▼ Improvement of chemical capture is an important part of wildlife conservation and animal welfare to minimise distress for the animals and the risk of morbidity and mortality. The aim of this thesis was to improve wildlife immobilisation and anaesthesia by studying physiological variables and clinically evaluate different drug combinations and methods of capture in selected species. Reversible anaesthetic protocols were developed for use in free-ranging lions and four species of South- East Asian primates. Capture and anaesthesia of free-ranging wolverines, brown bears, black and white rhinoceros were physiologically evaluated. The effect of intranasal oxygen supplementation on arterial oxygenation was assessed in brown bears and rhinoceros. Partial reversal of the opioid effect and different body positions were evaluated in rhinoceros. Capture methods used included darting from a helicopter or the ground, and physical restraint followed by drug injection. Medetomidine-ketamine was used in wolverines and medetomidine-zolazepam-tiletamine in primates, lions and brown bears. Rhinoceros were immobilised with a combination of an opioid, azaperone and an alpha2-agonist. Body temperature and cardiorespiratory variables were monitored in all animals. Arterial blood samples were analysed to interpret pulmonary gas exchange and acid-base status in carnivores and rhinoceros. Low doses of medetomidine-zolazepam-tiletamine rapidly anaesthetised primates and lions, and reversal with atipamezole resulted in a smooth and calm recovery. Physiological alterations varied with different protocols and species and included changes in body temperature, respiratory and heart rates, gas exchange and acid-base balance. Hypoxaemia was recorded in all rhinoceros and most carnivore species. The major contributor to hypoxaemia was likely ventilation-perfusion mismatch including shunt. In rhinoceros, hypoventilation contributed to an impaired gas exchange and the animals remained hypoxaemic despite partial reversal. However, in black rhinoceros arterial oxygenation was higher during sternal compared to lateral recumbency. Capture-induced lactic acidaemia was recorded in carnivores and rhinoceros. Intranasal oxygen supplementation improved arterial oxygenation. In conclusion, this thesis increases the understanding of the effects of capture and anaesthesia in several wildlife species. Physiological derangements were identified, potential causative factors were investigated and methods for improvement were evaluated.

▼ Introduction: Despite major advances in renal replacement therapy acute kidney
injury (AKI) is still associated with high morbidity and mortality. Beside
risk factors like mechanical ventilation, acidosis has been identified as one
factor contributing to AKI. However, in critically ill patients with multiple
risk factors a differentiated analysis of the influence of acidosis is hard to
perform. Therefore, the aim of our study was to evaluate the impact of an
acidaemia on renal function excluding sepsis or other infections. Methods: 35
young and healthy pigs were randomized into 6 groups. Three test pairs were
settled with or without exposition to a continuos veno-venous haemofiltration
(CVVH), respectively. The groups differed by: normoxaemia-balanced
acid-
base-
status (NB-CVVH and NB-CTRL), normoxaemia-acidaemic (NA-CVVH and NA-CTRL) and
hypoxaemia-acidaemic (HA-CVVH and HA-CTRL). Due to low-tidal-volume-
ventilation and
acid-infusion a mixed acidaemia (pH 7.19-7.24) was induced
into acidaemic groups. HA-groups underwent an additional hypoxaemia. Acidemia
and hypoxaemia were maintained for five hours. Haemodynamics, ventilation
parameter, blood gas analysis and diuresis were monitored constantly as well
as interleukin-6 (IL-6), TNF-aplha and interleukin-18 (IL-18) in blood and
tissue samples, neutrophil gelatinase-associated lipocalin (NGAL) was analyzed
in urine specimen. Tissue samples of the kidneys were (immuno-) histologically
examined. Results: All animals were haemodynamically stable throughout the
entire experiment. Animals had sufficient diuresis facing a physiologic
cardiac output at any time. Interestingly, diuresis increased in acidemia
groups while urine-pH decreased. Increased plasma levels of IL-18 and NGAL
indicated a developing AKI in the acidaemia groups. Concordantly, normoxic-
acidaemia groups had significantly more reversible and irreversible cell
damage in histopathological examination, just exceeded by hypoxaemia-
acidaemia groups. Due to measurements of proinflammatory cytokines in tissue
and plasma a biotrauma induced AKI was excluded, because subgroup analysis of
immunohistology revealed an inverse correlation between tissue levels of
proinflammatory cytokines and histopathological cell damage. Conclusion: We
were able to demonstrate that an isolated acidaemia without concomitant sepsis
already leads to a relevant histologically detectable damage of the kidneys
after only five hours exposition to acidaemia. Immunohistochemical
investigations showed that other than inflammatory factors induced the renal
damage. Results are leading to the hypothesis that the acidaemia works as a
kind of „first hit“ by increasing renal workload with respect to the
correction of
acid-
base-
status abnormalities and thereby contributing to a
depletion of renal energy reserves. Initially increasing diuresis clinically
masks the early onset of an AKI due to the compensation for acidosis by the
kidneys.

