaron sensores de registro continuo (temperatura y humedad) y se realizaron análisis de la composición del biogás generado cada semana. En el primer periodo se aplicaron correlaciones lineales a las variables, obteniendo que la temperatura del aire afectó en mayor proporción a la temperatura de descarga, estanque y la del biodigestor. Para el segundo periodo se analizaron las tendencias de las variables, se aplicaron análisis de anomalías para series de tiempo, promedios móviles y ajustes funcionales de las variables más relevantes en el estudio. De los resultados se obtuvo, que la temperatura interna del biodigestor mantuvo una temperatura promedio de 22°C y la temperatura media del suelo se ajustó mejor a la temperatura media máxima del aire. Los análisis de composición fluctuaron de 55 a 85% de CH4 (metano) y de 16 a 35% de CO2 (dióxido de carbono). La zonificación se realizó aplicando el “índice térmico” propuesto en un estudio previo, pero esta vez con información térmica actualizada para todo el país; obteniendo que a partir de la región de Los Lagos y a lo largo de la cordillera de Los Andes, no es apta la aplicación de esta tecnología. Así es posible afirmar que estos biodigestores favorecen la estabilidad térmica interna y que Chile posee un importante potencial para su implementación.; Temperature is an important parameter for the development of anaerobic digestion, base of biogas production. ▼ The genus Salmonella belongs to the Enterobacteriaceae family and comprises two species: S. bongori and S. enterica. Both species include more than 2500 serovars. Within S. enterica serovars, the most studied are S. Typhi, S. Typhimurium and S. Enteritidis, the latter being the main causative agent of salmonellosis worldwide. During its infective cycle, Salmonella needs to adapt to variations in different environmental conditions, such as changes in pH, osmolarity and oxygen availability. In addition, these conditions serve as environmental signals that modulate the expression of virulence factors. One of these factors is the lipopolysaccharide (LPS), the main component of the envelope of Gram negative bacteria. The LPS presents three structural domains: the O antigen (AgO), the core region and lipid A, also called endotoxin. The latter anchors the LPS to the outer membrane, and is a known virulence factor responsible for the induction of the immune response. The lipid A domain is capable of undergoing covalent modifications, which modulate its toxicity and helps bacteria to evade antimicrobial agents and the immune system of the host. One of the main environmental cues faced by Salmonella during its infectious cycle is the oxygen availability. During the aerobic-anaerobic transition participates the global regulators ArcA and Fnr. Results from our laboratory show that changes in the polymerization degree of the AgO in response to oxygen availability is modulated by these regulators. This observation led us to examine whether global regulators Fnr and ArcA are involved in the control of probable oxygen-dependent modifications of lipid A in response to oxygen availability. The main goal of this study was to determine the effect of oxygen availability on covalent modifications of lipid A in S. Enteritidis and the participation of transcriptional factors ArcA and Fnr in this process. First, we mounted a method for LPS extraction followed by a hydrolysis step to obtain a highly-purified lipid A that can be used to study the composition of this molecule by MALDI-TOF. Thus, in aerobiosis we observed four major signals corresponding to hexa-acylated and hepta-acylated lipid A molecules and their corresponding hydroxylated species. Under this environmental condition, hydroxylated species were more abundant than non-hydroxylated species. In the case of anaerobic cultures we detected the same four signal, but this relationship is reversed, being more abundant the nonhydroxylated species. To determine the participation of ArcA and Fnr in the modulation of covalent modifications of lipid A, we generated ΔarcA and Δfnr mutant strains and a strain overproducing ArcA (parcA). Our results indicate that both regulators are involved in controlling structural changes of lipid A in response to oxygen availability. ▼ Catalytic and regulatory properties of phosphofructokinase (PFK) (EC 2.7.1.11), pyruvate kinase (PK) (EC 2.7.1.40), and creatine kinase (CK) (EC 2.7.3.2) from the heart of the red-eared turtle (Pseudemys scripta elegans) were studied. Particular attention was given to those properties of the