▼ Amines formed by biosynthesis in germinated barley have been suggested as precursors for N-nitrosodimethylamine (NDMA) in barley malt dried by direct-fired kilning. This hypothesis was verified by germinating raw barley and exposing the resulting malt roots and clean malt to nitrous acid. Quantitation of the NDMA formed indicated that malt roots contained relatively large amounts of NDMA precursor which could only have been formed as a result of germination. The clean malt also contained NDMA precursor, but the amount of precursor in clean malt was only slightly higher than the level of precursor already present in raw barley. The two tertiary amine alkaloids, hordenine and gramine, which are biosynthesized in malt during germination were subjected to nitrosation. Nitrosation of both amines at 65° (pH 4.4 or pH 6.4) resulted in NDMA formation. Gramine was highly susceptible to nitrosation to yield NDMA. At 24° in dilute acetic acid (pH 3.4), the initial rate of nitrosation of gramine to yield NDMA was nearly equal to the initial rate of NDMA formation from dimethylamine. The ratio of initial rates of formation of NDMA from gramine and trimethylamine was 6250:1. At 23°, the ratio of initial rates of formation of NDMA from gramine and hordenine was 5200:1. N-Methyltyramine and N-methyl-S-aminomethylindole, the immediate biosynthetic precursors of hordenine and gramine, were synthesized. Nitrosation of N-methyltyramine yielded p-hydroxy-N-nitroso-N-methyl- 2-phenylethylamine as the major reaction product and p-hydroxy-m-nitro- N-nitroso-N-methyl-2-phenylethylamine as the minor product. Nitrosation of N-methyl-3-aminomethylindole yielded N-nitroso-N-methyl-3- aminomethylindole as the minor reaction product; the major product was a dinitroso compound identified as N¹-nitroso-N-nitroso-N-methyl-3- aminomethylindole. Investigation of the products of hordenine nitrosation at 65° (pH 4.4) indicated the formation of NDMA and p-hydroxy-m-nitro-N-nitroso- N-methyl-2-phenylethyl amine; p-hydroxy-N-nitroso-N-methyl-2- phenylethylamine was not observed as a product of hordenine nitrosation at 65°. NDMA appeared to be the major nitrosamine formed from hordenine. The rapid reaction of gramine with nitrous acid and investigation of the gramine nitrosation reaction products both indicated that gramine did not undergo nitrosation by the expected mechanism of nitrosative dealkylation. A new mechanism is proposed to explain the rapid reaction of gramine with nitrous acid and to account for the fact that NDMA was the only N-nitrosamine formed during the nitrosation of gramine. ▼ The reaction of several primary amines with nitrite was investigated with regard to the formation of N-nitrosamines. When reacted in a low moisture system at 160°C with a 1:2 molar ratio of amine to sodium nitrite, various primary amine hydrochloride salts were found to give rise to heterocyclic nitrosamines. The amines and yields of nitrosamines were as follows: putrescine, 22% nitrosopyrrolidine; cadaverine, 21.5% nitrosopiperidine; ornithine, 1% nitrosopyrrolidine and 3% nitrosoproline; lysine, 1% nitrosopiperidine and 2.5% nitrosopipecolic acid. Nitrosamine analysis was by gas chromatography and identification was confirmed by mass spectrometry. The analysis of the nitrosamino acids was facilitated by making the methyl ester derivatives. When the reactions were carried out in buffered solution at temperatures up to 100°C, the same products were identified except ornithine did not produce nitrosopyrrolidine and lysine did not produce nitrosopiperidine. The pH optimum for the formation of nitrosopyrrolidine was 3.8 and the pH optimuna for the production of nitrosopipecolic acid from lysine was 3.4. Substantial differences in nitrosamine yield were noted when various types of buffer were used. The effects of time and temperature were investigated with nitrosamines being produced at temperatures as low as 22°C. The concentration of nitrite influenced nitrosamine yield dramatically and the reaction rate appeared to be second order with respect to nitrite concentration. Theoretical yields of heterocyclic nitrosamines from primary diamines reacted with nitrite in buffer were as high as nine percent. On a comparative basis, yields of five-membered heterocyclic nitrosamines were higher than the six-membered ring compounds, and the amino acids gave higher yields of the respective nitrosamines than did the simple diamines. When butylamine was reacted with nitrite, dibutylnitrosamine was identified as a product. Yields were extremely low in solution, but as a high temperature-low moisture reaction, one percent of the nitrosamine was formed. A system similar to frying bacon was used to determine if putrescine could contribute to the formation of nitrosopyrrolidine in cooked bacon. Ground pork belly containing 200 ppm sodium nitrite was cooked to 177°C and the cooked meat, cooked-out fat, and cooking distillate were combined and analyzed for nitrosopyrrolidine. An average of 109 ppb