▼ The main purpose of this study was to assess the utilization of active learning, to identify factors affecting the implementation of active learning either positively or negatively in terms of teachers` commitment, classroom conditions, instructional facilities and teachers training. Mixed methods were used to conduct the study. The study mainly conducted in upper primary schools in Addis Ababa city administration, in Nifas Silk Lafto Sub City. Data were collected from 169 teachers, 282 students and 3 pedagogy instructors of Kotebe college of Teachers Education. Because 75% of teachers were graduates of Kotebe college of teachers education. In the selection of sample population purposive and random techniques were used. The instruments of data collection were questionnaire, interview and observation checklist. The data were analyzed using percentage. The findings of the study proved that the implementation of active learning was low. Among the factors affecting the implementation of active learning, teachers training, teachers` commitment, classroom conditions, class size, beliefs and interests of teachers in their profession were the major ones. Finally, the following recommendations were forwarded, it is important to conduct in- service training so that their use of active learning strategies will be improved, Even if appropriate class size may not always be possible, the implementers of active learning should see alternative solutions such as, dividing students into groups, demonstration fields, field trip, laboratories and others. In addition to this, government bodies and stake holders have to construct extra classes to solve the problem and In order to solve the problems of teachers‟ commitment and lack of interest in the profession, school administrators, wereda educational leaders, Addis Ababa City administration Education bureau and Ministry of Education have to do different awareness creation programs. ▼ The purpose of this study was to examine the practice of active learning in upper primary schools of North Western zone of Tigray. Furthermore, it treats factors that affect the proper implementation of active learning. To accomplish this purpose, the study employed a descriptive survey method, which is supplemented by qualitative research methods to enrich the data. With this respect, teachers, students, directors and cluster supervisors were the sources of data for the study. The data gathering instruments were questionnaires, interviews and observations. The questionnaires were dispatched to 200 students, 60 teachers, and interviews were made with 16 directors and 6 cluster supervisors drawn from 16 schools which were teachers and students using simple random sampling, and directors and supervisors using availability system in the academic year of 2010/11. Moreover, 32 sections of grade seven and eight observation was made, two sections from each school at the departments of Language and Natural Science. The data obtained through questionnaires were quantitatively analyzed and interpreted in light of available literature whereas the information obtained through interview and observation were qualitatively described to supplement the quantitative data. The results of this study reveal that though the attitude of teachers and students toward active learning was positive and not serious problem, the level of practice of active learning was found to be low and moderate. Especially to use different strategies of active learning were not fully performed., Appropriate teaching methods were not used to address the comprehensive benefits of active learning, and teachers’ selfevaluation were not performed sufficiently. Furthermore, the study revealed that teachers’ and students’ tendency to prefer traditional lecture method, majority of students become dependent on the minority, shortage of time, lack of support from educational leadership, lack of teachers’ commitment, inadequacy of teachers’ training, lack of teaching resources, unavailability of facilities, teachers’ high workload, inconclusive of the class-room, were among the factors that hindered the implementation of active learning. It has been recommended that creating awareness, continuous and intensive short-term and long-term training should be offered for teachers on the practice active learning and about different strategies of active learning to improve their instructional practice to over come the challenges of implementation active learning. Besides, to alleviate or solve the factors that hinder the proper implementations of the practice, the school administrative body ought to create conducive environments that helps for teachers’ sufficient preparation and working time and provision of educational materials and resources to implement active learning methods in their class. ▼ Recent advances in electromagnetics introduced tools that enable the creation of arti- cial electromagnetic structures with exotic properties such as negative material pa- rameters. The ability to express these parameters has experimentally demonstrated using passive metamaterial structures. These structures, based on their passivity and resonant properties, are typically associated with high loss and signicant bandwidth limitations. Enhancing and further exploring novel electromagnetic properties can be done through embedding active circuits in the constitutive unit cells. Active elements are able to supplement the passive inclusions to mitigate and overcome loss and bandwidth limitations. The inclusion of these circuits also signcantly expands the design space for the development of functional metamaterials and their potential applications. Due to the relative diculty of designing active circuits compared with passive circuits, using active circuits in the construction of metamaterials is still an under- developed area