enzymes which could help to explain the high glycolytic efficiency in this tissue and so provide insights into the selective forces involved in the evolutionary development of an extreme tolerance to anoxia. The control of glycolytic flux in turtle ventrical muscle has been vested primarily in phosphofructokinase and pyruvate kinase. Creatine phosphate and fructose diphosphate play pivotal roles in the channelling of carbon through the pathway and in the production of metabolic energy as ATP. The levels of these key metabolites are in turn tightly regulated. One of the enzymes studied (phosphofructokinase) controls the levels of fructose diphosphate produced and creatine kinase (itself modulated by the cell's redox balance) controls creatine phosphate levels. When oxygen levels are reduced, NADH accumulates because of a decrease in electron transport chain regeneration of NAD (129). This leads to an effective activation of creatine kinase by lowering the Kм for creatine phosphate. This activation causes a drop in creatine phosphate levels without the decline in ATP levels that are seen at the onset of hypoxia in other tissues ( 4 ). Since the high levels of creatine phosphate present in aerobic heart are responsible for the inhibition of phosphofructokinase by greatly increasing the for its substrate, fructose-6-phosphate, this drop in concentration deinhibits the enzyme and leads to a flush of its product, fructose diphosphate. The increase in fructose diphosphate (1) serves to activate phosphorructokinase by itself, and (2) further reduces the effect of creatine phosphate by a deinhibition of the enzyme. These two effects cause an autocatalytic increase in flux through this locus* Fructose diphosphate also feed-forward activates pyruvate kinase by decreasing the Km for its substrate, phosphoenolpyruvate, and serves to deinhibit this enzyme which is normally inactive due to alanine and ATP inhibition. The most important feature of such a regulatory system is that it is a kind of autocatalytic cascade. Once the activation of creatine kinase is initiated by the redox imbalance inherent in anaerobiosis, all the various regulatory interactions potentiate one another. Drops in inhibitor levels lead to increases in activator levels and these activators serve to further deinhibit. These interacting effects serve to potentiate anoxic production of energy to compensate for the temporary depletion of oxygen in diving stress and in long periods of hibernation in this turtle, Nature's premier vertebrate anaerobe.

The objective of the present study was to determine the nutritional and redox conditions that promote the growth and biosurfactant production by Bacillus spp.. Three strains belonging to the culture collection of the Laboratório de Biotecnologia e Biodiversidade para o Meio Ambiente (LBBMA), previously evaluated for their potential for application in Microbial Enhanced Oil Recovery, including biosurfactant production, were used in the study. Among the three strains, only LBBMA111A was able to grow in mineral medium reduced with 0,5 mmol L-1 of dithiothreitol in the absence of any growth factor. Strains LBBMA155 and LBBMA283 required supplementation of the growth medium with DNA at low redox potential. However, this behavior can be modified according to the physiological state of the cells. The redox potential was shown to affect the growth patter of all strains, but in different intensities. In addition, the redox potential of the growth media also affected the yield and activity of biosurfactants produced by all strains. The growth of the strains was affected by the carbon and nitrogen sources of the media. The three strains were not able to growth when starch or pyruvate were used as carbon source. Either the carbon source or the nitrogen source of the media affected the production of biosurfactants. The greatest growth and biosurfactant production was observed when proteose peptone was used as the nitrogen source. The greatest growth was obtained when sucrose was the carbon source; however, for strains LBBMA111A and LBBMA283, glucose provided the greatest reduction of the interfacial tension. Variations in carbon and nitrogen sources appear to affect the structure of the biosurfactants produced. It is concluded that the three strains Bacillus are potential candidates for Microbial enhanced oil recovery (MEOR) based on the production of biosurfactants in situ; nevertheless, more studies must be conduct to evaluate the range of physical- chemical conditions for their successful application.