nitrosopyrrolidine was detected without added putrescine. When 0.40% putrescine was added to the pork prior to cooking, an average of 321 ppb nitrosopyrrolidine was formed. In another set of cooking experiments, only the distillate was analyzed. Increased amounts of nitrosopyrrolidine were detected when the pork contained added putrescine or proline. Nowadays, both secondary and tertiary amines are playing vital roles in modern chemical industry as well as pharmaceutical industry. Various synthetic methods of amines have also been reported due to their promising applications. In this thesis, a simple and widely applicable one-pot aza-Wittig/reduction method is reported for synthesizing both secondary and tertiary amines. Firstly, we focused on tandem aza-Wittig/reduction reactions for synthesizing secondary amines. We noticed that an aza-Wittig reaction involving an iminophosphrane which is the product from a Staudinger reaction, followed by addition of an aldehyde can be used to construct a C=N double bond in the product. By making use of the PPh3O (byproduct of aza-Wittig reactions) as the catalyst along with trichlorosilane as the reducing agent, the imine products in the aza-Wittig reactions can be converted into amines in one pot. In addition to using PPh3, highly-efficient heterogeneous polystyrene-based rasta resin-supported PPh3 was also examined in our study. Secondly, in order to broaden the utility of this one-pot method, instead of using relatively dangerous organic azides as substrates, in situ azide formation reaction was added to the flow chart. By reacting alkyl bromides with sodium azide or tertabutylammonium azide can give various azides in one-pot. Thirdly, the secondary amines formed using our one-pot procedure can undergo reductive amination reaction with aldehyde to give diverse tertiary amines as the final products in one pot. In this reaction, trichlorosilane reduction was performed twice by utilizing the phosphine oxide byproduct from the aza-Wittig reaction as a catalyst. Finally, chemistry regarding tandem one-pot aza-Wittig reaction by using catalytic amount of phosphine oxide was discussed. Instead of using organic azides, various aryl isocyanates were used as substrates for the reaction, with the help of catalytic amount of a five-membered ring phosphine oxide to give high yields of secondary amines. Chemistry studied in the thesis stands as solid proof for the superiority of our method. Tandem one-pot reactions improved the efficiency of our method by avoiding lengthy workup and purification steps. Our chemistry may serve as a good alternative for both secondary and tertiary amine synthesis in the future.

▼ Our present knowledge of the acidity constants of acids which are too weak to "be measured with respect to the pH scale is limited. In this work discrepancies existing among the results published by previous investigators are reconciled by a reinterpretation of their data. No accepted basicity values are available for concentrated aqueous alkali solutions (greater than 0.1 Molar), since measurements of pH are unreliable in such solutions. The extreme insolubility of compounds which might be used as indicators has prevented evaluation of the basicity of these solutions by spectrophotometric measurement of indicator equilibria. The present work uses the Hammett acidity function concept to develop the H- function for the following systems: For water containing benzyltrimethylammonium hydroxide H- varies from 11.98 for 0.01 Molar to 16.20 for 2.38 Molar; For pyridine-water containing 0.011 Molar tetramethyl-ammonium hydroxide H- varies from 12.27 for water which contains 1.0 mole percent pyridine to 15.43 for water containing 64.7 mole percent pyridine; For a solution of 30 mole percent pyridine and 70 mole percent water H-varies from 14.06 when the concentration of benzyltrimethylammonium hydroxide is 0.03 Molar to 17-65 when the concentration of the base is 2.19; For a solution 50 mole percent pyridine and 50 mole percent water H- varies from 15.67 when the base concentration is 0.01 Molar to 18.91 when the base concentration is 2.12 Molar; For sulfolane-water containing 0.011 Molar tetramethylammonium hydroxide H- varies from 12.39 for 1.79 mole percent sulfolane to 19.18 for 93.4 mole percent sulfolane; For dimethylsulfoxide-water containing 0.011 Molar tetramethylammonium hydroxide H- varies from 12.17 for 5 mole percent dimethylsulfoxide to 18.61 for 70 mole percent dimethylsulfoxide: For aqueous solutions of lithium hydroxide H-varies from 13.43 for 1 normal lithium hydroxide to 14.31 for 5 normal. In addition to these results, adjustments are made to previously published scales for hydrazine-water and ethylenediamine water. The corrected values range from 12.76 for 9.2 mole percent ethylenediamine in water to 15.48 for 31.0 mole percent and 13.06 for 23.2 mole percent hydrazine in water to 15.08 for 51.1 mole percent. These results are obtained using the Hammett acidity function concept from the