Perante a suspeita de intoxicação aguda, a abordagem célere e sistemática ao doente, permite uma rápida prestação de cuidados específicos. A triagem, recolha da história clínica, avaliação e intervenção médica de urgência, o diagnóstico e a terapêutica são processos chave nessa abordagem. O diagnóstico, por norma, é suportado por um painel analítico inicial. O recurso a testes imediatos (“point of care”), permite avaliar não só a condição geral dos doentes em tempo real, mas também os efeitos que os xenobióticos possam ter a nível orgânico e que se traduzam, nomeadamente, em alterações do equilíbrio hidroeletrólitico e estado ácido base.
O presente estudo pretendeu avaliar as alterações analíticas, detetadas através da ferramenta ePOC (“Enterprise Point of Care”), em animais suspeitos de intoxicação por inseticidas anticolinesterásicos e a sua eventual influência no diagnóstico e implementação da terapêutica. A amostra estudada consistiu em 14 canídeos, que na triagem médica apresentavam sintomas inespecíficos e outros compatíveis com intoxicação aguda por insecticidas anticolinesterásicos: tremores musculares (93%), hipersiália (64%), hipertermia (57%), cianose (50%), diarreia (57%) e vómito (36%). Foram colhidas amostras de sangue para a realização do teste ePOC e para ulterior análise toxicológica.Os resultados obtidos através do ePOC mostraram diminuição da pressão parcial de dióxido de carbono (pCO2) (64%), aumento da concentração de lactato (36%), aumento dos valores de hemoglobina e hematócrito (50%), hiperglicémia (36%) e aumento dos valores de creatinina (50%), não havendo, contudo, relação estatisticamente significativa entre as variáveis testadas e o diagnóstico toxicológico (p>0.05). Porém, em todos os animais considerados acidémicos houve detecção de compostos do grupo dos inseticidas anticolinesterásicos na análise toxicológica. O estudo clínico individualizado de cada caso permitiu verificar que, apesar da sintomatologia similar, os animais apresentavam distúrbios ácido-base diferentes, sendo por isso também necessária uma abordagem terapêutica diferenciada, fulcral especialmente em condições críticas.
Estudos futuros, com uma amostra de maior dimensão e preferencialmente com exposição a diferentes xenobióticos, poderão permitir uma avaliação mais exata e abrangente da relação entre a etiologia das intoxicações e as alterações hidroeletrolíticas e de gases sanguíneas verificadas.