O presente trabalho objetivou determinar as condições nutricionais e de potencial redox que estimulem o crescimento e a produção de biossurfactantes por três isolados do gênero Bacillus. Foram utilizados três isolados pertencentes à coleção de culturas do Laboratório de Biotecnologia e Biodiversidade para o Meio Ambiente (LBBMA) que possuíam características desejáveis para sua aplicação na recuperação avançada de petróleo, incluindo a capacidade de produzirem biossurfactantes. Dos três isolados, somente o isolado LBBMA111A foi capaz de crescer em meio mineral reduzido com 0,5 mmol L-1 de ditiotreitol sem a adição de qualquer fator de crescimento. ▼ Abstract: Petroleum reservoirs are deep subsurface environments of global importance as energy resources and in biogeochemical cycles. Microorganisms living in petroleum reservoirs are able to use petroleum hydrocarbons as carbon sources, leading to biodegradation and negatively affecting the quality and economic value of oil. Previous studies suggest that anaerobic microorganisms are responsible for in-reservoir oil biodegradation. However, little is known about the actual microbial diversity and metabolic processes involved in biodegradation, mainly because of the difficulty in recovering the complex microbial community present in such extreme environments. This work aimed to characterize the microbiota of two petroleum reservoir samples using culture-independent approaches and assess the anaerobic hydrocarbon degradation potential of the indigenous microorganisms under different physicochemical conditions. The study revealed clear differences in the phylogenetic diversity between the degraded (BA-1) and non-degraded (BA-2) oil samples based on the analysis of bacterial and archaeal 16S rRNA gene clone libraries. The taxonomic and functional differences were also evident in the metagenomic profiles of the two petroleum reservoir samples. The metagenome from the BA-1 sample showed high abundance of genes related with the anaerobic hydrocarbon degradation, methanogenesis and syntrophic associations, while the BA-2 sample showed abundance of aerobic hydrocarbon degradation genes. Correspondingly, a microbial anaerobic active consortium was successfully cultivated from the BA-1 oil sample capable of metabolizing the crude oil components to methane. Higher rates of methane production were observed at the lowest salinity and at the in situ reservoir temperature. Together, the highest salt concentration (120 gl-1) and temperature (70 ºC) tested showed to inhibit the methanogenic activity of the consortium. Data obtained demonstrate that crude oil can harbour an indigenous microbiota with the ability to degrade petroleum hydrocarbons predominantly via methanogenic metabolism. Results gathered herein, and the ongoing work, will help us to understand the potential roles of microbes in hydrocarbon biodegradation and how such organisms respond to the environmental conditions in the petroliferous subsurface. ▼ O objetivo deste trabalho foi investigar o efeito da castração, tempo de confinamento, metabolismo celular e regiões do musculo sobre a oxidação proteica e lipídica e outras características de qualidade da carne bovina. Oitenta e quatro bovinos (castrados e inteiros) Nelore, confinados por diferentes períodos, foram usados para conduzir estudos em músculos Longissimus dorsi e Biceps femoris. O segundo musculo foi dividido em duas porções: origem (PO) e inserção (PI). No estudo com L. dorsi, bifes foram embalados sob condições de aerobiose (PVC) e anaerobiose (vácuo) e maturados por 1, 3, 5, 7 e 9, e 1, 7, 14 e 21 dias, respectivamente. Para este músculo, nenhuma diferença na estabilidade oxidativa [tióis, carbonilas e Substancias Reativas ao Ácido Tiobarbiturico (TBARS)] e cor entre as carnes dos animais inteiros e castrados foram encontradas. Isto poderia ser explicado pela falta de diferença no status oxidativo inicial, mensurados através da atividade de enzimas antioxidantes, conteúdo de glutationa total e composição de ácidos graxos, entre as condições sexuais. Os resultados também indicaram que a oxidação dos bifes embalados a vácuo leva o dobro de dias para iniciar, em comparação aos bifes em aerobiose. No estudo com o Bíceps femoris, os animais foram abatidos com 59 e 129 dias de confinamento e os bifes da PO e PI foram maturados por 1, 30, 60 e 100 dias. Os resultados da atividade das enzimas lactato desidrogenase e citrato sintase mostraram que a PO tem metabolismo mais oxidativo (aeróbio) e a PI glicolítico (anaeróbio). A carne dos