spectroscopic determination of the ionization equilibria of 24 substituted anilines and diphenylamines. pKa values are reported for these compounds ranging from 2.63 for symmetrical hexanitrodiphenylamine to 18.37 for paranitroaniline. All previous H- determinations have been based on paranitrobenzyl cyanide. This indicator is shown to be unsuitable for this purpose. Results which have been reported by other investigators are adjusted by eliminating the use of paranitrobenzyl cyanide and assigning another indicator as a starting point. PART I: Deamination of l-amino-4-bromo-7,7-dimethylbicyclo_ [2.2.1]heptan-2-ol (1) with nitrous acid formed 4-bromo-5, 5-dimethylbicyclo[2.1.1]hexane-l-carboxaldehyde (2). The structure of 2 was elucidated with infrared and nmr spectra. The corresponding carboxylic acid, semicarbazone, and p-toluenesulfonylhydrazone derivatives of 2 were prepared. The ring contraction during the deamination of 1 is similar to that of l-amino-3,3-dimethylbicyclo[2.2.1]heptan-2-ol (3), which formed 5,5-dimethylbicyclo[2.1.1]hexane-l-carboxaldehyde (4). Compounds 1 and 3 are different in the configuration of the hydroxyl groups and the substitution feature at C-3. Therefore, the successful deamination of 1 and 3 to the bicyclo[2.1.1]hexane derivatives demonstrates that the deamination of l-aminobicyclo[2.2.1]heptan-2-ols may be of general utility. During the ring contraction, the migrating group is anti-coplanar to the leaving group. PART II: Condensation of β-nitrostyrene with 1,3-cyclohexanedione in the presence of sodium methoxide did not yield a normal Michael addition product. The product was proved, with the support of spectral data, to be N-hydroxy-3-phenyl-4,5,6,7-tetrahydrooxindol-4-one (5). The acetyl derivative of 5, which gave a band in the infrared spectrum that is characteristic of an ester, ruled out the isomeric structure 6 as an alternative structure for 5. Upon hydrolysis, compound 5 could be converted into keto acid Z, from which a known compound, 2-phenyloctanedioic acid, could be obtained through Wolff-Kishner reduction. Hydrogenation of 5 over Raney nickel catalyst produced 3phenyl- 2,3,4,5,6,7-hexahydroindol-4-one (9), which was then dehydrogenated with Raney nickel in hot ethanol to form 3phenyl- 4,5,6,7-tetrahydroindol-4-one (2). Structures 8 and 9 were fully elucidated, and an authentic sample of 9 was synthesized. Stetter and Hoehne proposed oxazine 10 as the structure of the condensation product. This structure is reassigned as 5.

▼ Reaction of the pyrrolidine dienamines of ~1,8a-2-octalones with methyl vinyl ketone is complex. In methanol as solvent, the reaction occurs primarily with the linear dienamine isomer and results in annulation of the 8,8a-positions and, to a lesser extent, the 1,2-positions to give the corresponding octahydro-1 Hbenzo[d]naphthalene-2,10(3H,111-1)-dione and 3,4,5,6,7,8,8a,9-ctahydrophenanthren-2(10H)-one respectively. In toluene the dienamines react mainly in their cross-conjugated form. Diels-Alder addition of methyl vinyl ketone occurs across the 3,8a-positions to give the corresponding 9-acetylperhydro-2,4aethanonaphthalen-3-one and annulation of the 2,3-positions gives the 4,4a,5,5a,6,7,8,9-octahydroanthracen-2(3H)-one. The reaction of methyl vinyl ketone with the pyrrolidine dienamine of 4amethyl- 5-oxo-~ 1,8a-2-octalone is reported to give the aromatic product 3a, 7dimethyl- 2,3,3a,4,5,6-hexahydrophenalen-3,6-dionearisingfromp,o-annulation and in a related investigation with dienamines derived from isophorone it was reported that only products of Stork alkylation and Diels-Alder cycloaddition were isolated, there being no evidence for the reaction of methyl vinyl ketone with either the endo- or exocyclic o-positions of the dienamines. In an attempt to ascertain the reason for the apparently random changes in the regioselectivity of these related reactions some of this work was repeated and the reaction of methyl vinyl ketone applied to two other dienamines derived from 5,6,7,7a-tetrahydroindan-5-one and 4a,6-dimethyl-5-oxo-~1,8a-2-octalone. The reactions proved to be more complex than reported and several additional products have been isolated. Finally, reaction of phenyl vinyl ketone with dienamines derived from a1,8a_2-octalone was investigated. With the exception of the reaction of the 4amethyl dienamine, the main products isolated were those arising from addition of p.henyl vinyl ketone across the 3,8a-position to give the corresponding 9-benzoylperhydro-,4a-ethanonaphthalen-3-one. Reaction of the 4a-methyl dienamine with two equivalents of phenyl vinyl ketone gave a product tentatively assigned as the octahydrophenalenone. Polymer-supported reagents and catalysts, which allow for simple product separation and easy recycling, have been widely studied in the context of organic synthesis. The past decade has witnessed a number of new variations of polymeric materials, and among the most frequently immobilized functionalities are amines that possess versatile