of research. By combining the two elds of active circuit design and metamaterial design, we aim ll the functional active metamaterial design space. This document provides the basis for understanding the design and synthesis of functional active metamaterials. To provide necessary background matter, chapter 1 will function as an introduc- tion chapter, discussing how active electromagnetic metamaterials are created and characterized. There are also several required design techniques necessary to suc- cessfully engineer a functional active metamaterial. The introduction will emphasize on linking metamaterial unit cell response with RF/analog circuit design with a brief introduction to the semiconductor physics important to aid in the understanding of the full active metamaterial design and fabrication process. The subsequent chapters detail our specic contributions to the eld of func- tional active RF metamaterials. Chapter 2 introduces and characterizes a meta- material designed to have a tunable quality factor (tunable resonant bandwidth). This metamaterial is essentially passive but demonstrates the transistor's versatility as a combination of tunable elements, motivating the use of embedding transistors in metamaterials. After establishing a simple application of a transistor in a pas- sive metamaterial, chapter 3 outlines the design and characterization of an active metamaterial exhibiting the properties of loss cancellation and gain. Chapter 4 in- troduces another active metamaterial with the ability to self-adapt to an incident signal. Within the self-adapting system, several complex RF circuit systems are simulatenously developed and implemented such as a self-oscillating mixer and a phase locked loop. ▼ The elucidation of protein molecular function lags far behind the rate of high-throughput sequencing technology; thus, the development of accurate and efficient computational methods to define functional relationships is essential. Protein similarity networks have emerged as a simple, high-throughput method for defining protein relationships, but sequence-based techniques often inaccurately define molecular function details. Active site profiling (ASP) was previously developed to identify and compare molecular details of protein functional sites. In this work, protein similarity networks were created using active site similarity, sequence similarity, and structure similarity for four manually curated SFLD superfamilies. Results demonstrate that ASP-based clustering identifies detailed functional relationships more accurately than sequence- and structure-based clustering. Building on this, two iterative pipelines were developed utilizing active site profiling and profile-based protein database searches to cluster protein superfamilies into distinct, functionally relevant groups. First, the Two Level Iterative clustering Process (TuLIP) utilizes active site profiling and iterative PDB searches to divisively cluster protein structures into groups sharing functional site features. Across eight superfamilies, TuLIP clusters exhibit high correlation with SFLD functional annotations. Subsequently, the Multi-level Iterative Sequence Searching Technique (MISST) was developed to identify protein sequences sharing active site similarity with each TuLIP group using iterative profile-based GenBank searches. Though research is ongoing to fully parameterize MISST, preliminary results with multiple gold standard superfamilies indicate the ability to identify and cluster a large portion of each superfamily into functionally relevant groups. Furthermore, detailed analysis of the functionally relevant clusters generates hypotheses regarding previously unknown functional relationships. Lastly, DASP2 and DASP3 were created to improve the efficiency and accuracy of DASP at identifying proteins sharing active site similarity with a given set. Initial validation with the Prx superfamily indicates DASP3 TuLIP clusters match known functional groups more closely than DASP2, and DASP3 MISST groups are populated in fewer iterations. TuLIP and MISST provide an efficient, accurate way to define functionally relevant groups that can be applied systematically and on a large-scale, laying the foundation for functionally relevant clustering of the entire protein universe.

▼ ABSTRACT: One of the most important questions in modern astrophysics is the study of the formation and evolution of galaxies throughout the history of the Universe. In the last decades, several pieces of evidence have been found that support a coevolution of galaxies with the supermassive black hole (SMBH) that most of them (maybe every galaxy) have in their centres. Consequently, to make progress in this field it is necessary to understand the properties of these SMBH. Particularly of those in an active phase of growing (increasing their mass) accreting material from the host galaxy. They are the so-called active galactic nuclei (AGN). Of these, the most highly obscured systems are of special interest, since it is believed that during this highly obscured phase the relationship between SMBHs and their hosts was established. The obscuration is caused by a dust and gas structure, the so-called torus. The highly obscured AGN (known as Compton thick (CT)) are lost in optical surveys and, to a lesser extent, in X-ray surveys as the radiation emitted at these frequencies is absorbed. These AGN could be detected in mid-infrared wavelengths, where obscuration effect are less important. However, their study in X-rays is fundamental to determine the obscuration level. There are empirical relations in the literature that relate the mid-infrared and X-ray luminosity of AGN. The first objective of this work is to check whether these relations describe correctly the relation between luminosities for mid-infrared selected AGN samples (as