animais inteiros tiveram menor TBARS e maior luminosidade (L*), perda de peso por cocção (PPC) e força de cisalhamento (FC) em comparação aos animais castrados. A PO foi mais susceptível a oxidação proteica (menor tióis) em comparação a PI. A carne dos animais confinados por 129 dias tiveram maiores PPC e oxidação proteica (menores tióis) em comparação a carne dos animais confinados por 59 dias. Diferenças de estabilidade oxidativa entre a carne de animais castrados e inteiros confinados por menor período desapareceram quando os animais foram confinados por maior período. Valores de pH e tióis na carne dos animais castrados e inteiros foram afetados pelo tempo de maturação. Ambas as condições sexuais tiveram carne com maior valores de pH no dia 30 de maturação e este, se manteve ao longo do tempo. A PO teve maiores valores de TBARS no dia 60, PPC no dia 100 e FC nos dias 30 e 60 de maturação em comparação a PI. Foi observada uma interação entre tempo de confinamento e tempo de maturação para tióis, TBARS, metamioglobina, pH, L* e FC. ▼ Oxygen and its reactive species —hydrogen peroxide (H2O2), superoxide (O2-) and hydroxyl radicals (HO•)— can cause a plethora of damages in the cell, from DNA damage to inactivation of iron containing enzymes. Virtually all life forms experience oxidative stress to some degree, and they are all equipped with mechanisms to protect themselves and to repair the damaged moieties. While oxidative stress is an obvious caveat of oxic life, it can also affect the organisms in anoxic environments and at oxic- anoxic interfaces. The present body of work looks into the latter condition by investigation the effects of oxidative stress endured by a facultative anaerobe (Escherichia coli) and an obligate anaerobe (Bacteroides thetaiotaomicron). E. coli lives adjacent to the epithelia in mammalian intestines. The encounter of oxygen penetrating from the epithelial layer and the reduced molecules like H2S from the center of the lumen creates an oxic-anoxic interface with different reactive oxygen species that can affect E. coli. B. thetaiotaomicron is another gut-dwelling bacterium living in the intestinal lumen, where it occasionally experiences oxidative stress. Bacteroides species in general are staple members of the gut microbiome because of their role in breaking down complex carbohydrates and thereby providing carbon sources for the rest of the community. Both E. coli and B. thetaiotaomicron should deal with high levels of oxidative stress during pathogenesis and the transition periods between hosts. The first chapter is an introduction to oxidative stress, its targets and the defense mechanisms against it in both of the bacteria discussed above. The second chapter focuses on finding a new periplasmic H2O2 degrading enzyme in E. coli that is only induced in the absence of oxygen and in presence of H2O2. The regulation mechanism of this enzyme, along with the detailed calculations of H2O2 fluxes through cellular membranes, revealed that it cannot protect the cytoplasm against exogenous H2O2. Instead, it enables E. coli to use H2O2 as a terminal electron acceptor for anaerobic respiration. The third chapter of this thesis looks into the molecular basis of obligate anaerobiosis. Previous reports have identified three points of oxidative damage in the central metabolism of B. thetaiotaomicron. Two of these enzymes, fumarase and pyruvate formate lyse (PFL), were reported to be damaged by O2- and molecular O2 respectively. In this study, we show that the third enzyme, pyruvate ferredoxin oxidoreductase (PFOR), is also damaged by molecular O2 itself. Pyruvate dissimilation in this bacterium depends upon PFL and PFOR; therefore, even in the absence of other reactive species O2 can directly inhibit this key metabolic node and block growth. The rate of fumarase damage by O2- approaches the PFL and PFOR inactivation by O2. We infer that the titer of scavenging enzymes in B. thetaiotaomicron is set so targets of H2O2 and O2- are neither more nor less sensitive to aeration than is pyruvate dissimilation. L’effet de la température et de la composition du milieu en nutriments sur les propriétés des spores (résistance et germination) de B. cereus a été largement étudié contrairement à l'effet de l'anaérobiose. Or, les cellules végétatives de B. cereus peuvent se retrouver dans une grande variété de milieux naturels avec un faible niveau d'oxygène (intestin, sol, lignes de traitement des aliments…) où la sporulation peut avoir lieu. Les spores produites dans ces conditions anaérobies