▼ A insuficiência renal aguda (IRA) em cadelas com piometra foi avaliada retrospectivamente em 147 animais e prospectivamente em 351 animais selecionando 27 e 132 cadelas respectivamente, mediante os seguintes critérios de inclusão, creatinina ≥ 2,4 mg/dL e ou uréia ≥ 80 mg/dL e ou aumentos relativos de 100% da creatinina e ou uréia séricas em relação a qualquer mensuração anterior, aplicados em 3 momentos estratégicos, o diagnóstico, 24 horas após a ovário-salpingo-histerectomia e na retirada dos pontos. Os animais do estudo retrospectivo foram utilizados como controle, enquanto que os animais selecionados do estudo prospectivo receberam tratamento conservador com solução de Ringer Lactato até 90 ml/kg/h, durante 4 horas. Os parâmetros avaliados antes e após a terapia, peso, pressão arterial sistólica, freqüência cardíaca, função renal, hemogasimetria arterial, bioquímica sérica e urinária e "clearance" da creatinina endógena. Os animais que não responderam adequadamente a terapia convencional e apresentavam-se em "síndrome urêmica" foram submetidos à hemodiafiltração (N=5 animais) e prescrito sessões de 2 horas, durante 3 dias, soluções apropriadas para hemodiafiltração (HDF) e reposição e capilar de polissulfona. Durante a HDF 2 animais vieram a óbito, 1 animal morreu com 18 dias e outro 12 dias após a última sessão de diálise e 1 animal sobreviveu. A taxa de mortalidade para os animais selecionados retrospectivamente foi de 40,7% e 20,4% prospectivamente. Entretanto os animais selecionados do estudo prospectivo que não sobreviveram foram comparados com os animais selecionados que sobreviveram, mediante análise univariada da uréia e creatinina séricas, fração de excreção de eletrólitos, bicarbonato arterial e "clearance" da creatinina e foram significativamente diferentes (p<0,05). Porém quando estes dados foram submetidos a uma análise multivariada e realizada a curva ROC (Receiver Operating Characteristic), detectou-se que a área sob a curva era de 0,867 (p<0,0001) IC: 0,769 ? 0,966. Desta forma foi possível concluir que a creatinina maior que 2,6 mg/dL prediz a morte da cadela com piometra e IRA, com 93% de sensibilidade e 77% de especificidade. Quanto aos animais que receberam a HDF e morreram apresentavam uma sobrevida de 7 dias em relação aos animais que receberam o tratamento conservador apenas, podendo a HDF ser um método de aumentar a sobrevida, dando tempo para o animal se recuperar.

IN THIS INVESTIGATION WE STUDIED THE MATERNAL AND NEONATAL ACID-BASE STATUS OF THE RABBIT AFTER PROPOFOL ANAESTHESIA FOR CAESAREAN SECTION. WE USED 2 GROUPSOF 20 DOES EACH. THE ANAESTHESIA WAS INDUSED, IN BOTH GROUPS, WITH A SINGLE DOSE OF PROPOFOL, WHILE IT WAS MAINTAINED WITH BOLUS, IN THE FIRST GROUP, ANDCONTINUOUS, IN THE SECOND, INFUSIONS OF PROPOFOL. WE TOOK, EVERY 5 MINUTES DURING THE ANAESTHETIC PERIOD, AND ALSO AT THE FIRST 5 MINUTES OF RECOVERY, SAMPLES OF ARTERIAL BLOOD FROM THE PREGNANT DOES, WHERE PH, PCO2, PO2 AND HCO-3WERE CALCULATED IN ORDER TO EVALUATE THEIR ACID-BASE STATUS. SIMILAR SAMPLESWERE OBTAINED FROM, 60 PER GROUP, NEONATES IMMEDIATELY AFTER THEIR DELIVERY WITH CAESAREAN SECTION AND 24 HOURS LATER. BASED ON THE MATERNAL ARTERIAL BLOOD MEASUREMENTS, DURING THE ANAESTHETIC PERIOD, IN BOTH GROUPS, WE OBSERVED THE PRESENCE OF AN ACUTE RESPIRATORY ACIDOSIS. THIS ACID-BASE DISORDER WAS RESTORED DURING THE RECOVERY PERIOD. THE NEONATAL ACID-BASE STATUS IMMEDIATELY AFTER THEIR DELIVERY, IN BOTH GROUPS, WAS THAT OF A RESPIRATORY ACIDOSIS, WHICHWAS RESTORED 24 HOURS LATER. ALTHOUGH PROPOFOL CAUSES A MILD MATERNAL RESPIRATORY DEPRESSION OF THE PREGNANT DOE, IT DOESN'T SEEM TO ADVERSE EFFECT THE PHYSIOLOGICAL DEVELOPMENT OF THE NEONATAL ACID-BASE STATUS.

▼ The aim of this study was to get further information about the influence of
different factors to the effect of anionic rations, to optimize the use of
anionic salts in the prevention of milk fever of dairy cows. These factors are
the influence of a prolonged administration of anions, of different energy and
calcium supply, the effects of the sodium-potassium relation, the maximal
tolerated dose of calcium sulfate as well as the investigation about the
application of the daily dose of calcium chloride and calcium sulfate once a
day.