the one used in this work) and to obtain our own relation otherwise. The second objective is to estimate a lower-limit for the obscuration for mid-infrared selected AGN not detected in deep X-ray exposures with the X-ray observatory XMM-Newton. The last objective is to estimate the necessary exposure times for the detection of this kind of objects with the X-ray observatories XMM-Newton and Athena. To accomplish these scientific objectives we have selected a sample in the mid-infrared composed of 91 AGN: 68 detected in X-rays with XMM-Newton, 3 detected in X-rays but with low-quality X-ray spectra and 20 AGN with no X-ray detection. As it is a sample selected in the mid-infrared it is expected to include objects with obscuration levels all over the parameter space. These AGN (23) are our sample of CT candidates. The X-ray undetected AGN and those with low-quality X-ray spectra are expected to be highly obscured or CT. For the undetected AGN we have upper-limit estimated for their X-ray fluxes. For those AGN with low-quality X-ray spectra, we have the observed luminosity (not corrected by obscuration). These measurements will be used to put constrains on the level of obscuration for these sources. First, we obtained the intrinsic X-ray luminosity (corrected by obscuration) of the CT candidates. To do that we check if the empirical relations of the literatura between X-rays and mid-infrared luminosities are valid for mid-infrared selected samples. ▼ Emergent collective behaviors are ubiquitous in nature at a remarkably broad range of length scales, spanning from the living cells to fish schools. As a prototype of these biological systems, self-driven active colloidal particles have been of much interest in the context of both theoretical and technological relevance. Consequently, many new propulsion mechanisms and geometry of active particles have emerged over the last decade to develop functional materials with unique nonequilibrium features such as dynamic pattern formation and complex shape changes. Notably, a majority of these swimmers are limited to linear propulsion and simple shapes (it{e.g.}, spherical, cylindrical, and helical) where the actuation relies on the heterogeneity of surface property. This dissertation, in general, focuses on ensembles of self-rotating active particles or spinners where the external energy is harnessed into individual particle rotation. In particular, the study constitutes experimental and numerical investigations of the collective dynamics in a mixture of counter-rotating spinners. \par Firstly, the significant advances in the development of self-propelled particles (SPPs) powered by several propulsion mechanisms are briefly summarized. Here, SPPs of different shapes and compositions with both translational and rotational dynamics are reviewed. Afterward, a phenomenological, continuum model is developed to investigate the influence of self-generated flows on the coarsening dynamics of binary liquids comprising counter-rotating spinners. The model combines a convective Cahn-Hilliard equation with a modified Navier-Stokes equation to describe the fluid composition and velocity, respectively. Several new types of collective excitations are predicted by the continuum model including ``active coarsening'' and self-propelled ``vortex-doublets'', many of which can also be qualitatively captured by the microscopic model. The exciting numerical results motivate the need for a simple experimental system to study the collective dynamics in ensembles of counter-rotating spinners. With this goal, chiral particles with rotational symmetry are designed to rotate in MHz acoustic field. The acoustic spinners of opposite chirality (R/S) can rotate in both clockwise and counterclockwise directions at the same time depending on their chiral orientation. Importantly, the spinners rotate either toward the concave or convex end of the fins depending on the order of rotational symmetry it{i.e.}, the number of fins. Beyond acoustic propulsion, the concept of shape-directed actuation is further extended for other propulsion mechanisms such as induced-charge electrophoresis and self-electrophoresis to encode any desired dynamics it{via} particle shape. Finally, a roadmap for the future directions is provided in light of the remaining questions along with a brief discussion on the potential functionalities including active catalysis and it{in vivo} applications. ▼ Increasingly, predictive models are used to support routine business de- cisions and are integral to the strategic competitive business strategies for a wide range of industries. Most often, data-driven predictive models are in- duced from training data obtained through the businesss routine operations. However, recent research on policies for intelligent information acquisitions suggests that proactive acquisition of information can improve models at a lower cost. Most active information acquisition policies are accuracy centric; they aim to identify acquisitions of training data that are particularly benefi- cial for improving the predictive accuracy of a given model. In practice, however, inferences from a predictive model are often used along with inferences from other predictive models as well as constant factors to inform arbitrarily complex decisions. In this dissertation, I discuss how these settings motivate a new kind of collaborative information acquisition (CIA) policies that exploit knowledge of the decision to allow multiple predictive models to collaboratively prioritize the prospective information acquisitions, so as to best improve the decisions they inform jointly. I present a framework for CIA policies and two specific CIA policies: CIA for binary decisions (CIA-BD), and CIA for top-ranked opportu- nities in terms of expected revenue (CIA-TR). Extensive empirical evaluations of