pourraient donc avoir des propriétés particulières. Dans ce travail, un panel de 18 souches de B. cereus appartenant aux groupes phylogénétiques de II à VII a été étudié pour sa capacité à sporuler en anaérobiose dans un milieu de sporulation approprié que nous avons développé (MODS). En anaérobiose, la capacité de sporulation a été plus faible et plus hétérogène qu’en aérobiose. La souche AH187 a produit le niveau de spores le plus important en anaérobiose, elle a donc été choisie pour étudier les propriétés de ces spores. Les spores produites en anaérobiose étaient plus résistantes à la chaleur humide entre 90°C et 100°C, à 1M de NaOH, 1M d'acide nitreux et à la lumière pulsée. Aucune différence dans la résistance à 5 % de peroxyde d'hydrogène ou à 0.25 mM de formaldéhyde, ni aux UV-C, n'a été observée entre les deux conditions. En présence de L- alanine, les spores produites en anaérobiose germaient plus efficacement que celles produites en aérobiose tandis qu’aucune différence dans la germination n’a été observée en présence d'inosine. Aucune différence dans la taille des spores produites dans les deux conditions n’a été observée par microscopie électronique à transmission. Toutefois, les spores obtenues dans des conditions anaérobies avaient un exosporium endommagé ou dans certains cas un exosporium complètement détaché, contrairement aux spores produites dans des conditions aérobies. Afin de comprendre les différences dans la capacité de sporulation de B.cereus entre les 2 conditions, des PCR en temps réel (RT-PCR) ont été utilisées pour étudier l'expression des gènes de l'initiation de la sporulation spo0A, spo0B, spo0F, KinA et kinB. Les cinétiques d'expressions des gènes spo0A, spo0B, spo0F et KinA avaient la même tendance. Ils étaient caractérisés par une expression plus élevée en anaérobiose par rapport à l’aérobiose au début et à la fin de la phase exponentielle de croissance. En outre, l'expression du gène kinB était caractérisée par une augmentation en anaérobiose par rapport à l’aérobiose pour atteindre un pic entre 4 h (milieu de phase exponentielle) et 6 h (début de phase stationnaire) de croissance. Les gènes spo0A, spo0B, spo0F, KinA et kinB sont exprimés de manière différentielle entre l’aérobiose et l’anaérobiose. ▼ In the ethanol industry and research community, Saccharomyces cerevisiae is regarded as the most competitive fermentation yeast and therefore industrial yeast populations are rarely studied. This thesis investigates yeast and lactic acid bacteria (LAB) populations in a Swedish ethanol plant. Interestingly, the yeast population was dominated by Dekkera bruxellensis and not by the inoculated S. cerevisiae. High numbers of lactic acid bacteria (LAB) were also found, with Lactobacillus vini dominating. Since there was no indication of a reduction in productivity, we regarded D. bruxellensis together with L. vini as a production consortium. In test fermentations, the industrial D. bruxellensis strains had similar or higher ethanol yield than two industrial S. cerevisiae strains. Glycerol yield was lower and biomass yield higher, indicating more energy-efficient metabolism of D. bruxellensis. To test the ability of our isolates to produce ethanol from lignocellulosic substrate, we cultured D. bruxellensis in aspen sawdust hydrolysate. D. bruxellensis was slightly more sensitive to the hydrolysate than S. cerevisiae in batch culture, but was able to adapt to the inhibitors, achieving an ethanol production comparable to S. cerevisiae. Five years after the first isolation, the ethanol plant made substantial process changes. Renewed investigations showed that D. bruxellensis and L. vini were still the dominant microbial population. In continuous competition experiments, D. bruxellensis was only able to outcompete S. cerevisiae during glucose limitation. This suggests that in glucose-limited conditions, D. bruxellensis is more competitive due to its more energy-efficient metabolism, probably together with a higher affinity for glucose uptake in such conditions. Although D. bruxellensis is indeed facultatively anaerobic, it has a higher nutritional demand under anaerobic conditions than S. cerevisiae. This is probably due to a redox imbalance caused by low glycerol production. The results show that D. bruxellensis is a competitive yeast owing to its energy-efficient metabolism in glucose-limited conditions. It can act as production yeast for ethanol production from both first- and second-generation substrates under conditions of optimal aeration and nutrient supply.