From february 2003 to march 2004 the investigations were performed on cows
with rumen fistula. During the trials the animals got the
acid salts calcium
chloride and calcium sulfate through the rumen fistula twice a day at the
feedings (except for the trial about the once a day application and the trial
with sodium- and potassium bicarbonate). Several times a week blood and urine
samples were taken to determine the blood gas analysis (ph, be, CO2, O2) as
well as the net
acid-
base excretion. For each trial the feed was analysed and
the dcad of the ration was measured. In all 1401 blood and urine samples have
been taken and examined.
The prolonged administration of anions showed, that the entire effect of the
anionic salts is completely developed after 3 7 days. A significant
metabolic acidosis set in. The values of ph, be and bicarbonate of the blood
decreased. In the urine the effect of the acidosis was observed as well. There
was a significant decrease in ph and nsba. Because of the fact, that the
complete effects are developed within 3 7 days, and because of the risk of
strong disturbances of the
acid-
base equilibrium, the use of the
acid salts
should last 7 days at least, but not longer than 14 days. The results of the
investigation about the different energy supply showed, that the concentration
of energy should be sufficient to avoid a further increase of the acidosis by
the metabolic products, which result from the degradation of fatty and amino
acid at catabolism. The degree of the calcium supply had no significant
influence of the impact of the anionic salts relating to the
acid-
base status.
Never the less, when feeding
acid salts, it is very important to supply
calcium in sufficient amount (120 180g/d), because of the strong renal
calcium excretion. The investigation of the impact of sodium and potassium
showed, that cations can counteract and compensate the effects of the anions
and with it the mechanism of the prophylaxis of milk fever.
The application of anions and cations in equivalent amounts at the same time
had no effect of the
acid-
base-balance. The exclusive use of sodium- and
potassium bicarbonate did not show the expected alkaline effects. The
investigation of the maximal tolerated dose of calcium sulfate showed, that
the cows developed a strong metabolic acidosis after one week of application
of 3 equivalents of calcium sulfate. Because of that, the dose of 3
equivalents should not be exceeded. It is better to think of 2,5 equivalents

▼ The aim of this study was to find a solution how to valuate the effect of
anionic rations with herd datas and to make the use of anions safer. The first
part of the study shows the examinations about the practical use of the
measurement of urine ph to valuate the effects of anions on the
acid-
base
status. For that purpose, samples of urine were taken for eight weeks from the
two weeks prepartum cows of two different farms. The ph was measured with an
electrical ph-meter in the cowshed and with ph-stripes directly after the
winning. Apart from that, the urine was frozen and after defrosting measured
with an electrical ph-meter in the lab. The measurement of the ph by ph-
stripes showed nearly the similar results than the measurement of the urine ph
by electrical ph-meter directly after the winning. After defrosting the urine,
the results of the urine ph were slightly increased. Within the scope of herd
management fifty farms were visited and some of them more than one time. Blood
and urine samples were taken from close up dry cows and recently calved cows.
The ration of the close up dry cows was analysed. Ten single samples of blood
and urine were necessary to produce a pool sample. The values of the pool
samples of urine ph, net
acid-
base excretion and urine calcium excretion were
compared with the mean values of the single samples. There were high
correlations between the pool and the mean values for each of the parameters.
Because of that, the pool and the mean values have nearly the same content of
information. The analyses of the rations from 2002 to 2004 contain the dcad
and the amount of potassium, sodium, chloride and sulphur. Herds with and
without anionic rations were compared. The analysis of the feed of herds with
anionic rations showed a lower dcad and a higher amount of sulphur in all of
the three years. But the recommendations of the NRC were not achieved. That is
an indication that not enough anionic salts were used refer to the rations
content. To examine the influence of more factors of the ration on the effect
of the anions the urine ph, the net
acid-
base excretion and the urine calcium
excretion were compared with the parameters of the rations. The effect of
reducing the dcad and the amount of potassium as well as the effect of
increasing the amount of calcium and sulphur in the ration is a mild acidosis
and an increased urine calcium excretion. These are the effects of the anions.
Within feeding anions there should be a shift of the major elements. This
leads to a decreased dcad and to the metabolic effects as described. The same
effects on the
acid-
base status and the urine calcium excretion were observed
by feeding an increased amount of starch and a decreased amount of crude
fibre. This could simulate the effect of anionic rations. The parameters of
the energy concentration in the rations have a slight correlation with the
parameters of the
acid-
base status. At low amounts of energy there is an