the policies on real-world data suggest that the notion of CIA policies is indeed a valuable one. In particular, I demonstrate that these two new poli- cies lead to superior decision-making performances as compared to those of alternative policies that are either decision-centric or do not allow multiple models to collaboratively prioritize acquisitions. The performance exhibited by the CIA policies suggest that these policies are able to effectively exploit knowledge of the decisions to avoid greedy improvements in accuracy of any individual model informing the decisions; instead, they promote improvements in any one or all of the models when such improvements are likely to benefit the decisions. ▼ Oxygen is one of the most important molecules in human beings. Our research is focused on how the human body can respond and adapt to the physiological challenge posed by a lack of oxygen. Ascorbic acid (Vitamin C) is one of the most important and considered the most effective water-soluble, chain-breaking antioxidant in human plasma, with the capacity to prevents damage by free radicals. This thesis presents four studies investigating the phenomenon of Reactive Oxygen Species (ROS) generation in the many different surgical conditions in the animal and in the human. Study one investigated the geometry and thermodynamic properties of vitamin C. Calculations were carried out at the restricted and unrestricted B3LYP/6-31+G(d,p), B3LYP/6-311++G(d,p) and B3LYP/EPR-II levels for two conformers (1 and 2) of L-ascorbic acid and their respective oxidation products to monodehydroascorbates of ab-initio methods by Gaussian O3W package. Conformer 1, free radical properties are compared with previously published calculations in the gaseous and aqueous solution states and with experimental EPR values. Calculated molecular structures, EPR (electron paramagnetic resonance spectroscopy), the vibration spectral and energetic properties and all are reported including some proposed changes to previous EPR assignments. Conformer 2 of L-ascorbic acid is predicted to have lower energy than Conformer 1, under the method and basis sets used, by between 11 and 26 kJ mol-1 and is stabilised by internal hydrogen bonding. Relaxed potential energy surface (PES) scans were carried out for two proton transfer processes and relative energies of stable minima and barriers between them determined. Hydrogen transfer is predicted in two systems with favourable spatial arrangements of O–H and O groups for which relaxed potential energy surface scans are reported. Calculated vibrational wavenumber values are provided for selected C=C, C=O, C–H and O–H modes assigned to particular groups and significant calculated EPR hyperfine coupling constants (HCC) values for splitting by H(1) and C(13) for radical species are also reported. These calculations contribute to a better understanding of the complex role of L-ascorbic acid and its various oxidised, neutral, ionic and radical forms in biochemistry and medicine. Study two examined if vitamin C could ameliorate the damaging effects of I-R on myocardium and we postulated that the mechanism of vitamin C protection against iii I-R-induced cell death involved quenching of ROS. In the vitamin C group after 5 min of reperfusion a significant, sudden increase of diastolic pressure in the heart was noted and reached a maximum of 77 mmHg after 12 min of reperfusion and then gradually decreased to 51 mmHg after 60 min of reperfusion period but was quicker than in Control group reaching 37 mmHg by the end of the reperfusion period. ▼ Active fluids are ubiquitous in nature, spanning both microscopic and macroscopic length scales. They belong to an interesting new class of nonequilibrium systems in physics. In contrast with externally driven materials, active fluids are intrinsically forced out of equilibrium by their constituent active particles, which consume energy from the environment, using it to engage in nonequilibrium activities, such as motility and replication. Two of the most prevalent examples of active particles are bacteria and actomyosin complexes. In both cases, we observe rich collective behaviour giving rise to, respectively, the multicellular organisation of bacterial colonies and the intracellular structure in eukaryotes. A hydrodynamic description can be used to model the behaviour of active fluids. This approach is based on the fact that the active particles exert stresses on the surrounding fluid as they move through it. Within the hydrodynamic framework, we can study many interesting nonequilibrium phenomena, including hydrodynamic instabilities and spontaneous symmetry breaking. These provide an explanation for macroscopic motility patterns in active systems. In our work, we use a hybrid lattice Boltzmann method to simulate a range of motile states and their chiral characteristics. We start by studying the dynamics of an active fluid enclosed in a droplet with imposed orientational anchoring at the interface. Our results show that when the anchoring is strong enough, active extensile and contractile stresses lead to spontaneous droplet rotation. In contrast, if the anchoring is weak, the droplet instead translates. The signature of the observed rotating states is a significant deformation of the droplet shape, distinguishing it from the rotation of spiral defects, which has been previously reported. Moreover, the sense of rotation is selected via spontaneous symmetry breaking, so that the droplet is equally likely to start rotating clockwise or anticlockwise, acquiring a random chirality. Most biological active particles are also microscopically chiral themselves. Thus, active processes must involve chiral interactions at the microscale. We address this feature by considering a chiral active stress originating from a collection of active torque dipoles. We find that the active chiral stresses drive a nonequilibrium transition to a self-assembled cholesteric phase in both active fluids and dry active systems. This spontaneously twisted phase shares many of the characteristics of equilibrium cholesterics, including the formation of layered and fingering patterns, and the existence on non-singular defects. If the activity is suciently high, chiral active stresses are capable of untwisting an equilibrium cholesteric configuration (which is thermodynamically favoured). Finally, we look at active fluids with both achiral (extensile, contractile) and chiral active stresses. We observe that contractile activity suppresses the spontaneous twist, while extensile activity enhances it. We also show the existence of a pitchsplay…