IN AN ATTEMPT TO ELUCIDATE THE METABOLIC PATTERN OF RESPONSE UNDER CONDITIONS OF OXYGEN DEPRIVATION IN SUBTIDAL GASTROPODS, THE INTERMEDIARY METABOLISM OF DIFFERENT TISSUES WAS STUDIED UNDER AEROBIC AND ANAEROBIC CONDITIONS. THE TISSUES UNDER EXAMINATION INCLUDE THE HEPATOPANCREAS, THE MANTLE TISSUE AND THE FOOT MUSCLE. THE RESULTS SHOW THAT THE APPLICATION OF ANOXIC CONDITIONS INDUCES CHANGES IN METABOLISM AND PARTICULARLY IN GLYCOLYTIC RATE WHICH ARE DIFFERENT BETWEEN DIFFERENT TISSUES. THE HEPATOPANCREAS RESPONDS BY REDUCING ITS METABOLIC RATE. THIS PATTERN OF RESPONSE IS FOUND IN MUSCULAR AND NON MUSCULAR TISSUES OF INTERDAL INVERTEBRATES THAT ARE FREQUENTLY EXPOSED TO ANOXIC SITUATIONS. IN CONTRAST, THE FOOT MUSCLE AND PROBABLY THE MANTLE TISSUE RESPOND IN A WAY SIMILAR TO THAT FOUND IN VERTEBRATES, WHICH IS CHARACTERIZED BY AN INCREASE OF THE GLYCOLYTIC RATE IN ORDER TO GAIN THE NECESSARY ENERGY BULK FROM ANAEROBIC PROCEDURES.

Os óleos pesados (10-20 API) constituem mais da metade das reservas mundiais de petróleo, havendo previsões de que em 2025 sejam a principal fonte de petróleo no mundo. As dificuldades na exploração desse tipo de óleo estão associadas a características desfavoráveis, como densidade e viscosidade (100 a 10000 cP). Com a necessidade de melhorar o aproveitamento dos reservatórios tendo em vista os níveis relativamente baixos de eficiência das técnicas tradicionais tem-se ampliado as pesquisas para o desenvolvimento de novas técnicas de recuperação, destacando-se a recuperação avançada de petróleo melhorada por microrganismos (Microbially enhanced oil recovery MEOR). Na recuperação de óleos contidos em meios porosos, duas variáveis adimensionais são relatadas como importantes: o número capilar e a razão de viscosidade. À medida que o número capilar aumenta, o óleo residual decresce. Isso pode ser obtido por meio da redução da tensão interfacial entre o fluido de arraste e o óleo residual. Diversos autores têm reportado a obtenção de tensões interfaciais ultrabaixas entre biossurfactantes e hidrocarbonetos, incluindo o óleo pesado. Neste trabalho, testou-se a hipótese de que os biossurfactantes produzidos, em condição de reservatório, por dois isolados de Bacillus subtilis pertencentes à Coleção de Culturas do Laboratório de Biotecnologia e Biodiversidade para o Meio Ambiente (LBBMA/DMB/UFV), são capazes de mobilizar óleo pesado em sistemas porosos de areia. Inicialmente, os isolados de Bacillus subtilis LBBMA 155 e BacilIus subtilis spizizenii LBBMA 258 foram avaliados quanto ao crescimento e produção de biossurfactante em resposta a variações de temperatura e salinidade. Para tanto, foi empregada a Metodologia de Superfície de Resposta (MSR) através do delineamento experimental Composto Central Rotacional (DCCR), e observou-se que esses isolados foram capazes de produzir biossurfactantes em anaerobiose, sendo o crescimento influenciado pela temperatura e a produção de biossurfactante influenciada pela temperatura e pela salinidade. Posteriormente, os biossurfactantes produzidos por esses isolados, sozinhos ou em misturas, foram avaliados quanto à capacidade de mobilização de óleo pesado retido em núcleos porosos de areia. A injeção de três volumes porosos do extrato de biossurfactante (EB) produzidos por Bacillus subtilis LBBMA 155 e Bacillus subtilis subsp. spizizenii LBBMA 258 removeram 13,55% e 17,42%, respectivamente, de petróleo residual (14 API). A mistura dos extratos de biossurfactantes produzidos por esses dois isolados mostrou valores intermediários de DMC e recuperação de petróleo residual reportados para cada um. Sinteticamente, os extratos de biossurfactantes (EB) produzidos em anaerobiose por B. subtilis LBBMA 155 e B. subtilis subsp. spizizenii LBBMA 258 são efetivos na mobilização de óleo pesado em núcleos porosos de areia e possuem potencial de utilização na recuperação avançada de petróleo melhorada por microrganismos.