synthetic utilities. Polymers with new structures and improved properties for use in synthesis have been continuously developed since the support may impact the chemical reactions in which they are used in various ways. A new heterogeneous polystyrene-based amine, rasta resin-DMAP, has been synthesized and used in addition reactions of carbon dioxide to epoxides to afford cyclic carbonate products. This new material was found to be a more efficient catalyst than divinylbenzene cross-linked polystyrene supported DMAP, and was readily recycled without significant loss of catalytic activity. Compared to polymers bearing a single functionality, polymers possessing multiple different functional groups attached to a single polymer backbone would have greater potential utility, especially in reactions requiring multiple catalysts or reagents. As an example of this concept, a bifunctional polystyrene bearing both DMAP and piperazine groups has been prepared and applied as an organocatalyst for decarboxylative Doebner-Knoevenagel reactions of arylaldehydes and mono-ethyl malonate to produce (E)-,-unsaturated esters in high yields. Additionally, both non-cross-linked and cross-linked bifunctional polystyrenes featuring amine and thiourea groups have been developed, and their catalytic performance were evaluated in reactions of nitroalkenes with either nitroalkanes or sulfur ylides. Both polymers proved to be efficient catalysts in these reactions and the insoluble polymer demonstrated high recyclability. Control experiments using monofunctional polymers indicated that both catalytic groups of these bifunctional polymers are essential and they could work cooperatively to achieve efficient catalysis. Finally, a second generation bifunctional phosphine-amine polymer, rasta resin-PPh3-NBniPr2, was prepared and examined in tandem Wittig-reductive aldol reactions. In these reaction cascades, the phosphine oxide groups generated from the Wittig reaction served as the catalyst for the reductive aldol reaction, and moderate yields of structurally diverse -hydroxy ketones could be obtained from one-pot processes involving 5 sequential reactions.

▼ The dynamic interplay between the different types of cells in the brain involves the release and uptake of multiple chemical factors and their resulting signaling cascades in response to stimuli. This creates a constantly changing environment, which complicates the study of the organ’s function. Consequently, dissecting particular aspects of cellular communication is difficult in an in vivo environment. To better dissect particular aspects of cellular communication, controlled in vitro environments are combined with established techniques and novel platforms in this dissertation. Following an introduction to cellular communication and the methods utilized to investigate how it occurs, the development and fabrication of a microfluidic device to examine the interaction of a network of cells is described in chapter 2. This device, composed of a lower pharmacological layer and an upper bulk flow layer, utilizes hydrodynamic focusing to individually address cells without exposing the entire population. Fluorescent and electrochemical data illustrate how the flow streams introduced through the lower layer can be manipulated by fluid sent through the above bulk channels to limit diffusion. In chapter 3, a novel patterning technique, sucrose lift-off lithography, is reported as an alternative to traditional patterning schemes. With this technique, a sacrificial layer of sucrose is initially deposited onto a substrate, which is then coated with photoresist. The resist is then patterned lithographically to expose the substrate in defined well areas. The wells are subsequently loaded with adhesion proteins and a cell suspension, whose culture medium dissolves the sucrose over time, leaving only the patterned cells on the substrate. In chapters 4 and 5, amperometry is utilized to examine the effects of drug treatment and gene modification by knockout on exocytosis. Incubation with the D2 agonist, pramipexole, is found to cause an acute decrease in extracellular calcium influx and a decrease in the amount of dopamine released from in PC12 cells following potassium stimulation in chapter 4. Additionally, the vesicular monoamine transporter (VMAT) is found to package the agonist as well as DA into secretory vesicles. The effects of GODZ and SERZ-β gene knockout on calcium influx and release from intracellular stores in both murine hippocampal neurons and peritoneal mast cells are discussed in Chapter 5. The processes of double knockout neurons, lacking both GODZ and SERZ-β, were found to have decreased calcium influx and release when compared to wild type processes. Moreover, the exocytotic release of histamine and serotonin from mast cells was also examined following stimulation. The release kinetics for both compounds was observed to change with gene deletion as did the number of molecules released from the granules.