▼ Robotic assistants should be able to actively engage their human partner(s) to generalize knowledge about relevant tasks within their shared environment. Yet a key challenge is not all human partners will be proficient at teaching; furthermore, humans should not be held accountable for tracking a robot’s knowledge over time in a dynamically changing environment, across multiple tasks. Thus, it is important to enable these interactive robots to characterize their own uncertainty and equip them with an information gathering policy for asking the appropriate questions of their human partners to resolve that uncertainty. In this way, the robot shares the responsibility in guiding its own learning process and is a collaborator in the learning. Additionally, given the robot requires some tutelage from its partner, awareness of constraints on the teacher’s time and cognitive resources available for devoting to the interaction could help the agent to use the time allotted more wisely. This thesis examines the problem of enabling a robotic agent to leverage structured interaction with a human partner for acquiring concepts relevant to a task it must later perform. To equip the agent with the desired concept knowledge, we first explore the paradigm of Learning from Demonstration for the acquisition of (1) training instances as examples of task-relevant concepts and (2) informative features for appropriately representing and discriminating between task-relevant concepts. Given empirical evidence that a human partner can be helpful to the agent in solving the concept learning problem, we subsequently investigate the design of algorithms that enable the robot learner to autonomously manage interaction with its human partner, using a questioning policy to actively gather both instance and feature information. This thesis seeks to investigate the following hypothesis: In the context of robot learning from human demonstrations in changeable and resource-constrained environments, enabling the robot to actively elicit multiple types of information through questions, and to reason about what question to ask and when, leads to improved learning performance. Angiogenesis, the growth of new blood vessels from existing vessels, plays a key role in physiological processes including development, growth, and wound healing. Insufficient angiogenesis contributes to impaired wound healing in diabetic patients and the elderly. Vascularization is also essential for the successful engineering of large, complex tissues and organs in vitro. Although successful tissue fabrication of a few hundred microns has been achieved, tissue engineering is limited by the inability to vascularize constructs to provide nutrients to the tissue core.Plasma medicine is a rapidly expanding interdisciplinary field combining engineering, physics, biochemistry and life sciences. Plasma, the fourth state of matter, is an ionized gas composed of charged particles (electrons, ions), excited atoms and molecules, radicals, and UV photons. Non-thermal dielectric barrier discharge (DBD) plasma, which primarily produces reactive oxygen species (ROS) has previously been used primarily to sterilize tissue and equipment, coagulate blood, or kill bacteria and cancerous cells. A nitric oxide (NO) producing pin-to-hole spark discharge (PHD) plasma has been reported to sterilize liquids and surfaces, including living tissue.Low doses of reactive oxygen and nitrogen species (ROS/RNS) contribute to angiogenesis. This study demonstrates the potential of plasma in delivering ROS/RNS into liquids and cells for wound or tissue engineering vascularization. ROS produced by a DBD plasma penetrated into both treatment liquid as well as into endothelial cells. These ROS then enhanced endothelial cell proliferation, migration and tube formation through fibroblast growth factor-2 (FGF-2) release. Specifically, H2O2 and OH· were the primary ROS responsible for plasma-induced angiogenesis. In longer term studies, non-thermal DBD plasma induced FGF-2 release led to secondary intracellular ROS production, enhanced FGF-2 expression and subsequent enhanced FGF-2 release. In a parallel study, a PHD plasma produced NO that enhanced endothelial cell proliferation and migration while maintaining cell viability.This research improves both our understanding of plasma and ROS/RNS effects on endothelial cells, and may facilitate the development of inexpensive, portable, medical devices for wound treatment. Thus, this dissertation contributes significantly to vascular biology, plasma medicine and health care.

Chapter-2 comprises process development of zafirlukast, an anti-asthma drug, preparation of stable amorphous and new acetonitrile solvate polymorph. Chapter-3 deals with a complete impurity profile of zafirlukast including identification, synthesis, characterization and root cause of their formation. Chapter-4 explains the synthesis and characterization of new zafirlukast related analogs. Chapter-5 presents an alternative synthesis of tadalafil from D-tryptophan and sarcosine ethyl ester.