La pyocyanine (PYO) est une phénazine de couleur bleu-vert, produite spécifiquement par la bactérie pathogène opportuniste Pseudomonas aeruginosa (Pa). La toxicité aérobie de la PYO envers les cellules de mammifères, les levures et les bactéries a été décrite de longue date, mais la compréhension des mécanismes d’action est encore lacunaire, en particulier en conditions de limitation en O2 (conditions rencontrées dans le contexte infectieux). De plus, il a récemment été montré que la PYO peut apporter des effets bénéfiques pour la souche productrice en hypoxie. Au cours de ce travail, nous avons réexaminé les effets de la PYO sur un large panel de bactéries dont son propre producteur (Pa) ainsi que sur un modèle cellulaire eucaryote Saccharomyces cerevisiae exposées à différentes tensions en O2. Nos données suggèrent que la toxicité aérobie de la PYO envers S. cerevisiae est multifactorielle, impliquant à la fois une interaction avec le complexe III de la chaîne respiratoire et l’induction d’un stress oxydatif. Pour la première fois, nous avons mis en évidence une toxicité de la PYO exacerbée en anaérobiose chez un eucaryote (S. cerevisiae). Le mécanisme d’action impliquerait le PYO radical. Nous avons également montré que la PYO peut inhiber la croissance aérobie et anaérobie des microorganismes concurrents, plus particulièrement S. aureus en bloquant le complexe III de la chaîne respiratoire. A l’inverse, la PYO peut stimuler la respiration de Pa surtout dans les conditions mimant le contexte infectieux (hypoxie, vie ralentie). Le complexe III et/ou les oxydases terminales cbb3 serait impliqué favorablement. En conclusion, la PYO jouerait à la fois un rôle de poison hypoxique mais aussi un rôle de navette redox bénéfique pour la survie et la virulence de Pa en hypoxie.

Pyocyanin (PYO) is a blue-green phenazin, specifically produced by the opportunistic bacterium Pseudomonas aeruginosa (Pa). Aerobic toxicity of PYO toward mammalian cells, yeast and bacteria has been known for a long time, but the understanding of its mechanisms of action remains unclear, especially in conditions of limited O2 (conditions encountered during infection). In addition, it has recently been shown that PYO can bring benefits to the producer strain under hypoxia. In this study, we reexamined the effects of PYO toward a large panel of bacteria including its own producer Pa as well as a model of eukaryotic cells Saccharomyces cerevisiae exposed to different oxygen tensions. Our results suggest that the aerobic toxicity of PYO toward S. cerevisiae is multifactorial: involving both interaction with the respiratory chain at the level of complex III and induction of oxidative stress. For the first time, we have shown that PYO exerts an increased toxicity toward the eukaryotic cell, S. cerevisiae under anaerobiosis. The mechanism could involve the production of PYO radical.
